Catalytic Hydrogenation of Cyanohydrin Esters as a Novel Approach to N -Acylated ,-Amino Alcohols , Reaction Optimisation by a Design of Experiment Approach

Abstract The catalytic hydrogenation of acylated cyanohydrins and subsequent intramolecular migration of the acyl group to yield pharmaceutically interesting N -acyl ,-amino alcohols is shown to be a successful one-pot preparation method. The combination of a multistep DoE approach and high-throughput methodology proved to be an effective strategy for the optimisation of the reaction. With the favoured catalyst/solvent combination of nickel on alumina in dioxane, both hydrogenation and acyl group migration proceeded smoothly, giving the N -acyl ,-amino alcohols in yields (determined by GC) of up to 90,% for aliphatic substrates and up to 50,% for benzylic ones, the latter being more prone to side reactions. No racemisation was found to occur at the chiral centre of an aliphatic molecule when an enantiopure cyanohydrin ester was used, though a minor decrease in ee was observed with a benzylic substrate. Domain V of m-calpain shows the potential to form an oblique-orientated ,-helix, which may modulate the enzyme's activity via interactions with anionic lipid

The activity of m-calpain, a heterodimeric, Ca2+ -dependent cysteine protease appears to be modulated by membrane interactions involving oblique-orientated ,-helix formation by a segment, GTAMRILGGVI, in the protein's smaller subunit. Here, graphical and hydrophobic moment-based analyses predicted that this segment may form an ,-helix with strong structural resemblance to the influenza virus peptide, HA2, a known oblique-orientated ,-helix former. Fourier transform infrared spectroscopy showed that a peptide homologue of the GTAMRILGGVI segment, VP1, adopted low levels of ,-helical structure (, 20%) in the presence of zwitterionic lipid and induced a minor decrease (3 °C) in the gel to liquid-crystalline phase transition temperature, TC, of the hydrocarbon chains of zwitterionic membranes, suggesting interaction with the lipid headgroup region. In contrast, VP1 adopted high levels of ,-helical structure (65%) in the presence of anionic lipid, induced a large increase (10 °C) in the TC of anionic membranes, and showed high levels of anionic lipid monolayer penetration (,SP = 5.5 mN·m,1), suggesting deep levels of membrane penetration. VP1 showed strong haemolytic ability (LD50 = 1.45 mm), but in the presence of ionic agents, this ability, and that of VP1 to penetrate anionic lipid monolayers, was greatly reduced. In combination, our results suggest that m-calpain domain V may penetrate membranes via the adoption of an oblique-orientated ,-helix and electrostatic interactions. We speculate that these interactions may involve snorkelling by an arginine residue located in the polar face of this ,-helix. [source]


Trends in self-reported sleep duration and insomnia-related symptoms in Finland from 1972 to 2005: a comparative review and re-analysis of Finnish population samples

Summary A hypothesis concerning habitual sleep reduction and its adverse consequences among general population in modern societies has received wide publicity in the mass media, although scientific evidence supporting the hypothesis is scarce. Similarly, there is an extensively distributed belief, at least in Finland, that the prevalence of insomnia-related symptoms is increasing, but evidence for this is even sparser. These issues are important because of the known increased risk of mortality and health risks associated with sleep duration deviating from 7 to 8 h. To reveal possible trends in self-reported sleep duration and insomnia-related symptoms, we reanalyzed all available data from surveys carried out in Finland from 1972 to 2005. The main results were that a minor decrease of self-reported sleep duration has taken place in Finland, especially among working aged men. However, the size of the reduction (about 4%) was relatively small, approximately 5.5 min per each 10 years during the 33 years' time interval under study. The proportion of 7 h sleepers has increased and, correspondingly, the proportion of 8 h sleepers has decreased, but the extreme ends of the sleep duration distribution remained unchanged. Tentative evidence suggesting an increase in insomnia-related symptoms among working aged population during the last 10 years was found. In conclusion, the Finnish data during the past 33 years indicate a general decrease in self-reported sleep duration of about 18 min and an increase of sleep complaints, especially among the employed middle-aged population. Does dopexamine influence regional vascular tone and oxygenation during intestinal hypotension?

