Hoyle Abstract Aims:, To examine diadenosine tetraphosphate (Ap4A) for its ability to protect the eye from neurodegeneration induced by subconjunctival application of 6-hydroxydopamine (6-OHDA). Methods:, Intraocular neurodegeneration of anterior structures was induced by subconjunctival injections of 6-OHDA. Animals were pre-treated with topical corneal applications of Ap4A or saline. Results:, 6-OHDA caused miosis, abnormal pupillary light reflexes, a precipitous drop in intraocular pressure and loss of VMAT2-labelled (vesicle monoamine transporter-2, a marker for sympathetic neurones) intraocular neurones. Pre-treatment with Ap4A prevented all of these changes from being induced by 6-OHDA, demonstrably preserving the sympathetic innervation of the ciliary processes. This neuroprotective action of Ap4A was not shared with the related compounds adenosine, ATP or diadenosine pentaphosphate. P2-receptor antagonists showed that the effects of Ap4A were mediated via a P2-receptor. Conclusion:, Ap4A is a natural component of tears and aqueous humour, and its neuroprotective effect indicates that one of its physiological roles is to maintain neurones within the eye. Ap4A can prevent the degeneration of intraocular nerves, and it is suggested that this compound may provide the basis for a therapeutic intervention aimed at preventing or ameliorating the development of glaucoma associated with neurodegenerative diseases. Furthermore, subconjunctival application of 6-OHDA provides a useful model for studying diseases that cause ocular sympathetic dysautonomia. Robinson Abstract Suspended sediments are a natural component of aquatic ecosystems, but anthropogenic activity such as land development can result in significant increases, especially after rain events. Continuous exposures of suspended clay and silt have been shown to affect growth and reproduction of Cladocera, leading to a decrease in population growth rate. The mechanism of clay toxicity in these filter-feeding organisms is clogging of the gut tract, resulting in decreased food uptake and assimilation. When placed in clean water, daphnids can purge clay from their gut and recover. In many surface waters, aquatic organisms experience episodic exposures of high concentrations of suspended solids driven by rain events. However, little is known about the consequences of pulsed exposures on individuals and populations. The objective of the present study was to characterize the effects of continuous and pulsed exposures of natural and defined clays on survival, growth, and reproduction of Daphnia magna. Two defined clays, montmorillonite and kaolinite, as well as clay isolated from the Piedmont region of South Carolina, USA, were used. Continuous exposures of clays elicited a dose dependent decrease in survival. Toxicity varied depending on clay source with montmorillonite,>,natural clay,>,kaolinite. Pulsed exposures caused a decrease in survival in a 24 h exposure of 734 mg/L kaolinite. Exposure to 73.9 mg/L also caused an increase in the time to gravidity, although there was not a corresponding decrease in neonate production over 21 d. No significant effects resulted from 12 h exposures even at 730 mg/L, almost 10 times the 24-h reproductive effects concentration. This suggests that exposure duration impacted toxicity more than exposure concentration in these pulsed exposures. Environ. Toxicol. Hannon Abstract 1The history of a forest stand over the last 6000 years has been reconstructed by studying pollen, macrofossils and charcoal from a small, wet hollow in Suserup Skov on the island of Sjælland in eastern Denmark. 2The earliest recorded forest was Tilia -dominated but contained an intimate mixture of many different tree species that included Acer campestre, A. platanoides, Alnus glutinosa, Betula pubescens, Corylus avellana, Frangula alnus, Fraxinus excelsior, Malus sylvestris, Populus tremula, Pinus sylvestris, Quercus robur, Q. petraea, Salix spp., Sorbus aucuparia, Tilia cordata and T. platyphyllos. The preserved fruits of T. platyphyllos confirm its hitherto doubtful status as a native member of the Danish flora. 