Thirty-six mature females captured in the province of Buenos Aires showed different physiological reproductive stages. Most of them presented a postpartum oestrus in August,September. This characteristic is different from that reported in other geographical areas. The stages considered were: anoestrus, follicular phase, early pregnancy and late pregnancy. The ovaries were light-pink and smooth and presented a tortuous cord-like aspect. Many primordial follicles were found in all the females studied. Follicles in different states of maturation and primary and accessory corpora lutea were observed in the cortex. These structures were smaller than those present in other related species. Follicles did not project into the surface of the organ. Calcified follicles of diverse size were found in all the ovaries. Atretic follicles were found in all the stages analysed. Interstitial tissue surrounding the follicles and the corpora lutea was also observed. The number and proportion of different cortical structures varied in the physiological stages analysed. The ovaries of the viscacha have differential characteristics in comparison to other Hystricognathi, some of them related to polyovulation. [source] Putative reaction mechanism of heterologously expressed octopine dehydrogenase from the great scallop, Pecten maximus (L)FEBS JOURNAL, Issue 24 2007Andre Müller cDNA for octopine dehydrogenase (ODH) from the adductor muscle of the great scallop, Pecten maximus, was cloned using 5,- and 3,-RACE. The cDNA comprises an ORF of 1197 nucleotides and the deduced amino acid sequence encodes a protein of 399 amino acids. ODH was heterologously expressed in Escherichia coli with a C-terminal penta His-tag. ODH,5His was purified to homogeneity using metal,chelate affinity chromatography and Sephadex G-100 gel filtration. Recombinant ODH had kinetic properties similar to those of wild-type ODH isolated from the scallop's adductor muscle. Site-directed mutagenesis was used to elucidate the involvement of several amino acid residues for the reaction catalyzed by ODH. Cys148, which is conserved in all opine dehydrogenases known to date, was converted to serine or alanine, showing that this residue is not intrinsically important for catalysis. His212, Arg324 and Asp329, which are also conserved in all known opine dehydrogenase sequences, were subjected to site-directed mutagenesis. Modification of these residues revealed their importance for the catalytic activity of the enzyme. Conversion of each of these residues to alanine resulted in strong increases in Km and decreases in kcat values for pyruvate and l -arginine, but had little effect on the Km and kcat values for NADH. Assuming a similar structure for ODH compared with the only available structure of a bacterial opine dehydrogenase, these three amino acids may function as a catalytic triad in ODH similar to that found in lactate dehydrogenase or malate dehydrogenase. The carboxyl group of pyruvate is then stabilized by Arg324. In addition to orienting the substrate, His212 will act as an acid,base catalyst by donating a proton to the carbonyl group of pyruvate. The acidity of this histidine is further increased by the proximity of Asp329. [source] A brief review of the status, distribution and biology of wild Asian elephants Elephas maximusINTERNATIONAL ZOO YEARBOOK, Issue 1 2006R. SUKUMAR The Asian elephant Elephas maximus is distributed discontinuously across the Asian continent. The total wild population is 38 500-52 500, with a further c. 16 000 in captivity, the majority of which are in range countries. India has 60% of the global population of wild Asian elephants. The species has a multi-tiered social system with ,, living in matriarchal groups of five to 20 individuals that interact with other family units in the area. Adult ,, live alone or in small, temporary groups with weak social bonds. Asian elephants are megaherbivores that spend 12-18 hours per day feeding, and they eat browse and plants depending on availability and season. Home-range size is dependant on the availability of food, water and shelter in the region. Loss and fragmentation of habitat, human-elephant conflicts and poaching are the greatest threats to the species. Asian elephants are managed using traditional and modern methods but progress still needs to be made to improve welfare, training and breeding for these animals. HILDEBRANDT Elephants possess many unique qualities, including some that relate directly to their reproductive biology. Thus, comparative studies on elephants provide valuable information to the growing biological database for extant mammals. Left undisturbed, Asian Elephas maximus and African Loxodonta Africana elephants reproduce well in the wild. It is ironic then that most captive populations face possible,extinction'because of historically poor reproductive performance. Some of the problems with breeding elephants in captivity are logistical but others, like ovarian and uterine pathologies and bull infertility, have management-related aetiology. Through advances in endocrine monitoring and ultrasound imaging techniques, we are beginning to understand some of the complex mechanisms controlling reproductive function in elephants. Several reproductive characteristics appear to be