    SIZE INCREMENTS DUE TO INTERINDIVIDUAL FUSIONS: HOW MUCH AND FOR HOW LONG?,

    Size increments following interindividual fusions appear as a general benefit for organisms, such as coalescing seaweeds and modular invertebrates, with the capacity to fuse with conspecifics. Using sporelings of the red algae Gracilaria chilensis C. J. Bird, McLachlan et E. C. Oliveira and Mazzaella laminarioides (Bory) Fredericq, we measured the growth patterns of sporelings built with different numbers of spores, and the magnitude and persistence of the size increments gained by fusions. Then we studied three morphological processes that could help explain the observed growth patterns. Results indicate that in these algae, coalescence is followed by immediate increase in total size of the coalesced individual and that the increment is proportional to the number of individuals fusing. However, the size increments in sporelings of both species do not last >60 d. Increasing reductions of marginal meristematic cells and increasing abundance of necrotic cells in sporelings built with increasing numbers of initial spores are partial explanations for the above growth patterns. Since sporelings formed by many spores differentiate erect axes earlier and in larger quantities than sporelings formed by one or only a few spores, differentiation, emergence, and growth of erect axes appear as a more likely explanation for the slow radial growth of the multisporic sporelings. Erect axis differentiation involves significant morphological and physiological changes and a shift from radial to axial growth. It is concluded that the growth pattern exhibited by these macroalgae after fusion differs from equivalent processes described for other organisms with the capacity to fuse, such as modular invertebrates.     Activation of nervous system development genes in bone marrow derived mesenchymal stem cells following spaceflight exposure,

    Abstract Stalled cell division in precursor bone cells and reduced osteoblast function are considered responsible for the microgravity-induced bone loss observed during spaceflight. However, underlying molecular mechanisms remain unraveled. Having overcome technological difficulties associated with flying cells in a space mission, we present the first report on the behavior of the potentially osteogenic murine bone marrow stromal cells (BMSC) in a 3D culture system, flown inside the KUBIK aboard space mission ISS 12S (Soyuz TMA-8,+,Increment 13) from March 30 to April 8, 2006 (experiment "Stroma-2"). Flight 1g control cultures were performed in a centrifuge located within the payload. Ground controls were maintained on Earth in another KUBIK payload and in Petri dishes. Half of the cultures were stimulated with osteo-inductive medium. Differences in total RNA extracted suggested that cell proliferation was inhibited in flight samples. Affymetrix technology revealed that 1,599 genes changed expression after spaceflight exposure. A decreased expression of cell-cycle genes confirmed the inhibition of cell proliferation in space. Unexpectedly, most of the modulated expression was found in genes related to various processes of neural development, neuron morphogenesis, transmission of nerve impulse and synapse, raising the question on the lineage restriction in BMSC.     Associative effects of supplementing rice straw-based diet with cornstarch on intake, digestion, rumen microbes and growth performance of Huzhou lambs

    ABSTRACT Thirty-six male Hu lambs consuming a rice straw-based diet were used in a 60-day trial to study the associative effects of cornstarch supplementation on intake, digestion, ruminal microbial population and growth performance. All animals were fed rice straw ad libitum together with 160 g/day of rapeseed meal and supplemented with cornstarch at levels of 0 (control), 60, 120 or 180 g/day, respectively. Increment of supplementary cornstarch showed little influence on rice straw intake. Optimal growth performance and highest apparent digestibility of organic matter was achieved in the 120 g/day cornstarch group (P < 0.05), while the digestibilities of neutral detergent fiber and crude protein were significantly decreased by 180 g/day cornstarch (P < 0.05). Similar results were observed for carboxymethyl cellulose activity and relative populations of cellulolytic bacteria (Ruminococcus albus, Ruminococcus flavefaciens and Fibrobacter succinogene). Blood urea nitrogen was reduced by supplementary cornstarch, indicating enhanced protein utilization efficiency. Carcass traits were all significantly improved by supplementary cornstarch. These results suggested that proper amounts of starch supplementation (within 0.5% BW) has little adverse effect on forage utilization, but could effectively improve growth performance. High levels of cornstarch, however, would decrease cellulase activity and populations of cellulolytic bacteria, and hence the digestibility of forage. [source]


    Improved Stirling engine performance through displacer surface treatment

    Abstract This study intended to improve the performance of the beta-type Stirling engine, being developed by the authors for solar energy and low-grade heat sources, by means of displacer surface treatments. Three different displacers were manufactured and tested where one of them was without any surface treatment, other was zirconium coated with 0.15,mm thickness, and the other was helically knurled with 0.30,mm track depth. Because of good thermo-physical properties, helium was used as the working fluid. The heat was supplied by an LPG burner. Tests were conducted at 360±10°C hot end temperature. The highest engine power was obtained with knurled displacer as 250,W at 545,rpm speed and corresponding to this power 4.38,Nm torque was obtained. This was followed by coated and smooth displacers. Power increments provided by the knurled displacer are 40 and 60% compared with the zirconium-coated and untreated displacers.     Testing for the Presence of Self-Similarity of Gaussian Time Series Having Stationary Increments

