    A New Species of Halophilous Collembola (Insecta) from Korea

    ABSTRACT Three species of interstitial Collembola in Entomobryidae were identified from South Korean coast. One of them, Sinella (Coecobrya) maritima, is new to science.     Targeting clusters of transferred genes in Thermotoga maritima

    Summary We screened a Thermotoga sp. strain RQ2 lambda library for genes present in that strain but absent from the closely related completely sequenced relative Thermotoga maritima strain MSB8, by using probes generated in an earlier genomic subtraction study. Five lambda insert fragments were sequenced, containing, respectively, an archaeal type ATPase operon, rhamnose biosynthetic genes, ORFs with similarity to an arabinosidase, a Thermotoga sp. strain RQ2-specific alcohol dehydrogenase and a novel archaeal Mut-S homologue. All but one of these fragments contained additional Thermotoga sp. strain RQ2-specific sequences not screened for, suggesting that many such strain-specific genes will be found clustered in the genome. Moreover, phylogenetic analyses, phylogenetic distribution and/or G + C content suggests that all the Thermotoga sp. strain RQ2 specific sequences in the sequenced lambda clones have been acquired by lateral gene transfer. We suggest that the use of strain-specific small insert clones obtained by subtractive hybridization to target larger inserts for sequencing is an efficient, economical way to identify environmentally (or clinically) relevant interstrain differences and novel gene clusters, and will be invaluable in comparative genomics.     An early temperature-sensitive period for the plasticity of segment number in the centipede Strigamia maritima

    SUMMARY Geophilomorph centipedes show variation in segment number (a) between closely related species and (b) within and between populations of the same species. We have previously shown for a Scottish population of the coastal centipede Strigamia maritima that the temperature of embryonic development is one of the factors that affects the segment number of hatchlings, and hence of adults, as these animals grow epimorphically,that is, without postembryonic addition of segments. Here, we show, using temperature-shift experiments, that the main developmental period during which embryos are sensitive to environmental temperature is surprisingly early, during blastoderm formation and before, or very shortly after, the onset of segmentation.     Temperature-dependent plasticity of segment number in an arthropod species: the centipede Strigamia maritima

    SUMMARY The evolution of arthropod segment number provides us with a paradox, because, whereas there is more than 20-fold variation in this character overall, most classes and orders of arthropods are composed of species that lack any variation in the number of segments. So, what is the origin of the higher-level variation? The centipede order Geophilomorpha is unusual because, with the exception of one of its families, all species exhibit intraspecific variation in segment number. Hence it provides an opportunity to investigate how segment number may change in a microevolutionary context. Here, we show that segment number can be directly altered by an environmental factor (temperature),this is the first such demonstration for any arthropod. The direction of the effect is such that higher temperature during embryogenesis produces more segments. This potentially explains an intraspecific cline in the species concerned, Strigamia maritima, but it does not explain how such a cline is translated into the parallel interspecific pattern of lower-latitude species having more segments. Given the plastic nature of the intraspecific variation, its link with interspecific differences may lie in selection acting on developmental reaction norms. [source]


    Expression of trunk Hox genes in the centipede Strigamia maritima: sense and anti-sense transcripts

    SUMMARY We report the coding sequence and embryonic expression of the four trunk Hox genes Antennapedia (Antp), Ultrabithorax (Ubx), abdominal-A (abd-A), and Abdominal-B (Abd-B) in the geophilomorph centipede Strigamia maritima. In geophilomorph centipedes, all leg-bearing segments (LBS) are generated during embryogenesis, allowing us to define expression in relation to the full extent of the forming trunk. Persistent Antp expression characterizes the maxillipedal (poison claw) segment, whereas all LBS express the three Hox genes Antp, Ubx, and abd-A. Abd-B is never detectably expressed in segmented tissue, but is restricted to a zone around the proctodaeum that contributes to the hindgut. Expression of all these Hox genes initiates in the unsegmented tissue of the blastodisc, with expression of Antp respecting a sharply defined anterior border before the appearance of morphological segmentation in the trunk. The accumulation of Hox gene transcripts is strongly modulated by the maturing segment pattern, suggesting regulatory interactions with multiple levels of the segment patterning machinery. For one of these genes, Ubx, we detect both sense and anti-sense transcripts. The anti-sense transcripts originate 3, to the Ubx coding sequence and overlap the homeobox exon; they are expressed earlier than the Ubx coding transcripts and persistently, in an axially restricted pattern comparable to but distinct from those of the Hox coding transcripts. The pattern of accumulation of Ubx sense and anti-sense transcripts is strikingly complementary, suggesting the possibility of anti-sense regulation of Ubx expression. [source]


    Early development and segment formation in the centipede, Strigamia maritima (Geophilomorpha)

