    MORPHOLOGICAL CHANGES IN MITOCHONDRIAL AND CHLOROPLAST NUCLEOIDS AND MITOCHONDRIA DURING THE CHLAMYDOMONAS REINHARDTII (CHLOROPHYCEAE) CELL CYCLE,

    Morphological changes in the organellar nucleoids and mitochondria of living Chlamydomonas reinhardtii Dang were examined during the cell cycle under conditions of 12:12 light:dark. The nucleoids were stained with SYBR-Green I, and the mitochondria were stained with 3,3-dihexyloxacarbocyanine iodide. An mocG33 mutant, which contains one large chloroplast nucleoid throughout the cell cycle, was used to distinguish between the mitochondrial and chloroplast nucleoids. Changes in the total levels of organellar DNA levels were assessed by real-time PCR. Each of the G1, S, M, and Smt,cp phases was estimated. At the start of the light period, the new daughter cells were in G1 and contained about 30 mitochondrial and 10 chloroplast nucleoids, which were dispersed and had diameters of 0.1 and 0.2 ,m, respectively. During the G1 phase of the light period, and at the start of the S phase, both nucleoids formed short thread-like or bead-like structures, probably divided, and increased continuously in number, concomitantly with DNA synthesis. The nucleoids probably became smaller due to the decrease in DNA of each particle and were indistinguishable. The cells in the S and M phases contained extremely high numbers of scattered nucleoids. However, in the G1 phase of the dark period, the nucleoids again formed short thread-like or bead-like structures, probably fused, and decreased in number. The mitochondria appeared as tangled sinuous structures that extended throughout the cytoplasm and resembled a single large mitochondrion. During the cell cycle, the numbers of mitochondrial nucleoids and sinuous structures varied relative to one another. [source]


    Alcohol-Induced Lipid and Morphological Changes in Chick Retinal Development

    Abstract: Background: Alcohol exposure causes alterations in the lipid content of different organs and a reduction of long-chain fatty acids. During embryo development, the central nervous system is extremely vulnerable to the teratogenic effects of alcohol, and the visual system is particularly sensitive. Methods: White Leghorn chick embryos were injected with 10- and 20-,l alcohol doses into the yolk sac at day 6 of incubation. The lipid composition of the retina was analyzed in embryos at day 7 of incubation (E7), E11, E15, and E18. The percentages of phospholipids, free cholesterol, esterified cholesterol, diacylglycerides, and free fatty acids were estimated by using an Iatroscan thin layer chromatography flame ionization detector. Gas chromatography and mass spectrometry were used to determine fatty acid composition. The morphological study was performed at E7, E11, and E19 by means of semithin and immunohistochemical techniques. Results: In the retina, alcohol causes the total lipid content to change, with a remarkable increase in free cholesterol and a dramatic decrease in esterified cholesterol. Diacylglycerides and free fatty acids tend to increase. Phosphatidylcholine and phosphatidylethanolamine decrease, whereas phosphatidylserine, sphingomyelin, and phosphatidylinositol increase. The main fatty acids of the retina also undergo changes. At E7, myriotic acid increases, and oleic acid and polyunsaturated fatty acids such as arachidonic acid and docosahexaenoic acid decrease. From E18 onward, there is some recovery, except for fatty acids, which recover earlier. From a morphological point of view, alcohol effects on retinal development are various: increase of intercellular spaces in all cell layers, pyknosis with loss of cellularity in the inner nuclear cell layer and ganglion cell layer, retarded or disorderly cell migration, early cell differentiation, and loss of immunoreactivity for myelin oligodendrocyte,specific protein. Conclusions: Acute alcohol exposure during embryo development causes the lipid composition of the retina to change, with a trend to recovery in the last stages. These alterations are in line with the changes observed at a morphological level. [source]


    Age-Related Three-Dimensional Morphological Changes in Rat Motoneurons Innervating Diaphragm and Longissimus Muscles

    Summary We investigated age-related morphological changes of rat motoneurons innervating diaphragm muscle (DI-MN) and lumber longissimus muscle (LL-MN) in which quite different activation patterns exist. In young (2,4 months) and old (24,26 months) rats, the motoneurons innervating both muscles were labelled retrogradely by intramuscular injection of cholera toxin B subunit. After a 4-day survival, horizontal slices of the spinal cord were processed with immunohistochemical staining (first antibody to cholera toxin B subunit and second antibody with Cy3) and observed with a confocal microscope. Three-dimensional reconstruction of labelled motoneurons was performed to examine soma and dendrite morphology. As compared to the soma volume in young rats, significantly smaller values were found in old rats in both motoneurons and the degrees of decline were 16.1% in DI-MN and 20.3% in LL-MN. Significant decreases in the thickness of primary dendrites were also found in both motoneurons, and the degrees of decline were 17.5% in DI-MN and 22.3% in LL-MN. Smaller changes were found in DI-MN than in LL-MN, indicating the possibility that increased activation by central drives can attenuate age-related morphological changes of the motor system in the spinal cord.     Effects of Monochromatic Light on Proliferation Response of Splencyte in Broilers

