Selected AbstractsMeasured concentrations of combustion gases from the use of unvented gas fireplacesINDOOR AIR, Issue 5 2010P. W. Francisco Abstract, Measurements of combustion product concentrations were taken in 30 homes where unvented gas fireplaces were used. Measurements of CO, CO2, NOx, NO2, O2 (depletion), and water vapor were taken at 1-min interval. The analyzers were calibrated with certified calibration gases for each placement and were in operation for 3,4 days at each home. Measured concentrations were compared to published health-based standards and guidelines. The two combustion gases that exceeded published values were NO2 and CO. For NO2, the Health Canada guideline of 250 ppb (1-h average) was exceeded in about 43% of the sample and the World Health Organization (WHO) guideline of 110 ppb (1-h average) was exceeded in 80% of the sample. Carbon monoxide levels exceeded the U.S. EPA 8-h average standard of 9 ppm in 20% of the sample. Moisture problems were not evident in the test homes. An analysis of the distribution of CO showed that the CO is dispersed throughout the home almost immediately upon operation of the fireplace and that the concentrations throughout the home away from the immediate vicinity of the fireplace are 70,80% of the level near the fireplace. Decay analysis of the combustion gases showed that NO was similarly stable to CO and CO2 in the indoor environment but that both NO2 and water vapor were removed from the air at much greater rates. Practical Implications Previous studies on unvented gas fireplaces have made assumptions of how they are operated by users. This article presents the results of field monitoring of 30 unvented gas fireplaces under normal operation, regardless of whether users follow industry recommendations regarding installation, usage patterns, and maintenance. The monitoring found that health-based standards and guidelines were exceeded for CO in 20% of homes and for NO2 in most homes. There were no identified moisture problems in these homes. Nearly, half of the fireplaces were used at least once for longer than 2 h, counter to manufacturers' intended usage as supplemental heating. This demonstrates that given actual usage patterns and compared to current health-based thresholds, these appliances can produce indoor air concentrations considered to be unhealthy to at least sensitive or at-risk individuals. [source] Consequences of hyperconcentrated flow for process-based soil erosion modelling on the Chinese Loess PlateauEARTH SURFACE PROCESSES AND LANDFORMS, Issue 9 2006Rudi Hessel Abstract High sediment concentrations in runoff are a characteristic feature of the Chinese Loess Plateau, and are probably caused by factors such as the occurrence of erodible materials on steep slopes, the characteristics of the loess and the harsh climate that results in low plant cover. When sediment concentration increases, fluid density increases, viscosity increases and settling velocity decreases. These effects become increasingly important with increasing concentration and can result in flow behaviour that is quite different from that of clear water flow. Although the net effect of these changes on the flow is not always apparent, erosion models that deal with high sediment concentrations should consider such effects and could include corrections for some of these effects. A case study in a small catchment on the Loess Plateau indicated that sediment concentrations were considerable, and literature data suggested that for such sediment concentrations, corrections for settling velocity, fluid density and viscosity are needed. Furthermore, a number of corrections are necessary to be able to compare field measurements with results of soil erosion models: sediment volume should be subtracted from runoff volume and a density correction is needed to use data from a pressure transducer. For flumes that were used to measure discharge from smaller areas inside the catchment, the measured water level should be corrected by subtracting the sediment level in the flume from the water level, while the sediment volume should also be subtracted from the discharge. Finally, measured concentration should be corrected to give concentration expressed as grams per litre of clear water, since soil erosion models express sediment concentration in this way. [source] Multivariable real-time optimal control of a cooling and antisolvent semibatch crystallization processAICHE JOURNAL, Issue 10 2009Milana Trifkovic Abstract This article presents an experimental