Selected AbstractsSOCIAL ECOLOGY AND RECIDIVISM: IMPLICATIONS FOR PRISONER REENTRY,CRIMINOLOGY, Issue 2 2008DANIEL P. MEARS Despite the marked increase in incarceration over the past 30 years and the fact that roughly two thirds of released offenders are rearrested within 3 years of release, we know little about how the social ecology of the areas to which offenders return may influence their recidivism or whether it disproportionately affects some groups more than others. Drawing on recent scholarship on prisoner reentry and macrolevel predictors of crime, this study examines a large sample of prisoners released to Florida communities to investigate how two dimensions of social ecology,resource deprivation and racial segregation,may independently, and in interaction with specific populations, influence recidivism. The findings suggest that ecology indeed is consequential for recidivism, and it differentially influences some groups more than others. We discuss these findings and their implications for theory, research, and policy. [source] Micronization of the officinal component baicalin by SEDS-PA processCRYSTAL RESEARCH AND TECHNOLOGY, Issue 6 2007Wen Zhi He Abstract Application of micronizing technologies in processing Chinese herbal medicines is very important to improve the forms of prepared Chinese herbal medicines and promote their therapeutic efficacy. Baicalin, a major active component of the typical Chinese herb medicine Scullateria baicallensis Georgi, was micronized using the Solution Enhanced Dispersion by Supercritical fluids though Prefilming Atomization (SEDS-PA) process with the aim of evaluating the efficiency of applying supercritical fluid precipitation technologies in Chinese herb medicine. This study has shown that acicula or rod-like baicalin crystals with Particle Size (PS) of about 20×100 ,m were successfully micronized by the SEDS-PA process to long rod-like, twisted fiber-like or fibrous net-like microparticles with PS of 0.1-2.2 ,m in width within the range of experiments performed. It was found that a substantial reduction of baicalin microparticles' sizes could lead to a marked increase of adhesions among them and subsequent microparticles agglomeration. With the increase of supercritical CO2 flow rate and the decrease of solution concentration and solution flow rate, smaller and much more agglomerated microparticles were obtained. Increasing pressure led to formation of smaller microparticles. A larger tendency of particles agglomeration was produced at a higher temperature. (© 2007 WILEY-VCH Verlag GmbH & Co. KGaA, Weinheim) [source] Delivery of small interfering RNA with a synthetic collagen poly(Pro-Hyp-Gly) for gene silencing in vitro and in vivoDEVELOPMENT GROWTH & DIFFERENTIATION, Issue 8 2010Taro Adachi Silencing gene expression by small interfering RNAs (siRNAs) has become a powerful tool for the genetic analysis of many animals. However, the rapid degradation of siRNA and the limited duration of its action in vivo have called for an efficient delivery technology. Here, we describe that siRNA complexed with a synthetic collagen poly(Pro-Hyp-Gly) (SYCOL) is resistant to nucleases and is efficiently transferred into cells in vitro and in vivo, thereby allowing long-term gene silencing in vivo. We found that the SYCOL-mediated local application of siRNA targeting myostatin, coding a negative regulator of skeletal muscle growth, in mouse skeletal muscles, caused a marked increase in the muscle mass within a few weeks after application. Furthermore, in vivo administration of an anti-luciferase siRNA/SYCOL complex partially reduced luciferase expression in xenografted tumors in vivo. These results indicate a SYCOL-based non-viral delivery method could be a reliable simple approach to knockdown gene expression by RNAi in vivo as well as in vitro. [source] Academic performance in children with rolandic epilepsyDEVELOPMENTAL MEDICINE & CHILD NEUROLOGY, Issue 5 2008P Piccinelli MD The aim of this study was to investigate the frequency of reading, writing, and calculation disabilities in children with typical rolandic epilepsy (RE) and healthy control children. We also aimed to define the possible electroclinical markers of specific cognitive dysfunctions in RE. School abilities were evaluated and compared in 20 children (eight males, 12 females; mean age 10y 3mo [SD 1y 7mo]; range 7y 9mo-12y 9mo) consecutively diagnosed with typical RE, and a group of 21 healthy controls (nine males, 12 females; mean age 10y 4mo [SD 1y 8mo]; range 7y 6mo-13y 3mo). All the children received standardized neuropsychological tests. For each patient an exhaustive seizure diary was kept and all the sleep electroencephalogram (EEG) recordings were reviewed. Specific difficulties with reading, writing, and calculation (diagnosed according to the criteria of the Diagnostic and Statistical Manual of Mental Disorders, 4th edition) were found in nine out of 20 children with RE and two out of 21 healthy controls (,2=0.01). The specific learning disabilities in the RE group were correlated with a marked increase in epileptiform discharges during sleep (,2=0.02) and an early onset of epilepsy (,2=0.02). Our findings suggest that seizure onset before age 8 years and epileptiform discharges (more than 50% of the sleep EEG recording) in several tracings over more than a year are relevant markers for identifying patients at risk of developing academic difficulties. [source] Serotonin decreases generation of dopaminergic neurons from mesencephalic precursors via serotonin type 7 and type 4 receptorsDEVELOPMENTAL NEUROBIOLOGY, Issue 1 2007J. Parga Abstract Inductive signals mediating the differentiation of neural precursors into serotonergic (5-HT) or dopaminergic neurons have