Objective:, Adults with non-affective psychosis show subtle deviations in a range of developmental trajectories as children and adolescents. Method:, Based on a birth-cohort (n = 3801), we examined the Peabody Picture Vocabulary Test (PPTV) at age 5, and Raven's Standard Progressive Matrices (RSPM) and Wide Range Achievement Test reading scale (WRAT-R) at age 14. Items related to speech problems and attentional dysfunction were available from maternal- or self-report. At age 21, we identified 60 cohort members who were screen-positive for non-affective psychosis (SP-NAP). Results:, Impaired performance on the PPVT and RSPM (but not WRAT-R) predicted SP-NAP for males only. Male cohort members in the highest quartile for attentional dysfunction at ages 5 and 14 were about 5,8 times more likely to develop SP-NAP. SP-NAP in males was significantly associated with speech problems at age 14. Conclusion:, Males who develop non-affective psychoses have subtle impairments in cognitive capacity prior to the development of their psychotic disorder. The current status of urban,rural differences in psychiatric disorders. Objective:, Reviews of urban,rural differences in psychiatric disorders conclude that urban rates may be marginally higher and, specifically, somewhat higher for depression. However, pooled results are not available. Method:, A meta-analysis of urban,rural differences in prevalence was conducted on data taken from 20 population survey studies published since 1985. Pooled urban,rural odds ratios (OR) were calculated for the total prevalence of psychiatric disorders, and specifically for mood, anxiety and substance use disorders. Results:, Significant pooled urban,rural OR were found for the total prevalence of psychiatric disorders, and for mood disorders and anxiety disorders. No significant association with urbanization was found for substance use disorders. Adjustment for various confounders had a limited impact on the urban,rural OR. Conclusion:, Urbanization may be taken into account in the allocation of mental health services. The presence of nitric oxide synthase (NOS) and nerve fibres in the photophores of seven bioluminescent fish species (Hygophum benoiti, Myctophum punctatum, Electrona risso, Cyclothone braueri, Vinciguerria attenuata, Maurolicus muelleri and Porichthys notatus) with endogenous photocytes, were investigated. Antibodies directed against neuronal and inducible NOS (n and iNOS respectively) and NADPH-diaphorase activity were used to reveal the locations of NOS, while antibodies directed against acetylated tubulin were used to visualize nerve fibres. The nNOS antibody labelled structures in all investigated photophores except in the organs from P. notatus. The photocytes of P. notatus showed NADPH-diaphorase activity. In the myctophid species, NOS-like immunoreactivity was found in small intracellular structures of the photocytes and in nerve fibres reaching the photocytes. nNOS-positive fibres were also found among lens/filter cells in V. attenuata, and in M. muelleri the cytoplasm of lens/filter cells contained NOS-like material. In C. braueri, a cell type located at a collecting chamber for luminous products in the photophore contained NOS-like material. All photophores received an innervation reaching the photocytes, as well as other components including lens/filter areas. The results of this study comply with an involvement of nitric oxide in the control of bioluminescence in several fish species. The tritocerebrum and the clypeolabrum in mandibulate arthropods: segmental interpretations. ,Acta Zoologica (Stockholm) 91: 249,266 Different interpretations of the segmental composition of the head in mandibulate arthropods are critically reviewed, with particular focus on three closely associated structures: the tritocerebrum, the stomatogastric nervous system and the clypeolabrum. The main conclusions arising from the different discussions are the following. (1) Each tritocerebral ganglion has a dual composition, clearly discernable in some crustacean and hexapod species, including a dorsal portion connected with the second antennae and a ventral portion connected with the stomatogastric nervous system via the frontal ganglion. (2) The suboesophageal commissure linking the tritocerebral lobes of the two sides, can be wholly ascribed to the tritocerebral segment. (3) The stomatogastric nervous system is a morphologically autonomous system that is not fundamentally affected by head