    Summary To investigate the effects of various monochromatic lights on splenocyte proliferation responses, a total of 260 Arbor Acre male broilers on P1 (post-hatching day 1) were exposed to blue light (BL), green light (GL), red light (RL) and white light treatments by light emitting diode system for 7 weeks, respectively. All light sources were equalized on the intensity of 15 lx and light period of 23 h daily. Morphological change of spleen and response of splenocyte proliferation were assessed by using histochemistry staining and colorimetric test in cultures of purified splenic cells. The results were as follows: (1) At P21, GL increased significantly the spleen weight by 163.6% and spleen index by 118.8% compared with RL (P < 0.05). Until P49, BL enhanced significantly the spleen weights by 42.2% compared with RL (P < 0.05), but no significant difference was found in the spleen index among four light-treated groups (P > 0.05). (2) Compared with RL, GL increased significantly the diameter of splenic nodule and area of periarterial lymphatic sheath at P21 by 87.2 and 58.1%, respectively (P < 0.05); BL increased significantly the diameter of splenic nodule and area of periarterial lymphatic sheath at P49 by 64.4 and 50.5%, respectively (P < 0.05). (3) At P21, GL enhanced spleen lymphocytes proliferation in response to concanavalin A compared with RL by 50.0% (P < 0.05). Until P49, the mitogenic response in BL was significantly higher (29.4%) than that of RL (P < 0.05). (4) The interleukin-2 (IL-2) bioactivity was significantly increased to 34.3% in GL than in RL at P21 (P < 0.05). Until P49, the IL-2 bioactivity in BL was significantly higher (62.2%) than that of RL (P < 0.05). (5) There was no significant difference in the nitric oxide (NO) concentration of splenocyte among RL, GL and BL groups at P21 (P > 0.05), but the concentration in RL group at P49 was significantly increased, 59.0 and 63.7% compared to that of GL and BL groups, respectively (P < 0.05). These results suggested that the monochromatic light affected splenocyte proliferation mainly because of alterations in IL-2 bioactivity and NO production in splenocyte of broiler. In early stage of broiler growth, the action of GL was obvious, while the response of BL was stronger in later stage. [source]


    Characteristics and dynamics of multiple intertidal bars, north Lincolnshire, England

    Abstract Multiple intertidal bars and troughs, often referred to as ,ridges and runnels', are significant features on many macrotidal sandy beaches. Along the coastline of England and Wales, they are particularly prevalent in the vicinity of estuaries, where the nearshore gradient is gentle and a large surplus of sediment is generally present. This paper examines the dynamics of such bar systems along the north Lincolnshire coast. A digital elevation model of the intertidal morphology obtained using LIDAR demonstrates that three to five intertidal bars are consistently present with a spacing of approximately 100 m. The largest and most pronounced bars (height = 0·5,0·8 m) are found around mean sea level, whereas the least developed bars (height = 0·2,0·5 m) occur in the lower intertidal zone. Annual aerial photographs of the intertidal bar morphology were inspected to try to track individual bars from year to year to derive bar migration rates; however, there is little resemblance between concurrent photographs, and ,resetting' of the intertidal profile occurs on an annual basis. Three-dimensional beach surveys were conducted monthly at three locations along the north Lincolnshire coast over a one-year period. The intertidal bar morphology responds strongly to the seasonal variation in the forcing conditions, and bars are least numerous and flattest during the more energetic winter months. Morphological changes over the monthly time scale are strongly affected by longshore sediment transport processes and the intertidal bar morphology can migrate along the beach at rates of up to 30 m per month. The behaviour of intertidal bars is complex and varies over a range of spatial and temporal scales in response to a combination of forcing factors (e.g. incident wave energy, different types of wave processes, longshore and cross-shore sediment transport), relaxation time and morphodynamic feedback.     The actin-binding protein profilin I is localized at synaptic sites in an activity-regulated manner

    Abstract Morphological changes at synaptic specializations have been implicated in regulating synaptic strength. Actin turnover at dendritic spines is regulated by neuronal activity and contributes to spine size, shape and motility. The reorganization of actin filaments requires profilins, which stimulate actin polymerization. Neurons express two independent gene products , profilin I and profilin II. A role for profilin II in activity-dependent mechanisms at spine synapses has recently been described. Although profilin I interacts with synaptic proteins, little is known about its cellular and subcellular localization in neurons. Here, we investigated the subcellular distribution of this protein in brain neurons as well as in hippocampal cultures. Our results indicate that the expression of profilin I varies in different brain regions. Thus, in cerebral cortex and hippocampus profilin I immunostaining was associated predominantly with dendrites and was present in a subset of dendritic spines. In contrast, profilin I in cerebellum was associated primarily with presynaptic structures. Profilin I immunoreactivity was partially colocalized with the synaptic molecules synaptophysin, PSD-95 and gephyrin in cultured hippocampal neurons, indicating that profilin I is present in only a subset of synapses. At dendritic spine structures, profilin I was found primarily in protrusions, which were in apposition to presynaptic terminal boutons. Remarkably, depolarization with KCl caused a moderate but significant increase in the number of synapses containing profilin I. These results show that profilin I can be present at both pre- and postsynaptic sites and suggest a role for this actin-binding protein in activity-dependent remodelling of synaptic structure.     Effect of endodontic irrigation on bonding of resin cement to radicular dentin

    The influence of endodontic irrigation on shear bond strengths of resin cement to radicular dentin was investigated. Human radicular dentin blocks were divided into four groups and subjected to one of four endodontic irrigations: ethylenediaminetetraacetic acid (EDTA) group, 17% EDTA for 60 s; EDTA/sodium hypochlorite (NaOCl) group, 17% EDTA for 60 s followed by 10 ml of 5% NaOCl for 15 s; NaOCl group, 10 ml of 5% NaOCl for 15 s; and control group, no treatment. Morphological changes of dentin surface after endodontic irrigation were observed by scanning electron microscopy (SEM). A resin block was bonded to the radicular dentin after irrigation using resin cement with either wet-bonding (Uni-Etch/One-Step; Bisco) or self-etching (Tyrian SPE/One-Step Plus; Bisco) adhesives. Shear bond strengths were measured and the penetration of resin tags into dentinal tubules at resin,dentin interface was observed by SEM. With the wet-bonding system, the shear bond strengths for the EDTA/NaOCl group, in which dentinal tubules openings and uniform resin tag penetration into dentinal tubules were observed, were significantly higher than the EDTA and control groups. With the self-etching system, the shear bond strengths were significantly lower in the EDTA group compared with the NaOCl and control groups. The effects of endodontic irrigation on the bonding of resin cement to radicular dentin depended on the dentin bonding system used. [source]


