Kinds of Mycelium Terms modified by Mycelium Selected AbstractsSPECIFIC DETECTION OF AMANITA PHALLOIDES MYCELIUM AND SPORES BY PCR AMPLIFICATION OF THE GPD (GLYCERALDEHYDE-3-PHOSPHATE DEHYDROGENASE) GENE FRAGMENTJOURNAL OF FOOD BIOCHEMISTRY, Issue 3 2000OWSKI, ROMAN KOT ABSTRACT Oligonucleotide primers designed to flank a 635 bp fragment of the gene encoding glyceraldehyde-3-phosphate dehydrogenase (gpd) from Araanita muscaria were used to amplify the corresponding gpd fragment from Amanita phalloides. The A. phalloides PCR product was cloned, sequenced and found to be 70 - 77% similar to the known basidiomycetes gpd genes within the exon part and 25 - 52% within the intron part. Based on these data, species-specific amplification was achieved using a pair of oligonucleotide primers complementary to the A. phalloides gpd intron sequences. These primers allowed the amplification of the corresponding gpd fragment from the A. phalloides but not from various other basidiomycetes, ascomycetes and human matrices. PCR amplification of the A. phalloides DNA gave the predicted PCR product of 284 bp. The created PCR system is an efficient tool for the specific, rapid and sensitive detection of A. phalloides mycelium and spores. [source] A Suspension Culture Method for the Rapid Mass Culture of Cistella japonica MyceliumJOURNAL OF PHYTOPATHOLOGY, Issue 9 2006T. Yamanobe Abstract Different methods were investigated for the rapid mass culture of Cistella japonica by using water extracts of some nutritional sources. In an agar culture test, there was little difference in mycelial growth in water extracts of wheat bran, rice bran and potato. In suspension culture with wheat bran extract, which is easily and cheaply available, the mycelium of C. japonica increased seven times more than that in agar culture after a month's incubation. C. japonica from suspension culture was pathogenic to Chamaecyparis obtusa. These results suggest that suspension culture in water extract of wheat bran can be adopted for the rapid mass culturing of C. japonica for use in inoculation tests. [source] Patterns of Interaction between Populus Esch5 and Piriformospora indica: A Transition from Mutualism to AntagonismPLANT BIOLOGY, Issue 2 2005M. Kaldorf Abstract: Piriformospora indica (Sebacinaceae, Basidiomycota) is an axenically cultivable, plant growth promoting root endophyte with a wide host range, including Populus. Rooting of Populus Esch5 explants started within 6 days after transfer to WPM medium. If such plantlets with roots were inoculated with P. indica, there was an increase in root biomass, and the number of 2nd order roots was increased significantly. A totally different observation was recorded when the explants were placed into WPM with pre-grown P. indica. The interaction led to complete blocking of root production and severely inhibited plant growth. Additionally, branched aerial roots appeared which did not penetrate the medium. On contact with the fungal colony or the medium, the ends of the aerial roots became inflated. Prolonged incubation stimulated the fungus to colonize aerial parts of the plant (stem and leaves). Mycelium not only spread on the surface of the aerial parts, but also invaded the cortical tissues inter- and intracellularly. Detached Populus leaves remained vital for 4 - 5 weeks on sterile agar media or on AspM medium with pre-grown P. indica. When the fungus was pre-grown on culture media such as WPM, containing ammonium as the main source of nitrogen, leaves in contact with the cultures turned brownish within 4 - 12 h. Thereafter, the leaves bleached, and about one day later had become whitish. Thus, cultural conditions could alter the behaviour of the fungus drastically: the outcome of the interaction between plant and fungus can be directed from mutualistic to antagonistic, characterized by fungal toxin formation and extension of the colonization to Populus shoots. [source] Plants, gall midges, and fungi: a three-component systemENTOMOLOGIA EXPERIMENTALIS ET APPLICATA, Issue 1 2008Odette Rohfritsch Abstract Larvae of gall midges (Diptera: Cecidomyiidae) induce the activation of plant cells, partial cell lysis, and differentiation of nutritive tissue. Specialized nutritive tissue is essential for larval development and plays a key role in gall organization. Midges of the tribes Lasiopterini and Asphondyliini, however, do not induce nutritive tissues as part of the formation of their galls. Instead, these ,ambrosia galls' contain fungal mycelia that line the interior surface of the chambers. The fungi not only provide Lasiopterini with nutrition, they also penetrate the stems, induce the lysis of the middle lamella of host cells, and open a channel to the vascular bundles. Larvae of Lasioptera arundinis (Schiner) (Lasiopterini) follow the fungus and feed on its mycelium along with adjoining stem cells of Phragmites australis (Cav.) Trin. (Poaceae). Eggs together with fungal conidia are deposited by the imago on the host. Asphondyliini use a needle-like ovipositor to introduce fungal conidia and eggs into the organs they attack. Larvae of Schizomyia galiorum Kieffer (Asphondyliini) are unable to initiate the gall or to develop in the flowers of Galium mollugo L. (Rubiaceae) without their fungal associate. In this article, I provide an overview of oviposition behaviour in the Asphondyliini, as well as descriptions of the ovipositor and the female post-abdominal segments. Gall formation by Lasiopterini and Asphondyliini and the role of associated fungi are discussed, as is the