metamerization. (4) The clypeolabrum, the epistome,labrum and the hypostome are regarded as homologous formations. The clypeolabrum represents a fundamental structure of the head probably present in the arthropod ground plan. Its close spatial and developmental association with the stomodeum and its derivative, the stomatogastric nervous system, suggests that it is an anterior outgrowth of the forehead arising from a preoral territory (presegmental acron or protocerebral,ocular region?) and secondarily connected with the tritocerebrum, rather than derived from a pair of reduced appendages. In Xylocopa frontalis the reproductive tract is composed of testes, deferent ducts, seminal vesicles, accessory glands and an ejaculatory duct. Each testis comprises four testicular tubules in which multiple cysts are present containing approximately 64 spermatozoa per cyst. The seminal vesicle consists of an epithelium, a thick basement lamina and a muscular external sheet. In the luminal region some vesicles can be observed; however, the epithelial cells of the seminal vesicle do not display morphological features associated with secretory functions. The spermatozoa, measuring approximately 260 µm long, are similar to the hymenopteran pattern. The head region consists of an acrosome with an inner perforatorium that penetrates an asymmetrical nuclear tip. The nucleus is linear, electron-dense and its posterior tip projects into the beginning of the axoneme. The centriolar adjunct is asymmetric with many electron-lucent lacunae interspersed throughout. The axoneme has the 9 + 9 + 2 pattern of microtubules and in the posterior region the central microtubules finish first, followed by the doublets and finally the accessory microtubules. The mitochondrial derivatives are asymmetric in both length and diameter with paracrystalline material present only in the larger one. These features may be useful characters for taxonomy and phylogenetic studies. A novel exocrine structure of the bicellular unit type in the thorax of termites , Acta Zoologica (Stockholm) 91: 193,198 Studying the thorax of some Termitidae species, we found two pairs of hitherto unknown lateral glands in the mesothorax and metathorax of both workers and soldiers. The glands consist of distinct clusters of class 3 secretory cells accompanied by their duct cells, located in the upper lateral portion of the thoracic wall. Ultrastructural observations reveal numerous mitochondria, a well-developed Golgi apparatus and vesicular smooth endoplasmic reticulum, indicating a cytoplasm with intensive metabolic activity. The gland is reported to occur in Microcerotermes strunckii, Cornitermes cumulans and Nasutitermes minor, three species comprising an interesting morpho-behavioural gradient, respectively, from only mechanical, through mechanical,chemical, to only chemical defence systems. The extent of such a gradient allows speculations that this gland would be related to the general needs of termites, rather than to some specificities of a single group. We warn, however, that complementary studies are needed, before any conclusions can be drawn. Spermiogenesis and spermatozoon ultrastructure of the davaineid cestode Raillietina micracantha. , Acta Zoologica (Stockholm) 91: 212,221 The spermiogenesis and the ultrastructural organization of the spermatozoon of the davaineid cestode Raillietina micracantha are described by means of transmission electron microscopy. Spermiogenesis begins with the formation of a zone of differentiation containing two centrioles. One of the centrioles develops a free flagellum that later fuses with a cytoplasmic extension. The nucleus migrates along the spermatid body after the proximodistal fusion of the flagellum and the cytoplasmic extension. During advanced stages of spermiogenesis a periaxonemal sheath and intracytoplasmic walls appear in the spermatids. Spermiogenesis finishes with the appearance of two helicoidal crested bodies at the base of spermatids and, finally, the narrowing of the ring of arched membranes detaches the fully formed spermatozoon. The mature spermatozoon of R. micracantha is a long and filiform cell, tapered at both ends, which lacks mitochondria. It exhibits two crested bodies of different lengths, one axoneme of the 9 + ,1' pattern of trepaxonematan Platyhelminthes, twisted cortical microtubules, a periaxonemal sheath, intracytoplasmic walls, granules of glycogen and a spiralled nucleus. The anterior extremity of the spermatozoon is