    Post-embryonic development of the Furongian (late Cambrian) trilobite Tsinania canens: implications for life mode and phylogeny

    SUMMARY The current concept of the order Asaphida was proposed to accommodate some Cambrian and Ordovician trilobite clades that are characterized by the possession of a ventral median suture. The family Tsinaniidae was recently suggested to be a member of the order Asaphida on the basis of its close morphological similarity to Asaphidae. Postembryonic development of the tsinaniid trilobite, Tsinania canens, from the Furongian (late Cambrian) Hwajeol Formation of Korea, reveals that this trilobite had an adult-like protaspis. Notable morphological changes with growth comprise the effacement of dorsal furrows, sudden degeneration of pygidial spines, regression of genal spines, and loss of a triangular rostral plate to form a ventral median suture. Programmed cell death may be responsible for degenerating the pygidial and genal spines during ontogeny. Morphological changes with growth, such as the loss of pygidial spines, modification of pleural tips, and effacement of dorsal furrows, suggest that T. canens changed its life mode during ontogeny from benthic crawling to infaunal. The protaspid morphology and the immature morphology of T. canens retaining genal and pygidial spines suggest that tsinaniids bear a close affinity to leiostegioids of the order Corynexochida. Accordingly, development of a ventral median suture in T. canens demonstrates that the ventral median suture could have evolved polyphyletically, and thus the current concept of the order Asaphida needs to be revised.     Morphological changes in the shape of the non-pathological bony knee joint with age: a morphometric analysis of the distal femur and proximal tibia in three populations of known age at death

    Abstract This study examines possible morphological variation in the knee joint of Homo sapiens with increasing age in ostensively healthy and non-pathological distal femora and proximal tibiae. Throughout the lifetime of each individual, the hard tissue of the knee undergoes considerable remodelling as a response to biomechanical stresses, changes in bone microarchitecture and reduction of bone mineral content as a concomitant of ageing. The knee is also subject to greater levels of degenerative joint disease than any other joint. If death occurs whilst such diseases are in the earliest stages, initial bone changes may not be visually obvious in museum specimens. If such specimens are used for comparative analyses, it is hypothesised that changes might render it problematic if all ages are conglomerated into discrete samples. This study therefore investigates the degree to which the distal femur and proximal tibia change shape during ageing and, if changes are present, whether they are expressed similarly in males and females. It also examines whether changes are of greater magnitude than those morphological differences which might exist between populations. In an example population of African-Americans, results indicate that there is a statistically significant difference in shape between age groups and those differences become progressively greater between the youngest and oldest adults. Results also show that although morphological variation caused by ageing is apparent, those shape differences attributable to sexual dimorphism are more powerful. When two additional populations are analysed jointly with the African-Americans (Caucasian Americans and the European Spitalfields sample), results indicate that inter-population shape differences are considerably greater than differences caused by increasing age.     Combined pulmonary toxicity of cadmium chloride and sodium diethyldithiocarbamate

    Abstract The pulmonary toxicity of sodium diethyldithiocarbamate and cadmium chloride, each separately and in combination, was compared in Sprague-Dawley rats after single intratracheal instillation in sequential experiments by chemical, immunological and morphological methods. With combined exposure, the cadmium content of the lungs increased permanently relative to that of the lungs of just cadmium-treated animals. Immunoglobulin levels of the whole blood did not change, whereas in bronchoalveolar lavage the IgA and IgG levels increased significantly. Morphological changes were characteristic of the effects of cadmium but were more extensive and more serious than in the case of cadmium administration alone: by the end of the first month, interstitial fibrosis, emphysema and injury of membranes of type I pneumocytes developed and hypertrophy and loss of microvilli in type II pneumocytes were detectable. These results showed that although dithiocarbamates as chelating agents are suitable for the removal of cadmium from organisms, they alter the redistribution of cadmium within the organism, thereby increasing the cadmium content in the lungs, and structural changes are more serious than observed upon cadmium exposure alone.     Paradoxical enhancement of oxidative cell injury by overexpression of heme oxygenase-1 in an anchorage-dependent cell ECV304

    Abstract There has been increasing evidence suggesting the potent anti-inflammatory roles of heme oxygenase-1 (HO-1) in protecting renal tubular epithelial cells, vascular endothelial cells, and circulating monocytes. Based on these findings, novel therapeutic interventions have been proposed to control the expression of endothelial HO-1 levels to ameliorate various vascular diseases. We evaluated the effect of HO-1 gene transfer into an anchorage-dependent cell, ECV304. Effect of HO-1 production on the cell injury induced by hydrogen peroxide was evaluated after hemin stimulation and after HO-1 gene transfection. Morphological changes and the induction of various anti-apoptotic proteins were examined at the same time. Levels of HO-1 expression were variable in different clones of HO-1-transfected ECV304 cells. Among these, the clones with moderate levels of HO-1 expression were significantly more resistant to oxidative stress. In contrast, those with the highest levels of HO-1 exhibited paradoxically enhanced susceptibility to oxidative injury. Interestingly, the cell survival after oxidative stress was in parallel with the levels of Bcl-2 expression and of fibronectin receptor, ,5 integrin. It is suggested from these results, that excessive HO-1 not only leads to enhanced cell injury, but also prolongs the repair process of the injured endothelial tissue. However, HO-1 reduces the oxidative cell injury and protects the endothelial cells, if its expression is appropriately controlled. © 2004 Wiley-Liss, Inc. [source]


