Selected AbstractsPositive diversity,stability relationships in forest herb populations during four decades of community assemblyECOLOGY LETTERS, Issue 10 2010Martin Dov Ecology Letters (2010) Abstract It is suggested that diversity destabilizes individual populations within communities; however, generalizations are problematic because effects of diversity can be confounded by variation attributable to community type, life history or successional stage. We examined these complexities using a 40-year record of reassembly in forest herb communities in two clearcut watersheds in the Andrews Long-term Ecological Research Site (Oregon, USA). Population stability was higher among forest than colonizing species and increased with successional stage. Thus, life history and successional stage may explain some of the variability in diversity,stability relationships found previously. However, population stability was positively related to diversity and this relationship held for different forest communities, for species with contrasting life histories, and for different successional stages. Positive relationships between diversity and population stability can arise if diversity has facilitative effects, or if stability is a precursor, rather than a response, to diversity. [source] IL-23-driven encephalo-tropism and Th17 polarization during CNS-inflammation in vivoEUROPEAN JOURNAL OF IMMUNOLOGY, Issue 7 2009Gabor Gyülvészi Abstract IL-23 but not IL-12 is essential for the development of autoimmune tissue inflammation in mice. Conversely, IL-12 and IL-23 impact on the polarization of Th1 and Th17 cells, respectively. While both polarized T helper populations can mediate autoimmune inflammation, their redundancy in the pathogenesis of EAE indicates that IL-23 exerts its crucial influence on the disease independent of its T helper polarizing capacity. To study the impact of IL-23 and IL-12 on the behavior of encephalitogenic T cells in vivo, we generated BM-chimeric mice in which we can trace individual populations of IL-23 or IL-12 responsive T helper cells during EAE. We observed that T cells, which lack IL-12R,1 (no IL-12 and IL-23 signaling), fail to invade the CNS and do not acquire a Th17 phenotype. In contrast, loss of IL-12 signaling prevents Th1 polarization but does not prevent T-cell entry into the CNS. The loss of IL-12R engagement does not appear to alter T-cell expansion but leads to their accumulation in secondary lymphoid organs. We found that IL-23 licenses T cells to invade the target tissue and to exert their effector function, whereas IL-12 is critical for Th1 differentiation, but does not influence the pathogenic capacity of auto-reactive T helper cells in vivo. Soto-Ortolaza Background and purpose:, Calcium levels have been proposed to play an important role in the selective vulnerability of nigrostriatal dopaminergic neurons in Parkinson's disease (PD). Recently, an association was reported between the calcium buffer, calbindin (rs1805874) and risk of PD in a Japanese patient,control series. Methods:, We genotyped rs1805874 in four independent Caucasian patient,control series (1543 PD patients, 1771 controls). Results:, There was no evidence of an association between rs1805874 and disease risk in individual populations or in the combined series (odds ratio: 1.04, 95% CI: 0.82,1.31, P = 0.74). Discussion:, Our study shows there is no association between rs1805874 and risk for PD in four Caucasian populations. This suggests the effect of calbindin on PD risk displays population specificity. [source] NATURAL SELECTION ON A POLYMORPHIC DISEASE-RESISTANCE LOCUS IN IPOMOEA PURPUREAEVOLUTION, Issue 2 2007Joel M. Kniskern Although disease-resistance polymorphisms are common in natural plant populations, the mechanisms responsible for this variation are not well understood. Theoretical models predict that balancing selection can maintain polymorphism within a population if the fitness effects of a resistance allele vary from a net cost to a net benefit, depending upon the extent of pathogen damage. However, there have been a few attempts to determine how commonly this mechanism operates in natural plant,pathogen interactions. Ipomoea purpurea populations are often polymorphic for resistance and susceptibility alleles at a locus that influences resistance to the fungal pathogen, Coleosporium ipomoeae. We measured the fitness effects of resistance over three consecutive years at natural and manipulated levels of damage to characterize the type of selection acting on this locus. Costs of resistance varied in magnitude from undetectable to 15.5%, whereas benefits of resistance sometimes equaled, but never exceeded, these costs. In the absence of net benefits of resistance at natural or elevated levels of disease, we conclude that selection within individual populations of I. purpurea probably does not account completely for maintenance of this polymorphism. Rather, the persistence of this polymorphism is probably best explained by a combination of variable selection and meta-population processes. [source] Invasions and niche width: does niche width of an introduced crayfish differ from a native crayfish?FRESHWATER BIOLOGY, Issue 8 2009KARIN OLSSON Summary 1. Human activities have promoted the spread of species worldwide. Several crayfish species have been introduced into new areas, posing a threat to native crayfish and other biota. Invader success may depend on the ability to utilise a wide variety of habitats and resources. Successful invaders are generally expected to have broader niches and to be more plastic than non-invasive species. 