Selected AbstractsTreatment development for systemic Tetrahymena sp. infection in guppies, Poecilia reticulata PetersJOURNAL OF FISH DISEASES, Issue 6 2010M Pimenta Leibowitz Abstract Antibacterial and antiparasitic agents and a cysteine protease inhibitor (E-64) were tested against Tetrahymena infection, a serious problem in guppy production worldwide. Chemicals were tested in vitro by a colorimetric assay for Tetrahymena survival. The most effective were niclosamide, albendazole and chloroquine, with 23%, 35% and 60% survival, respectively, following 2-h exposure to 100 ppm. Longer incubation periods resulted in greater reductions in survival. Niclosamide was further studied in vivo at different dosages, administered orally to Tetrahymena -infected guppies. Mortality rates were significantly lower in all treatment groups; in trial I, 30% and 33% mortality in 5 and 40 mg kg,1 niclosamide-fed fish vs. 59% mortality in controls; in trial II, 35%, 13% and 10% in 50, 100 and 200 mg kg,1 niclosamide-fed fish vs. 64% in controls. The effect of the cysteine protease inhibitor E64 was tested in tissue culture, by measuring histolytic activity of the parasite (Tet-NI) on a guppy-fin cell line, based on cell depletion. Tet-NI feeding activity was significantly reduced following pretreatment with E-64 relative to non-treated Tet-NI. E-64-pretreated Tet-NI was injected i.p. into guppies: recorded mortality rates were significantly lower (35%) than that in non-treated Tet-NI (60%), suggesting inhibition of the parasite's cysteine protease as a possible therapeutic approach. [source] Sensitive detection of Ralstonia solanacearum in soil: a comparison of different detection techniquesPLANT PATHOLOGY, Issue 4 2000P. M. Pradhanang The sensitivity and specificity of various methods were compared for routine detection of Ralstonia solanacearum in a sandy loam soil. Populations fewer than 102 CFU per g soil were detected by dilution plating on a modified semiselective medium (SMSA). In comparison, a tomato bioassay was shown consistently to detect populations at or greater than 7·5 × 105 CFU per g soil. An indirect enzyme-linked immunosorbent assay (ELISA) was as sensitive as the tomato bioassay, but detected as few as 104 CFU per g soil when the suspension was first incubated in SMSA broth prior to testing. Detection using a nested polymerase chain reaction (PCR) was equally as sensitive as that using culture on SMSA agar, but only when the infested soil sample was first enriched overnight in SMSA broth prior to the nested PCR. Longer incubation periods in SMSA broth also increased the sensitivity of pathogen detection using a conventional PCR method, permitting detection of as few as 102 CFU per g soil after 60 h enrichment in SMSA broth. When evaluated using naturally infected field soils in Nepal, isolation of R. solanacearum on SMSA was reliable only when pathogen populations were higher than those of saprophytic soilborne bacteria. As few as 5 × 102 CFU of R. solanacearum per g were recovered from naturally infested soil, whereas the sensitivity of indirect ELISA was 106 CFU g,1. TIELEMAN Summary 1The wide range in incubation periods among bird species has puzzled biologists for decades, because an extended egg-phase increases time-dependent mortality of the eggs. 2We investigated a recently proposed mechanistic explanation inspired by life-history theory, suggesting that adults may increase their own survival by reducing nest attentiveness, the percentage of daytime spent incubating eggs, in exchange for reduced offspring (egg) survival due to a longer incubation period. Incubation behaviour and egg temperatures (Tegg) of 14 bird species in the humid lowland tropics were studied to test the hypothesis that lower nest attentiveness and reduced Tegg cause longer incubation periods. 3Increased nest attentiveness correlated with higher average Tegg. However, neither nest attentiveness nor average Tegg was associated with the length of the incubation period. Longer off-bouts resulted in lower Tegg, but neither number of off-bouts nor off-bout length was associated with incubation period. In addition, we reanalysed a previously published negative association between Tegg and incubation period based on literature data from temperate passerine birds using a larger data set and found no significant correlation. 4In conclusion, our results do not support the hypothesis that longer incubation periods are caused by reduced nest attentiveness and corresponding lower Tegg. Mamidi Abstract This study demonstrates the utility of an in vitro , in vivo correlative approach in the selection and optimization of a prodrug candidate of celecoxib (CBX), a COX2 inhibitor. As an initial screening step, a comparative single oral dose pharmacokinetic study was conducted in rats for CBX and its three aliphatic acyl water-soluble prodrugs viz., CBX-acetyl (CBX-AC), CBX-propionyl (CBX-PR) and CBX-butyryl (CBX-BU) at high equimolar dose, 100 mg/kg. Only CBX-BU and CBX-PR converted rapidly to CBX and yielded approximately five-fold greater systemic exposure of CBX than CBX alone or CBX-AC. Rank order of systemic exposure of prodrugs in its intact form was CBX-AC >CBX-PR >CBX-BU. Further in vitro hydrolysis studies of CBX prodrugs in intestinal mucosal suspensions and liver homogenates indicated that CBX-BU is rapidly and completely converted to CBX, whereas CBX-PR and CBX-AC require longer incubation period for complete conversion to CBX. There was a very good correlation of the in vitro and in vivo data supporting CBX-BU as the prodrug of choice. Further in vitro pharmacological studies showed that COX2 selective inhibition is improved for CBX-BU as compared to CBX-AC and CBX-PR. Dose proportionality in pharmacokinetic studies of CBX-BU and CBX at equimolar oral doses confirmed that relative oral bioavailability of CBX was improved following CBX-BU administration and there was linearity in pharmacokinetics of CBX over a wide dose range (10,100 mg/kg), whereas CBX in its conventional form showed poor bioavailability and lack of dose linearity in pharmacokinetics. The oral bioavailability of CBX from CBX-BU was dose independent and was in the range 78,96%. At a 50% reduced molar dose, CBX-BU showed an equivalent efficacy to that of CBX in the in vivo carrageenan model. [source] Influence of copper-alloying of austenitic stainless steel on multi-species biofilm developmentLETTERS IN APPLIED MICROBIOLOGY, Issue 2 2001J. Kielemoes Aims: To investigate the bactericidal influence of copper-alloying of stainless steel on microbial colonization. Methods and Results: Inhibition of bacterial adherence was investigated by monitoring (192 h) the development of a multi-species biofilm on Cu-alloyed (3·72 wt%) stainless steel in a natural surface water. During the first 120 h of exposure, lower numbers of viable bacteria in the water in contact with copper-containing steel relative to ordinary stainless steel were observed. Moreover, during the first 48 h of exposure, lower colony counts were found in the biofilm adhering to the Cu-alloyed steel. No lower colony or viable counts were found throughout the remainder of the experimental period. Conclusions: The presence of Cu in the steel matrix impedes the adhesion of micro-organisms during an initial period (48 h), while this bactericidal effect disappears after longer incubation periods. Significance and Impact of the Study: The application of Cu-alloyed stainless steels for bactericidal purposes should be restricted to regularly-cleaned surfaces. [source] |
