Curriculum vitae of intestinal intraepithelial T cells: their developmental and behavioral characteristics

Summary:, The alimentary tract has an epithelial layer, consisting mainly of intestinal epithelial cells (IECs), that is exposed to the exterior world through the intestinal lumen. The IEC layer contains many intestinal intraepithelial T cells (IELs), and the total number of IELs constitutes the largest population in the peripheral T-cell pool. Virtually all ,,-IELs and many ,,-IELs in the mouse small intestine are known to express CD8,, homodimers. A wide range of evidence that supports extrathymic development of these CD8,,+ IELs has been collected. In addition, while several studies identified cells with precursor T-cell phenotypes within the gut epithelium, how these precursors, which are dispersed along the length of the intestine, develop into ,,-IELs and/or ,,-IELs has not been clarified. The identification of lymphoid cell aggregations named ,cryptopatches' (CPs) in the intestinal crypt lamina propria of mice as sites rich in T-cell precursors in 1996 by our research group, however, provided evidence for a central site, whereby precursor IELs could give rise to T-cell receptor-bearing IELs. In this review, we discuss the development of IELs in the intestinal mucosa and examine the possibility that CPs serve as a production site of extrathymic IELs. [source]


SHRIMP U,Pb zircon chronology of ultrahigh-temperature spinel,orthopyroxene,garnet granulite from South Altay orogenic belt, northwestern China

Abstract Diagnostic mineral assemblages, mineral compositions and zircon SHRIMP U,Pb ages are reported from an ultrahigh-temperature (UHT) spinel,orthopyroxene,garnet granulite (UHT rock) from the South Altay orogenic belt of northwestern China. This Altay orogenic belt defines an accretionary belt between the Siberian and Kazakhstan,Junggar Plates that formed during the Paleozoic. The UHT rock examined in this study preserves both peak and retrograde metamorphic assemblages and microstructures including equilibrium spinel + quartz, and intergrowth of orthopyroxene, spinel, sillimanite, and cordierite formed during decompression. Mineral chemistry shows that the spinel coexisting with quartz has low ZnO contents, and the orthopyroxene is of high alumina type with Al2O3 contents up to 9.3 wt%. The peak temperatures of metamorphism were >950°C, consistent with UHT conditions, and the rocks were exhumed along a clockwise P,T path. The zircons in this UHT rock display a zonal structure with a relict core and metamorphic rim. The cores yield bimodal ages of 499 ± 8 Ma (7 spots), and 855 Ma (2 spots), with the rounded clastic zircons having ages with 490,500 Ma. Since the granulite was metamorphosed at temperatures >900°C, exceeding the closure temperature of U,Pb system in zircon, a possible interpretation is that the 499 ± 8 Ma age obtained from the largest population of zircons in the rock marks the timing of formation of the protolith of the rock, with the zircons sourced from a ,500 Ma magmatic provenance, in a continental margin setting. We correlate the UHT metamorphism with the northward subduction of the Paleo-Asian Ocean and associated accretion-collision tectonics of the Siberian and Kazakhstan,Junggar Plates followed by rapid exhumation leading to decompression. Features of Saccharomyces cerevisiae as a culture starter for the production of the distilled sugar cane beverage, cachaça in Brazil