    Summary Geophilomorph centipedes exhibit a number of unique characteristics that make them of particular developmental and evolutionary interest. Segment numbers in geophilomorphs are higher than in any other centipedes, ranging from 27 to 191. They may be constant within a species, presenting in extreme form the "counting" problem in development, or they may vary,a situation that provides us with the opportunity to study naturally occurring variation in segment numbers. All their segments are generated during embryogenesis, a situation unlike that in the more basal centipede orders, which generate only a fraction of their 15 trunk segments in the embryo and develop the rest postembryonically. Here we provide a foundation for further developmental studies of the Geophilomorpha, building on the one study that has been conducted to date, on the coastal species Strigamia maritima. Development begins with the migration of nuclei to the surface of the egg, which then condense to form an embryonic rudiment of more than 20,000 cells, covering an entire hemisphere. During early development, the embryo can be divided into two distinct areas: a large terminal disc of apparently undifferentiated tissue and the germ-band, which has a clear anteroposterior axis and differentiated segments. The germ-band forms from the anterior of the terminal disc and extends anteriorly as the disc contracts. New segments are formed at the posterior margin of the germ-band. Once the process of segmentation ends, the germ-band folds and sinks into the yolk. We note that the classic description of centipede development, by Heymons more than a century ago, contains a fundamental error in the identification of the axes and hence in the interpretation of early segmentation.     Characterization and mode of action of an exopolygalacturonase from the hyperthermophilic bacterium Thermotoga maritima

    An intracellular pectinolytic enzyme, PelB (TM0437), from the hyperthermophilic bacterium Thermotoga maritima was functionally produced in Escherichia coli and purified to homogeneity. PelB belongs to family 28 of the glycoside hydrolases, consisting of pectin-hydrolysing enzymes. As one of the few bacterial exopolygalacturonases, it is able to remove monogalacturonate units from the nonreducing end of polygalacturonate. Detailed characterization of the enzyme showed that PelB is highly thermo-active and thermostable, with a melting temperature of 105 °C and a temperature optimum of 80 °C, the highest described to date for hydrolytic pectinases. PelB showed increasing activity on oligosaccharides with an increasing degree of polymerization. The highest activity was found on the pentamer (1000 U·mg,1). In addition, the affinity increased in conjunction with the length of the oligoGalpA chain. PelB displayed specificity for saturated oligoGalpA and was unable to degrade unsaturated or methyl-esterified oligoGalpA. Analogous to the exopolygalacturonase from Aspergillus tubingensis, it showed low activity with xylogalacturonan. Calculations on the subsite affinity revealed the presence of four subsites and a high affinity for GalpA at subsite +1, which is typical of exo-active enzymes. The physiological role of PelB and the previously characterized exopectate lyase PelA is discussed. [source]


    Glucose-6-phosphate dehydrogenase from the hyperthermophilic bacterium Thermotoga maritima: expression of the g6pd gene and characterization of an extremely thermophilic enzyme

    Abstract The gene (open reading frame Tm1155, g6pd) encoding glucose-6-phosphate dehydrogenase (G6PD, EC 1.1.1.49) of the hyperthermophilic bacterium Thermotoga maritima was cloned and functionally expressed in Escherichia coli. The purified recombinant enzyme is a homodimer with an apparent molecular mass of 95 kDa composed of 60-kDa subunits. Rate dependence (at 80°C) on glucose-6-phosphate and NADP+ followed Michaelis,Menten kinetics with apparent Km values of 0.15 mM and 0.03 mM, respectively; apparent Vmax values were about 20 U mg,1. The enzyme also reduced NAD+ (apparent Km 12 mM, Vmax 12 U mg,1). The 1000-fold higher catalytic activity (kcat/Km) with NADP+ over NAD+ defines the G6PD as NADP+ specific in vivo. G6PD activity was competitively inhibited by NADPH with a Ki value of 0.11 mM. With a temperature optimum of 92°C the enzyme is the most thermoactive G6PD described. [source]


    Experiments on the susceptibility of conifers to Heterobasidion annosum in Great Britain

    During the period 1960,71, experimental plantings were established at three sites in western Britain that were infested with Heterobasidion annosum: Ceri in mid-Wales, Lael in north-west Scotland and Red Marley in the West Midlands of England. At each site a randomized block experiment involving at least four species was supplemented with an ancillary trial of other species. In two of the experiments various treatments were applied to the previous stand of trees before or at felling, but only stump removal reduced the amount of disease in the succeeding crops. At Ceri, the incidence of H. annosum in stems removed at first thinning was: Picea sitchensis, 14%; Pseudotsuga menziesii 11%; Pinus contorta 3% and Abies procera 1%. At Lael, the figures were Larix decidua 59%; P. menziesii 51%; Chamaecyparis lawsoniana 37%; Abies amabilis 33% and Tsuga heterophylla 21%. There was negligible disease in A. procera;Abies grandis and Pinus sylvestris. At Red Marley, the incidence of disease was: P. menziesii 28%; T. heterophylla 18%; A. grandis 7%; Picea abies 1% and Pinus nigra var. maritima 0%. In the ancillary trial at Lael, the incidence of H. annosum in P. sitchensis was 55% and in P. abies 16%. The mean height of colonization by H. annosum within the diseased stems removed at first thinning at Lael (age 21,22 years) was 2.1 m for L. decidua, 1.4 m for P. sitchensis and 1.3 m for P. abies. Armillaria sp. caused mortality and decay in two of the experiments and these data are also presented. The results are discussed in relation to other information on the susceptibility of these species to H. annosum in the UK and elsewhere. Das Auftreten der Krankheit im Folgebestand konnte nur durch die Rodung der Stümpfe reduziert werden.     Medium-term vegetation dynamics and their association with edaphic conditions in two Hungarian saline grassland communities

