Selected AbstractsImpact of Chronic Alcohol Ingestion on Cardiac Muscle Protein ExpressionALCOHOLISM, Issue 7 2010Rachel L. Fogle Background:, Chronic alcohol abuse contributes not only to an increased risk of health-related complications, but also to a premature mortality in adults. Myocardial dysfunction, including the development of a syndrome referred to as alcoholic cardiomyopathy, appears to be a major contributing factor. One mechanism to account for the pathogenesis of alcoholic cardiomyopathy involves alterations in protein expression secondary to an inhibition of protein synthesis. However, the full extent to which myocardial proteins are affected by chronic alcohol consumption remains unresolved. Methods:, The purpose of this study was to examine the effect of chronic alcohol consumption on the expression of cardiac proteins. Male rats were maintained for 16 weeks on a 40% ethanol-containing diet in which alcohol was provided both in drinking water and agar blocks. Control animals were pair-fed to consume the same caloric intake. Heart homogenates from control- and ethanol-fed rats were labeled with the cleavable isotope coded affinity tags (ICATÔ). Following the reaction with the ICATÔ reagent, we applied one-dimensional gel electrophoresis with in-gel trypsin digestion of proteins and subsequent MALDI-TOF-TOF mass spectrometric techniques for identification of peptides. Differences in the expression of cardiac proteins from control- and ethanol-fed rats were determined by mass spectrometry approaches. Results:, Initial proteomic analysis identified and quantified hundreds of cardiac proteins. Major decreases in the expression of specific myocardial proteins were observed. Proteins were grouped depending on their contribution to multiple activities of cardiac function and metabolism, including mitochondrial-, glycolytic-, myofibrillar-, membrane-associated, and plasma proteins. Another group contained identified proteins that could not be properly categorized under the aforementioned classification system. Conclusions:, Based on the changes in proteins, we speculate modulation of cardiac muscle protein expression represents a fundamental alteration induced by chronic alcohol consumption, consistent with changes in myocardial wall thickness measured under the same conditions. [source] The effect of GHRH antagonists on human glioblastomas and their mechanism of action,INTERNATIONAL JOURNAL OF CANCER, Issue 10 2010Eva Pozsgai Abstract The effects of new growth hormone-releasing hormone (GHRH) antagonists JMR-132 and MIA-602 and their mechanism of action were investigated on 2 human glioblastoma cell lines, DBTRG-05 and U-87MG, in vitro and in vivo. GHRH receptors and their main splice variant, SV1 were found on both cell lines. After treatment with JMR-132 or MIA-602, the cell viability decreased significantly. A major decrease in the levels of phospho-Akt, phospho-GSK3, and phosho-ERK 1/2 was detected at 5 and 10 min following treatment with the GHRH antagonists, whereas elevated levels of phospho-p38 were observed at 24 hr. The expression of caspase-3 and poly(ADP-ribose) (PARP), as the downstream executioners of apoptosis were found to be significantly elevated after treatment. Following treatment of the glioblastoma cells with GHRH antagonists, nuclear translocation of apoptosis inducing factor (AIF) and Endonuclease G (Endo G) and the mitochondrial release of cytochrome c (cyt c) were detected, indicating that the cells were undergoing apoptosis. In cells treated with GHRH antagonists, the collapse of the mitochondrial membrane potential was shown with fluorescence microscopy and JC-1 membrane potential sensitive dye. There were no significant differences between results obtained in DBTRG-05 or U-87MG cell lines. After treatment with MIA-602 and JMR-132, the reduction rate in the growth of DBTRG-05 glioblastoma, xenografted into nude mice, was significant and tumor doubling time was also significantly extended when compared with controls. Our study demonstrates that GHRH antagonists induce apoptosis through key proapoptotic pathways and shows the efficacy of MIA-602 for experimental treatment of glioblastoma. [source] Heathland restoration in The Netherlands: Effects of turf cutting depth on germination of Arnica montanaAPPLIED VEGETATION SCIENCE, Issue 2 2003Leon J.L. van der Berg Abstract. Germination experiments were conducted in a heathland after turf cutting and in a climate chamber to investigate the effects of turf cutting depth, aluminium toxicity and aluminium detoxification by humic acids and base cations on the germination and establishment of Arnica montana. Turfs were cut at three different depths, creating a gradient from organic to mineral soils. Germination and establishment of A. montana were negatively correlated with turf cutting depth. The removal of organic matter resulted in a major decrease in organic fraction of the soil and its nutrients. It also resulted in a considerable decrease in moisture content and humic acids. Additional liming after turf cutting increased germination and establishment in all plots and at all depths. Germination experiments under controlled conditions in a climate chamber revealed a significantly higher germination at low aluminium/calcium (Al:Ca) ratios. High Al:Ca ratios resulted in poor germination, suggesting Al toxicity. Addition of humic acids increased germination, even at high Al:Ca ratios, suggesting immobilization of Al by humic acids. It is concluded that turf cutting can have a marked effect on the success of heathland restoration. It results in the intended removal of the eutrophic layer but also in the unintentional removal of much of