    A method for testing for the presence of self-similarity of a Gaussian time series with stationary increments is presented. The test is based on estimation of the distance between the time series and a set of time series containing all the fractional Brownian motions. This distance is constructed from two estimations of multiscale generalized quadratic variations expectations. The second one requires regression estimates of the self-similarity index H. Two estimations of H are then introduced. They present good robustness and computing time properties compared with the Whittle approach, with nearly similar convergence rate. The test is applied on simulated and real data. The self-similarity assumption is notably accepted for the famous Nile River data. [source]


    NMR diffusion measurements under chemical exchange between sites involving a large chemical shift difference

    Abstract This study concerns the thallium-205 cation in aqueous solution in the presence of a calixarene molecule. Although the measurement of the self-diffusion coefficient of pure thallium (without calixarene in the aqueous solution) does not pose any particular problem, major difficulties are encountered with the standard method using gradient strength increment as soon as thallium is partly complexed by calixarene. With static magnetic field gradients, the NMR signal is so weak that it prevents any reliable measurement, whereas radio frequency (rf) field gradients lead to an unrealistic value of the diffusion coefficient. This failure is explained by the fact that thallium is in fast exchange between two sites (complexed and free thallium) thus exhibiting a single NMR signal although, in the course of the experiment, two signals, with an important difference in resonance frequencies (due to the large thallium chemical shift range), are effectively involved. With the objective to understand these quite unexpected observations, the theory underlying NMR diffusion experiments is first reviewed, and criteria of fast exchange are discussed for three parameters: chemical shifts, relaxation rates, and diffusion coefficients. It turns out that off-resonance effects are responsible for unwanted defocusing due to rf pulses in the static magnetic field gradient method and for time-dependent gradients in the rf field gradient method. Concerning the latter, a remedy is proposed which consists in applying the stronger gradient and incrementing the gradient pulse durations.     Aldosterone Receptor Antagonism: Interface With Hyperkalemia in Heart Failure

    Aldosterone receptor antagonism (ARA) is an increasingly well-accepted element of heart failure therapy. The experimental underpinnings for the use of ARA in heart failure are strong being linked to a variety of tissue-based cardiac effects characteristic of drugs in this class. However, the benefits of ARA therapy do not come without some risk since drugs in this class are potent inhibitors of renal potassium (K+) elimination. Thus, some increment in serum K+, up to and including the development of overt hyperkalemia (typically defined as a serum K+ value in excess of 6.0 mEq/L), is to be expected whenever they are used. Hyperkalemia attributable to ARA relates to several factors including ARA dose, patient predisposition to hyperkalemia, as in the case of renal failure, and dietary intake of K+. The risk of some change in serum K+ with ARA should not be a deterrent to use of drugs in this class but, rather should prompt careful surveillance for the onset of this potentially life-threatening electrolyte disturbance. The frequency of such scrutiny should be patient-specific and based on the constellation of risk factors for hyperkalemia. [source]


    Estimation of endogenous adenosine activity at adenosine receptors in guinea-pig ileum using a new pharmacological method

    Abstract Aim:, Adenosine modulates neurotransmission and in the intestine adenosine is continuously released both from nerves and from smooth muscle. The main effect is modulation of contractile activity by inhibition of neurotransmitter release and by direct smooth muscle relaxation. Estimation of adenosine concentration at the receptors is difficult due to metabolic inactivation. We hypothesized that endogenous adenosine concentrations can be calculated by using adenosine receptor antagonist and agonist and dose ratio (DR) equations. Methods:, Plexus-containing guinea-pig ileum longitudinal smooth muscle preparations were made to contract intermittently by electrical field stimulation in organ baths. Schild plot regressions were constructed with 2-chloroadenosine (agonist) and 8-(p -sulfophenyl)theophylline (8-PST; antagonist). In separate experiments the reversing or enhancing effect of 8-PST and the inhibiting effect of 2-chloroadenosine (CADO) were analysed in the absence or presence of an adenosine uptake inhibitor (dilazep), and nucleoside overflow was measured by HPLC. Results:, Using the obtained DR, baseline adenosine concentration was calculated to 28 nm expressed as CADO activity, which increased dose dependently after addition of 10,6 m dilazep to 150 nm (P < 0.05). HPLC measurements yielded a lower fractional increment (80%) in adenosine during dilazep, than found in the pharmacological determination (440%). Conclusion:, Endogenous adenosine is an important modulator of intestinal neuro-effector activity, operating in the linear part of the dose,response curve. Other adenosine-like agonists might contribute to neuromodulation and the derived formulas can be used to calculate endogenous agonist activity, which is markedly affected by nucleoside uptake inhibition. The method described should be suitable for other endogenous signalling molecules in many biological systems. [source]


