Cell-specific elevation of NRF2 and sulfiredoxin-1 as markers of oxidative stress in the lungs of idiopathic pulmonary fibrosis and non-specific interstitial pneumonia. APMIS 2010; 118: 703,12. Human idiopathic pulmonary fibrosis (IPF) and non-specific interstitial pneumonia (NSIP) have been proposed to be attributable to oxidative stress. The nuclear factor, erythroid derived 2, like protein (NRF2),sulfiredoxin-1 (SRX1) pathway was hypothesized to be associated with the pathogenesis of human pulmonary fibrosis. Several methods including digital morphometry were used in the assessment of the cell-specific localization and expression of NRF2 and SRX1 and selected proteins linked to their activation/stability in human IPF/usual interstitial pneumonia (UIP) and NSIP lung. The proteins of the NRF2 pathway were localized in the hyperplastic alveolar epithelium and inflammatory cells in IPF and NSIP, but were absent in the fibroblastic foci characteristic of IPF. Morphometric evaluation revealed NRF2 and KEAP1 to be significantly elevated in the hyperplastic alveolar epithelium compared with the normal alveolar epithelium, and NRF2 was remarkably expressed in the nuclear compartment of the hyperplastic cells. SRX1 was expressed mainly in alveolar macrophages, and the number of SRX1-positive macrophages/surface area was elevated in NSIP, a disease which contains more marked inflammatory reaction compared with the IPF/UIP lung. The expression of the NRF2 pathway in human IPF and NSIP is further evidence that the pathogenesis of human fibrotic lung diseases is oxidant-mediated and originates from the alveolar epithelium. [source] Morphometric evaluation of the repair of critical-size defects using demineralized bovine bone and autogenous bone grafts in rat calvariaCLINICAL ORAL IMPLANTS RESEARCH, Issue 8 2008Rodrigo Cardoso de Oliveira Abstract Objective: To evaluate the repair of critical-size bone defects in rats treated with demineralized bovine bone (DBB) compared with autogenous bone (AB). Material and method: A bone defect of 8 mm in diameter was created in the calvaria of 50 Rattus norvegicus, treated either with DBB or AB. Sub-groups of five rats of each group were killed at 7, 14, 21, 30 and 90 days post-operatively, and the skulls were removed and processed histologically. Histological sections were stained with hematoxylin and eosin. Result: Histological analysis showed complete closure of the defects with new bone at 90 days in group AB, and substitution of the biomaterial by fibrotic connective tissue in the DBB group at 21 days. Morphometric analysis showed that DBB was rapidly absorbed at 14 days, with its volume density decreasing from 47%±0.8% at 7 days to 1.2%±0.41% at 14 days. Subsequently, volume densities of the connective tissue and neoformed bone increased from 51.1%±11.17% to 86.8%±7.92% and from 1.9%±1.13% to 12%±8.02%, respectively, for the same time interval. The volume density of AB particles did not change throughout the experimental periods, but the amount of new bone increased markedly between 7 and 90 days, from 4.5%±1.57% to 53.5%±6.42% (P<0.05). Conclusion: DBB did not provide complete repair of the defects, with significantly less new bone formation than in the AB group. [source] Bovine-HA spongiosa blocks and immediate implant placement in sinus augmentation proceduresCLINICAL ORAL IMPLANTS RESEARCH, Issue 4 2002Histopathological, histomorphometric observations on different histological stainings in 10 consecutive patients Abstract: Bovine mineral spongiosa block (B-SB) was used as a bone substitute in sinus augmentation procedures in 10 consecutive patients. Implants were placed during the same session. The purpose of this study was to histopathologically examine the healing of the grafted site around the implants at 12 months. Radiographic follow-up showed apparent bone apposition in the augmented area around the implants. Clinically, all 36 implants were stable and integrated with the surrounding tissue. Histopathologically, new bone formation was evident in all specimen hard tissue cores. Hydroxyapatite particles were present in direct contact with the remodeled osseous tissue. Mallory trichrome staining showed different stages of mineralization and maturation of the newly formed bone around the grafted mineral particles. Morphometric evaluation of Picrosirius red stained slides under polarized light microscopy was performed at the peripheral/external and deep section slides of all specimens. The average bone area fraction was 34.2%, with a 1 : 5.4 mean lamellar/woven bone ratio at the peripheral side and 53.0%, with 1 : 2.5 mean ratio at the deep side. The differences of both parameters between the two sites were statistically significant. B-SB proved to be a suitable grafting material with simultaneously placed implants in sinus floor augmentation procedures. [source] Evaluation of processed bovine cancellous bone matrix seeded with syngenic osteoblasts in a critical size calvarial defect rat modelJOURNAL OF CELLULAR AND MOLECULAR MEDICINE, Issue 3 2006U. Kneser Abstract Introduction: Biologic bone substitutes may offer alternatives to bone grafting procedures. The aim of this study was to evaluate a preformed bone substitute based on processed bovine cancellous bone (PBCB) with or without osteogenic cells in a critical size calvarial defect rat model. Methods: Discs of PBCB (Tutobone®) were seeded with second passage fibrin gel-immobilized syngenic osteoblasts (group A, n = 40). Cell-free matrices (group B, n = 28) and untreated defects (group C; n=28) served as controls. Specimens were explanted between day 0 and 4 months after implantation and were subjected to histological and morphometric evaluation. Results: At 1 month, bone formation was limited to small peripheral areas. At 2 and 4 months, significant bone formation, matrix resorption as well as integration of the implants was evident in groups A and B. In group C no significant regeneration of the defects was observed. Morphometric analysis did not disclose differences in bone formation in matrices from groups A and B. Carboxyfluorescine-Diacetate-Succinimidylester (CFDA) labeling demonstrated low survival rates of transplanted cells. Discussion: Osteoblasts seeded into PBCB matrix display a differentiated phenotype following a 14 days cell culture period. Lack of initial vascularization may explain the absence of added osteogenicity in constructs from group A in comparison to group B. PBCB is well integrated and represents even without osteogenic cells a promising biomaterial for reconstruction of critical size calvarial bone defects. [source] Quantitative analysis of epithelial papillae in patients with oral lichen planusJOURNAL OF THE EUROPEAN ACADEMY OF DERMATOLOGY & VENEREOLOGY, Issue 6 2009P López-Jornet Abstract Background, The oral mucosa is relatively vulnerable to pathological processes, and is often affected by autoimmune and malignant diseases. The oral epithelium is normally non-homogeneous, and joins to the connective tissue through interlocking of its downward projections in the form of papillae. Objective, This study aims to conduct a histomorphometric study of the epithelial papillae in patients with oral lichen planus (OLP). Material and method, This study was based on 100 cheek mucosa biopsies from patients with OLP (66 white reticular and 34 atrophic-erosive) (13 males and 87 females, with a mean age of 54.95 ± 13.64 years). A histological and morphometric evaluation was made, based on imaging analysis with MIP software 4.5 for studying the papillary structure in the patients with OLP. Results, The mean epithelial thickness was 227.5 ± 78.5 µm. The different papillary measures , BLS (distance from basal layer to epithelial surface), DPS (distance from dermal papilla top to epithelial surface), DPW (dermal papilla width), and DPD (interdermal papilla distance between two papillae) , yielded no statistically significant differences with respect to age, sex, smoking and clinical form. However, a significant correlation was observed in relation to papilla width and inflammatory infiltrate (P = 0.031). Conclusions, The application of this imaging system is useful for measuring variations in epithelial papillary architecture. Conflicts of interest None declared. [source] |
