Selected AbstractsFrost tolerance in excised leaves of the common bugle (Ajuga reptans L.) correlates positively with the concentrations of raffinose family oligosaccharides (RFOs)PLANT CELL & ENVIRONMENT, Issue 8 2009SHAUN PETERS ABSTRACT Mass increases in raffinose family oligosaccharides (RFOs, ,1,6-galactosyl extensions of sucrose) are well documented in the generative tissues of many plants upon cold acclimation, and they (i.e. mainly the two shortest RFO members, raffinose and stachyose) have been suggested as frost stress protectants. Our focus here was on the longer RFO members as they commonly occur in the frost-hardy evergreen labiate Ajuga reptans in its natural habitat, and accumulate to their highest concentrations in winter when the plant is faced with sub-zero temperatures. We examined the effects of RFO concentration and chain length on frost tolerance using excised leaves which accumulate long-chain RFOs under both cold and warm conditions, thereby uncoupling the acclimation temperature from RFO production. We demonstrated that frost tolerance in excised A. reptans leaves correlates positively with long-chain RFO accumulation under both acclimation temperatures. After 24 d post-excision in the warm, the leaves had increased their RFO concentrations (mainly long-chain RFOs) 22-fold to 78 mg g,1 fresh weight, and decreased their EL50 values (temperature at which 50% leakage occurred) from ,10.5 to ,24.5 °C, suggesting a protective role for these oligosaccharides in the natural frost tolerance of A. reptans. [source] Distribution and growth of benthic macroinvertebrates among different patch types of the littoral zones of two arctic lakesFRESHWATER BIOLOGY, Issue 12 2006STEVEN R. BEATY Summary 1. To evaluate the effect of habitat patch heterogeneity on abundance and growth of macroinvertebrates in arctic lakes, macroinvertebrate abundance, individual biomass, and potential food resources were studied in three patch types in two arctic lakes on the Alaskan North Slope near the Toolik Lake Field Station. An experiment was conducted to determine which sediment patch type supported higher growth rates for Chironomus sp., a commonly occurring macroinvertebrate. 2. Potential organic matter (OM) resources were significantly higher in both rock and macrophyte patches than in open-mud patches. Total macroinvertebrate densities in both lakes were highest in rock patches, intermediate in macrophytes and lowest in open-mud. The open-mud patches also had lower species richness compared with other patch types. Additionally, individual biomass for one clam species and two chironomid species was significantly greater in rock patches than in open-mud. 3. In a laboratory experiment, Chironomus showed two to three times greater mass increase in sediments from macrophyte and rock patches than from open-mud patches. Rock and macrophyte experimental sediments had at least 1.5 × the percentage OM as open-mud sediments. 4. Chlorophyll a appeared to be the best predictor for invertebrate abundances across all patch types measured, whereas OM content appeared to be the variable most closely associated with Chironomus growth. 5. Our results combined with previous studies show that the relationships between macroinvertebrate community structure, individual growth, and habitat heterogeneity are complex, reflecting the interaction of multiple resources, and biotic interactions, such as the presence or absence of a selective vertebrate predator (lake trout, Salvelinus namaycush). [source] Analysis of coupled seepage and stress fields in rock mass around the Xiaowan arch damINTERNATIONAL JOURNAL FOR NUMERICAL METHODS IN BIOMEDICAL ENGINEERING, Issue 8 2004Chai Junrui Abstract The Xiaowan arch dam, with a maximum height of 292 m, is located across the Lancangjiang River in Yunnan Province of China, and once completed will be the highest arch dam in China. Because of the high water head and the arch action, it is necessary to analyse the interaction between seepage and stress fields in rock mass around the Xiaowan arch dam. Numerical solution of coupled seepage and stress fields in rock mass around the Xiaowan arch dam is analysed by means of the multi-level fracture network model and the finite element method. It can be shown from the computation results that storage of the reservoir makes the seepage field change much, and makes the effective vertical stress in rock foundation near the dam and the tensile stress in the abutment rock mass increase, and that the coupled action between seepage and stress fields should