Rumyantsev Abstract In addition to the great number of publications focused on the leading role of virus mutations and reassortment in the origin of pandemic influenza, general opinion emphasizes the victim side of the epidemic process. Based on the analysis and integration of relevant ecological, epidemiological, clinical, genetic and experimental data, the present article is focused on the evolution of ,virus , victim' ecological systems resulting in the formation of innate (i.e. genetic, constitutional) immunity in the involved species and populations. This kind of immunity functions today as the greatest natural barrier to the pandemic spread of influenza among humans and ecologically related kinds of animals. Global influenza pandemics can arise when the worldwide population contains at least a minimum number of people susceptible to a known or mutant influenza virus. Special attention is paid in this article to individual tests for the presence of this barrier, including the implications of specific findings for public health policy. Such tests could be based on in vitro observation of the action of relevant virus strains on primary cell cultures or on their cellular or molecular components extracted from individuals. Godavarthy Abstract One promising way to breach the skin's natural barrier to drugs is by the application of chemicals called penetration enhancers. However, identifying potential enhancers is difficult and time consuming. We have developed a virtual screening algorithm for generating potential chemical penetration enhancers (CPEs) by integrating nonlinear, theory-based quantitative structure,property relationship models, genetic algorithms, and neural networks. Our newly developed algorithm was used to identify seven potential CPE molecular structures. These chemical enhancers were tested for their toxicity on (a) mouse embryonic fibroblasts (MEFs) with MTT assay, and (b) porcine abdominal skin by histology using H/E staining at the end of a 48-h exposure period to the chemicals. Further, melatonin permeability in the presence of the enhancers was tested using porcine skin and Franz diffusion cells. Careful toxicity tests showed that four of the seven "general" CPEs were nontoxic candidate enhancers (menthone, 1-(1-adamantyl)-2-pyrrolidinone, R(+)-3-amino-1-hydroxy-2-pyrrolidinone, and 1-(4-nitro-phenyl)-pyrrolidine-2,5-dione). Further testing of these four molecules as potential melatonin-specific CPEs revealed that only menthone and 1-dodecyl-2-pyrrolidinone provided sufficient enhancement of the melatonin permeation. The results from our permeability and toxicity measurements provide validation of the efficacy and ability of our virtual screening algorithm for generating potential chemical enhancer structures by virtual screening algorithms, in addition to providing additional experimental data to the body of knowledge. © 2009 Wiley-Liss, Inc. and the American Pharmacists Association J Pharm Sci 98:4085,4099, 2009 [source] Preliminary evaluation of ,interpreter' role plays in teaching communication skills to medical undergraduatesMEDICAL EDUCATION, Issue 3 2001K C J Lau Rationale and objectives Multiculturalism presents linguistic obstacles to health care provision. We explored the early introduction of ,interpreter' role-play exercises in teaching medical undergraduates communication skills. The interpreter role creates a natural barrier in communication providing an active prompt for recognizing learning needs in this area. Methods Bilingual Cantonese first-year medical students (n=160) were randomly allocated to either ,Observer' or ,Interpreter' role plays at a small-group introductory communication skills workshop using a quasi experimental design, counterbalanced across tutors. Students assessed their own skill competence before and, together with their perceptions of the different role plays' effectiveness, again after the workshop, using an anonymous 16 item Likert-type scale, analysed using ANOVA and MANOVA. Results Students' assessments of their skills improved significantly following the workshop (F=73·19 [1,156], P=0·0009). Students in the observer group reported greater changes in their scores following the workshop than did students in the interpreter group (F=4·84 [1,156], P=0·029), largely due to improvement in perceived skill (F=4·38 [1,156], P=0·038) rather than perceived programme effectiveness (F=3·13 [1,156], P > 0·05). Subsequent MANOVA indicated no main effect of observer/interpreter conditions, indicating these differences could be attributed to chance alone (F=1·41 [16 141], P > 0·05). Conclusion The workshop positively influenced students' perceived communication skills, but the ,Interpreter' role was less effective than the ,Observer' role in achieving this. Future studies should examine whether interpreter role plays introduced later in the medical programme are beneficial. [source] The importance of mangrove forest in tsunami disaster mitigationDISASTERS, Issue 2 2009Rabindra Osti Tsunamis and storm surges have killed more than one million people and some three billion people currently live with a high risk of these disasters, which are becoming more frequent and devastating worldwide. Effective mitigation of such disasters is possible via healthy coastal forests, which can reduce the energy of tsunamis. In recent years, these natural barriers have declined due to adverse human and natural activities. In the past 20 years, the world has lost almost 50 per cent of its mangrove forests, making them one of the most endangered landscapes. It is essential to recover them and to use them as a shield against a tsunami and as a resource to secure optimal socio-economic, ecological and environmental benefits. This paper examines the emerging scenario facing mangrove forests, discusses protection from tsunamis, and proposes a way to improve the current situation. We hope that practical tips will help communities and agencies to work collectively to achieve a common goal. BERGHOFF Abstract The army ant Eciton burchellii is probably the most important arthropod predator in the Neotropics, and many animal species depend upon it. Sex-biased dispersal with winged males and permanently wingless queens may render this species especially sensitive to habitat fragmentation and natural barriers, which might have severe impacts on population structure and lead to population decline. Using nuclear microsatellite markers and mitochondrial sequences, we investigated genetic differentiation in a fragmented population in the Panama Canal area. While nuclear markers showed little differentiation between subpopulations (FST = 0.017), mitochondrial differentiation was maximal in some cases (,ST = 1). This suggests that, while females are not capable of crossing barriers such as large rivers, flying males are able to promote nuclear gene flow between the studied forest patches. Consistent with this interpretation, we did not find any evidence for inbreeding or genetic deterioration on Barro Colorado Island over the last 90 years since its formation. [source] Effects of natural barriers on the spillover of a marine mollusc: implications for fisheries reservesAQUATIC CONSERVATION: MARINE AND FRESHWATER ECOSYSTEMS, Issue 6 2003Alexander Tewfik Abstract 1.The movement of organisms and dispersal of propagules is fundamental to the maintenance of populations over time. However, the existence of barriers, created through the spatial configuration of habitats, may significantly affect dispersal patterns and thus influence community dynamics and resource sustainability. 2.Within marine environments unstructured or open habitats may form partial or complete ecological barriers due to elevated risk of predation or physical stresses associated with them. The existence and effects of such barriers may be of particular importance when considering the establishment of marine protected areas with a fisheries enhancement focus. 3.In this paper, the spillover of post-settlement queen conch (Strombus gigas) from a protected area in the Turks and Caicos Islands is investigated. It is hypothesized that the reserve boundaries overlap with a series of shallow, sand habitats that effectively enclose the protected population, reducing the spillover of conch into the adjacent fished areas. 4.To test this, density gradient maps for juvenile and adult conch populations were constructed using underwater visual survey data at 68 sites within and surrounding the protected area. These maps illustrate very low densities coinciding with poor, shallow sand habitats along the two marine boundaries of the reserve where spillover is expected to take place. 5.These sand habitats are thought to create ecological barriers to a slow, sedentary gastropod largely due to their shallowness (physical stresses of solar exposure or anoxia) and lack of food reducing the tendency of individuals to move across these areas, despite the 10-times higher density of adult queen conch observed in the protected area compared with outside.