unique to the taxon, such as luteal steroidogenic function, follicular development patterns, pituitary gonadotrophin secretion, a 22 month-long gestation and musth (in ,,). One example is the,double LH surge'occurring 3 weeks apart during the follicular or non-luteal phase of the cycle, with only the second surge inducing ovulation. These qualities have at times both enhanced and hampered efforts to understand and control reproduction. We have learned that techniques developed for domestic or laboratory species are not always directly applicable to elephants. However, the recent success of artificial insemination based on new ultrasound and endocrine methodology offers hope that establishing selfsustaining populations is possible. This paper reviews our current knowledge of elephant reproduction and how it is being used to aid species conservation for maximal reproductive efficiency and enhancement of genetic management. Bouley Abstract Both Asian (Elephas maximus) and African (Loxodonta africana) elephants produce low-frequency, high-amplitude rumbles that travel well through the ground as seismic waves, and field studies have shown that elephants may utilize these seismic signals as one form of communication. Unique elephant postures observed in field studies suggest that the elephants use their feet to ,listen' to these seismic signals, but the exact sensory mechanisms used by the elephant have never been characterized. The distribution, morphology and tissue density of Pacinian corpuscles, specialized mechanoreceptors, were studied in a forefoot and hindfoot of Asian elephants. Pacinian corpuscles were located in the dermis and distal digital cushion and were most densely localized to the anterior, posterior, medial and lateral region of each foot, with the highest numbers in the anterior region of the forefoot (52.19%) and the posterior region of the hindfoot (47.09%). Pacinian corpuscles were encapsulated, had a typical lamellar structure and were most often observed in large clusters. Three-dimensional reconstruction through serial sections of the dermis revealed that individual Pacinian corpuscles may be part of a cluster. By studying the distribution and density of these mechanoreceptors, we propose that Pacinian corpuscles are one possible anatomic mechanism used by elephants to detect seismic waves. [source] Habitat-specific normal and reverse diel vertical migration in the plankton-feeding basking sharkJOURNAL OF ANIMAL ECOLOGY, Issue 4 2005DAVID W. SIMS Summary 1Megaplanktivores such as filter-feeding sharks and baleen whales are at the apex of a short food chain (phytoplankton,zooplankton,vertebrate) and are sensitive indicators of sea-surface plankton availability. Even though they spend the majority of their time below the surface it is still not known how most of these species utilize vertical habitat and adapt to short-term changes in food availability. 2A key factor likely to control vertical habitat selection by planktivorous sharks is the diel vertical migration (DVM) of zooplankton; however, no study has determined whether specific ocean-habitat type influences their behavioural strategy. Based on the first high-resolution dive data collected for a plankton-feeding fish species we show that DVM patterns of the basking shark Cetorhinus maximus reflect habitat type and zooplankton behaviour. 3In deep, well-stratified waters sharks exhibited normal DVM (dusk ascent,dawn descent) by tracking migrating sound-scattering layers characterized by Calanus and euphausiids. Sharks occupying shallow, inner-shelf areas near thermal fronts conducted reverse DVM (dusk descent,dawn ascent) possibly due to zooplankton predator,prey interactions that resulted in reverse DVM of Calanus. 4These opposite DVM patterns resulted in the probability of daytime-surface sighting differing between these habitats by as much as two orders of magnitude. Ship-borne surveys undertaken at the same time as trackings reflected these behavioural differences. 5The tendency of basking sharks to feed or rest for long periods at the surface has made them vulnerable to harpoon fisheries. Ship-borne and aerial surveys also use surface occurrence to assess distribution and abundance for conservation purposes. Our study indicates that without bias reduction for habitat-specific DVM patterns, current surveys could under- or overestimate shark abundance by at least 10-fold. [source] Studies on feed digestibilities in captive Asian elephants (Elephas maximus)JOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 3-4 2003M. Clauss Summary In order to test the suitability of the horse as a nutritional model for elephants, digestibility studies were performed with six captive Asian elephants on six different dietary regimes, using the double marker method with acid detergent lignin as an internal and chromium oxide as an external digestibility marker. Elephants resembled horses in the way dietary supplements and dietary crude fibre content influenced digestibility, in calcium absorption parameters and in faecal volatile fatty acid composition. However, the absolute digestibility coefficients achieved for all nutrients are distinctively lower in elephants. This is because of much faster ingesta passage rates reported for elephants. No