characterized by the presence of an electron-dense apical cone and two spiralled crested bodies while the posterior extremity of the male gamete exhibits only the axoneme and an electron-dense posterior tip. A prime inference on genetic diversity (RAPDs) in the marine fish Atherinella brasiliensis (Teleostei, Atherinopsidae) from Southern Brazil. ,Acta Zoologica (Stockholm) 91: 242,248 As a result of the importance of Atherinella brasiliensis in estuarine environments, random amplified polymorphic DNA (RAPD) markers were used to verify the genetic diversity in A. brasiliensis from two different places in Paranaguá Bay (Paraná State) and one from the Conceição Lagoon (Santa Catarina State). Cytogenetic data have shown a high karyotypic diversity in some populations, although in others this peculiarity demonstrates rearrangements such as heterochromatinization. In the present study, a low level of genetic structuring between the samples from Conceição Lagoon compared with the others was observed through principal coordinate analysis (PCO), analysis of molecular variance and Mantel test according to 79 RAPD markers. As this specie does not perform horizontal migration and the individuals of Conceição Lagoon are isolated, three hypotheses are proposed to explain the results: (i) similar environments may show homogeneous populations not depending on the geographical distance, (ii) because vicariant events that formed the bays occurred in a recent period, the fragmentation effects over the structuring of the genetic diversity may still be low and not totally detectable by the RAPD technique and (iii) the isolation time or the number of generations may not be enough to promote a possible differentiation and genetic structuring between the specimens of these three places. The specimens of these places present a low level of differentiation and genetic structuring so we can consider them as a unique homogeneous population. A shortening of the manus precedes the attenuation of other wing-bone elements in the evolution of flightlessness in birds. ,Acta Zoologica (Stockholm) 91: 115,122 This is the first study to present evidence for a general pattern of wing-bone attenuation during the early stages of the evolution of flightlessness. A comparative analysis using phylogenetic independent contrasts showed that in families that contain both flighted (volant) and flightless species, the volant species have shorter wings and total-arm (humerus + ulna + manus) lengths relative to their body masses than the species within their wholly volant sister families. A shortening of the manus may typify the early stages of the evolution of flightlessness, with the humerus and ulna attenuating later, perhaps because of their role in maintaining the position of the aerodynamically important alula. A shorter wing relative to body mass was not the result of the inverse (i.e. heavier body mass relative to wing length) because mean body masses of volant members of flightless families were similar to or lower than those of their wholly volant sister families. Despite finding a common trend in the wing morphologies of volant members of flightless families, it seems unlikely that a general model of selection pressures driving the evolution of flightlessness exists. At the very least, a dichotomy between those birds that have lost the ability to fly in order to gain the ability to swim and terrestrial forms, may persist. Functional morphology of the postpharyngeal gland of queens and workers of the ant Monomorium pharaonis (L.). ,Acta Zoologica (Stockholm) 87: 101,111 The postpharyngeal gland (PPG) is unique to ants and is the largest exocrine gland in their head. In queens of the pharaoh's ant, Monomorium pharaonis, the gland contains approximately 15 finger-like epithelial extensions on each side and opens dorsolaterally in the posterior pharynx. In these ants the PPG morphology varies considerably according to age and mating status. The epithelial thickness increases with age and reaches a maximum at 3 weeks in both virgin and mated queens. A considerable expansion of the lumen diameter occurs in both groups between 4 and 7 days. Virgin queens release their secretion into the gland lumen from an age of 7 days, whereas mated queens accumulate large amounts of secretion in their epithelium. The increasing epithelial thickness, together with the increasing lumen diameter, the presence of numerous inclusions in the epithelium and the release of secretion, are