be taken into account. [source] The highly specialized vocal tract of the male Mongolian gazelle (Procapra gutturosa Pallas, 1777 , Mammalia, Bovidae)JOURNAL OF ANATOMY, Issue 5 2003R. Frey Abstract The entire head and neck of a wild adult male Mongolian gazelle (Procapra gutturosa) was dissected with special reference to its enlarged larynx. Two additional adult male specimens taken from the wild were analysed by computer tomography. The sternomandibularis, omohyoideus, thyrohyoideus and hyoepiglotticus muscles are particularly enlarged and improve laryngeal suspension and stabilization. The epiglottis is exceptionally large. A permanent laryngeal descent is associated with the evolution of an unpaired palatinal pharyngeal pouch. A certain momentary descent seems to occur during vocalization. The high lateral walls of the thyroid cartilage are ventrally connected by a broad keel. The large thyroarytenoid muscle is divided into two portions: a rostral ventricularis and a caudal vocalis muscle. A paired lateral laryngeal ventricle projects between these two muscles. The massive vocal fold is large and lacks any rostrally directed flexible structures. It is supported by a large cymbal-like fibroelastic pad. Vocal tract length was measured in the course of dissection and in computer tomographic images. Two representative spectrograms, one of an adult male and one of a juvenile, recorded in the natural habitat of the Mongolian gazelle are presented. In the spectrograms, the centre frequency of the lowest band is about 500 Hz in the adult male and about 790 Hz in the juvenile. The low pitch of the adult male's call is ascribed to the evolutionary mass increase and elongation of the vocal folds. In the habitat of P. gutturosa a call with a low pitch and, thus, with an almost homogeneous directivity around the head of the vocalizing animal may be optimally suited for multidirectional advertisement calls during the rut. The signal range of an adult male's call in its natural habitat can therefore be expected to be larger than the high-pitched call of a juvenile. [source] Energy allocation in juvenile sablefish: effects of temperature, ration and body sizeJOURNAL OF FISH BIOLOGY, Issue 3 2004S. M. Sogard The lipid deposition of juvenile sablefish Anoplopoma fimbria was examined, in particular, the changes in allocation over time. Growth rates of early juveniles (initial size 36,50 mm total length, LT) were manipulated using two temperatures (10 and 20° C) and two rations (ad libitum and 3,4% body mass day,1). Fish LT, mass and lipid content were measured every 3 weeks for 15 weeks. Irrespective of treatment, the relationship of total lipid content with body size was clearly hyperallometric; small juveniles allocated relatively more energy to growth and less to lipid storage than large juveniles. After adjusting for the influence of body size, temperature and ration significantly influenced body composition but these effects varied over the course of the experiment. In the first 3 week time period, fish on the high ration, high temperature treatment had reduced lipid storage relative to other treatments, but in all subsequent time periods their lipid concentrations were similar to or higher than those of fish on other treatments. In contrast, fish held at low rations and low temperatures initially had average levels of lipid concentration, but after 6 weeks their levels were lower than other treatments. Estimation of allocation to lipid storage over time (proportion of dry mass increase comprised of lipid) suggested that fish in all of the treatments were approaching an asymptotic level of lipid concentration (c. 50,60% of dry mass) but with different rates of lipid increase. Within a treatment, it was predicted that individual differences in allocation would result in trade-offs between somatic growth and storage. This trade-off was evident only for fish held on low rations at low temperatures. In contrast, fish held on high rations at high temperatures exhibited the opposite pattern of a positive correlation between somatic growth and storage. These results suggest that lipostatic regulation of appetite is unlikely in juvenile sablefish. When resources are unlimited, this species appears to adopt a maximizing strategy for both somatic growth and lipid accumulation. [source] The High-Temperature Oxidation Behavior of Reaction-Bonded Porous Silicon Carbide Ceramics in Dry OxygenJOURNAL OF THE AMERICAN CERAMIC SOCIETY, Issue 7 2010Chuanwei Zheng The oxidation behavior of reaction-bonded porous silicon carbide (RPSC) ceramics in dry oxygen between 1100° and 1500°C was investigated based on four specimens with different porosities. RPSC ceramics exhibited a rapid mass increase in the initial stage of oxidation but a slow mass increase in the following oxidation, which was considerably different from the oxidation behavior of dense SiC. The oxidation kinetics for RPSC can be better represented by an asymptotic law rather than the parabolic law for dense SiC. We suppose that, although oxidation occurred in the entire pore channels at the beginning, the pores were rapidly blocked by the oxide as their growth rate near the pore mouth was very fast due to sufficient oxygen. As the result, the oxidation of the pore interior was stopped in the absence of further oxygen supply. [source] Determination of the Oxygen Deficiency in Vacuum-Hot-Pressed Ferroelectric Ceramics Using a Thermogravimetric TechniqueJOURNAL OF THE AMERICAN CERAMIC SOCIETY, Issue 2 2000Qi Tan An attempt to measure the oxygen deficiency in vacuum-hot-pressed ferroelectric target materials was made, using a thermogravimetric method. The approach was intended to measure the relative mass increase of oxygen-deficient sheet samples, with respect to an oxidized reference that was annealed in air. A saturation of mass change was achieved, which indicated a saturated oxidation state and provided a basis for further calculation of the oxygen deficiency in the samples. Results were obtained for barium strontium titanate and lead lanthanum zirconate titanate compositions, which shows the feasibility of applying the thermogravimetric method to ferroelectric materials. Chandrashekara Abstract The diffusion behavior of a commercial permanent liquid hair dye in human hair has been investigated using Positron Annihilation Lifetime Spectroscopy (PALS) and gravimetric sorption method. The o-Ps lifetime parameters ,3 and I3 decrease rapidly during the first 60 minutes of sorption time. This is understood in terms of dye molecules filling the free volume cavities in hair. The sorption results suggest that the dye molecule diffusion is essentially a Fickian process. In the latter part of the sorption, where positron parameters remain almost constant, mass increase might be due to surface adhesion. These two stages of sorption are well separated by the positron technique. The study shows that the free volume theory and positron technique, widely used in polymer research, may expediently be used to understand hair properties, more importantly diffusion of dye molecules. KGaA, Weinheim) [source] Do changes in Atlantic salmon, Salmo salar L., fillet fatty acids following a dietary switch represent wash-out or dilution?AQUACULTURE RESEARCH, Issue 13 2003Test of a dilution model, its application Abstract The fatty acid compositions of fish tissue lipids usually reflect those of the feed lipids, but few attempts have been made to predict the way in which the profiles change or assess the time required for the fatty acid profile to stabilize following a dietary change. The present focus on the influences of vegetable oils and fish oils on the fatty acid compositions and sensory attributes of fish fillets increases the interest in the ability to make such predictions. A dilution model was tested using data for the influences of feed oils (rape/linseed (V) vs. sand-eel (F)) and dietary fat concentrations (ca. 30% (H) vs. ca. 20% (L)) on the body growth and fatty acid compositions of the fillets of Atlantic salmon, Salmo salar L., parr and post smolt. Fish given HV or LV feeds during freshwater rearing (mass increase from ca. 19 g to ca. 130 g) were switched to HF and LF feeds following parr,smolt transformation. The changes in fillet percentages of 18:1, 18:2 (n-6) and 18:3 (n-3) during 98 days of on-growing in seawater (mass increase from ca. 130 g to ca. 380 g) conformed closely to predictions made on the basis of the dilution model. Model applications require information about the proportionate increase in fillet fat over time, but the relative changes in body mass can be used as a surrogate provided that both fillet yield (as a % of body mass) and fillet fat percentage change little over time. This is not the case for small salmon, but does seem to apply to larger salmon as they approach harvest size. This means that, for large salmon, ratios of changes in body mass can be substituted for ratios in the quantitative change in fillet fat without the introduction of a large error in the prediction of the change in fillet fatty acid profile following the introduction of a