role of the fungus as an inquiline or an organizer of gall tissues and a nutritive device. [source] The nature of tobacco resistance against Botrytis cinerea depends on the infection structures of the pathogenENVIRONMENTAL MICROBIOLOGY, Issue 1 2010Mohamed El Oirdi Summary To protect themselves, plants have evolved an armoury of defences in response to pathogens and other stress situations. These include the production of pathogenesis-related (PR) proteins and the accumulation of antimicrobial molecules such as phytoalexins. Here we report that resistance of tobacco to Botrytis cinerea is cultivar specific. Nicotiana tabacum cv. Petit Havana but not N. tabacum cv. Xanthi or cv. samsun is resistant to B. cinerea. This resistance is correlated with the accumulation of the phytoalexin scopoletin and PR proteins. We also show that this resistance depends on the type of B. cinerea stage. Nicotiana tabacum cv. Petit Havana is more resistant to spores than to mycelium of B. cinerea. This reduced resistance of N. tabacum cv. Petit Havana to the mycelium compared with spores is correlated with the suppression of PR proteins accumulation and the capacity of the mycelium, not the spores, to metabolize scopoletin. These data present an important advance in understanding the strategies used by B. cinerea to establish its disease on tobacco plants. [source] Volatile organic compounds: a potential direct long-distance mechanism for antagonistic action of Fusarium oxysporum strain MSA 35ENVIRONMENTAL MICROBIOLOGY, Issue 4 2009Daniela Minerdi Summary Fusarium oxysporum MSA 35 [wild-type (WT) strain] is an antagonistic Fusarium that lives in association with a consortium of bacteria belonging to the genera Serratia, Achromobacter, Bacillus and Stenotrophomonas in an Italian soil suppressive to Fusarium wilt. Typing experiments and virulence tests provided evidence that the F. oxysporum isolate when cured of the bacterial symbionts [the cured (CU) form], is pathogenic, causing wilt symptoms identical to those caused by F. oxysporum f. sp. lactucae. Here, we demonstrate that small volatile organic compounds (VOCs) emitted from the WT strain negatively influence the mycelial growth of different formae speciales of F. oxysporum. Furthermore, these VOCs repress gene expression of two putative virulence genes in F. oxysporum lactucae strain Fuslat10, a fungus against which the WT strain MSA 35 has antagonistic activity. The VOC profile of the WT and CU fungus shows different compositions. Sesquiterpenes, mainly caryophyllene, were present in the headspace only of WT MSA 35. No sesquiterpenes were found in the volatiles of ectosymbiotic Serratia sp. strain DM1 and Achromobacter sp. strain MM1. Bacterial volatiles had no effects on the growth of the different ff. spp. of F. oxysporum examined. Hyphae grown with VOC from WT F. oxysporum f. sp. lactucae strain MSA 35 were hydrophobic whereas those grown without VOCs were not, suggesting a correlation between the presence of volatiles in the atmosphere and the phenotype of the mycelium. This is the first report of VOC production by antagonistic F. oxysporum MSA 35 and their effects on pathogenic F. oxysporum. The results obtained in this work led us to propose a new potential direct long-distance mechanism for antagonism by F. oxysporum MSA 35 mediated by VOCs. Antagonism could be the consequence of both reduction of pathogen mycelial growth and inhibition of pathogen virulence gene expression. Smits Summary A method for the quantitative analysis of the hydrophobicity of the mycelial mat of filamentous fungi based on contact angle measurements is presented. It was tested for a range of fungi belonging to the classes of basidiomycetes, ascomycetes and deuteromycetes. The measured contact angles of the mycelial mats ranged between hydrophilic (<30°) for the deuteromycetes Fusarium oxysporum Fo47 GUS1 and Trichoderma harzianum P1[pZEGA1] and hydrophobic (>60°) for the ascomycete Cladosporium sp. DSE48.1b and the basidiomycetes Paxillus involutus WSL 37.7, Hebeloma crustiliniforme WSL 6.2, Suillus bovinus WSL 48.1 and Laccaria bicolor WSL 73.1. For some fungi, variations in the hydrophobicity of the mycelium depending on the growth medium, the physiological state and the exposure to water were distinguished. [source] Intracellular pH homeostasis in the filamentous fungus Aspergillus nigerFEBS JOURNAL, Issue 14 2002Stephan J. A. Hesse Intracellular pH homeostasis in the filamentous fungus Aspergillus niger was measured in real time by 31P NMR during perfusion in the NMR tube of fungal biomass immobilized in Ca2+ -alginate beads. The fungus maintained constant cytoplasmic pH (pHcyt) and vacuolar pH (pHvac) values of 7.6 and 6.2, respectively, when the extracellular pH (pHex) was varied between 1.5 and 7.0 in the presence of citrate. Intracellular metabolism did not collapse until a ,pH over the cytoplasmic membrane of 6.6,6.7 was reached (pHex 0.7,0.8). Maintenance of these large pH differences was possible without increased respiration compared to pHex 5.8. Perfusion in the presence of various hexoses and pentoses (pHex 5.8) revealed that the magnitude of ,pH values over the cytoplasmic and vacuolar membrane could be linked to the carbon catabolite repressing properties of the carbon source. Also, larger ,pH values coincided with a higher degree of respiration and increased accumulation of polyphosphate. Addition of protonophore (carbonyl cyanide m -chlorophenylhydrazone, CCCP) to the perfusion buffer led to decreased ATP levels, increased respiration and a