Frontal nasal prominence expression driven by Tcfap2a relies on a conserved binding site for STAT proteins

Abstract The AP-2 transcription factor family is linked with development of the head and limbs in both vertebrate and invertebrate species. Recent evidence has also implicated this gene family in the evolution of the neural crest in chordates, a critical step that allowed the development and elaboration of the vertebrate craniofacial skeleton. In mice, the inappropriate embryonic expression of one particular AP-2 gene, Tcfap2a, encoding AP-2,, results in multiple developmental abnormalities, including craniofacial and limb defects. Thus, Tcfap2a provides a valuable genetic resource to analyze the regulatory hierarchy responsible for the evolution and development of the face and limbs. Previous studies have identified a 2-kilobase intronic region of both the mouse and human AP-2, locus that directs expression of a linked LacZ transgene to the facial processes and the distal mesenchyme of the limb bud in transgenic mice. Further analysis identified two highly conserved regions of ,200,400 bp within this tissue-specific enhancer. We have now initiated a transgenic and biochemical analysis of the most important of these highly conserved regions. Our analysis indicates that although the sequences regulating face and limb expression have been integrated into a single enhancer, different cis -acting sequences ultimately control these two expression domains. Moreover, these studies demonstrate that a conserved STAT binding site provides a major contribution to the expression of Tcfap2a in the facial prominences. Effects of experimental small-scale grassland fragmentation on spatial distribution, density, and persistence of ant nests

Abstract., 1.,Grassland fragmentation is expected to influence the abundance of different invertebrate species to a different extent. Fragmentation-related effects are of particular importance in species that interact with many other species. 2.,The density and spatial distribution of nests of 15 ant species in experimentally fragmented calcareous grasslands at three sites in the Northern Swiss Jura mountains were examined. Fragments of different size (0.25 m2, 2.25 m2, and 20.25 m2) were isolated by a 5-m wide strip of frequently mown vegetation. Control plots of corresponding size were situated in adjacent undisturbed grassland. 3.,Three years after initiation of the experiment, ant nest density did not differ between fragments and control plots. Six years after initiation of the experiment, however, ant nest density and forager abundance were higher in large fragments than in large control plots. Ant nests tended to occur more frequently along the edge of fragments than in the core area. Persistence time of nests of the most abundant species, Lasius paralienus, tended to be shorter in fragments than in control plots. Furthermore, persistence time was longer in nests situated close to the fragment edge than in nests in the core area. 4.,Effects on nest density, edge effects on the spatial distribution of nests, and the relationships between nest density and environmental factors were more pronounced when only nests of L. paralienus were considered. The implications of these findings for plant and other invertebrate species are discussed. [source]


Effects of 4-nonylphenol on the endocrine system of the shore crab, Carcinus maenas

Abstract There is a considerable body of evidence to suggest that many anthropogenic chemicals, most notably xeno-estrogens, are able to disrupt the endocrine system of vertebrates. There have been few comparable studies on the effects of exposure to these chemicals that may serve as biomarkers of endocrine disruption in aquatic invertebrate species. In addition, the evidence available is complex, conflicting, and far from conclusive. The present study aimed to investigate the impact of the xeno-estrogen 4-nonylphenol (4-NP, nominal concentrations 10,100 ,g L,1) on the regulation and functioning of the endocrine system of the shore crab Carcinus maenas. It also set out to establish whether 4-NP are causing the effects (i.e., changes of exoskeletons including secondary sexual characteristics, pheromonally mediated behavior and ecdysone levels, and the presence of vt in the male hepatopancreas) found recently in wild shore crabs (Lye et al.,2005). The study utilizes morphological (e.g., gonadosomatic and hepatosomatic indices) and hormonal (ecdysteroid moulting hormone levels and the induction of female specific proteins, vitellins) biomarkers using radioimmunoassay and an indirect enzyme linked immunosorbent assay applied to the soluble protein fraction of adult male hepatopancreatic homogenates. Exposure of C. maenas to an effective concentration as low as 1.5 ,g L,1 4-NP resulted in a reduced testis weight, increased liver weight, and altered levels of ecdysone equivalents compared to controls. Induction of vitellin-like proteins was absent in all samples tested. The ecological implications and the possible mechanisms for the action of 4-NP on the response of the shore crab to xeno-estrogen exposure are discussed. © 2008 Wiley Periodicals, Inc. The role of biomarkers to assess oil-contaminated sediment quality using toxicity tests with clams and crabs,