indicative for increasing gland activity. The gland ultrastructure indicates involvement in lipid metabolism and de novo synthesis of lipids. The PPG of workers consists of 12 finger-like tubes at each side. There is a significant difference in epithelial thickness between nurses and repletes and between nurses and foragers. We suggest the PPG serves different purposes in pharaoh's ants: it is likely that the PPG of workers and virgin queens is used to feed larvae. In mated queens the gland probably plays a role in providing the queen with nutritious oils for egg production. The PPG may also function in signalling species nestmate and caste identity, as well as in the reproductive capacity of the queens. Unlike other carapids whose sounds have been recorded (C. boraborensis, C. homei and Encheliophis gracilis), these two species have a central constriction in their swimbladder and are unlikely to encounter heterospecific carapids within their hosts. We evoked sound production in Carapus acus and Carapus mourlani by adding several individuals to a tank with a single host and found that their sounds differ substantially from the sounds emitted by other carapids in pulse length, peak frequency and sharpness of tuning (Q3 dB). Unlike the other carapids, C. mourlani and C. acus produce shorter and less repetitive sounds and do not produce sounds when they enter their host. Since sounds produced within a sea cucumber have the potential to be heard by distant carapids and are typically recorded outside the sea cucumber, we examined the effect of the sea cucumber tegument on acoustic transmission. Attenuation by the tegument was negligible at the frequencies within carapid sounds. Therefore, carapids have the potential to call from the relative safety of a sea cucumber without sacrificing the distance over which their transmissions are heard. Ultrastructure of the seminal receptacle and the dimorphic sperm in the commensal bivalve Mysella bidentata (Veneroida: Galeommatoidea: Montacutidae). ,Acta Zoologica (Stockholm) 82: 107,115 The seminal receptacle and the euspermatozoa and paraspermatozoa of Mysella bidentata were examined at an ultrastructural level and the results were compared with earlier findings of the same and other species of the Montacutidae. The euspermatozoon has a slender 13 µm long nucleus and a 1.1 µm long bullet-shaped acrosome. The acrosome of the paraspermatozoon is almost identical in ultrastructure to that of the euspermatozoa but is longer (1.9 µm) and more slender and is bent at an angle to the diminutive nucleus (1.1 µm long). The unpaired seminal receptacle is lined by a heavily ciliated epithelium and a non-ciliated epithelium with short and broad microvilli. Euspermatozoa only are stored in the receptacle. They are densely packed and orientated with their heads towards the non-ciliated epithelium. In this position they develop numerous extremely fine microvilli from the acrosome which apparently serve to attach them to the epithelial microvillar surface. Stored sperm may presumably remain functional for at least six months. A possible function of paraspermatozoa could be to clump sperm into sperm bags to keep them in suspension. Locomotory and feeding effectors of the tornaria larva of Balanoglossus biminiensis. ,Acta Zoologica (Stockholm) 82: 117,126 The tornaria ciliary bands and oesophagus were examined ultrastructurally to identify the neural components that control larval behaviour. The circumoral ciliary band is known to be innervated in part by fibres from the apical plate and adoral nerve centres. Within the band itself, however, the only neurones we could find were multipolar cells, an unusual cell type with apical processes that traverse the surface of the band. Similar cells occur in the circumoral bands of echinoderm larvae. The tornaria telotroch has a much larger nerve, but no neurones were found either in the band or nearby, so the source of the fibres in the telotroch nerve remains unknown. In addition to having different innervation, the two bands also respond differently to cholinergic agonists, which elicit telotroch arrests but have no visible effect on the circumoral band. The oesophagus has a well-developed musculature and an extensive nerve plexus. During feeding, the oesophagus repeatedly contracts, forcing excess water out along two lateral channels prior to swallowing. These channels are also sites of gill slit formation, so there is evidently a continuity between the water