A molecular analysis of biclonal follicular lymphoma: further evidence for bone marrow origin and clonal selection

Abstract We report a follicular lymphoma (FL) case presenting the coexistence of two tumor cell subpopulations in lymph node (LN) and bone marrow (BM), which exhibited an inverse pattern of immunoglobulin light (IgL) chain gene rearrangement and expression: Ig,,,+ in LN and Ig,+,, in BM. These tumor clones shared an identical BCL2-IgH recombination, accompanying t(14;18)(q32;q21) translocation, and an identical variable, diversity and joining segments joining with clone-specific VH somatic hypermutations on the untranslocated IgH allele. Our study provides further evidence that FL clones, originating from common progenitor cells, can be developed independently at different sites and with different IgL expression after immune selection. Neutrophil gelatinase-associated lipocalin is a marker for dysregulated keratinocyte differentiation in human skin

Abstract: Neutrophil gelatinase-associated lipocalin (NGAL) is a 25-kDa protein initially isolated from the specific granules of human neutrophils. It is a member of the highly heterogeneous lipocalin protein family, which shares a common tertiary structure. Its synthesis is induced in gastrointestinal epithelium in association with inflammation and malignancy. To gain insight into its potential role in other epithelia we have investigated the expression of NGAL in human skin embryonic development, in normal adult skin, and in skin associated with inflammation and neoplastic transformation. In the present study we report that the embryonic expression of NGAL appears to be regulated in a spatio-temporal pattern. It was induced in the interfollicular epidermis at 20,24 weeks of gestational age but thereafter progressively receded towards the hair follicles. In normal adult skin, NGAL was detected solely in association with hair follicles. However, strong induction of NGAL in the epidermis was seen in a variety of skin disorders characterized by dysregulated epithelial differentiation such as psoriasis, pityriasis rubra and squamous cell carcinoma. In these tissues production of NGAL was confined to spatially distinct subpopulations of keratinocytes underlying areas of parakeratosis, whereas skin samples lacking parakeratotic epithelium such as lichen ruber planus, acute contact eczema and basal cell carcinoma were negative for NGAL. Consistent with being a marker for disturbed terminal differentiation, NGAL immunoreactivity showed an inverse pattern when compared with that of the differentiation marker filaggrin. The biologic functions of NGAL in epithelia are not fully known, although an immunomodulatory role in host defense has been proposed. In addition, the transient interfollicular NGAL expression during skin embryogenesis along with the induction of NGAL in adult parakeratotic epidermis suggests it play a role in epithelial differentiation pathways. Prey selection by flounder, Platichthys flesus, in the Douro estuary, Portugal

Summary Prey selection by the flounder, Platichthys flesus (Linnaeus, 1758), in an estuarine nursery was investigated and the major factors influencing food choice by this species were assessed. Diet breadth was narrow, reflecting the low prey diversity observed in the benthos. A gradual ontogenetic shift from small prey such as amphipods to larger prey like polychaetes and bivalves was observed. Amphipods had positive electivity values in the upper estuary and negative values in the lower estuary.Polychaetes showed the inverse pattern. Bivalve electivity values were always positive. Differential selectivity throughout the estuary was mainly related to spatial segregation of flounders according to size, with the smaller individuals concentrating in the upper estuary and larger individuals concentrating in the lower estuary. Amphipods such as Corophium spp. play a crucial role in the flounder diet because of their small size, low mobility and diel activity pattern. As prey, the polychaete value increases throughout flounder ontogeny since the flounder size range is compatible with the larger mouth gapes and detection ability of larger fish. Bivalve electivity values for flounder are mainly related to high calorific values. The absence of Crangon crangon (Linnaeus, 1758) in the diet may be due to low water temperature since the cost,benefit involved in the capture of highly mobile prey is too high at low temperatures. It was concluded that flounder must use several sensory features to detect and capture prey in turbid estuarine waters and that field studies provide important background information on the actual predator preferences under natural conditions. Increased wild-type p53-induced phosphatase 1 (Wip1 or PPM1D) expression correlated with downregulation of checkpoint kinase 2 in human gastric carcinoma

Phosphorylation of checkpoint kinase 2 (Chk2) at Thr68 (pChk2) induced by DNA double-strand breaks is required for inhibition of cell cycle progression in the G2 phase. The purpose of the present paper was to investigate the expression of wild-type p53-induced phosphatase 1 (Wip1 or PPM1D), a negative regulator of Chk2, to better understand its role in human gastric cancer. In non-neoplastic gastric mucosa, most epithelial cells exhibited Wip1-positive and pChk2-negative immunoreactivity, whereas an inverse pattern of protein expression was detected at the surface of the foveolar epithelium. In tumor tissues, 74% of 53 gastric cancers had intense Wip1 immunoreactivity and close correlation with both tumor size (P = 0.0497) and Chk2 dephosphorylation (P = 0.0213). In MKN-74 gastric cancer cells, ionizing radiation (IR)-induced Wip1 upregulation was detected at protein levels, but the Chk2-mediated cell cycle regulatory mechanism was disrupted. In addition, protease inhibitor Z-Leu-Leu-Leu (ZLLL) effectively upregulated Wip1 levels in the presence or absence of IR, suggesting that Wip1 expression can be modulated post-transcriptionally. Understanding the Wip1-mediated signaling pathway in gastric cancer may provide useful information for the development of new chemo- and radiotherapies. [source]


I'll Take the High Road: Two Pathways to Altruistic Political Mobilization Against Regime Repression in Argentina

What led Argentine human rights activists to risk challenging state repression in the late 1970s? Chi-square analyses of 78 interviews with early activists and nonactivists suggested few commonalities among activists but revealed two distinct and inverse routes to high-risk other-centered political activism. Activists directly affected by regime violence tended to be relatively inexperienced politically, to have little experience with fear, and to see groups as comprising individuals rather than as monolithic wholes. An inverse pattern characterized activists not directly affected by regime violence: Their activism was preceded by experience in politics and survival of previous fear-evoking episodes. [source]


Effects of graded levels of dietary myo -inositol on non-specific immune and specific immune parameters in juvenile Jian carp (Cyprinus carpio var. Jian)

Abstract The aim of this study was to evaluate the effects of myo -inositol (MI) on non-specific immune and specific immune defence in fish. A total of 1050 Jian carp (Cyprinus carpio var. Jian) (22.28±0.07 g) were randomly distributed into seven groups, of three replicates each, of feeding diets containing graded levels of MI (163.5, 232.7, 384.2, 535.8, 687.3, 838.8 and 990.3 mg MI kg,1 diet). After a 60-day growth trial, an infectious trial was conducted by injection of Aeromonas hydrophila for 17 days. Results showed that the red blood cell (RBC) and the white blood cell count were significantly increased with increasing MI levels up to 535.8 mg kg,1 diet (P<0.05). The spleen index showed a tendency similar to RBC, whereas the head kidney index showed the inverse pattern (P<0.05). The phagocytic activity of leucocytes, haemagglutination titre, lysozyme activity, anti- A. hydrophila antibody titre and immunoglobulin M, after being injected with A. hydrophila, were all improved with an increase in the MI levels up to 232.7,687.3 mg kg,1 diet respectively (P<0.05). Myo -inositol did not influence serum acid phosphatase activity and total iron-binding capacity (P>0.05). These results suggested that MI could enhance non-specific immune and specific immune responses in fish. [source]