2. Using stable isotope ratios of carbon and nitrogen we compared the niche widths of native noble crayfish and introduced signal crayfish, a successful invader of Swedish streams. The calculation of niche width took account of between-site differences in basal resource isotope signature ranges. We also assessed whether population density, prey biomass or prey diversity affected niche width. 3. At the species level, signal crayfish had twice the niche width of noble crayfish. However, individual populations of noble crayfish and signal crayfish in Swedish streams had similar niche widths. This suggests that signal crayfish has greater plasticity with respect to habitat utilisation and feeding than noble crayfish. Niche width in both species correlated positively with benthic invertebrate biomass and diversity, indicating that animal food sources are important for crayfish. 4. We find that assessing niche width in relation to invader success can be a useful tool trying to predict the impact of invasions on different scales. The findings in this study suggest that invaders and natives will have a similar impact on the stream scale whereas the invader will have a larger impact on the regional scale due to the ability to utilise a wider range of streams. [source] Contrasting phylogeographical patterns of two closely related species, Machilus thunbergii and Machilus kusanoi (Lauraceae), in TaiwanJOURNAL OF BIOGEOGRAPHY, Issue 5 2006Su-Hwa Wu Abstract Aim, The purpose of this paper was to study the patterns of genetic variation, demographic history, haplotype relationships and potential location of diversity centres of two closely related species, Machilus thunbergii and Machilus kusanoi. Location, The phylogeography of M. thunbergii and M. kusanoi was examined by sampling 110 and 106 individuals from 25 and 16 sampling sites, respectively, across their distributional range in Taiwan. Machilus thunbergii is distributed on the Asian mainland, South Korea, southern Japan, the Ryukyus, Taiwan and the Philippines, whereas M. kusanoi is endemic to Taiwan. These two species are closely related, and both are widely distributed in Taiwan but occupy different altitudinal zones and habitats. Methods, The range-wide variation of M. thunbergii and M. kusanoi in Taiwan was studied using chloroplast DNA (cpDNA) variations. A haplotype network was constructed with the computer program tcs. Nested clade analysis was conducted with the computer program ceodis, and various parameters of genetic diversity were calculated and neutrality tested by the computer program DnaSP. Population differentiation was estimated using the programs arlequin and hapstep. The contribution of the populations to gene diversity and to allelic richness was calculated using the software contrib. The level of divergence for each population from the remaining populations was calculated as the mean value of pairwise FST for each population against the rest of the populations. Results, Extremely low levels of genetic differentiation were found for both species. This result suggested that these two species probably survived in multiple relict refugia with different population sizes throughout the island during low-temperature periods of the Pleistocene. In addition, nested clade analysis (NCA) of cpDNA haplotypes indicated that restricted gene flow with isolation-by-distance characterized the recolonization after the Pleistocene by Tashueshan and Shiouhluan populations of M. thunbergii in the north-central area west of the Central Mountain Range (CMR). In contrast, NCA analysis indicated that a major diversity centre on the southern tip of the island (Kending population) and contiguous range expansion characterized the recolonization by M. kusanoi of northern areas along the east side of the CMR. The major diversity centres found for the two species examined were further supported by the results of the mean FST for individual populations in comparison with other populations, and of the contribution of the divergence component to the total diversity. Main conclusions, This research supports the multiple relict refugia hypothesis for both species investigated. Populations of M. thunbergii at Shiouhluan and Tashueshan in the north-central area west of the CMR represent a diversity centre currently expanding its size. A diversity centre at the southern-edge population of M. kusanoi, and a contiguous range expansion from Kending, were found. These results indicate that the M. thunbergii populations at Tashueshan and Shiouhluan and the M. kusanoi population at Kending, and