novel feed. [source] Crystallization of a pentapeptide-repeat protein by reductive cyclic pentylation of free amines with glutaraldehydeACTA CRYSTALLOGRAPHICA SECTION D, Issue 5 2009Matthew W. Vetting The pentapeptide-repeat protein EfsQnr from Enterococcus faecalis protects DNA gyrase from inhibition by fluoroquinolones. EfsQnr was cloned and purified to homogeneity, but failed to produce diffraction-quality crystals in initial crystallization screens. Treatment of EfsQnr with glutaraldehyde and the strong reducing agent borane,dimethylamine resulted in a derivatized protein which produced crystals that diffracted to 1.6,Å resolution; their structure was subsequently determined by single-wavelength anomalous dispersion. Analysis of the derivatized protein using Fourier transform ion cyclotron resonance mass spectrometry indicated a mass increase of 68,Da per free amino group. Electron-density maps about a limited number of structurally ordered lysines indicated that the modification was a cyclic pentylation of free amines, producing piperidine groups. [source] Inactivation of Aeromonas hydrophila metallo-,-lactamase by cephamycins and moxalactamFEBS JOURNAL, Issue 13 2001Astrid Zervosen Incubation of moxalactam and cefoxitin with the Aeromonas hydrophila metallo-,-lactamase CphA leads to enzyme-catalyzed hydrolysis of both compounds and to irreversible inactivation of the enzyme by the reaction products. As shown by electrospray mass spectrometry, the inactivation of CphA by cefoxitin and moxalactam is accompanied by the formation of stable adducts with mass increases of 445 and 111 Da, respectively. The single thiol group of the inactivated enzyme is no longer titrable, and dithiothreitol treatment of the complexes partially restores the catalytic activity. The mechanism of inactivation by moxalactam was studied in detail. Hydrolysis of moxalactam is followed by elimination of the 3, leaving group (5-mercapto-1-methyltetrazole), which forms a disulfide bond with the cysteine residue of CphA located in the active site. Interestingly, this reaction is catalyzed by cacodylate. [source] Why are very large herbivores absent from Australia?JOURNAL OF BIOGEOGRAPHY, Issue 4 2000A new theory of micronutrients Abstract Aim We propose a Megacatalyst Theory, based on the pivotal role of the micronutrients iodine (I), cobalt (Co) and selenium (Se), in answer to the body size anomaly of herbivores on different continents, and the previously unexplained absence of megaherbivores in certain environments. Location It is anomalous that megaherbivores are absent from Australia while present in even dry and nutrient-poor parts of southern Africa, and that they have been exterminated from the Americas, but not south-east Asia. Methods We hypothesize that I, Co and Se are micronutrients in quantity, but megacatalysts in effect, determining maximum body size and pace of life, hence whether energy is used by animals or fire. The Megacatalyst Theory suggests that the greater the reproductive rate and brain size relative to body size, the greater the probable demand for I, Co and Se. Results Balanced supply of I, Co and Se, within narrow tolerances, is elusive because of disparate cycles: I gravitates towards the sea, whereas Co and Se are concentrated in ultramafics and organic shales, respectively. Sufficiency of these micronutrients, at less than toxic concentrations, is vital for rapid metabolism and growth, particularly of the nervous system. Iodine controls thermogenesis, Co controls the gut fermentation supplying herbivores, and Se controls biochemical damage where both processes occur rapidly. The supply of Co allows vegetation to be metabolized instead of combusted, by promoting digestion of fibre by gut microbes. Herbivores demand I, Co and Se in greater concentrations than palatable plants necessarily contain, as an increasing proportion of energy is fermented from fibre with increasing body size. Economy of scale is limited by loss of I in urine (partly compensated by thyroid size), Co in faeces (partly compensated by gut compartments), and Se both ways. Main conclusions The larger the herbivore species, the more it may depend on supplementation in order to survive predation by humans. As body mass increases, Co becomes deficient before I, because it is essential for rumination, and cannot be absorbed by the skin. Moderate uplift of a fairly flat landscape sustainably supplies I from mineralized springs, and Co from rocks (and Se from both), avoiding the excess of I in the sea and the excess of Co on high