Selected AbstractsRhizoctonia solani AG 2-1 as a causative agent of cotyledon rot on European beech (Fagus sylvatica)FOREST PATHOLOGY, Issue 6 2005A. M. Hietala Summary Rhizoctonia solani was frequently isolated in the Italian Alps from nursery-grown European beech (Fagus sylvatica) seedlings displaying symptoms of cotyledon rot. Koch's postulates were verified and mode of infection of the associated isolates was investigated with light and scanning electron microscopy. Population structure of the pathogen was investigated by scoring the anastomosis reaction type in pairings between different isolates from the same seedbed. One pathogen genotype showed a large distribution area within the seedbed, this implying that the inoculum had been building up in the seedbed over a longer time period. Hyphal anastomosis tests and sequence analysis of the internal transcribed spacer (ITS) region of ribosomal DNA indicated that the pathogen belongs to AG 2-1 of R. solani. ITS sequence analysis indicates that the isolates from beech are closely related to R. solani isolates causing a disease on tulip. The striking similarities between the two diseases are discussed. Les postulats de Koch ont été vérifiés et le mode d'infection étudié par microscopie optique et électronique à balayage. La structure de la population de l'agent pathogène a étéétudiée en examinant les réactions d'anastomoses dans les confrontations par paires des isolats d'un même lit de semences. Un génotype particulier s'est avéré largement distribué dans le lit de semence, suggérant soit une accumulation de l'inoculum pendant une longue période soit que ce génotype est capable de reproduction homocaryotique, favorisant sa dispersion. Les tests d'anastomose et l'analyse de la séquence de la région ITS de l'ADN ribosomal indiquent que l'agent pathogène appartient au groupe AG 2-1 de R. solani. L'analyse de la séquence de l'ITS montre que les isolats de hêtre sont proches d'isolats de R. solani pathogènes sur tulipe. Les ressemblances frappantes entre les deux maladies et la gestion de la maladie sur hêtre sont discutées. Die Koch'schen Postulate wurden erfüllt und die Art der Infektion der beteiligten Isolate wurde licht- und rasterelektronen-mikroskopisch untersucht. Die Populationsstruktur des Pathogens wurde anhand der Reaktionstypen (Anastomosierungsverhalten) in Paarungsversuchen mit den unterschiedlichen Isolaten aus demselben Saatbeet untersucht. Ein Kompatibilitätstyp war innerhalb des Saatbeetes weit verbreitet, was darauf hindeutet, dass sich das Inokulum über einen längeren Zeitraum dort angereichert hatte und/oder der Genotyp homokaryotisch fruchtet, was seine Ausbreitung fördert. Die Anastomosierungstests und die ITS-Sequenzanalyse der ribosomalen DNA ergaben, dass der Erreger zu der AG 2-1 Gruppe R. solani gehört. Die ITS-Sequenzen deuten darauf hin, dass die Isolate von Buche mit den R. solani, Isolaten verwandt sind, die an Tulpen pathogen sind. We complement their results by analyzing data over a longer time period and, more importantly, contend that most of the unexplained gender difference in total pay among executives was due to gender differences in the portion of variable pay, in particular a different cash payout from stock option exercises. Koprinkova For the purpose of dynamic systems modeling, it was proposed to include feedback connections or delay elements in the classical feed-forward neural network structure so that the present output of the neural network depends on its previous values. These delay elements can be connected to the hidden and/or output neurons of the main neural network. Each delay element gets a value of a state variable at a past time instant and keeps this value during a single sampling period. The groups of delay elements record the values of the state variables for a given time period in the past. Changing the number of the delay elements, which belongs to one group, a shorter or a longer time period in the past can be accounted for. Thus, the connection weights determine the influence of the past process states on the present state in a similar way as it is in the time delay kernel or cause-effect relation membership function (CER-MF) models. Specific feed-forward neural networks with time delay connections are used to solve the problem of neural network chemostat modeling as well as specific kinetic rates modeling. The weights of the feedback connections obtained during model training are discussed as the points of a time delay kernel or as the strength levels in a CER model (the points in the CER-MF). [source] The (Ir)Relevance of Militarized Interstate Disputes for International TradeINTERNATIONAL STUDIES QUARTERLY, Issue 1 2002Quan Li Do military disputes between two states suppress trade between their firms? Both liberals and realists suggest that conflict occurrence reduces bilateral trade. However, using a rational expectation argument, Morrow (1999) proposes that conflict occurrence and trade should be uncorrelated statistically. Empirical evidence to date both supports expectations and appears contradictory and inconclusive. We offer a theory that reconciles, encompasses, and extends the competing arguments, explaining the empirical inconsistency. By incorporating rational expectations and uncertainty into the profit calculus of trading firms, the theory identifies the conditions under which various properties of a conflict (onset, duration, and severity) should and should not reduce bilateral trade ex ante and ex post. We test the ex post