Particle clusters in gravel-bed rivers: an experimental morphological approach to bed material transport and stability concepts

Abstract Structured gravel river beds clearly exert a major influence on bed stability. Indexing structural stability by field measurements of bed strength neglects the processes operating to entrain and transport bed material in different parts of each structure. This study takes a morphological approach to interpreting the critical processes, using particle tracing to determine the movement of individual cluster particles over a range of flood event magnitudes and durations. The experiment was carried out on the River South Tyne, UK; it uses flow hydrographs measured nearby and also benefits from previous studies of historical development, channel morphology and sediment transport at the same site. More than 30 clusters were monitored over a seven-month period during which clusters occupied 7,16 per cent of the bed. Threshold flows delimiting three apparently contrasting bed sediment process regimes for cluster particles are tentatively set at 100 m3 s,1 and 183 m3 s,1; durations of flow at these levels are critical for cluster development, rather than flow peak values. Wake particles are transported most easily. Flow straightening in the wandering channel planform reduces the stability of clusters, since mechanical strength is markedly reduced by this change of direction. Close range digital photogrammetric analysis of experimental drainage basin evolution

Abstract Despite the difficulties of establishing formal hydraulic and geometric similarity, small-scale models of drainage basins have often been used to investigate the evolution and dynamics of larger-scale landforms. Historically, this analysis has been restricted to planform basin characteristics and only in the last decade has the topographic similarity of experimental landscapes been explored through explicitly three-dimensional parameters such as the distributions of cumulative drainage area, area,slope and catchment elevation. The current emphasis on three-dimensional morphometry reflects a growing awareness of the descriptive paucity of planform data and the need for more robust analysis of spatial scaling relationships. This paradigm shift has been significantly facilitated by technological developments in topographic survey and digital elevation modelling (DEM) which now present the opportunity to acquire and analyse high-resolution, distributed elevation data. Few studies have, however, attempted to use topographic modelling to provide information on the changing pattern and rate of sediment transport though an evolving landscape directly by using multitemporal DEM differencing techniques. This paper reports a laboratory study in which digital photogrammetry was employed to derive high-resolution DEMs of a simulated landscape in declining equilibrium at 15 minute frequency through a 240 minute simulation. Detailed evaluation of the DEMs revealed a vertical precision of 1·2 mm and threshold level of change detection between surfaces of ±3 mm at the 95 per cent confidence level. This quality assurance set the limits for determining the volumetric change between surfaces, which was used to recover the sediment budget through the experiment and to examine local - and basin-scale rates of sediment transport. A comparison of directly observed and morphometric estimates of sediment yield at the basin outlet was used to quantify the closure of the sediment budget over the simulation, and revealed an encouragingly small 6·2 per cent error. The application of this dynamic morphological approach has the potential to offer new insights into the controls on landform development, as demonstrated here by an analysis of the changing pattern of the basin sediment delivery ratio during network growth. Diverse developmental mechanisms contribute to different levels of diversity in horned beetles

Summary An ongoing challenge to evolutionary developmental biology is to understand how developmental evolution on the level of populations and closely related species relates to macroevolutionary transformations and the origin of morphological novelties. Here we explore the developmental basis of beetle horns, a morphological novelty that exhibits remarkable diversity on a variety of levels. In this study, we examined two congeneric Onthophagus species in which males develop into alternative horned and hornless morphs and different sexes express marked sexual dimorphism. In addition, both species differ in the body region (head vs. thorax) that develops the horn. Using a comparative morphological approach we show that prepupal growth of horn primordia during late larval development, as well as reabsorption of horn primordia during the pupal stage, contribute to horn expression in adults. We also show that variable combinations of both mechanisms are employed during development to modify horn expression of different horns in the same individual, the same horn in different sexes, and different horns in different species. We then examine expression patterns of two transcription factors, Distal-less (Dll) and aristaless (al), in the context of prepupal horn growth in alternative male morphs and sexual dimorphisms in the same two species. Expression patterns are qualitatively consistent with the hypothesis that both transcription factors function in the context of horn development similar to their known roles in patterning a wide variety of arthropod appendages. Our results suggest that the origin of morphological novelties, such as beetle horns, rests, at least in part, on the redeployment of already existing developmental mechanisms, such as appendage patterning processes. Our results also suggest, however, that little to no phylogenetic distance is needed for the evolution of very different modifier mechanisms that allow for substantial modulation of trait expression at different time points during development in different species, sexes, or tissue regions of the same individual. We discuss the implications of our results for our understanding of the evolution of horned beetle diversity and the origin and diversification of morphological novelties. [source]