Abstract Aims:, To evaluate the dominance and persistence of strains of Saccharomyces cerevisiae during the process of sugar cane fermentation for the production of cachaça and to analyse the microbial compounds produced in each fermentative process. Methods and Results:, Three S. cerevisiae strains were evaluated during seven consecutive 24-h fermentation batches using recycled inocula. The UFLA CA 116 strain had the largest population of viable organisms, and the maximum population was achieved in the fourth batch after 96 h of fermentation. The UFLA CA 1162 and UFLA CA 1183 strains grew more slowly, and the maximum population was reached in the seventh batch. Molecular characterization of isolated yeast cells using PFGE (pulse field gel electrophoresis) revealed that more than 86% of the isolates corresponded to the initially inoculated yeast strain. The concentration of aldehydes, esters, methanol, alcohol and volatile acids in the final-aged beverages were within the legal limits. Conclusions:, Cachaça produced by select yeast strains exhibits analytical differences. UFLA CA 1162 and UFLA CA 116 S. cerevisiae isolates can be considered the ideal strains for the artisanal production of cachaça in Brazil. Significance and Impact of the Study:, The use of select yeast strains can improve the quality and productivity of cachaça production. Our findings are important for the appropriate monitoring of yeast during sugar cane fermentation. In addition, we demonstrate that UFLA CA 116 and UFLA CA 1162, the ideal yeast strains for cachaça production, are maintained at a high population density. The persistence of these yeast strains in the fermentation of sugar cane juice promotes environmental conditions that prevent or decrease bacterial contamination. Thus, the use of select yeast strains for the production of cachaça is a viable economic alternative to standardize the production of this beverage. [source]


Cardiometabolic Syndrome and Its Association With Education, Smoking, Diet, Physical Activity, and Social Support: Findings From the Pennsylvania 2007 BRFSS Survey

The authors aimed to examine the prevalence of cardiometabolic syndrome (CMS) and its association with education, smoking, diet, physical activity, and social support among white, black, and Hispanic adults using data from the 2007 Pennsylvania Behavior Risk Factor Surveillance System (BRFSS) survey, the largest population-based survey in the state. The authors examined associations between CMS and associated factors cross-sectionally using univariate and multivariate methods. The study included a representative sample of 12,629 noninstitutionalized Pennsylvanians aged ,18. Components of CMS included obesity, hypercholesterolemia, angina (as a surrogate for decreased high-density lipoprotein), prehypertension or hypertension, and prediabetes or diabetes. CMS was identified as the presence of ,3 CMS components. The results show that the prevalence of CMS was 20.48% in blacks, followed by Hispanics (19.14%) and whites (12.26%), (P<.01). Multivariate logistic regression analyses indicated that physical inactivity, lower educational levels, smoking, daily consumption of vegetables and/or fruits <3 servings, and lack of social support were significantly associated with the odds of having CMS. In conclusion, black and Hispanic adults have a significantly higher prevalence of CMS than whites. The significant association between CMS and risk factors provides new insights in the direction of health promotion to prevent and control CMS in those who are at high risk. [source]

Development of Issoria lathonia (Lepidoptera: Nymphalidae) on zinc-accumulating and nonaccumulating Viola species (Violaceae)

Abstract The larvae of Issoria lathonia L. feed in natural conditions on several Viola spp., among which are the zinc-accumulating Viola calaminaria (Gingins) Lej. and the nonmetal-accumulating Viola tricolor L. To examine how I. lathonia caterpillars cope with the naturally high foliar zinc concentration of V. calaminaria, we compared the growth of caterpillars reared on leaves varying in zinc concentration. Larvae were fed in controlled conditions with V. calaminaria and V. tricolor grown on noncontaminated soil (i.e., two low-Zn diets) and with V. calaminaria grown on zinc-enriched soil (i.e., one high-Zn diet). Larvae had a higher growth rate when fed with noncontaminated V. calaminaria compared to zinc-enriched V. calaminaria, suggesting that zinc slows down larval growth. However, larvae consumed more leaves of zinc-enriched V. calaminaria (+45%; estimated from fecal mass) compared with noncontaminated V. calaminaria, suggesting that zinc accumulation would not be advantageous to plants. Caterpillars reared on high-zinc leaves regulate their internal zinc concentration through excretion of highly metal-concentrated feces. When kinetics of growth on both low-zinc diets were compared, it appeared that larval development was faster on noncontaminated V. calaminaria than on V. tricolor. This suggests that more nutrients or less feeding inhibitors in V. calaminaria account for fastest growth. Developmental rates on V. tricolor and on zinc-enriched V. calaminaria were similar, despite the high leaf zinc concentration of the latter species. Together with the abundance of V. calaminaria on calamine soils, this may explain why the largest populations of I. lathonia develop on V. calaminaria in Belgium. [source]