    Effect of NaCl filler on ferroelectric phase and polaron configurations of PVDF films

    Abstract Polyvinyldene fluoride (PVDF) films filled with NaCl of mass fraction range 1 , W , 6 % were prepared by casting technique. Their crystalline structure, thermal, optical properties and Electron spin resonance (ESR) were examined. X-ray diffraction (XRD) and differential thermal analysis (DTA) measurements indicated a maximum ferroelectric ,-phase increment at 4%. DTA was used to identify the phase transition temperatures, the order of reaction and the activation energy of melting. The UV-Visible optical absorption implied a minimum value of the estimated optical energy gap at W = 4%. ESR spectra contained a Lorentizian signal exhibiting a minimum value of the symmetry factor at W = 4%. The energy levels of the optical gap boundaries were though to contribute to ESR transitions.     Optimal Design of the Online Auction Channel: Analytical, Empirical, and Computational Insights,

    ABSTRACT The focus of this study is on business-to-consumer (B2C) online auctions made possible by the advent of electronic commerce over an open-source, ubiquitous Internet Protocol (IP) computer network. This work presents an analytical model that characterizes the revenue generation process for a popular B2C online auction, namely, Yankee auctions. Such auctions sell multiple identical units of a good to multiple buyers using an ascending and open auction mechanism. The methodologies used to validate the analytical model range from empirical analysis to simulation. A key contribution of this study is the design of a partitioning scheme of the discrete valuation space of the bidders such that equilibrium points with higher revenue structures become identifiable and feasible. Our analysis indicates that the auctioneers are, most of the time, far away from the optimal choice of key control factors such as the bid increment, resulting in substantial losses in a market with already tight margins. With this in mind, we put forward a portfolio of tools, varying in their level of abstraction and information intensity requirements, which help auctioneers maximize their revenues. [source]


    Homozygous thermolabile variant of the methylenetetrahy-drofolate reductase gene: a potential risk factor for hyperhomo-cysteinaemia, CVD, and stroke in childhood

    In this study of 118 children (median age 5.1 years; range 6 months to 17 years) with ischaemic stroke or transient ischaemic attack (TIA), 22 children (19%) were homozygous for the thermolabile variant of the methylenetetrahydrofolate reductase allele (t-MTHFR), compared with nine of 78 (12%) of a reference population (p=0.18, OR 1.76, 95% CI 0.76 to 4.04). Of those with cerebrovascular disease (CVD), 17 of 84 were homozygous for the t-MTHFR allele (p=0.13 compared with the reference population (OR 1.95, 95% CI 0.81 to 4.65). There was a significant (p<0.025) increment of plasma total homocysteine concentration in homozygotes for the t-MTHFR allele compared with heterozygotes, negatives for the t-MTHFR allele, and control children with no history of stroke. In four of 12 homozygotes for the t-MTHFR allele, plasma homocysteine levels were raised, compared with three of 38 of those who were negative or heterozygous (p=0.047; OR 5.8, 95% CI 1.1 to 31.2). Homozygotes for the t-MTHFR allele were significantly more likely to have a recurrent event than those who were negative or heterozygous (Cox regression p=0.031, hazard ratio 2.18, 95% CI 1.08 to 4.42). These data suggest that homozygosity for the t-MTHFR allele is associated with raised homocysteine levels in children and is a risk factor for primary and secondary stroke and TIA. [source]


    Twice daily biphasic insulin aspart improves postprandial glycaemic control more effectively than twice daily NPH insulin, with low risk of hypoglycaemia, in patients with type 2 diabetes

    Objective:, Biphasic insulin aspart 30 (BIAsp30) is a dual release formulation, containing 30% soluble and 70% protamine-crystallized insulin aspart. This study compared the glycaemic control and safety profiles achieved with either twice daily BIAsp30 or NPH insulin in patients with type 2 diabetes not optimally controlled by oral hypoglycaemic agents (OHAs), NPH insulin or a combination of both. Methods:, In this 16-week multinational, parallel-group, double-blind trial, 403 such patients were randomized to receive either BIAsp30 or NPH insulin immediately before breakfast and evening meals. OHAs were discontinued at randomization. Efficacy was assessed by glycosylated haemoglobin (HbA1c) and self-recorded daily 8-point blood glucose (BG) profiles. Hypoglycaemic and other adverse events were the chosen safety parameters. Results:, HbA1c concentration decreased by >0.6% (p < 0.0001 vs. baseline) in both groups, with metabolic control continuing to improve throughout the trial without reaching a stable level. Patients who switched from once or twice daily NPH monotherapy to twice daily BIAsp30 achieved a significantly greater reduction in HbA1c (0.78%) than those randomized to twice daily NPH insulin (0.58%; p = 0.03). BIAsp30 decreased mean daily postprandial glycaemic exposure to a greater extent than NPH insulin (mean difference = 0.69 mmol/l; p < 0.0001), reflecting greater decreases in the postbreakfast and postdinner increments (of 1.26 and 1.33 mmol/l, respectively), although postlunch increment was relatively increased (by 0.56 mmol/l). Despite the greater reduction in overall postprandial glycaemic exposure in the BIAsp30 group, the overall safety profile of BIAsp30 was equivalent to that of NPH insulin with <2% of patients experiencing major hypoglycaemia, and approximately 33% reporting minor hypoglycaemic episodes, in both groups. Conclusion:, Twice daily BIAsp30 reduced postprandial glucose exposure to a significantly greater extent than NPH insulin and was at least as effective at reducing HbA1c in patients with type 2 diabetes. Both insulins were well tolerated. In patients poorly controlled on OHAs or NPH alone, glycaemic control can be improved by switching to twice daily BIAsp30, without increasing hypoglycaemic risk.     Impact of Scribes on Performance Indicators in the Emergency Department

