Selected AbstractsTHE ANTARCTIC PSYCHROPHILE, CHLAMYDOMONAS RAUDENSIS ETTL (UWO241) (CHLOROPHYCEAE, CHLOROPHYTA), EXHIBITS A LIMITED CAPACITY TO PHOTOACCLIMATE TO RED LIGHT,JOURNAL OF PHYCOLOGY, Issue 4 2005Rachael M. Morgan-Kiss The psychrophilic Antarctic alga, Chlamydomonas raudensis Ettl (UWO241), grows under an extreme environment of low temperature and low irradiance of a limited spectral quality (blue-green). We investigated the ability of C. raudensis to acclimate to long-term imbalances in excitation caused by light quality through adjustments in photosystem stoichiometry. Log-phase cultures of C. raudensis and C. reinhardtii grown under white light were shifted to either blue or red light for 12 h. Previously, we reported that C. raudensis lacks the ability to redistribute light energy via the short-term mechanism of state transitions. However, similar to the model of mesophilic alga, C. reinhardtii, the psychrophile retained the capacity for long-term adjustment in energy distribution between PSI and PSII by modulating the levels of PSI reaction center polypeptides, PsaA/PsaB, with minimal changes in the content of the PSII polypeptide, D1, in response to changes in light quality. The functional consequences of the modulation in PSI/PSII stoichiometry in the psychrophile were distinct from those observed in C. reinhardtii. Exposure of C. raudensis to red light caused 1) an inhibition of growth and photosynthetic rates, 2) an increased reduction state of the intersystem plastoquinone pool with concomitant increases in nonphotochemical quenching, 3) an uncoupling of the major light-harvesting complex from the PSII core, and 4) differential thylakoid protein phosphorylation profiles compared with C. reinhardtii. We conclude that the characteristic low levels of PSI relative to PSII set the limit in the capacity of C. raudensis to photoacclimate to an environment enriched in red light. [source] The Role of Support Networks in the Initial Stages of Integration: The Case of West African Newcomers in the NetherlandsINTERNATIONAL MIGRATION, Issue 2 2010Magali Chelpi-den Hamer This study focuses on the role of support networks in West African migration to the Netherlands. The authors examine the interactions of non-State support groups with West African newcomers and explore the nature, scope and function of support. What type of support networks do respondents rely on, under which circumstances, and for how long? What is the scope of support? Is it to be taken for granted? What alternatives are there for those left out of the loop? The findings suggest that support is mainly punctual, in response to a specific need, and for a limited period of time. It is not to be taken for granted, as support groups have limited capacities, and protect themselves from excessive demands. They play a significant role in providing assistance to migrants, yet this role should not be overestimated. [source] Regulation of nickel-induced T-cell responsiveness by CD4+CD25+cells in contact allergic patients and healthy individualsCONTACT DERMATITIS, Issue 2 2005H. Moed In this study, we investigated the capacity of CD4+CD25+ regulatory T cells to suppress nickel-specific effector T cells, both in nickel-allergic patients and healthy controls. CD4+ cells isolated from allergic patients showed an increased proliferative response to nickel, whereas CD4+ cells from negative controls did not respond to allergen. When CD4+CD25+ cells were depleted, nickel-specific responsiveness was strongly increased both in allergic and in non-allergic individuals, with the most pronounced effect in allergic patients. These regulatory T cells were anergic to nickel but inhibited nickel-specific CD4+CD25, effector T cells in coculture experiments. CD4+CD25+ cells from nickel-allergic patients showed only a limited capacity to suppress effector T-cell responsiveness, because an increased nickel reactivity could still be detected in these cocultures. None of the isolated CD4+CD25+ cells, either isolated from healthy controls or allergic patients, produced IL-10 in response to nickel. Overall, these results support the view that CD4+CD25+ cells can control the activation of nickel-specific effector T cells in non-allergic individuals, whereas this regulatory capacity is impaired in allergic patients. To investigate the presence of allergen-specific regulatory T cells in truly naïve, non-sensitized individuals, T-cell reactivity should also be studied with non-environmental contact allergens, such as para-phenylenediamine. [source] Channel Strategies and Stocking Policies in Uncapacitated and Capacitated Supply Chains,DECISION SCIENCES, Issue 2 2002Jayashree Mahajan ABSTRACT A supply chain consisting of a single supplier distributing two independent products through multiple retailers is analyzed in this paper. The supplier needs to incentivize its retailers to adopt stocking policies that are mutually advantageous and that result in the optimal level of market coverage. The focus is on determining the optimal stocking policies