partial (1 µm CCCP), transient (2 µm CCCP) or permanent (10 µm CCCP) collapse of the vacuolar membrane ,pH. Nonlethal levels of the metabolic inhibitor azide (N3,, 0.1 mm) caused a transient decrease in pHcyt that was closely paralleled by a transient vacuolar acidification. Vacuolar H+ influx in response to cytoplasmic acidification, also observed during extreme medium acidification, indicates a role in pH homeostasis for this organelle. Finally, 31P NMR spectra of citric acid producing A. niger mycelium showed that despite a combination of low pHex (1.8) and a high acid-secreting capacity, pHcyt and pHvac values were still well maintained (pH 7.5 and 6.4, respectively). [source] Mycoparasitism of arbuscular mycorrhizal fungi: a pathway for the entry of saprotrophic fungi into rootsFEMS MICROBIOLOGY ECOLOGY, Issue 2 2010Nathalie De Jaeger Abstract Within the rhizosphere, arbuscular mycorrhizal (AM) fungi interact with a cohort of microorganisms, among which is the biological control agent, Trichoderma spp. This fungus parasitizes a wide range of phytopathogenic fungi, a phenomenon also reported in the extraradical mycelium (ERM) of AM fungi. Here, we question whether the mycoparasitism of the ERM could be extended to the intraradical mycelium (IRM), thus representing a pathway for the entry of Trichoderma harzianum within the root. Microcosm experiments allowing interactions between Glomus sp. MUCL 41833 placed in a clade that contains the recently described species Glomus irregulare and T. harzianum were set up under in vitro autotrophic culture conditions using potato as a host. A microscope camera-imaging system, coupled with succinate dehydrogenase staining, was used to assess the mycoparasitism in the ERM and IRM. Trichoderma harzianum colonized the ERM of the AM fungus and spread into the IRM, before exiting into the root cells. Intrahyphal growth of T. harzianum caused protoplasm degradation, decreasing the ERM and IRM viability. ERM of the AM fungus represented a pathway for the entry of T. harzianum into the roots of potato. It further sets off the debate on the susceptibility of the AM fungi of being infected by microorganisms from the rhizosphere. [source] Influence of arbuscular mycorrhizal mycelial exudates on soil bacterial growth and community structureFEMS MICROBIOLOGY ECOLOGY, Issue 2 2007Jonas F. Toljander Abstract Plant root systems colonized by arbuscular mycorrhizal (AM) fungi have previously been shown to influence soil bacterial populations; however, the direct influence of the AM extraradical mycelium itself on bacterial growth and community composition is not well understood. In this study, we investigated the effects of exudates produced by AM extraradical mycelia on the growth and development of an extracted soil bacterial community in vitro. The chemical composition of the mycelial exudates was analysed using proton nuclear magnetic resonance spectrometry. Following the addition of exudates to a bacterial community extracted from soil, bacterial growth and vitality were determined using a bacterial vitality stain and fluorescence microscopy. Changes in community composition were also analysed at various times over the course of 3 days by terminal restriction fragment length polymorphism analysis, in combination with cloning and sequencing of 16S rRNA genes. Mycelial exudates increased bacterial growth and vitality and changed bacterial community composition. Several Gammaproteobacteria, including a taxon within the Enterobacteriaceae, increased in frequency of occurrence in response to AM mycelial exudates. This study is the first attempt to identify carbohydrates from the extraradical mycelium of an AM fungus, and demonstrates the direct effects of mycelial exudates on a soil bacterial community. [source] Lead and cadmium uptake in the marine fungi Corollospora lacera and Monodictys pelagicaFEMS MICROBIOLOGY ECOLOGY, Issue 3 2005Michael A.S. Taboski Abstract This study provides observations on the effects of lead and cadmium ions on the growth of two species of marine fungi, Corollospora lacera and Monodictys pelagica. On solid media lead appeared to have no effect on the radial rate of growth of fungi. Exposure to increasing cadmium concentrations on solid media resulted in significant reduction (P < 0.05) in the radial mycelial growth rates of both fungi, especially in M. pelagica. These results reveal significant difference in species sensitivity toward cadmium and, essentially, insensitivity toward lead exposure. In liquid cultures, the metal content of mycelia (metal mass found in mycelium, in mg), and the concentration of metal in dry mycelium (metal mass in 1 g of mycelium, in mg g,1) were both found to increase (P < 0.05) with the increase in the metal cation concentration, while mycelium dry mass decreased. As it was observed on solid media, cadmium cation affected more severely (P < 0.05) the growth of M. pelagica in liquid cultures. Ergosterol content of mycelia of C. lacera exposed to increasing cadmium cation concentration decreased, similarly to the trend observed for dry mycelial mass. It was found that ca. 93% of all lead sequestered by C. lacera is located extracellularly. M. pelagica was found to bioaccumulate over 60 mg of cadmium and over 6 mg of lead per 1 g of mycelium, while C. lacera bioaccumulated over 7 mg of cadmium and up to 250 mg of lead per 1 g of mycelium. Overall, the results indicate that both metal ions affect the growth of marine fungi with lead being accumulated extracellularly in the mycelia. Both metals accumulated