    Experimental study of the safety of the selective COX-2 inhibitor, celecoxib, for gastric mucosa

    OBJECTIVE: To compare the gastric mucosal damage induced by a COX-2 inhibitor, celecoxib, and a conventional NSAID, indomethacin. METHODS: A rat model of NSAID-induced gastric mucosal damage was prepared for indomethacin and celecoxib separately (n = 8). After gastric damage was induced by 100% ethanol, celecoxib was administered by gastric gavage (n = 8). Gastric mucosal concentrations of 6-keto-PGF1, and TXB2 and the lesion index (LI) were measured. Morphological changes of the gastric mucosa were assessed under light and scanning electron microscopy. RESULTS: Indomethacin caused marked gastric damage (LI: 13.38 ± 2.06) and significant reduction of the concentrations of 6-keto-PGF1, and TXB2 (P < 0.01), Celecoxib did not produce necrotic injuries on healthy gastric mucosa (LI: 0), but the mucosal injuries previously induced by ethanol worsened after its administration (LI: 37.19 ± 3.34 vs 19.90 ± 2.28, P < 0.01). CONCLUSIONS: Inhibition of COX-1 is the major mechanism of NSAIDs in producing gastric mucosal damage. As a selective COX-2 inhibitor, celecoxib does not produce toxic injuries of the healthy gastric mucosa, and is thus safer than conventional NSAID. However, when administered in the presence of an altered gastric mucosa, gastric injuries were worsened. [source]


    Beneficial effects of growth hormone on bacterial translocation during the course of acute necrotizing pancreatitis in rats

    OBJECTIVE: Because bacterial translocation from the gut is one of the important sources of bacterial infection in acute necrotizing pancreatitis (ANP), and growth hormone (GH) has the ability to promote intestinal epithelial proliferation, we investigated the effects of GH on bacterial translocation in a rat ANP model. METHODS: Acute necrotizing pancreatitis was induced in rats via injection of 5% sodium taurocholate into the biliopancreatic duct. The rats with ANP were treated with either human recombinant GH or a placebo. Laparotomized animals without ANP induction (sham operation) served as controls. Twenty-four hours after the operation, blood was drawn for bacterial culture and determinations of amylase, lipase and endotoxin. Peritoneal fluid and specimens of mesenteric lymph nodes (MLN), liver, pancreas and spleen were taken for bacterial culture by standard techniques. Intestinal mucosal permeability was assessed by measuring the movement of [125I]-labeled albumin from blood to the intestinal lumen. Insulin-like growth factor-1 (IGF-1) mRNA was detected in the liver and ileum by reverse transcription,polymerase chain reaction (RT-PCR). Morphological changes in the pancreas and ileum were also analyzed. RESULTS: Administration of GH significantly decreased the activity of serum amylase and lipase, decreased the plasma endotoxin level and reduced the incidence of bacterial translocation. Moreover, the survival rate of ANP rats was improved. The severity of inflammation in the pancreas and ileum was reduced by GH treatment. Ileal mucosal thickness, villus height and crypt depth in GH-treated rats were obviously increased as compared with those of ANP rats. The intestinal permeability was markedly decreased in the GH group as compared with the ANP group. GH treatment resulted in upregulation of IGF-1 mRNA expression in ileum, but not in liver. CONCLUSIONS: These results suggest that exogenous GH has beneficial effects in maintaining the integrity of the intestinal mucosal barrier and reducing the incidence of bacterial translocation in rats with ANP. One of the mechanisms might be the upregulation of IGF-1 mRNA in the intestine by GH. [source]


    Morphological changes during postembryonic development in two species of neotropical harvestmen (Opiliones, Laniatores, Cranaidae)

    Abstract Morphological changes during postembryonic development in the Cranaidae are described on the basis of the examination of an incomplete series of larvae, nymphs, and adults of Phareicranaus calcariferus and Santinezia serratotibialis. The life histories of these species are hypothesized to consist of six nymphal stages, featuring the appearance of secondary male sexual characteristics in the antepenultimate nymph (N5). Color and body shape change dramatically during development. Growth rates for nymphs based upon leg measurements were similar for both species. In S. serratotibialis, the greatest increase in leg size occurred from larva to 1st nymph. The tarsomeres of legs I,IV varied by 1,2 segments per leg for each nymph stage, with the number of tarsal segments increased by 1,2 segments at each stage. Adults had nearly twice as many tarsomeres on leg II than other legs. Ontogenetic changes were observed in the armature of the proximal cheliceral segment, ocularium, pedipalp, opisthosoma, distitarsus III and IV, and leg IV.     Abstract Activated astrocytes show various patterns of Ca2+ mobilization under pathological conditions. In the present study we revealed a novel function of astrocytic Ca2+ dynamics through investigation of thrombin-induced unique Ca2+ entry. Using 1321N1 human astrocytoma cells, which have been shown to be a good model for detecting morphological dynamics, we observed rapid retraction of bipolar protrusions that were reversibly evoked by 0.03,3 U/mL thrombin. Morphological changes were predominantly dependent on a specific thrombin receptor subtype, proteinase-activated receptor 1 (PAR-1). In parallel, Fura-2 imaging of intracellular Ca2+ concentration ([Ca2+]i) showed that thrombin induced heterogeneous Ca2+ responses with asynchronous repetitive peaks. These oscillations were found to be a result of repetitive Ca2+ release from intracellular stores, followed by refilling of Ca2+ from the extracellular region without a direct [Ca2+]i increase. Pharmacological manipulation with BAPTA-AM, cyclopiazonic acid, and 2-aminoethoxydiphenyl borate indicated that Ca2+ mobilization was involved in thrombin-induced morphological changes. We further addressed the role of Ca2+ entry using small interfering RNA (siRNA) for transient receptor potential canonical 3 (TRPC3). As a result, both thrombin-induced morphological changes and oscillatory Ca2+ responses were significantly attenuated in siRNA-transfected cells. Inhibition of TRPC3 with pyrazole-3 also provided support for the contribution of Ca2+ influx. Moreover, TRPC3-mediated Ca2+ dynamics regulated thrombin-induced phosphorylation of myosin light chain 2. These results suggest a novel function of astrocytic Ca2+ dynamics, including Ca2+ entry, in the pathophysiological effects of PAR-1-mediated astrocytic activation.     Solvent-mediated solid phase transformations of carbamazepine: Effects of simulated intestinal fluid and fasted state simulated intestinal fluid