for retailers and the resulting distribution strategy given that the supplier has either unlimited or limited capacity. The results provide insights on the optimal distribution strategy and stocking policies for the supply chain. In general, the paper shows that it is optimal for the supplier to use an intensive distribution strategy (i.e., the products are stocked by all retailers). Selective or exclusive strategies are optimal only when retailers are risk averse, stocking synergies exist, and there are differences in demand or supply uncertainties across products. The analysis also shows that retailers hold larger stocks of a product which generates higher supplier margins but only when the supplier has unlimited capacity. If the supplier has limited capacity, then their margins have no effect on retailers' stocking decisions. Contrary to conventional wisdom, retailers hold larger stocks of a product that has less demand uncertainty as compared to one that has more demand uncertainty. [source] Identification of novel markers expressed during fin regeneration by microarray analysis in medaka fishDEVELOPMENTAL DYNAMICS, Issue 9 2007Masanobu Nishidate Abstract Urodeles and fish have a remarkable ability to regenerate lost body parts, whereas many higher vertebrates, including mammals, retain only a limited capacity. It is known that the formation of specialized cell populations such as the wound epidermis or blastema is crucial for regeneration; however, the molecular basis for their formation has not been elucidated. Recently, approaches using differential display and microarray have been done in zebrafish for searching molecules involved in regeneration. Here, we used the medaka fish, a distantly diverged fish species, for microarray screening of transcripts up-regulated during regeneration. By setting criteria for selecting transcripts that are reliably and reproducibly up-regulated during regeneration, we identified 140 transcripts. Of them, localized in situ expression of 12 transcripts of 22 tested was detected either in differentiating cartilage, basal wound epidermis, or blastema. [source] East Timor Emerging from Conflict: The Role of Local NGOs and International AssistanceDISASTERS, Issue 1 2001Ian Patrick International assistance efforts have represented a conundrum for East Timorese seeking to assert their new independence and autonomy. While urgent needs have been met, local participation, involvement and capacity building have not been given adequate attention. This outcome is aptly demonstrated in the case of local non-government organisations (LNGOs). This paper specifically examines the role of LNGOs in the recovery of East Timor within the international assistance programme. It examines the challenges of rehabilitation efforts in East Timor with a particular focus on capacity building of East Timorese NGOs as part of a broader effort to strengthen civil society. The initial crisis response in East Timor highlighted tension between meeting immediate needs while simultaneously incorporating civil society actors such as NGOs and communities. It has been argued that local NGOs and the community at large were not sufficiently incorporated into the process. While it is acknowledged that many local NGOs had limited capacity to respond, a greater emphasis on collaboration, inclusion and capacity building was desirable, with a view to supporting medium and longer term objectives that promote a vibrant civil society, sustainability and self-management. [source] Post-European settlement response gradients of river sensitivity and recovery across the upper Hunter catchment, AustraliaEARTH SURFACE PROCESSES AND LANDFORMS, Issue 7 2009Kirstie Fryirs Abstract Most analyses of river adjustment have focused on parts of catchments where metamorphosis has occurred. This provides a non-representative view of river responses to human-disturbance. Although many rivers have been subjected to systematic land-use change and disturbance, significant variability is evident in the form, extent and consequences of adjustment. This study documents the catchment-wide distribution of river sensitivity and adjustment in the upper Hunter catchment, New South Wales, Australia in the period since European settlement. The spatial distribution and timing of lateral, vertical and wholesale river adjustments are used to assess river sensitivity to change. The type and pattern of rivers, influenced largely by valley setting, have induced a fragmented pattern of river adjustment in the upper Hunter catchment. Adjustments have been largely non-uniform and localized, reflecting the predominance of bedrock-controlled rivers which have limited capacity to adjust and are resilient to change. Less than 20% of river courses have experienced metamorphosis. Phases of reach-scale geomorphic adjustment to human disturbance are characterized as a gradient of primary, secondary and tertiary responses. In general terms, primary responses such as cutoffs or straightening were followed by secondary responses such as