Selected AbstractsUsing Profile Monitoring Techniques for a Data-rich Environment with Huge Sample SizeQUALITY AND RELIABILITY ENGINEERING INTERNATIONAL, Issue 7 2005Kaibo Wang Abstract In-process sensors with huge sample size are becoming popular in the modern manufacturing industry, due to the increasing complexity of processes and products and the availability of advanced sensing technology. Under such a data-rich environment, a sample with huge size usually violates the assumption of homogeneity and degrades the detection performance of a conventional control chart. Instead of charting summary statistics such as the mean and standard deviation of observations that assume homogeneity within a sample, this paper proposes charting schemes based on the quantile,quantile (Q,Q) plot and profile monitoring techniques to improve the performance. Different monitoring schemes are studied based on various shift patterns in a huge sample and compared via simulation. Guidelines are provided for applying the proposed schemes to similar industrial applications in a data-rich environment. Copyright © 2005 John Wiley & Sons, Ltd. [source] Brain Protection During Pediatric Cardiopulmonary BypassARTIFICIAL ORGANS, Issue 4 2010Xiaowei W. Su Abstract Improvements in peri- and postoperative surgical techniques have greatly improved outcomes for pediatric patients undergoing cardiopulmonary bypass (CPB) in the treatment of congenital heart defects (CHDs). With decreased mortality rates, the incidence of adverse neurological outcomes, comprising cognitive and speech impairments, motor deficits, and behavioral abnormalities, has increased in those patients surviving bypass. A number of mechanisms, including ischemia, reperfusion injury, hypothermia, inflammation, and hemodilution, contribute to brain insult, which is further confounded by unique challenges presented in the pediatric population. However, a number of brain monitoring and preventative techniques have been developed or are being currently evaluated in the practice of pediatric CPB. Monitoring techniques include electroencephalography, near-infrared as well as visible light spectroscopy, transcranial Doppler ultrasound, and emboli detection and classification quantitation. Preventative measures include hypothermic perfusion techniques such as deep hypothermic circulatory arrest, low-flow CPB, blood gas management, and pharmacologic prophylaxes, among others. The present review summarizes the principles of brain insult, neurodevelopmental abnormalities, monitoring techniques, methods of prevention, as well as preexisting morbidities and risk factors in pediatric CPB, with a focus on brain protection. Clinical and translational research is presented with the aim of determining methods that may optimize neurological outcomes post CPB and guiding further study. [source] Operative Results without Invasive Monitoring in Patients with Frontal Lobe Epileptogenic LesionsEPILEPSIA, Issue 10 2001A. Mariottini Summary: ,Purpose: To further explore the still controversial issues regarding whether all or most candidates for epilepsy surgery should be investigated preoperatively with invasive long-term video-EEG monitoring techniques (ILTVE). Methods: We studied five patients with intractable seizures since early childhood using the same protocol: clinical evaluation, magnetic resonance imaging (MRI) with fluid-attenuated inversion recovery (FLAIR) sequences, long-term video-EEG (LTVE) monitoring with scalp electroencephalogram (EEG), interictal single photon emission computed tomography (SPECT), positron emission tomography (PET), and neuropsychological testing. The patients' seizures had clinical features suggesting a frontal lobe (FL) origin. MRI scans revealed focal cortical dysplasia (CD) in four patients and a probable gliotic lesion in the fifth. The findings in both PET and SPECT images were congruent with those of the MRI. Scalp LTVE failed to localize the ictal onset, although the data exhibited features suggestive of both CDs and FL seizures. On the basis of these results, surgery was performed with intraoperative corticography, and the cortical area exhibiting the greatest degree of spiking was ablated. Results: Histopathologic study of four of the resected specimens confirmed the presence of CD, whereas in the fifth, there were features consistent with a remote encephaloclastic lesion. There were no postoperative deficits. Seizures in three of the patients were completely controlled at 2,3.5 years of follow-up; a fourth patient is still having a few seizures, which have required reinstitution of pharmacotherapy, and the fifth has obtained 70% control. All patients have had significant improvement in psychosocial measures. For comparison, five patients with generally similar clinical and neuroradiologic features to the previous group underwent preoperative ILTVE monitoring. The surgical outcomes between the two groups have not differed significantly. Conclusions: We conclude that patients with FL epilepsies may be able to undergo successful surgery without preoperative ILTVE monitoring, provided there is high concordance between neuroimaging tests (MRI, SPECT, PET) and the seizure phenotypes, even when routine EEGs and scalp LVTE fail to localize ictal onset unambiguously. The surgical outcomes of these patients generally paralleled those of the other subjects who also had FL epilepsy but who were operated on only after standard ILTVE monitoring. ERZINGER Abstract Continuous