Restricted randomization and multiple responses in industrial experiments

Abstract Two issues regarding designed experiments are discussed; restrictions on randomization and multiple responses. The former is typically related to hard-to-vary factors and factors appearing in separate stages of a process experiment. Randomization restrictions should be taken into account in the construction of the design as well as in the statistical modelling. In the paper, a case study of sausage production is discussed, having a split-plot model with correlated multiple responses. Multiple responses are handled in two ways, by principal component analysis (PCA) followed by ANOVA of the principal components, and by a newly developed alternative, the ,50,50 MANOVA'. Multiple tests of correlated response variables are also described. Practical aspects of the planning, performing, response measurements, and statistical analysis are emphasized throughout. Hence, the paper aims to extend the utility of statistical methods in industry by linking design of experiments to multivariate analysis of the responses. Copyright © 2007 John Wiley & Sons, Ltd. The value of a questionnaire in assessing the acquisition and retention of basic science knowledge by dental students

This cross-sectional study aimed to assess and compare, by performances in a questionnaire, the level of knowledge of basic medical sciences in 6th-form school pupils studying science subjects as entrance requirements to University and in 2nd, 3rd and 4th-year undergraduate dental students. A 40-part multiple response, true/false questionnaire, testing recall of factual knowledge in anatomy, biochemistry, physiology and oral biology, was used as the method of assessment. The results suggested that this simple format was an acceptable and useful method of assessment of the knowledge level of the study groups. The difference in scores of knowledge, expected to be higher in 2nd-year students compared to 6th form groups, was greatest in anatomy and oral biology, less in biochemistry and, unexpectedly, was not apparent in physiology. A difference in performance on the knowledge questionnaire was observed between 4th and 2nd year dental students, attributable primarily to decreased scores for 4th year dental students in biochemistry and, to a lesser extent, anatomy. The results obtained with this standardised test of factual knowledge recall may be of value to teachers compiling medical sciences courses for dental undergraduates and to those planning and evaluating new curricula with a less didactic approach. Calibration of Multivariate Scatter plots for Exploratory Analysis of Relations Within and Between Sets of Variables in Genomic Research

Abstract The scatter plot is a well known and easily applicable graphical tool to explore relationships between two quantitative variables. For the exploration of relations between multiple variables, generalisations of the scatter plot are useful. We present an overview of multivariate scatter plots focussing on the following situations. Firstly, we look at a scatter plot for portraying relations between quantitative variables within one data matrix. Secondly, we discuss a similar plot for the case of qualitative variables. Thirdly, we describe scatter plots for the relationships between two sets of variables where we focus on correlations. Finally, we treat plots of the relationships between multiple response and predictor variables, focussing on the matrix of regression coefficients. We will present both known and new results, where an important original contribution concerns a procedure for the inclusion of scales for the variables in multivariate scatter plots. We provide software for drawing such scales. We illustrate the construction and interpretation of the plots by means of examples on data collected in a genomic research program on taste in tomato. Mechanisms of Hedgehog gradient formation and interpretation

Abstract Morphogens are molecules that spread from localized sites of production, specifying distinct cell outcomes at different concentrations. Members of the Hedgehog (Hh) family of signaling molecules act as morphogens in different developmental systems. If we are to understand how Hh elicits multiple responses in a temporally and spatially specific manner, the molecular mechanism of Hh gradient formation needs to be established. Moreover, understanding the mechanisms of Hh signaling is a central issue in biology, not only because of the role of Hh in morphogenesis, but also because of its involvement in a wide range of human diseases. Here, we review the mechanisms affecting the dynamics of Hh gradient formation, mostly in the context of Drosophila wing development, although parallel findings in vertebrate systems are also discussed. © 2005 Wiley Periodicals, Inc. Predicting species distributions from museum and herbarium records using multiresponse models fitted with multivariate adaptive regression splines