    Abstract Solvent-mediated transformations of carbamazepine (CBZ) anhydrate form III were investigated in Simulated Intestinal Fluid, a simple USP buffer medium, and in FaSSIF, which contains sodium taurocholate (STC) and lecithin, important surfactants that solubilize lipophilic drugs and lipids in the gastrointestinal tract. Raman spectroscopy (in situ) was utilized to reveal the connection between the changes in solid phase composition and dissolution rate while simultaneously detecting the solid state and the dissolved amount of CBZ. Initial dissolution rate was clearly higher in FaSSIF, while the solid phase data revealed that the crystallization of CBZ dihydrate was inhibited in both the dissolution media, albeit by different mechanisms. In SIF this inhibition was related to extensive needle growth, which impeded medium contact with the solid surface by forming a sterical barrier leading to retarded crystallization rates. Morphological changes from the needle-like dihydrate crystals to plate-like counterparts in FaSSIF, combined with the information that the transformation process was leveled off, evidenced strong hydrogen bonding behavior between the CBZ and STC molecules.     Morphological changes in the organellar nucleoids and mitochondria of living Chlamydomonas reinhardtii Dang were examined during the cell cycle under conditions of 12:12 light:dark. The nucleoids were stained with SYBR-Green I, and the mitochondria were stained with 3,3-dihexyloxacarbocyanine iodide. An mocG33 mutant, which contains one large chloroplast nucleoid throughout the cell cycle, was used to distinguish between the mitochondrial and chloroplast nucleoids. Changes in the total levels of organellar DNA levels were assessed by real-time PCR. Each of the G1, S, M, and Smt,cp phases was estimated. At the start of the light period, the new daughter cells were in G1 and contained about 30 mitochondrial and 10 chloroplast nucleoids, which were dispersed and had diameters of 0.1 and 0.2 ,m, respectively. During the G1 phase of the light period, and at the start of the S phase, both nucleoids formed short thread-like or bead-like structures, probably divided, and increased continuously in number, concomitantly with DNA synthesis. The nucleoids probably became smaller due to the decrease in DNA of each particle and were indistinguishable. The cells in the S and M phases contained extremely high numbers of scattered nucleoids. However, in the G1 phase of the dark period, the nucleoids again formed short thread-like or bead-like structures, probably fused, and decreased in number. The mitochondria appeared as tangled sinuous structures that extended throughout the cytoplasm and resembled a single large mitochondrion. During the cell cycle, the numbers of mitochondrial nucleoids and sinuous structures varied relative to one another. [source]


    Morphological changes in human gastric tumours after eradication therapy of Helicobacter pylori in a short-term follow-up

    Summary Background :,It is controversial as to whether the development of gastric cancer is influenced by Helicobacter pylori eradication. If eradication itself influences the tumour morphology, this may affect the tumour discovery rate. Aim :,To investigate the morphological changes in the gastric neoplasm after H. pylori eradication. Methods :,We studied 37 patients with eradication therapy. After a 1-month follow-up, endoscopic re-evaluation was performed and the appearance was compared with first image. All lesions were resected endoscopically, and were subjected to histological assessment and to immunohistochemistry. Serum gastrin levels were determined before and after eradication. Results :,Twenty-nine of 37 patients underwent successful eradication. The appearance of 11 lesions (33% of 33 lesions) became indistinct after successful eradication. All lesions were of the superficial-elevated type and the height of the lesions decreased. We detected normal columnar epithelium over the neoplasm in eight of the lesions. Higher expression of single-stranded deoxyribonucleic acid in the deep area was characteristic in tumours with an indistinct appearance. These changes did not correlate with the serum gastrin levels. Conclusions :,The morphology of the gastric neoplasm change after eradication in the short-term. This may contribute to the decreased tumour discovery rate. [source]


    Experimental autoimmune cholangitis: a mouse model of immune-mediated cholangiopathy