study of simultaneous optimization with respect to two variables (cooling rate and flow-rate of antisolvent) in an offline and online (real-time) manner on a semibatch crystallizer. The nucleation and growth kinetic parameters of paracetamol in an isopropanol-water cooling, antisolvent batch crystallizer were estimated by nonlinear regression in terms of the moments of the crystal population density. Moments of crystal population were estimated from the measured chord length distribution, generated by the FBRM®, and the supersaturation was calculated from the measured concentration by attenuated total reflectance-fourier transform infrared spectroscopy. The results of real-time optimization showed a substantial improvement of the end of batch properties (the volume-weighted mean size and yield). For the same objective function, the real-time optimization method resulted in an increase in the volume-weighted mean size by ,100 ,m and 15% of theoretical yield compared with the best result obtained in an offline optimization manner. © 2009 American Institute of Chemical Engineers AIChE J, 2009 [source] Accelerator mass spectrometry offers new opportunities for microdosing of peptide and protein pharmaceuticalsRAPID COMMUNICATIONS IN MASS SPECTROMETRY, Issue 10 2010Mehran Salehpour Accelerator Mass Spectrometry (AMS) is an ultra-sensitive analytical method which has been instrumental in developing microdosing as a strategic tool in early drug development. Considerable data is available for AMS microdosing using typical pharmaceutical drugs with a molecular weight of a few hundred Daltons. The so-called biopharmaceuticals such as proteins offer interesting possibilities as drug candidates; however, experimental data for protein microdosing and AMS is scarce. The analysis of proteins in conjunction with early drug development and microdosing is overviewed and three case studies are presented on the topic. In the first case study AMS experimental data is presented, for the measured concentration of orally administered recombinant insulin in the blood stream of laboratory rabbits. Case study 2 concerns minimum sample size requirements. AMS samples normally require about 1,mg of carbon (10,µL of blood) which makes AMS analysis unsuitable in some applications due to the limited availability of samples such as human biopsies or DNA from specific cells. Experimental results are presented where the sample size requirements have been reduced by about two orders of magnitude. The third case study concerns low concentration studies. It is generally accepted that protein pharmaceuticals may be potentially more hazardous than smaller molecules because of immunological reactions. Therefore, future first-in-man microdosing studies might require even lower exposure concentrations than is feasible today, in order to increase the safety margin. [source] Modelling sources and sinks of CO2, H2O and heat within a Siberian pine forest using three inverse methodsTHE QUARTERLY JOURNAL OF THE ROYAL METEOROLOGICAL SOCIETY, Issue 590 2003M. Siqueira Abstract Source/sink distributions of heat, CO2 and water vapour in a Siberian Scots pine forest were estimated from measured concentration and temperature profiles using three inverse analysis methods. These methods include: a Eulerian second-order closure model (EUL); a localized near-field Lagrangian dispersion model (LNF); and a hybrid model (HEL) which uses the Eulerian second-order turbulence model to calculate the flow statistics combined with the regression analysis used with the Lagrangian model. Model predictions were compared to heat flux profiles measured at five levels in the canopy, and to CO2 and water-vapour fluxes measured close to the ground and above the forest. Predictions of sensible-heat flux profiles by the LNF and HEL schemes were systematically better than results from the EUL analysis. This improvement was attributed to the redundancy in the measured profile (scalar concentration and temperature) data for LNF and HEL and to the imposed smoothness condition used in the regression analyses, whereas the EUL approach calculates a source for each level without any redundancy. The LNF and HEL schemes were also better than EUL in predicting source distributions for CO2 and water vapour, although errors were larger than for sensible heat. The main novelty in our study is the use of EUL to decompose the vertical variability in scalar (or heat) sources into variability produced by the inhomogeneity in flow statistics and variability inferred