answer is given to why elephants do not make use of their high digestive potential theoretically provided by their immense body weight. Differences in volatile fatty acid concentrations between these captive elephants and those reported from elephants from the wild are in accord with a reported high dependence of free-ranging elephants on browse forage. Dabei wurde die Doppelindikatormethode mit Lignin als internem und Chromoxid als externem Marker verwendet. Elefanten ähnelten Pferden hinsichtlich der Art und Weise, wie sich Ergänzungsfuttermittel und der Rohfasergehalt der Ration auf die Verdaulichkeit auswirkten, hinsichtlich der Kalziumabsorption, und hinsichtlich der Zusammensetzung der flüchtigen Fettsäuren im Kot. [source] Population structure in the South American tern Sterna hirundinacea in the South Atlantic: two populations with distinct breeding phenologiesJOURNAL OF AVIAN BIOLOGY, Issue 4 2010Patrícia J. Faria The South American tern Sterna hirundinacea is a migratory species for which dispersal, site fidelity and migratory routes are largely unknown. Here, we used five microsatellite loci and 799,bp partial mitochondrial DNA sequences (Cytochrome b and ND2) to investigate the genetic structure of South American terns from the South Atlantic Ocean (Brazilian and Patagonian colonies). Brazilian and Patagonian colonies have two distinct breeding phenologies (austral winter and austral summer, respectively) and are under the influence of different oceanographic features (e.g. Brazil and Falklands/Malvinas ocean currents, respectively), that may promote genetic isolation between populations. Results show that the Atlantic populations are not completely panmictic, nevertheless, contrary to our expectations, low levels of genetic structure were detected between Brazilian and Patagonian colonies. Such low differentiation (despite temporal isolation of the colonies) could be explained by demographic history of these populations coupled with ongoing levels of gene flow. Interestingly, estimations of gene flow through Maximum likelihood and Bayesian approaches has indicated asymmetrical long term and contemporary gene flow from Brazilian to Patagonian colonies, approaching a source,sink metapopulation dynamic. Genetic analysis of other South American tern populations (especially those from the Pacific coast and Falklands,Malvinas Islands) and other seabird species showing similar geographical distribution (e.g. royal tern Thalasseus maximus), are fundamental in gaining a better understanding of the main processes involved in the diversification of seabirds in the southern hemisphere. [source] Sexual development and reproductive seasonality of hogfish (Labridae: Lachnolaimus maximus), an hermaphroditic reef fishJOURNAL OF FISH BIOLOGY, Issue 5 2007R. S. McBride The seasonality, size, age, colour phases and sexual dimorphism of 13 reproductive classes of hogfish Lachnolaimus maximus are described. Analysis of histological sections of gonads (n = 1662) confirmed earlier conclusions that L. maximus is a monandric, protogynous hermaphrodite. Sex change was initiated at the end of the spawning season and over a broad range of sizes and ages. It occurred after a functional female phase (postmaturation) and proceeded more slowly (months) than previously believed. Eventually all individuals changed sex to a terminal male phase. Females were batch spawners, spawning as often as every day during winter and spring. There was no evidence of precocious sperm crypts in active females, sperm competition or other alternative male sexual strategies. Mating has been reported elsewhere to be haremic. The sexual development of L. maximus appears to be adaptive in terms of Ghiselin's size-advantage model, which links monandric protogyny and polygyny. The slow rate of sex change, however, poses problems when fishing pressure is high because harvest of a single male has the potential to reduce the reproductive output of an entire harem. [source] Non-specific immune response of turbot, Scophthalmus maximus (L.), experimentally infected with a pathogenic Vibrio pelagiusJOURNAL OF FISH DISEASES, Issue 6 2003L Villamil Abstract The effect of a pathogenic Vibrio pelagius, isolated during a mass mortality of turbot larvae, on the non-specific immune response of turbot, Scophthalmus maximus (L.), macrophages was studied both in vitro and in vivo. The in vitro treatment of head kidney (HK) macrophages with viable V. pelagius caused a significant inhibition of the chemiluminescence (CL) response in comparison with untreated macrophages, while incubation with heat-killed bacteria did not affect this response. In vivo, the intraperitoneal injection of V. pelagius resulted in a significant inhibition of the CL response in infected fish at days 1 and 4 post-infection compared with the control fish response. The HK macrophage nitric oxide (NO) production was enhanced by in vitro incubation with intermediate doses of viable V. pelagius (5 × 103 and 5 × 104 bacteria mL,1) and higher doses of the heat-killed bacteria (5 × 104,5 × 106 bacteria mL,1). In both cases, the NO inhibitorN- , -nitro-L-arginine was capable of down-regulating the specific NO induction caused by incubation with the bacterial treatments. In contrast, incubation with ECPs at