    Abstract:Background: Primary biliary cirrhosis (PBC) is characterised by intra-hepatic immune-mediated cholangiopathy (non-suppurative destructive cholangitis (NSDC)). Although auto-reactive immune responses against pyruvate dehydrogenase complex (PDC) have been characterised in PBC, the lack of an animal model of the disease has limited study of the mechanisms of disease induction and the development of novel approaches to therapy. Aims: To develop and validate a mouse model of immune-mediated cholangiopathy relevant for future use in the study of the aetio-pathogenesis and therapy of PBC. Methods: Female SJL/J, C57BL/6, NOD and BALB/c mice were sensitised with PDC, its purified E2/E3BP component, and a PDC-E2 derived peptide p163 (a dominant T-cell epitope in humans) in complete Freund's adjuvant (CFA). Morphological changes were assessed under light microscopy by a hepatic histopathologist blinded to the experimental details. Antibody responses to PDC were studied by ELISA and PDC inhibition assay. Results: An initial series of experiments was performed to survey the susceptibility of female mice of a range of strains to the induction of NSDC by i.p. sensitisation with PDC, PDC-E2/E3BP or p163 in CFA. Although each animal showed a specific antibody response following sensitisation, it was found that NSDC development (assessed at 30 weeks post-sensitisation) was restricted to SJL/J mice following sensitisation with any of the mitochondrial antigen preparations. A subsequent series of experiments was performed to examine the specificity and aetiology of this disease. Significant bile duct lesions were only seen in SJL/J animals following sensitisation with CFA containing PDC, and were absent from CFA only and un-sensitised controls. Kinetic analysis revealed that this pathology developed slowly, but a high incidence of animals with severe lesions was observed after 30 weeks. Conclusions: We have described a model of experimental autoimmune cholangitis (EAC) with immunological (anti-PDC antibodies) and histological (immune-mediated cholangiopathy) features suggestive of PBC. This model may be useful in further defining the role of self-tolerance breakdown in the development of this condition. [source]


    Morphological changes in mouse embryos cryopreserved by different techniques

    Abstract Cryopreservation of mammalian embryos is an important tool for the application of reproductive biotechnologies. Subjective evaluation to determine embryo viability is often used. The determination of the best cryopreservation protocol depends on morphological and molecular analysis of cellular injuries. The main objective of this study was to compare two methods of cryopreservation by assessing morphological alterations of frozen embryos using light, fluorescence, and transmission electron microscope. Fresh (control), slow frozen, and vitrified mouse embryos were composed. To evaluate the viability of the embryos, the cell membrane integrity was assessed using Hoechst33342 and propidium iodide (H/PI) staining. Morphological analyses using hematoxylin and eosin (HE) staining were performed to test different techniques (in situ, paraffin, and historesin) by both light and fluorescence microscopy. Transmission electron microscope was used to detect ultrastructural alterations in Spurr- and Araldite-embedded samples. H/PI staining detected more membrane permeability in the vitrification (69.8%) than in the slow freezing (48.4%) or control (13.8%) groups (P < 0.001). Historesin-embedded samples showed to be more suitable for morphological analyses because cellular structures were better identified. Nuclear evaluation in historesin sections showed the induction of pycnosis in slow freezing and vitrification groups. Cytoplasm evaluation revealed a condensation and an increase in eosinophilic intensity (indicating apoptosis) in the slow freezing group, and weakly eosinophilic structures and degenerated cells (indicating oncosis) in the vitrification group (P < 0.05). Ultrastructural analyses confirmed HE morphological findings. It was concluded that both cryopreservation techniques resulted in oncosis and apoptosis injuries.     Short-term changes in temporomandibular joint function in subjects with cleft lip and palate treated with maxillary distraction osteogenesis

Design,,, Morphological changes in the maxillofacial region were measured using lateral cephalometric radiographs taken immediately before (pre-DO) and after DO (post-DO) and 1 year after DO (1-year follow-up). A questionnaire was evaluated using a visual analog scale. A chi-square test was used to compare the prevalence of TMJ symptoms between pre-DO and 1-year follow-up. The Spearman correlation coefficient was used to determine the correlation between changes in cephalometric variables and TMJ symptoms in association with maxillary DO. Statistical significance was set at p < 0.05. Results,,, The ANB (anteroposterior relationship of the maxilla with the mandible) angle and the mandibular plane angle at pre-DO, post-DO, and 1-year follow-up were ,4.3°, +5.8°, +4.3° and 32.1°, 33.5°, 33.6°, respectively. The average amounts of anterior and downward movement of the maxilla at post-DO and 1-year follow-up were 8.3, ,1.3 and 0.9, 1.1 mm, respectively. The prevalence of TMJ symptoms showed no significant increase in association with maxillary DO. Moreover, there was no significant correlation between changes in cephalometric variables and TMJ symptoms. Conclusion,,, These results suggest that there was no short-term (i.e., up to 1 year after DO) effect of maxillary DO on TMJ function in subjects with CLP. [source]


    Morphological changes of sperm nuclei during spermatogenesis in the brown alga Cystoseira hakodatensis (Fucales, Phaeophyceae)

    SUMMARY Morphological changes and chromatin condensation of sperm nuclei were observed during spermatogenesis in the fucalean brown alga Cystoseira hakodatensis (Yendo) Fensholt. Ultrastructural studies have shown that the mature spermatozoid has an elongated and concave nucleus with condensed chromatin. The morphological changes and the chromatin condensation process during spermatogenesis was observed. Nuclear size decreased in two stages during spermatogenesis. During the first stage, spherical nuclei decreased in size as they were undergoing meiotic divisions and the subsequent mitoses within the antheridium. During the second stage, the morphological transformation from a spherical into an elongated nucleus occurred. Afterwards, chromatin condensed at the periphery in each nucleus, and chromatin-free regions were observed in the center of the nucleus. These chromatin-free regions in the center of nucleus were compressed by the peripheral chromatin-condensed region. As the result, the elongated and concave nucleus of the mature sperm consisted of uniformly well-condensed chromatin. [source]

