Limited Distribution of a Cardiomyopathy-Associated Variant in India

Summary Heart failure is a leading cause of death of people in South Asia, and cardiomyopathy is a major cause of heart failure. Myosin binding protein C (MYBPC3) is expressed in the heart muscle, where it regulates the cardiac response to adrenergic stimulation and is important for the structural integrity of the sarcomere. Mutations in the MYBPC3 gene are associated with hypertrophic or dilated cardiomyopathies. A 25-base-pair deletion in intron 32 causes skipping of the downstream exon and is associated with familial cardiomyopathy. To date, this deletion is found primarily in India and South Asia, although it is also found at low frequency in Southeast Asia. In order to better characterise the distribution of this variant, we determined its frequency in 447 individuals from 19 populations, including 10 populations from India and neighbouring populations from Pakistan and Nepal. The deletion frequency is over 8% in some of our Indian samples, and it is not present in any of the populations we sampled outside of India. The differences in the deletion frequencies among populations in India are consistent with patterns of variation previously reported and with patterns we observed among Indian populations based on high-density SNP chip data. Our results indicate that the MYBPC3 deletion is primarily found among Indian populations and that its distribution is consistent with genome-wide patterns of variation in India. [source]


EVIDENCE FOR HISTORICAL INTROGRESSION ALONG A CONTACT ZONE BETWEEN TWO SPECIES OF CHAR (PISCES: SALMONIDAE) IN NORTHWESTERN NORTH AMERICA

Abstract Phylogeographic analyses can yield valuable insights into the geographic and historical contexts of contact and hybridization between taxa. Two species of char (Salmonidae), Dolly Varden (Salvelinus malma) and bull trout (S. confluentus) have largely parapatric distributions in watersheds of northwestern North America. They are, however, sympatric in several localities and hybridization and some introgression occurs across a broad area of contact. We conducted a comparative phylogenetic analysis of Dolly Varden and bull trout to gain a historical perspective of hybridization between these species and to test for footprints of historical introgression. We resolved two major Dolly Varden mitochondrial DNA (mtDNA) clades (with 1.4,2.2% sequence divergence between haplotypes) that had different geographical distributions. Clade N is distributed across most of the range of Dolly Varden, from southern British Columbia through to the Kuril Islands in Asia. Clade S had a much more limited distribution, from Washington state, at the southern limit of the Dolly Varden range, to the middle of Vancouver Island. The distribution and inferred ages of the mtDNA clades suggested that Dolly Varden survived the Wisconsinan glaciation in a previously unsuspected refuge south of the ice sheet, and that Dolly Varden and bull trout were probably in continuous contact over most of the last 100,000 years. When bull trout were included in the phylogenetic analysis, however, the mtDNA of neither species was monophyletic: Clade S Dolly Varden clustered within the bull trout mtDNA clade. This pattern was discordant with two nuclear phylogenies produced (growth hormone 2 and rRNA internal transcribed sequence 1), in which Dolly Varden and bull trout were reciprocally monophyletic. This discordance between mtDNA- and nDNA-based phylogenies indicates that historical introgression of bull trout mtDNA into Dolly Varden occurred. Percent sequence divergence within these introgressed Dolly Varden (clade S) was 0.2,0.6%, implying that the introgression occurred prior to the most recent glaciation. Our analysis and other evidence of contact between divergent lineages in northwestern North America strongly suggests that the area may be the site of previously unsuspected suture zones of aquatic biotas. Stratigraphic and Morphologic Constraints on the Weichselian Glacial History of Northern Prins Karls Forland, Western Svalbard

Uncertainty remains if ice,free marginal areas existed on the west coast of Svalbard during the Late Weichselian. Field mapping and correlation to well dated raised beach sequences on nearby Brøggerhalvøya reveal the existence of two generations of raised beach deposits on northern Prins Karls Forland. Distinct beach ridges rise up to the inferred Late Weichselian marine limit at 18 m a.s.l. Discontinuous pre,Late Weichselian beach deposits rise from the Late Weichselian marine limit up to approximately 60 m a.s.l. Expansion of local glaciers during the Late Weichselian is indicated by the limited distribution of a till that overlies parts of the older beach sequence. Stratigraphic data and chronological control indicate deposition in a shallow marine environment before 50 ka bp. Correlation to stratigraphic sites on western Svalbard suggests deposition at c. 70 ±10 ka. Glaciotectonic structures disclose expansion of local glaciers into the For,landsundet basin during stage 4 or late stage 5 high relative sea level. Palaeotemperature estimates derived from amino acid ratios indicate that during the time interval c. 70 to 10 ka the area was exposed to cold subaerial temperatures with low rates of racemization. Pedogenesis and frost,shattered clasts at the contact between c. 70 ka deposits and Holocene deposits further indicate a prolonged period of subaerial polar desert conditions during this time interval. The evidence suggests that the Barents Sea ice sheet did not extend across northern Prins Karls Forland during the Weichselian. It is inferred that during the Late Weichselian, ice was drained throughout the major fjords on the west coast of Svalbard and that relatively large marginal areas experienced polar desert conditions and minor expansions of local glaciers. Spatial and ontogenetic variability in the microhabitat use of stream-dwelling spined loach (Cobitis taenia) and stone loach (Barbatula barbatula)