not been clarified. We have recently shown that in cell aggregates obtained from rat mesencephalic precursors, reduction of serotonin levels induces a marked increase in generation of dopaminergic neurons. In the present study we treated rat neurospheres with antagonists of the main subtypes of 5-HT receptors, 5-HT transport inhibitors, or 5-HT receptor agonists, and studied the effects on generation of dopaminergic neurons. Cultures treated with Methiothepin (5-HT1,2,5,6,7 receptor antagonist), the 5-HT4 receptor antagonist GR113808;67:00,.or the 5-HT7 receptor antagonist SB 269970 showed a significant increase in generation of dopaminergic cells. Treatment with the 5-HT1B/1D antagonist GR 127935, the 5-HT2 antagonist Ritanserin, the 5-HT transporter inhibitor Fluoxetine, the dopamine and norepinephrine transport inhibitor GBR 12935, or with both inhibitors together, or 5-HT4 or 5-HT7 receptor agonists induced significant decreases in generation of dopaminergic cells. Cultures treated with WAY100635 (5-HT1A receptor antagonist), the 5-HT3 receptor antagonist Ondasetron, or the 5-HT6 receptor antagonist SB 258585 did not show any significant changes. Therefore, 5-HT4 and 5-HT7 receptors are involved in the observed serotonin-induced decrease in generation of dopaminergic neurons from proliferating neurospheres of mesencephalic precursors. 5-HT4 and 5-HT7 receptors were found in astrocytes and serotonergic cells using double immunolabeling and laser confocal microscopy, and the glial receptors appeared to play a major role. © 2006 Wiley Periodicals, Inc. Belch The morbidity and mortality associated with peripheral arterial disease (PAD) creates a huge burden in terms of costs both to the patient and to the health service. PAD is a deleterious and progressive condition that causes a marked increase in the risk of cardiovascular and cerebrovascular events. Further, PAD has a major negative impact on quality of life and mortality, and is associated with an increased risk of limb amputation. The clinical profile of patients at risk of PAD overlaps considerably with the known cardiovascular risk factors. These include, increasing age, smoking habit, diabetes, hypertension, dyslipidaemia, male sex and hyperhomocysteinaemia. For women, hormone replacement therapy appears to be associated with a reduced risk of PAD. Published PAD guidelines recommend aggressive management of risk factors, stressing the importance of lifestyle modification, antiplatelet agents, treating dyslipidaemia and diabetes. However, a large number of patients with PAD go undetected, either because they do not report their symptoms or because they are asymptomatic. It is therefore important to improve detection rates so that these patients can receive appropriate risk factor management. [source] Retinal capillary basement membrane thickness in diabetic mice genetically modified at the haptoglobin locusDIABETES/METABOLISM: RESEARCH AND REVIEWS, Issue 2 2007Rachel Miller-Lotan Technion Faculty of Medicine Abstract Background Individuals with diabetes mellitus (DM) homozygous for the haptoglobin (Hp) 1 allele are at decreased risk of retinopathy as compared to DM individuals with the Hp 2 allele. We sought to recapitulate these findings in DM mice genetically modified at the Hp locus. Methods An early morphological characteristic of the microangiopathy seen in diabetic retinal disease is retinal capillary basement membrane (RCBM) thickening. RCBM thickness as assessed by electron microscopy was performed on a total of 12 eyes taken from three mice in each of the four study groups (three eyes from C57Bl/6 Hp 1 and C57Bl/6 Hp 2 mice with and without streptozotocin-induced diabetes). Results The non-parametric Kruskal,Wallis ANOVA test demonstrated that there was a highly significant difference between the four groups of mice (P < 0.0001). Mann,Whitney tests for specific pair-wise comparisons demonstrated that there was no significant difference in the RCBM thickness between Hp 1 and Hp 2 mice (p = 0.70) or between DM Hp 1 and non-DM Hp 1 mice (p = 0.42). However, induction of diabetes resulted in a marked increase in RCBM thickness in Hp 2 mice compared to non-DM Hp 2 mice (p = 0.0004) and compared to DM Hp 1 mice (p = 0.0005). Conclusions A highly significant increase in RCBM thickness was observed in DM mice with the Hp 2 genotype. These data provide important support for association studies done in humans showing an increased prevalence of diabetic retinopathy in individuals with the Hp 2 genotype. Tamborlane Abstract While treatment of Type 1 diabetes mellitus (T1DM) in children and adolescents is especially difficult, recent technological advances have provided new therapeutic options to clinicians and patients. The urgency to achieve strict diabetes control and the introduction of new and improved insulin pumps have been accompanied by a marked increase in use of continuous subcutaneous insulin infusion (CSII) therapy in youth with diabetes. Results of clinical outcome studies indicate that CSII provides a safe and effective alternative to multiple daily injection (MDI) therapy, even when employed in a regular clinic setting in a large number of children. The safety and efficacy of CSII is further enhanced by the introduction of lispro and aspart insulin. The sharper peaks and shorter duration of action of these very rapid-acting insulin analogues provides a means to achieve better control of post-prandial hyperglycaemia with less late post-prandial and nocturnal hypoglycaemia. Glargine insulin, a soluble and essentially peakless long-acting insulin analogue, may provide a better basal insulin for MDI regimens, but there are limited published data with this agent in children with T1DM. A number of systems