channel expansion. These secondary responses occurred between 50,70 years after initial disturbance. A subsequent tertiary phase of river recovery, denoted as a transition from predominantly erosional to predominantly depositional geomorphic processes such as channel contraction, occurred around 70,120 years after initial disturbance. [source] The influence of strenuous exercise on collagen characteristics of articular cartilage in Thoroughbreds age 2 yearsEQUINE VETERINARY JOURNAL, Issue 6 2000P. A. J. BRAMA Summary In order to assess the influence of strenuous exercise on collagen characteristics of articularcartilage, the response of the collagen network was studied in seven 2-year-old Thoroughbreds subjected to strenuous exercise compared to 7 nontrained individuals. After 13 weeks, the animals were subjected to euthanasia, fetlock joints of the forelimbs were scored macroscopically after Indian Ink staining, and articular cartilage from different locations of the articular surface of the proximal first phalanx was sampled and analysed for water content, collagen content, hydroxylysine content and amount of hydroxylysylpyridinoline (HP) crosslinks. Gross lesions were significantly more severe in the exercised than in the nonexercised group. In the control animals, the characteristic site-specific differences in collagen parameters were found as described earlier, but in the strenuously exercised animals this physiological biochemical heterogeneity had disappeared. In the exercised animals, an increase in water content and a sharp decrease in HP crosslinking was found that was correlated with the presence of wear lines. It is concluded that the strenuous exercise provoked significant alterations in the characteristics of the collagen network of the articular cartilage of the fetlock joint which were suggestive of microdamage and loosening of the collagen network. The collagen component of cartilage, in contrast to the proteoglycan component, is known to have a very limited capacity for repairand remodelling due to an extremely low turnover rate. Therefore, alterations within the articular collagen network might be expected to play an important role in the pathophysiology of degenerative joint disorders. [source] Cardiovascular Response to Graded Lower Body Negative Pressure in Young and Elderly ManEXPERIMENTAL PHYSIOLOGY, Issue 3 2001R. van Hoeyweghen Lower body negative pressure (LBNP) reduces central venous pressure (CVP) and cardiac output. The elderly are reported to have a limited capacity to increase cardiac output by increasing heart rate (HR), are especially dependent on end diastolic volume to maintain stroke volume and therefore should be especially vulnerable to LBNP. The present study compared the effects of LBNP in the young and old. Stroke volume was assessed non-invasively as stroke distance (SD) by aortovelography. Two groups of healthy male volunteers were studied: eight young (29.7 ± 2.0 years, mean ± S.E.M.) and nine old (70.1 ± 0.9 years). LBNP was applied progressively at 17.5, 35 and 50 mmHg in 20 min steps, with measurements taken during each steady state. There were similar, significant, falls in CVP in both groups. SD fell significantly in both groups from respective control values of 24.8 ± 1.6 and 16.6 ± 0.9 cm to 12.5 ± 1.3 and 8.9 ± 0.4 cm at a LBNP of 50 mmHg. Although SD in the elderly was significantly lower than in the young, the LBNP-induced changes were not different between groups. Both groups produced similar significant increases in vascular resistance, HR, plasma vasopressin (AVP) and noradrenaline. Mean arterial blood pressure (MBP) and plasma adrenaline did not change significantly. Therefore healthy old men respond to LBNP in a similar manner to the young, although MBP and SD are regulated around different baselines in the two groups. [source] Long-term carbon exchange in a sparse, seasonally dry tussock grasslandGLOBAL CHANGE BIOLOGY, Issue 10 2004John E. Hunt Abstract Rainfall and its seasonal distribution can alter carbon dioxide (CO2) exchange and the sustainability of grassland ecosystems. Using eddy covariance, CO2 exchange between the atmosphere and a sparse grassland was measured for 2 years at Twizel, New Zealand. The years had contrasting distributions of rain and falls (446 mm followed by 933 mm; long-term mean=646 mm). The vegetation was sparse with total above-ground biomass of only 1410 g m,2. During the dry year, leaf area index peaked in spring (November) at 0.7, but it was <0.2 by early summer. The maximum daily net CO2 uptake rate was only 1.5 g C m,2 day,1, and it occurred before mid-summer in both years. On an annual basis, for the dry year, 9 g C m,2 was lost to the atmosphere. During the wet year, 41 g C m,2 was sequestered from the atmosphere. The net exchange rates were determined mostly by the timing and intensity of spring rainfall. The components of ecosystem respiration were measured using chambers. Combining scaled-up measurements with the eddy CO2 effluxes, it was estimated that 85% of ecosystem respiration