Abstract A 28-d bioassay was conducted with two invertebrate species with different feeding habits, the clam Ruditapes philippinarum and the shore crab Carcinus maenas. The purpose of the present study was to assess the quality of sediments affected by oil spills in different areas of the Spanish coast. The organisms were exposed to environmental samples of oil-contaminated sediments during four weeks and, after the experiment, a suite of biomarkers of exposure was measured: The phase one detoxification system was assessed by ethoxyresorufin- O -deethylase (EROD) activity; glutathione- S -transferase (GST) is a phase-two detoxification enzyme but also is implicated in oxidative stress events; glutathione peroxidase (GPX), glutathione reductase (GR), and the ferric reducing ability of plasma (FRAP) assay were analyzed to determine the antioxidant activity of the tissues. The biomarker results were correlated with the chemical compounds bound to sediments (polycyclic aromatic hydrocarbons [PAHs], polychlorinated biphenyls [PCBs], Zn, Cd, Pb, Cu, Ni, Co, V) and a principal component analysis was carried out with the purpose of linking all the variables and to detect those contaminated sediments potentially harmful to the biota. Results showed induction of biomarkers in both invertebrate species and significant differences (p < 0.05; p < 0.01) were established among sediments affected by different spills. The use of the selected biomarkers together with the sediment chemical analysis assesses the bioavailability of contaminants and has proven to be a suitable tool to monitor the environmental quality of sediments affected by oil spills. 4-nonylphenol-induced toxicity and apoptosis in Hydra attenuata

Abstract Effects of 4-nonylphenol (4-NP) onthe morphology and survival of the cnidarian Hydra attenuata were studied under acute exposure conditions. The lethal concentration value inducing 50% mortality after 96 h was 97.5 ± 20 ,g/L, whereas the lethal concentration value inducing 10% mortality after 96 h was 64 ± 25.5 ,g/L. The no-observed-effect concentration based on morphological criteria was less than 25 ,g/L. Hydra was one of the most sensitive freshwater invertebrate species behind the amphipod Hyalella azteca. Toxicity effects appeared rapidly and did not evolve substantially between 24 and 96 h of exposure. Induction of apoptosis was registered during the first hour of exposure to 4-NP at lethal concentrations, indicating rapid effects of the chemical. Abnormal increase of apoptosis may explain the acute toxicity of 4-NP in hydra. Results show that hydra viability is affected in the short term at 4-NP concentrations normally found in contaminated sites, but not at those concentrations reflecting lower levels of environmental contamination. Toxicity and fate of two munitions constituents in spiked sediment exposures with the marine amphipod Eohaustorius estuarius

Abstract The lethal toxicity of the explosive compounds 14C-labeled 2,4,6-trinitrotoluene (TNT) and nonradiolabeled hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) to the estuarine amphipod Eohaustorius estuarius was investigated in 10-d spiked sediment exposures. The 10-d median lethal concentration (LC50) was determined using the sum molar initial concentration of TNT, ami-nodinitrotoluenes (ADNTs), and diaminonitrotoluenes (DANTs), as determined by high-performance liquid chromatography (HPLC), and collectively referred to as HPLC-TNT*. Despite expectations of higher toxicity in sandy sediment (Yaquina Bay [YB], OR, USA) compared to relatively fine-grained sediment (San Diego Bay [SDB], CA, USA), LC50 values were similar: 159 and 125 ,mol/kg, for YB and SDB sediments, respectively. When expressed as the sum of TNT and all its degradation products (14C-TNT*), LC50s were approximately two times the corresponding LC50s determined by HPLC. The HPLC-TNT* fraction likely corresponds to the most bioavailable and toxic transformation products. The concentrations of 14C-TNT* in tissues were substantially higher than those for HPLC-TNT*, suggesting that compounds other than TNT and its major aminated transformation products were prevalent. Critical body residues were similar for exposures to SDB (11.7 ,mol/kg) and YB sediments (39.4 ,mol/kg), despite marked differences in the nature of compounds available for uptake in the exposure media. The critical body residues for E. estuarius are lower than those reported for other aquatic invertebrates (83,172 ,mol/kg). Unlike observations for TNT, RDX was only loosely associated with SDB sediment, with near complete recovery of the parent compound by chemical analysis. Exposure to RDX did not result in significant mortality even at the highest measured sediment concentration of 10,800 ,mol/kg dry weight, nor tissue concentrations as high as 96 ,mol/kg wet weight. The lack of RDX lethal effects in this study is consistent with results reported for other invertebrate species. [source]