for pulmonary delivery of insulin are in development and preliminary results of Phase III studies have been promising. Like CSII, inhaled insulin allows the child to take bolus insulin doses before each meal without having to take a premeal injection. A major obstacle to effective treatment is that self-monitoring of three to four blood glucose levels a day often misses the marked glycaemic excursions that characterize T1DM in young patients. On the other hand, new continuous glucose sensing systems provide a wealth of data that can be used to optimize basal and bolus therapy, regardless of how insulin is administered. Med. 18, 864,870 (2001) [source] Discussion of landslide self-organized criticality and the initiation of debris flowEARTH SURFACE PROCESSES AND LANDFORMS, Issue 2 2007Chen Chien-Yuan Abstract The study contains descriptions of landslides and debris flows in a small upland catchment in Taiwan between 1986 and 2004. This catchment, the Chushui Creek, is situated near the epicentre of the 1999 M7·6 Chi-Chi earthquake. This is an area with high background erosion rate. The Chi-Chi earthquake caused a sharp increase in the rate of mass wasting in the epicentral area, and the data presented in this study illustrates the geomorphic change associated with the earthquake. The measurements of the geometry of the trunk stream of the Chushui catchment also show its change due to the impact of a strong typhoon in 1996. Two channel reaches that were affected by separate debris flows during this event were identified. Each reach has three sections: scour, transfer and deposition. Cross profiles of these sections show a systematic change from V-shaped bedrock channel in the scour areas to flat-floored channel in the transfer and deposition areas. Debris flows also occurred on other occasions in this channel, and their frequency has increased since the 1999 earthquake. In addition, this study contains precise geographic and statistical descriptions of the landslides triggered by three typhoons and an earthquake that affected the catchment since 1996. The total landslide area is measured for different time intervals, and a marked increase in landslide incidence is found after the 1999 earthquake. Frequency,area plots of the 126 landslides in the catchment indicate a power-law scaling with an exponent of about -1·5 of these quantities, as has been observed in other studies. We propose that a link exists between landslide frequency,area distributions and initialization of debris flow in the catchment. It may be classified as a self-organized criticality process with a critical frequency,area distribution for the landslide population. In this case, the landslide frequency,area distribution is useful in quantifying the severity of the trigger and the contribution of landslides to debris flow. [source] Bioaccumulation and biotransformation of arsenic in the Mediterranean polychaete Sabella spallanzanii experimental observationsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 6 2007Alessandra Notti Abstract The Mediterranean fan worm Sabella spallanzanii is characterized by elevated basal levels of arsenic in branchial crowns (>1,000 ,g/g) and an unusual prevalence of dimethylarsinic acid (DMA), a relatively toxic compound with a possible antipredatory role. The aim of this work was to obtain further insights on the capability of this polychaete to accumulate arsenic from different compounds and to operate biotransformation reactions. Laboratory exposures to arsenate (AsV), dimethylarsinic acid (DMA), trimethylarsine (TMA), and arsenobetaine (AsB) revealed significant differences among tissues and kind of experiments. The highest increases of arsenic content were observed in branchial crowns of organisms treated with arsenate, which can enter the cell through the phosphate carrier system; lower variations were measured with DMA and TMA, while not-significant changes of total As occurred after treatments with AsB. In body tissues, exposure to AsV, DMA, and TMA confirmed a progressively lower accumulation of total arsenic, while a marked increase was caused by AsB. Obtained results suggested that accumulated arsenic could be chemically transformed, thus explaining the elevated basal levels of DMA typical of S. spallanzanii; during all the experiments, DMA was the most accumulated molecule, suggesting that this species possesses the enzymatic pathways for methylation and demethylation reactions of inorganic and trimethylated arsenicals. Only arsenobetaine was not converted into DMA, which would confirm a microbial pathway for degradation for this molecule, particularly important in body tissues of S. spallanzanii for the presence of bacteria associated to digestive tracts. Overall, the present study suggests future investigations on the biological role of arsenic and DMA in S. spallanzanii as a potential adaptive mechanism against predation in more vulnerable tissues. [source] Oxidative stress biomarkers in bivalves transplanted to the Guadalquivir estuary after Aznalcóllar spillENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 1 2003Antonio Romero-Ruiz Abstract Biological effects of metals were studied in clams (Scrobicularia plana) transplanted to Guadalquivir estuary (Spain) at several times after the spill of toxic metals from Aznalcóllar pyrite mine (southwest Spain) (April 1998) using biochemical biomarkers responsive to reactive oxygen species. Significant As, Cd, Fe, Mn, Ni, and Pb uptake was found in clams living for seven months at the estuary (from July 1999). Increased activity of antioxidant (catalase, glucose-6-phosphate, and 6-phosphogluconate dehydrogenase) and glutathione-related (glutathione reductase and glyoxalase I and II) enzymes was also found after