Side population cells expressing ABCG2 in human adult dental pulp tissue

Abstract Aim, To investigate the presence of side population (SP) cells by the Hoechst exclusion method in human adult dental pulp tissue. Methodology, Human adult dental pulp-derived cells were generated from third molar teeth. The cells were stained with Hoechst 33342 and sorted into SP cells or non-SP cells [main population (MP) cells]. Both cell types were compared with cell growth and RT,PCR analyses. Results, SP cells that express ABCG2, Nestin, Notch-1 and ,-smooth muscle actin were found at frequencies ranging from 0.67% to 1.02%. This SP profile disappeared in the presence of verapamil. These SP cells expressed dentine sialophosphoprotein and dentine matrix protein-1 when cultured in osteogenic medium. Conclusion, Human adult dental pulp tissue contains SP cells that differentiate into odontoblast-like cells. [source]


The effects of telomerase inhibition on prostate tumor-initiating cells

Abstract Prostate cancer is the most common malignancy in men, and patients with metastatic disease have poor outcome even with the most advanced therapeutic approaches. Most cancer therapies target the bulk tumor cells, but may leave intact a small population of tumor-initiating cells (TICs), which are believed to be responsible for the subsequent relapse and metastasis. Using specific surface markers (CD44, integrin ,2,1 and CD133), Hoechst 33342 dye exclusion, and holoclone formation, we isolated TICs from a panel of prostate cancer cell lines (DU145, C4-2 and LNCaP). We have found that prostate TICs have significant telomerase activity which is inhibited by imetelstat sodium (GRN163L), a new telomerase antagonist that is currently in Phase I/II clinical trials for several hematological and solid tumor malignancies. Prostate TICs telomeres were of similar average length to the telomeres of the main population of cells and significant telomere shortening was detected in prostate TICs as a result of imetelstat treatment. These findings suggest that telomerase inhibition therapy may be able to efficiently target the prostate TICs in addition to the bulk tumor cells, providing new opportunities for combination therapies. Extra-pair paternity in waved albatrosses

Abstract We estimated the rate of extra-pair fertilizations (EPFs) in waved albatrosses (Phoebastria irrorata) on Isla Española, Galápagos, Ecuador, using multilocus minisatellite DNA fingerprinting. Waved albatrosses are socially monogamous, long-lived seabirds whose main population is on Española. Aggressive extra-pair copulation (EPC) attempts have been observed in the breeding colony during the days preceding egg-laying. Our genetic analyses of 16 families (single chicks and their attending parents) revealed evidence of EPFs in four families. In all cases males were the excluded parent. These data suggest that waved albatrosses have an unusually high rate of EPF relative to taxa with similar life histories. Future behavioural observations will determine the extent to which forced vs. unforced EPCs contribute to this high EPF rate. [source]


The stellar content of the isolated transition dwarf galaxy DDO210,

ABSTRACT We use Subaru Suprime-Cam and VLT FORS1 photometry of the dwarf galaxy DDO210 to study the global stellar content and structural properties of a transition-type galaxy (with properties intermediate between dwarf irregular and dwarf spheroidal systems). This galaxy is sufficiently isolated that tidal interactions are not likely to have affected its evolution in any way. The colour,magnitude diagrams of DDO210 show a red giant branch (RGB) population (with an RGB bump), a bright asymptotic giant branch population, a red clump, young main-sequence stars and blue-loop stars. The youngest stars formed within the last 60 Myr and have a distinct radial distribution compared to the main population. Whereas the overall stellar spatial distribution and H i spatial distribution are concentric, the young stars are offset from the centre of DDO210 and are coincident with a ,dent' in the H i distribution. The implied recent star formation rate required to form the young population is significantly higher than the derived current star formation rate, by a factor of >10. Most of the stars in DDO210 are found in a red clump, and its mean I -band magnitude suggests that the majority of stars in DDO210 have an average age of 4+2,1 Gyr. Given this age, the colour of the RGB implies a mean metallicity of [Fe/H],,1.3. By comparing the shape of the red clump with models for a variety of star formation histories, we estimate that an old (>10 Gyr) stellar population can contribute ,20,30 per cent of the stars in DDO210 at most. The unusual star formation history of DDO210, its low-mass estimate and its isolated nature, provide insight into how star formation proceeds in the lowest mass, unperturbed, dwarf galaxy haloes. Genetic diversity and structure of the West Balkan Pramenka sheep types as revealed by microsatellite and mitochondrial DNA analysis

