Selected AbstractsThe Effects of Antihypertensive Treatment on the Doppler-Derived Myocardial Performance Index in Patients with Hypertensive Left Ventricular Hypertrophy: Results from the Swedish Irbesartan in Left Ventricular Hypertrophy Investigation Versus Atenolol (SILVHIA)ECHOCARDIOGRAPHY, Issue 7 2009Stefan Liljedahl M.D. Objectives: To investigate the effects of antihypertensive treatment on the Doppler-derived myocardial performance index (MPI) in patients with hypertensive left ventricular hypertrophy. Methods: The MPI was measured at baseline and after 48 weeks of antihypertensive treatment in 93 participants of the SILVHIA trial, where individuals with primary hypertension and left ventricular hypertrophy were randomized to double blind treatment with either irbesartan or atenolol. Results: Antihypertensive treatment lowered MPI (mean difference ,0.03 ± 0.01, P = 0.04). Changes in MPI by treatment were associated with changes in left ventricular ejection fraction (,-coefficient ,0.35 P = 0.005), stroke volume/pulse pressure (reflecting arterial compliance, ,-coefficient ,0.39 P < 0.001) and peripheral vascular resistance (,-coefficient 0.28 P < 0.04). Furthermore, there was a borderline significant association between changes in MPI and changes in E-wave deceleration time (reflecting diastolic function, ,-coefficient 0.23, P = 0.06). No associations were found between changes in MPI and changes in blood pressure, E/A-ratio, left ventricular mass index, relative wall thickness or heart rate. A stepwise multivariable regression model confirmed the association between changes in MPI and changes in ejection fraction and stroke volume/pulse pressure (all P < 0.05), as well as the trend for E-wave deceleration time (P = 0.08), but not in the case of peripheral vascular resistance. Conclusion: The MPI exhibited a modest decrease after 48 weeks of antihypertensive treatment in patients with hypertensive left ventricular hypertrophy. Changes in MPI were associated with changes in left ventricular function and vascular compliance, rather than with changes in left ventricular remodeling or blood pressure. MacLaren PharmD Summary Objective:, Review the literature regarding the use of recombinant human erythropoietin (rHuEPO) to prevent red blood cell (RBC) transfusion in critically ill patients. Data sources:, A computerized search of MEDLINE and EMBASE from 1966 through June 2003 was conducted using the terms erythropoietin, anemia, hemoglobin, critical care, intensive care, surgery, trauma, burn, and transfusion. References of selected articles were reviewed. A manual search of critical care, surgery, trauma, burn, hematology, and pharmacy journals was conducted to identify relevant abstracts. Results:, Six randomized studies have evaluated exogenous administration of erythropoietin to prevent RBC transfusions in critically ill patients. Studies vary with respect to rHuEPO dosage regimens, dose of concurrently administered iron, patient characteristics, and transfusion thresholds. Administration of rHuEPO rapidly produces erythropoiesis to reduce the need for RBC transfusions. The largest study conducted to date used weekly rHuEPO administration and found a modest decrease in transfusion requirements although the time to first transfusion was delayed. Reduced intensive care unit (ICU) length of stay (LOS) was shown in only one study of surgical/trauma patients. Reduced LOS after ICU discharge was found in another study of severely ill patients (APACHE II score >22). Other clinical outcomes were not altered by rHuEPO use. No adverse events were associated with rHuEPO use although studies were not designed to evaluate safety. Conclusions:, rHuEPO reduces the need for transfusions. A cost-effectiveness analysis of rHuEPO for this indication is needed. Defining an optimal dosage regimen, identifying patients most likely to respond to rHuEPO, and determining risk factors for ICU associated anaemia would provide information for appropriate rHuEPO utilization. [source] Ethanol inhibits cold-menthol receptor TRPM8 by modulating its interaction with membrane phosphatidylinositol 4,5-bisphosphateJOURNAL OF NEUROCHEMISTRY, Issue 1 2007Jan Benedikt Abstract Ethanol has opposite effects on two members of the transient receptor potential (TRP) family of ion channels: it inhibits the cold-menthol receptor TRPM8, whereas it potentiates the activity of the heat- and capsaicin-gated vanilloid receptor TRPV1. Both thermosensitive cation channels are critically regulated by the membrane lipid, phosphatidylinositol 4,5-bisphosphate (PIP2). The effects of this phospholipid on TRPM8 and TRPV1 are also functionally opposite: PIP2 is necessary for the activation of TRPM8 but it constitutively inhibits TRPV1. This parallel led us to investigate the possible