    Abstract Medium-term (5.5 years) changes in the cover of major species in "Artemisia saline puszta" (Ass) and "Pannonic Puccinellia limosa hollow" (PPlh) grassland communities in the Kiskunság region, Hungary, were monitored and analyzed in relation to abiotic factors (e.g. air temperature, precipitation, soil moisture, salinity and alkalinity). Soil salinity varied considerably, indicating leaching and desalinization of surface layers as the most typical process occurring in the region. Yearly average covers of Artemisia santonicum and Plantago maritima were negatively and positively related to surface soil salinity, respectively, in accordance with their salt tolerance. Multiple regression analysis showed soil pH and salinity to be the most important factors determining yearly average cover of plants at Ass. Increasing pH increased the cover of A. santonicum and P. maritima, but decreased the cover of Podospermum canum. Increasing salinity decreased the cover of A. santonicum and P. canum. At PPlh, pH of groundwater had a positive effect and the lakewater level had a negative effect on the cover of Puccinellia limosa. The results provide information on the ongoing changes in the soil properties and the resulting changes in plant composition in these Hungarian salt-affected grasslands. [source]


    Plant colonization after managed realignment: the relative importance of diaspore dispersal

    Summary 1Deliberate breaching of sea defences is frequently practised with the aim of restoring salt-marsh vegetation on previously embanked land. However, experience so far has shown that it may take several years before salt-marsh vegetation is fully established, and it is possible that limited diaspore dispersal plays a role in this. In order to ascertain whether salt-marsh development may be constrained by limited diaspore dispersal, we studied the dispersal of salt-marsh species by tidal water. 2From October 2001 to the end of March 2002 a total of 38 species, of which 18 were salt-marsh species, was trapped in a restoration site and adjacent marsh. Aster tripolium, Limonium vulgare, Puccinellia maritima, Salicornia spp., Spergularia media and Suaeda maritima were the most abundant salt-marsh species, with more than 3 diaspores m,2 trapped during the study period. 3For most species, the number of diaspores trapped was representative of their abundance in nearby vegetation. Hence, despite the potential for long-distance transport by tidal water, our results indicate a predominantly local dispersal of salt-marsh species. 4Synthesis and applications. For the restoration of salt-marsh vegetation after de-embankment, relatively rapid colonization may be expected from pioneer and low-marsh species, provided they are present in a nearby source area and the restoration site is at the appropriate altitude. The establishment of species absent from the adjacent marsh may be dependent on the presence of birds or humans as the main dispersal agents. Breaching of sea defences should preferably take place before or during September, in order to take advantage of the peak in dispersal of salt-marsh species in the first year after breaching.     Physiological and biochemical traits involved in the genotypic variability to salt tolerance of Tunisian Cakile maritima

    Abstract Cakile maritima (family: Brassicaceae) was collected from three provenances belonging to different bioclimatic stages (humid, semi arid and arid) in Tunisia to study their eco-physiological and biochemical responses to salinity. Seedlings were cultivated on inert sand for 20 days under NaCl treatments (0, 100, 200, 400 mm NaCl). Plant response to salinity was provenance- and salt-dependent. At 100 mm NaCl, growth parameters (leaf biomass, area, number per plant and relative growth rate) were improved in plants from Jerba (originating from arid bioclimatic stage) compared with the control, while growth was reduced in those from Tabarka (from humid area). High salt levels (400 mm NaCl) decreased the plant growth in the three provenances, but plants in Tabarka were the most salt sensitive. The relative salt tolerance of plants from Jerba and Bekalta provenances was associated with low levels of malondialdehyde as well as of electrolyte leakage and endoproteolytic activity. Salt reduced leaf hydration, the decrease in water content being dose-dependent and more pronounced in Tabarka. Increase in salinity led to significant increase in leaf succulence and decrease in leaf water potential, especially in Jerba plants. The plants from the latter displayed the highest leaf levels of Na+ and Cl,, proline, soluble carbohydrates, soluble proteins, and polyphenols. Overall, the higher salt tolerance of plants from Jerba provenance, and to a lower extent of those from Bekalta, may be partly related to their better capacity for osmotic adjustment and to limit oxidative damage when salt-challenged. Résumé Cakile maritima a été collecté (famille des Brassicaceae) dans trois provenances appartenant à des étages bioclimatiques différentes (humide, semi-aride et aride) de la Tunisie, dans le but d'étudier leurs réponses éco-physiologique et biochimique à la salinité. Des plantules ont été cultivées dans du sable inerte pendant vingt jours avec des doses croissantes de NaCl (0, 100, 200 et 400 mm NaCl). La réponse de Cakile maritima dépend de la provenance et de la salinité du milieu. A 100 mm de NaCl, les paramètres de croissance (biomasse, surface et nombre des feuilles par plante ainsi que le taux de la croissance relative) ont été améliorés chez Djerba (zone bioclimatique aride) par comparaison aux plantes témoins, tandis que la croissance a été réduite chez Tabarka (zone humide). A la plus forte dose de sel (400 mm), une réduction de la croissance des trois provenances a été enregistrée avec une nette sensibilité chez les plantes de la provenance Tabarka. La tolérance relative des deux provenances Djerba et Bekalta est associée à une faible teneur en malondialdéhyde ainsi qu'une fuite d'électrolyte et activité endo-protéolytique modérées. Le traitement salin a réduit l'hydratation des feuilles et cette diminution du contenu en eau est dose-dépendante et elle est plus prononcée chez Tabarka. En outre, l'augmentation de la salinité du milieu a entrainé une élévation de la succulence des feuilles concomitante à une diminution du potentiel hydrique notamment chez Djerba. Les plantes de cette dernière ont été les plus riches en Na+ et Cl - , en proline, carbohydrates, en protéines solubles et en polyphénols. En général, la tolérance au sel de la provenance Djerba, et à moindre degré Bekalta, est en partie reliée à la meilleure capacité d'ajustement osmotique et la limitation des dommages oxydatifs sous stress salin.     Vernalization requirement of wild beet Beta vulgaris ssp. maritima: among population variation and its adaptive significance