short exposures; the levels of malondialdehyde and metallothionein increased also, particularly with long exposures. Clams living four weeks at the estuary (from March 2000) but not at a reference site also accumulated metals. The higher malondialdehyde and lower reduced-glutathione levels and the more oxidized glutathione status confirmed the oxidative stress of clams living at the estuary, while no marked increase of antioxidant activities was found this time. Lower metal availability along the second transplant could explain the limited responses in this shorter experiment. Although the status of Guadalquivir estuary has recovered since Aznalcóllar spill, continuous monitoring is needed to confirm its progress and to be alert to possible deterioration after heavy rains. [source] Spindles-Inducing Mechanism Modulates Sleep Activation of Interictal Epileptiform Discharges in the Landau,Kleffner SyndromeEPILEPSIA, Issue 2 2000L. Nobili Summary: Purpose: Landau,Kleffner syndrome (LKS) is characterized by a marked increase of interictal epileptiform discharges (IEDs) during sleep. During nonrapid eye movement (NREM) sleep, neuronal membrane potential oscillations lead to the appearance of spindles and delta waves in the surface EEG and might develop into paroxysmal synchronization. Spectral analysis allows the quantitative description of the dynamics of delta (slow-wave activity, SWA, 0.5-4.5 Hz) and sigma activity (SA, 12.0,16.0 Hz) and can be used to assess the relation between SA, SWA, and IEDs during sleep. Methods: We performed six overnight continuous EEG-polysomnographic studies in three patients with LKS. The temporal series of SWA and SA were obtained from a spike-free derivation lead. The IEDs count was performed on the most active lead. Relations between sigma and SWA and time series of lEDs were tested by means of correlation techniques after data normalization. Results: Our results revealed a significantly higher correlation between IEDs and SA with respect to SWA in all the subjects, in total sleep time. The same analysis limited to NREM sleep highlights the better correlation between SA and IEDs. Conclusions: Our data suggest that neural mechanisms involved in the generation of sleep spindles facilitate IEDs production in LKS. [source] Effect of intravenous lidocaine administration on laminar inflammation in the black walnut extract model of laminitisEQUINE VETERINARY JOURNAL, Issue 3 2010J. M. WILLIAMS Summary Reasons for performing study: Laminitis is a serious complication of horses suffering from sepsis/endotoxaemia-related events. Laminitis in horses and organ injury in human sepsis are both reported to involve inflammatory injury to the laminae/organs including early activation of endothelium and leucocytes leading to emigration of neutrophils into the tissue interstitium. In the black walnut extract (BWE) model, systemic inflammatory events coincide with marked increase in laminar mRNA concentrations of inflammatory genes including proinflammatory cytokines (i.e. IL-1,, IL-6), COX-2, chemokines (i.e. IL-8) and endothelial adhesion molecules (i.e. ICAM-1 and E-selectin). In models of human sepsis, i.v. lidocaine has been reported to decrease leucocyte and endothelial activation, and the expression of proinflammatory cytokines and chemokines. Objectives: To evaluate the effect of i.v. lidocaine therapy on the inflammatory processes documented to occur in the BWE model of laminitis. Methods: Twelve horses were administered BWE and treated immediately with either lidocaine (1.3 mg/kg bwt bolus, followed by 0.05 mg/kg bwt/min CRI, n = 6) or saline (n = 6) for 10 h. At 10 h post BWE administration, laminar samples were obtained under general anaesthesia for assessment of proinflammatory gene expression (using RT-qPCR) and leucocyte emigration (via CD13 immunohistochemistry). At 0, 3 and 10 h post BWE administration, skin samples were obtained for assessment of leucocyte emigration (via calprotectin immunohistochemistry). Results: No significant differences between groups were noted for inflammatory gene mRNA concentrations (IL-1,, IL-6, IL-8, COX-2) or for number of leucocytes present within the laminar interstitium or skin dermis. Increased (P<0.05) laminar E-selectin mRNA concentrations were present in the LD group (vs. SAL group). Conclusions: Continuous administration of i.v. lidocaine does not inhibit inflammatory events in either the laminae or skin in the horse administered black walnut extract. Potential relevance: This work questions the use of continuous i.v. administration of lidocaine as an effective anti-inflammatory therapy for systemic inflammation. [source] The ,Malbouffe' Saga La Saga de la ,Malbouffe' Die Saga von ,Malbouffe"EUROCHOICES, Issue 1 2007Alain Rérat summary The ,Malbouffe' Saga After the end of the Second World War, a marked increase in animal and plant production was observed in France, little by little considered by consumers to be obtained at the expense of product quality. The pejorative term ,malbouffe' soon emerged, in connection not only with the hygiene of food, but also with its organoleptic and technological characteristics. This article focuses on food safety in France, with special attention paid to the incidence of toxi-infections and food contaminations of biological and chemical origin. The Mad Cow outbreak is reviewed, along with its consequences for human health in the form of new variant Creutzfeldt-Jakob's disease. It is emphasized that food-related human mortality , almost exclusively due to biological contaminations , represented only 647 cases in 1995, i.e., 0.12 per cent of the overall mortality rate. The main contaminants were Salmonella, whose number is steadily decreasing, and Campylobacter, but parasite