Summary Ontogenetic and spatial variability in microhabitat use of spined loach Cobitis taenia (Linnaeus), considered as one species for the purposes of this study, and stone loach Barbatula barbatula (Linnaeus) were examined in the River Great Ouse basin, England, using multivariate and habitat suitability methods, including a technique for handling spatial variation in collections of preference curves. Distinct ordinations of spined age classes and stone loach developmental stages, respectively, in canonical correspondence analysis of species × variables × samples relationships suggest that the two species occupy completely different microhabitats; however, young-of-the-year spined loach occurred more often than expected with all developmental stages of stone loach except young larvae. Water velocity and filamentous algae were the most influential microhabitat variables, the latter decreasing in importance with increasing age of both fish species. Preferred water velocities generally decreased with age in spined loach and increased in stone loach, with substratum size generally increasing with fish age in both species. Spatial variation in microhabitat preferences was great in both species but less so in the spined loach, suggesting that limited plasticity in habitat use could account, at least in part, for the latter species' limited distribution and abundance in the catchment. Preference curves for a species, if generated and verified for all life intervals and all seasons, could be used as a management tool for a given stream or sector of river basin. But preference curves should be generated for each location to ensure that river management decisions with regard habitat and species conservation consider local-level species requirements. Thus, a multi-(eco)species and multi-scale approach is required in habitat suitability assessments. [source]


In situ chemical oxidation of residual LNAPL and dissolved-phase fuel hydrocarbons and chlorinated alkenes in groundwater using activated persulfate,

A treatablity study (TS) was conducted to evaluate the efficacy of in situ chemical oxidation (ISCO) using activated persulfate, alone and in combination with air sparging (AS), for treating a source area contaminated with residual light nonaqueous-phase liquid (LNAPL), dissolved-phase fuel hydrocarbons (HCs), and dissolved-phase chlorinated alkenes at Edwards Air Force Base (AFB), California. The TS was implemented in two phases. Phase I included injecting a solution of sodium persulfate and sodium hydroxide (NaOH) into groundwater via an existing well where residual LNAPL and dissolved-phase contaminants were present. Because the results of Phase I indicated a limited distribution of the activated persulfate, Phase II was performed to assess whether AS could enhance the distribution of the sodium persulfate. Each phase was followed by groundwater monitoring and sampling at the injection well and at three monitoring wells, located 20 to 44 feet from the injection well. Results from Phases I and II of the TS indicated that (1) alkaline-activated persulfate was effective in promoting the dissolution of LNAPL and the degradation of dissolved-phase contaminants, but only at the injection well; (2) the addition of AS was effective in enhancing the radius of persulfate distribution from less than 20 feet to greater than 44 feet, and (3) persulfate alone (i.e., not in an activated state) was effective in reducing the concentrations of dissolved-phase fuel HC and chlorinated alkenes. © 2009 Wiley Periodicals, Inc. Distribution of Lectin-Bindings in the Testis of the Lesser Mouse Deer, Tragulus javanicus

Summary The distribution of lectin bindings in the testis of the smallest ruminant, lesser mouse deer (Tragulus javanicus), was studied using 12 biotinylated lectins specific for d -galactose (peanut agglutinin PNA, Ricinus communis agglutinin RCA I), N -acetyl- d -galactosamine (Dolichos biflorus agglutinin DBA, Vicia villosa agglutinin VVA, Soybean agglutinin SBA), N -acetyl- d -glucosamine and sialic acid (wheat germ agglutinin WGA, s-WGA), d -mannose and d -glucose (Lens culinaris agglutinin LCA, Pisum sativum agglutinin PSA, Concanavalin A Con A), l -fucose (Ulex europaeus agglutinin UEA I), and oligosaccharide (Phaseolus vulgaris agglutinin PHA-E) sugar residues. In Golgi-, cap-, and acrosome-phase spermatids, lectin-bindings were found in the acrosome (PNA, RCA I, VVA, SBA, WGA and s-WGA), and in the cytoplasm (PNA, RCA I, VVA, SBA, WGA, LCA, PSA, Con A and PHA-E). s-WGA binding was confined to the spermatid acrosome, but other lectins were also observed in spermatocytes. In spermatogonia, VVA, WGA, Con A, and PHA-E bindings were observed. Sertoli cells were intensely stained with DBA and Con A, and weakly with PHA-E. In interstitial Leydig cells, RCA I, DBA, VVA, Con A, PSA, LCA, WGA and PHA-E were positive. UEA I was negative in all cell types including spermatogenic cells. Unusual distribution of lectin-bindings noted in the testis of lesser mouse deer included the limited distribution of s-WGA only in the spermatid acrosome, the distribution of DBA in Sertoli cells, Leydig cells and lamina propria, and the absence of UEA I in all type cells. The present results were discussed in comparison with those of other animals and their possible functional implications. [source]