    Summary 1Seven populations of Beta vulgaris ssp. maritima (wild beet) situated along a latitudinal cline were studied for their vernalization requirement and its consequences for fitness. 2Various cold regimes were applied in glasshouses and experimental gardens with plants of different ages. Three additional experimental sites (on the French Mediterranean, Atlantic and North Sea coasts) situated near three of the sampled populations, and thus including a reciprocal transplant design, were used to evaluate the influence of latitude under natural conditions. Survival and plant size were measured over 3 years. 3The vernalization requirement was greater in plants from more northern origins. The level of cold required to allow flowering overcompensated for the colder springs, so that northern plants in northern sites flowered less than southern plants in southern sites. 4Young seedlings were more difficult to vernalize than plants that had already developed vegetative rosettes. 5Differences in vernalization requirement seem to be an adaptive response to spring temperatures and season length in a particular latitude. A whole winter vernalization almost always led to flowering in the subsequent year whatever the latitude or geographical origin. 6Plants from the Atlantic and Channel coasts showed the highest lifetime reproductive success at all sites. Southern populations were better adapted to disturbed habitats as shown by their higher first-year reproductive success. The North Sea population had a lower reproductive success than the Atlantic populations, even in its native environment. [source]


    Short-term epidemic dynamics in the Cakile maritima,Alternaria brassicicola host,pathogen association

    Summary 1Studies combining within- and among-population processes are crucial for understanding ecological and co-evolutionary dynamics in host,pathogen interactions. We report on work over an 18-month period involving multiple beach populations of the plant Cakile maritima and its fungal pathogen Alternaria brassicicola along the south-east coast of Australia. 2Results from permanent transects replicated on several beaches show that disease incidence and prevalence vary significantly with plant age, class and distance from the sea, as well as time during the season. Plant density is also positively related to disease levels. 3Results from three subregions indicate that disease persistence depends on survival of infected plants behind the foredunes of protected beaches. Population extinction was more likely on beaches with greater wind exposure and sea access, with the latter also related to colonization consistent with the dispersal of Cakile seeds via ocean currents. 4Although disease dynamics during the epidemic were similar across subregions, the severity of the epidemic varied significantly among these areas, suggesting that large-scale environmental factors may influence the timing and development of the epidemic. 5Estimates of synchrony in disease dynamics indicated that populations within a subregion were significantly correlated with respect to epidemic development. There was, however, no evidence for spatial synchrony in disease dynamics based on among-population covariances in disease prevalence and interbeach distances. Populations within a subregion were thus often at quite different phases of the epidemic at any given time. [source]


    Influence of environmental factors on the growth and interactions between salt marsh plants: effects of salinity, sediment and waterlogging

    Summary 1,Artificial environmental gradients were established in a series of pot experiments to investigate the effect of salinity, sediment type and waterlogging on the growth, and interactions between Spartina anglica and Puccinellia maritima. In each experiment, one environmental variable was manipulated and plants grown in pairwise combinations to examine the effect of the environmental factor on the intensity of intra- and interspecific interactions, quantified using the Relative Neighbour Effect (RNE) index. 2,Puccinellia was found to exert an asymmetric, one-way competitive dominance above ground over Spartina in experiments where gradients of sediment type and waterlogging were established. The intensity of the competition was highest in conditions with the least abiotic stress and lower or non-existent where stress was increased. 3,The intensity of the above-ground competition was greatest in loam and least in sand sediments. Reduction in competitive intensity in sand was accompanied by an increase in below-ground Spartina biomass and it is suggested that the production of rhizomes is a potential mechanism by which this species can expand vegetatively into areas without competition. 4,Interspecific competition on Spartina from Puccinellia also varied in intensity in the waterlogging experiment, being more intense in non-immersed treatments, where abiotic stress was reduced. 5,The competitive dominance of Puccinellia and the competition avoidance mechanism shown by Spartina in these experiments help to explain the successional interactions between the species along environmental gradients in natural salt marsh communities.     Effects of resource competition and herbivory on plant performance along a natural productivity gradient