and phycotoxic risks are increasing. Mortality due to chemical contaminants is very low i.e., 10 cases or 0.002 per cent of overall mortality These contaminants, either accidental (dioxin, hydrocarbons, radioactive isotopes) or unavoidable (residues from phytochemicals, fertilisers) may be at the source of acute or chronic intoxications with sometimes unknown consequences. Nevertheless, food safety in France does not merit the spiteful term ,malbouffe'. Nach dem Ende des Zweiten Weltkriegs war in Frankreich im Bereich der Tier- und Pflanzenproduktion ein deutlicher Zuwachs zu beobachten, welcher in den Augen der Verbraucher zunehmend auf Kosten der Produktqualität erreicht wurde. Der abwertende Begriff ,Malbouffe" (in etwa ,schlechtes Essen") entstand bald darauf nicht nur im Hinblick auf die Nahrungsmittelhygiene, sondern auch in Bezug auf die organoleptischen und technologischen Eigenschaften der Nahrungsmittel. Dieser Beitrag konzentriert sich auf die Nahrungsmittelsicherheit in Frankreich unter besonderer Berücksichtigung der aufgetretenen Infektionen durch Giftstoffe und der Kontamination von Nahrungsmitteln biologischen und chemischen Ursprungs. Der BSE-Ausbruch und dessen Auswirkungen auf die Gesundheit des Menschen in Form von einer neuen Variante der Creutzfeldt-Jakob-Krankheit werden noch einmal betrachtet. Es wird hervor gehoben, dass die nahrungsmittelbedingte Sterblichkeit bei Menschen, die nahezu ausschließlich auf biologische Kontaminationen zurückzuführen ist, 1995 bei nur 647 Fällen lag, d.h. bei 0,12 Prozent der gesamten Sterblichkeitsrate. Die Nahrungsmittel wurden hauptsächlich durch Salmonellen (die Anzahl dieser Fälle nimmt kontinuierlich ab) und Campylobacter kontaminiert, die parasitären und phykotoxischen Risiken nehmen jedoch zu. Ce rapport se focalise uniquement sur la salubrité alimentaire en France, soulignant, en particulier, l'évolution de l'incidence des toxi-infections et des contaminations alimentaires d'origine biologique et chimique. Après avoir rappelé l'épizootie de la vache folle (1000 cas en France depuis 1996 et actuellement en cours d'extinction) et de ses conséquences sur la santé humaine (nouvelle variante de la maladie de Creutzfeldt-Jakob) limitées actuellement à 13 cas mortels dans notre pays, ce rapport précise que la mortalité humaine liée à l'alimentation , presque totalement due à des contaminations biologiques - ne représentait en 1995 que 647 cas, i.e. 0.12% de la mortalité générale. Pour l'essentiel, ces contaminants sont des salmonelles, en baisse constante, et des campylobacter, mais on peut craindre la progression des risques parasitaires et phycotoxiques, encore réduits actuellement. Oudit Abstract Background, Angiotensin converting enzyme 2 (ACE2), a monocarboxylase that degrades angiotensin II to angiotensin 1,7, is also the functional receptor for severe acute respiratory syndrome (SARS) coronavirus (SARS-CoV) and is highly expressed in the lungs and heart. Patients with SARS also suffered from cardiac disease including arrhythmias, sudden cardiac death, and systolic and diastolic dysfunction. Materials and methods, We studied mice infected with the human strain of the SARS-CoV and encephalomyocarditis virus and examined ACE2 mRNA and protein expression. Autopsy heart samples from patients who succumbed to the SARS crisis in Toronto (Canada) were used to investigate the impact of SARS on myocardial structure, inflammation and ACE2 protein expression. Results, Pulmonary infection with the human SARS-CoV in mice led to an ACE2-dependent myocardial infection with a marked decrease in ACE2 expression confirming a critical role of ACE2 in mediating SARS-CoV infection in the heart. The SARS-CoV viral RNA was detected in 35% (7/20) of autopsied human heart samples obtained from patients who succumbed to the SARS crisis during the Toronto SARS outbreak. Macrophage-specific staining showed a marked increase in macrophage infiltration with evidence of myocardial damage in patients who had SARS-CoV in their hearts. The presence of SARS-CoV in the heart was also associated with marked reductions in ACE2 protein expression. Conclusions, Our data show that SARS-CoV can mediate myocardial inflammation and damage associated with down-regulation of myocardial ACE2 system, which may be responsible for the myocardial dysfunction and adverse cardiac outcomes in patients with SARS. [source] Aberrant increase in the immature platelet fraction in patients with myelodysplastic syndrome: a marker of karyotypic abnormalities associated with poor prognosisEUROPEAN JOURNAL OF HAEMATOLOGY, Issue 1 2009Naomi Sugimori Abstract Objectives:, Some patients with myelodysplastic syndrome (MDS) show a marked increase in the percentage of immature platelet fraction (IPF%) despite the absence of severe thrombocytopenia. To determine the significance of such an unbalanced increase in the IPF%, we investigated the IPF% and other laboratory findings of 51 patients recently diagnosed with MDS. Method:, Subjects consisted of 80 healthy males, 90 healthy females, and 51 patients with MDS and 20 patients with idiopathic thrombocytopenic purpura (ITP). The IPF and IPF% were determined using a Sysmex XE-2100 system loaded with IPF Master software (XE IPF Master, Sysmex). Platelet counts were measured simultaneously. Results:, IPF% and platelet counts of these patients ranged from 1.1% to 25.1% (median, 5.3%) and from 6 to 260 × 109/L (median, 71 × 109/L), respectively. Twelve patients showed platelet counts more than 50 × 109/L with 10% or more IPF%. All of the 12 patients had chromosome abnormalities including monosomy 7 and complex abnormalities involving 7 or 5q. In the other 39 patients who did not show the aberrant IPF% increase, chromosomal abnormalities