emanated from the soil surface. Under well-watered conditions, 26% of the soil surface CO2 efflux came from soil microbial activity. Rates of soil microbial CO2 production and net mineral-N production were low and indicative of substrate limitation. Soil respiration declined by a factor of four as the soil water content declined from field capacity (0.21 m3 m,3) to the driest value obtained (0.04 m3 m,3). Rainfall after periods of drought resulted in large, but short-lived, respiration pulses that were curvilinearly related to the increase in root-zone water content. Coupled with the low leaf area and high root : shoot ratio, this sparse grassland had a limited capacity to sequester and store carbon. Assuming a proportionality between carbon gain and rainfall during the summer, rainfall distribution statistics suggest that the ecosystem is sustainable in the long term. [source] Anti-oxidant activity of added tea catechins on lipid oxidation of raw minced red meat, poultry and fish muscleINTERNATIONAL JOURNAL OF FOOD SCIENCE & TECHNOLOGY, Issue 6 2001Shuze Tang The comparative anti-oxidative effects of added tea catechins (TC) and ,-tocopherol to raw minced red meat (beef and pork), poultry (chicken, duck and ostrich) and fish (whiting and mackerel) muscle on susceptibility to lipid oxidation were investigated during 10 days of refrigerated (4 °C) display. Fresh meats, poultry and fish, purchased from a local market, were trimmed to remove bones, skin and surface fat and minced through a 4 mm plate. The minced muscle of each species was treated with either the addition of 300 mg TC kg,1 minced muscle (TC300) or 300 mg ,-tocopherol kg,1 minced muscle (VE300). Minced muscle without any additives served as control (C). Oxidative stability (TBARS) was measured at 3-day intervals. Total lipids, fatty acid composition, total iron and haem iron from minced muscle for each species were also analysed. The susceptibility of untreated minced muscle to lipid oxidation was in the decreasing order: mackerel > beef > duck > ostrich > pork , chicken > whiting. This may be because of the different content of total fat, iron and fatty acid composition between species. The TC300 significantly (P < 0.05) reduced lipid oxidation compared with controls for all seven species as shown by lower TBARS values. The anti-oxidant potential of TC was two to fourfold greater than that of ,-tocopherol at the same concentration and this potential was species dependent. The VE300 showed limited capacity in inhibiting lipid oxidation for pork, chicken, duck and whiting. The results obtained show that TCs are powerful natural antioxidants when used in minced muscle food. This paper asserts that routine standardized outcome measurement in its current form can only provide a crude and narrow lens through which to witness recovery. It has only a limited capacity to capture the richness of people's recovery journeys or provide information that can usefully inform care. Indeed, in its implementation nurses may be required to collude in practices or account for practice in ways which run counter to the personal recovery paradigm. Nurses should view a focus on outcomes as an opportunity for critical reflection as well as to seek ways to account for recovery stories in meaningful ways. It provides results of an experimental implementation developed for this purpose in a wireless LAN (WLAN) environment. Multimedia streaming is an application that is gaining in popularity for mobile devices and in particular mobile Internet-based content broadcasting is rapidly emerging as a key feature on mobile devices. In these scenarios, the wireless link (last hop) is normally the performance bottleneck due to the dynamic and limited capacity of the wireless medium. The use of ULLA in this context can provide the ability to tailor the video transmission to the wireless link performance and also to configure the links in response to performance problems or environmental changes. For this purpose the focus of multimedia streaming has been on WLAN link technology and dynamic adaptation (i.e., dynamic channel selection and video transcoding) using a dynamic resource reservation overlay protocol. Given the limited capacity of the satellite component, to meet the diverse quality of service (QoS) demands of multimedia applications, it is highly desired that the available resources can be adaptively utilized in an optimized way. In this paper, we draw our attention on the development and evaluation of an efficient packet scheduling scheme in a representative broadband satellite system, namely satellite digital multimedia broadcasting (SDMB), which is positioned as one of the most attractive solutions in the convergence of a closer integration with the terrestrial mobile networks for a cost-effective delivery of point-to-multipoint services. By taking into account essential aspects of a successful QoS provisioning while preserving the system power/resource constraints, the proposed adaptive multidimensional QoS-based (AMQ) packet scheduling scheme