    Summary 1,The effects of resource competition and herbivory on a target species, Triglochin maritima, were studied along a productivity gradient of vegetation biomass in a temperate salt marsh. 2,Transplants were used to measure the impact of grazing, competition and soil fertility over two growing seasons. Three parts of the marsh were selected to represent different successional stages; Triglochin reached local dominance at intermediate biomass of salt-marsh vegetation. At each stage, three competition treatments (full plant competition, root competition only, and no competition) and three grazing treatments (full grazing, no grazing on Triglochin, and no grazing on Triglochin or neighbours) were applied to both seedlings and mature plants. 3,Competition and herbivory reduced biomass and flowering of Triglochin. The impact of grazing was strongest at the stage with the lowest biomass, while both herbivory and competition had a significant impact at the stage with the highest biomass. When plants were protected from direct herbivory, competition operated at all three successional stages. 4,Grazing reduced light competition when vegetation biomass was low or intermediate, but at high biomass there was competition for light even when grazing occurred. Herbivore exclusion increased the effects of plant competition. Except at low biomass, the negative impact of plant competition on Triglochin performance was greater than the positive effect of not being grazed. 5,Grazing played a minor role in seedling survival and establishment which were largely controlled by competitive and facilitative effects. 6,Once established, the persistence of Triglochin will be determined largely by grazing. Intense grazing in the younger marsh and increasing competition for light in the older marsh will restrict the distribution to sites with intermediate biomass. [source]


    The role of spatio-temporal heterogeneity in the establishment and maintenance of Suaeda maritima in salt marshes

The effects of disturbance and microtopography on the organization and dynamics of plant communities were studied in a European salt marsh located in the Bay of Mont St. Michel, France. The existence of seed trapping mechanisms was also tested. The study took place in the lower and middle marsh plant communities dominated by the perennials Puccinellia maritima and Halimione portulacoides, respectively and associated with the annual Suaeda maritima. Three treatments were used in series of plots placed in each community: (1) vegetation removal and root destruction to a depth of 10 cm and refilling, (2) non-remnant herbicide treatment without vegetation removal and (3) creation of depressions (20 cm deep). These treatments were compared with adjacent control plots. The first year of the experiment showed that the perennials facilitated the establishment of Suaeda by trapping its seeds. Estimation of cover, density and biomass over 5 yr following the disturbances showed that in the first 2 yr Suaeda dominated the disturbed plots. Thereafter Suaeda was gradually eliminated by competitive exclusion after ca. 3 yr in the zone originally dominated by Puccinellia maritima and after 4 yr in the zone occupied by Halimione portulacoides. Depressions constituted refuge habitats for Suaeda by limiting competition with the perennials but also led to a high risk of mortality with temporal fluctuations in density. Despite a period of investigation limited to 5 yr, our study demonstrated that natural disturbances of various types occurred and influenced the dynamics of Suaeda, Halimione and Puccinellia. We deduced that natural disturbances and microtopography are responsible for the maintenance of the habitat in a state of non-equilibrium by favouring the establishment of both spatial and temporal environmental heterogeneity. These conditions appear to be particularly favourable for the maintenance of annual species such as Suaeda maritima. [source]


    Kinetics of TmHU binding to DNA as observed by optical tweezers

    Abstract The kinetics of binding for the histone-like protein TmHU (from Thermotoga maritima) to DNA is analyzed on a single molecule level by use of optical tweezers. For the reaction rate a pronounced concentration-dependence is found with an "all or nothing"-limit which suggests the cooperative nature of the binding-reaction. By analyzing the statistics of mechanically induced dissociation-events of TmHU from DNA multiple reaction sites are observed to become more likely with increasing TmHU concentration. This is interpreted as a hint for a secondary organizational level of the TmHU/DNA complex.     The enigmatic invasive Spartina densiflora: A history of hybridizations in a polyploidy context

    Abstract The aim of this study was to explore the origin of the invasive Spartina densiflora by analysing samples from the native region (South America) and from a recently colonized area (California). A combination of various molecular data (chloroplast and nuclear sequences, molecular fingerprint) and ploidy level estimations was used to answer the question whether the reticulate phylogenetic pattern previously detected in S. densiflora was restricted to California, or alternatively, whether a more ancient hybrid origin preceded formation of this species in its native area. We found that this species is heptaploid in both its native and introduced range. Identification of nuclear homeologous sequences indicate that this species has a reticulate origin in its native range, involving a lineage related to the hexaploid clade formed by S. alterniflora, S. foliosa, and S. maritima, and another lineage related to the sub-Antarctic endemic S. arundinacea that provided the chloroplast genome. The samples from California displayed similar multilocus patterns to the samples from Chile, supporting the hypothesis that this species originated on the southeast American coast (Argentina), from where it eventually spread to the west coast of South America (Chile) first and to the Northern Hemisphere (California) later.     Low level of gene flow from cultivated beets (Beta vulgaris L. ssp. vulgaris) into Danish populations of sea beet (Beta vulgaris L. ssp. maritima (L.) Arcangeli)