from the measured mean scalar concentration (or temperature) profile. Hence, it is possible with this analysis to assess how much ,new information' about the source variability is attributed to vertical variation in the measured mean scalar concentration (or temperature) profiles. The analysis shows that measured water vapour concentration profiles provide little information on the inferred source distribution, whereas the CO2 profiles contain more information. Monte Carlo simulations show that computed sources from all three inverse methods have similar sensitivities to errors in measured temperatures. Errors are reduced when the reference temperature above the canopy is held fixed, implying that errors in this temperature propagate throughout the entire domain. When information content and error estimations are combined, a valuable tool to assess the quality of source prediction by inverse methods can be generated. Copyright © 2003 Royal Meteorological Society [source] Determination of bevantolol enantiomers in human plasma by coupled achiral,chiral high performance-liquid chromatographyCHIRALITY, Issue 7 2007Joung Weon Oh Abstract A coupled achiral,chiral high performance liquid chromatographic method was developed and fully validated for the determination of bevantolol enantiomers, (,)-(S)-bevantolol and (+)-(R)-bevantolol, in human plasma. Plasma samples were prepared by solid phase extraction with Sep-Pak Plus C18 cartridges followed by HPLC. Bevantolol enantiomers and (+)-(R)-Propranolol as internal standard (IS) were preseparated from interfering components in plasma on a Phenomenex silica column and bevantolol enantiomers and IS were resolved and determined on a Chiralcel OJ-H chiral stationary phase. The two columns were connected by a switching valve equipped with silica precolumn. The Precolumn was used to concentrate bevantolol in the eluent from the achiral column before back flushing onto chiral phase. A detailed validation of the method was performed accordingly to FDA guidelines. For each enantiomer the assay was linear between 20 and 1600 ng/ml. The quantification limits of both bevantolol enantiomers were 20 ng/ml. The intraday variation was between 1.07 and 12.64% in relation to the measured concentration and the interday variation was 0.91 and 11.79%. The method has been applied to the determination of (,)-(S)- and (+)-(R)-bevantolol in plasma from healthy volunteers dosed with racemic bevantolol hydrochloride. hydrochloride. [source] Risk assessment of herbicide mixtures in a large European lakeENVIRONMENTAL TOXICOLOGY, Issue 2 2008Nathalie Chèvre Abstract Lake Geneva is one of the largest European lakes with a surface area of 580 km2. Its catchment area covers 7400 km2, of which ,20% is arable land. Monitoring campaigns have been carried out in 2004 and 2005 to determine the contamination of the lake by pesticides. The results highlight the widespread presence of herbicides in water, the measured concentrations for most substances remaining constant in 2004 and 2005. However, for some individual herbicides the concentrations increased drastically (e.g., the herbicide foramsulfuron). We assessed the environmental risk of the herbicides detected in the lake using water quality criteria recently determined for the Swiss environmental protection agency. Furthermore, we assessed the risk of herbicide mixtures, grouped based upon their mode of action. Generally, the risk estimated for all single substances is low, except for some sulfonylurea compounds. For these substances, the measured concentrations are higher than the predicted no-effect concentration. Impact on the flora of the lake can therefore not be excluded. When mixtures of pesticides with similar mode of action are taken into account, the risk remains lower than the mixture water quality criteria for all groups, but can reach as high as one third of this quality criteria. [source] Exposure and effects assessment of resident mink (Mustela vison) exposed to polychlorinated dibenzofurans and other dioxin-like compounds in the Tittabawassee River basin, Midland, Michigan, USA,ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 10 2008Matthew J. Zwiernik Abstract Historically, sediments and floodplain soils of the Tittabawassee River (TR; MI, USA) have been contaminated with polychlorinated dibenzofurans (PCDFs), polychlorinated dibenzo- p -dioxins (PCDDs), and polychlorinatedbiphenyls (PCBs). Median concentrations of 2,3,7,8-tetrachlorodibenzo- p -dioxin equivalents (TEQs) based on 2006 World