in this paper aims to effectively satisfy diverse QoS requirements and adaptively optimize the resource utilization for the satellite multimedia broadcasting. The proposed scheme is formulated via an adaptive service prioritization algorithm and an adaptive resource allocation algorithm. By taking into account essential performance criteria, the former is capable of prioritizing contending flows based on the QoS preferences and performance dynamics, while the latter allocates the resources, in an adaptive manner, according to the current QoS satisfaction degree of each session. Simulation results show that the AMQ scheme achieves significantly better performance than those of existing schemes on multiple performance metrics, e.g. delay, throughput, channel utilization and fairness. [source] A genetic algorithm for a bicriteria supplier selection problemINTERNATIONAL TRANSACTIONS IN OPERATIONAL RESEARCH, Issue 2 2009Chuda Basnet Abstract In this paper, we discuss the problem of selecting suppliers for an organisation, where a number of suppliers have made price offers for supply of items, but have limited capacity. Selecting the cheapest combination of suppliers is a straightforward matter, but purchasers often have a dual goal of lowering the number of suppliers they deal with. This second goal makes this issue a bicriteria problem , minimisation of cost and minimisation of the number of suppliers. We present a mixed integer programming (MIP) model for this scenario. Quality and delivery performance are modelled as constraints. Smaller instances of this model may be solved using an MIP solver, but large instances will require a heuristic. We present a multi-population genetic algorithm for generating Pareto-optimal solutions of the problem. The performance of this algorithm is compared against MIP solutions and Monte Carlo solutions. Wong Abstract The Dial-a-Ride Problem (DARP) consists of planning routes and schedules for picking up and delivering users within user-specified time windows. Vehicles of a given fleet with limited capacity depart from and end at a common depot. The travel time of passengers cannot exceed a given multiple of the minimum ride time. Other constraints include vehicle capacity and vehicle route duration. In practice, scheduling is made more complicated by special user requirements and an inhomogeneous vehicle fleet. The transportation of elderly and handicapped people is an important example, as space for wheelchairs is limited and a lift is required. In this study, we present a modified insertion heuristic to solve the DARP with multi-dimensional capacity constraints, and the performance of the proposed algorithm is tested in simulation. We show that the proposed methodology is effective when compared with the classic algorithms. Johnson Summary 1As many species of marine benthic invertebrates have a limited capacity for movement as adults, dispersal mode is often considered as a determinant of geographical ranges, genetic structure and evolutionary history. Species that reproduce without a larval stage can only disperse by floating or rafting. It is proposed that the colonization processes associated with such direct developing species result in spatial distributions that show relatively greater fine scale patchiness than the distributions of species with a larval dispersal stage. This hypothesis was tested by collecting molluscs at different spatial scales in the Isle of Man. 2Spatial distribution patterns supported the predictions based on dispersal mode. Estimated variance components for species with larval dispersal suggested that the majority of the spatial variation was associated with variation between shores. In comparison, there was relatively more variability within shores for abundance counts of species with direct development. 3Multivariate analyses reflected the univariate results. An assemblage of direct developers provided a better discrimination between sites (100 m separation) but the group of species with larval dispersal gave a clearer separation of shores (separated by several km). 4The fine scale spatial structure of direct developing species was reflected in higher average species diversity within quadrats. Species richness also reflected dispersal mode, with a higher fraction of the regional species pool present for direct developers in comparison to species with larval dispersal. This may reflect the improved local persistence of taxa that avoid the larval dispersal stage. We assessed the views of researchers, decision makers and research policy makers on how the development and usage of evidence from systematic reviews can be promoted in a country with limited resources. Methods, We surveyed 131 participants in six systematic review workshops for their views on important items influencing the production and usage of systematic reviews in a developing country. They were also asked to propose interventions to deal with potential barriers. We analysed the quantitative data using multidimensional scaling methods, and the qualitative