Inheritance of resistance of wheat to eyespot at the adult stage

Abstract Moderate resistance to eyespot was first incorporated in the variety Cappelle-Desprez (CD). Later the gene Pch1, which could confer a higher level of resistance, was introduced from Aegilops ventricosa. However Pch1 -carrying varieties can sustain significant eyespot-induced yield losses in severe attack situations. A strategy to further enhance the resistance of wheat is by pyramiding Pch1 and the genes for resistance in CD. The first requirement to achieve this is a better understanding of the genetics of resistance in CD. The resistance of the 21 Cappelle-Desprez (Bezostaya) disomic substitution lines was evaluated. Chromosome 7A was confirmed as carrying a major gene for resistance to eyespot at the seedling stage. However, this study demonstrates that this chromosome has no effects at the adult stage. Chromosome 5A was shown to carry a major gene for resistance to eyespot at the adult stage, which was stably expressed each year of testing. Chromosomes 1A and 2B had significant effects for only two years among four. [source]


Effect of leg exercises on popliteal venous blood flow during prolonged immobility of seated subjects: implications for prevention of travel-related deep vein thrombosis

Summary.,Background:,Venous stasis is an important contributing factor in the development of travel-related deep vein thrombosis. This study examined factors affecting popliteal venous blood flow in order to determine the most effective exercise regimen to prevent venous stasis. Methods:,Twenty-one healthy subjects were randomly assigned to various activities over a 9-week period. Subjects remained seated throughout the investigation and 3660 duplex ultrasound examinations were performed by a single examiner using a SonoSite 180 Plus handheld ultrasound. Baseline popliteal vein blood flow velocity, cross-sectional area and volume flow in subjects sitting motionless were assessed in the first 3 weeks. The remaining 6 weeks involved subjects performing airline-recommended activities, foot exercises, foot exercises against moderate resistance and foot exercises against increased resistance in order to determine the most beneficial method for enhancing popliteal venous flow. Sitting with feet not touching the floor and the effect of sleeping were also assessed. Results:,The median age of the subjects was 22 years (range: 18,25.5 years), height 171 cm (162.5,180.5 cm) and body mass index 25.3 kg m,2 (23.2,26.3 kg m,2). Blood volume flow in the popliteal vein was reduced by almost 40% with immobility of seated subjects and by almost 2-fold when sitting motionless with feet not touching the floor. Foot exercises against increased resistance positively enhanced volume flow (P < 0.0001). Conclusion:,Leg exercise regimens enhanced popliteal venous flow during prolonged immobility of seated subjects, reinforcing the importance of regular leg movement to prevent venous stasis during prolonged sitting, such as in long-distance travel. Genome scan in the mosquito Aedes rusticus: population structure and detection of positive selection after insecticide treatment

Abstract Identification of genes involved in local adaptation is particularly challenging for species functioning as a network of interconnected populations undergoing frequent extinctions,recolonizations, because populations are submitted to contrasted evolutionary pressures. Using amplified fragment length polymorphism markers, population genetic structure of the mosquito Aedes rusticus was analysed in five geographical areas of the French Rhône-Alpes region. We included a number of sites that were treated with the bio-insecticide Bacillus thuringiensis israelensis (Bti) for more than 15 years. Analysis of molecular variance revealed that most of the genetic variability was found within populations (96%), with no significant variation among geographical areas, although variation among populations within areas (4%) was significant. The global genetic differentiation index FST was low (0.0366 ± 0.167). However, pairwise FST values were significant and no isolation-by-distance at the regional level was observed, suggesting a metapopulation structure in this species. Bti -treatment had no effect on genetic structure and on within-population genetic diversity. Potential signatures of positive selection associated with Bti -treatment were detected for five loci, even though toxicological bioassays performed on field-collected larvae showed no significant difference in mortality between Bti -treated and nontreated sites. The difficulty of detecting moderate resistance in field-collected larvae together with possible differential persistence of toxins in the environment may explain our inability to detect a toxicological response to Bti in treated sites. The evidence for positive selection occurring at several genomic regions suggests a first step towards Bti resistance in natural mosquito populations treated with this bio-insecticide. Furthermore, this signal was detectable using genomic tools before any toxicological evidence for resistance could be identified. [source]