by fungi may then enter the marine ecosystem food web, of which marine fungi are integral members. [source] Allocation of plant carbon to foraging and storage in arbuscular mycorrhizal fungiFEMS MICROBIOLOGY ECOLOGY, Issue 2 2003Mayra E Gavito Abstract Foraging strategies, the cost,benefit associated with the search for new resources, have only begun to be explored in arbuscular mycorrhizal fungi (AMF). We show the use of 13C-labelling, via shoot photosynthesis, of the 16:1,5 fatty acid biomarker (the dominant and rather specific fatty acid in AMF storage lipids) to study the immediate patterns of carbon allocation to fungal lipids in response to inorganic and organic nutrient amendments. Signature fatty acid measurements, the incorporation of the label and complementary hyphal length density measurements showed that the extraradical mycelium of AMF proliferated in response to all the amendments provided whereas its development into unamended sand was minor in all treatments. We demonstrate the foraging capacity of AMF, linked to plant carbon, through their hyphal proliferation and accumulation of energy reserves. [source] Deprogrammed sporulation in StreptomycesFEMS MICROBIOLOGY LETTERS, Issue 1 2002Yasuo Ohnishi Abstract The bacterial genus Streptomyces forms chains of spores by septation at intervals in aerial hyphae and subsequent maturation on solid medium. Substrate hyphae undergo extensive lysis, liberating nutrients on which aerial hyphae develop. Some mutant strains, however, ectopically form spores by septation in substrate hyphae on solid medium or in vegetative hyphae in liquid medium, which suggests that all hyphae have the potential to differentiate into spores. A Streptomyces griseus mutant strain NP4, which has a mutation in the regulatory system for an ATP-binding cassette (ABC) transporter gene, forms ectopic spores in substrate hyphae only on glucose-containing medium. In addition, overexpression of a substrate-binding protein of the ABC transporter in the wild-type strain causes ectopic septation in very young substrate hyphae and subsequent sporulation in response to glucose. These ectopic spores germinate normally. The ectopic sporulation is independent of A-factor, a microbial hormone that determines the timing of aerial mycelium formation during normal development. Thus, substrate hyphae of Streptomyces have a potential to develop into spores without formation of aerial hyphae. For programmed development, therefore, the strict repression of septum formation in substrate mycelium should be necessary, as well as the positive signal relay leading to aerial mycelium formation followed by septation and sporulation. [source] Characterization and functional analysis of the ,-1,3-glucanosyltransferase 3 of the human pathogenic fungus Paracoccidioides brasiliensisFEMS YEAST RESEARCH, Issue 1 2009Nadya Da Silva Castro Abstract The fungus Paracoccidioides brasiliensis causes paracoccidioidomycosis, a systemic granulomatous mycosis prevalent in Latin America. In an effort to elucidate the molecular mechanisms involved in fungus cell wall assembly and morphogenesis, ,-1,3-glucanosyltransferase 3 (PbGel3p) is presented here. PbGel3p presented functional similarity to the glucan-elongating/glycophospholipid-anchored surface/pH-regulated /essential for pseudohyphal development protein families, which are involved in fungal cell wall biosynthesis and morphogenesis. The full-length cDNA and gene were obtained. Southern blot and in silico analysis suggested that there is one copy of the gene in P. brasiliensis. The recombinant PbGel3p was overexpressed in Escherichia coli, and a polyclonal antibody was obtained. The PbGEL3 mRNA, as well as the protein, was detected at the highest level in the mycelium phase. The protein was immunolocalized at the surface in both the mycelium and the yeast phases. We addressed the potential role of PbGel3p in cell wall biosynthesis and morphogenesis by assessing its ability to rescue the phenotype of the Saccharomyces cerevisiae gas1, mutant. The results indicated that PbGel3p is a cell wall-associated protein that probably works as a ,-1,3-glucan elongase capable of mediating fungal cell wall integrity. [source] Seasonal effect on infection and development of lesions caused by Cryphonectria parasitica in Castanea sativaFOREST PATHOLOGY, Issue 4 2003L. Guérin Summary Seasonal variation in the development of chestnut blight, caused by Cryphonectria parasitica, was investigated by inoculating in situ chestnut trees and in vitro excised chestnut segments, at either monthly or 3-monthly intervals throughout 30 months. Inoculations were made with conidia and mycelium of a virulent isolate and with mycelium of a hypovirulent isolate. Conidial inoculations of living sprouts or excised segments between May and July resulted in the greatest incidence of infection whereas inoculations in autumn and winter, in vitro as well as in situ, did not reveal any visible disease. However, from these symptomless inoculated stems, C. parasitica was isolated 3 months after inoculation. Inoculations with the mycelium of the virulent isolate always resulted in lesions, except in January 1999, and the greatest rate of lesion development occurred for inoculations made in the spring and summer. There was a significant seasonal effect on lesion development. Lesions caused by the hypovirulent isolate, smaller than those caused by the virulent isolate, followed a similar seasonal pattern. The same seasonal variations were observed for inoculations in vitro of excised