role of PIP2 in the ethanol-induced modulation of rat TRPM8, heterologously expressed in HEK293T cells. In this study, we characterize the effects of ethanol (0.1,10%) on whole-cell currents produced by menthol and by low temperature (< 17°C). We show that the inclusion of PIP2 in the intracellular solution results in a strong reduction in the ethanol-induced inhibition of menthol-evoked responses. Conversely, intracellular dialysis with anti-PIP2 antibody or with the PIP2 scavenger, poly l -lysine, enhanced the ethanol-induced inhibition of TRPM8. A 20 min pre-incubation with wortmannin caused a modest decrease in inhibition produced by 1% ethanol, indicating that the ethanol-induced inhibition is not mediated by lipid kinases. These findings suggest that ethanol inhibits TRPM8 by weakening the PIP2,TRPM8 channel interaction; a similar mechanism may contribute to the ethanol-mediated modulation of some other PIP2 -sensitive TRP channels. [source] Allometric analysis reveals relatively little variation in nitrogen versus biomass accrual in four plant species exposed to varying light, nutrients, water and CO2PLANT CELL & ENVIRONMENT, Issue 10 2007CARL J. BERNACCHI ABSTRACT Nitrogen concentrations in plant tissues can vary as a function of resource availability. Altered rates of plant growth and development under varying resource availabilities were examined to determine their effects on changes in whole-plant N use efficiency (NUE). Three species of old-field annuals were grown at broadly varying light, nutrient and water levels, and four species at varying atmospheric concentrations of CO2. Study results show highly variable N accrual rates when expressed as a function of plant age or size, but similar patterns of whole-plant N versus non-N biomass accrual over a wide range of environmental conditions. However, severely light-limited plants showed increased N versus biomass accrual for two of three species, and severely nutrient-limited plants had decreased N versus biomass accrual for all species. Whole-plant N accrual versus age and N versus biomass accrual increased under saturating water for two of three species. A marginally significant, modest decrease in N versus biomass accrual was found at high CO2 levels for two of four species. Physiological adjustments in NUE, expressed as N versus biomass accrual, were limited to environments with severely limited or overabundant resources. [source] A Method for Evaluating Outcomes of Restoration When No Reference Sites ExistRESTORATION ECOLOGY, Issue 1 2009J. Stephen Brewer Abstract Ecological restoration typically seeks to shift species composition toward that of existing reference sites. Yet, comparing the assemblages in restored and reference habitats assumes that similarity to the reference habitat is the optimal outcome of restoration and does not provide a perspective on regionally rare off-site species. When no such reference assemblages of species exist, an accurate assessment of the habitat affinities of species is crucial. We present a method for using a species by habitat data matrix generated by biodiversity surveys to evaluate community responses to habitat restoration treatments. Habitats within the region are rated on their community similarity to a hypothetical restored habitat, other habitats of conservation concern, and disturbed habitats. Similarity scores are reinserted into the species by habitat matrix to produce indicator (I) scores for each species in relation to these habitats. We apply this procedure to an open woodland restoration project in north Mississippi (U.S.A.) by evaluating initial plant community responses to restoration. Results showed a substantial increase in open woodland indicators, a modest decrease in generalists historically restricted to floodplain forests, and no significant change in disturbance indicators as a group. These responses can be interpreted as a desirable outcome, regardless of whether species composition approaches that of reference sites. The broader value of this approach is that it provides a flexible and objective means of predicting and evaluating the outcome of restoration projects involving any group of species in any region, provided there is a biodiversity database that includes habitat and location information. [source] Effect of long-term belimumab treatment on b cells in systemic lupus erythematosus: Extension of a phase II, double-blind, placebo-controlled, dose-ranging study,ARTHRITIS & RHEUMATISM, Issue 1 2010Annett M. Jacobi Objective To understand the effects of long-term BLyS inhibition in human systemic lupus erythematosus (SLE). Methods Seventeen patients with SLE who were enrolled in a clinical trial of belimumab, a BLyS-specific inhibitor, plus standard of care therapy were studied. Phenotypic analysis