    Abstract Objectives:, The objective was to quantify the effect of scribes on three measures of emergency physician (EP) productivity in an adult emergency department (ED). Methods:, For this retrospective study, 243 clinical shifts (of either 10 or 12 hours) worked by 13 EPs during an 18-month period were selected for evaluation. Payroll data sheets were examined to determine whether these shifts were covered, uncovered, or partially covered (for less than 4 hours) by a scribe; partially covered shifts were grouped with uncovered shifts for analysis. Covered shifts were compared to uncovered shifts in a clustered design, by physician. Hierarchical linear models were used to study the association between percentage of patients with which a scribe was used during a shift and EP productivity as measured by patients per hour, relative value units (RVUs) per hour, and turnaround time (TAT) to discharge. Results:, RVUs per hour increased by 0.24 units (95% confidence interval [CI] = 0.10 to 0.38, p = 0.0011) for every 10% increment in scribe usage during a shift. The number of patients per hour increased by 0.08 (95% CI = 0.04 to 0.12, p = 0.0024) for every 10% increment of scribe usage during a shift. TAT was not significantly associated with scribe use. These associations did not lose significance after accounting for physician assistant (PA) use. Conclusions:, In this retrospective study, EP use of a scribe was associated with improved overall productivity as measured by patients treated per hour (Pt/hr) and RVU generated per hour by EPs, but not as measured by TAT to discharge.     Color M-Mode Flow Propagation Velocity: Is It Really Preload Independent?

    Objective: This study investigates the change in flow propagation velocity (FPV) in uremic patients who undergo regular hemodialysis (H/D). Materials and Methods: We studied 93 uremic patients (44 men and 49 women; 59 ± 14-years-old) receiving regular hemodialysis. Patients were separated by baseline left ventricular ejection fraction (LVEF): 71 patients with LVEF > 50% (group 1), 13 patients with LVEF 35,50% (group 2), 7 patients with LVEF < 35% (group 3). All patients were in sinus rhythm before H/D. They received complete transthoracic echocardiographic examinations. Flow propagation velocity was measured by color M-mode echocardiography in apical four chambers view. All these parameters were obtained before and after H/D. Paired data were compared. According to different H/D amounts, we viewed the FPV response after H/D in variant baseline LVEF groups. Result: The baseline FPV became lower in patients with low LVEF. After H/D, obvious decrement of FPV occurred in group 1, but there were no obvious changes in groups 2 and 3. In fact, a slight increment of FPV was found in group 3. In patients with baseline LVEF > 50%, FPV after H/D was almost always lower, regardless of H/D amount. But there was different response in patients with baseline LVEF < 50%. Conclusion: Flow propagation velocity is preload independent in patients with LVEF < 50%, but it is preload-dependent in patients with LVEF > 50%. [source]


    Transient response of closed-loop MHD experimental facility

    Abstract Transient responses of a closed-loop MHD experimental facility from nonpower generation to power generation have been investigated by means of time-dependent quasi-one-dimensional numerical simulations. For the long-time continuous power generation experiment, the time required to obtain the steady state for the power generation is estimated to be approximately 20 hours. By increasing the electrical input power to the heater as an exponential function of time, the temperature increment of ceramics can be moderated. When the duration of the experiment is around 10 minutes, argon gas temperature at the exit of the heater hardly changes because of the large heat capacity of structure materials. It is found that the fluid disturbances are induced at the instant of the power generation and they propagate as they repeatedly reflect at the sudden change of duct shape. Since all of the induced disturbances attenuate approximately 0.4 second after the power generation, the time scale that the disturbances exist in the facility is estimated to be 1 second at most. © 2006 Wiley Periodicals, Inc.     Microstructure of microemulsion in MEEKC