Willan's itch and other causes of pruritus in the elderly

Itch in the elderly presents a diagnostic and therapeutic challenge. A thorough history, review of systems, and physical examination are critical to determining its cause. Examination of the skin may be misleading. There are frequently only secondary lesions, eczematous changes, lichenification, and excoriation, which may be misdiagnosed as a primary dermatitis. Xerosis may be the cause, but it is sometimes merely coincidental. If primary lesions are present, a skin biopsy can enable a diagnosis to be made. Systemic causes of itch, such as cholestasis, uremia, hyperthyroidism, medications, or lymphoma, must be considered. If the cause remains elusive, idiopathic itching of the elderly or so-called "senile pruritus" may be considered. However, we propose to discard the term "senile pruritus", which can be offensive and frightening. We propose to replace it with "Willan's itch". Robert Willan (1757,1812) is honored as one of the founders of modern dermatology thanks to his book, On Cutaneous Diseases, and its morphological approach to skin disease. He was probably the first to give a good clinical description of itching in the elderly. The diagnosis of Willan's itch should be reserved for generalized pruritus in the absence of xerosis or other recognizable cause. The pathophysiology of this form of pruritus is poorly understood, but it is likely that age-related changes of the skin, cutaneous nerves, and other parts of the nervous system play a role. Anecdotal and limited data suggest that gabapentin, cutaneous field stimulation, serotonin antagonists, and ultraviolet B phototherapy may attenuate itch in some of these patients. [source]


Adaptive radiation in African weakly electric fish (Teleostei: Mormyridae: Campylomormyrus): a combined molecular and morphological approach

Abstract We combined multiple molecular markers and geometric morphometrics to revise the current taxonomy and to build a phylogenetic hypothesis for the African weakly electric fish genus Campylomormyrus. Genetic data (2039 bp DNA sequence of mitochondrial cytochrome b and nuclear S7 genes) on 106 specimens support the existence of at least six species occurring in sympatry. We were able to further confirm these species by microsatellite analysis at 16 unlinked nuclear loci and landmark-based morphometrics. We assigned them to nominal taxa by comparisons to type specimens of all Campylomormyrus species recognized so far. Additionally, we showed that the shape of the elongated trunk-like snout is the major source of morphological differentiation among them. This finding suggests that the radiation of this speciose genus might have been driven by adaptation to different food sources. [source]


Underlying mechanism of portal hypertensive gastropathy in cirrhosis: A hemodynamic and morphological approach