Abnormalities in sexual development of the amphipod Gammarus pulex (L.) found below sewage treatment works

Abstract Increasing numbers of widely used industrial, agricultural, and natural chemicals are known to elicit endocrine-disrupting effects in a wide range of vertebrate and invertebrate species. The objective of this study was to determine whether the sexual development of the freshwater crustacean Gammarus pulex (L.) was affected below sewage treatment works (STW) previously known to contain endocrine-disrupting chemicals in their effluent. The gonadal structure, external sexual characteristics, and size of gammarids from exposed sites were compared to those of gammarids from a reference site. No significant difference was found in the gonadal structure of males collected below two STW. However, a highly significant number of females collected from a site known to elicit high estrogenic responses in vertebrates displayed an abnormal structure of oocytes in vitellogenesis. Body size was significantly shorter and male/female size differential was significantly reduced below one of the STW. Analysis of gnathopod and genital papillae length data suggests that different allometric relationships of these organs to body size exist between sample sites. [source]


A novel role for polyamines in adult neurogenesis in rodent brain

Abstract Although neurogenesis in the adult is known to be regulated by various internal cues such as hormones, growth factors and cell-adherence molecules, downstream elements underlying their action at the cellular level still remain unclear. We previously showed in an insect model that polyamines (putrescine, spermidine and spermine) play specific roles in adult brain neurogenesis. Here, we demonstrate their involvement in the regulation of secondary neurogenesis in the rodent brain. Using neurosphere assays, we show that putrescine addition stimulates neural progenitor proliferation. Furthermore, in vivo depletion of putrescine by specific and irreversible inhibition of ornithine decarboxylase, the first key enzyme of the polyamine synthesis pathway, induces a consistent decrease in neural progenitor cell proliferation in the two neurogenic areas, the dentate gyrus and the subventricular zone. The present study reveals common mechanisms underlying birth of new neurons in vertebrate and invertebrate species. [source]


EVOLUTION OF POECILOGONY AND THE BIOGEOGRAPHY OF NORTH AMERICAN POPULATIONS OF THE POLYCHAETE STREBLOSPIO

Abstract. Invertebrate interspecific developmental patterns can be highly variable and, taxonomically, are considered only weakly constrained. Intraspecifically, some invertebrate species possess multiple developmental modes,a condition known as poecilogony. Closer examination of most putative poecilogenous species, however, has not supported poecilogony, but rather has uncovered hidden or cryptic species. The polychaete Streblospio benedicti is a well-known, poecilogenous species found along the coast of North America. We collected mitochondrial cytochrome subunit I DNA sequence data from 88 individuals taken from 11 locations along the Atlantic, Gulf, and Pacific Coasts of the United States to provide a phylogenetic framework from which to interpret intraspecific variation in larval life history and brooding structure morphology in this species. Our results are consistent with a recent revision of the species into two separate species: S. benedicti, a pouched brooding form distributed along the Atlantic and Pacific Coasts, and S. gynobranchiata, a branchiate brooding form in the Gulf of Mexico. Contrary to the redescription, S. benedicti is paraphyletic because the pouched brooding population in Vero Beach, Florida shows strong genetic affinity with Gulf of Mexico populations (S. gynobranchiata). However, S. benedicti is a true poecilogenous species, with both lecithotrophic and planktotrophic individuals possessing identical mitochondrial DNA haplotypes. Crossbreeding experiments further support the molecular phylogeny with reproductive isolation demonstrated between, but not within, the major phylogenetic clades consistent with the previously described species. The genetic break near Vero Beach, Florida, corresponds to a well-known phylogeographic boundary, but the estimated time of separation for the Streblospio spp., approximately 10 million years before present, predates all other known phylogeographic subdivisions in this area. This suggests that biogeographic sundering in this region is a recurrent event. Divergence times within the major Streblospio spp. clades are recent and indicate that changes in larval life history as well as brooding structure morphology are highly plastic and can evolve rapidly. Visualisation of standardized life-history patterns