    Abstract Gene flow from sugar beets to sea beets occurs in the seed propagation areas in southern Europe. Some seed propagation also takes place in Denmark, but here the crop,wild gene flow has not been investigated. Hence, we studied gene flow to sea beet populations from sugar beet lines used in Danish seed propagation areas. A set of 12 Danish, two Swedish, one French, one Italian, one Dutch, and one Irish populations of sea beets, and four lines of sugar beet were analysed. To evaluate the genetic variation and gene flow, eight microsatellite loci were screened. This analysis revealed hybridization with cultivated beet in one of the sea beet populations from the centre of the Danish seed propagation area. Triploid hybrids found in this population were verified with flow cytometry. Possible hybrids or introgressed plants were also found in the French and Italian populations. However, individual assignment test using a Bayesian method provided 100% assignment success of diploid individuals into their correct subspecies of origin, and a Bayesian Markov chain Monte Carlo (MC MC) approach revealed clear distinction of individuals into groups according to their subspecies of origin, with a zero level of genetic admixture among subspecies. This underlines that introgression beyond the first hybridization is not extensive. The overall pattern of genetic distance and structure showed that Danish and Swedish sea beet populations were closely related to each other, and they are both more closely related to the population from Ireland than to the populations from France, the Netherlands, and Italy.     The linkage disequilibrium between chloroplast DNA and mitochondrial DNA haplotypes in Beta vulgaris ssp. maritima (L.): the usefulness of both genomes for population genetic studies

    Abstract The structure and evolution of the plant mitochondrial genome may allow recurrent appearance of the same mitochondrial variants in different populations. Whether the same mitochondrial variant is distributed by migration or appears recurrently by mutation (creating homoplasy) in different populations is an important question with regard to the use of these markers for population genetic analyses. The genetic association observed between chloroplasts and mitochondria (i.e. two maternally inherited cytoplasmic genomes) may indicate whether or not homoplasy occurs in the mitochondrial genome. Four-hundred and fourteen individuals sampled in wild populations of beets from France and Spain were screened for their mitochondrial and chloroplast polymorphisms. Mitochondrial DNA (mtDNA) polymorphism was investigated with restriction fragment length polymorphism (RFLP) and chloroplast DNA (cpDNA) polymorphism was investigated with polymerase chain reaction PCR,RFLP, using universal primers for the amplification. Twenty and 13 variants for mtDNA and cpDNA were observed, respectively. Most exhibited a widespread geographical distribution. As a very strong linkage disequilibrium was estimated between mtDNA and cpDNA haplotypes, a high rate of recurrent mutation was excluded for the mitochondrial genome of beets. Identical mitochondrial variants found in populations of different regions probably occurred as a result of migration. We concluded from this study that mtDNA is a tool as valuable as cpDNA when a maternal marker is needed for population genetics analyses in beet on a large regional scale. [source]


    Species identification of Alnus (Betulaceae) using nrDNA and cpDNA genetic markers

    Abstract One nuclear and three chloroplast DNA regions (ITS, rbcL, matK and trnH-psbA) were used to identify the species of Alnus (Betulaceae). The results showed that 23 out of all 26 Alnus species in the world, represented by 131 samples, had their own specific molecular character states, especially for three morphologically confused species (Alnus formosana, Alnus japonica and Alnus maritima). The discriminating power of the four markers at the species level was 10% (rbcL), 31.25% (matK), 63.6% (trnH-psbA) and 76.9% (ITS). For ITS, the mean value of genetic distance between species was more than 10 times the intraspecific distance (0.009%), and 13 species had unique character states that differentiated them from other species of Alnus. The trnH-psbA region had higher mean values of genetic distance between and within species (2.1% and 0.68% respectively) than any other region tested. Using the trnH-psbA region, 13 species are distinguished from 22 species, and seven species have a single diagnostic site. The combination of two regions, ITS and trnH-psbA, is the best choice for DNA identification of Alnus species, as an improvement and supplement for morphologically based taxonomy. This study illustrates the potential for certain DNA regions to be used as novel internet biological information carrier through combining DNA sequences with existing morphological character and suggests a relatively reliable and open taxonomic system based on the linked DNA and morphological data.     Genetic variation and biogeography of Mertensia maritima (Boraginaceae)

    To elucidate the population dynamics and biogeographic connections of Mertensia maritima in the North Atlantic we studied the genetic variation within and among 16 local populations in the Oslofjord region (south-east Norway), Finnmark (northern Norway), Iceland and Svalbard, using starch gel isozyme electrophoresis. We also tested the self-compatibility experimentally. Mertensia maritima was found to be self-compatible and able to self-pollinate by autodeposition. The total level of genetic variation was extremely low. All the screened individuals were homozygous at all loci. All individuals from the populations of mainland Norway and Iceland were isozymatically identical, whereas individuals from Svalbard differed from these at three loci (AAT-1, PGM-3 and SKD-2). The genetic identity between any population at Svalbard and any population on the mainland was 0.86. These genetic patterns may have resulted from a combination of inbreeding, frequent local population extinctions, and long-distance dispersal. The isozymatic distinctness of the Svalbard populations is consistent with the arctic distribution of the morphologically based var. tenella, here raised to subspecific rank as Mertensia maritima ssp. tenella.     Interspecific and intraspecific interactions between salt marsh plants: integrating the effects of environmental factors and density on plant performance