Abstract Background and Aim:, Portal hypertensive gastropathy (PHG) is an important cause of bleeding in patients with cirrhosis associated with portal hypertension. Histologically, the condition is characterized by dilation of the mucosal and submucosal vessels of the stomach; however, its mechanisms remain unclear. The aim of the present cross-sectional study was to evaluate the role of portal and systemic hemodynamic features, humoral factors and hepatocellular function in the development and severity of PHG in patients with cirrhosis. Methods:, Forty-six patients with cirrhosis of different etiologies underwent endoscopy. Portal hypertension was evaluated by hepatic venous pressure gradient (HVPG). The gastric mucosa was analyzed using two diagnostic methods: endoscopy according to the McCormack criteria and histological by histomorphometric analysis. Results:, The prevalence of PHG according to the endoscopic and histomorphometric methods was 93.4% and 76.1%, respectively. There were no statistically significant differences in HVPG measurements between the patients with mild (16.0 ± 5.9 mmHg) and severe PHG (16.9 ± 6.5 mmHg; P = 0.80) or between patients who did not have (15.2 ± 8.0 mmHg) and those who had PHG (16.3 ± 5.7 mmHg). No correlation was found between the presence or severity of PHG and systemic vascular resistance index (P = 0.53 and 0.34, respectively), Child,Pugh classification (P = 0.73 and 0.78, respectively) or glucagon levels (P = 0.59 and 0.62, respectively). Conclusions:, The present data show no correlation between the presence or the severity of PHG and portal pressure, Child,Pugh classification or systemic hemodynamics, suggesting that other factors may be involved in the physiopathology of PHG, such as local gastric mucosal factors or other underlying factors. A four-fold humanity: Margaret Mead and psychological types

Beginning in 1933, while working in New Guinea, Margaret Mead developed her so-called squares hypothesis. Mead never published its terms, though she made a brief comment on it in her autobiography, Blackberry Winter (1972), and the arguments found in Sex and Temperament in Three Primitive Societies (1935) and the research leading to Balinese Character (Bateson & Mead, 1942) bore its imprint. Beginning with William McDougall's distinction between temperament (innate predispositions) and character (learned organization of habit), Mead articulated a morphological approach to the interplay between biology and culture that yielded four primary and four intermediary personality types. Under specified but not inevitable circumstances, the conscious choices of a given people could render one or another of these types characteristic or predominantly stable within their population, giving each of the other types a definite relation to the dominant type and thereby the cultural ethos of its society. Persons of each type followed a developmental path specific to their type different both from that of other types and in its manifestations given the various relations of the individual's type to the dominant type. Mead's hypothesis was, therefore, a vision of the unity and diversity of a single human species as well as an approach to the differing psychological positioning of individuals in cultures. In examining Mead's hypothesis, this essay also takes up Mead's debts to several leading psychologists (McDougall, C. G. The human premotor oculomotor brainstem system , can it help to understand oculomotor symptoms in Huntington's disease?

Recent progress in oculomotor research has enabled new insights into the functional neuroanatomy of the human premotor oculomotor brainstem network. In the present review, we provide an overview of its functional neuroanatomy and summarize the broad range of oculomotor dysfunctions that may occur in Huntington's disease (HD) patients. Although some of these oculomotor symptoms point to an involvement of the premotor oculomotor brainstem network in HD, no systematic analysis of this functional system has yet been performed in brains of HD patients. Therefore, its exact contribution to oculomotor symptoms in HD remains unclear. A possible strategy to clarify this issue is the use of unconventional 100-µm-thick serial tissue sections stained for Nissl substance and lipofuscin pigment (Nissl-pigment stain according to Braak). This technique makes it possible to identify the known nuclei of the premotor oculomotor brainstem network and to study their possible involvement in the neurodegenerative process. Studies applying this morphological approach and using the current knowledge regarding the functional neuroanatomy of this human premotor oculomotor brainstem network will help to elucidate the anatomical basis of the large spectrum of oculomotor dysfunctions that are observed in HD patients. This knowledge may aid clinicians in the diagnosis and monitoring of the disease. [source]


Fluorescence in situ hybridization analysis with a tissue microarray: ,FISH and chips' analysis of pathology archives

Practicing pathologists expect major somatic genetic changes in cancers, because the morphological deviations in the cancers they diagnose are so great that the somatic genetic changes to direct these phenotypes of tumors are supposed to be correspondingly tremendous. Several lines of evidence, especially lines generated by high-throughput genomic sequencing and genome-wide analyses of cancer DNAs are verifying their preoccupations. This article reviews a comprehensive morphological approach to pathology archives that consists of fluorescence in situ hybridization with bacterial artificial chromosome (BAC) probes and screening with tissue microarrays to detect structural changes in chromosomes (copy number alterations and rearrangements) in specimens of human solid tumors. The potential of this approach in the attempt to provide individually tailored medical practice, especially in terms of cancer therapy, is discussed. [source]