bypass mechanism of the larva and that of the postmetamorphic juvenile. Structure and Ultrastructure of the Spermatozoa of Trichogramma pretiosum Riley and Trichogramma atopovirilia Oatman and Platner (Hymenoptera: Trichogrammatidae). ,Acta Zoologica (Stockholm) 81: 205,211 Spermatozoa of the Trichogramma pretiosum and T. atopovirilia are very slender and long, about 0.35 µm in diameter and 283 µm and 106 µm in length, respectively. Under light microscopy, they appear wavy along their entire length. The head contains a small acrosome which, together with the initial nuclear region is surrounded by an ,extracellular sheath', from which innumerable filaments irradiate. The nucleus is filled with homogeneous, compact chromatin and is attached to the flagellum by an electron dense centriolar adjunct, which extends anteriorly from the nuclear base. The flagellum consists of an axoneme with the 9 + 9 + 2 microtubule arrangement pitched in a long helix, as well as a pair of spiralling mitochondrial derivatives which coil around the axoneme. Based on these characteristics, the sperm of these Trichogramma are very similar to the chalcidoids studied to date and differ from non-chalcidoid Hymenoptera. They differ widely from the sperm of T. dendrolimi and T. ostriniae studied, where no helically twisted structure is shown. However, based on these results we argue that the spiralling of the flagellar structures is a synapomorphy for Trichogrammatidae as well as for Eulophidae + Eurytomidae + Pteromalidae. [source] Microchip-based small, dense low-density lipoproteins assay for coronary heart disease risk assessmentELECTROPHORESIS, Issue 9 2008Hua Wang Abstract Small, dense low-density lipoprotein (sdLDL) has been accepted as an emerging cardiovascular risk factor, and there has been an increasing interest in analytical methods for sdLDL profiling for diagnosis. Serum sdLDL may be measured by different laboratory techniques, but all these methods are laborious, time-consuming, and costly. Recently, we have demonstrated that a low-temperature bonding of quartz microfluidic chips for serum lipoproteins analysis (Zhuang, G., Jin, Q., Liu, J., Cong, H. et al., Biomed. Microdevices 2006, 8, 255,261). In contrast to this previous study, we chose SDS as anionic surfactant to modify both lipoproteins and the channel surface to minimize lipoprotein adsorption and improve the resolution of lipoprotein separation. Two major LDL subclass patterns including large, buoyant LDL (lLDL), sdLDL, and high-density lipoprotein (HDL) were effectively separated with high reproducibility. RSD values of the migration time (min) and peak areas of standard LDL and HDL were 6.28, 4.02, 5.02, and 2.5%, respectively. Serum lipoproteins of 15 healthy subjects and 15 patients with coronary heart disease (CHD) were separated by microchip CE. No peaks of sdLDL were detected in serum samples of healthy subjects while sdLDL fractional peaks were observed in patients' entire serum samples. These results suggested that the microchip-based sdLDLs assay was a simple, rapid, and highly efficient technique and significantly improved the analysis of CHD risk factors. (2010) European Journal of Cancer Care Mapping the information resources available to patients with colorectal cancer The objectives of this study were to identify gaps in information provision along the colorectal cancer (CRC) treatment pathway as provided by health services within the North Eastern Metropolitan Integrated Cancer Service in Victoria Australia; to evaluate the information and recommend consistent, high quality health information resources; and to recommend strategies to improve delivery of patient information. A random sample of health professionals (n= 47) from various disciplines at eight health service sites participated in semi-structured interviews regarding the types of information they provided to CRC patients. Information items were mapped against a published CRC patient management framework and evaluated. A total of 193 information items were collected with 24 items specific to CRC. Gaps in information provision were evident in the community, at diagnosis, in clinics, when treatment was determined and when completed. The quality of information delivery to CRC patients across the public health sites was variable. Resources were often unavailable, out of date and inaccessible in other languages. Results indicate a need to improve health information availability and resource delivery to