segments. Relative water content (RWC) of chestnut bark significantly varied with bark sampling date. The rate of lesion development in sprouts significantly correlated with average minimum (ATn) and maximum (ATx) temperatures and the sum of rainfall during inoculation period, with the rate of lesion development measured in excised segments 10 days after inoculation (R10d) and with RWC measured on the day of inoculation. In multiple regression models, variables ATx and R10d best explained variation in lesion development. Résumé La variation saisonnière du développement de la maladie du chancre causée par Cryphonectria parasitica a étéétudiée en réalisant, une fois par mois ou par trimestre pendant deux ans et demi, des inoculations de châtaigniers in situ et des inoculations in vitro de branches coupées. Les inoculations ont été réalisées avec des conidies et du mycélium d'un isolat virulent et avec du mycélium d'un isolat hypovirulent. En Mai et Juin, les inoculations conidiennes ont provoqué le taux d'infection le plus élevé, in situ et in vitro. A l'inverse, en automne et en hiver, ces inoculations n'ont pas causé de symptômes. Cependant C. parasitica a été réisolé 3 mois plus tard, sur ces tiges ne présentant aucune lésion. Les inoculations avec du mycélium ont toujours résulté en des lésions, excepté en Janvier 1999, et le développement des lésions (R1m) a été le plus rapide au printemps et en été. La date d'inoculation a eu un effet significatif sur l'extension des lésions. Le même effet saisonnier était observé sur le développement des lésions causées par l'isolat hypovirulent, plus petites que celles causées par l'isolat virulent. Les mêmes variations saisonnières de l'extension des lésions étaient observées in vitro sur tiges excisées. Un effet saisonnier significatif a été mis en évidence sur la teneur en eau relative des écorces de châtaignier (RWC). Zusammenfassung Der jahreszeitlich bedingte Einfluß auf die Entstehung von Kastanienrindenkrebs, verursacht von Cryphonectria parasitica, wurde durch Inokulation von Kastanienbäumen in situ und durch Beimpfung von geschnittenen Triebsegmenten in vitro in monatlichen oder drei-monatlichen Intervallen über dreißig Monate untersucht. Zur Inokulation wurden Myzel und Konidien eines virulenten Isolates und Myzel eines hypovirulenten Isolates verwendet. Mit Konidien wurde zwischen Mai und Juni die größte Infektionshäufigkeit auf lebenden Sprossen oder Triebsegmenten erreicht, während Inokulationen im Herbst und Winter keine sichtbaren Symptome hervorriefen. Jedoch konnte C. parastica drei Monate nach Inokulation von lebenden Sprossen isoliert werden, auch wenn keine Läsion sichtbar war. Außer im Januar 1999 führte Inokulation mit Myzel des virulenten Isolates immer zu Läsionen. Am häufigsten traten Läsionen bei Inokulationen im Frühling und im Sommer auf. Der Einfluß des Inokulationszeitpunktes auf die Ausbildung von Läsionen war signifikant. Inokulationen mit dem hypovirulenten Isolat zeigten eine ähnliche jahreszeitliche Schwankung wie jene mit dem virulenten Isolat, führten aber zu kleineren Läsionen. Die gleichen jahreszeitlichen Variationen wurden bei in vitro Inokulation von Kastanientrieben beobachtet. Der relative Wassergehalt (RWC) der Borke variierte signifikant mit dem Beprobungszeitpunkt der Borke. Die Bildung von Läsionen an lebenden Sprossen korrelierte signifikant mit den durchschnittlichen Temperaturminima (ATn) und -maxima (ATx), mit der Gesamtregenmenge (SR) während der Inokulationsperiode, mit der Bildung von Läsionen an Triebsegmenten (R10d) und mit dem am Inokulationstag gemessenen RWC. In einem multiplen Regressionsanalysenmodell zeigte sich, dass die jahreszeitliche Schwankung der Ausbildung von Kastanienrindenkrebs beim virulenten Isolat am besten durch die Variablen ATx und R10d, beim hypovirulenen Isolat durch ATx erklärt wurde. [source] Incomplete movement of Cryphonectria hypovirus 1 within a vegetative compatibility type of Cryphonectria parasitica in natural cankers on grafted American chestnut treesFOREST PATHOLOGY, Issue 6 2002E. Hogan Summary American chestnut trees, grafted in 1980 from large survivors, were inoculated in 1982 and 1983 with four white (European) hypovirulent strains of Cryphonectria parasitica, infected with C. hypovirus 1 (CHV1); this hypovirus has been shown to be capable of moving rapidly within the mycelium of a vegetative compatibility (vc) type of C. parasitica in blight cankers. Using a 49-cell lattice plot, 17.8×17.8 cm, the spatial patterns and frequencies of white and pigmented isolates and white and pigmented vc types were investigated within superficial cankers on the grafts located outside the hypovirulent-strain-inoculated zone. Four of six cankers assayed contained white isolates, and three of the four had random spatial patterns of white isolates, based on join-count statistics. Vc tests, using pigmented isolates and pigmented single-spore colonies of white isolates, indicated that the majority of white and pigmented isolates recovered from each of two cankers assayed were in one vc type. White and pigmented lattice-plot cells of the same vc type were frequently in contact with each other, indicating incomplete movement of CHV1 within a vc type. Nine and 10 vc types were found in the two cankers; it is hypothesized that small, white vc type areas in each canker may be a source of CHV1 transmission to the major vc types. Based on join-count statistics, the spatial pattern of the single, major vc type in one canker was non-random (aggregated), whereas the