The use of volunteers for conducting sponge biodiversity assessments and monitoring using a morphological approach on Indo-Pacific coral reefs

Abstract 1.Sponges are an important component of coral reef ecosystems, but even though they are widespread with the ability to significantly influence other benthic community members they rarely feature to any great extent in current monitoring or biodiversity assessment programmes conducted by volunteer and professional groups. This exclusion is usually because of the taxonomic problems associated with sponge identification. 2.A potential alternative to monitoring temporal or spatial change in sponge assemblages and assessing biodiversity levels is to characterize sponges using morphologies present rather than collecting species data. Quantifying sponge biodiversity (for monitoring and biodiversity assessments) at the morphological level is less time and resource consuming than collecting species data and more suited to groups with little training and experience of sponge taxonomy or in regions where detailed taxonomic information on sponges is sparse. 3.This paper considers whether the same differences and similarities in sponge richness and assemblage composition can be identified using species and morphological data in response to environmental gradients at two coral reef ecosystems in south-east Sulawesi, Indonesia, and whether volunteers can be used to reliably collect morphological information. Sponge morphologies were classified into 14 groups and different morphological assemblages were found by the author at the two sites and between depth intervals. Comparisons of sponge species and morphological composition data showed that common patterns in assemblage structuring and richness could be identified irrespective of whether morphological or species data were used. In addition, a positive linear relationship was found between sponge species and morphological richness. 4.The morphological data recorded by volunteer divers (n=10) were compared with that collected by the author. Although volunteers recorded fewer sponges than the author (approximately 15% less), missing mainly small encrusting specimens, similar assemblage structure could be identified from both the volunteers' and the author's data. 5.The results showed that the same differences in sponge assemblages between sites and depths could be identified from both species and morphological data. Biology, ecology and status of Iberian ibex Capra pyrenaica: a critical review and research prospectus

ABSTRACT 1The Iberian ibex Capra pyrenaica is endemic to the Iberian Peninsula and of the four subspecies originally recognized, recent extinctions mean that only two now persist. Recent genetic analyses have cast doubt on the generally accepted taxonomy of the species, where four subspecies were distinguished by coat colour and horn morphology, and propose the distinction of two subspecies based on their mitochondrial DNA sequence polymorphism. These analyses make clear the need for a comprehensive revision that integrates genetic and morphological approaches resulting in a definitive description and differentiation of the subspecies. 2Studies of ibex behavioural ecology and health status are scarce and generally descriptive. They should be implemented in an integrative way, taking into account the ecological requirements of the species, current population status, the presence of other sympatric wild and domestic ungulates, and the type of hunting regime and management in their distribution areas. 3A natural expansion of the species is currently taking place. Ibexes are present and well established in all the main mountain ranges of the Spanish Iberian Peninsula, and have recently expanded their range into the north of Portugal. Other authors estimated a total population of more than 50 000 individuals 10 years ago, distributed over more than 60 000 km2, with an average population density of 2.7 ibex/km2. However, these estimates were obtained prior to the species' recovery from recent epizootics of sarcoptic mange and should be updated. Survey methods, mainly direct count-based methods, should be adjusted to suit mountainous conditions, where it is difficult to estimate accurately the surveyed surface. 4A series of threats to ibex conservation have been identified, such as population overabundance, disease prevalence and potential competition with domestic livestock and invasive ungulates, along with negative effects of human disturbance through tourism and hunting. 5Applied ecological issues focused on the proper management of populations should be prioritized, along with the identification of current threats based on empirical, ecological data obtained from populations living in various ecological conditions in different regions. Patterns of cryptic hybridization revealed using an integrative approach: a case study on genets (Carnivora, Viverridae, Genetta spp.) from the southern African subregion