Abstract The life-history patterns of fish and invertebrate species are complex. But much of this complexity can be captured in simple diagrams of coastal transects, where juveniles usually occur in larger numbers in shallow waters, while adults generally inhabit deeper, offshore waters. Iconographic representations of generalized life-history patterns and depth profiles, with specific key life-history parameters can capture much of these standardized patterns, including spawning areas, nursery/juvenile distributions, adult distributions and spawning migrations. Several examples presented here from a wide range of habitats and ecosystems (temperate and tropical waters, demersal, deep water, pelagic and coral reefs), including an example of different stocks of the same species, illustrate some general patterns with regard to water depth and distance from shore. The present approach should be viewed as a first step towards obtaining standardized patterns about key life-history parameters, and will hopefully lead to incorporation into management of life-history interconnectivity between different fishery sectors or gears. This may contribute to sustainable, ecosystem-based approaches to management by informing policy options when faced with decisions to rationalize overcapitalized fisheries. [source]


Effects of increased flow in the main stem of the River Rhine on the invertebrate communities of its tributaries

Summary 1. We hypothesised that increased flow in the main stem of the River Rhine would influence the invertebrate communities of its tributaries and therefore investigated the invertebrate fauna of six tributaries over 2 years. 2. We collected quantitative invertebrate samples at three sites in each tributary: in the tributary mouth (influenced by Rhine water whenever flow in the Rhine exceeded mean annual level), in the zone reached by average floods (return period 1.5 years) in the Rhine (average flood level sites), and immediately upstream of the range of extreme Rhine floods (reference sites). Samples were taken in spring, summer and autumn of each year, at different flow levels of the Rhine. We also compared substratum composition at the three sites. 3. Tributary mouth sites had the finest substratum, the lowest total invertebrate density and the lowest taxon richness. At average flood level and reference sites, these three parameters were similar. 4. Taxa known to prefer larger rivers were mostly confined to the tributary mouth sites, and species preferring upland streams dominated at the average flood level and reference sites. 5. Multivariate analyses confirmed the influence of the Rhine on the tributary mouth sites. Invasive invertebrate species, which usually appear only in the Rhine itself, were found at the tributary mouth sites but not further up in the tributaries. 6. Our study shows that increased flow in the main stem of the Rhine influenced substratum composition and invertebrate communities at the tributary mouth sites. These results imply that the relationship between the main stem of a river and its tributaries is not one-way (from tributary to main stem), but rather a two-way interaction. [source]


Persistent effects of a discrete warming event on a polar desert ecosystem

Abstract A discrete warming event (December 21, 2001,January 12, 2002) in the McMurdo Dry Valleys, Antarctica, enhanced glacier melt, stream flow, and melting of permafrost. Effects of this warming included a rapid rise in lake levels and widespread increases in soil water availability resulting from melting of subsurface ice. These increases in liquid water offset hydrologic responses to a cooling trend experienced over the previous decade and altered ecosystem properties in both aquatic and terrestrial ecosystems. Here, we present hydrological and meteorological data from the McMurdo Dry Valleys Long Term Ecological Research project to examine the influence of a discrete climate event (warming of >2 °C) on terrestrial environments and soil biotic communities. Increases in soil moisture following this event stimulated populations of a subordinate soil invertebrate species (Eudorylaimus antarcticus, Nematoda). The pulse of melt-water had significant influences on Taylor Valley ecosystems that persisted for several years, and illustrates that the importance of discrete climate events, long recognized in hot deserts, are also significant drivers of soil and aquatic ecosystems in polar deserts. Thus, predictions of Antarctic ecosystem responses to climate change which focus on linear temperature trends may miss the potentially significant influence of infrequent climate events on hydrology and linked ecological processes. Decline in a dominant invertebrate species contributes to altered carbon cycling in a low-diversity soil ecosystem