    ABSTRACT Introduction and Aim., This study evaluated the possible differences between an angiotensin converting enzyme (ACE) inhibitor and a beta-blocker concerning their potential protective role on female external genitalia in spontaneously hypertensive rats (SHR). Main Outcome Measures., Morphological changes in the clitoris after antihypertensive treatments. Methods., For 6 months, SHR received no treatment; SHR + ramipril (RAM), SHR + atenolol (AT), and control Wistar Kyoto (WKY) rats received no treatment. Clitorises were processed for immunohistochemistry using anti-,-smooth muscle actin (,-SMA), anti-collagen I and III, anti-transforming growth factor ,1 (TGF,1), and anti-endothelial nitric oxide synthase (eNOS) antibodies. Results., SHR + RAM and SHR + AT presented significantly lower blood pressure in both groups vs. untreated SHR. Compared with WKY, ,-SMA was increased in the arteries and in the cavernous spaces of the clitoris together with a marked increase in wall/lumen ratio in clitoral vessels in untreated SHR. All these alterations were diminished in SHR + AT (P < 0.01). SHR + RAM presented differences with respect to SHR + AT in the reduction of these variables. TGF,1 expression in the vessel wall from the clitoris and collagen I and III deposition in the interstitium from the clitoris in untreated SHR were significantly more (P < 0.01) than in WKY. While SHR + AT showed a mild decrease in these variables, SHR + RAM presented a significant reduction (P < 0.01) in TGF,1 expression interstitial fibrosis and in both types of collagens. Positive immunostaining of eNOS in the sinusoidal endothelium from the clitoris was less (P < 0.01) in untreated SHR (3.4 ± 1.3%) and SHR + AT (5.1 ± 1.2%) than in SHR + RAM (17.2 ± 1.6%) and WKY (15.9 ± 1.7%). Untreated SHR and SHR + AT presented more surrounding connective tissue at the perineurium in the clitoris (P < 0.01) than SHR + RAM. Conclusion., ACE inhibition provided a considerable protective role on the female external genitalia structures in SHR by a mechanism that may be, at least in part, independent of the degree of blood pressure lowering.     Alterations in the expression of inhibitors of apoptosis during differentiation of prostate epithelial cells

    OBJECTIVE To investigate alterations in the apoptotic phenotype, specifically the inhibitors of apoptosis (IAP) family, in prostate epithelial cells after differentiation from an apoptotic-resistant basal cell to an apoptotic-susceptible secretory cell. MATERIALS AND METHODS Cells of the immortalized human prostate epithelial line HPr-1AR were cultured with and with no 5,-dihydrotestosterone (DHT) to drive differentiation. Western blotting and reverse transcription-polymerase chain reaction (RT-PCR) were used to determine changes in differentiation markers such as cytokeratins (CK) 14 and 18, and in XIAP, cIAP-1 and cIAP-2. Flow cytometry was used to assess viability and apoptosis, by propidium iodide DNA staining of the cells during differentiation. RESULTS Morphological changes and the increased CK-18 and decreased CK-14 expression confirmed differentiation of cells towards a secretory phenotype. Real-time PCR and Western blotting confirmed the expression of the IAPs in the HPr-1AR cells. There was a time-dependent decrease in the mRNA expression of XIAP, cIAP-1 and cIAP-2 after treatment with DHT. Differentiation also resulted in decreased cIAP-1 and XIAP protein expression, but cIAP-2 remained unchanged. Spontaneous apoptosis was significantly increased during cellular differentiation. CONCLUSION We show for the first time that differentiation of HPr-1AR prostate epithelial cells results in the development of a transient end-stage cell that might be explained by the loss of the IAP family of proteins.     Neuroprotection by donepezil against glutamate excitotoxicity involves stimulation of ,7 nicotinic receptors and internalization of NMDA receptors

    BACKGROUND AND PURPOSE Glutamate excitotoxicity may be involved in ischaemic injury to the CNS and some neurodegenerative diseases, such as Alzheimer's disease. Donepezil, an acetylcholinesterase (AChE) inhibitor, exerts neuroprotective effects. Here we demonstrated a novel mechanism underlying the neuroprotection induced by donepezil. EXPERIMENTAL APPROACH Cell damage in primary rat neuron cultures was quantified by lactate dehydrogenase release. Morphological changes associated with neuroprotective effects of nicotine and AChE inhibitors were assessed by immunostaining. Cell surface levels of the glutamate receptor sub-units, NR1 and NR2A, were analyzed using biotinylation. Immunoblot was used to measure protein levels of cleaved caspase-3, total NR1, total NR2A and phosphorylated NR1. Immunoprecipitation was used to measure association of NR1 with the post-synaptic protein, PSD-95. Intracellular Ca2+ concentrations were measured with fura 2-acetoxymethylester. Caspase 3-like activity was measured using enzyme substrate, 7-amino-4-methylcoumarin (AMC)-DEVD. KEY RESULTS Levels of NR1, a core subunit of the NMDA receptor, on the cell surface were significantly reduced by donepezil. In addition, glutamate-mediated Ca2+ entry was significantly attenuated by donepezil. Methyllycaconitine, an inhibitor of ,7 nicotinic acetylcholine receptors (nAChR), inhibited the donepezil-induced attenuation of glutamate-mediated Ca2+ entry. LY294002, a phosphatidyl inositol 3-kinase (PI3K) inhibitor, had no effect on attenuation of glutamate-mediated Ca2+ entry induced by donepezil. CONCLUSIONS AND IMPLICATIONS Decreased glutamate toxicity through down-regulation of NMDA receptors, following stimulation of ,7 nAChRs, could be another mechanism underlying neuroprotection by donepezil, in addition to up-regulating the PI3K-Akt cascade or defensive system.     Compression-Inhibited Pore Formation of Polyelectrolyte Multilayers Containing Weak Polyanions: A Scanning Force Microscopy Study