data using content analysis approach. Results, We identified seven clusters of items that contribute to the promotion of conducting and using systematic reviews. For each cluster a set of interventions are proposed that health care decision makers and research policy makers may use for promoting conduct and use of systematic reviews. The clusters are ,importance for policy makers', ,access to international research', ,priority and support for systematic reviews', ,competency and willingness of researchers to conduct reviews', ,importance for end-users', ,quality of local primary research' and ,visibility and access to local research'. Discussion, The proposed interventions focus on national level initiatives for making the systematic reviews ,wanted' and improving the capacity to conduct research. Our findings emphasize the essential role of policy makers for promoting systematic reviews. They demonstrate that many barriers stem from the lower quality of and lack of access to primary research originating from developing countries. [source] Temperature-dependent changes in energy metabolism, intracellular pH and blood oxygen tension in the Atlantic codJOURNAL OF FISH BIOLOGY, Issue 6 2003F. J. Sartoris The effect of acute increase in temperature on oxygen partial pressure (Po2) was measured in the gill arches of Atlantic cod Gadus morhua between 10 and 19° C by use of oxygen microoptodes. Oxygen saturation of the gill blood under control conditions varied between 90 and 15% reflecting a variable percentage of arterial or venous blood in accordance with the position of each optode in the gill arch. The data obtained suggested that arterial Po2 remained more or less constant and arterial oxygen uptake did not become limiting during warming. A progressive drop in venous Po2, however, was observed at >10° C indicating that excessive oxygen uptake from the blood is not fully compensated for by circulatory performance, until finally, Po2 levels fully collapse. In a second set of experiments energy and acid,base status of white muscle of Atlantic cod in vivo was measured by magnetic resonance (31P-NMR) spectroscopy in unanaesthetized and unimmobilized fish in the temperature range between 13 and 21° C. A decrease in white muscle intracellular pH (pHi) with temperature occurred between 10 and 16° C (,pH per ° C = ,0·025 per ° C). In white muscle temperature changes had no influence on high-energy phosphates such as phosphocreatine (PCr) or ATP except during exposure to high critical temperatures (>16° C), indicating that white muscle energy status appears to be relatively insensitive to thermal stress if compared to the thermal sensitivity of the whole animal. The data were consistent with the hypothesis of an oxygen limitation of thermal tolerance in animals, which is set by limited capacity of oxygen supply mechanisms. Although chondrocyte transplantation is a useful therapeutic strategy, the repair process can be lengthy. Previously we have shown that over expression of bone morphogenetic protein-7 (BMP-7) in chondrocytes by adenovirus-mediated gene transfer leads to increased matrix synthesis and cartilage-like tissue formation in vitro. In this context we hypothesized that implantation of genetically modified chondrocytes expressing BMP-7 would accelerate the formation of hyaline-like repair tissue in an equine model of cartilage defect repair. Methods: Chondrocytes treated with adenovirus vector encoding BMP-7 (AdBMP-7) or as control, an adenovirus vector encoding an irrelevant gene (Escherichia coli cytosine deaminase, AdCD) were implanted into extensive (15 mm diameter) articular cartilage defects in the patellofemoral joints of 10 horses. Biopsies were performed to evaluate early healing at 4 weeks. At the terminal time point of 8 months, repairs were assessed for morphology, MRI appearance, compressive strength, biochemical composition and persistence of implanted cells. Results: Four weeks after surgery AdBMP-7-treated repairs showed an increased level of BMP-7 expression and accelerated healing, with markedly more hyaline-like morphology than control. Quantitative real-time polymerase chain reaction (PCR) analysis of the repair tissue 8 months after surgery showed that few implanted cells persisted. By this time, the controls had healed similarly to the AdBMP-7-treated defects, and no difference was detected in the morphologic, biochemical or biomechanical properties of the repair tissues from the two treatment groups. Conclusions: Implantation of genetically modified chondrocytes expressing BMP-7 accelerates the appearance of hyaline-like repair tissue in experimental cartilage defects. Clinical relevance: Rehabilitation after cell-based cartilage repair can be prolonged, leading to decreased patient productivity and quality of life. Baer Cells of the intervertebral disc have a limited capacity for matrix repair that may contribute to the onset and progression of degenerative disc changes. In this study, the biosynthetic capacity of cells isolated from specific regions