    Abstract The influences of the composition of microemulsion on the microstructure including dimensions and , potentials of microdroplets were measured in details. The average dynamic dimension of microdroplets was measured by dynamic laser light scattering, and , potential was determined to characterize average surface charge density of microdroplets. The experiment results showed that increase of the amount of surfactant resulted in decrease of microdroplet size but almost invariant , potential, which would enlarge migration time of the microdroplet in MEEKC. With increment of cosurfactant concentration, the microdroplet size had an increasing trend, whereas the , potential decreased. Thus, observed migration velocity of microdroplets increased, which made the separation window in MEEKC shortened. Neither dimension nor , potential of microdroplets changed by varying both the type and the amount of the oil phase. Adding organic solvent as modifier to microemulsion did not change the microdroplet size, but lowered , potential. The migration time of microdroplet still became larger, since EOF slowed down owing to organic solvent in capillary. So, besides increment of surfactant concentration, organic additive could also enlarge the separation window. Increase of cosurfactant concentration was beneficial for separation efficiency thanks to the looser structure of swollen microdroplet, and the peak sharpening might compensate for the resolution and peak capacity owing to a narrow separation window. Except the oil phase, tuning the composition of microemulsion would change the microstructure, eventually could be exploited to optimize the resolution and save analysis time in MEEKC. [source]


    Chiral ion-exchange capillary electrochromatography of arylglycine amides with dextran sulfate as a pseudostationary phase

    Abstract A low-cost tunable chiral ion-exchange capillary electrochromatographic method has been developed for the separation of arylglycine amide racemic mixtures with dextran sulfate (DS) as an anionic and chiral pseudostationary phase and Tris-tartrate as a buffer system. The concentrations of DS and Tris had opposite influences on retention and resolution and could serve as ideal factors to finely tune the running speed and chiral resolution. Tartrate and pH largely impact the separation but pH should be confined within 3.0,5.5, only suitable for coarse tuning, while tartrate was preserved as the key buffering reagent, normally maintained at 40 mmol/L. With a working system composed of 0.1,1.0% DS, 20,60 mmol/L Tris, and 40 mmol/L tartrate at pH 3.50,4.50, the enantioresolution of arylglycine amides was shown to be dependent on their chemical structure: The chiral resolution increased when the hydrogen at the ,-amino group or at the p -position of phenyl ring was replaced by other larger group(s) but the resolution decreased when the group at the o- or m- site on the phenyl ring was enlarged. Further, the electronegative substitute of -Cl had larger resolution increment than methyl or methoxy at the position p- of phenyl ring but much lower increment at position m- . It is possible to well explain the resolution variation phenomenon by considering the group resistance and the variation of hydrogen-bonds formed inside the amino amides and between the solutes and DS. The amido group was shown irreplaceable to have chiral resolution with DS alone as an ionic and chiral pseudostationary phase. [source]

Objectives: To examine the effects of NO synthase inhibition on O2 extraction and anaerobic metabolism at rest, and during submaximal and maximal exertion. Methods: Placebo and NO synthase inhibition (with N,-nitro-L-arginine methyl ester [l -NAME] administered at 20 mg/kg bwt i.v.) studies were performed in random order, 7 days apart on 7 healthy, exercise-trained Thoroughbred horses at rest and during incremental exercise leading to 120 sec of maximal exertion at 14 m/sec on a 3.5% uphill grade. Results: At rest, NO synthase inhibition significantly augmented the arterial to mixed-venous blood O2 content gradient and O2 extraction as mixed-venous blood O2 tension and saturation decreased significantly. While NO synthase inhibition did not affect arterial blood-gas tensions in exercising horses, the exercise-induced increment in haemoglobin concentration and arterial O2 content was attenuated. In the l -NAME study, during submaximal exercise, mixed-venous blood O2 tension and haemoglobin-O2 saturation decreased to a greater extent causing O2 extraction to increase significantly. During maximal exertion, arterial hypoxaemia, desaturation of haemoglobin and hypercapnia of a similar magnitude developed in both treatments. Also, the changes in mixed-venous blood O2 tension and haemoglobin-O2 saturation, arterial to mixed-venous blood O2 content gradient, O2 extraction and markers of anaerobic metabolism (lactate and ammonia production, and metabolic acidosis) were not different from those in the placebo study. Conclusion: Endogenous NO production augments O2 extraction at rest and during submaximal exertion, but not the during short-term maximal exercise. Also, NO synthase inhibition does not affect anaerobic metabolism at rest or during exertion. Potential relevance: It is unlikely that endogenous NO release modifies aerobic or anaerobic metabolism in horses performing short-term maximal exertion. [source]


    Irradiated cultured apoptotic peripheral blood mononuclear cells regenerate infarcted myocardium