Abstract Low-diversity ecosystems cover large portions of the Earth's land surface, yet studies of climate change on ecosystem functioning typically focus on temperate ecosystems, where diversity is high and the effects of individual species on ecosystem functioning are difficult to determine. We show that a climate-induced decline of an invertebrate species in a low-diversity ecosystem could contribute to significant changes in carbon (C) cycling. Recent climate variability in the McMurdo Dry Valleys of Antarctica is associated with changes in hydrology, biological productivity, and community composition of terrestrial and aquatic ecosystems. One of the greatest changes documented in the dry valleys is a 65% decrease in the abundance of the dominant soil invertebrate (Scottnema lindsayae, Nematoda) between 1993 and 2005, illustrating sensitivity of biota in this ecosystem to small changes in temperature. Globally, such declines are expected to have significant influences over ecosystem processes such as C cycling. To determine the implications of this climate-induced decline in nematode abundance on soil C cycling we followed the fate of a 13C tracer added to soils in Taylor Valley, Antarctica. Carbon assimilation by the dry valley nematode community contributed significantly to soil C cycling (2,7% of the heterotrophic C flux). Thus, the influence of a climate-induced decline in abundance of a dominant species may have a significant effect on ecosystem functioning in a low-diversity ecosystem. [source]


Predicting the effects of marine climate change on the invertebrate prey of the birds of rocky shores

By the end of the 21st century models of climate change predict that the air temperature over most of the British Isles will increase by between 2 and 3 °C and sea-level will rise by 40,50 cm. Over that period it will become windier and mean wave height will increase, as will the frequency of storms. These changes in climate and weather will impact the intertidal zone of the UK and will cause distribution changes in many of the common invertebrate species that live there. Where these changes are severe they may well impact on patterns of distribution of ducks and wading birds. In the British Isles a number of organisms live close to their geographical limits of distribution. Some of these species might be expected to extend their range as climatic restraints are relaxed. Species currently limited by cool summers or winter cold will move northwards. In most cases the effects on the distribution of waterbirds will be small. For example, the replacement of the Northern Limpet Patella vulgata by the Southern Limpet P. depressa is unlikely to adversely affect Eurasian Oystercatchers Haematopus ostralegus. Of wider concern is the possibility that as climate warms the abundance and productivity of brown algae will decrease. This is likely to have two significant effects for waders. First, it would represent a loss of potentially rich feeding grounds for species such as Ruddy Turnstone Arenaria interpres that feed on small easily desiccated invertebrates living on or below the seaweed. Secondly, as algae die or are broken away the resulting debris is exported to sediment habitats where it considerably boosts the in situ production of bacteria at the base of the food web. An increase in sea-level will only have a major impact on the extent of rocky shore invertebrate communities where shore topography prevents the upward migration of the biota. Where a seawall limits shores, for example, biological production will be curtailed as the area available for colonization decreases. Increases in the size of waves and the frequency of storms will mimic increasing exposure and there will be a significant reduction in algal production in areas that are affected. [source]


Conservation strategy maps: a tool to facilitate biodiversity action planning illustrated using the heath fritillary butterfly

Summary 1.,The UK Biodiversity Action Plan (UKBAP) identifies invertebrate species in danger of national extinction. For many of these species, targets for recovery specify the number of populations that should exist by a specific future date but offer no procedure to plan strategically to achieve the target for any species. 2.,Here we describe techniques based upon geographic information systems (GIS) that produce conservation strategy maps (CSM) to assist with achieving recovery targets based on all available and relevant information. 3.,The heath fritillary Mellicta athalia is a UKBAP species used here to illustrate the use of CSM. A phase 1 habitat survey was used to identify habitat polygons across the county of Kent, UK. These were systematically filtered using relevant habitat, botanical and autecological data to identify seven types of polygon, including those with extant colonies or in the vicinity of extant colonies, areas managed for conservation but without colonies, and polygons that had the appropriate habitat structure and may therefore be suitable for reintroduction. 4.,Five clusters of polygons of interest were found across the study area. The CSM of two of them are illustrated here: the Blean Wood complex, which contains the existing colonies of heath fritillary in Kent, and the Orlestone Forest complex, which offers opportunities for reintroduction. 5.,Synthesis and applications. Although the CSM concept is illustrated here for the UK, we suggest that CSM could be part of species conservation programmes throughout the world. CSM are dynamic and should be stored in electronic format, preferably on the world-wide web, so that they can be easily viewed and updated. CSM can be used to illustrate opportunities and to develop strategies with scientists and non-scientists, enabling the engagement of all communities in a conservation programme. CSM for different years can be presented to illustrate the progress of a plan or to provide continuous feedback on how a field scenario develops. [source]


Modelling the efficacy of entomopathogenic nematodes in the regulation of invertebrate pests in glasshouse crops