of the porcine intervertebral disc was evaluated in vitro. Using a competitive reverse transcription-polymerase chain reaction technique, gene expression levels for types I and II collagen were quantified in cells cultured for up to 21 d in a three-dimensional alginate culture system and compared to levels obtained for cells in vivo. The mechanical properties of cell-alginate constructs were measured in compression and shear after periods of culture up to 16 weeks. Cells from the anulus fibrosus expressed the most type I collagen mRNA in vivo and in vitro, while cells from the transition zone expressed the most type II collagen mRNA in vivo and in vitro. Mechanical testing results indicate that a mechanically functional matrix did not form at any time during the culture period; rather, decreases of up to 50% were observed in the compressive and shear moduli of the cell,alginate constructs compared to alginate with no cells. Together with results of prior studies, these results suggest that intervertebral disc cells maintain characteristics of their phenotype when cultured in alginate, but the molecules they synthesize are not able to form a mechanically functional matrix in vitro. © 2001 Orthopaedic Research Society. Ralph Sea ice is the dominant feature of polar oceans and contains significant quantities of microalgae. When sea ice forms and melts, the microalgal cells within the ice matrix are exposed to altered salinity and irradiance conditions, and subsequently, their photosynthetic apparatuses become stressed. To simulate the effect of ice formation and melting, samples of sea-ice algae from Cape Hallett (Antarctica) were exposed to altered salinity conditions and incubated under different levels of irradiance. The physiological condition of their photosynthetic apparatuses was monitored using fast and slow fluorescence-induction kinetics. Sea-ice algae exhibited the least photosynthetic stress when maintained in 35, and 51, salinity, whereas 16, 21, and 65, treatments resulted in significant photosynthetic stress. The greatest photosynthetic impact appeared on PSII, resulting in substantial closure of PSII reaction centers when exposed to extreme salinity treatments. Salinity stress to sea-ice algae was light dependent, such that incubated samples only suffered photosynthetic damage when irradiance was applied. Analysis of fast-induction curves showed reductions in J, I, and P transients (or steps) associated with combined salinity and irradiance stress. This stress manifests itself in the limited capacity for the reduction of the primary electron receptor, QA, and the plastoquinone pool, which ultimately inhibited effective quantum yield of PSII and electron transport rate. These results suggest that sea-ice algae undergo greater photosynthetic stress during the process of melting into the hyposaline meltwater lens at the ice edge during summer than do microalgae cells during their incorporation into the ice matrix during the process of freezing. The guidelines vary between countries, but the principles of respect for persons, beneficence, and justice are constant. These principles are reflected in requirements to obtain free and informed consent, to minimize risk or harm, and to not overly burden or disadvantage particular populations. For research to be ethical, it must also be of such a standard, and be conducted in such a manner that it will generate knew and useful knowledge. Children have limited capacity for understanding and may be more open to coercion. Therefore, they are regarded as a particularly vulnerable population, and specific clauses regarding children are incorporated into many guidelines. A key concept in these clauses is the degree of risk acceptable for children involved in research. While it is generally agreed that children require particular attention because of their vulnerability, there is also increasing concern that children in general should not be disadvantaged by lack of knowledge due to reduced research activity. Finally, an increasingly active area of research in children involves genetics and biobanking. Research in these areas raises new and challenging ethical issues. Eriksson The growth of every human fetus is constrained by the limited capacity of the mother and placenta to deliver nutrients to it. At birth, boys tend to be longer than girls at any placental weight. Boy's placentas may therefore be more efficient than girls, but may have less reserve capacity. In the womb boys grow faster than girls and are therefore at greater risk of becoming undernourished. Fetal undernutrition leads to small size at birth and cardiovascular disorders, including hypertension, in later life. We studied 2003 men and women aged around 62 years who were born in Helsinki, Finland, of whom 644 had hypertension: we examined their body and placental size at birth. In both sexes, hypertension was associated with low birth weight. In men, hypertension was also associated with a long minor diameter of the placental surface. The dangerous growth strategy of