mud gas loggings during drilling as well as offline mud gas sampling are standard procedures in oil and gas operations, where they are used to test reservoir rocks for hydrocarbons while drilling. Our research group has developed real-time mud gas monitoring techniques for scientific drilling in non-hydrocarbon formations mainly to sample and study the composition of crustal gases. We describe in detail this technique and provide examples for the evaluation of the continuous gas logs, which are not always easy to interpret. Hydrocarbons, helium, radon and with limitations carbon dioxide and hydrogen are the most suitable gases for the detection of fluid-bearing horizons, shear zones, open fractures, sections of enhanced permeability and permafrost methane hydrate occurrences. Off-site isotope studies on mud gas samples helped reveal the origin and evolution of deep-seated crustal fluids. [source] Quality-by-Design (QbD): An integrated process analytical technology (PAT) approach for real-time monitoring and mapping the state of a pharmaceutical coprecipitation process,JOURNAL OF PHARMACEUTICAL SCIENCES, Issue 3 2010Huiquan Wu Abstract In this work, an integrated PAT approach was developed for monitoring a pharmaceutical (naproxen) and a polymer (eudragit) coprecipitation process: real-time in-line near-infrared (NIR) absorbance monitoring, real-time on-line turbidity monitoring, and in situ crystal size monitoring. The data and information obtained through these three monitoring techniques confirmed the observation of the onsets of three distinct stages: incubation, nucleation, and crystal growth. The process trajectory constructed based on results of applying principal component analysis (PCA) to either process NIR spectra data or process turbidity profile, clearly demonstrated that various distinguishable process events, including incubation, nucleation, and crystal growth, could be accurately tracked and differentiated. These findings were further supported by process knowledge and information, such as process design, process sequence, thermodynamic and mass-transfer analysis. J. O'Donnell ABSTRACT Historical anecdotes and preliminary monitoring since 1990 indicate that New Zealand Long-tailed Bats (Chalinolobus tuberculatus, Vespertilionidae) are now rare or absent at many sites where formerly they were common. Chalinolobus tuberculatus appeared to be common throughout New Zealand in the 1800s but by 1900,30 it was becoming scarce in many districts. Formal surveys in the South Island since 1990 either failed to find C. tuberculatus, or recorded bats in low numbers. Of eight sites where transect counts were undertaken, bats were recorded frequently at two sites (45,66% of counts; Eglinton and Dart Valleys), rarely at four sites (2.4,10.7% of counts), and were not recorded at the remaining two sites despite considerable survey effort. Of 10 sites where stationary counts using automatic detector units were used, no C. tuberculatus were recorded in three areas (153 nights combined), they were found rarely at six sites (2.1,21.0% of nights; 461 nights combined) and were recorded commonly only in the Eglinton Valley (85% of 120 nights). Assertions that C. tuberculatus are ,common' and that the conservation status is ,secure' are questionable and this review supports suggestions that the species should be classed as ,Vulnerable'. Possible causes of decline have been suggested including clearance and logging of lowland forests, predation by introduced mammals and owls, competition for roost sites by introduced mammals, birds and wasps, and human interference and disturbance at roosting sites. However, authors' claims have all been speculative and unsubstantiated. There has been no research undertaken to quantify these claims, and this is required urgently. The results of these preliminary surveys provide a new baseline against which future population trends might be compared. Increased effort using standardized monitoring techniques, applied at a national level, is required to confirm the possible trends and to help identify the best sites where conservation managers may attempt to restore the population level. [source] Monitoring the corrosion and remediation of reinforced concrete on-site: An alternative approachMATERIALS AND CORROSION/WERKSTOFFE UND KORROSION, Issue 4 2005N. Birbilis Abstract Difficulties associated with the interpretation of site data collected over long periods of time from commonly used electrochemical corrosion monitoring techniques often make it difficult to assess the corrosion of reinforcing steel. An alternative approach for the interpretation of data is proposed, based on a model of the quality of passive film upon the steel surface. This model leads to a representation of the corrosion state by means of the relationship, over a long period of time, of the corrosion potential and the logarithm of the Linear Polarization Resistance, since both are functions inter alia of corrosion rate. It is shown that for the reinforced concrete panels tested, data points representing this relationship closely fitted a family of results; allowing the development of a "monitoring control diagram", MCD. The MCD reveals that for a fixed geometry and experimental conditions, a relationship between the corrosion potential and polarization resistance of steel exists, facilitating a useful monitoring tool for assessment of both the corrosion and remediation of reinforced concrete structures. Particular emphasis is