Health Organization tetrachloro-dibenzo- p -dioxin toxic equivalency factors (TEFs) in the diet of mink (Mustela vison) ranged from 6.8 × 10,1 ng TEQ/kg wet weight upstream of the primary source of PCDF to 3.1 × 101 ng TEQ/kg wet weight downstream. Estimates of toxicity reference values (TRVs) derived from laboratory studies with individual PCDDs/PCDFs and PCB congeners or mixtures of those congeners, as well as application of TEFs, were compared to site-specific measures of mink exposure. Hazard quotients based on exposures expressed as concentrations of TEQs in the 95th percentile of the mink diet or liver and the no-observable-adverse-effect TRVs were determined to be 1.7 and 8.6, respectively. The resident mink survey, however, including number of mink present, morphological measures, sex ratios, population age structure, and gross and histological tissue examination, indicated no observable adverse effects. This resulted for multiple reasons: First, the exposure estimate was conservative, and second, the predominantly PCDF congener mixture present in the TR appeared to be less potent than predicted from TEQs based on dose,response comparisons. Given this, there appears to be great uncertainty in comparing the measured concentrations of TEQs at this site to TRVs derived from different congeners or congener mixtures. Based on the lack of negative outcomes for any measurement endpoints examined, including jaw lesions, a sentinel indicator of possible adverse effects, and direct measures of effects on individual mink and their population, it was concluded that current concentrations of PCDDs/PCDFs were not causing adverse effects on resident mink of the TR. [source] Effect of maternal exposure to tributyltin on reproduction of the pearl oyster (Pinctada fucata martensii)ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 5 2004Suguru Inoue Abstract We examined the effect of tributyltin (TBT) on reproduction of the pearl oyster (Pinctada fucata martensii). In a maternal exposure test, five female pearl oysters were exposed to TBT at measured concentrations of 0 (control), 0.092, or 0.191 ,g/L at 25°C for one week, and the embryo developmental success (the ratio of normal D-larvae to all larvae) was measured. The embryo developmental success was significantly decreased in the 0.191-,g/L treatment group (65.5%) compared to that in the control group (82.5%; p = 0.031). Concentrations of TBT in the ovary reached 0.088 ,g/g in the 0.191-,g/L treatment group. In a waterborne exposure test, inseminated eggs were exposed to TBT at measured concentrations of 0 (control), 0.020, 0.045, 0.091, 0.192, or 0.374 ,g/L for 24 h. The embryo developmental success also was significantly decreased in the 0.192-,g/L treatment group (78.3%; p = 0.020) and no development at all was observed in the 0.374-,g/L treatment group compared with that in the control group (95.4%). These results clearly demonstrate that TBT accumulating in the bodies of bivalves has the potential to inhibit reproduction. [source] Studies on the effect of soil aging on the toxicity of pyrene and phenanthrene to a soil-dwelling springtailENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 3 2002Line E. Sverdrup Abstract Soil samples spiked with five concentrations of pyrene and phenanthrene were aged for 0, 10, 40, and 120 d before toxicity was investigated using a standardized bioassay with the soil-dwelling collembolan Folsomia fimetaria L. Toxic effects were measured as reductions in survival and reproductive output after 3 weeks of exposure. Both pyrene and phenanthrene were degraded in the test system during storage, phenanthrene to a higher degree than pyrene. However, when toxic effects of the tested substances were calculated on the basis of measured concentrations of the parent compounds, toxicity was unaffected by storage for up to 120 d. Many studies have shown a negative correlation between aging and the biodegradation of polycyclic aromatic hydrocarbons (PAHs), indicating a reduction in the bioavailability of PAHs to micro-organisms with time. Our results indicate that a reduction in toxicity, as related to the measured concentrations of the parent compounds in the soil, may not always be expected for aged soils. Provided that metabolites of pyrene and phenanthrene did not significantly contribute to the toxicity in aged soil samples, a possible explanation for the absence of aging effects is the high test concentrations used in combination with the low content of organic carbon (1.6%) in the test