higher doses caused a reduction in NO production. In vivo, a significant enhancement in NO production was also observed in macrophage supernatants at day 10 post-infection. Lysozyme concentration in the serum was also significantly increased in the experimentally infected fish at days 4 and 10 post-injection. In addition, viable V. pelagius and its ECPs significantly reduced HK macrophage viability in vitro, whereas no significant differences in viability were observed during the incubation with heat-killed bacteria. As NO production was enhanced in the experimentally infected fish, the inhibitory effect of the NO donor, S-nitroso-acetyl-penicillamine (SNAP), was tested in vitro in a cell-free assay. The results showed that growth of V. pelagius was significantly inhibited using SNAP at a high concentration (1 mm). [source] A study of the susceptibility of Atlantic halibut, Hippoglossus hippoglossus (L.), to viral haemorrhagic septicaemia virus isolated from turbot, Scophthalmus maximus (L.)JOURNAL OF FISH DISEASES, Issue 4 2003T J Bowden Abstract The susceptibility of Atlantic halibut, Hippoglossus hippoglossus (L.), to viral haemorrhagic septicaemia virus (VHSV) was tested. Juvenile halibut of approximately 5 g weight were subjected to challenge by intraperitoneal injection, cohabitation and immersion to a VHSV isolate from an outbreak of the disease in turbot, Scophthalmus maximus (L.). The intraperitoneal injection gave the highest mortality rate of 28% after 50 days. The cohabitee group suffered 19% mortality rate and the immersion group only 2%. Control groups included turbot exposed either by intraperitoneal injection or immersion which suffered mortality rates of 93 and 50%, respectively. The results suggest that halibut are markedly less susceptible to VHSV than turbot. [source] Use of probiotics to control furunculosis in rainbow trout, Oncorhynchus mykiss (Walbaum)JOURNAL OF FISH DISEASES, Issue 6 2002A Irianto Aerobic heterotrophic bacteria were isolated from the intestinal contents of Atlantic salmon, Salmo salar, rainbow trout, Oncorhynchus mykiss, and turbot, Scophthalmus maximus, on tryptone soya agar and De Man Rogosa and Sharpe agar, of which 11 of 177 (6% of the total) of the isolates were antagonistic to Aeromonas salmonicida. Four of these cultures, which were identified tentatively as A. hydrophila, Vibrio fluvialis, Carnobacterium sp. and an unidentified Gram-positive coccus, were beneficial to fish when fed singly or as an equi-mixture. Feed supplemented with the putative probiotics indicated survival of the organisms in the gastrointestinal tract for 7 days. Feeding with the probiotics for 7 and 14 days led to better survival following challenge with A. salmonicida. There was no indication of serum or mucus antibodies to A. salmonicida, but there was an increased number of erythrocytes, macrophages, lymphocytes and leucocytes, and enhanced lysozyme activity in the fish. [source] Gill disease of marine fish caused by infection with Neoparamoeba pemaquidensisJOURNAL OF FISH DISEASES, Issue 9 2001B L Munday Amoebic gill disease (AGD) of maricultured salmonids, turbot, Scophthalmus maximus (L.), European seabass, Dicentrarchus labrax (L.), and sharpsnout seabream, Diplodus puntazzo (Cetti), caused by Neoparamoeba pemaquidensis has been reported from Australia (Tasmania), Ireland, France, Chile, North America (Washington State and California) and Spain. Of the salmonids, Atlantic salmon, Salmo salar L., appears to be the most susceptible with rainbow trout, Oncorhynchus mykiss (Walbaum), also suffering significant disease. Only minor outbreaks have been reported in coho, O. kisutch (Walbaum), and chinook salmon, O. tshawytscha (Walbaum). The disease now accounts for 10,20% of production costs of Atlantic salmon in Tasmania and has lead to temporary abandonment of culture of this species in parts of Spain. It is of lesser, but still significant, importance in other countries. Much is known about the pathology of AGD but the pathophysiology of the disease is poorly understood. There is evidence that non-specific immunity is involved in fish acquiring resistance to AGD, but no unequivocal evidence exists for protection as a result of specific immune responses. To date, for salmonids, the only effective treatment for AGD is a freshwater bath. Control procedures based on modification of management strategies have been minimal and virtually unresearched. [source] Electromyographic analysis of a modified maneuver for quadriceps femoris muscle setting with co-contraction of the hamstringsJOURNAL OF ORTHOPAEDIC RESEARCH, Issue 3 2003Masaaki Nakajima Abstract A ,quadriceps femoris muscle setting" is isometric quadriceps femoris exercise which can be widely used in early knee rehabilitation. However this exercise cannot obtain enough co-contraction of the hamstrings. Isolated quadriceps femoris contraction in knee extension imposes severe strain to anterior cruciate ligament. We succeeded in developing a simple training maneuver that is effective in obtaining co-contraction of the hamstrings,a modified maneuver for the quadriceps femoris muscle setting with the contralateral lower limb raised (MQS). In this study, we analyzed the effect of this maneuver by EMG quantification. Twenty-eight healthy young