were seen only in seven patients and none of them had chromosome 7 abnormalities. The IPF% of two patients increased to more than 10% in association with the appearance of monosomy 7. Conclusions:, These findings suggest that a high IPF% in MDS patient may be a marker for karyotypic abnormalities with a poor prognosis, including chromosome 7 abnormalities. To determine the relative role of these subsets in disease exacerbation, we examined mice deficient in T-bet (T-bet,/,), which is required for Th1 differentiation and IFN-, production. We now report that immunization with soluble egg Ag in CFA caused a significantly greater enhancement of egg-induced hepatic immunopathology in T-bet,/, mice compared with WT controls, and analysis of their granulomas disclosed a higher proportion of activated DC and CD4+ T cells, as well as a marked influx of neutrophils. The absence of IFN-, in the T-bet,/, mice correlated with a marked increase in IL-23p19, IL-17 and TNF-, in granulomas and MLN. In contrast, T-bet,/, mice had lower levels of IL-4, IL-5 and IL-10 and a reduction in FIZZ1 and FoxP3 expression, suggesting diminished regulatory activity, respectively, by alternatively activated macrophages and Treg. These T cells, represented by the clone 178-4 Ts, restrict the Dex-specific IgG antibody repertoire such that the J558 idiotype dominates. Antibodies with other structures in the heavy-chain variable region (VH region), predominantly within the CDR3 domain, occur when the T cell control fails. This increase of antibody diversity caused by a lack of CD25+ Ts cells, e.g. in nude mice, does not result in the appearance of antibodies with enhanced affinity to the antigen Dex, but often leads to a crossreactivity with autologous proteins. Twenty-two out of sixty Dex-specific hybridomas from nude mice, but no hybridomas from euthymic mice, crossreact with a nuclear protein, as tested by ELISA. This nuclear protein was identified as histone H3. Ten of the sixty hybridomas from nude mice were sequenced and show VH sequences that deviate from the original J558 sequence. Three of these ten hybridomas crossreact with the histone H3. Adoptive transfer of CD25+ Ts cells to nude mice leads to a marked increase of antibodies carrying the original J558 idiotype within the IgG pool after immunization with Dex. Our data demonstrate a CD25+ Ts cell-mediated restriction of VH usage, which prevents the appearance of crossreactive autoantibodies. Cooper Abstract Growing evidence suggests that pharmacological inhibition of Na/H exchange and Na/HCO3 transport provides protection against damage or injury in cardiac ischemia. In this study, we examined the contribution of the sodium/bicarbonate cotransporter NBCn1 (slc4a7) to cytotoxicity in cultured hippocampal neurons of rats. In neurons exposed to extracellular pH (pHo) ranging from 6.2 to 8.3, NBCn1 protein expression increased by fivefold at pH < 6.5 compared to the expression at pHo 7.4. At pHo 6.5, the intracellular pH of neurons was ,1 unit lower than that at pH 7.4. Immunochemistry showed a marked increase in NBCn1 immunofluorescence in plasma membranes and cytosol of the soma as well as in dendrites, at pHo 6.5. NBCn1 expression also increased by 40% in a prolonged Mg2+ -free incubation at normal pHo. Knockdown of NBCn1 in neurons had negligible effect on cell viability. The effect of NBCn1 knockdown on cytotoxicity was then determined by exposing neurons to 0.5 mm glutamate for 10 min and measuring lactate dehydrogenase (LDH) release from neurons. Compared to normal incubation (pHo 7.2 for 6 h) after glutamate exposure, acidic incubation (pHo 6.3 for 6 h) reduced cytotoxicity by 75% for control neurons and 78% for NBCn1-knockdown neurons. Thus, both controls and knockdown neurons showed acidic protection from cytotoxicity. However, in Mg2+ -free incubation after glutamate exposure, NBCn1 knockdown progressively attenuated cytotoxicity. This attenuation was unaffected by acidic preincubation before glutamate exposure. We conclude that NBCn1 has a dynamic upregulation in low pHo and Mg2+ depletion. NBCn1 is not required for acidic protection, but increases cytotoxicity in Mg2+ -free conditions. Androulidakis Abstract Oscillatory synchronization in the beta (,20 Hz) band is a common feature of human motor control, manifest at cortical and muscular levels during tonic contraction. Here we test the hypothesis that the influence of visual feedback on performance in a positional hold task is increased during bursts of beta-band synchrony in the corticospinal motor system. Healthy subjects were instructed to extend their forefinger while receiving high-gain visual feedback of finger position on a PC screen. Small step displacements of the feedback signal were triggered either by bursts of beta oscillations in scalp electroencephalogram or randomly with respect to cortical beta activity, and the resulting positional corrections expressed as a percentage of the step displacement. Corrective responses to beta and randomly triggered step changes in visual feedback were 41.7 ± 4.9 and 31.5 ± 6.8%, respectively (P < 0.05). A marked increase in the coherence in the beta band was also found between muscle activity and cortical activity during the beta-triggered condition. The results suggest that phasic elevations of beta activity in the corticospinal motor system are associated with an increase in the gain of the motor response to visual feedback during a tonic hold task. Beta activity may index a motor state in which processing relevant to the control of positional hold tasks is promoted, with behavioural consequences. Animals were subjected to either 30 min of MCAO, which causes damage confined to the striatum, or 2 h of MCAO, which leads to both