all CRC patients across different health services particularly at diagnosis and after treatment. Further research is required to determine patient preferences for information about CRC. (2010) European Journal of Cancer Care19, 346,351 Complementary and alternative medicine practitioner consultations among those who have or have had cancer in a Norwegian total population (Nord-Trøndelag Health Study): prevalence, socio-demographics and health perceptions The aim of the study was to identify the use of complementary and alternative medicine (CAM) practitioners among current and previous cancer patients in a total population. A secondary analysis of data from the Nord-Trøndelag Health Study (otherwise known as the HUNT 2 Study) , a total population survey conducted in central Norway , was undertaken. Analysis focused upon the response of 1406 individuals who reported to have or have had cancer and who answered a question on visits to CAM practitioners. The study identified 16.1% of respondents had visited a CAM practitioner in the prior 12 months compared with 12.8% in the total population, and the likelihood of consulting a CAM practitioner was significantly increased among those who had a university degree, who reported a lower perceived global health and who had experienced a health complaint during the last 12 months. Complementary and alternative medicine practitioner consultations among individuals with a previous or current malignant disease were highest for those with poor self-reported health status and with a recent health complaint. We have previously reported that AgC10, a glycosylphosphatidylinositol-anchored mucin from T. cruzi, inhibits TNF secretion by activated macrophages (de Diego, J., Punzon, C., Duarte, M. and Fresno, M., Alteration of macrophage function bya Trypanosoma cruzi membrane mucin. J. Immunol. 1997. 159: 4983,4989). In this report we have further investigated the molecular mechanisms underlying this inhibition. AgC10 inhibited TNF, IL-10 and cyclooxygenase-2 (COX-2) synthesis by macrophages activated with LPS or LPS plus IFN-, in a dose-dependent manner. AgC10 did not affect other aspects of macrophage activation induced by LPS, such as inducible nitric oxide synthase (iNOS) expression. AgC10 also had no effect on TNF or COX-2 transcription or the induction of their promoters but inhibited the stability of TNF and COX-2 mRNA, which are regulated post-transcriptionally by the mitogen-activated protein kinase (MAPK) p38 pathway. AgC10 was found to inhibit both the activation and the activity of p38 MAPK, since MAPK activated protein kinase-2 (MAPKAP-K2 or MK-2) phosphorylation was also strongly inhibited. This led to TNF and COX-2 mRNA destabilization. In contrast, AgC10 did not affect p38 activation induced by TNF. Furthermore, AgC10 inhibition must lie upstream in the MAPK activation pathway by LPS, since this mucin also inhibited extracellularly regulated kinase (ERK) and Jun kinase (JNK)activation. Hippocampal memory is associative and, to study it more effectively than with previous paradigms, a new learning task is described which is unusual in requiring the incidental encoding of flavour,place paired associates, with the readout of successful storage being successful recall of a place given the flavour with which it was paired. NMDA receptor-dependent synaptic plasticity is shown to be critical for the encoding and intermediate storage of memory traces in this task, while AMPA receptor-mediated fast synaptic transmission is necessary for memory retrieval. Typically, these rapidly encoded traces decay quite rapidly over time. Synaptic potentiation also decays rapidly, but can be rendered more persistent by a process of cellular consolidation in which synaptic tagging and capture play a key part in determining whether or not it will be persistent. Synaptic tags set at the time of an event, even many trivial events, can capture the products of the synthesis of plasticity proteins set in train by events before, during or even after an event to be remembered. Tag,protein interactions stabilize synaptic potentiation and, by implication, memory. The behavioural implications of tagging are explored. Finally, using a different protocol for flavour,place paired associate learning, it is shown that rats can develop a spatial schema which represents the relative locations of several different flavours of food hidden at places within a familiar space. This schema is learned gradually but, once