    There has been much debate about the role of plant interactions in the structure and function of vegetation communities. Here the results of a pot experiment with controlled environments are described where three environmental variables (nutrients, sediment type and waterlogging) were manipulated factorially to identify their effects on the growth and intensity of interactions occurring between Spartina anglica and Puccinellia maritima. The two species were grown in split-plot planting treatments, representing intraspecific and interspecific addition series experiments, to determine individual and interactive effects of environmental factors and plant interactions on plant biomass. Above-ground growth of both species involved interactions between the environmental and planting treatments, while below-ground, environmental factors affected the biomass irrespective of planting treatments. It was suggested that this difference in growth response is evidence that in our experiment plant interactions between the two species occur primarily at the above-ground level. The intensity of plant interactions varied in a number of ways. First, interactions between Spartina and Puccinellia were distinctly asymmetrical, Puccinellia exerting a competitive effect on Spartina, with no reciprocal effect, and with a facilitative effect of Spartina on Puccinellia in low nutrient conditions. Second, the interactions varied in intensity in different environmental conditions. Interspecific competitive effects of Puccinellia on Spartina were more intense in conditions favourable to growth of Puccinellia and reduced or non-existent in environments with more abiotic stress. Third, intraspecific competition was found to be less intense for both species than interspecific interactions. Finally, the intensity of plant interactions involving both species was more intense above ground than below ground, with a disproportionate reduction in the intensity of interspecific competition below relative to above ground in treatments with less productive sediments and greater immersion. This is interpreted as reflecting a potential mechanism by which Spartina may be able to evade competitive neighbours. [source]


    Osmoprotectant , -alanine betaine synthesis in the Plumbaginaceae: S -adenosyl- l -methionine dependent N -methylation of , -alanine to its betaine is via N -methyl and N,N -dimethyl , -alanines

    , -Alanine betaine is an osmoprotective compound accumulated by most members of the plant family Plumbaginaceae. Leaf and root tissues of Limonium latifolium known to accumulate , -alanine betaine readily convert supplied , -alanine to , -alanine betaine. To identify the intermediates and the enzymes involved in , -alanine betaine synthesis, radiotracer experiments using [ C] formate were employed. These studies demonstrate that , -alanine betaine is synthesized from , -alanine via N -methyl and N,N- dimethyl , -alanines. A rapid and sensitive radiometric assay was developed to measure N -methyltransferase (NMT) activities by using [methyl- 14C] or [methyl- 3H] S -adenosyl- l -methionine (AdoMet) as the methyl donor. Leaf extracts from , -alanine betaine accumulators ,Armeria maritima, L. latifolium and L. ramosissimum, had detectable NMT activities while none were found in L. perezii, a species that does not accumulate , -alanine betaine. The NMT activities were further characterized from the leaves of L. latifolium. The activities had a pH optimum of 8.0, were soluble and inhibited by S -adenosyl- l -homocysteine. Extractable activities were similar from plants grown under control and salinity stress conditions. Radiolabeling with [ C] l -aspartic acid indicated that, unlike in bacteria, decarboxylation of l -aspartic acid is not the source of , -alanine in the Plumbaginaceae. [source]


    In vitro leaf inoculation studies as an indication of tree foliage susceptibility to Phytophthora ramorum in the UK

    Leaves of 11 coniferous and 23 broad-leaved tree species important to UK forestry were tested for their susceptibility to the quarantine pathogen Phytophthora ramorum using a detached leaf assay. Two European and two USA isolates were used. Wounded and unwounded leaves were dipped in zoospore suspensions during summer; conifers were also tested in winter. Successful infection of tissue and amount of necrosis were assessed. Highly susceptible broad-leaved hosts included Aesculus hippocastanum, Fraxinus excelsior, Quercus ilex, Ulmus procera and, to a lesser extent, Castanea sativa, Q. cerris and Q. petraea, together with Umbellularia californica and rhododendrons. Acer pseudoplatanus, Alnus glutinosa, Carpinus betulus, Corylus avellana, Fagus sylvatica, Prunus avium, Q. robur, Q. rubra and Q. suber showed consistently low susceptibility. Conifer species including Abies procera, Picea abies, P. sitchensis, Pseudotsuga menziesii, Sequoia sempervirens and Tsuga heterophylla were also susceptible. Pseudotsuga menziesii and A. procera were severely affected. Pinus contorta, P. nigra var. maritima and P. sylvestris were virtually resistant, while Taxus baccata was only slightly affected. Increased necrosis was apparent on leaves that were wounded prior to inoculation. These results extend the known range of trees that P. ramorum is able to attack and confirm its relative host-nonspecificity. [source]


    Variation in pathogen aggressiveness within a metapopulation of the Cakile maritima,Alternaria brassicicola host,pathogen association