    Abstract Background, Acute myocardial infarction (AMI) is followed by post AMI cardiac remodelling, often leading to congestive heart failure. Homing of c-kit+ endothelial progenitor cells (EPC) has been thought to be the optimal source for regenerating infarcted myocardium. Methods, Immune function of viable peripheral blood mononuclear cells (PBMC) was evaluated after co-culture with irradiated apoptotic PBMC (IA-PBMC) in vitro. Viable PBMC, IA-PBMC and culture supernatants (SN) thereof were obtained after 24 h. Reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay were utilized to quantify interleukin-8 (IL-8), vascular endothelial growth factor, matrix metalloproteinase-9 (MMP9) in PBMC, SN and SN exposed fibroblasts. Cell suspensions of viable- and IA-PBMC were infused in an experimental rat AMI model. Immunohistological analysis was performed to detect inflammatory and pro-angiogenic cells within 72 h post-infarction. Functional data and determination of infarction size were quantified by echocardiography and Elastica van Gieson staining. Results, The IA-PBMC attenuated immune reactivity and resulted in secretion of pro-angiogenic IL-8 and MMP9 in vitro. Fibroblasts exposed to viable and IA-PBMC derived SN caused RNA increment of IL-8 and MMP9. AMI rats that were infused with IA-PBMC cell suspension evidenced enhanced homing of endothelial progenitor cells within 72 h as compared to control (medium alone, viable-PBMC). Echocardiography showed a significant reduction in infarction size and improvement in post AMI remodelling as evidenced by an attenuated loss of ejection fraction. Conclusion, These data indicate that infusion of IA-PBMC cell suspension in experimental AMI circumvented inflammation, caused preferential homing of regenerative EPC and replaced infarcted myocardium.     Effect of daytime, weekday and year of admission on outcome in acute ischaemic stroke patients treated with thrombolytic therapy

    Background:, Since doubts were raised, if a challenging medical procedure such as acute stroke treatment including thrombolysis with recombinant tissue plasminogen activator (rTPA) is available with identical standard and outcome 24 h and 7 days a week our aim was to examine if acute stroke patients defined by onset-admission time (OAT) of , 3 h were treated differently or had distinct outcome when admitted during off duty hours (day versus night and weekend versus weekdays) and if any differences in treatment or outcome were apparent when comparing patients admitted in the year 2003 with patients admitted in the year 2006. Methods:, We analyzed 2003,2006 data of a prospective registry and grouped patients by time, day, and year of admission. The evaluation was limited to patients that were diagnosed with ischaemic stroke and with OAT of , 3 h. Medical and sociodemographic items, use of thrombolytic treatment, complications during clinical course and place of discharge were obtained. Clinical state on admission and discharge was assessed using the modified Rankin scale. Comparison with chi-square test, t -test and logistic regression was performed. Results:, Patient's characteristics, rate of thrombolysis, and outcome were independent from time or day of admission. Proportion of patients with good clinical state at discharge increased significantly from 2003 to 2006 together with a higher rate of rTPA treatment without increase of intracranial hemorrhage. Proportion of patients discharged in good clinical condition after rTPA treatment increased from 34% to 44%. Conclusions:, Stroke treatment in potential candidates for thrombolytic therapy revealed no impairment on weekend or at night already in 2003. During 4 years, it was possible to increase rate of rTPA treatment from 8.9% to 21.8% without increment of complications or death, confirming that rTPA is safe and can be implemented with full daily and weekly coverage.     Intrauterine proximity to male fetuses affects the morphology of the sexually dimorphic nucleus of the preoptic area in the adult rat brain

    Abstract Previous studies on polytocous rodents have revealed that the fetal intrauterine position influences its later anatomy, physiology, reproductive performance and behavior. To investigate whether the position of a fetus in the uterus modifies the development of the brain, we examined whether the structure of the sexually dimorphic nucleus of the preoptic area (SDN-POA) of rat brains accorded to their intrauterine positions. Brain sections of adult rats gestated between two male fetuses (2M) and between two female fetuses (2F) in the uterus were analysed for their immunoreactivity to calbindin-D28k, which is a marker of the SDN-POA. The SDN-POA volume of the 2M adult males was greater than that of the 2F adult males, whereas the SDN-POA volume of the 2M and 2F adult females showed no significant difference. This result indicated that contiguous male fetuses have a masculinizing effect on the SDN-POA volume of the male. To further examine whether the increment of SDN-POA volume in adulthood was due to exposure to elevated steroid hormones during fetal life, concentrations of testosterone and 17,-estradiol in the brain were measured with 2M and 2F fetuses during gestation, respectively. On gestation day 21, the concentrations of testosterone and 17,-estradiol in the brain were significantly higher in the 2M male rats as compared with the 2F male rats. The results suggested that there was a relationship between the fetal intrauterine position, hormone transfer from adjacent fetuses and the SDN-POA volume in adult rat brains. [source]


    Long-term potentiation of mGluR1 activity by depolarization-induced Homer1a in mouse cerebellar Purkinje neurons