boys may be compounded by the costs of compensatory placental enlargement in late gestation. In women, hypertension was associated with a small placental area, which may reduce nutrient delivery to the fetus. In men, hypertension was linked to the mothers' socioeconomic status, an indicator of their diets: in women it was linked to the mothers' heights, an indicator of their protein metabolism. Boys' greater dependence on their mothers' diets may enable them to capitalize on an improving food supply, but it makes them vulnerable to food shortages. MCGRATH This article examines the implementation of the State Children's Health Insurance Program (SCHIP) in three states: Massachusetts, Georgia, and Ohio. It examines the effectiveness of four theoretical driving forces in explaining implementation using a multiple case study analysis. Data were compiled using legislative histories, key informant interviews, public record, and media content analysis and were analyzed using a triangulation of sources. Findings suggest that the driving forces as conceptualized in the literature are only partially helpful when examining the implementation of federal redistributive health policy in these states. A pursuit of rationality approach was the most explanatory of the driving forces followed by an organizational-policy fit when there was limited capacity to implement new policy. Overall, implementation was found to be more related to state-level capacity and the state's previous programmatic experiences. Policy innovation was more likely to occur when capacity was high and where goals agreement drove the process. [source] The challenges and opportunities of implementing the integrity pact as a strategy for combating corruption in Nigeria's oil rich Niger Delta regionPUBLIC ADMINISTRATION & DEVELOPMENT, Issue 4 2010Uwafiokun Idemudia Abstract In recent years, significant local and transnational concerted initiatives have been instituted to curb the incidence of corruption that has undermined socio-economic development in Nigeria. Drawing on the critiques of such initiatives, and the experience from the process of implementing the Integrity Pact in the Niger Delta Development Commission, this article suggests that the Integrity Pact in principle offers real opportunities that can both reinforce and complement existing anti-corruption initiatives in Nigeria's public sector. However, political instability, lack of continuity in civil service leadership and limited capacity are core challenges that confront the successful implementation and institutionalisation of the Integrity Pact as a means of fighting corruption and meeting sustainable development objectives in the Niger Delta. Nevertheless, it has limited capacity to preview male fertility. As calorimetry may be used to evaluate directly the metabolic activity of a biological system, the aim of this study was to use microcalorimetry as an additive method for sperm metabolism evaluation of cooled equine semen. Two ejaculates of four stallions were collected and motility, viability (eosin 3%) and membrane functional integrity (hyposmotic swelling test) of spermatozoa were evaluated. Sperm samples were processed following different protocols and the metabolism of these samples was accessed by calorimetry. Centrifugation is part of some of these processing protocols and although this procedure has been deleterious for sperm viability and plasma membrane integrity, no decrease in sperm motility was observed. Microcalorimetry was capable of detecting the positive effect of re-suspending the sperm pellet with Kenney extender. Thus, the use of microcalorimetry offered additional information for equine sperm metabolism evaluation and was efficient in detecting important information from sperm cell metabolism. PIROZZI Abstract The objectives of this study were to describe the interactive effects of varying digestible protein (DP) and digestible energy (DE) contents on the feed intake, growth, protein utilization and whole body composition of juvenile mulloway (Argyrosomus japonicus) and to determine the optimal DP : DE ratio for growth. This was achieved by feeding mulloway diets containing one of four different DP levels (250,550 g kg,1) at two DE levels (16 or 21 MJ kg,1). Juvenile mulloway were stocked at each of two different sizes (70 or 200 g) in triplicate groups for each dietary treatment and fed twice daily to apparent satiation over 58 days. The results indicated that feed intake was not governed solely by energy demands but was also dependant on the DP content of the diet. Protein utilization did not improve with diets containing decreasing protein and increasing lipid content indicating that mulloway have a limited capacity to spare DP. Optimal DP content was found to be 444,491 g kg,1 depending on the DE content of the diet and the size of mulloway and is within the range reported for other sciaenid species. The use of formulated diets with 28.6 g of DP MJ DE,1 will achieve optimal growth and protein deposition for 70,275 g mulloway. [source] |