    Variation in aggressiveness and its consequences for disease epidemiology were studied in the Cakile maritima,Alternaria brassicicola host,pathogen association. Variability in pathogen growth rates and spore production in vitro, as well as disease severity and lesion growth rate on C. maritima in glasshouse inoculation trials, were investigated. Substantial variation was found in growth rates among individual A. brassicicola isolates, as well as among pathogen populations. A significant trade-off also existed between growth and spore production, such that faster-growing isolates produced fewer spores per unit area. While there was little evidence for a link between growth in vitro and either disease severity or lesion development among fast- vs slow-growth isolate classes at the individual isolate level, the results suggest that variation in pathogen fitness components associated with aggressiveness may influence disease dynamics in nature. An analysis using an independent data set of disease prevalence in the associated host populations found a significant positive relationship between the average growth rate of pathogen populations in vitro and disease progress over the growing season in wild host populations. Trade-offs such as those demonstrated between growth rate and spore production may contribute to the maintenance of variation in quantitatively based host,pathogen interactions. [source]


    Estuarine Restoration of Submersed Aquatic Vegetation: The Nursery Bed Effect

    The historic decline of submersed aquatic vegetation (SAV) in mesohaline regions of Chesapeake Bay, United States involved a diversity of plant species. The recent modest recovery is mostly, however, associated with a single, prolific but ephemeral species, Ruppia maritima. Two previously abundant and more stable species, Potamogeton perfoliatus and Stuckenia pectinata, have shown virtually no evidence of recovery. Based on previous studies that demonstrated the ability of R. maritima stands to enhance water clarity and nutrient conditions for SAV growth, we hypothesized that these beds would serve as effective "nursery" areas to incite transplant success for other SAV. We conducted experiments in a two-phase study at small and large spatial scales designed to explore this "nursery effect" as a restoration approach to increase plant species diversity. The first phase was conducted at small spatial scales to test effects of patch density by planting P. perfoliatus and S. pectinata into bare, sparse, and densely vegetated areas within three similar R. maritima beds in a tributary of Chesapeake Bay. Mean seasonal percent survivorship and shoot density were significantly higher in bare patches compared to vegetated patches. In the second phase of the study, P. perfoliatus was transplanted into separate R. maritima beds of different densities to test the effect of bed scale plant density on P. perfoliatus survival and growth. Transplant success of P. perfoliatus was positively correlated with the density of R. maritima among all sites. [source]


    Vegetation change in a man-made salt marsh affected by a reduction in both grazing and drainage

    Abstract. In order to restore natural salt marsh in a 460-ha nature reserve established in man-made salt marsh in the Dollard estuary, The Netherlands, the artificial drainage system was neglected and cattle grazing reduced. Vegetation changes were traced through two vegetation surveys and monitoring of permanent plots over 15 yr after the management had been changed. Exclosure experiments were started to distinguish grazing effects from effects of increased soil waterlogging caused by the neglect of the drainage system. Both vegetation surveys and permanent plots demonstrated a dichotomy in vegetation succession. The incidence of secondary pioneer vegetation dominated by Salicornia spp. and Suaeda maritima increased from 0 to 20%, whereas the late-successional (Phragmites australis) vegetation from 10 to 15%. Grazing intensity decreased towards the sea. The grazed area contracted landward, which allowed vegetation dominated by tall species to increase seaward. Grazing and increased waterlogging interacted in several ways. The impact of trampling increased, and in the intensively grazed parts soil salinity increased. This can probably be explained by low vegetation cover in spring. Framework Ordination, an indirect-gradient-analysis technique, was used to infer the importance of environmental factors in influencing changes in species composition. Many changes were positively or negatively correlated with soil aeration and soil salinity, whereas elevation was of minor importance. Grazing accounted for only a few changes in species frequency. Changes in permanent plots were greater during the first than during the second half of the study period. In exclosures that were installed halfway through the study period, there was a relatively rapid recovery of previously dominant species that had decreased during the first half of the study period. Species richness per unit area in the reserve increased. At the seaward side of the marsh, the altered management allowed succession to proceed leading to establishment of stands of Phragmites australis, whereas on the landward side, the combination of moderate grazing with neglect of the drainage system appeared an effective measure in maintaining habitats for a wider range of halophytic species.     Structure of nondiscriminating glutamyl-tRNA synthetase from Thermotoga maritima

    Aminoacyl-tRNA synthetases produce aminoacyl-tRNAs from the substrate tRNA and its cognate amino acid with the aid of ATP. Two types of glutamyl-tRNA synthetase (GluRS) have been discovered: discriminating GluRS (D-GluRS) and nondiscriminating GluRS (ND-GluRS). D-GluRS glutamylates tRNAGlu only, while ND-GluRS glutamylates both tRNAGlu and tRNAGln. ND-GluRS produces the intermediate Glu-tRNAGln, which is converted to Gln-tRNAGln by Glu-tRNAGln amidotransferase. Two GluRS homologues from Thermotoga maritima, TM1875 and TM1351, have been biochemically characterized and it has been clarified that only TM1875 functions as an ND-GluRS. Furthermore, the crystal structure of the T. maritima ND-GluRS, TM1875, was determined in complex with a Glu-AMP analogue at 2.0,Å resolution. The T. maritima ND-GluRS contains a characteristic structure in the connective-peptide domain, which is inserted into the catalytic Rossmann-fold domain. The glutamylation ability of tRNAGln by ND-GluRS was measured in the presence of the bacterial Glu-tRNAGln amidotransferase GatCAB. Interestingly, the glutamylation efficiency was not affected even in the presence of excess GatCAB. Therefore, GluRS avoids competition with GatCAB and glutamylates tRNAGln. [source]