Recent years have seen the development of molecular-based methodologies to investigate hybridization and its impact on the evolutionary process. However, morphological characterization of hybrid zones has only scantily been considered, especially in zootaxa. Thus, the level of congruence between molecular and morphological characters when attempting to detect hybrids remains a poorly tackled area. The genets (genus Genetta) provide an ideal case study for further investigation of the respective contribution of morphology and DNA in hybrid zone characterization because (1) their morphology has recently been exhaustively explored and (2) the existence of hybrid zones in southern Africa was proposed in the literature. We assessed levels of hybridization among the southern African genets, and questioned the role of ecological factors on the hybridization patterns detected. We used an integrative approach involving nine discrete morphological characters and a diagnostic discriminant function, geometric morphometrics and sequences of cytochrome b including collection specimens. The combination of independent materials allowed us to accurately reassess the level of hybridization in southern African genets, and revealed cryptic, interspecific gene flows. Morphology unambiguously detected a low number of G. maculata × G. tigrina hybrids and rejected the hypothesis of a large intergradation zone in KwaZulu-Natal, thus supporting the species status of the two genets. Cytochrome b analyses revealed: (1) cryptic, massive hybridization between G. tigrina and the sympatric G. felina, and (2) a trace of reticulation (one sequence) between G. tigrina and the allopatric G. genetta. The type specimen of G. mossambica Matschie, 1902 is considered to be a morphological hybrid between G. maculata and G. angolensis. Remarkably, the morphological approaches (discrete characters and morphometrics) proved complementary to conclusions derived from cytochrome b sequences. Whilst morphometrics was generally unable to accurately identify all putative hybrids, this approach revealed diagnostic cranial shape differences between recognized species as well as the cryptic G. ,letabae' (included in the super-species G. maculata). Morphometrics also confirmed the diagnostic value and age dependency of discrete characters. Our integrative approach appeared necessary to the detection of cryptic hybridizations and to the comprehensive characterization of hybrid zones. The recurrent detection of hybrids exhibiting tigrina -like coat patterns may suggest (1) asymmetric hybridization of G. tigrina males to females of other species and (2) positive selection for tigrina -like phenotype in South African habitats, but these hypotheses will have to be further tested using other sources of evidence. Despite the precise mosaic of hybrid zones identified in southern African genets, the environmental factors that shape patterns of distribution of hybrids remain unclear. Nevertheless, in the light of our range reassessment, it appears that seasonality of precipitation and periods of annual frost may play stringent roles in the distribution of genets. Microtubule-interacting drugs induce moderate and reversible damage to human bone marrow mesenchymal stem cells

Objectives:, This study aimed to investigate molecular and cellular changes induced in human bone marrow mesenchymal stem cells (hMSCs) after treatment with microtubule-interacting agents and to estimate damage to the bone marrow microenvironment caused by chemotherapy. Materials and methods:, Using an in vitro hMSC culture system and biochemical and morphological approaches, we studied the effect of nocodazole and taxol® on microtubule and nuclear envelope organization, tubulin and p53 synthesis, cell cycle progression and proliferation and death of hMSCs isolated from healthy donors. Results and conclusions:, Both nocodazole and taxol reduced hMSC proliferation and induced changes in the microtubular network and nuclear envelope morphology and organization. However, they exhibited only a moderate effect on cell death and partial arrest of hMSCs at G2 but not at M phase of the cell cycle. Both agents induced expression of p53, exclusively localized in abnormally shaped nuclei, while taxol, but not nocodazole, increased synthesis of ,-tubulin isoforms. Cell growth rates and microtubule and nuclear envelope organization gradually normalized after transfer, in drug-free medium. Our data indicate that microtubule-interacting drugs reversibly inhibit proliferation of hMSCs; additionally, their cytotoxic action and effect on microtubule and nuclear envelope organization are moderate and reversible. We conclude that alterations in human bone marrow cells of patients under taxol chemotherapy are transient and reversible. [source]
