Selected AbstractsDevelopment of a natural preservative system using the mixture of chitosan- Inula helenium L. extractINTERNATIONAL JOURNAL OF COSMETIC SCIENCE, Issue 4 2002S.-B. Seo Synopsis The aim of this study was to develop a new natural preservative system making up for the weak points of chitosan as a preservative. As reported in a previous manuscript (20th IFSCC Congress, Cannes, France, 1998), the minimum inhibitory concentrations (MICs) of water-soluble chitosan against bacteria and yeast were 0.9,3.0 mg mL,1, whereas MICs of chitosan against Aspergillus niger were over 5.0 mg mL,1. However, the result of recent study showed that the MICs of Inula helenium L. extract against A. niger were below 1.0 mg mL,1. Thus, we could develop a new preservative system containing both chitosan and I. helenium L. extract named CI-mixture. MICs of CI-mixture against bacteria and fungi (yeast and mould) were 2.0,4.0 mg mL,1. When 10.0% of the mixture (the ratio of chitosan to I. helenium L. extract = 7.5% : 2.5%) was applied to cosmetic formulae such as skin lotion, milk lotion, cream and pack, it revealed appropriate preservative efficacy. Our result of the patch test also showed that this preservative system reduced skin irritation by about 30,50%, as compared to the organic preservative system. Therefore, the good natural preservative system including chitosan and Inula helenium L. extract could be incorporated in cosmetic formulations. Résumé Le but de cette étude était de développer un nouveau système conservateur naturel en améliorant les propriétés du chitosan en tant que conservateur. Comme décrit dans un manuscrit précédent (20ième Congrès IFSCC Cannes, France, 1998), les C.M.I (concentrations minimales inhibitrices) du chitosan hydro-soluble contre les bactéries et levures étaient de 0.9,3 mg mL,1 et supérieures à 5 mg mL,1 conte A. niger. Cependant, des résultats récents conféraient à un extrait d' Inula helenium L. des CMI contre A niger inférieures à 1 mg mL,1. Ainsi, il devenait possible de développer un système conservateur combinant ces deux composés (chitosan et I. helenium L.) dénommé ici mélange CI. Les CMI du mélange CI contre les bactéries et champignons (levures et moisissures) étaient de 2,4 mg mL,1. Lorsque ce mélange CI (rapport chitosan/Inula = 7.5% : 2.5%) était introduit à 10% dans des formules cosmétiques telles que lotion cutanée, lait cutané, crème. il montrait de bonnes propriétés conservatrices. Des résultats de patch tests cutanés montraient aussi que ce système conservateur réduisait l'irritation cutanée d'environ 30à 50% par rapport à un système conservateur organique. However, the effects of Vimang on allergic response have not been reported until now. In this study, the effects of Vimang and mangiferin, a C-glucosylxanthone isolated from the extract, on different parameters of allergic response are reported. Vimang and mangiferin showed a significant dose-dependent inhibition of IgE production in mice and anaphylaxis reaction in rats, histamine-induced vascular permeability and the histamine release induced by compound 48/80 from rat mast cells, and of lymphocyte proliferative response as evidence of the reduction of the amount of B and T lymphocytes able to contribute to allergic response. In these experiments, ketotifen, promethazine and disodium cromoglicate were used as reference drugs. Furthermore, we demonstrated that Vimang had an effect on an in-vivo model of inflammatory allergy mediated by mast cells. These results constitute the first report of the anti-allergic properties of Vimang on allergic models, as well as suggesting that this natural extract could be successfully used in the treatment of allergic disorders. Mangiferin, the major compound of Vimang, contributes to the anti-allergic effects of the extract. [source] Adsorption separation of terpene lactones from Ginkgo biloba L. extract using glass fiber membranes modified with octadecyltrichlorosilaneJOURNAL OF SEPARATION SCIENCE, JSS, Issue 11 2005I-Fang Su Abstract In this study porous glass fiber membranes were modified by reaction with octadecyltrichlorosilane to form C18 hydrophobic membranes. The contact angle and the CH2 vibration bands at 2855 and 2920 cm,1 found by FTIR measurements verified the successful immobilization of C18 groups on the glass fiber membranes. The resulting C18 hydrophobic membranes were used to adsorb terpene lactones from crude Ginkgo biloba L. extracts. In batch adsorption processes, the modified C18 membranes exhibited a better adsorption performance than commercial C18 solid phase extraction adsorbents. Different desorption solvents were tested and ethyl acetate was found to preferentially desorb terpene lactones from the modified C18 membranes. In flow adsorption experiments at 1 mL/min, terpene lactone contents higher than 6 wt% (the standardized content) could be achieved in the elution step using ethyl acetate. [source] Effect of X-Radiation on Lipid Peroxidation and Antioxidant Systems in Rats Treated with Saponin-containing CompoundsPHOTOCHEMISTRY & PHOTOBIOLOGY, Issue 1 2008Omer Yalinkilic The aim of this study was to investigate the effect of three saponin-containing plant species extracts (Aesculuc hippocastanum L. seed extract [AHE], Medicago sativa L. extract [MSE] and Spinacia oleracea L. extract [SOE]) on lipid peroxidation and on antioxidant systems in rats exposed to X-rays (XR). The rats were divided into three categories. The first category served as controls and received only a standard diet. The second category served as the radiation group and received 5 and 10 Gy XR dose. The third category (XR+extract-treated) received plant