of lymphocytes was performed using flow cytometry. Circulating antibody-secreting cells were enumerated using enzyme-linked immunospot assay. Serum was analyzed by enzyme-linked immunosorbent assay using an antibody that recognizes products of the VH4,34 gene. Lymphocyte counts, Ig levels, and anti,double-stranded DNA antibody levels were available as part of the clinical trial analyses. Results Samples were collected on days 0, 84, 168, 365, and 532 and after day 730. The total number of B cells started to decrease from baseline between days 84 and 168. This was due to a decrease in naive and transitional B cells. CD27+IgD+ memory B cells and plasmablasts decreased only after 532 days, whereas CD27+IgD, memory B cells were not affected, and there were no changes in T cells. Serum IgM levels began to decline between days 84 and 168, but there were no changes in serum levels of IgG, IgG anti-DNA antibodies, or VH4,34 antibodies during the study. SLE patients had more IgM-, IgG-, and autoantibody-producing B cells than did normal controls on day 0. There was only a modest decrease in the frequency of total IgM-producing, but not IgG-producing, cells on days 365 and 532, consistent with the phenotypic and serologic data. Conclusion Our data confirm the dependence of newly formed B cells on BLyS for survival in humans. In contrast, memory B cells and plasma cells are less susceptible to selective BLyS inhibition. [source] Proarrhythmic potential of halofantrine, terfenadine and clofilium in a modified in vivo model of torsade de pointesBRITISH JOURNAL OF PHARMACOLOGY, Issue 4 2002Andrew J Batey This study was designed to compare the proarrhythmic activity of the antimalarial drug, halofantrine and the antihistamine, terfenadine, with that of clofilium a K+ channel blocking drug that can induce torsade de pointes. Experiments were performed in pentobarbitone-anaesthetized, open-chest rabbits. Each rabbit received intermittent, rising dose i.v. infusions of the ,-adrenoceptor agonist phenylephrine. During these infusions rabbits also received increasing i.v. doses of clofilium (20, 60 and 200 nmol kg,1 min,1), terfenadine (75, 250 and 750 nmol kg,1 min,1), halofantrine (6, 20 and 60 ,mol kg,1) or vehicle. Clofilium and halofantrine caused dose-dependent increases in the rate-corrected QT interval (QTc), whereas terfenadine prolonged PR and QRS intervals rather than prolonging cardiac repolarization. Progressive bradycardia occurred in all groups. After administration of the highest dose of each drug halofantrine caused a modest decrease in blood pressure, but terfenadine had profound hypotensive effects resulting in death of most rabbits. The total number of ventricular premature beats was highest in the clofilium group. Torsade de pointes occurred in 6 out of 8 clofilium-treated rabbits and 4 out of 6 of those which received halofantrine, but was not seen in any of the seven terfenadine-treated rabbits. These results show that, like clofilium, halofantrine can cause torsade de pointes in a modified anaesthetized rabbit model whereas the primary adverse effect of terfenadine was cardiac contractile failure. A. Tarantili Abstract The effect of accelerated aging on the structure and properties of single, metalized, and multilayer films used in food packaging was studied through the exposure of specimens of those films to repeated aging cycles in a weather meter under the combined action of ultraviolet, humidity, and heat. The aged specimens were tested for their mechanical properties and water vapor transmission characteristics, and the results were compared to those obtained from the original specimens. The property changes introduced into the films by aging were further explored by attenuated total reflectance spectroscopy and differential scanning calorimetry in an attempt to correlate the changes in the properties with structural characteristics. The results showed that the films made of polypropylene (PP) underwent severe chain scission upon irradiation and lost mechanical properties but still retained their impermeability to water vapor. The metallic coating could not prevent PP from degrading, as it seemed to oxidize under the aging conditions. Therefore, the metalized film showed the same mechanical response as PP, but its water impermeability dropped dramatically. Polyethylene (PE) and poly(ethylene terephthalate) (PET) films showed modest decreases in their mechanical properties, which could be attributed to crosslinking reactions taking place with PE and to the increased ultraviolet stability of PET, respectively. On the other hand, the multilayer films presented a decrease in their mechanical properties according to those of their weak component, which would be expected for a composite structure. © 2008 Wiley Periodicals, Inc.