soil. [source] Induction of morphological deformities in Chironomus tentans exposed to zinc- and lead-spiked sedimentsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 11 2001Edward A. Martinez Abstract Laboratory experiments were used to assess morphological responses of Chironomus tentans larvae exposed to three levels of zinc and lead. Chironomus tentans egg masses were placed into triplicate control and metal-spiked aquaria containing the measured concentrations 1,442, 3,383, and 5,562 ,g/g Pb dry weight and 1,723, 3,743, and 5,252 ,g/g Zn dry weight. Larvae were collected at 10-d intervals after egg masses were placed in aquaria until final emergence. Larvae were screened formouthpart deformities and metal body burdens. Deformities increased with time of exposure in both Zn and Pb tanks. Deformity rates between the three Zn concentrations differed statistically, with low and medium Zn levels containing the highest overall deformity rates of 12%. Deformity rates for larvae held in the Pb aquaria were found to differ significantly. Larvae in the low-Pb tanks had a deformity rate of 9%. Larvae and water from both the Zn and Pb aquaria had increasing metal concentrations with increasing sediment metal concentration. Results demonstrate that Zn and Pb each induce chironomid mouthpart deformities at various concentrations. However, a clear dose-related response was not demonstrated. Our research provides more support for the potential use of chironomid deformities as a tool for the assessment of heavy metal pollution in aquatic systems. [source] Substances with estrogenic activity in effluents of sewage treatment plants in southwestern Germany.ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 10 2001Abstract A gas chromatography/mass spectrometry method for the simultaneous quantitative determination of natural and synthetic estrogens (17,-estradiol, estrone, 17,-ethinylestradiol, and mestranol), phytoestrogens (genistein and ,-sitosterol), and xenoestrogens (benzyl butyl phthalate, dibutyl phthalate, bisphenol A, 4-nonylphenol [NP], 4-nonylphenoxyacetic acid [NP1EC], 4-nonyl-phenol diethoxylate [NP2EO], and ,-endosulfan) in effluents of sewage treatment plants (STPs) was developed. Identification and quantification were carried out with the standard addition method using analyte-specific and, in some cases, deuterium-labeled internal standards. The effluents of 18 STPs were investigated. Apart from ,-endosulfan and mestranol, all selected substances were detected in the majority of samples. The median concentrations of steroidal estrogens were between 0.4 ng/L (17,-ethiny-lestradiol) and 1.6 ng/L (17,-estradiol). The metabolites of the nonylphenol polyethoxylates, NP, NP1EC, and NP2EO were found in concentrations ranging from the upper-ng/L-range (NP) to the lower-,g/L range (NP1EC). For all substances except mestranol and ,-endosulfan, median values were calculated and compared to the results of other investigations in Europe and the United States. Possible dependencies of measured concentrations on the geographical location, the capacity, the influent composition, and the technical fitting of the STPs are discussed. [source] Acute toxicity of para -nonylphenol to saltwater animals,ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 3 2000Suzanne M. Lussier Abstract para -Nonylphenol (PNP), a mixture of alkylphenols used in producing nonionic surfactants, is distributed widely in surface waters and aquatic sediments, where it can affect saltwater species. This article describes a database for acute toxicity of PNP derived for calculating a national saltwater quality criterion. Using a flow-through exposure system with measured concentrations, we tested early life stages of four species of saltwater invertebrates and two species of fish. Static 96-h tests were also conducted on zoeal Homarus americanus, embryo-larval Mulinia lateralis, and larval Pleuronectes americanus. The number of organisms surviving the flow-through test was measured at 2, 4, 8, and 12 h and daily through day 7. Mortality for most species plateaued by 96 h. The ranked sensitivities (96-h 50% lethal concentrations, measured in micrograms per liter) for the species tested were 17 for Pleuronectes americanus, 37.9 (48-h 50% effective concentration) for Mulinia lateralis, 59.4 for Paleomonetes vulgaris, 60.6 for Americamysis bahia (formerly Mysidopsis bahia), 61.6 for Leptocheirus plumulosos, 70 for Menidia beryllina, 71 for Homarus americanus, 142 for Cyprinodon variegatus, and > 195 for Dyspanopius