other canker had a random major vc type pattern. White and pigmented in vitro variants (sectors) of C. parasitica, that resemble white and pigmented in vivo variants in spatial contact and vc compatibility, were intermediate hypovirulent and virulent on forest American chestnuts, and dsRNA positive and negative, respectively. Murphy Interactions between two isolates (Avondhu and CBS) of Tricholomopsis rutilans and ectomycorrhizal fungi (Hebeloma crustuliniforme, Laccaria laccata and Paxillus involutus) were examined on agar medium in the presence or absence of woodchips. The CBS isolate showed more competitiveness than the Avondhu isolate, when paired with ectomycorrhizal fungi. There was an inhibition of the growth of mycelium of the ectomycorrhizal fungi ranging from overgrowth to avoidance. The ectomycorrhizal fungi exhibited hyphal abnormalities such as discoloration, excessive vacuolation and curling. Formation of mycorrhizas by H. crustuliniforme in Pinus contorta and Picea abies was unaffected by the presence of the CBS isolate, whereas a reduction occurred in the presence of the Avondhu isolate. In non-mycorrhizal seedlings of both conifers, the Avondhu isolate reduced root and shoot dry mass and number of root laterals and caused a lower number of short roots in lodgepole pine. The significance of these interactions between T. rutilans and ectomycorrhizal fungi in paired culture and during symbiosis is discussed. Yuan A study of lesion development in stems of Eucalyptus nitens following artificial inoculations with canker fungi was carried out on 16-year-old plantation trees. In a first trial cambium bark wounds on smooth- and rough-barked trees were inoculated with the mycelium of nine species of canker fungi, including Endothia gyrosa. In a second trial spores or mycelium of E. gyrosa were applied directly onto undamaged or superficially wounded bark surfaces. Infection subsequent to artificial inoculation via wounding (whatever the wounding technique or type of inoculum) resulted in significantly larger external lesions (mean lesion area up to 35.6 cm2 20 months after inoculation) on smooth bark compared with those on rough bark (up to 19.0 cm2). Microscopic studies of infected rough and smooth bark suggest that, once smooth bark is compromised by wounding and artificial inoculation, the particular anatomical structure of smooth bark may offer less mechanical resistance to post-penetration hyphal spread in comparison with rough bark. It is suggested that at a pre-penetration stage under natural conditions spores of E. gyrosa more easily infect rough bark via cracks associated with this type of bark but not present in smooth bark. Przyby Summary Bacteria including Pseudomonas putida, Pseudomonas fluorescens biovar I, Pseudomonas fluorescens biovar V, Pseudomonas aureofaciens and Erwinia herbicola were isolated from discoloured zones in birch trunks. Antagonistic effects of these bacteria to growth of Piptoporus betulinus mycelium were tested in vitro, both in dual culture and using bacterial cell-free culture filtrates. In dual cultures, P. putida was most effective at inhibiting mycelial growth of Piptoporus betulinus. Filtrates of P. putida inhibited growth of P. betulinus mycelium irrespective of filtrate concentration, incubation time of bacteria and timing of recording mycelium growth. The strongest antagonistic effect (inhibition of fungal growth) was observed on a medium containing 80% of sterile filtrate obtained from 15-day-old bacterial cultures. The highest stimulating effect on mycelium growth was noted on medium containing 80% filtrate obtained from 7-day-old E. herbicola cultures. [source] Colonization of beech leaves by two endophytic fungi in northern JapanFOREST PATHOLOGY, Issue 2 2000N. Sahashi Summary Leaves of Japanese beech (Fagus crenata) were collected monthly during the vegetation period at five sites in the Tohoku district in Japan to isolate endophytic fungi. Leaves were also collected only once at two additional sites. Two endophytic fungi were dominant, a Discula species and a sterile mycelium. This result strongly suggests that these two fungi are generally associated with leaves of the Japanese beech at different sites. At most sites the isolation frequency of Discula sp. was greatest in June and gradually decreased from July to October whereas the isolation frequency of the sterile mycelium increased during the vegetation period and remained at a high isolation frequency in October. Spores of Discula sp. were released for a very short time in late May, just after the disappearance of the snow cover on the forest floor. These spores may be important for the infection of newly sprouting leaves. Pers. and Hypholoma fasciculare (Huds: Fr) Kumner over 152,155 days, determined as ability to capture or share territory in soil and wood, in terms of decay partitioning and the carbon cost of interactions. 2.,The outcome of interactions in wood alone differed from those in which the fungi competed for an opponents' inoculum in soil microcosms. Competitive ability (the ability to capture or co-occupy an opponent's inoculum) varied according to species and inoculum age. In wood block pairings in the absence of soil there was evidence that P. velutina opportunistically utilized C previously mobilized within an opponent's inoculum. 