even Soukar, are evolutionarily significant units for conservation programmes. [source] RAPD variation and population genetic structure in Prunus mahaleb (Rosaceae), an animal-dispersed treeMOLECULAR ECOLOGY, Issue 9 2000Pedro Jordano Abstract We examined the patterns of random amplified polymorphic DNA (RAPD) variation among seven Prunus mahaleb (Rosaceae) populations extending over , 100 km2 to examine local differentiation in relation to spatial isolation due to both geographical distance and differences in elevation. No less than 51.4% of the RAPD loci were polymorphic, but very few were fixed and among-population variation accounted for 16.46% of variation in RAPD patterns. Mean gene diversity was 0.1441, with mean Nei's genetic diversity for individual populations ranging between 0.089 and 0.149. Mean GST value across loci was 0.1935 (range, 0.0162,0.4685), giving an average estimate for Nm of 1.191. These results suggest extensive gene flow among populations, but higher GST and lower Nm values relative to other outcrossing, woody species with endozoochorous dispersal, also suggest a process of isolation by distance. The combined effect of both geographical and elevation distances and nonoverlapping flowering and fruiting phenophases on the GST matrix was partially significant, revealing only marginal isolation of the P. mahaleb populations. The matrix correlation between estimated Nm values among populations and the geographical + elevation distance matrices (r = ,0.4623, P = 0.07), suggests a marginal trend for more isolated populations to exchange less immigrants. Long-distance seed dispersal by efficient medium-sized frugivorous birds and mammals is most likely associated to the high levels of within-population genetic diversity. However, vicariance factors and demographic bottlenecks (high postdispersal seed and seedling mortality) explain comparatively high levels of local differentiation. [source] Phylogeography and genetic structure of northern populations of the yellow warbler (Dendroica petechia)MOLECULAR ECOLOGY, Issue 6 2000Emmanuel Milot Abstract Phylogeographic patterns of intraspecific variation can provide insights into the population-level processes responsible for speciation and yield information useful for conservation purposes. To examine phylogeography and population structure in a migratory passerine bird at both continental and regional geographical scales, we analysed 344 bp of mitochondrial DNA (mtDNA) control region sequence from 155 yellow warblers (Dendroica petechia) collected from seven locations across Canada and from Alaska. There is a major subdivision between eastern (Manitoba to Newfoundland) and western (Alaska and British Columbia) populations which appears to have developed during the recent Pleistocene. Some localities within these two regions also differ significantly in their genetic composition, suggesting further subdivision on a regional geographical scale. Eastern and western birds form distinct phylogeographic entities and the clustering of all western haplotypes with two eastern haplotypes suggests that the western haplotypes may be derived from an eastern lineage. Analyses based on coalescent models support this explanation for the origin of western haplotypes. These results are consistent with important features of Mengel's model of warbler diversification. From a conservation perspective they also suggest that individual populations of migrant birds may form demographically isolated management units on a smaller scale than previously appreciated. Cross-species amplification of the loci was tested in eight species of the family Thamnophilidae and all loci successfully amplified in five or more related species. [source] Indigenous domestic breeds as reservoirs of genetic diversity: the Argentinean Creole cattleANIMAL GENETICS, Issue 5 2001G. Giovambattista Contrary to highly selected commercial breeds, indigenous domestic breeds are composed of semi-wild or feral populations subjected to reduced levels of artificial selection. As a consequence, many of these breeds have become locally adapted to a wide range of environments, showing high levels of phenotypic variability and increased fitness under natural conditions. Genetic analyses of three loci associated with milk production (,S1 -casein, , -casein and prolactin) and the locus BoLA-DRB3 of the major histocompatibility complex indicated that the Argentinean Creole cattle (ACC), an indigenous breed from South America, maintains high levels of genetic diversity and population structure. In contrast to the commercial Holstein breed, the ACC showed considerable variation in heterozygosity (He) and allelic diversity (A) across populations. As expected, bi-allelic markers showed extensive variation in He whereas the highly polymorphic BoLA-DRB3 showed substantial variation in A, with individual populations having 39,74% of the total number of alleles characterized for the breed. An analysis of molecular variance (AMOVA) of nine populations throughout the distribution range of the ACC revealed that 91.9,94.7% of the total observed