3The present-day woodland developed after a period of intensive anthropogenic disturbance between , 600 bc and ad 900, during which time open canopy conditions prevailed at Suserup. Fagus sylvatica and Fraxinus excelsior are the dominant trees at present, together with some Quercus robur and Ulmus glabra.4,Charcoal was present in the sediments from most time periods except at the Ulmus decline. In the last 1000 years of the sequence , the period of Fagus dominance , charcoal counts were consistently low. 5Pinus sylvestris was a natural component of this primarily deciduous forest, and the last macrofossil find dates from c. ad 900. Macrofossil Pinus cone scales recorded c. ad 1800 originate from planted individuals. Prior to Fagus dominance, the forest had an open structure partly caused by frequent, low-intensity fires associated with the presence of Pinus sylvestris. 6The replacement of Tilia by Fagus in this forest was catalysed by human activity. If the forest had not been so disturbed, the rich diversity of trees would most probably have persisted up to the present time, with only a moderate-sized Fagus population. [source] Palaeovegetational and palaeoenvironmental trends in the summit of the Guaiquinima massif (Venezuelan Guayana) during the Holocene,JOURNAL OF QUATERNARY SCIENCE, Issue 2 2005Valentí Rull Abstract The summits of the table mountains (tepuis) from the Neotropical Guayana region are remote environments suitable for palaeoecological studies with evolutionary, biogeographical and palaeoclimatic implications. Here, using palynological analyses of two radiocarbon-dated peat bogs from a tepui summit, the Holocene palaeovegetational trends are reconstructed, and related to possible forcing factors. Because of the pristine character of the Guaiquinima summit, the recorded palaeoenvironmental changes are probably due to natural causes, which makes them valuable archives of the natural component of climatic change at a millennial time scale. The sequence begins with pioneer communities or meadows similar to present-day ones, between about 8.4 and 4.5,ky BP. After this date, and until about 2,kyr BP the expansion of gallery forests suggests an increase in precipitation, documented also at regional (Neotropical) level. Between ca. 2,kyr BP and the last century, gallery forests are replaced by forests characteristic of the upper Guaiquinima altitudes, coinciding with a regional phase of reduced moisture. The present-day meadows, established relatively quickly during the last century, substituted the former upland forests. In the locality studied, the main controlling factor of the vegetation during the Holocene seems to have been the moisture balance. In contrast to other tepui summits, there is no clear evidence for changes linked to temperature oscillations. This could be due to the elevation of the site, far from any characteristic ecological boundary, that makes it insensitive to this parameter. Copyright © 2005 John Wiley & Sons, Ltd. [source] Effects of Bifidobacterium bifidum on adaptive immune senescence in aging miceMICROBIOLOGY AND IMMUNOLOGY, Issue 10 2010Yu-Rong Fu ABSTRACT Bifidobacteria are a natural component of the bacterial flora of the human body and have a symbiotic bacteria-host relationship with human beings. Aging is associated with reduced numbers of beneficial colonic Bifidobacteria and impaired immunity. The possible anti-senescence effects of Bifidobacteria are presently unknown. The aims of the present study were to investigate possible anti-senescence effects of B. bifidum on naturally senescent mice and to explore their mechanisms. After treatment with B. bifidum, mice were killed and samples collected. Cytokine production in serum and lymphocyte culture supernatant, anti-oxidation activity and gene expression were measured. B. bifidum significantly increased cytokine IL-2 and IFN-, levels but decreased proinflammatory cytokines IL-6 and TNF-, concentrations. Moreover, B. bifidum improved anti-oxidation activity and reduced lipid peroxidation in thymus and spleen. In addition, B. bifidum down-regulated p16 expression in thymus and spleen. Taken together, the results indicate, for the first time, that B. bifidum delays senescence by several mechanisms, including enhancement of anti-oxidation activity in thymus and spleen, alteration of gene expression and improvement in immune function. [source] Intracerebral schwannoma in a child with infiltration along perivascular spaces resembling meningioangiomatosisPATHOLOGY INTERNATIONAL, Issue 8 2009Misa Ishihara Schwannoma arising within brain parenchyma is a rare lesion, usually found in children. Reported herein is a case of intracerebral schwannoma in a 5-year-old boy, with a review of the English-language literature on the subject, in