Summary 1. ,Entomopathogenic nematodes belonging to the families Heterorhabditidae and Steinernematidae are lethal obligate parasites of a wide range of invertebrate species. These nematodes exhibit many characteristics that make them ideal candidates as biological control agents of insect pests (rapid host death, high reproductive rates, easily mass-reared in vitro, easy application techniques). 2. ,However, at present, the number of pest species to which these nematodes are applied successfully is small. Clearly, there is a need to develop existing knowledge of the nematode into a more complete understanding of the nematode,pest system as a whole. 3. ,To consider the potential of entomopathogenic nematodes as biological control agents, we adopted a generalized analytical modelling approach and, using realistic parameter estimates, determined the conditions under which these nematodes can regulate a pest population. 4. ,Stability analyses suggested that entomopathogenic nematodes may not be capable of regulating a host population to a stable equilibrium. Long-term persistence of the host and nematode population is unlikely, due to the highly destabilizing effects of the parasite,host relationship. As such, these nematodes may be better suited to short-term control through inundative application techniques rather than long-term regulation. 5. ,This preliminary generalized model highlights areas where further work is needed. This includes estimation of the probability of nematode infection in the field, the effect of host size on the transmission cycle and the influence of spatial heterogeneity on stability. [source]


Fishing near sea-cage farms along the coast of the Turkish Aegean Sea

Summary This study reports on the yield of commercially important species captured intentionally around and under fish-cage farms using specific types of gear from both sea-cage farmers and artisanal fishermen, and to identify the species composition of these catches. The 2004,2008 research was carried out in 21 fish farms, randomly chosen along the coast of Izmir Province on the Aegean Sea. A total of 91 persons were interviewed, including directors, fishers from the fish farms and artisanal fishermen. The technical characteristics of a special trap used by some fish farmers were diagramed. Total wild fish catch quantities as well the species raised in the sea-cages were recorded. A total of 34 finfish and four invertebrate species were identified from the sea-cage farm fishery. Eight fish species according to the descending quantities raised were bogue (Boops boops), grey mullet (Mugil spp.), blackspot seabream (Pagellus bogaraveo), salema (Sarpa salpa), common two-banded seabream (Diplodus vulgaris), annular seabream (Diplodus annularis), striped seabream (Lithognathus mormyrus) and gilthead seabream (Sparus aurata). Mean quantities of wild fish caught from 2004 to 2008 in a sampled farm were 13 998 ± 210, 34 434 ± 482, 30 116 ± 529, 27 893 ± 429, and 32 366 ± 808 kg, respectively. Landed and discarded catches from commercial prawn trammel net fishery

Summary This study presents estimates of numbers and biomass of discarded and landed catches from the commercial prawn trammel net fishery in ,zmir Bay. A total of 43 finfish and 29 invertebrate species were identified in catches sampled between May and October 2003, with 26 species landed by fishers and 46 species discarded. Melicertus kerathurus, Mullus barbatus and Solea solea dominated the landed catches. Average ratio of landed to discarded fish by number and weight was estimated as 1 : 2.32 and 1 : 0.77, respectively, demonstrating relatively low by-catch ratios compared to other shrimp by-catch studies. This study shows that trammel nets have a much lower discard ratio than the shrimp trawls in other parts of the world. The benefits of shifting toward passive trammel nets in coastal areas are discussed. [source]


Marine biogeographical structure in two highly dispersive gastropods: implications for trans-Tasman dispersal

Abstract Aim, Recent genetic and ecological studies of marine invertebrate species with planktotrophic larvae have inferred high rates of gene flow across wide oceanic barriers. We therefore aim to test for the genetic signature of long-distance dispersal in two widespread and abundant marine gastropod taxa. Location, The intertidal and shallow subtidal zones of southern Australia and New Zealand (NZ), which house similar marine invertebrate assemblages despite being separated by the 2000-km-wide Tasman Sea. Methods, We used mtDNA cytochrome oxidase I gene sequence analysis of two gastropod genera exhibiting trans-Tasman distributions, namely Austrolittorina (Littorinidae) (139 specimens; 28 localities) and Scutus (Fissurellidae) (154 specimens; 32 localities). The cool-temperate Australian (A. unifasciata; S. antipodes) and NZ (A. antipodum; S. breviculus) taxa within each genus are morphologically similar but of uncertain taxonomic status. Results, The mtDNA analyses indicate major trans-Tasman genetic discontinuities for both gastropod genera, with no evidence of recent or ongoing intercontinental gene flow. Although both Scutus and Austrolittorina show significant east,west structure within southern Australia , consistent with recent studies of regional marine phylogeography , neither taxon exhibits significant differentiation within NZ. Main conclusions, Morphologically conserved but biogeographically disjunct gastropod populations may exhibit striking phylogeographic discontinuities, even when dispersal abilities appear to be high. On the basis of these data we reject recent calls for the synonymy of NZ and Australian lineages. [source]