placed on the latter in this work. [source] Host specificity and reproductive success of yucca moths (Tegeticula spp.MOLECULAR ECOLOGY, Issue 24 2009Lepidoptera: Prodoxidae) mirror patterns of gene flow between host plant varieties of the Joshua tree (Yucca brevifolia: Agavaceae) Abstract Coevolution between flowering plants and their pollinators is thought to have generated much of the diversity of life on Earth, but the population processes that may have produced these macroevolutionary patterns remain unclear. Mathematical models of coevolution in obligate pollination mutualisms suggest that phenotype matching between plants and their pollinators can generate reproductive isolation. Here, we test this hypothesis using a natural experiment that examines the role of natural selection on phenotype matching between yuccas and yucca moths (Tegeticula spp.) in mediating reproductive isolation between two varieties of Joshua tree (Yucca brevifolia var. brevifolia and Y. brevifolia var. jaegeriana). Using passive monitoring techniques, DNA barcoding, microsatellite DNA genotyping, and sibship reconstruction, we track host specificity and the fitness consequences of host choice in a zone of sympatry. We show that the two moth species differ in their degree of host specificity and that oviposition on a foreign host plant results in the production of fewer offspring. This difference in host specificity between the two moth species mirrors patterns of chloroplast introgression from west to east between host varieties, suggesting that natural selection acting on pollinator phenotypes mediates gene flow and reproductive isolation between Joshua-tree varieties. [source] A practice survey on vasopressor and inotropic drug therapy in Scandinavian intensive care unitsACTA ANAESTHESIOLOGICA SCANDINAVICA, Issue 6 2003A. Oldner Background: This practice survey was performed to analyse the indications for use of vasopressor/inotropic drugs, preferred drugs and doses as well as concomitant monitoring and desired haemodynamic target values in Scandinavian ICUs. An internet-based reporting system was implemented. Methods: A total of 223 ICUs were identified in the Scandinavian countries and invited to participate in a one-day point-prevalence study. An internet-based database was constructed and a practice survey protocol designed to identify haemodynamic monitoring, indications for vasopressor/inotropic drug-therapy, fluids used for volume loading, pretreatment circulatory state, actual and targeted haemodynamic variables. Patients were eligible for the study if on vasopressor/inotropic drug-therapy for more than 4 h. Results: A total of 114 ICUs participated. A total of 114 adult patients matched the inclusion criteria. Sixty-seven per cent of the patients had received vasopressor/inotropic drug-treatment for >24 h and 32% received more than one drug. Arterial hypotension (92%) and oliguria (50%) were most common indications. Fluid loading prior to therapy was reported in 87% of patients. Dopamine (47%) and noradrenaline (44%) were the most commonly used drugs followed by dobutamine (24%). No other drug exceeded 6%. Non-catecholamine drugs were rarely used even in cardiac failure patients. Invasive arterial pressure was monitored in 95% of patients, pulmonary artery catheters were used in 19%. Other cardiac output monitoring techniques were used in 8.5% of the patients. Conclusion: Dopamine and noradrenaline seem to be the most commonly used inotropic/vasopressor drugs in Scandinavia. Traditional indications for inotropic/vasopressor support as hypotension and oliguria seem to be most common. Invasive monitoring was used in almost all patients, whereas a limited use of pulmonary artery catheters was noted. The internet-based reporting system proved to be an efficient tool for data collection. Identifying vascular access at risk of thrombosis is an evolving and important aspect to the management of any haemodialysis patient. Haemodynamic profiles of native fistulas and arteriovenous grafts differ significantly, and, as such, the sensitivity of the specific monitoring techniques to detect dysfunction varies depending on access type. Multiple strategies are now available for monitoring vascular access including measuring intra-access pressure and access blood flow, or screening for significant stenoses with Doppler ultrasonography. the ability of each strategy to detect significant stenosis is reviewed by looking at the evidence for both arteriovenous fistula (AVF) and arteriovenous grafts (AVG), separately. the majority of published literature involves AVG, and measuring access blood flow is the modality of choice. the best method for measuring flow is not known. For AVF, much less is known, and it is not clear whether monitoring will decrease the thrombosis risk and prolong access life. Recommendations for AVF cannot be made until more evidence becomes available. [source] Developing recovery and monitoring strategies for the endemic Mount Graham red squirrels (Tamiasciurus hudsonicus grahamensis) in ArizonaANIMAL CONSERVATION, Issue 1 2004Kate E. Buenau An important challenge in conservation biology is extracting pertinent information from the available data for endangered species. Rarely do we have enough information to precisely determine an organism's risk of extinction and other factors that affect its management. How, then, can we use limited information to make responsible conservation decisions on controversial species such as