acquired, enables new paired associates to be encoded and stored in one trial. Their incorporation into the schema prevents rapid forgetting and suggests that schema play a key and hitherto unappreciated role in systems-level memory consolidation. (2004) Science 303, 1831,1838). However, its function has not been fully clarified. Here we used thermoluminescence and flash-induced chlorophyll fluorescence measurements to characterize the effect of the D1-H333E, D1-H332D and D1-H332S mutations on the electron transport of Photosystem II in intact cells of the cyanobacterium Synechocystis 6803. Although the mutants are not photoautotrophic they all show flash-induced thermoluminescence and chlorophyll fluorescence, which originate from the S2QA, and S2QB, recombinations demonstrating that charge stabilization takes place in the water oxidizing complex. However, the conversion of S2 to higher S states is inhibited and the energetic stability of the S2QA, charge pair is increased by 75, 50 and 7 mV in the D1-H332D, D1-H332E and D1-H332S mutants, respectively. This is most probably caused by a decrease of Em(S2/S1). Concomitantly, the rate of electron donation from Mn to Tyr-Z, during the S1 to S2 transition is slowed down, relative to the wild type, 350- and 60-fold in the D1-H332E and D1-H332D mutants, respectively, but remains essentially unaffected in D1-H332S. A further effect of the D1-H332E and D1-H332D mutations is the retardation of the QA to QB electron transfer step as an indirect consequence of the donor side modification. Our data show that although the His residue in the D1-332 position can be substituted by other metal binding residues for binding photo-oxidisable Mn it is required for controlling the functional redox energetics of the Mn cluster. Biochem.269, 5564,5571], we showed that the precursor of outer-membrane protein PhoE of Escherichia coli with a Gly to Leu substitution at position ,10 in the signal sequence (G-10L) is targeted to the SecYEG translocon via the signal-recognition particle (SRP) route, instead of via the SecB pathway. Here, we studied the fate of the mutant precursor in a prlA4 mutant strain. prlA mutations, located in the secY gene, have been isolated as suppressors that restore the export of precursors with defective signal sequences. Remarkably, the G-10L mutant precursor, which is normally exported in a wild-type strain, accumulated strongly in a prlA4 mutant strain. In vitro cross-linking experiments revealed that the precursor is correctly targeted to the prlA4 mutant translocon. However, translocation across the cytoplasmic membrane was defective, as appeared from proteinase K-accessibility experiments in pulse-labeled cells. Furthermore, the mutant precursor was found to accumulate when expressed in a secY40 mutant, which is defective in the insertion of integral-membrane proteins but not in protein translocation. Together, these data suggest that SecB and SRP substrates are differently processed at the SecYEG translocon. [source] Interleukin-6-induced proliferation of pre-B cells mediated by receptor complexes lacking the SHP2/SOCS3 recruitment sites revisitedFEBS JOURNAL, Issue 24 2001Kerstin Friederichs Interleukin-6 (IL-6) induces B-cell proliferation by binding to receptor complexes composed of a specific ,-receptor (gp80; CD126) and the signal transducing receptor subunit gp130 (CD130). Immediately after receptor complex activation, signal transducers and activators of transcription (STATs) 1 and 3 and the Src-homology domain-containing protein tyrosine phosphatase 2 (SHP2) are recruited to gp130 and subsequently tyrosine phosphorylated. The activated dimerized STATs translocate to the nucleus and bind to enhancer elements of IL-6-inducible genes. SHP2 acts as an adapter and links the Jak/STAT pathway to the Ras/Raf/MAPK cascade but it is also involved in signal attenuation. Whereas STAT3 activation appears to be crucial for all biological activities of IL-6, the requirement of SHP2-activation depends on the individual biological response analyzed. The requirement of SHP2 activation for the pre-B cell (Ba/F3) proliferation has been reported previously [Fukada, T., Hibi, M., Yamanaka, Y., Takahashi-Tezuka, M., Fujitani, Y., Yamaguchi, T., Nakajima, K. & Hirano, T. (1996) Immunity5, 449,460]. [source] Three pheromone-binding proteins in olfactory sensilla of the two silkmoth species Antheraea polyphemus and Antheraea pernyiFEBS JOURNAL, Issue 10 2000Rosario Maida Females of the