effects in two datasets that cover either a wider range of countries or a longer time period than previous quantitative studies. Both an unexpected MID onset and the unexpectedness of a MID onset reduce bilateral trade substantially ex post. Preliminary tests suggest that MID duration and severity also affect bilateral trade ex post. We conclude by discussing the implications of our research. [source] Oestrogenic activity of benzyl salicylate, benzyl benzoate and butylphenylmethylpropional (Lilial) in MCF7 human breast cancer cells in vitroJOURNAL OF APPLIED TOXICOLOGY, Issue 5 2009A. K. Charles Abstract Benzyl salicylate, benzyl benzoate and butylphenylmethylpropional (Lilial) are added to bodycare cosmetics used around the human breast. We report here that all three compounds possess oestrogenic activity in assays using the oestrogen-responsive MCF7 human breast cancer cell line. At 3 000 000-fold molar excess, they were able to partially displace [3H]oestradiol from recombinant human oestrogen receptors ER, and ER,, and from cytosolic ER of MCF7 cells. At concentrations in the range of 5 × 10,5 to 5 × 10,4 m, they were able to increase the expression of a stably integrated oestrogen-responsive reporter gene (ERE-CAT) and of the endogenous oestrogen-responsive pS2 gene in MCF7 cells, albeit to a lesser extent than with 10,8 m 17, -oestradiol. They increased the proliferation of oestrogen-dependent MCF7 cells over 7 days, which could be inhibited by the antioestrogen fulvestrant, suggesting an ER-mediated mechanism. Although the extent of stimulation of proliferation over 7 days was lower with these compounds than with 10,8 m 17, -oestradiol, given a longer time period of 35 days the extent of proliferation with 10,4 m benzyl salicylate, benzyl benzoate or butylphenylmethylpropional increased to the same magnitude as observed with 10,8 m 17, -oestradiol over 14 days. This demonstrates that benzyl salicylate, benzyl benzoate and butylphenylmethylpropional are further chemical components of cosmetic products which give oestrogenic responses in a human breast cancer cell line in culture. Further research is now needed to investigate whether oestrogenic responses are detectable using in vivo models and the extent to which these compounds might be absorbed through human skin and might enter human breast tissues. [source] Twenty-Four Hours Postoperative Results After Orthotopic Cardiac Transplantation in SwineJOURNAL OF CARDIAC SURGERY, Issue 4 2007Matthias Siepe M.D. However, there is no functional data available for a longer time period after transplantation. We have established a pig model to investigate myocardial function 24 hours after orthotopic transplantation.Materials and Methods: Orthotopic cardiac transplantations (HTx) in pigs were performed with a postoperative observation period of 24 hours (n = 6). To analyze myocardial function after transplantation, hemodynamical parameters (Swan-Ganz- and impedance-catheter data) as well as tissue and blood samples were obtained. Regional myocardial blood flow (RMBF) was assessed using fluorescent microspheres. Results: The impedance-catheter parameters demonstrated a preserved contractility in both ventricles 24 hours post-transplantation. In contrast, cardiac output 24 hours after HTx was diminished by 50% as compared to the preoperative value. Conversely, pulmonary vascular resistance increased significantly. The RMBF was increased in both ventricles. Metabolic and histological analyses indicate myocardial recovery 24 hours after HTx with no irreversible damage. Conclusions: For the first time, we were able to establish a porcine model to investigate myocardial function 24 hours after heart transplantation. While the contractility of the transplanted hearts was well-preserved, impaired cardiac output was going along with an increase in pulmonary vascular resistance. Using this clinical relevant model, improvements of human cardiac transplantation and post-transplant contractile dysfunction, especially, could be investigated. [source] On-line optical and X-ray spectroscopies with crystallography: an integrated approach for determining metalloprotein structures in functionally well defined statesJOURNAL OF SYNCHROTRON RADIATION, Issue 5 2008Mark J. Ellis X-ray-induced redox changes can lead to incorrect assignments of the functional states of metals in metalloprotein crystals. The need for on-line monitoring of the status of metal ions (and other chromophores) during protein crystallography experiments is of growing importance with the use of intense synchrotron X-ray beams. Significant efforts are therefore being made worldwide to combine different spectroscopies in parallel with X-ray crystallographic data collection. Here the implementation and utilization of optical and X-ray absorption spectroscopies on the modern macromolecular crystallography (MX) beamline 10, at the SRS, Daresbury Laboratory, is described. This beamline is equipped with a dedicated monolithic energy-dispersive X-ray fluorescence detector, allowing X-ray absorption spectroscopy (XAS) measurements to be made in situ on the same crystal used to record the diffraction data. In addition, an optical microspectrophotometer has been incorporated on the beamline, thus facilitating combined MX, XAS and optical spectroscopic measurements. By uniting these techniques it is also possible to monitor the status of optically active and optically silent metal centres present in a crystal at the same time. This unique capability has been applied