Chronic interleukin-6 alters the level of synaptic proteins in hippocampus in culture and in vivo

Abstract There is now considerable evidence that the level of expression of the proinflammatory cytokine, interleukin-6 (IL-6), is increased in the central nervous system (CNS) during neuroinflammatory conditions such as occurs in neurological disorders and in disease and injury. However, our understanding of the consequences of increased expression of IL-6 on the CNS is still limited, especially with respect to the developing nervous system, which is known to be particularly vulnerable to environmental factors. To address this issue, we investigated the properties of cultured hippocampal neurons exposed chronically to IL-6 during the main period of morphological and physiological development, which occurs during the first 2 weeks of culture. IL-6 was tested at 500 U/mL, considered to reflect a pathophysiologic concentration. The morphological features of neuronal development in the control and IL-6-treated cultures appeared similar. However, Western blot analysis showed a significant reduction in the level of Group-II metabotropic receptors (mGluR2/3) and L-type Ca2+ channels in the IL-6-treated cultures. A similar reduction in mGluR2/3 and L-type Ca2+ channel protein was observed in transgenic mice that over-express IL-6 in the CNS through astrocyte production starting early in development. Analysis of Ca2+ signals produced by spontaneous synaptic network activity in the hippocampal cultures and effects of a mGluR2/3 agonist and antagonist showed that the reduced levels of mGluR2/3 impact on the functional properties of hippocampal synaptic network activity. These results have important implications relative to the mechanisms responsible for altered CNS function during conditions associated with increased levels of IL-6 in the CNS. [source]


Characteristics of woods used recently and historically by Lesser Spotted Woodpeckers Dendrocopos minor in England

Lesser Spotted Woodpecker Dendrocopos minor numbers have declined greatly in England since the early 1980s for reasons that are not yet fully understood. It has been suggested that the species' decline may be linked to the increase in Great Spotted Woodpeckers Dendrocopos major, changes in woodland habitat quality (such as deadwood abundance) and landscape-scale changes in tree abundance. We tested some of these hypotheses by comparing the characteristics of woods in southern England where the species is still relatively numerous with those of woods used in the 1980s before the major decline. In each time period, habitat, predator and landscape information from woods known to be occupied by Lesser Spotted Woodpeckers was compared with those found to be unoccupied during surveys. Before the main period of decline, Lesser Spotted Woodpeckers used oak-dominated, mature, open woods with a large amount of standing deadwood. Habitat use assessed from recent data was very similar, the species being present in mature, open, oak-dominated woodlands. There was a strong relationship between wood use probability and the extent of woodland within a 3-km radius, suggesting selection for more heavily wooded landscapes. In recent surveys, there was no difference in deadwood abundance or potential predator densities between occupied and unoccupied woods. Habitat management should focus on creating and maintaining networks of connected woodlands in areas of mature, open woods. Finer-scale habitat selection by Lesser Spotted Woodpecker within woodlands should be assessed to aid development of beneficial management actions. [source]


Abundance, Population Structure and Production of Scrobicularia plana and Abra tenuis (Bivalvia: Scrobicularidae) in a Mediterranean Brackish Lagoon, Lake Ichkeul, Tunisia

Abstract Abundance, growth and production of the deposit-feeding bivalves were studied in the Ichkeul wetland, northern Tunisia, from July 1993 , April 1994. Scrobicularia plana(Da Costa, 1778) occurred at annual mean densities (biomasses) of 299 ± 65 to 400 ± 100 individuals/m2 (22.54 ± 3.00 to 34.27 ± 3.96 g ash-free dry mass (AFDM)/m2) depending on the study area. The annual mean density of Abra tenuis(Montagu, 1803) amounted to 640 ± 74 individuals/m2 during the whole study period, in contrast the biomass rose from 2.87 g AFDM/m2 in July to 10.29 g AFDM/m2 in April. Both species were largely dominated by age class I. Although not very successful, recruitment presented a two-period pattern: the main period at the beginning of spring, and a secondary one in late summer/autumn. S. plana rarely exceeded 40 mm and lived for only 2 years, while most individuals of A. tenuis lived for only 15,18 months growing to a length of 12 mm. The annual bivalve deposit-feeder production for the whole lagoon system (90 km2) was 8.24 g AFDM/m2 (5.26 g C/m2, 0.65 g N/m2). Kinetic analysis of the cross reaction between dithioester and alkoxyamine by a Monte Carlo simulation,