striatal and cortical infarction. When compared to sham-operated rats, MCAO-rats showed a marked increase of the number of cells double-labelled for 5-bromo-2,-deoxyuridine-5,-monophosphate (BrdU; injected during 4,6 days postischemia) and neuronal-specific antigen (NeuN; a marker of postmitotic neurons) in the ipsilateral dentate granule cell layer and subgranular zone at 5 weeks following the 2 h insult. Only a modest and variable increase of BrdU-labelled cells was found after 30 min of MCAO. The enhanced neurogenesis was not dependent on cell death in the hippocampus, and its magnitude was not correlated to the degree of cortical damage. Systemic administration of the N -methyl- d -aspartate (NMDA) receptor blocker dizocilpine maleate (MK-801) completely suppressed the elevated neurogenesis following 2 h of MCAO. Our findings indicate that stroke leads to increased neurogenesis in the adult rat dentate gyrus through glutamatergic mechanisms acting on NMDA receptors. This modulatory effect may be mediated through changes in the levels of several growth factors, which occur after stroke, and could influence various regulatory steps of neurogenesis. In adult rats, delayed dopaminergic neuron cell death induced by intrastriatal 6-hydroxydopamine injection led to a marked increase in the number of both c-Jun- and phosphorylated c-Jun-immunoreactive nuclei in the substantia nigra pars compacta. The response was maximal before any significant loss of nigral neurons could be detected (on day 7 post lesion) and was confined to the dopaminergic neurons. Similarly, 6-hydroxydopamine lesion of the striatal dopaminergic terminals or excitotoxic lesion of the striatal target neurons in neonatal rats resulted in an increased number of c-Jun- and phosphorylated c-Jun-immunoreactive nigral nuclei that preceded the loss of nigral dopaminergic neurons. By contrast, after an excitotoxic lesion of the striatal target neurons in the adult rat, resulting in atrophy but not cell death of the nigral dopaminergic neurons, no upregulation of either c-Jun or phosphorylated c-Jun was found. A single injection of 10 µg of glial-cell-line-derived-neurotrophic factor given at day 3 after the intrastriatal 6-hydroxydopamine lesion reduced the number of c-Jun- and phosphorylated c-Jun-immunoreactive nuclei in the substantia nigra and protected the dopaminergic neurons from the ensuing cell death. L. Wardrop The natural resistance associated macrophage protein 2 (Nramp2) is a transporter that is involved in iron (Fe) uptake from transferrin (Tf) and low molecular mass Fe complexes. Here we describe the effect of the inflammatory mediators interferon-, (IFN-,) and lipopolysaccharide (LPS) on the expression of Nramp2 mRNA and Fe uptake by cells of the macrophage lineage. After incubation of the RAW264.7 macrophage cell line with LPS there was a sevenfold increase in the expression of the 2.3 kb Nramp2 mRNA transcript when compared with the control, but little effect on the Nramp2 3.1 kb transcript. These results indicate differential regulation of the two transcripts. Treatment with LPS resulted in an increase in 59Fe uptake from 59Fe,nitrilotriacetic acid, while transferrin receptor (TfR) mRNA levels and 59Fe uptake from 59Fe,Tf were decreased. Paradoxically, at the same time, an increase in iron regulatory protein (IRP)1 RNA-binding activity was observed. Incubation with IFN-, (50 U·mL,1) resulted in a marked decrease in TfR mRNA levels but had no effect on Nramp2 mRNA expression. Exposure of RAW264.7 cells to both IFN-, and LPS resulted in a fourfold increase in the Nramp2 2.3-kb transcript and a four to fivefold decrease in the 3.1-kb transcript when compared with the control. Furthermore, there was a decrease in TfR mRNA levels despite an increase in IRP1 RNA-binding activity and a marked increase in inducible nitric oxide synthase mRNA expression. Hence, TfR and Nramp2 mRNA expression did not appear to be regulated in a concerted manner. Similar responses to those found above for RAW264.7 cells were also observed in the J774 macrophage cell line and also for primary cultures of mouse peritoneal macrophages. Apoptosis was observed in the spleen but not in the liver of infected mice, whereas the induction of liver necrosis was evident by rising levels of serum aminotransferases in these animals. Apoptosis was detected in the liver of L. monocytogenes -infected mice which had been treated with monoclonal antibody (mAb) against tumor necrosis factor-, (TNF-,) or interleukin-6 (IL-6), or in TNF-,,/, mice, but not in ,- interferon (IFN-,),/, mice or mice which had been treated with mAb against IL-4 or IL-10. Augmentation of liver apoptosis in mice treated with mAb against TNF-, or IL-6 or in TNF-,,/, mice correlated with the increase in bacterial numbers in the organ, while no augmentation of apoptosis was observed in the liver of IFN-,,/, mice irrespective of the marked increase in bacterial numbers in the organs, indicating that augmentation of liver apoptosis may not be merely due to the increase in bacterial growth in the organs. [source] Content and biosynthesis of polyamines in salt and osmotically stressed cells of Synechocystis sp.FEMS MICROBIOLOGY LETTERS, Issue 1 2003PCC 680 Abstract The effects of various NaCl and sorbitol concentrations in the growth medium on polyamine content and on two enzymes of the polyamine biosynthesis pathway, arginine decarboxylase (ADC) and S -adenosyl methionine decarboxylase (SAMDC), were investigated in the unicellular cyanobacterium Synechocystis sp. PCC 6803. Synechocystis cells showed no difference in growth rate when the concentration of NaCl was raised up to 550 mM. The growth rate decreased at 300 mM sorbitol, and complete inhibition of growth occurred at