to observe the results of crystallographic data collection on crystals of nitrite reductase from Alcaligenes xylosoxidans, which contains both type-1 and type-2 Cu centres. It is found that the type-1 Cu centre photoreduces quickly, resulting in the loss of the 595,nm peak in the optical spectrum, while the type-2 Cu centre remains in the oxidized state over a much longer time period, for which independent confirmation is provided by XAS data as this centre has an optical spectrum which is barely detectable using microspectrophotometry. This example clearly demonstrates the importance of using two on-line methods, spectroscopy and XAS, for identifying well defined redox states of metalloproteins during crystallographic data collection. [source] Caught in a Trap?ECONOMICA, Issue 268 2000Wage Mobility in Great Britain: 197 In this paper I study wage mobility in Great Britain using the New Earnings Surveys of 1975,94 and the British Household Panel Surveys of 1991,94. Measuring mobility in terms of decile transition matrices, I find a considerable degree of immobility within the wage distribution from one year to the next. Mobility is higher when measured over longer time periods. Those in lower deciles in the wage distribution are much more likely to exit into unemployment and non-employment. Measuring mobility by studying changes in individuals' actual percentile rankings in the wage distribution, I find evidence that short-run mobility rates have fallen since the late 1970s. This has potentially important welfare implications, given the rise in cross-section earnings inequality observed over the last two decades. [source] Comparison of conventional and ultrasound-assisted extraction of carvone and limonene from caraway seedsFLAVOUR AND FRAGRANCE JOURNAL, Issue 3 2004Smain Chemat Abstract Extraction experiments with hexane were carried out at atmospheric pressure in Soxhlet, conventional and ultrasound extractions of ,aked caraway seeds, and detailed results are given for two major plant extract components, carvone and limonene. The results indicate that carvone yield and plant extract quality are better in ultrasound extraction compared to those given by conventional methodology. Extraction rates of carvone and limonene reported that ultrasound-assisted extraction was 1.3,2 times more rapid, depending on temperature, than a conventional system, according to the rate constant obtained during the initial 10 minutes of extraction. Moreover, the SEM micrographs provided more evidence for the mechanical effects of ultrasound, mainly appearing on cell walls and shown by the destruction of cells, faulitating the release of their contents, in contrast to conventional maceration or extraction, which involve diffusion of plant extracts across glandular walls and causing cell rupture over longer time periods. Resco Summary 1A major research focus in population and community ecology is to establish a mechanistic understanding of plant interactions and demographic responses. The first step towards this mechanistic approach relies on understanding the differences in stress caused by different environmental conditions. Leaf-level photosynthetic rate (A) within and among plant populations provides important insight into population and community processes, but is difficult to acquire with sufficient replication under field conditions. Instead, chlorophyll fluorescence (Fv/Fm) and predawn water potential (,pd) are often used in arid and semi-arid ecosystems. 2Fv/Fm reflects the photoactivation status of photosystem II (PSII), whereas ,pd indicates water availability in the rhizosphere. Here we compare these indices with A in two perennial C4 grasses (native Heteropogon contortus and invasive Eragrostis lehmanniana) and in seedlings of the C3 shrub Prosopis velutina growing on highly contrasting sandy loam and loamy clay soils in experimental plots. Measurements were made the day prior to and up to 7 days following a 39-mm rainfall pulse after 2 months of drought. 3A was more sensitive across a broad range of environmental conditions, whereas Fv/Fm and ,pd only responded to periods of protracted drought. The use of these measures was further complicated because their values varied daily and we observed different time-lags in their response to precipitation pulses. 4We suggest sampling schemes and a priori measurements to capture the value that is representative for the question of interest, and that match the pulsed biological activity in these ecosystems. Finally, we suggest the use of these measures in combination with measurements providing integration over longer time periods, such as ,13C, ,18O and N concentration in bulk leaf tissue. [source] Genetic analysis of phenotypes derived from longitudinal data: Presentation Group 1 of Genetic Analysis Workshop 13GENETIC EPIDEMIOLOGY, Issue S1 2003Konstantin Strauch Abstract The participants of Presentation Group 1 used the GAW13 data to derive new phenotypes, which were then analyzed for linkage and, in one case, for association to the genetic markers. Since the trait measurements ranged over longer time periods, the participants looked at the time dependence of particular traits in addition to the trait itself. The phenotypes analyzed with the Framingham data can be roughly divided into 1) body weight-related traits, which also include a type 2 diabetes progression trait, and 2) traits related to systolic blood pressure. Both trait classes are associated with metabolic syndrome. For traits related to body weight, linkage