mountains. Iodine and Se leached to groundwater under dry climates are inaccessible to herbivores on a continent as flat as Australia, where even kangaroos have limited fecundity and intelligence compared to southern African ruminants of similar body mass. Where springs and associated earth-licks were available in the late Pleistocene, megaherbivores could evolve to survive the era of domestication. [source] Locomotor impairment of gravid lizards: is the burden physical or physiological?JOURNAL OF EVOLUTIONARY BIOLOGY, Issue 2 2000Olsson Pregnancy is associated with reduced locomotor performance in several reptile species, but the reasons for this reduction remain unclear. Previous authors generally have assumed that the decreased maternal mobility is due to the physical burden of the clutch, but our data on a viviparous Tasmanian scincid lizard (Niveoscincus microlepidotum) suggest a different interpretation. Running speeds of gravid female skinks decrease during gestation (as litter mass increases), but this locomotor impairment is due to physiological changes associated with pregnancy, rather than simple physical burdening. Maternal running speeds are unrelated to litter masses, and do not increase in the week after parturition. Females with very large abdominal fat-bodies (due to ad libitum feeding in the laboratory), equivalent in mass to the litter, nonetheless run rapidly. If the locomotor ,costs' of reproduction reflect all-or-none physiological changes associated with pregnancy, then the magnitude of such costs may correlate only weakly with the actual level of reproductive investment. Because life-history models predict that the relationship between fecundity and ,cost' has important evolutionary consequences, our results highlight the need to clarify the causal basis for locomotor impairment in gravid reptiles. [source] Peak capacity of ion mobility mass spectrometry: the utility of varying drift gas polarizability for the separation of tryptic peptidesJOURNAL OF MASS SPECTROMETRY (INCORP BIOLOGICAL MASS SPECTROMETRY), Issue 4 2004Brandon T. Ruotolo Abstract Ion mobility mass spectrometry (IM-MS) peptide mass mapping experiments were performed using a variety of drift gases (He, N2, Ar and CH4). The drift gases studied cover a range of polarizabilities ((0.2,2.6) × 10,24 cm3) and the peak capacities obtained for tryptic peptides in each gas are compared. Although the different gases exhibit similar peak capacities (5430 (Ar) to 7580 (N2)) in some cases separation selectivity presumably based on peptide conformers (or conformer populations), is observed. For example the drift time profiles observed for some tryptic peptide ions from aldolase (rabbit muscle) show a dependence on drift gas. The transmission of high-mass ions (m/z > 2000) is also influenced by increased scattering cross-section of the more massive drift gases. Consequently the practical peak capacity for IM-MS separation cannot be assumed to be solely a function of resolution and the ability of a gas to distribute signals in two-dimensional space; rather, peak capacity estimates must account for the transmission losses experienced for peptide ions as the drift gas mass increases. [source] IgG binding kinetics to oligo B protein A domains on lipid layers immobilized on a 27,MHz quartz-crystal microbalanceJOURNAL OF MOLECULAR RECOGNITION, Issue 2 2007Hideyuki Mitomo Abstract Although molecular recognitions between membrane receptors and their soluble ligands have been analyzed using their soluble proteins in bulk solutions, molecular recognitions of membrane receptors should be studied on lipid membranes considering their orientation and dynamics on membrane surfaces. We employed Staphylococcal Protein A (SpA) oligo B domains with long trialkyl-tags from E. coli (LppBx, x,=,1, 2, and 5) and immobilized LppBx on lipid layers using hydrophobic interactions from the trialkyl-tag, while maintaining the orientation of B domain-chains on a 27,MHz quartz-crystal microbalance (QCM; AT-cut shear mode). The binding of IgG Fc regions to LppBx on lipid layers was detected by frequency decreases (mass increases) on the QCM. The maximum amount bound (,mmax), association constants (Ka), association and dissociation rate constants (k1 and k,1, respectively) were obtained. Binding kinetics of IgG to LppB2 and LppB5 were quite similar, showing a simple 1:1 binding of the IgG Fc region to the B domain, when the surface coverage of LppB2 and LppB5 on the lipid surface is low (1.4%). When LppB5 was immobilized at the high surface coverage of 3.5%, the