concentrations of ,700 mM sorbitol. Salt stress induced a moderate increase in the total cellular polyamine content, spermine in particular. Osmotic stress caused an apparent increase in the total cellular polyamine content with a marked increase of spermidine induced by 700 mM sorbitol. Importantly, a low level of spermine, which so far has never been detected in cyanobacteria, could be found in Synechocystis sp. PCC 6803. ADC, a key enzyme for putrescine synthesis, was unaffected by salt stress but showed a six-fold increase in enzyme activity upon osmotic stress imposed by 700 mM sorbitol. SAMDC, another important enzyme for spermidine and spermine synthesis, responded to salt and osmotic stresses similarly to the pattern observed for ADC. An analysis by reverse transcription-polymerase chain reaction revealed an increase of ADC mRNA level in cells under salt and osmotic stresses. Most importantly, the increase of ADC mRNA was attributed to its slower turnover rate under both stress conditions. Interestingly, the samdc gene(s) of Synechocystis appear to be unique since comparisons with known gene sequences from other organisms resulted in no homologous sequences identified in the Synechocystis genome. [source] Use of biological reference points for the conservation of Atlantic salmon, Salmo salar, in the River Lune, North West EnglandFISHERIES MANAGEMENT & ECOLOGY, Issue 1 2006M. W. APRAHAMIAN Abstract, The development and use of biological reference points (BRPs) for salmon, Salmo salar L., conservation on the River Lune, England were examined. The Lune supports recreational and net fisheries with annual catches averaging 1332 and 2146 salmon, respectively. Using models transported from other river systems, BRPs were developed that were exclusive to the Lune; specifically the number of eggs deposited and carrying capacity estimates for age 0+ and 1+ parr. The conservation limit was estimated at 11.9 million eggs, and to ensure that the conservation limit was exceeded 80% of the time, the management target was set at 14.4 million eggs (equivalent to ,5000 adults). Between 1989 and 1998 the management target was exceeded in only 2 years. Comparison of juvenile salmon densities in 1991 and 1997 with estimates of carrying capacity indicated that 0+ and 1+ parr densities were at around 60% of carrying capacity and may relate to the number of eggs deposited in 1990 and 1996 being approximately 70% of the target value. From, and including, the 2000 fishing season, regulations to ensure that the conservation limit is met 4 years out of 5 were introduced. These consisted of a reduction from 26 to 12 haaf nets, from 10 to seven drift nets and a four-fish bag limit for the rod fishery. In the period between 2000 and 2004 there was a marked increase in the estimated number of salmon spawning and the management target value of ,5000 spawning adults was exceeded in all years. There was also an increase in the juvenile salmon population. The estimated level of exploitation in the net and rod fisheries reduced from 29.9% to 13.8% and from 26.4% to 14.8% respectively, after the introduction of the regulations. However, the roles of the PI3K pathway in neurite branch formation were not fully understood. Also, it was not clear where the PI3K pathway is activated during branch formation. Results:, We found that the treatment of PC12 cells with the PI3K inhibitor LY294002 resulted in a marked increase in the number of neurite branch points, suggesting a suppressive role of PI3K in neurite branch formation. Expression of a constitutively active form of Akt, a downstream effector of PI3K, decreased the number of branch points, whereas that of a dominant-negative form of Akt increased it. In contrast, inhibition of neither Rac, mTOR nor GSK3, other effectors of PI3K, promoted branch formation. Importantly, the phosphorylated form of endogenous Akt was localized at the tips of growth cones, but devoid of small branches in NGF-treated PC12 cells. The Debeli Namet glacier in the Durmitor massif, Montenegro, is one of the lowest altitude glaciers (2050,2300 m) at this latitude (42,44°N) in the northern hemisphere. The glacier survives well below the climatological equilibrium line altitude because of substantial inputs from avalanching and windblown snow. The glacier survived two of the hottest summers on record in 2003 and 2007, although it experienced significant retreat. However, during the intervening years (2004,2006) the glacier increased in size and advanced, forming a new frontal moraine. This rapid advance was primarily in response to much cooler summer temperatures, close to or cooler than average, and a marked increase in winter precipitation. The rapid growth and decay of the Debeli Namet glacier in response to inter-annual climate variability highlights the sensitivity of small cirque glaciers to short-term climate change. Platelet count on admission was 5 × 103/µL, with a high serum concentration of platelet-associated immunoglobulin G. Bone marrow examination showed a marked increase in megakaryocytes. The patient was diagnosed with idiopathic thrombocytopenic purpura accompanied by chronic subdural hematoma. An increase in platelet count and gradual resolution of the subdural hematoma were obtained with prednisolone and azathioprine administration, but platelet count decreased when steroid tapering was attempted. Ultimately we administered the Chinese herbal medicine EK-49 and ascorbic acid, with a gradual increase in platelet count and no adverse effects. Some elderly patients with chronic subdural hematoma can be treated non-invasively. Further, a combination of EK-49 and ascorbic acid may be an effective treatment for refractory idiopathic thrombocytopenic purpura. [source] |