was consistently identified by at least two participating groups to genetic regions on chromosomes 4, 8, 11, and 18. For systolic blood pressure, or its derivatives, at least two groups obtained linkage for regions on chromosomes 4, 6, 8, 11, 14, 16, and 19. Five of the 13 participating groups focused on the simulated data. Due to the rather sparse grid of microsatellite markers, an association analysis for several traits was not successful. Linkage analysis of hypertension and body mass index using LODs and heterogeneity LODs (HLODs) had low power. For the glucose phenotype, a combination of random coefficient regression models and variance component linkage analysis turned out to be strikingly powerful in the identification of a trait locus simulated on chromosome 5. Haseman-Elston regression methods, applied to the same phenotype, had low power, but the above-mentioned chromosome 5 locus was not included in this analysis. Faster growth in non-audit fees and longer time periods over which non-audit services are purchased might reduce the auditor's independence from that client. Our results do not provide any support for a relationship between non-audit fee growth rates or the length of time of the non-audit fee relationship with the client and discretionary accruals, our measure of earnings management. We do find some evidence that the interaction of the non-audit fee time-period measures and client importance is positive and significantly related to discretionary accruals. [source] Establishing native plants on newly-constructed and older-reclaimed sites along West Virginia highwaysLAND DEGRADATION AND DEVELOPMENT, Issue 4 2008J.G. Skousen Abstract Many state highway departments in the USA must use native plants for revegetating roadsides. We conducted two field studies in West Virginia to assess native plant establishment under two different conditions. On newly-constructed sites, native species were seeded alone or combined with non-native species. On older roadsides, native species were seeded in disturbed existing vegetation. In the first study, we used four seed mixtures comprised of seeds of native and non-native species, and two N-P-K fertilizer treatments at three newly-constructed sites. Native, warm-season grasses were slow to establish and only contributed 25 per cent cover in some plots after three years. Indiangrass (Sorghastrum nutans [L.] Nash), big bluestem (Andropogon gerardii Vitman), Brown-Eyed Susan (Rudbeckia triloba L.), and wild senna (Cassia hebecarpa Fernald) were the only seeded native species found. Fertilizer at 150,kg,ha,1 of 10-20-10 showed little influence on increasing plant cover. In the second study, we disturbed three different-aged established stands of vegetation composed of tall fescue (Festuca arundinacea Screb.) and crownvetch (Coronilla varia L.) by mowing, herbicide, or tillage, and native plants were seeded with and without fertilizer. Native cover was <10 per cent in all plots during the first year, but greatly increased by the second year to as much as 45 per cent in tilled plots, indicating that disturbance was necessary for natives to become important contributors within 2 years. Only switchgrass (Panicum virgatum L.), little bluestem (Andropogon scoparius Vitman), partridge pea (Chamaecrista fasciculate Michx.), and Brown-Eyed Susan were observed in plots. Fertilizer at 300,kg,ha,1 of 10-20-10 did not increase native plant cover on these sites. Based on our results, introducing or increasing the cover of native species along roadsides requires (1) reducing competition from non-native species, and (2) longer time periods for these slower-establishing species to be observed. Mendell MD Objective ,-Sarcoglycan deficiency results in a severe form of muscular dystrophy (limb-girdle muscular dystrophy type 2D [LGMD2D]) without treatment. Gene replacement represents a strategy for correcting the underlying defect. Questions related to this approach were addressed in this clinical trial, particularly the need for immunotherapy and persistence of gene expression. Methods A double-blind, randomized controlled trial using rAAV1.tMCK.hSGCA injected into the extensor digitorum brevis muscle was conducted. Control sides received saline. A 3-day course of methylprednisolone accompanied gene transfer without further immune suppression. Results No adverse events were encountered. SGCA gene expression increased 4,5-fold over control sides when examined at 6 weeks (2 subjects) and 3 months (1 subject). The full sarcoglycan complex was restored in all subjects, and muscle fiber size was increased in the 3-month subject. Adeno-associated virus serotype 1 (AAV1)-neutralizing antibodies were seen as early as 2 weeks. Neither CD4+ nor CD8+ cells were increased over contralateral sides. Scattered foci of inflammation could be found, but showed features of programmed cell death. Enzyme-linked immunospot (ELISpot) showed no interferon-, response to ,-SG or AAV1 capsid peptide pools, with the exception of a minimal capsid response in 1 subject. Restimulation to detect low-frequency capsid-specific T cells by ELISpot assays was negative. Results of the first 3 subjects successfully achieved study aims, precluding the need for additional enrollment. Interpretation The finding of this gene replacement study in LGMD2D has important implications for muscular dystrophy. Sustained gene expression was seen, but studies over longer time periods without immunotherapy will be required for design of vascular delivery gene therapy trials.