Resistance of Pakistani field populations of spotted bollworm Earias vittella (Lepidoptera: Noctuidae) to pyrethroid, organophosphorus and new chemical insecticides

Abstract BACKGROUND: The spotted bollworm Earias vittella (Fab.) is a serious pest of cotton and okra in Pakistan. Owing to persistent use of insecticides, this pest has developed resistance, especially to pyrethroids. The present studies aimed at determining the extent of resistance to pyrethroid, organophosphorus and new chemical insecticides in Pakistani populations of E. vittella. RESULTS: Field populations of E. vittella were monitored at Multan, Pakistan, from 1999 to 2007 for their resistance against six pyrethroid, four organophosphorus and six new chemical insecticides using a leaf-dip bioassay. Of the pyrethroids, resistance was generally low to zeta-cypermethrin and moderate to high or very high to cypermethrin, deltamethrin, esfenvalerate, bifenthrin and lambda-cyhalothrin. Resistance to organophosphates chlorpyrifos, profenofos, triazophos and phoxim was recorded at very low to low levels. Among new chemicals, E. vittella had no or a very low resistance to spinosad, emamectin benzoate and methoxyfenozide, a very low to low resistance to abamectin, a very low to moderate resistance to indoxacarb and a moderate resistance to chlorfenapyr. CONCLUSION: The results indicate a lack of cross-resistance between pyrethroid and organophosphorus insecticides in E. vittella. Rotation of insecticides showing no, very low or low resistance, but belonging to different insecticide classes with unrelated modes of action, may prevent or mitigate insecticide resistance in E. vittella. Lack of cross-resistance to indoxacarb in insecticide-resistant Spodoptera frugiperda (Lepidoptera: Noctuidae) and Plutella xylostella (Lepidoptera: Yponomeutidae)

Abstract Two field strains of the fall armyworm, Spodoptera frugiperda (JE Smith), collected from corn in north Florida showed high resistance to carbaryl (626- and 1159-fold) and moderate resistance to parathion-methyl (30- and 39-fold) as compared with a laboratory susceptible strain. A field strain of the diamondback moth, Plutella xylostella (L.) collected from cabbage in north Florida and selected for 20 generations with permethrin showed high resistance to permethrin (987-fold) as compared with a susceptible strain. However, in all instances, no cross-resistance to indoxacarb, a novel oxidiazine insecticide, was observed in these two species. Biochemical studies revealed that, in S. frugiperda, activities of detoxification enzymes (microsomal oxidase, glutathione S -transferase and general esterase) were significantly higher in the field strains than in the susceptible strain, indicating that these detoxification enzymes were not actively involved in the resistance to indoxacarb. The lack of cross-resistance between indoxacarb and permethrin in P. xylostella further supports the notion that the mode of action of these insecticides on the insect sodium channel is different. Cotton whitefly (Bemisia tabaci) resistance to organophosphate and pyrethroid insecticides in Pakistan

Abstract Resistance to three organophosphate and four pyrethroid insecticides was monitored from 1992 to 2000 in field populations of adult whiteflies, Bemisia tabaci, from Pakistan using a leaf-dip method. There was generally a very high resistance to dimethoate and deltamethrin, and a moderate resistance to monocrotophos during 1992 to 1996. From 1997 to 2000, resistance to these insecticides dropped to low levels because of less reliance on them for whitefly control, and introduction of new chemistries with novel modes of action that had no cross-resistance to conventional insecticides. Concurrently, whitefly resistance to acephate, fenpropathrin, lambda-cyhalothrin and bifenthrin mostly remained low. An insecticide resistance management strategy is recommended that particularly emphasizes the rotation of still-effective insecticides from different chemical classes along with the use of novel chemicals and other tactics of integrated pest management. © 2001 Society of Chemical Industry [source]