ABSTRACT Current circumstances , that the majority of species distribution records exist as presence-only data (e.g. from museums and herbaria), and that there is an established need for predictions of species distributions , mean that scientists and conservation managers seek to develop robust methods for using these data. Such methods must, in particular, accommodate the difficulties caused by lack of reliable information about sites where species are absent. Here we test two approaches for overcoming these difficulties, analysing a range of data sets using the technique of multivariate adaptive regression splines (MARS). MARS is closely related to regression techniques such as generalized additive models (GAMs) that are commonly and successfully used in modelling species distributions, but has particular advantages in its analytical speed and the ease of transfer of analysis results to other computational environments such as a Geographic Information System. MARS also has the advantage that it can model multiple responses, meaning that it can combine information from a set of species to determine the dominant environmental drivers of variation in species composition. We use data from 226 species from six regions of the world, and demonstrate the use of MARS for distribution modelling using presence-only data. We test whether (1) the type of data used to represent absence or background and (2) the signal from multiple species affect predictive performance, by evaluating predictions at completely independent sites where genuine presence,absence data were recorded. Models developed with absences inferred from the total set of presence-only sites for a biological group, and using simultaneous analysis of multiple species to inform the choice of predictor variables, performed better than models in which species were analysed singly, or in which pseudo-absences were drawn randomly from the study area. The methods are fast, relatively simple to understand, and useful for situations where data are limited. A tutorial is included. [source]


Overexpression of the Wounding-Responsive Gene AtMYB15 Activates the Shikimate Pathway in Arabidopsis

Abstract The MYB transcription factor genes play important roles in many developmental processes and various defense responses of plants. The shikimate pathway is a major biosynthetic pathway for the production of three aromatic amino acids and other aromatic compounds that are involved in multiple responses of plants, including protection against UV and defense. Herein, we describe the characterization of the R2R3-MYB gene AtMYB15 as an activator of the shikimate pathway in Arabidopsis. The AtMYB15 protein is nuclear localized and a transcriptional activation domain is found in its C-terminal portion. Northern blots showed that AtMYB15 is an early wounding-inducible gene. Resutls of microarray analysis, confirmed using quantitative real-time polymerase chain reaction, showed that overexpression of AtMYB15 in transgenic plants resulted in elevated expression of almost all the genes involved in the shikimate pathway. Bioinformatics analysis showed that one or more AtMYB15-binding AC elements were detected in the promoters of these upregulated genes. Furthermore, these genes in the shikimate pathway were also found to be induced by wounding. These data suggest an important role of AtMYB15 as a possible direct regulator of the Arabidopsis shikimate pathway in response to wounding. (Managing editor: Ya-Qin Han) [source]


Discovering EFL learners' perception of prior knowledge and its roles in reading comprehension

This paper investigates EFL learners' perception of prior knowledge and its roles in reading comprehension. It is based on a survey conducted among 400 EFL students studying at secondary and tertiary levels in China. Through the analysis of the ranked multiple responses to the questions posed in the questionnaire, the paper shows that EFL students in mainland China believe that their command of English vocabulary plays a crucial role in their reading comprehension. However as their linguistic knowledge increases, they tend to attach less importance to their linguistic knowledge, especially the knowledge of English syntax and formal structures. At the same time, conceptual and sociocultural knowledge seems to gain greater importance. Furthermore, the perceived importance of linguistic knowledge seems to start diminishing around the end of secondary education, when EFL students have acquired a vocabulary of about 3000 words and the basic knowledge of English syntax and formal structures. The final replacement of linguistic knowledge by conceptual or sociocultural knowledge as the top factor that affects their reading comprehension seems to take place one year after the beginning of the tertiary EFL course. This may well be considered as an indication of the EFL threshold level for EFL students in China. [source]