adult men performed sequential trials consisting of normal quadriceps femoris muscle setting (NQS) and MQS. Electromyographic activity was recorded from surface electrodes on the gluteus maximus, vastus medialis, rectus femoris, vastus lateralis, semitendinosus and biceps femoris (long head), and normalized to values derived from maximal isometric trials. The % maximal voluntary isometric contraction (%MVIC) of the vastus medialis, vastus lateralis and rectus femoris did not vary in the each maneuver. However, the %MVIC of the hamstrings varied significantly in the MQS. This study suggests that effective co-contraction of the hamstrings can be obtained in MQS by adjusting the load to the raised lower limb. © 2002 Orthopaedic Research Society. [source] Molecular identification of five commercial flatfish species by PCR,RFLP analysis of a 12S rRNA gene fragmentJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 8 2003Angel S Comesaña Abstract Refrigerated or frozen fillets of commercial flatfish species are sometimes mislabelled, and identification of those products is needed to avoid fraudulent substitution. Molecular identification of five commercial flatfish species (order Pleuronectiformes), ie Lepidorhombus whiffiagonis (megrim), Platichthys flesus (flounder), Reinhardtius hippoglossoides (Greenland halibut), Scophthalmus maximus (turbot) and Solea vulgaris (= S solea) (sole), has been carried out on the basis of the amplification of an approximately 433 bp segment from the mitochondrial 12S rRNA gene using the polymerase chain reaction (PCR) and universal primers. Direct DNA sequencing from two PCR products for each flatfish species was carried out, and sequences were used to select six restriction enzymes. PCR products of 15 individuals of each species were cut with each enzyme, resulting in species-specific restriction fragment length polymorphism (RFLP). The five flatfish species could be identified by application of the restriction enzyme AluI as well as by using different combinations of a pair of enzymes, ie DdeI and either AciI or MwoI. No intraspecific genetic polymorphism was found for any of the six enzymes. Results confirmed the usefulness of this technique to distinguish and genetically characterise refrigerated or frozen pieces of these five flatfish species. Imsland The aim of this study was to investigate the possible benefit of "temperature-steps" (T-steps) rearing for juvenile turbot (initial weight 15.1 g) under realistic production scale and to determine whether initial growth advantage is maintained throughout the rearing period to market size. One group (called T-step 22-19-16) of juvenile turbot was reared at three different temperatures, that is, 22 C (from 17 to 60 g) followed by 19 C (from 60 to 100 g) and 16 C (>100 g); another group (called T-step 19-16) at two temperatures, that is, 19 C (from 17 to 100 g) and lowered to 16 C (>100 g); and the third group (called C16) at one constant temperature, that is, 16 C. Relative growth was significantly higher in the two T-step groups, with the T-step 19-16 showing the highest overall growth. Feed conversion efficiency was highest in the 19-16 group. Only minor effects of the experimental rearing on blood physiology were found, with one notable exception of inverse relationship between plasma glucose and growth. Overall, these findings indicate that a short interval of rearing fish at high temperature during the early juvenile phase may have a long-term effect on biomass increment in turbot. This is an important finding for the turbot industry. [source] Population pharmacokinetics of pyrazinamide in elephantsJOURNAL OF VETERINARY PHARMACOLOGY & THERAPEUTICS, Issue 5 2005M. ZHU This study was undertaken to characterize the population pharmacokinetics (PK), therapeutic dose, and preferred route of administration for pyrazinamide (PZA) in elephants. Twenty-three African (Loxodonta africana) and Asian (Elephas maximus) elephants infected with or in contact with others culture positive for Mycobacterium tuberculosis were dosed under treatment conditions. PZA was dosed daily at 20,30 mg/kg via oral (fasting or nonfasting state) or rectal (enema or suppository) administration. Blood samples were collected 0,24 h postdose. Population PK was estimated using nonlinear mixed effect modeling. Drug absorption was rapid with Tmax at or before 2 h regardless of the method of drug administration. Cmax at a mean dose of 25.6 (±4.6) mg/kg was 19.6 (±9.5 ,g/mL) for PZA given orally under fasting conditions. Under nonfasting conditions at a mean dose of 26.1 ± 4.2 mg/kg, Cmax was 25% (4.87 ± 4.89 ,g/mL) and area under concentration curve (AUC) was 30% of the values observed under fasting conditions. Mean rectal dose of 32.6 ± 15.2 mg/kg yielded Cmax of 12.3 ± 6.3 ,g/mL, but comparable AUC to PZA administered orally while fasting. Both oral and rectal administration of PZA appeared to be acceptable and oral dosing is preferred because of the higher Cmax and lower inter-subject variability. A starting dose of 30 mg/kg is recommended with drug monitoring between 1 and 2 h postdose. Higher doses may be required if the achieved Cmax values are below the recommended 20,50 ,g/mL range. N. MASLOW We recently described the clinical presentation and treatment