3.,In soil systems, short-term (28-day) respiratory losses of preloaded 14C (supplied as glucose) indicated that interaction could have a substantial C cost, depending on the resource quality of the opponents' inocula. Muñoz Abstract Aim:, To evaluate the activity against fungal phytopathogens of two synthetic peptides derived from the protein bovine lactoferricin: the antibacterial active core of six amino acid residues (LfcinB20,25) and an extension of 15 amino acids (LfcinB17,31). Methods and Results:,In vitro activity against fungal pathogens was determined and compared with that against model micro-organisms. Activity was demonstrated against fungi of agronomic relevance. Distinct antimicrobial properties in vitro were found for the two peptides. LfcinB17,31 had growth inhibitory activity higher than LfcinB20,25. However, LfcinB17,31 was not fungicidal to quiescent conidia of Penicillium digitatum at the concentrations assayed, while LfcinB20,25 killed conidia more efficiently. Microscopical observations showed that the mycelium of P. digitatum treated with LfcinB17,31 developed alterations of growth, sporulation and chitin deposition, and permeation of hyphal cells. In experimental inoculations of mandarins, both peptides showed limited protective effect against the disease caused by P. digitatum. Conclusions:, LfcinB20,25 and LfcinB17,31 peptides were shown to have antimicrobial activity against plant pathogenic filamentous fungi, with distinct properties and mode of action. Leung Abstract Aims:, To examine and illustrate the morphological characteristics and growth kinetics of Cs-HK1, a Tolypocladium fungus, isolated from wild Cordyceps sinensis in solid and liquid cultures, and the major chemical constituents and antitumour effects of Cs-HK1 mycelium. Methods and Results:, The Cs-HK1 fungus was isolated from the fruiting body of a wild C. sinensis and identified as a Tolypocladium sp. fungus. It grew rapidly at 22,25°C on a liquid medium containing glucose, yeast extract, peptone and major inorganic salts, with a specific growth rate of 1·1 day,1, reaching a cell density of 23·0 g dw l,1 in 7,9 days. Exopolysaccharides accumulated in the liquid culture to about 0·3 g l,1 glucose equivalent. In comparison with natural C. sinensis, the fungal mycelium had similar contents of protein (11·7,,g) and carbohydrate (654·6,,g) but much higher contents of polysaccharide (244·2 mg vs 129·5 mg), adenosine (1116·8,,g vs 264·6 ,g) and cordycepin (65·7 ,g vs 20·8 ,g) (per gram dry weight). Cyclosporin A, an antibiotic commonly produced by Tolypocladium sp., was also detected from the mycelium extract. The hot water extract of mycelium showed low cytotoxic effect on B16 melanoma cells in culture (about 25% inhibition) but significant antitumour effect in animal tests, causing 50% inhibition of B16 cell-induced tumour growth in mice. Conclusions:, The Tolypocladium sp. fungus, Cs-HK1, can be easily cultivated by liquid fermentation. The mycelium biomass contained the major bioactive compounds of C. sinensis, and the mycelium extract had significant antitumour activity. Methods and Results:, The microbial hydrolysis of various racemic esters of 1,2-O-isopropylidene glycerol (IPG) was attempted by screening among Streptomyces spp. previously selected on the basis of their carboxylesterase activity. The best results were observed in the hydrolysis of butyrate ester and two strains appeared promising as they showed opposite enantioselectivity: Streptomyces sp. 90852 gave predominantly (S)-IPG, while strain 90930 mostly gave the R -alcohol. Streptomyces sp. 90930 was identified as Streptomyces violaceusniger, whereas Streptomyces sp. 90852 is a new species belonging to the Streptomyces violaceus taxon. The carboxylesterase belonging to strain 90852 gave a maximum value of enantiomeric ratio (E) of 14,16. This strain was lyophilized and used as dry mycelium for catalysing the synthesis of isopropylidene glycerol butyrate in heptane showing reaction rate and enantioselectivity (E = 6·6) lower than what observed for the hydrolysis. Conclusions:, A new esterase with enantioselective activity towards (R,S)-IPG butyrate has been selected. The best enantioselectivity is similar or even better than the highest reported value in the literature with commercial enzymes. The enzyme is produced by a new species belonging to the S. violaceus taxon. Significance and Impact of the Study:, New esterases from streptomycetes can be employed for the enantioselective hydrolysis of chiral esters derived from primary alcohols, not efficiently resolved with commercial enzymes. [source] Oil and fungal biomass dispersion in a stirred tank containing a simulated fermentation brothJOURNAL OF CHEMICAL TECHNOLOGY & BIOTECHNOLOGY, Issue 11 2001Ma Soledad Córdova-Aguilar Abstract The production of ,-decalactone by the filamentous fungus Trichoderma harzianum involves four phases (oil,water,air,mycelium) and its dispersion is crucial during fermentation. Oil and biomass (when present) dispersion, as a function of the volumetric power drawn (P/V), was characterized, in two; three- and four-phase systems agitated with Rushton turbines. Trichoderma harzianum mycelium was used as the solid phase in the four-phase system. Two stages of the fermentation were simulated: the beginning (15% oil and 1.4,kgm,3 of mycelium) and the end (2% oil and 10.6,kg,m,3 of mycelium). In the two-phase system, the use of exhausted broth achieved higher oil dispersions at low P/V values as compared with distilled water. Aeration decreased the oil dispersion for the high-oil system, but enhanced oil dispersion for the low-oil system. Compared with the P/V used in the actual fermentation (0.2,kW m,3), a high segregation of the system was observed for the high-oil/low-biomass