the buffering mechanisms and/or Al immobilizing compounds. Additional buffering and/or less deep turf cutting may be necessary to allow germination and establishment of rare herbaceous species such as A. montana. [source] ,Paper Clinics', a model for improving delivery of outpatient colorectal servicesCOLORECTAL DISEASE, Issue 4 2004T. R. C. Porrett Abstract Objectives The overburdening of colorectal out-patient clinics necessarily leads to delays in time from referral to consultation and subsequent clinic attendance. This study aimed to ascertain the feasibility of ,paper clinic' follow-up rather than all patients receiving a routine follow-up appointment following investigation. A more efficient outpatient follow-up process should reduce unnecessary follow-up, thereby facilitating the speedy investigation and diagnosis of patients through changes in clinic profiles. Methods From August 2001 all patients seen in the outpatient clinic of one (part time) Consultant colorectal surgeon, who required investigation, were prospectively recorded on a ,paper clinic' form. These patients were given the necessary test request forms but were not given a further outpatient appointment. The results of the investigations were reviewed, together with the patients' medical records at a formal fortnightly ,paper clinic' session carried out by the Consultant and Nurse Consultant, and a treatment plan derived. Patients then followed one of 5 follow-up pathways and were notified in writing with a copy to their GP. Results During a 24-month period a total of 897 patients were reviewed using the ,paper clinic' follow-up system. Of these, 285 (31.8%) patients were discharged without further follow-up. In a given 3-month period when the clinic was well established, 152 patients were reviewed, of whom 27% were discharged from follow-up, 17% received SOS appointments, 13% required further investigation (and consequently were returned to ,paper clinic' follow-up), and 7% received Nurse led follow-up. In this 3-month period 64% of patients reviewed by ,paper clinic' follow-up did not return to Surgical Outpatient's and 12% received a Surgical Outpatient appointment for review. Conclusion ,Paper clinic' follow-up is an effective and feasible follow-up alternative, resulting in a major decrease in outpatient follow-up burden. This has allowed the redesign of the outpatient clinic profile allowing for an increase in new urgent slots, and more rapid clinic follow up review of those patients who need it. Re-design and rationalization of existing services can result in considerable service improvement. Expanding clinics should not be considered the only option when faced with capacity and demand issues. [source] LIVE-BIRTH IN VIPERS (VIPERIDAE) IS A KEY INNOVATION AND ADAPTATION TO GLOBAL COOLING DURING THE CENOZOICEVOLUTION, Issue 9 2009Vincent J. Lynch The identification of adaptations and key innovations has long interested biologists because they confer on organisms the ability to exploit previously unavailable ecological resources and respond to novel selective pressures. Although it can be extremely difficult to test for the effects of a character on the rate of lineage diversification, the convergent evolution of a character in multiple lineages provides an excellent opportunity to test for the effect of that character on lineage diversification. Here, I examine the effect of parity mode on the diversification of vipers, which have independently evolved viviparity in at least 13 lineages. I find strong statistical evidence that viviparous species diversify at a greater rate than oviparous species and correlate major decreases in the diversification rate of oviparous species with periods of global cooling, such as the Oligocene. These results suggest that the evolution of viviparity buffered live-bearing species against the negative effects of global climate change during the Cenozoic, and was a key innovation in the evolution and diversification of live-bearing vipers. Dormann Summary 1,The effects of global climate change are predicted to be strongest in the Arctic. This, as well as the suitability of tundra as a simple model ecosystem, has led to many field experiments investigating consequences of simulated environmental change. 2,On the basis of 36 experiments reviewed here, minor light attenuation by clouds, small changes in precipitation, and increases in UV-B radiation and atmospheric CO2 concentrations will not affect arctic plants in the short term. However, temperature elevation, increases in nutrient availability and major decreases in light availability will cause an immediate plant-growth response and alter nutrient cycling, possibly creating positive feedbacks on plant biomass. The driver of future change in arctic vegetation is likely to be increased nutrient availability, arising for example from temperature-induced increases in mineralization. 3,Arctic plant species differ widely in their response to environmental manipulations. Classification into plant functional types proved largely unsatisfactory for generalization of responses and predictions of effects. 4,Nevertheless, a few generalizations and consistent differences between PFTs were detected. Responses to fertilization were the strongest, particularly in grasses. Shrubs and grasses were most responsive to elevated temperature. 5,Future studies should focus on interactive effects of environmental factors, investigate long-term responses to manipulations, and incorporate interactions with other trophic levels. With respect to plant functional types, a new approach is advocated, which groups species according to their responses to environmental manipulations. [source] |