of 18 elephants from six herds infected with TB. Treatment protocols and methods varied between herds to include both oral and rectal dosing using multiple drug doses and formulations. In this paper we present information regarding the pharmacokinetics (PK) of isoniazid (INH) in elephants and provide suggestions regarding initial treatment regimens. Forty-one elephants received INH daily by either oral or rectal administration with different formulations. Population PK analysis was performed using Non-linear Mixed Effect Modeling (NONMEM). Results of oral administration indicated that compared with premixed INH solution, the drug exposure was highest with a suspension prepared freshly with INH powder. When INH was concomitantly given as an admixture over food, Tmax was delayed and variability in drug absorption was significantly increased. Compared with oral administration, similar drug exposures were found when INH was dosed rectally. The data generated suggest that a starting dose of 7.5 mg/kg of INH is appropriate for initial TB treatment in elephants when premixed solution is administered directly into the oropharynx or rectal vault and 4 mg/kg are when INH is administered following immediate suspension from powdered form. Hansen Metomidate was administered to halibut (Hippoglossus hippoglossus) and turbot (Scophthalmus maximus) intravenously at a dose of 3 mg/kg bodyweight, as a bath treatment at a dose of 9 mg/L water for 5 min to study the disposition of metomidate, and as bath treatment (9 mg/L) for 10 min to study the absorption and effect of metomidate on respiration and balance/motor control. Additionally, turbot were given metomidate orally at a dose of 7 mg/kg. The studies were performed in seawater at a temperature of 10.3 ± 0.4 °C (halibut) and 18.0 ± 0.3 °C (turbot). Pharmacokinetic modeling of the data showed that metomidate had shorter elimination half-life and higher plasma concentrations in turbot compared with halibut, both species displaying a rapid uptake, distribution and excretion. Following intravenous administration, the volumes of distribution at steady state (Vd(ss)) were 0.21 L/kg (halibut) and 0.44 L/kg (turbot). Plasma clearances (Cl) were 0.099 L/h·kg in halibut and 0.26 L/h·kg in turbot and the elimination half-lives (t½,z) were calculated to be 5.8 h and 2.2 h in halibut and turbot, respectively. Mean residence times (MRT) were 2.2 h in halibut and 1.7 h in turbot. Following oral administration, the t½,z was 3.5 h in turbot. The maximum plasma concentration (Cmax) was 7.8 mg/L in turbot 1 h after administration. The oral bioavailability (F) was calculated to 100% in turbot. Following 5 min bath the maximum plasma concentrations (Cmax), which were observed immediately after end of the bath, were 9.5 mg/L and 13.3 mg/L in halibut and turbot, respectively. Metomidate rapidly immobilized the fish, with respiratory depression, reduced heart rate, and loss of balance/motor control within 1 min (mean). Recovery was slow, with resumed balance/motor control after 26.4 min. Opercular respiration movements were resumed more rapidly with a recorded mean of 1.7 min. Oral administration was demonstrated to be a way of immobilizing fish, for example in large aquariums, without exposing them to unwanted stress. The physical relationships between somatic motor nerves, which control muscle fiber contraction, and arterioles, which control microvascular perfusion, are unexplored. The authors tested the hypothesis that motor axons align with arterioles in adult skeletal muscle. Methods: Transgenic mice (C57BL/6 background, n = 5; 10 months of age) expressing yellow fluorescent protein in all motor nerves underwent vascular casting (Microfil). Excised epitrochlearis, gracilis, gluteus maximus, and spinotrapezius muscles were imaged at 380× and 760× and a computer-integrated tracing system (Neurolucida) was used to acquire 3-dimensional digital renderings of entire arteriolar and neural networks within each muscle. Results: Arteriolar networks were typically ,3-fold longer than neural networks. Nerves coursed with arterioles until terminating at motor endplates. Across muscles, proximity analyses revealed that , 75% of total nerve length (9.8,48.8 mm) lay within 200 ,m of the nearest arteriole (diameters of 15,60 , m). Conclusions: Somatic motor nerves and arterioles align closely within adult mammalian skeletal muscle. Understanding the signals governing neurovascular alignment may hold important clues for the advancement of tissue engineering and regeneration. No significant linkage disequilibrium between pairs of loci was found, but two loci significantly deviated from Hardy,Weinberg equilibrium after Bonferroni correction. Methods.,We trained four men with sensorimotor-complete (ASIA A) SCI, who achieved gait through FES of the quadriceps femoris, gluteus maximus, and common peroneal nerve on each side on a motorized treadmill. Training involved progressive interval walking exercise, consisting of periods of activity followed by equal periods of rest, repeated until muscle fatigue. We used time to muscle fatigue during continuous treadmill ambulation as the primary outcome measure. We also recorded the patterns of incremental stimulation for all training and testing sessions. Results.,All