sibling silkmoth species Antheraea polyphemus and A. pernyi use the same three sex pheromone components in different ratios to attract conspecific males. Accordingly, the sensory hairs on the antennae of males contain three receptor cells sensitive to each of the pheromone components. In agreement with the number of pheromones used, three different pheromone-binding proteins (PBPs) could be identified in pheromone-sensitive hairs of both species by combining biochemical and molecular cloning techniques. MALDI-TOF MS of sensillum lymph droplets from pheromone-sensitive sensilla trichodea of male A. polyphemus revealed the presence of three major peaks with m/z of 15702, 15752 and 15780 and two minor peaks of m/z 15963 and 15983. In Western blots with four antisera raised against different silkmoth odorant-binding proteins, immunoreactivity was found only with an anti-(Apol PBP) serum. Free-flow IEF, ion-exchange chromatography and Western blot analyses revealed at least three anti-(Apol PBP) immunoreactive proteins with pI values between 4.4 and 4.7. N-Terminal sequencing of these three proteins revealed two proteins (Apol PBP1a and Apol PBP1b) identical in the first 49 amino acids to the already known PBP (Apol PBP1) [Raming, K., Krieger, J. & Breer, H. (1989) FEBS Lett.256, 2215,2218] and a new PBP having only 57% identity with this amino-acid region. Screening of antennal cDNA libraries with an oligonucleotide probe corresponding to the N-terminal end of the new A. polyphemus PBP, led to the discovery of full length clones encoding this protein in A. polyphemus (Apol PBP3) and in A. pernyi (Aper PBP3). By screening the antennal cDNA library of A. polyphemus with a digoxigenin-labelled A. pernyi PBP2 cDNA [Krieger, J., Raming, K. & Breer, H. (1991) Biochim. Biophys. Acta1088, 277,284] a homologous PBP (Apol PBP2) was cloned. Binding studies with the two main pheromone components of A. polyphemus and A. pernyi, the (E,Z)-6,11-hexadecadienyl acetate (AC1) and the (E,Z)-6,11-hexadecadienal (ALD), revealed that in A. polyphemus both Apol PBP1a and the new Apol PBP3 bound the 3H-labelled acetate, whereas no binding of the 3H-labelled aldehyde was found. Electrostatic interactions between negatively charged residues of the antibody Fv and peptide residues were studied by mutagenesis of both antibody and peptide residues and double-mutant cycles. Several interactions could be assigned unambiguously: F96(L) of the antibody interacts with Pro13 of the peptide, H52(H) interacts with Ile7, Ile9 and Gln10 and D56(H) interacts with Arg11. The interactions of the light-chain tyrosines with Pro13 and Gly14 could be assigned to either Y30a(L) and Y32(L), respectively, or Y32(L) and Y49(L), respectively. Three heavy-chain tyrosines interact with Ile7, Ile20 and Phe17. Several combinations of assignments involving Y32(H), Y53(H), Y96(H) and Y100a(H) may satisfy the NMR and mutagenesis constraints, and therefore at this stage the interactions of the heavy-chain tyrosines were not taken into account. The unambiguous assignments [F96(L), H52(H) and D56(H)] and the two possible assignments of the light-chain tyrosines were used to dock the peptide into the antibody-combining site. According to sequence homology, mature LPH contains two different regions (III and IV), each of them homologous to family 1 glycosidases and each with a putative active site. There has been some discrepancy with regard to the assignment of enzymatic activity to the two active sites. Here we show differential reactivity of the two active sites with mechanism-based glycosidase inhibitors. When LPH is treated with 2,,4,-dinitrophenyl 2-deoxy-2-fluoro-,- d -glucopyranoside (1) and 2,,4,-dinitrophenyl-2-deoxy-2-fluoro-,- d -galactopyranoside (2), known mechanism-based inhibitors of glycosidases, it is observed that compound 1 preferentially inactivates the phlorizin hydrolase activity whereas compound 2 is selective for the lactase active site. On the other hand, glycals (d -glucal and d -galactal) competitively inhibit lactase activity but not phlorizin hydrolase activity. This allows labeling of the phlorizin site with compound 1 by protection with a glycal. By differential labeling of each active site using 1 and 2 followed by proteolysis and MS analysis of the labeled fragments, we confirm that the phlorizin hydrolysis occurs mainly at the active site located at region III of LPH and that the active site located at region IV is responsible for the lactase activity.