sayii. Values for the seven most sensitive of these species ranged over a factor of only 4.2. The narrow range of responses for PNP implies that exceeding a threshold concentration would endanger a large proportion of the aquatic community. [source] Absence of veno-occlussive disease in a cohort of multiple myeloma patients undergoing autologous stem cell transplantation with targeted busulfan dosageEUROPEAN JOURNAL OF HAEMATOLOGY, Issue 1 2006A. Clopés Abstract:,Background:,Plasma concentrations of oral busulfan (BU) were measured in multiple myeloma (MM) patients undergoing autologous peripheral blood stem cell transplantation (ASCT) with a double alkylating conditioning protocol in order to individualise doses of BU based on individual pharmacokinetic parameters and to reduce toxicities related to BU exposure. Patients and methods:,Forty-four consecutive patients with MM participating in the co-operative Spanish protocol were prospectively evaluated. Conditioning regimen prior to autologous infusion consisted of BU followed by melphalan. BU pharmacokinetic parameters were estimated for each patient after the first dose based on measured concentrations and subsequent doses were modified as necessary to achieve target exposure. Results:,Mean BU exposure (AUCss) (±DS) before dosage modification range from 3192 to 12 180 ng h/mL. Twenty-six out of 44 (59%) patients required dose adjustment. None of the patients developed hepatic veno-occlusive disease (VOD). Grade , II oropharyngeal mucositis was observed in the majority of patients (95%) and the severity of mucositis increased with increasing average steady-state BU plasma concentration. There were four treatment-related deaths: two patients died from multiorgan failure and two of respiratory infections. Of the remaining 40 patients, 15 were in complete remission with negative immunofixation, 21 in partial remission and four in stable disease 3 months after ASCT. Conclusions:,The results of the present study show the variability in BU pharmacokinetic parameters and suggest the possible relationship between toxicities and BU exposure. Individualising BU dosage in MM patients undergoing ASCT we observed the absence of VOD. [source] Cadmium leaching from some New Zealand pasture soilsEUROPEAN JOURNAL OF SOIL SCIENCE, Issue 1 2003C. W. Gray Summary Cadmium (Cd) inputs and losses from agricultural soils are of great importance because of the potential adverse effects Cd can pose to food quality, soil health and the environment in general. One important pathway for Cd losses from soil systems is by leaching. We investigated loss of Cd from a range of contrasting New Zealand pasture soils that had received Cd predominantly from repeated applications of phosphate fertilizer. Annual leaching losses of Cd ranged between 0.27 and 0.86 g ha,l, which are less than most losses recorded elsewhere. These losses equate to between 5 and 15% of the Cd added to soil through a typical annual application of single superphosphate, which in New Zealand contains on average 280 mg Cd kg,1 P. It appears that Cd added to soil from phosphate fertilizer is fairly immobile and Cd tends to accumulate in the topsoil. The pH of the leachate and the total volume of drainage to some extent control the amount of Cd leached. Additional factors, such as the soil sorption capacity, are also important in controlling Cd movement in these pasture soils. The prediction of the amount of Cd leached using the measured concentrations of Cd in the soil solution and rainfall data resulted in an overestimation of Cd losses. Cadmium concentrations in drainage water are substantially less than the current maximum acceptable value of 3 µg l,1 for drinking water in New Zealand set by the Ministry of Health. [source] Development of a mechanistic model for biological nutrient removal activated sludge systems and application to a full-scale WWTPAICHE JOURNAL, Issue 6 2010Bing-Jie Ni Abstract In wastewater treatment plants (WWTPs) the production of nitrite as an intermediate in the biological nutrient removal (BNR) process has been widely observed, but not been taken into account by most of the conventional activated sludge models yet. This work aims to develop a mechanistic mathematical model to evaluate the BNR process after resolving such a problem. A mathematical model is developed based on the Activated Sludge Model No.3 (ASM3) and the EAWAG Bio-P model with an incorporation of the two-step nitrification,denitrification, the anoxic P uptake, and the associated two-step denitrification