Selected AbstractsGabapentin Increases Slow-wave Sleep in Normal AdultsEPILEPSIA, Issue 12 2002Nancy Foldvary-Schaefer Summary: ,Purpose: The older antiepileptic drugs (AEDs) have a variety of effects on sleep, including marked reduction in rapid-eye-movement (REM) sleep, slow-wave sleep (SWS), and sleep latency, and an increase in light sleep. The effects of the newer AEDs on sleep are unknown. Our purpose was to study the effect of gabapentin (GBP) on sleep. Methods: Ten healthy adults and nine controls were the subjects of this study. All underwent baseline and follow-up polysomnography (PSG) and completed sleep questionnaires. After baseline, the treated group received GBP titrated to 1,800 mg daily. Polygraphic variables and Epworth Sleepiness Scale (ESS) scores, a subjective measure of sleep propensity, were compared by using the Wilcoxon signed rank test. Results: Nine of the treated subjects achieved the target dose; one was studied with 1,500 mg daily because of dizziness experienced at the higher dose. GBP-treated subjects had an increase in SWS compared with baseline. No difference in the ESS or other polygraphic variables was observed. However, a minor reduction in arousals, awakenings, and stage shifts was observed in treated subjects. Conclusions: GBP appears to be less disruptive to sleep than are some of the older AEDs. These findings may underlie the drug's therapeutic effect in the treatment of disorders associated with sleep disruption. [source] LEARNING ABILITY AND LONGEVITY: A SYMMETRICAL EVOLUTIONARY TRADE-OFF IN DROSOPHILAEVOLUTION, Issue 6 2008Joep M. S. Burger Learning ability can be substantially improved by artificial selection in animals ranging from Drosophila to rats. Thus these species have not used their evolutionary potential with respect to learning ability, despite intuitively expected and experimentally demonstrated adaptive advantages of learning. This suggests that learning is costly, but this notion has rarely been tested. Here we report correlated responses of life-history traits to selection for improved learning in Drosophila melanogaster. Replicate populations selected for improved learning lived on average 15% shorter than the corresponding unselected control populations. They also showed a minor reduction in fecundity late in life and possibly a minor increase in dry adult mass. Selection for improved learning had no effect on egg-to-adult viability, development rate, or desiccation resistance. Because shortened longevity was the strongest correlated response to selection for improved learning, we also measured learning ability in another set of replicate populations that had been selected for extended longevity. In a classical olfactory conditioning assay, these long-lived flies showed an almost 40% reduction in learning ability early in life. This effect disappeared with age. Our results suggest a symmetrical evolutionary trade-off between learning ability and longevity in Drosophila. [source] Temporary amelioration of bilirubin conjugation defect in Gunn rats by transplanting conditionally immortalized hepatocytesJOURNAL OF GASTROENTEROLOGY AND HEPATOLOGY, Issue 6 2002BYUNG-HO KIM Abstract Background: Conditionally immortalized hepatocytes (CIH) have been used in hepatocyte transplantation as an alternative to primary hepatocytes to cope with the shortage of donor organs. However, CIH are known to undergo apoptosis at body temperature and survive in vivo for a short period. In the present study, we investigated whether CIH function or not and how long their function is maintained in vivo. Methods: Various CIH cell lines that were established with temperature-sensitive Simian virus 40 large T antigen were transplanted into the spleen of Gunn rats, which are defective in bilirubin uridine diphosphate glucuronoside transferase (BUGT). Then, we measured biological changes over 3 months. Results: Serum bilirubin of the syngeneic CIH recipients decreased by 30%, which was maintained for 8 weeks. Thereafter, it began to rise to basal levels. The recipients of allogeneic CIH showed a minor reduction of bilirubin, although this was not statistically significant. However, there was no significant change in the bilirubin level in recipients of BUGT-defective congeneic CIH throughout the study period. Bilirubin monoglucuronides in the bile were not detected in the recipients of BUGT-defective CIH. However, they appeared in recipients of non-defective CIH and made up approximately 41% of total bile pigments. Conclusions: Conditionally immortalized hepatocytes expressed hepatocyte function in vivo as well as in vitro, but the function lasted for a couple of months. According to our previous study, the limited functional duration may be related to the inevitable occurrence of apoptosis of these cells at body temperature. These data suggest that CIH can be used in hepatocyte transplantation only for temporary hepatic support. © 2002 Blackwell Publishing Asia Pty Ltd [source] The Action of a Novel Fluoroquinolone Antibiotic Agent Antofloxacin Hydrochloride on Human-Ether- à-go-go- Related Gene Potassium ChannelBASIC AND CLINICAL PHARMACOLOGY & TOXICOLOGY, Issue 2 2010Jia Guo In this study, the effects of a novel fluoroquinolone, antofloxacin hydrochloride (AX) on human- ether-à-go-go -related gene (HERG) encoding potassium channels and the biophysical mechanisms of drug action were performed with whole-cell patch-clamp technique in transiently transfected HEK293 cells. The administration of AX caused voltage- and time-dependent inhibition of HERG K+ current (IHERG/MiRP1) in a concentration-dependent manner but did not markedly modify the properties of channel kinetics, including activation, inactivation, deactivation and recovery from inactivation as well. In comparison with sparfloxacin (SPX), levofloxacin lactate (LVFX), the potency of AX to inhibit HERG tail currents was the least one, with an IC50 value of 460.37 ,M. By contrast, SPX was the most potent compound, displaying an IC50 value of 2.69 ,M whereas LVFX showed modest potency, with an IC50 value of 43.86 ,M, respectively. Taken together, our data suggest that AX only causes a minor reduction of IHERG/MiRP1 at the estimated free plasma level. [source] DAM-INDUCED MODIFICATIONS TO UPPER ALLEGHENY RIVER STREAMFLOW PATTERNS AND THEIR BIODIVERSITY IMPLICATIONS,JOURNAL OF THE AMERICAN WATER RESOURCES ASSOCIATION, Issue 1 2002C. Mark Cowell ABSTRACT: This study evaluates the streamflow characteristics of the upper Allegheny River during the periods preceding (1936 to 1965) and following (1966 to 1997) completion of the Kinzua Dam in northwestern Pennsylvania. Inter-period trends in seasonal patterns of discharge and peak flow at three downstream sites are compared to those at two upstream sites to determine the influence of this large dam on surface water hydrology. Climatic records indicate that significant changes in annual total and seasonal precipitation occurred over the twentieth century. Increased runoff during the late summer through early winter led to increased discharge both upstream and downstream during these months, while slightly less early-year rainfall produced minor reductions in spring flood peaks since 1966. The Kinzua Dam significantly enhanced these trends downstream, creating large reductions in peak flow, while greatly augmenting low flow during the growing season. This reduction in streamflow variability, coupled with other dam-induced changes, has important biodiversity implications. The downstream riparian zone contains numerous threatened/endangered species, many of which are sensitive to the type of habitat modifications produced by the dam. Flood dynamics under the current post-dam conditions are likely to compound the difficulties of maintaining their long-term viability. [source] |