Abstract A model reaction of dithioester and alkoxyamine is proposed to probe the reversible addition,fragmentation chain transfer (RAFT) process. The kinetics of the model reaction is analyzed and compared with that of pure alkoxyamine homolysis with a Monte Carlo simulation. Although the pure alkoxyamine obeys the law of persistent radical effect, the model reaction results in higher concentration of the persistent radical during the main period of the reaction. However, for a very fast RAFT process or a very low addition rate constant, the time dependence of the persistent radical concentration is quite close to that of pure alkoxyamine. Furthermore, the cross termination between the intermediate and alkyl radicals causes a retardation effect for the model reaction when the intermediate is relatively long-lived. The Monte Carlo simulation indicates that it is feasible to measure the individual rate constants of the RAFT process, such as the rate constant of addition, with a large excess of alkoxyamine. In addition, the special feature of the system with different leaving groups in the alkoxyamine and dithioester is also discussed. © 2006 Wiley Periodicals, Inc. A Doppler-based study on the prevalence of varicocele in German children and adolescents

Summary The prevalence of varicocele was estimated among pupils of forms 4 and 8 in the city of Hamburg and the severity of the associated venous reflux was analysed. In the school year of 1998/99, a genital examination was performed on 2756 children (median age 10.2 years) and 2008 adolescents (14.6 years). The varicocele degree was determined according to the WHO. Venous reflux was proven by CW-Doppler sonography, distinguishing between Valsalva-induced reflux (VR) and continuous reflux (CR). As a result, varicoceles were detected in 18.0% of the children (1.2% bilaterally) and 42.7% of the adolescents (7.2%). High-graded forms (palpable/visible) occurred with increasing rate (from 7% to 22.9%) on the left side. Subtle forms (subclinical/during Valsalva) counted for >90% of all right-sided findings, whilst an age-related shift towards higher degrees was noted left-sided. VR occurred bilaterally, CR was almost only established left-sided. VR was mainly associated with subtle varicoceles, CR was predominantly found in the high-graded forms. These results suggest that even in children varicoceles are not a rare phenomenon. However, adolescence is the main period of manifestation. A major venous malfunction is already evident in maturing boys, which seems to be associated with the formation of high-graded varicoceles. [source]


Diagenesis and Restructuring Mechanism of Oil and Gas Reservoir in the Marine Carbonate Formation, Northeastern Sichuan: A Case Study of the Puguang Gas Reservoir

Abstract: Based on the technology of balanced cross-section and physical simulation experiments associated with natural gas geochemical characteristic analyses, core and thin section observations, it has been proven that the Puguang gas reservoir has experienced two periods of diagenesis and restructuring since the Late Indo-Chinese epoch. One is the fluid transfer controlled by the tectonic movement and the other is geochemical reconstruction controlled by thermochemical sulfate reduction (TSR). The middle Yanshan epoch was the main period that the Puguang gas reservoir experienced the geochemical reaction of TSR. TSR can recreate the fluid in the gas reservoir, which makes the gas drying index higher and carbon isotope heavier because C2+ (ethane and heavy hydrocarbon) and 12C (carbon 12 isotope) is first consumed relative to CH4 and 13C (carbon 13 isotope). However, the reciprocity between fluid regarding TSR (hydrocarbon, sulfureted hydrogen (H2S), and water) and reservoir rock results in reservoir rock erosion and anhydrite alteration, which increases porosity in reservoir, thereby improving the petrophysical properties. Superimposed by later tectonic movement, the fluid in Puguang reservoir has twice experienced adjustment, one in the late Yanshan epoch to the early Himalayan epoch and the other time in late Himalayan epoch, after which Puguang gas reservoir is finally developed. Palaeomonsoon variability in the southern fringe of the Badain Jaran Desert, China, since 130 ka BP

Abstract Taking the Chagelebulu Stratigraphic Profile as a typical example, a comprehensive study has been conducted to elucidate the palaeoclimatic and geomorphic evolution patterns in the southern fringe of the Badain Jaran Desert, which were found to be complex and polycyclic in the past 130 ka. However, the fluctuating magnitude is not as remarkable as that in the eastern China sandy region. The shift in climate from interglacial to glacial and the uplift process of the Qinghai,Xizang Plateau are the two leading forces driving the evolution of the climate and desert landforms in this area. Seventeen cycles of cold, dry and warm, humid climatic stages were recognized in the Upper Pleistocene Series of the profile. The sharp uplift of the Qinghai,Xizang Plateau superimposed a cool and arid climatic trend in this area. As a result of the climatic changes, the desert in this area has undergone multiple stages of expansion and contraction since 130 ka bp. The middle Holocene Epoch and the early stage of the Late Pleistocene Period were the main periods when the sand dunes became stabilized, and the early and late phases of the Holocene Epoch and late phase of the Pleistocene Epoch were the main periods when the previously stabilized sand dunes became mobile. The late phase of the Pleistocene Epoch was the most mobile stage, when the aeolian sand activities formed the essential geomorphic pattern of the Badain Jaran Desert.