variance was explained by differences within populations whereas 5.3,8.1% was the result of differences among populations. In addition, the ACC breed consistently showed higher levels of genetic differentiation among populations than Holstein. Results from this study emphasize the importance of population genetic structure within domestic breeds as an essential component of genetic diversity and suggest that indigenous breeds may be considered important reservoirs of genetic diversity for commercial domestic species. [source] Inheritance and reliability of random amplified polymorphic DNA-markers in two consecutive generations of common carp (Cyprinus carpio L.)AQUACULTURE RESEARCH, Issue 2 2010Noel D Novelo Abstract Random amplified polymorphic DNA (RAPD) markers have been used in a variety of genetic studies in fisheries and aquaculture. Most population studies are performed without preliminary data demonstrating the Mendelian inheritance and reproducibility of RAPD markers. In this study, the inheritance and reproducibility of RAPD markers was examined in two consecutive generations of common carp, Cyprinus carpio L. Variability and segregation of RAPD markers were investigated in one F1 progeny and three F2 progenies. Seventy-four RAPD markers were generated by five primers using DNA extracted from the initial ornamental (koi) common carp female and wild-type colour common carp male. Fifty-five of these RAPD markers were transmitted to the F1 progeny and the inheritance patterns were analysed. Twenty RAPD markers were fully reproducible and demonstrated dominant simple Mendelian inheritance patterns in two consecutive generations. Twenty-four RAPD markers were not reproducible in all progenies. Thirteen markers displayed inheritance ratios in the progenies that did not fit simple Mendelian inheritance patterns. Non-reproducibility of RAPD markers and distorted ratios may be caused by the absence of amplification, poor amplification or by the appearance of artefact bands. Random amplified polymorphic DNA markers with poor reproducibility and non-Mendelian inheritance can lead to misinterpretations of data in population studies, resulting in errors in the estimation of genetic diversity within and between individual populations. Therefore, it is recommended to first identify the set of reproducible RAPD markers that demonstrate Mendelian inheritance before application of the RAPD technique in population studies. Lee Objective Previous studies have demonstrated that the PRL ,1149 T (minor) allele decreases prolactin expression and may be associated with autoimmune disease. The aim of this study was to determine the role of the PRL ,1149 G/T polymorphism (rs1341239) in rheumatoid arthritis (RA) susceptibility. Methods We examined the association between PRL ,1149 G/T and RA risk in 4 separate study populations, consisting of a total of 3,405 RA cases and 4,111 controls of self-reported white European ancestry. Samples were genotyped using 1 of 3 genotyping platforms, and strict quality control metrics were applied. We tested for association using a 2-tailed Cochran-Mantel-Haenszel additive, fixed-effects model. Results In the individual populations, odds ratios (ORs) for an association between PRL ,1149 T and RA risk ranged from 0.80 to 0.97. In a joint meta-analysis across all 4 populations, the OR for an association between PRL ,1149 T and RA risk was 0.90 (95% confidence interval 0.84,0.96, P = 0.001). Conclusion Our findings indicate a possible association between the PRL ,1149 T allele and decreased RA risk. The effect size is small but similar to ORs for other genetic polymorphisms associated with complex traits, including RA. [source] Phenetic analysis of the Mexican midland pinyon pines, Pinus cembroides and Pinus johannisBOTANICAL JOURNAL OF THE LINNEAN SOCIETY, Issue 2 2000A. ROMERO This account presents an analysis of allopatric, sympatric and juxtaposed populations of the pinyon pines, Pinus cembroides Zucc. and P. johannis M.-F. Robert, based on phenetic analysis of morphological data, and draws on a comparison and differentiation of P. culminicola Andresen & Beaman. A qualitative and quantitative database for populations has been assembled and used for a cluster and ordination analysis using NTSYS. It was found that individual populations of the taxa were distinguishable. The emergence of morphological intermediate populations between P. cembroides and P. johannis was confirmed. The pattern of variation inP. johannis was found to be discrete, showing three geographical groups. The phenetic similarity betweenP. johannis from Zacatecas and P. johannis from San Luis Potosi and Queretaro was lower than that of P. johannis from Zacatecas and P. culminicola from Nuevo Leon. The following traits distinguish P. cembroides and the other two species: colour and fertility of seeds; cone shape; colour and width of needles; physiognomy; seed dimensions. This last trait can be used to distinguish three subgroups of pines with white endosperm in the northern, central and southern regions of the Mexican highlands. [source] |