subjects increased their ambulation capacity; however, the responses varied from subject to subject. Some subjects increased the total distance walked by as much as 300% with progressive improvement over the entire training period; however, others made more modest gains and appeared to reach a performance plateau within a few training sessions. Conclusions.,FES-supported interval training offers a useful and effective strategy for strength-endurance improvement in the large muscle groups of the lower limb in motor-complete SCI. We believe that this training protocol offers a viable alternative to that of continuous walking training in people with SCI using FES to aid ambulation. Morphologically, it is formed by specific secretory cells, folliculostellate cells, and migratory cells coming from the pars distalis. The purpose of this work was to investigate differences in specific cellular characteristics in the PT of viscachas captured in summer (long photoperiod) and winter (short photoperiod), as well as the effects of chronic melatonin administration in viscachas captured in summer and kept under long photoperiod. In summer, the PT-specific cells exhibited cell-like characteristics with an important secretory activity and a moderate amount of glycogen. In winter, the PT-specific granulated cells showed ultrastructural variations with signs of a reduced synthesis activity. Also, PT showed a high amount of glycogen and a great number of cells in degeneration. After melatonin administration, the ultrastructural characteristics were similar to those observed in winter, but the amount of glycogen was higher. These results suggest possible functional implications as a result of morphological differences between long and short photoperiods, and are in agreement with the variations of the pituitary-gonadal axis, probably in response to the natural photoperiod changes through the pineal melatonin. The ultrastructural differences observed in PT, after melatonin administration, were similar to those observed in the short photoperiod, thus supporting the hypothesis that these cytological changes are induced by melatonin. [source] Morphological and Morphometric Changes of Pituitary Lactotrophs of Viscacha (Lagostomus maximus maximus) in Relation to Reproductive Cycle, Age, and SexTHE ANATOMICAL RECORD : ADVANCES IN INTEGRATIVE ANATOMY AND EVOLUTIONARY BIOLOGY, Issue 1 2010Verónica Filippa Abstract Lactotrophs in pituitary pars distalis (PD) of viscacha were studied by immunohistochemistry and morphometric analysis in the following groups: 1) adult males throughout the reproductive cycle (reproductive, gonadal regression, and recovery periods), 2) melatonin-treated adults, 3) castrated adults, 4) prepubertal, 5) non-pregnant females, and 6) pregnant females (early, mid, and late pregnancy). Immunopositive percentage area (%IA), cell percentage in PD (% PDC), number of cells per reference area (no.cell/RA), major cellular and nuclear diameters were analyzed. Lactotrophs were mainly localized in the ventro,medial region and the caudal extreme of PD. In the male viscachas, they were isolated in small and big groups, close to blood vessels and near follicles. These cells were pleomorphic and with a heterogeneous cytoplasmic immunolabeling pattern. In the adult males of the gonadal regression period the morphometric parameters were the lowest. Most parameters of lactotrophs in the prepubertal were significantly lower than in the adult males in the reproductive period. In the melatonin-treated animals and in castrated animals there was a decrease in %IA, %PDC, and no.cell/RA. In the females, the morphometric parameters increased at the end of pregnancy. Non-pregnant females exhibited a higher immunopositive area and number, but a smaller size of cells than males. Our results showed that in the adult male viscacha, lactotrophs vary seasonally, probably due to the photoperiod effect through melatonin. Besides the changes observed after castration, in prepubertal animals, in adults of different sex, and during pregnancy suggest that the gonadal steroid hormones might modify the lactotrophs activity. Vidya This study examines the population genetic structure of Asian elephants (Elephas maximus) across India, which harbours over half the world's population of this endangered species. Mitochondrial DNA control region sequences and allele frequencies at six nuclear DNA microsatellite markers obtained from the dung of free-ranging elephants reveal low mtDNA and typical microsatellite diversity. Both known divergent clades of mtDNA haplotypes in the Asian elephant are present in India, with southern and central India exhibiting exclusively the , clade of Fernando et al. (2000), northern India exhibiting exclusively the , clade and northeastern India exhibiting both, but predominantly the , clade. A nested clade analysis revealed isolation by distance as the principal mechanism responsible for the observed haplotype distributions within the , and , clades. Analyses of molecular variance and pairwise population FST tests based on both mitochondrial and microsatellite DNA suggest that northern-northeastern India, central India, Nilgiris (in southern India) and Anamalai-Periyar (in southern India) are four demographically autonomous population