by phosphorus accumulating organisms. The database used for simulations originates from a full-scale BNR municipal wastewater treatment plant. The influent wastewater composition is characterized using batch tests. Model predictions are compared with the measured concentrations of chemical oxygen demand (COD), NH -N, NO -N, NO -N, PO -P, and mixed liquid volatile suspended solids. Simulation results indicate that the calibrated model is capable of predicting the microbial growth, COD removal, nitrification and denitrification, as well as aerobic and anoxic P removal. Lee ABSTRACT: A dynamic, compartmental, simulation model (WETLAND) was developed for the design and evaluation of constructed wetlands to optimize nonpoint source (NPS) pollution control. The model simulates the hydrologic, nitrogen, carbon, dissolved oxygen (DO), bacteria, vegetative, phosphorous, and sediment cycles of a wetland system. Written in Fortran 77, the WETLAND models both free-water surface (FWS) and subsurface flow (SSF) wetlands, and is designed in a modular manner that gives the user the flexibility to decide which cycles and processes to model. WETLAND differs from many existing wetland models in that the interactions between the different nutrient cycles are modeled, minimizing the number of assumptions concerning wetland processes. It also directly links microbial growth and death to the consumption and transformations of nutrients in the wetland system. The WETLAND model is intended to be utilized with an existing NPS hydro-logic simulation model, such as ANSWERS or BASINS, but also may be used in situations where measured input data to the wetland are available. The model was calibrated and validated using limited data from a FWS wetland located at Benton, Kentucky. The WETLAND predictions were not statistically different from measured values for of five-day biochemical oxygen demand (BOD5), suspended sediment, nitrogen, and phosphorous. Effluent DO predictions were not always consistent with measured concentrations. A sensitivity analysis indicated the most significant input parameters to the model were those that directly affected bacterial growth and DO uptake and movement. The model was used to design a hypothetical constructed wetland in a subwatershed of the Nomini Creek watershed, located in Virginia. Two-year simulations were completed for five separate wetland designs. Predicted percent reductions in BOD5 (4 to 45 percent), total suspended solids (85 to 100 percent), total nitrogen (42 to 56 percent), and total phosphorous (38 to 57 percent) were similar to levels reported by previous research. HILLARP Summary.,Background, objectives and methods:,An accurate, sensitive and precise assay for reliable determination of the ristocetin cofactor activity of von Willebrand factor (VWF:RCo) in plasma and von Willebrand Factor (VWF)-containing concentrates has been evaluated. The assay is based on a commercially available automated protocol with modifications including a combination of adding additional ristocetin and the use of two calibration curves for the high and low measuring ranges. Results:,Addition of extra ristocetin resulted in improved measurement of VWF recoveries from various VWF-containing concentrates that were underestimated using the standard automated protocol. The modifications resulted in improved assay performance over an extended measuring range (2.00,0.03 IU mL,1). Accuracy was tested using VWF deficiency plasma spiked with the 1st international standard (IS) for VWF concentrate. Seven dilutions, ranging from 1.80 to 0.05 IU mL,1, were analyzed and resulted in measured concentrations between 80% and 100% of the assigned potency of the standard. Linearity was determined from the regression plot of the same concentrate dilutions and resulted in a correlation coefficient of 0.998. The repeatability, expressed as coefficient of variation, was 2% in the normal range (0.90 IU mL,1) and 8% at the level of 0.05 IU mL,1. The corresponding reproducibility results were 2% and 15% at the normal and low measuring ranges, respectively. Conclusions:,Analysis of patients with von Willebrand disease (VWD) indicates that the modified automated BCS® protocol has a superior discrimination power compared with the standard protocol. [source] Trace metal concentration in water and sediments of satellite lakes within Lake Victoria (Kenya) basinLAKES & RESERVOIRS: RESEARCH AND MANAGEMENT, Issue 3 2009Job Mwamburi Abstract Lakes Kanyaboli, Sare and Namboyo are three important freshwater