Hydrophilic interaction LC of peptides: Columns comparison and clustering

Abstract A wide variety of hydrophilic interaction chromatography (HILIC) stationary phase surface chemistries are currently available. Although their selectivity can be considerably different, column comparison or clustering using peptides is limited. In this study, ten pharmaceutically relevant model peptides are analyzed on seven different HILIC columns (bare silica, amide, poly-hydroxyethyl aspartamide, diol and zwitterionic) for the evaluation of their performance and classification. The responses examined include single and multiple responses: plate number, asymmetry factor, LOD, geometric mean resolution, resolution product, time corrected resolution product, peak capacity and chromatographic response function. Column classification was performed using hierarchical clustering and principal component analysis. Moreover, the overall performance quality of the HILIC columns was compared using a linear desirability function. Hierarchical clustering and principal component analysis showed consistent clusters. The zwitterionic phase was clustered apart from the other HILIC columns and both poly-aspartamide columns were clustered together. In addition, the two bare silica phases represent two different clusters, and thus different selectivities. Overall, the responses showed the best performance for one of the bare silica columns (Alltima-Alltech), followed by the zwitterionic phase (ZIC)-HILIC. Thus, these columns, belonging to different clusters, were found to be the best performing systems in pharmaceutical peptide analysis for the selected peptide set. [source]

Summary., A fully Bayesian analysis of directed graphs, with particular emphasis on applica- tions in social networks, is explored. The model is capable of incorporating the effects of covariates, within and between block ties and multiple responses. Inference is straightforward by using software that is based on Markov chain Monte Carlo methods. Examples are provided which highlight the variety of data sets that can be entertained and the ease with which they can be analysed. [source]


Precues enable multiple response preprogramming: Evidence from startle

Abstract When one or more response dimensions in a choice reaction time (RT) task are provided beforehand (or precued), RT decreases, indicating that the precued part of a response was prepared in advance. In this study, a startling acoustic stimulus was used to investigate the amount of preprogramming that occurs when part of the response is precued because a startling stimulus has been shown to directly trigger preprogrammed responses. Participants performed wrist flexion/extension movements in a precued one to four choice RT paradigm. A control auditory stimulus (82 dB) or a startling acoustic stimulus (124 dB) accompanied the visual "go" signal on each trial. Although RT latencies were significantly reduced by the startle, many more errors were observed in the startled conditions. Importantly, the errors seen at short RT latencies largely reflected multiple movements to the cued response alternatives, suggesting that multiple responses were preprogrammed. [source]

Partition experimental designs for sequential processes: Part II,second-order models

Abstract Second-order experimental designs are employed when an experimenter wishes to fit a second-order model to account for response curvature over the region of interest. Partition designs are utilized when the output quality or performance characteristics of a product depend not only on the effect of the factors in the current process, but the effects of factors from preceding processes. Standard experimental design methods are often difficult to apply to several sequential processes. We present an approach to building second-order response models for sequential processes with several design factors and multiple responses. The proposed design expands current experimental designs to incorporate two processes into one partitioned design. Potential advantages include a reduction in the time required to execute the experiment, a decrease in the number of experimental runs, and improved understanding of the process variables and their influence on the responses. Wetland Restoration in the New Millennium: Do Research Efforts Match Opportunities?

Abstract Of 311 papers on wetland restoration, only 15 concerned large-scale experimentation in restoration sites. Most papers described what happened, reported on small field experiments, or discussed restoration targets. While these are important topics, our opinion is that we lose significant opportunities to learn how to recover populations, community structure, and ecosystem processes, and we limit our ability to document variability and whole-system responses, when we do not experiment at large scales. We suggest that, wherever possible, large projects facilitate field tests of alternative restoration approaches. Furthermore, we encourage researchers to take advantage of major restoration efforts by conducting large field experiments, assessing multiple responses, and offering restoration guidance in an adaptive framework. Robust optimization for multiple responses using response surface methodology

Abstract Typically in the analysis of industrial data for product/process optimization, there are many response variables that are under investigation at the same time. Robustness is also an important concept in industrial optimization. Here, robustness means that the responses are not sensitive to the small changes of the input variables. However, most of the recent work in industrial optimization has not dealt with robustness, and most practitioners follow up optimization calculations without consideration for robustness. This paper presents a strategy for dealing with robustness and optimization simultaneously for multiple responses. In this paper, we propose a robustness desirability function distinguished from the optimization desirability function and also propose an overall desirability function approach, which makes balance between robustness and optimization for multiple response problems. Simplex search method is used to search for the most robust optimal point in the feasible operating region. Finally, the proposed strategy is illustrated with an example from the literature.