extracts (25.0 or 50.0 mg kg,1 live weight) and 5 or 10 Gy XR dose. Blood samples were analyzed for their content of malondialdehyde (MDA), reduced glutathione (GSH), plasma vitamin C, ,-carotene and retinol. In animals receiving XR, the plasma MDA (P < 0.001) value significantly increased but the level of GSH (P < 0.01), vitamin C (P < 0.001), retinol and ,-carotene (P < 0.001) decreased significantly with increasing XR doses. In the XR+extract-treated groups, the concentrations of MDA increased significantly with increasing radiation but their concentrations decreased significantly with increasing extract concentrations. Plasma concentrations of GSH, ,-carotene, retinol and vitamin C in XR+extract-treated groups decreased significantly with increasing XR dose but their concentrations increased with increasing extract doses. Further, comparison of blood samples of XR+extract-treated groups with those from the control group showed that GSH, ,-carotene, retinol and vitamin C values increased significantly but that MDA values decreased significantly. The results showed that all extracts have enhanced the antioxidant status and decreased the incidence of free radical-induced lipid peroxidation in blood samples of rats exposed to XR. However, the antioxidant effect of AHE-administered animals was more effective than that of MSE- and SOE-administered whole-body XR rats. We conclude that the supplementation with saponin-containing extracts may serve to reinforce the antioxidant systems, thus having protective effect against cell damage by XR. [source] Studying the anti-tyrosinase effect of Arbutus andrachne L. extractsINTERNATIONAL JOURNAL OF COSMETIC SCIENCE, Issue 4 2008R. A. Issa Synopsis Arbutus andrachne L. is widely distributed in Jordan. Tyrosinase is the key enzyme in the biosynthesis of melanin. This preliminary study was carried out to assess the possible anti-tyrosinase activity of A. andrachne extracts. Arbutin, hydroquinone and kojic acid were selected as inhibitor standards. Five different extracts (chloroform, butanol, ethanol, methanol and water) were prepared from A. andrachne stems and their activities were compared with the selected tyrosinase inhibitors. IC50 was measured for both, standard and plant extracts. Among the different extracts, the methanolic extract exhibited the highest anttyrosinase activity with an IC50 value (1 mg mL,1). Furthermore, 9 mg A. andrachne methanolic extract showed 97.49% inhibition of tyrosinase activity. Arbutin, hydroquinone, ,-sitosterol and ursolic acid were identified in the different extracts of A. andrachne by thin layer chromatography (TLC) and isolated by preparative TLC from the methanolic and chloroform stem extracts, respectively. Résumé Arbutus andrachne L. est largement répandu en Jordanie. La tyrosinase est un enzyme clé dans la biosynthèse de la mélanine. Cette étude préliminaire est menée dans le but de juger de la possible activité anti-tyrosinase des extraits d'A. andrachne L. L'arbutine, l'hydroquinone et l'acide kojique ont été sélectionnés comme inhibiteurs de référence. Cinq extraits différents (chloroforme, butanol, éthanol, méthanol et eau) ont été préparés à partir de tiges d'A. andrachne L. et leurs activités ont été comparées à celles des inhibiteurs de tyrosinase sélectionnés. L'IC50 a été mesurée à la fois pour les références et les extraits de plantes. Parmi les différents extraits, l'extrait méthanolique montre l'activité anti-tyrosinase la plus élevée avec une valeur d'IC50 de 1 mg mL,1. De plus, 9 mg d'extrait méthanolique d'A. andrachne L. possède une activité inhibitrice de la tyrosinase de 97.49%. L'arbutine, l'hydroquinone, le ,-sitostérol et l'acide ursolique ont été identifiés dans les différents extraits par chromatographie sur couches minces et isolés par chromatographie préparative, respectivement à partir des extraits méthanoliques et chloroformiques de tiges. [source] Effect of lyophilisation, refrigerated storage and frozen storage on the coagulant activity and microbiological quality of Cynara cardunculus L. extractsJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 8 2008Luis Tejada Abstract BACKGROUND:Cheese-makers have traditionally kept vegetable coagulants refrigerated until use, even though little was known of their microbiological quality or coagulant activity during storage. This study aimed to assess the efficacy of lyophilisation, refrigerated storage and frozen storage of fresh vegetable extract as a means of standardising coagulant activity in terms of coagulation times, pH and microbiological quality. RESULTS:Neither the pH nor the coagulation time of lyophilised extracts was significantly modified during 1 year; however, changes were observed following frozen storage, and more notable following refrigerated storage. Lyophilisation of aqueous extracts prompted the destruction of most micro-organisms; low counts initially noted for total mesophiles, lactic acid bacteria and yeasts disappeared during the first few days of storage, due to low water activity. There was a generalised decrease in micro-organism counts during frozen storage. Refrigeration was found to be unsuitable for storing of cardoon extract; an increase of roughly 2 log unit counts was recorded in total mesophile, lactic acid bacteria, yeast and mould counts after 1 year of refrigerated storage. CONCLUSION:Refrigerated storage cannot be considered a suitable method for prolonged conservation of aqueous cardoon extract. Both lyophilisation and frozen storage of aqueous extracts proved ideal for prolonged storage of vegetable coagulant. Lyophilisation additionally had certain advantages over frozen storage.
