Scatà Summary The objective of this work was to characterize the complete coding region of the ovine acylCoA:diacylglycerol acyltransferase 1 (DGAT1) gene of three Italian sheep breeds: Sarda, Altamurana and Gentile di Puglia. Characterization was accomplished by direct sequencing of 8676 bp of the relevant DNA, including introns and partial 5, and 3, untranslated regions (UTRs). We detected five novel SNPs; one SNP (g.5553C>T) is located in intron 2, has similar frequencies in the three breeds and showed a negative association with milk fat content. More interesting is an SNP in the 5, UTR (g.127C>A), the occurrence of which is rare in the higher milk-fat breeds (Altamurana and Gentile di Puglia); it is located in the core sequence of Sp1, a putative binding site of transcription factors. This SNP showed a significant negative association with milk fat content in the Sarda sheep. Because DGAT1 plays a fundamental role in triacylglycerol synthesis, the novel detected SNP in the 5, UTR of the DGAT1 gene might explain, at least partially, the variation of fat content in the milk of Sarda sheep. Calvo Summary On the basis of comparisons between cattle and sheep genome mapping information the ovine , - amylase gene was examined as a possible genetic marker for milk traits in sheep. The objective of the present study was to isolate, map and determine whether this gene is a candidate gene for milk traits. DNA fragments (832 and 2360 bp) corresponding to two different AMY genes were isolated, and one SNP in intron 3 and one GTG deletion in exon 3 of the 2360 bp DNA fragment were found. The 2360 bp ovine AMY DNA fragment was located on chromosome 1 by linkage mapping using the International Mapping Flock. No association was found between estimated breeding values for milk yield, protein and fat contents and AMY genotypes in a daughter design comprising 13 Manchega families with an average of 29 daughters (12,62) per sire. [source] Relationship between milk cortisol concentration and the behavioral characteristics of postpartum cows introduced to a new groupANIMAL SCIENCE JOURNAL, Issue 5 2010Michiru FUKASAWA ABSTRACT The objective of this study was to examine the relationship between milk cortisol concentration (MC) and behavioral responses to stress. As a stress response, the behaviors of 37 postpartum cows were observed when they were introduced into a new group. Milk samples for MC measurement and milk trait data were collected on the test day every 2 months after calving. The best linear unbiased prediction (BLUP) was estimated from each behavioral and milk trait data. Behavioral BLUP data were subjected to principal component (PC) analysis and three PCs were extracted. The first PC, with large loadings for eating and allogrooming, was positively correlated with milk yield. Therefore, individual differences in behaviors closely related with PC1 would have been unconsciously paid a lot of attention in terms of productivity. However, the second PC, with large loadings for drinking and self-grooming, uncorrelated with any milk traits, had a negative correlation with MC. The third PC, with large loadings for exploring, fighting and the frequency of agonistic behavior, was uncorrelated with all traits. Both PC2 and PC3 were independent of productivity, and individual differences in behaviors related with these PCs may be scarcely considered. [source] |