system, due to the difficulty of mixing the thick oil,air emulsion present at the top of the tank. The system simulating the end of the fermentation reached almost complete homogeneity of oil and biomass, a phenomenon due to the high biomass/oil ratio and the biomass acting as an oil carrier. © 2001 Society of Chemical Industry [source] SPECIFIC DETECTION OF AMANITA PHALLOIDES MYCELIUM AND SPORES BY PCR AMPLIFICATION OF THE GPD (GLYCERALDEHYDE-3-PHOSPHATE DEHYDROGENASE) GENE FRAGMENTJOURNAL OF FOOD BIOCHEMISTRY, Issue 3 2000OWSKI, ROMAN KOT ABSTRACT Oligonucleotide primers designed to flank a 635 bp fragment of the gene encoding glyceraldehyde-3-phosphate dehydrogenase (gpd) from Araanita muscaria were used to amplify the corresponding gpd fragment from Amanita phalloides. The A. phalloides PCR product was cloned, sequenced and found to be 70 - 77% similar to the known basidiomycetes gpd genes within the exon part and 25 - 52% within the intron part. Based on these data, species-specific amplification was achieved using a pair of oligonucleotide primers complementary to the A. phalloides gpd intron sequences. These primers allowed the amplification of the corresponding gpd fragment from the A. phalloides but not from various other basidiomycetes, ascomycetes and human matrices. PCR amplification of the A. phalloides DNA gave the predicted PCR product of 284 bp. The created PCR system is an efficient tool for the specific, rapid and sensitive detection of A. phalloides mycelium and spores. [source] First Report of Rhizoctonia solani AG-7 on Cotton in EgyptJOURNAL OF PHYTOPATHOLOGY, Issue 4 2010Kamel A. Abd-Elsalam Abstract Eighty-two isolates of Rhizoctonia solani were recorded from roots of naturally-infected seedlings of the Egyptian cotton (Gossypium barbadense L.). Anastomosis groups (AGs) of the isolates were determined by using 13 different AGs testers. Three (3.7%) of the isolates were identified as R. solani AG7, while the remaining isolates were belonging to the AG 2-1, AG4 and AG5. The identification of the three isolates was based on the frequency of the C2 reaction with the AG7 tester isolate. No fusion was observed between AG7 and isolates representing the other 13 AGs. Colonies of AG7 isolates grown on potato dextrose agar (PDA), malt yeast agar (MYA) and melt peptone agar (MPA) were brown to dark brown with aerial mycelium and sclerotia. The isolates had pitted sclerotial clusters and brownish exudates after 21 days of culturing on PDA, but without clear zonation. Pathogenicity test under greenhouse conditions revealed that AG7 caused the common symptoms of damping,off, which included seed rot, lesions on the hypocotyls and root rot. [source] Development of a Sensitive Serological Method for Specific Detection of Latent Infection of Macrophomina phaseolina in CowpeaJOURNAL OF PHYTOPATHOLOGY, Issue 1 2009Leonard Afouda Abstract A double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) was developed for the specific detection and quantification of Macrophomina phaseolina in plant tissue. Both polyclonal antisera produced against immunogens from mycelium and culture filtrate of M. phaseolina detected the fungus in mycelial and plant extracts, although the antibodies raised against mycelium were more sensitive. No cross-reaction occurred with Rhizopus stolonifer, Pythium ultimum, Mucor hiemalis, Fusarium oxysporum, Septoria nodorum, Rhizoctonia solani, Sclerotinia sclerotiorum, Phytophthora infestans and Aspergillus niger. In enzyme assays, activity of the endo-acting hydrolytic enzymes 1,3-,-glucanase and, less, cellulase, but not xylanase was detected in infected plants. DAS-ELISA was more sensitive than the 1,3-,-glucanase assay. In polyacrylamide gel electrophoresis (PAGE) up to 18 protein bands were observed, with four bands occurring in the 12 tested isolates deriving from various geographical origin in Niger and Nigeria. The enzyme assays and protein patterns were considered not suitable for specific M. phaseolina detection. Macrophomina phaseolina was essentially located in the roots and hypocotyls, and less in epicotyls and leaves of infected plants. The antibodies were also useful to detect latent infection and the infection of cowpea seeds. Yamanobe Abstract Different methods were investigated for the rapid mass culture of Cistella japonica by using water extracts of some nutritional sources. In an agar culture test, there was little difference in mycelial growth in water extracts of wheat bran, rice bran and potato. In suspension culture with wheat bran extract, which is easily and cheaply available, the mycelium of C. japonica increased seven times more than that in agar culture after a month's incubation. C. japonica from suspension culture was pathogenic to Chamaecyparis obtusa. These results suggest that suspension culture in water extract of wheat bran can be adopted for the rapid mass culturing of C. japonica for use in inoculation tests. The activity of aminopeptidase and esterase was located in tissues adjacent to dry rot caused by Phoma exigua var. foveata and in other apparently healthy tissues. The activity of glucuronidase, succinic and glucose-6-phosphate dehydrogenases (G-6-PDH), however, was confined to tissues adjacent to the rotted tissue. The pathogen produces very active , - and , -glycosidases, so their highest activity occurred in rotten tissue that was filled with fungal mycelium. Results suggest that all these enzymes are involved in alteration of cell metabolism and the destruction of diseased tuber tissue. [source] |