Discovering new invertebrate neuropeptides using mass spectrometry

Abstract Neuropeptides are a complex set of messenger molecules controlling a wide array of regulatory functions and behaviors within an organism. These neuromodulators are cleaved from longer protein molecules and often experience numerous post-translational modifications to achieve their bioactive form. As a result of this complexity, sensitive and versatile analysis schemes are needed to characterize neuropeptides. Mass spectrometry (MS) through a variety of approaches has fueled the discovery of hundreds of neuropeptides in invertebrate species in the last decade. Particularly successful are direct tissue and single neuron analyses by matrix-assisted laser desorption/ionization (MALDI) MS, which has been used to elucidate approximately 440 neuropeptides, and examination of neuronal homogenates by electrospray ionization techniques (ESI), also leading to the characterization of over 450 peptides. Additional MS methods with great promise for the discovery of neuropeptides are MS imaging and large-scale peptidomics studies in combination with a sequenced genome. © 2005 Wiley Periodicals, Inc. Abstract (1)Assessing species diversity is a basic requirement for conservation, and protecting biodiversity is a major goal of marine area conservation. (2)A case study is presented on the development of a literature-based (1870s to 2000), museum collection-based, georeferenced inventory of marine invertebrate species of the Haida Gwaii (Queen Charlotte Islands) region, Canada. (3)Database structure and quality assurance are described, along with including indigenous people's words for species towards using traditional knowledge within cooperative marine conservation area management. (4)The utility of this type of inventory is proposed as a starting point for gathering regional biodiversity knowledge, and facilitating addition of other knowledge types, towards marine area conservation. Rhodolith bed diversity in the Gulf of California: the importance of rhodolith structure and consequences of disturbance

Abstract 1.Rhodolith beds, unattached coralline reefs, support a high diversity and abundance of marine species from both hard and soft benthos. We used surveys in multiple shallow (3,20 m) beds in the Gulf of California to (1) examine seasonal patterns in associated floral and faunal diversity and abundance, (2) compare differences in faunal associations between rhodolith beds and adjacent sedimentary habitats, (3) examine the importance of complexity of rhodolith structure to community structure, and (4) estimate the impact of anthropogenic disturbance on rhodoliths and associated species. 2.Macroalgal richness was seasonal, and beds supported higher richness in winter (to 36 species) than summer (6,7 species), primarily due to foliose red algae. Strong seasonal variation in the abundance of dominant cover organisms was due to a shift from macroalgae and mat-forming colonial invertebrate species to microalgae. 3.The community in a rhodolith bed of high-density thalli (El Coyote average ,11000 thalli/ m,2) had higher richness (52 versus 30 species) and abundance of epibenthic and crypto- and in-faunal species compared with an adjacent sand community. Species diversity and abundance was particularly high for unique cryptofaunal organisms associated with rhodolith interstices. Cryptofauna reached average densities of 14.4 organisms/ cm,3 rhodolith, the majority of which were crustaceans, polychaetes and cnidarians along with rhodolith-specific chitons. 4.Results from sampling across a range of rhodolith morphs in the El Requeson bed (with lower average cryptofaunal densities of 2.3 organisms/ cm,3) revealed that the total organisms supported by a rhodolith significantly increased with both complexity (branching density) and space available (thallus volume). These data suggest that reducing the population size structure, structural complexity and cover of living rhodoliths could decrease species richness and abundance. 5.While disturbance is a natural feature of these free-living beds, increased anthropogenic disturbance from trawling, anchoring and changes in water quality can directly impact the bed community through substrate alteration. Commercial fishing threatens rhodolith beds in the Gulf of California by decreasing rhodolith size and increasing sedimentation and burial rates.