satellite lakes in the northern region of the Lake Victoria (Kenya) basin. Lake Simbi, a small alkaline -saline crater-lake is located near the southern shoreline of the Nyanza Gulf of Lake Victoria. The three freshwater lakes provide unique aquatic habitats for the important indigenous fish species and other aquatic biodiversity, as well as serving as water resources for the surrounding communities. Surface and sediment samples were collected and examined to characterize these lake systems. Metal partitioning among the various defined geochemical phases also was determined. Based on the measured concentrations, the satellite lake waters are relatively uncontaminated with regard to Al (aluminium), manganese (Mn), zinc (Zn), copper (Cu) and chromium (Cr). Levels of most elements were below detection limits, as well as being well below the desirable drinking water guideline values proposed by the World Health Organization. The exception was the elevated iron (Fe) contents (>300 ,g L,1). The sediments exhibited a relatively low level of contamination with regard to heavy metals. The mean values of the measured metals in the sediments were compared to levels in adjacent Lake Victoria. The study results from these poorly-studied, and relatively unperturbed ecosystems, compared to the main body of lake Victoria, highlight their importance as conservation areas, despite the small-scale fishery activity, potential threats from human activities and their sensitivity to fluctuating environmental conditions. [source] Peak quantification in surface-enhanced laser desorption/ionization by using mixture modelsPROTEINS: STRUCTURE, FUNCTION AND BIOINFORMATICS, Issue 19 2006Martijn Dijkstra Abstract Surface-enhanced laser desorption/ionization (SELDI) time of flight (TOF) is a mass spectrometry technology for measuring the composition of a sampled protein mixture. A mass spectrum contains peaks corresponding to proteins in the sample. The peak areas are proportional to the measured concentrations of the corresponding proteins. Quantifying peak areas is difficult for existing methods because peak shapes are not constant across a spectrum and because peaks often overlap. We present a new method for quantifying peak areas. Our method decomposes a spectrum into peaks and a baseline using so-called statistical finite mixture models. We illustrate our method in detail on 8 samples from culture media of adipose tissue and globally on 64 samples from serum to compare our method to the standard Ciphergen method. Both methods give similar estimates for singleton peaks, but not for overlapping peaks. The Ciphergen method overestimates the heights of such peaks while our method still gives appropriate estimates. Peak quantification is an important step in pre-processing SELDI-TOF data and improvements therein will pay off in the later biomarker discovery phase. [source] Contribution of specific binding to the central benzodiazepine site to the brain concentrations of two novel benzodiazepine site ligandsBIOPHARMACEUTICS AND DRUG DISPOSITION, Issue 6 2007Andrew Pike Abstract The in vivo occupancy of brain benzodiazepine binding sites by compounds A and B was measured using a [3H]Ro 15-1788 binding assay and related to plasma and brain drug concentrations. The plasma concentration associated with 50% occupancy was higher for compound A than compound B (73 and 3.7 nM, respectively), however, there was little difference in the brain concentrations required (73 and 63 nM). Both compounds showed a non-linear relationship between plasma and brain concentrations such that above brain concentrations of ,100 nM increasing plasma concentrations did not result in a concomitant increase in brain concentrations. This is consistent with brain concentrations being dependent on a saturable compartment which was postulated to be the benzodiazepine binding site-containing GABAA receptors. This hypothesis was tested in ,1H101R mice, in which the ,1 subunit of the GABAA receptor is rendered insensitive to benzodiazepine binding resulting in an approximate 50% reduction in the total benzodiazepine-containing GABAA receptor population. It was shown that the Occ50 brain concentrations in the ,1H101R animals was lower (17 nM) than in wild type mice (63 nM), as was the plateau concentration in the brain (105 and 195 nM, respectively). These data suggest measured concentrations of compounds A and B in brain tissue are dependent on receptor expression with a minimal contribution from unbound and non-specifically bound compound.