complex bindings of IgG such as one IgG bound to one or two LppB5 on the membrane could be observed. [source] The distribution of ejected subhaloes and its implication for halo assembly biasMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 4 2009Huiyuan Wang ABSTRACT Using a high-resolution cosmological N -body simulation, we identify the ejected population of subhaloes, which are haloes at redshift z= 0 but were once contained in more massive ,host' haloes at high redshifts. The fraction of the ejected subhaloes in the total halo population of the same mass ranges from 9 to 4 per cent for halo masses from ,1011 to ,1012 h,1 M,. Most of the ejected subhaloes are distributed within four times the virial radius of their hosts. These ejected subhaloes have distinct velocity distribution around their hosts in comparison to normal haloes. The number of subhaloes ejected from a host of given mass increases with the assembly redshift of the host. Ejected subhaloes in general reside in high-density regions, and have a much higher bias parameter than normal haloes of the same mass. They also have earlier assembly times, so that they contribute to the assembly bias of dark matter haloes seen in cosmological simulations. However, the assembly bias is not dominated by the ejected population, indicating that large-scale environmental effects on normal haloes are the main source for the assembly bias. [source] The UV properties of E+A galaxies: constraints on feedback-driven quenching of star formationMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 3 2007S. Kaviraj ABSTRACT We present the first large-scale study of E+A galaxies that incorporates photometry in the ultraviolet (UV) wavelengths. E+A galaxies are ,post-starburst' systems, with strong Balmer absorption lines indicating significant recent star formation, but without [O ii] and H, emission lines which are characteristic of ongoing star formation. The starburst that creates the E+A galaxy typically takes place within the last Gyr and creates a high fraction (20,60 per cent) of the stellar mass in the remnant over a short time-scale (<0.1 Gyr). We find a tight correlation between the luminosity of our E+A galaxies and the implied star formation rate (SFR) during the starburst. While low-luminosity E+As [M(z) > ,20] exhibit implied SFRs of less than 50 M, yr,1, their luminous counterparts [M(z) < ,22] show SFRs greater than 300 and as high as 2000 M, yr,1, suggesting that luminous and ultra-luminous infrared galaxies in the low-redshift Universe could be the progenitors of massive nearby E+A galaxies. We perform a comprehensive study of the characteristics of the quenching that truncates the starburst in E+A systems. We find that, for galaxies less massive than 1010 M,, the quenching efficiency decreases as the galaxy mass increases. However, for galaxies more massive than 1010 M,, this trend is reversed and the quenching efficiency increases with galaxy mass. Noting that the mass threshold at which this reversal occurs is in excellent agreement with the mass above which active galactic nuclei (AGN) become significantly more abundant in nearby galaxies, we use simple energetic arguments to show that the bimodal behaviour of the quenching efficiency is consistent with AGN and supernovae (SN) being the principal sources of negative feedback above and below M, 1010 M,, respectively. The arguments assume that quenching occurs through the mechanical ejection or dispersal of the gas reservoir and that, in the high-mass regime (M > 1010 M,), the Eddington ratios in this sample of galaxies scale as M,, where 1 < , < 3. Finally, we use our E+A sample to estimate the time it takes for galaxies to migrate from the blue cloud to the red sequence. We find migration times between 1 and 5 Gyr, with a median value of 1.5 Gyr. [source] Examining the extinction risk of specialized folivores: a comparative study of Colobine monkeysAMERICAN JOURNAL OF PRIMATOLOGY, Issue 9 2008Jason M. Kamilar Abstract Species extinctions are nonrandom with some taxa appearing to possess traits that increase their extinction risk. In this study, eight predictors of extinction risk were used as independent variables to predict the IUCN category of a subfamily of specialized folivorous primates, the Colobinae. All data were transformed into phylogenetically independent contrasts and were analyzed using bivariate regressions, multiple regression, and a maximum likelihood approach using Akaike's Information Criterion to assess model performance. Once an outlier was removed from the data