units and should be managed separately. In addition, evidence for female philopatry, male-mediated gene flow and two possible historical biogeographical barriers is described. Ruff Abstract The present study investigated the distribution of , -tocopherol (vitamin E) in fillets of turbot (Scophthalmus maximus) and Atlantic halibut (Hippoglossus hippoglossus). Turbot and Atlantic halibut were fed commercial diets, supplemented with different levels of , -tocopheryl acetate at the dietary target levels of 100, 500 and 1000 mg , -tocopheryl acetate kg,1 diet. The actual levels were 72, 547 and 969 for turbot, while halibut received 189, 613 and 875 mg , -tocopheryl acetate kg,1 diet. Turbot were fed the diets for 24 weeks, while Atlantic halibut were fed for 20 weeks prior to slaughter. At the end of the feeding periods fish had reached a final weight of around 1 kg. Fish were slaughtered and filleted. From the four fillets obtained per fish, 22 samples were taken from designated areas and analysed for their , -tocopherol content. The average concentrations of , -tocopherol incorporated in turbot and Atlantic halibut increased with increasing levels of , -tocopheryl acetate in the diet. Atlantic halibut had significantly (P < 0.05) more , -tocopherol in positions 2/II and 1/I than in position 9/IX. Turbot had significantly (P < 0.05) more , -tocopherol in position 2/II than in positions 1/I, 4/IV and 11/XI. By mapping , -tocopherol concentrations in fish fillets, a high degree of quality prediction may be established. Moreover, this study may help scientists in their choice of sampling position, when investigating if , -tocopheryl acetate supplementation resulted in successful , -tocopherol incorporation. [source] Soybean protein concentrate as a protein source for turbot Scophthalmus maximus L.AQUACULTURE NUTRITION, Issue 4 2000O.J. Day In the first of two experiments, the effect of a gradual substitution of dietary fish meal with soybean protein concentrate (SPC) on growth, feed consumption and protein digestibility was examined in 13 g turbot Scophthalmus maximus. Five isonitrogenous and isoenergetic diets (50% protein and 22 kJ g,1) containing SPC at protein replacement levels of 0, 25, 50, 75 and 100% were offered by hand twice daily. Growth rates of fish fed diets with zero and 25% replacement were not significantly different, with SGRs of 2.47 and 2.28, respectively. At higher replacement levels, growth rates decreased significantly with SGRs of 2.00, 1.33 and 0.68, respectively. Feed conversion ratios increased with soya replacement, with values of 0.68, 0.75, 0.89, 1.27 and 2.32, respectively, although there was no significant difference between the first two. Feed consumption rates remained constant up to 50% replacement, above which they decreased significantly, possibly because of reduced diet palatability. Apparent protein digestibility (APD) was not affected by the incorporation of SPC and ranged from 82.8 to 87.5%. Results suggest that protein catabolism increases in SPC-rich diets, possibly because of rapid assimilation and utilization of the methionine supplement. In the second experiment, the importance of amino acid supplements and the beneficial effects of protecting these, either by coating them in protein or incorporating them in a protein,lipid emulsion, was investigated. Growth data provided some indication that the utilization of SPC may be improved by incorporating the methionine and lysine supplement in a protein,lipid emulsion prior to diet preparation, although this finding was not found to be statistically significant (0.1 < P < 0.2). Over the 297-day experimental period, the overall growth rate of the 12L:12D (0.82% d,1) treatment was higher than for both 08L:16D (0.80% d,1) and 20L:04D (0.77% d,1). Present results show that the long-term extended fixed photoperiod may act as an irritant, inducing stress, suppressing growth and reducing feed utilization. It is hypothesized that the progression of size-dependent hierarchies over time can be divided into two distinct phases herein referred to as ,hierarchy resolution' and ,hierarchy stabilization' phases (or phases 1 and 2) characterized by increasing and decreasing growth heterogeneity respectively. Hall-Spencer Abstract 1.Maerl beds are carbonate sediments, built by a surface layer of slow-growing coralline algae, forming structurally fragile habitats. 2.They are of international conservation significance, often supporting a high biodiversity and abundant bivalve molluscs. 3.Experimental fishing for scallops (Pecten maximus) on French and UK grounds has shown that although large epifauna are often killed, many organisms escape harm as they burrow deeply or are small enough to pass through the dredges. 4.Bivalve dredging is currently one of the main threats to European maerl grounds as it reduces their biodiversity and structural complexity and can lead to long-term degradation of the habitat. 5.Protecting maerl grounds is of importance for fisheries since they provide structurally complex feeding areas for juvenile fish (e.g. Atlantic cod - Gadus morhua) and reserves of commercial brood stock (e.g. Ensis spp., P. maximus and Venus verrucosa). 6.We outline improved mechanisms to conserve these ancient and unique biogenic habitats.