which 47 cases were found. Few detailed histological reviews of intracerebral schwannoma exist. The tumor had a distinctive plexiform growth pattern, and small aggregates of Schwann cells spread extensively into the surrounding brain tissue along perivascular spaces adjacent to the tumor nodule. Histological differential diagnoses included perivascular schwannosis and meningioangiomatosis. A few intratumoral axons, seen on immunostaining for neurofilament protein, were trapped at the periphery of the main lesion, but there was no evidence of intralesional axons in the multiple nodules of Schwann cell proliferations that extended into the perivascular spaces, suggesting that the lesions are neoplastic. Because Schwann cells are not a natural component of the central nervous system, the origin of intracerebral schwannomas remains unknown. The histology suggests that Schwann cells of the perivascular nerve plexus are a likely site of origin. Luo (Cancer Sci 2010; 101: 707,712) (,)-Epigallocatechin-3-gallate (EGCG) is one of the major polyphenol components in green tea. It effectively induces apoptosis in prostate cancer cells. The anticancer effect of this reagent is appealing because it is a natural component of a popular daily beverage that has proven harmless for thousands of years, making it a good candidate chemopreventive agent. EGCG suppresses cell growth and causes cell death, but the mechanisms are not well characterized, especially in androgen-independent prostate cancer cells. In the present study, using Affymetrix genechip Hu133 2.0, we analyzed the gene expression patterns of the androgen-independent prostate cancer cell line Du145, treated with or without EGCG, and found 40 genes whose expression levels were altered (>twofold, either upregulated or downregulated, P < 0.01) upon treatment with EGCG. These gene products are involved in the functions of transcription, RNA processing, protein folding, phosphorylation, protein degradation, cell motility, and ion transport. Among them, inhibitor of DNA binding 2 (ID2), known as a dominant anti-retinoblastoma (Rb) helix-loop-helix protein, was found to be downregulated fourfold by EGCG treatment. Forced expression of ID2 in Du145 cells reduced apoptosis and increased cell survival in the presence of EGCG, and knockdown ID2 expression in Du145 cells using a morpholino oligonucleotide specific for ID2 mimicked the apoptosis effect generated by EGCG treatment, although it was milder. To our knowledge, this is the first report indicating that ID2 is one of the critical factors in the signaling pathway of Du145 cell death induced by EGCG. [source] Effective Insect Repellent Formulation in both Surfactantless and Classical Microemulsions with a Long-Lasting Protection for Human BeingsCHEMISTRY & BIODIVERSITY, Issue 6 2009Jeremy Drapeau Abstract The aim of this work is to develop a new generation of repellent products with a long-lasting protection based on a natural component, para -menthane-3,8-diol (PMD). The active is first rendered soluble in a surfactantless microemulsion (H2O/iPrOH/PMD) and then in classical microemulsions. The presence of self-associated nanostructures is detected by dynamic light scattering (DLS). A synergetic system of surfactants (Cremophor®RH40 and Texapon®N70) is used. Additionally, 2-ethylhexane-1,3-diol and ethyl (,)-(S)-lactate are incorporated. The final product contains, as main components, 46% of H2O, 25% of iPrOH, 20% of non-H2O-soluble PMD, and only 4% of surfactants. Investigations of lasting protection on human volunteers are carried out using a cage test bioassay protocol and Aedes aegypti mosquitoes. A complete protection of 315,min is found on the test persons using the surfactantless microemulsion. An extension is observed with the final formulation to reach a mean of complete protection of 385,min. This study demonstrates that alternative formulations using a natural active instead of synthetic chemicals like N,N -diethyl- m -methylbenzamide (DEET) can be efficient for human protection against mosquitoes. Their presence in food, either as natural components (e.g. phytoestrogens) or chemical contaminants (e.g. xenoestrogenic pesticides) have been implicated in a number of human effects (e.g. declining sperm count). We present calculations which ranks the importance of the different classes of dietary endocrine disrupters, and show that only phytoestrogens are likely to have a health impact. [source] |