    Abstract Metabotropic glutamate receptor 1 (mGluR1) plays a crucial role in synaptic plasticity and motor learning in the cerebellum. We have studied activity-dependent changes in mGluR1 function in mouse cultured Purkinje neurons. Depolarizing stimulation potentiated Ca2+ and current responses to an mGluR1 agonist for several hours in the cultured Purkinje neurons. It also blocked internalization of mGluR1 and increased the number of mGluR1s on the cell membrane. We found that depolarization simultaneously increased transcription of Homer1a in Purkinje neurons. Homer1a inhibited internalization and increased cell-surface expression of mGluR1 when coexpressed in human embryonic kidney (HEK)-293 cells. Depolarization-induced Homer1a expression in Purkinje neurons was blocked by a mitogen-activated protein kinase (MAPK) inhibitor. Changes in internalization and mGluR1-mediated Ca2+ response were also blocked by inhibition of MAPK activity, suggesting that localization and activity of mGluR1 were regulated in the same signalling pathway as Homer1a expression. It is thus suggested that depolarization of the Purkinje neuron leads to the increment in mGluR1 responsiveness through MAPK activity and induction of Homer1a expression, which increases active mGluR1 on the cell surface by blocking internalization of mGluR1.     Predicting academic success with the Big 5 rated from different points of view: Self-rated, other rated and faked

    Abstract Self-ratings of personality predict academic success above general intelligence. The present study replicated these findings and investigated the increment of other-ratings or intentionally distorted self-ratings. Participants (N,=,145) had to compile a personality questionnaire twice. First they were given neutral instructions. The second time they were asked to imagine a specific applicant setting. Furthermore, two peers rated each participant. Additionally, verbal, numerical and figural reasoning scores were obtained. Grades on a statistics exam obtained 2 months later served as the criterion. Results replicated prior findings and showed incremental validity for self- and other-rated personality, which was stable after controlling for intelligence.     Falsification cueing in collective reasoning: example of the Wason selection task

    This paper focuses on the effectiveness of groups, as opposed to individuals, in benefiting from falsification cueing in solving the Wason selection task. Consistent with the idea that groups use information that often individuals fail to use, Experiment 1 showed that groups (but not individuals) that received falsification cueing focused more on cue-consistent evidence in their reasoning. Experiments 2 and 3 showed that the increment in focus on cue-consistent evidence is moderated by the distribution of the falsification cue within a group. Finally, Experiment 3 demonstrated that the cue distribution affects collective focus on cue-consistent evidence through the content of the group discussion, namely through mentioning the cue during the discussion.     CYTO-NUCLEAR EPISTASIS: TWO-LOCUS RANDOM GENETIC DRIFT IN HERMAPHRODITIC AND DIOECIOUS SPECIES

    Abstract We report the findings of our theoretical investigation of the effect of random genetic drift on the covariance of identity-by-descent (ibd) of nuclear and cytoplasmic genes. The covariance in ibd measures of the degree to which cyto-nuclear gene combinations are heritable, that is, transmitted together from parents to offspring. We show how the mating system affects the covariance of ibd, a potentially important aspect of host-pathogen or host-symbiont coevolution. The magnitude of this covariance influences the degree to which the evolution of apparently neutral cytoplasmic genes, often used in molecular phylogenetics, might be influenced by selection acting on unlinked nuclear genes. To the extent that cyto-nuclear gene combinations are inherited together, genomic conflict is mitigated and intergenomic transfer it facilitated, because genes in both organelle and nuclear genomes share the same evolutionary fate. The covariance of ibd also affects the rate at which cyto-nuclear epistatic variance is converted to additive variance necessary for a response to selection. We find that conversion is biased in species with separate sexes, so that the increment of additive variance added to the nuclear genome exceeds that added to the cytoplasmic genome. As a result, the host might have an adaptive advantage in a coevolutionary arms race with vertically (maternally) transmitted pathogens. Similarly, the nuclear genome could be a source of compensatory mutations for its organellar genomes, as occurs in cytoplasmic male sterility in some plant species. We also discuss the possibility that adaptive cytoplasmic elements, such as favorable mitochondrial mutations or endosymbionts (e.g., Wolbachia), have the potential to release heritable nuclear variation as they sweep through a host population, supporting the view that cytoplasmic introgression plays an important role in adaptation and speciation.     Stimulation of intramembranous bone repair in rats by ghrelin