Background: Local effects of dopexamine on intestinal vascular tone and oxygenation were investigated during intestinal hypotension. To this end, we employed an experimental model, in which the superior mesenteric arterial pressure (PSMA) was controlled by an adjustable perivascular clamp. This approach enabled us to keep the intestinal perfusion pressure (IPP) constant in the face of any systemic circulatory alterations. Methods: In 11 barbiturate-anesthetized pigs, we instrumented the superior mesenteric circulation for assessments of vascular resistance (RMES), IPP, jejunal mucosal perfusion (Laser Doppler) and intestinal tissue oxygenation (microoximetry). Measurements were carried out before and during dopexamine infusions (0.5 and 1.0 µg·kg,1·min,1) at a freely variable PSMA (i.e. the perivascular clamp fully open) and at a PSMA of 50 mmHg and 30 mmHg. Results: At a constant PSMA of 50 mmHg, dopexamine had no significant intestinal vascular effects. However, at a constant PSMA of 30 mmHg, both doses of dopexamine were associated with decreases in RMES. Effects of dopexamine on intestinal oxygen delivery and extraction were minimal during these procedures, while a minor decrease in intestinal tissue oxygen tension was observed during dopexamine administration at the lowest IPP level. Conclusion: At very low intestinal perfusion pressures (approximately 30 mmHg) dopexamine produces intestinal vasodilation in excess of what is produced by intrinsic autoregulation. This suggests that there is a vasodilatory reserve in the intestine under such conditions and that a pharmacological vasodilator like dopexamine may improve intestinal circulation during regional severe hypotension. Long-term stability of dye-sensitised solar cells

Accelerated ageing tests on large numbers of nanocrystalline dye-sensitised solar cells (nc-DSC) show that, to first order, separation between the effects of the stresses of visible light soaking, UV illumination and thermal treatment on long-term stability is possible. The corresponding mechanisms are electrochemical, photochemical and purely chemical in nature. It was found that visible light soaking alone is not a dominant stress factor. A dramatic improvement in UV stability has been achieved by using MgI2 as additive to the electrolyte. Thermal stress appears to be one of the most critical factors determining the long-term stability of nc-DSC and is strongly related to the chemical composition of electrolyte solvents and additives. Encouraging stability results have been obtained for cells based on pure nitrile-based solvents: (1) A minor decrease in performance of initially 5.5% solar efficient cells has been found after 2000 h at 60°C without light soaking; (2) After 900 h ageing at 85°C, a decrease of 30% in maximum power has been observed; (3) After 3400 h of combined thermal stress and continuous light soaking (45°C, 1 sun equivalent) good stability with 15% decrease in maximum power can be demonstrated. WATER QUALITY MODELING OF ALTERNATIVE AGRICULTURAL SCENARIOS IN THE U.S. CORN BELT,

ABSTRACT: Simulated water quality resulting from three alternative future land-use scenarios for two agricultural watersheds in central Iowa was compared to water quality under current and historic land use/land cover to explore both the potential water quality impact of perpetuating current trends and potential benefits of major changes in agricultural practices in the U.S. Corn Belt. The Soil Water Assessment Tool (SWAT) was applied to evaluate the effect of management practices on surface water discharge and annual loads of sediment and nitrate in these watersheds. The agricultural practices comprising Scenario 1, which assumes perpetuation of current trends (conversion to conservation tillage, increase in farm size and land in production, use of currently-employed Best Management Practices (BMPs)) result in simulated increased export of nitrate and decreased export of sediment relative to the present. However, simulations indicate that the substantial changes in agricultural practices envisioned in Scenarios 2 and 3 (conversion to conservation tillage, strip intercropping, rotational grazing, conservation set-asides and greatly extended use of best management practices (BMPs) such as riparian buffers, engineered wetlands, grassed waterways, filter strips and field borders) could potentially reduce current loadings of sediment by 37 to 67 percent and nutrients by 54 to 75 percent. Results from the study indicate that major improvements in water quality in these agricultural watersheds could be achieved if such environmentally-targeted agricultural practices were employed. Traditional approaches to water quality improvement through application of traditional BMPs will result in little or no change in nutrient export and minor decreases in sediment export from Corn Belt watersheds.