Summary Several different phenotypes of the native Pramenka sheep have been developed in the Balkan region for different environmental and socio-cultural conditions. Animals from seven West Balkan Pramenka sheep types were analysed for 15 microsatellite markers and for mitochondrial DNA (mtDNA) and the results were used to assess genetic variation within and among the types and to infer the genetic population structure of the Pramenka sheep. Mean expected heterozygosity and allelic richness over the microsatellite loci and sheep types were 0.78 and 7.9, respectively. A Bayesian statistical method for estimating hidden genetic structure suggested that a core of the largest panmictic population was formed by Serbian, Kosovan, Bosnian, Montenegrin and Albanian types, while Croatian and Macedonian types comprised two other main populations, respectively. Mitochondrial DNA analysis revealed two mtDNA haplogroups in the Pramenka sheep, B and A, with a frequency of 93.7% and 6.3%, respectively. A total of 60 mtDNA haplotypes were found in 64 animals sequenced, and the mean nucleotide and haplotypic diversities over the types were 0.013 and 0.945, respectively. Molecular analysis suggests that the West Balkan Pramenka sheep types have their origins in two distinct maternal lineages of domestic sheep and different Pramenka phenotypes tend to form few panmictic populations. The Pramenka sheep represents a valuable resource of genetic diversity in sheep. [source]


Evolutionary genetics and biogeographic structure of Rhizobium gallicum sensu lato, a widely distributed bacterial symbiont of diverse legumes

Abstract We used phylogenetic and population genetics approaches to evaluate the importance of the evolutionary forces on shaping the genetic structure of Rhizobium gallicum and related species. We analysed 54 strains from several populations distributed in the Northern Hemisphere, using nucleotide sequences of three ,core' chromosomal genes (rrs, glnII and atpD) and two ,auxiliary' symbiotic genes (nifH and nodB) to elucidate the biogeographic history of the species and symbiotic ecotypes (biovarieties) within species. The analyses revealed that strains classified as Rhizobium mongolense and Rhizobium yanglingense belong to the chromosomal evolutionary lineage of R. gallicum and harbour symbiotic genes corresponding to a new biovar; we propose their reclassification as R. gallicum bv. orientale. The comparison of the chromosomal and symbiotic genes revealed evidence of lateral transfer of symbiotic information within and across species. Genetic differentiation analyses based on the chromosomal protein-coding genes revealed a biogeographic pattern with three main populations, whereas the 16S rDNA sequences did not resolve that biogeographic pattern. Both the phylogenetic and population genetic analyses showed evidence of recombination at the rrs locus. We discuss our results in the light of the contrasting views of bacterial species expressed by microbial taxonomist and evolutionary biologists. [source]


Colocalization of GABA and glycine in the ventral nucleus of the lateral lemniscus in rat: An in situ hybridization and semiquantitative immunocytochemical study

Abstract We have studied by in situ hybridization for GAD65 mRNA in thick sections and by semiquantitative postembedding immunocytochemistry in consecutive semithin sections, the expression of ,-aminobutyric acid (GABA) and glycine in cell bodies and axosomatic puncta of the rat ventral nucleus of the lateral lemniscus (VNLL), a prominent monaural brainstem auditory structure. The in situ hybridization and the densitometric analysis of the immunostaining suggest that the rat VNLL contains two main populations of neurons. Approximately one-third of neurons are unstained with either technique and are presumably excitatory; their cell bodies are enveloped by a large number of glycine-immunoreactive puncta. Most if not all of the remaining two-thirds colocalize GABA and glycine and are assumed to be inhibitory. These two populations show a complementary distribution within the VNLL, with inhibitory neurons located mainly ventrally and excitatory neurons dorsally. In scatterplots of gray values measured from cell bodies, the double-labeled cells appear to form a single cluster in terms of their staining intensities for the two transmitter candidates. However, this cluster may have to be further subdivided because cells with extreme GABA/glycine ratios differ from those with average ratios with respect to location or size. The VNLL seems unique among auditory structures by its large number of neurons that colocalize GABA and glycine. Although the functional significance of this colocalization remains unknown, our results suggest that the VNLL exerts convergent excitatory and inhibitory influences over the inferior colliculus, which may underlie the timing processing in the auditory midbrain. U-Pb SHRIMP Dating of Zircon from Quartz Veins of the Yangshan Gold Deposit in Gansu Province and Its Geological Significance

Abstract, The Yangshan gold deposit is a super-large fine-grained disseminated gold deposit located in southern Gansu Province. Its metallogenic age has been determined by using the cathodoluminescence image and ion probe U-Pb dating techniques. It is found that zircons from quartz veinlet of the fine-grained disseminated gold ore show characters of magmatic origin with prism idiomorphism, oscillatory zoning and dominant Th/U ratios of 0.5,1.5. Three main populations of zircons are obtained, giving average 206Pb/238U ages of 197.6±1.7 Ma, 126.9±3.2 Ma and 51.2±1.3 Ma respectively. The first age corresponds to the K-Ar age of the plagiogranite dike, while the latter two ages indicate that buried Cretaceous and Tertiary intrusives exist in the orefield, suggesting that the Yangshan gold deposit was genetically related to the three magmatic hydrothermal activities. By contrast, zircons from coarse gold-bearing quartz vein in the mining area are much older than the host rock, indicating that the vein was formed earlier and was not contaminated by later magmatic fluids. It is concluded that the coupling of multiperiodic hydrothermal activities in the mining area has contributed a lot to mineralization of the Yangshan gold deposit. [source]