set, species that devote a smaller proportion of their diet to mature leaf consumption appear to be at a greater risk of extinction. Also, as female body mass increases, so does extinction risk. In contrast, as maximum latitude and the number of habitat types increase, extinction risk appears to decrease. These findings emphasize the importance of examining detailed dietary variation for predicting extinction risk at a relatively fine taxonomic scale and, consequently, may help improve conservation management. [source] Adiposity, fatty acid composition, and delta-9 desaturase activity during growth in beef cattleANIMAL SCIENCE JOURNAL, Issue 5 2006Stephen B. SMITH ABSTRACT Oleic acid (18:1n-9) is the most abundant fatty acid in bovine adipose tissue. Because most of the lipid in bovine muscle is contributed by intramuscular adipocytes, oleic acid also is the predominant fatty acid in beef. In many species, the concentration of oleic acid in adipose tissue is dictated by the average concentration of oleic acid in the diet, but in ruminant species such as beef cattle, oleic acid is hydrogenated largely to stearic acid by ruminal microorganisms. In these species, the concentration of oleic acid in adipose tissue is dependent upon the activity of ,9 desaturase, encoded by the stearoyl coenzyme A desaturase (SCD) gene. Expression of the SCD gene is essential for bovine preadipocyte differentiation, and desaturase gene expression and catalytic activity increase dramatically as adipose tissue mass increases after weaning. Feeding a hay-based diet to American Wagyu steers to a typical Japanese bodyweight endpoint (650 kg) markedly stimulated desaturase enzyme activity as well as the accumulation of both oleic acid and intramuscular lipid, but the increase in oleic acid and intramuscular lipid was much less in hay-fed Angus steers. Increasing the concentration of oleic acid improves the palatability and healthiness of beef, and Korean Hanwoo and Japanese Black (and American Wagyu) seem especially well adapted to accumulate oleic acid in their adipose tissue. [source] Osmoregulation, growth and moulting cycles of the giant freshwater prawn (Macrobrachium rosenbergii) at different salinitiesAQUACULTURE RESEARCH, Issue 9 2010Do Thi Thanh Huong Abstract The giant freshwater prawn, Macrobrachium rosenbergii, is a species with a high commercial value in aquaculture. Two experiments were performed to determine the effects of salinities on the osmoregulation, growth and molting cycles of M. rosenbergii during growout. The first experiment was designed to determine whether these animals are capable of adapting to the changes in salinity seen in salinity intrusions in tropical deltas, with an incremental increase in salinity of 3, per day from 0, to 30, Haemolymph osmolality was rapidly regulated up to salinities of 15, , whereas animals conformed at higher salinities. The second experiment determined the growth, moulting cycle, osmolality, muscle water content and mortality during a 4-month experiment at 0,, 15, or 25, salinity. The weight gains in 0, and 15, were not significantly different and were comparable to the growth rates achieved in production farms with body mass increases of 2.6 and 2.3-fold their initial body mass, respectively, after 4 months. The 25, group suffered from low growth, high mortality and a significantly lower moulting frequency. These data show that this species can be reared in brackish water up to 15,, allowing for farming in the large areas impacted by salt water intrusions in tropical deltas. The KID (kinase-inducible domain) of transcription factor CREB (cyclic AMP response element binding protein) is known to interact with the KIX domain of coactivator CBP (CREB binding protein), facilitated by phosphorylation at Ser-133 of the KID. The KIX domain peptide (86,aa) was immobilized on the QCM gold electrode surface by means of a poly(ethylene glycol) spacer. Binding of the KID peptide (46,aa) to the KIX peptide was detected by frequency decreases (mass increases) of the QCM. Both maximum binding amount (,mmax) and association constants (Ka) obtained from the QCM measurements increased as a result of phosphorylation of Ser-133 of the KID peptide. The Ka values for KIX peptide to the phosphorylated (pKID) and unphosphorylated KID peptides were (93±2)×103 and (5±1)×103,M,1, respectively. This difference was explained by the dissociation rate constant (k,1) of the pKID being 20 times smaller than that of the KID, while association rate constants (k1) were independent of phosphorylation.