    Abstract Morphological changes of poly(acrylic acid)/poly(diallyldimethylammonium chloride) multilayers induced by low pH were investigated by scanning force microscopy. The weakened interaction between the charged polymer chains in the protonation process is believed to be the reason for this variation. Kinetic studies have shown that during protonation phase separation and dissociation of the multilayers took place successively. The compression of the multilayers, however, caused a transition of the multilayers from a rubbery state to a glassy state. As a result, the closely compacted multilayers lost their sensitivity to pH change. An increase of electrostatic and hydrophobic interactions, can decrease the free energy of the multilayers, and stabilize the films. By compression of the multilayers with a rubber stamp having geometric patterns, films with spatially localized pores were produced. [source]


    Can a failed ileal pouch anal anastomosis be left in situ?

    Abstract Objective, Failure after ileal pouch-anal anastomosis (IPAA) is reported with a frequency of 10,20%. The failed IPAA can be excised or defunctioned. Indications for excision and further management of an indefinitely diverted pouch are poorly described. The aim of the present investigation was to investigate pouch-related problems and the histopathological pattern of the pouch mucosa in this group of patients. Method, In a cohort of 620 patients having IPAA with a median follow-up of 14 years, 56 patients with failure were identified. The patients with defunctioned pouches were assessed with regard to pouch-related problems and endoscopy with biopsies was performed. Biopsies were stained with haematoxylin-eosin, PAS for neutral mucins and Alcian blue/high iron diamine for sialomucins/sulphomucins. Morphological changes were grouped into three types modified according to Veress and assessed for dysplasia. Results, Twenty-two patients with an indefinitely diverted pouch were found. The follow-up time after surgery for failure was 10 years. Thirteen patients completed the follow-up. Except for two patients with pelvic/perineal pain, there were no clinical problems. The majority of patients displayed mild to moderate macroscopic signs of inflammation. Morphologically, findings ranged from a preserved mucosal pattern to intense inflammatory reaction. No case of dysplasia or carcinoma was found. Conclusion, Most patients with an indefinitely diverted pouch had no complaints regarding the pouch. There was no case of dysplasia. Indefinite diversion may be preferable to pouch excision, especially given the associated morbidity. [source]


    Dynamics of multiple intertidal bars over semi-diurnal and lunar tidal cycles, North Lincolnshire, England

    Abstract Multiple intertidal bars are common features of wave-dominated sandy beaches, yet their short-term (<1 month) and small-scale (<1 km) morphology and dynamics remain poorly understood. This study describes the morphodynamics of multiple intertidal bars in North Lincolnshire, England, during single and lunar tidal cycles under two contrasting conditions , first when significant wave height was <0·5 m and second when significant wave height frequently exceeded 1 m. The relative importance of swash, surf and shoaling processes in determining morphological change was examined using detailed field observations and a numerical model. The beach featured four intertidal bars and both cross-shore and longshore bar morphology evolved during the field investigation, particularly under medium to high wave-energy conditions. Numerical modelling suggests shoaling processes are most common on the seaward two bars under calm wave conditions (Hs < 0·5 m) and that surf zone processes become more common during neap tides and under more energetic (Hs < 0·5 m) conditions. Surf processes dominate the inner two bars, though swash influence increases in a landward direction. The numerical modelling results combined with low tide survey data and high-resolution morphological measurements strongly suggest changes in the intertidal bar morphology are accomplished by surf zone processes rather than by shoaling wave or swash processes. This is because shoaling waves do not induce significant sediment transport to have any morphological effect, whereas swash action generally does not have enough scope to act as the swash zone is much narrower than the surf zone. It was found, however, that the absolute rate of morphological change under swash action and surfzone processes are of similar magnitudes and that swash action may induce a significant amount of local morphological change when the high tide mark is located on the upper bar, making this process important for bar morphodynamics.     Geostatistical analysis of ground-survey elevation data to elucidate spatial and temporal river channel change

    Abstract A digital elevation model (DEM) of a fluvial environment represented landform surface variability well and provided a medium for monitoring morphological change over time. Elevation was measured above an arbitrary datum using a ground-based three-dimensional tacheometric survey in two reaches of the River Nent, UK, in July 1998, October 1998 (after flood conditions) and June 1999. A detailed geostatistical analysis of the elevation data was used to model the spatial variation of elevation and to produce DEMs in each reach and for each survey period. Maps of the difference in elevation were produced and volumetric change was calculated for each reach and each survey period. The parameters of variogram models were used to describe the morphological character of each reach and to elucidate the linkages between process and the form of channel change operating at different spatial and temporal scales. The analysis of channel change on the River Nent shows the potential of geostatistics for investigating the magnitude and frequency of geomorphic work in other rivers. A flood modified the channel features, but low magnitude and high frequency flows rationalized the morphology. In spite of relatively small amounts of net flux the channel features changed as a consequence of the reworking of existing material. The blocking of chute entrances and redirection of the channel had a considerable effect on the behaviour of the channel. Such small changes suggested that the distributary system was sensitive to variation in sediment regime. Plots of the kriging variances against sampling intervals were used to quantify the temporal variation in sampling redundancy (ranging between ,11 per cent and +93 per cent). These curves illustrated the importance of bespoke sampling designs to reduce sampling effort by incorporating anisotropic variation in space and geomorphic information on flow regime. Variation in the nugget parameter of the variogram models was interpreted as sampling inaccuracy caused by variability in particle size and is believed to be important for future work on surface roughness.