    Researchers in our laboratory have previously shown that ghrelin, a gastric peptide hormone, may regulate mesenchymal cell differentiation into adipocytes and myocytes. Here we show that ghrelin promotes osteogenesis of intramembranous bone and improves the repair of calvarial bone defects in rats. Rats with a 9 mm full-thickness calvarial bone defect received either Bio-Oss® (control group) or Bio-Oss® mixed with 20 ,g ghrelin (treatment group), followed by local administration of saline or ghrelin (10 ,g), respectively, on days 5, 10 and 15. After 6 and 12 weeks, new bone formation was assessed. Animals treated with ghrelin showed a significant increase in new bone formation as demonstrated by an increment in bone mineral density and fluorescence labelling of tetracycline relative to the control group. At 6 weeks, bone mineral density increased from 54 ± 7 (control group) to 78 ± 9 mg cm,2 in the treatment group, while the tetracycline fluorescence labelling increased by 61 ± 15%. A similar increment was observed at 12 weeks. Quantitative reverse transcriptase-polymerase chain reaction showed that expression of alkaline phosphatase (ALP), osteocalcin and collagen type I was elevated. Relative to the control animals, mRNAs for ALP, osteocalcin and collagen type I increased 2.4 ± 0.4-, 4.7 ± 1.9- and 4.0 ± 1.7-fold, respectively, in animals treated with ghrelin for 6 weeks (P < 0.05). At 12 weeks, mRNA levels of ALP, osteocalcin and collagen type I showed a decline relative to levels at 6 weeks but still remained significantly higher than in the control group, with fold changes of 2.4 ± 0.8, 2.4 ± 1.2 and 2.1 ± 0.7, respectively (P < 0.05). This study demonstrated that ghrelin stimulates intramembranous osteogenesis. [source]


    Experimental and numerical studies on dynamic crack growth in layered slate rock under wedge impact loads: part II , non-plane strain problem

    ABSTRACT Dynamic crack propagation in non-plane strain (or 3D) slate blocks under wedge impact loads was investigated numerically in this part of the paper. A parabolic-shaped crack trajectory was taken into consideration to model the crack propagation in slate blocks for analyzing the impact splitting of layered slate rock. Major and minor axes of the parabola were determined from the condition of equal mode I stress intensity factors (SIFs) along the crack front. Mode I SIFs were determined for experimental breaking loads for each increment of crack growth in a manner similar to that mentioned in part I of this paper. These values were compared with the plane strain material fracture toughness value obtained from experimental studies and very good agreement was obtained between them, for the case of actual load applied on the specimen. Numerical analysis of a field problem, i.e., separation of a large-sized slate slab from the rock strata in a slate quarry using wedge impacting, was also carried out in this paper. It can be observed that a large magnitude of load is required to break large-sized slate blocks; but this load is applied through a number of smaller load-capacity actuators-in-parallel, requiring large power capacity for the hydraulic pumps. However, this required power could be reduced considerably if the load applied on the line of hydraulic actuators is cascaded across the (line of) actuators (starting from centrally placed actuators) with a small time delay (equal to the initial crushing time in slate rock).     Study of crack growth in solid propellants

    The stress and displacement fields in an edge-cracked sheet specimen made of a solid propellant and subjected to a uniform displacement along its upper and lower faces was studied. The solid propellant was simulated as a hyperelastic material with constitutive behaviour described by the Ogden strain energy potential. A non-linear finite deformation analysis was performed based on the finite element code ABAQUS. A detailed analysis of the stress field in the vicinity of the crack tip was undertaken. The deformed profiles of the crack faces near the crack tip were determined. The results of stress analysis were coupled with the strain energy density theory to predict the crack growth behaviour including crack initiation, stable crack growth and final termination for two specimens with different dimensions. Crack growth resistance curves representing the variation of crack growth increment versus applied displacement were drawn. [source]


    [Na+]i -induced c-Fos expression is not mediated by activation of the 5,-promoter containing known transcriptional elements

    In vascular smooth muscle cells and several other cell types, inhibition of Na+/K+ -ATPase leads to the expression of early response genes, including c-Fos. We designed this study to examine whether or not a putative Na+i/K+i -sensitive element is located within the c-Fos 5,-UTR from ,,650 to +,103 containing all known response elements activated by ,classic' stimuli, such as growth factors and Ca2+i -raising compounds. In HeLa cells, the highest increment of c-Fos mRNA content was noted after 6 h of Na+/K+ -ATPase inhibition with ouabain that was abolished by actinomycin D, an inhibitor of RNA synthesis. c-Fos protein accumulation in ouabain-treated cells correlated with a gain of Na+i and loss of K+i. Augmented c-Fos expression was also observed under inhibition of Na+/K+ -ATPase in K+ -free medium and in the presence of the Na+ ionophore monensin. The effect of ouabain on c-Fos expression was sharply attenuated under dissipation of the transmembrane Na+ gradient, but was preserved in the presence of Ca2+ chelators and the extracellular regulated kinase inhibitor PD98059, thus indicating an Na+i -mediated, Ca2+i - and extracellular regulated kinase-independent mechanism of gene expression. In contrast to massive c-Fos expression, we failed to detect any effect of ouabain on accumulation of luciferase driven by the c-Fos 5,-UTR. Negative results were also obtained in ouabain-treated vascular smooth muscle cells and C11 Madin,Darby canine kidney cells possessing augmented c-Fos expression. Our results reveal that Na+i -induced c-Fos expression is not mediated by the 5,-UTR containing transcriptional elements activated by growth factors and other ,classic stimuli'. [source]