the Mount Graham red squirrel? We used several analytical approaches to examine 15 years of abundance data for the Mount Graham red squirrel in order to propose recovery criteria and to evaluate alternative conservation strategies. We analysed the historical population dynamics using a diffusion approximation model and showed that the main threat to the population was not the overall growth rate (which may well be greater than 1) but rather the wide range of variation in annual growth rates. We used information on the distribution of growth rates and abundance to classify the species as threatened under the Endangered Species Act (ESA). We then presented a simple demographic model to examine the effects of proportional changes in vital rates on the population growth rate (,). The elasticity values obtained for Mount Graham red squirrels indicate that the population is far more sensitive to changes in survival rates (particularly adult survival) compared to reproduction. Our analyses suggest that management should focus on refining monitoring techniques, reducing sources of variability, improving the survival of adult animals and filling the gaps in the currently available data. [source] A novel view on stem cell development: analysing the shape of cellular genealogiesCELL PROLIFERATION, Issue 2 2009I. Glauche Objectives: The analysis of individual cell fates within a population of stem and progenitor cells is still a major experimental challenge in stem cell biology. However, new monitoring techniques, such as high-resolution time-lapse video microscopy, facilitate tracking and quantitative analysis of single cells and their progeny. Information on cellular development, divisional history and differentiation are naturally comprised into a pedigree-like structure, denoted as cellular genealogy. To extract reliable information concerning effecting variables and control mechanisms underlying cell fate decisions, it is necessary to analyse a large number of cellular genealogies. Materials and Methods: Here, we propose a set of statistical measures that are specifically tailored for the analysis of cellular genealogies. These measures address the degree and symmetry of cellular expansion, as well as occurrence and correlation of characteristic events such as cell death. Furthermore, we discuss two different methods for reconstruction of lineage fate decisions and show their impact on the interpretation of asymmetric developments. In order to illustrate these techniques, and to circumvent the present shortage of available experimental data, we obtain cellular genealogies from a single-cell-based mathematical model of haematopoietic stem cell organization. Results and Conclusions: Based on statistical analysis of cellular genealogies, we conclude that effects of external variables, such as growth conditions, are imprinted in their topology. Moreover, we demonstrate that it is essential to analyse timing of cell fate-specific changes and of occurrence of cell death events in the divisional context in order to understand the mechanisms of lineage commitment. [source] Capillary electrophoresis for chimerism monitoring by PCR amplification of microsatellite markers after allogeneic hematopoietic cell transplantationCLINICAL TRANSPLANTATION, Issue 3 2005Alexandros Spyridonidis Abstract:, Background:, Hematopoietic chimerism has been demonstrated to be relevant for donor cell engraftment and detection of minimal residual disease after allogeneic hematopoietic cell transplantation (aHCT). In the light of increasing numbers of non-myeloablative aHCT as a treatment modality sensitive, rapid, and accurate chimerism monitoring techniques acquire novel relevance. Methods:, We evaluated the informativeness of five microsatellite markers in 376 donor/recipient pairs and evaluated the ability of capillary electrophoresis to detect mixed chimerism after aHCT. The sensitivity for capillary electrophoresis with respect to different markers was determined by limiting dilution assays with mixed chimerism samples containing defined amounts of cells or DNA. Furthermore, capillary electrophoresis was applied in 17 retrospectively selected patients with a mixed chimerism detected previously by gel electrophoresis, having undergone aHCT for different hematologic diseases and initially achieving a complete donor chimerism. Results:, In 163 of 165 (98%) of all related and 210 of 211 (99%) unrelated transplants the microsatellites identified informative alleles. The sensitivity and accuracy was higher with capillary electrophoresis when compared with gel electrophoresis with three of five microsatellites. Potential pitfalls with the application of capillary electrophoresis was preferential amplification and the occurrence of stutter peaks in the representative area. Investigation of the selected patient samples demonstrated that detection of a mixed chimerism was earlier with capillary electrophoresis when compared with gel electrophoresis. The detected recipient genotype by capillary electrophoresis examination, despite a negative gel electrophoresis result, ranged from 0.7 to 7.1%. Conclusions:, We conclude that chimerism assessment with our five microsatellites identified informative alleles in 99% of all donor/recipient pairs and may therefore be of use when establishing an institutional chimerism testing procedure. Capillary electrophoresis displayed a high sensitivity and accuracy for detecting a mixed chimerism in vitro and in vivo. [source] |
