    Increased growth and recruitment of piscivorous perch, Perca fluviatilis, during a transient phase of expanding submerged vegetation in a shallow lake

    Summary 1. In this study, we examine how a 7-year period of expanding submerged stonewort (Chara spp.) vegetation during a shift from turbid to clear water in a shallow lake influenced individual growth and population size structure of perch (Perca fluviatilis). We expected that a shift from phytoplankton to macrophyte dominance and clear water would improve feeding conditions for perch during a critical benthivorous ontogenetic stage, and enhance the recruitment of piscivorous perch. 2. Growth analysis based on opercula showed that growth during the second year of life was significantly higher in years with abundant vegetation than in years with turbid water and sparse vegetation. Growth was not affected during the first, third and fourth year of life. Stable isotope analyses on opercula from 2-year-old perch showed that the increase in growth coincided with a change in carbon source in the diet. Stable nitrogen ratio did not change, indicating that the increased growth was not an effect of any change in trophic position. 3. Following the expansion of submerged vegetation, perch size range and abundance of piscivorous perch increased in central, unvegetated areas of the lake. In stands of stoneworts, however, mainly benthivorous perch were caught, and size range did not change with time. 4. Our findings provide empirical support for the notion that establishment of submerged vegetation may lead to increased recruitment of piscivorous perch, because of improved competitive conditions for perch during the benthivorous stage. This is likely to constitute a benthic-pelagic feedback coupling, in which submerged vegetation and clear water promote the recruitment of piscivorous perch, which, in turn, may increase water clarity through top-down effects in the pelagic.     Is treatment with growth hormone effective in children with cerebral palsy?

    Children with cerebral palsy (CP) often have poor linear growth during childhood, resulting in a diminished final adult height. Here we report a female with CP and short stature but without growth hormone (GH) deficiency who exhibited increased growth during treatment with GH. We also report two other children with CP who were treated with GH: one female with a history of leukemia, and a male with Klinefelter syndrome. These two children were both found to be GH-deficient by insulin provocative GH testing and responded to treatment with increased growth rate. Growth improved to a greater extent in the two children with apparent GH deficiency. In summary, it is felt that GH therapy might be beneficial for children with CP and warrants further investigation. [source]


    Eradication of the invasive Pseudorasbora parva results in increased growth and production of native fishes

    Abstract,,, The topmouth gudgeon, Pseudorasbora parva, has been described as Europe's most invasive fish. To control their UK invasion, some lentic populations at risk of causing fluvial dispersal have been eradicated. The first of these operations was from a lake in north-west UK in March 2005 using rotenone application; prior to eradication, their mean density was estimated as 6.1 m,2 whereas since eradication, no P. parva have been recorded. Prior to rotenone application, the majority of native fishes were removed, held off-site and reintroduced following degradation of rotenone to safe levels. In the three growth seasons since their reintroduction and P. parva eradication, the abundance, somatic growth rate and production of roach Rutilus rutilus and common bream Abramis brama have increased significantly; production is now driven by a lower number of comparatively larger, faster growing individuals. These data suggest that the eradication of this P. parva population has been highly beneficial for the growth, recruitment and production of these native species. [source]


    Bryozoan populations reflect nutrient enrichment and productivity gradients in rivers

    Summary 1. The hypothesis that nutrient enrichment will affect bryozoan abundance was tested using two complementary investigations; a field-based method determining bryozoan abundance in 20 rivers of different nutrient concentrations by deploying statoblast (dormant propagule) traps and an experimental laboratory microcosm study measuring bryozoan growth and mortality. These two methods confirmed independently that increased nutrient concentrations in water promote increases in the biomass of freshwater bryozoans. 2. Statoblasts of the genus Plumatella were recorded in all rivers, regardless of nutrient concentrations, demonstrating that freshwater bryozoans are widespread. Concentrations of Plumatella statoblasts were high in rivers with high nutrient concentrations relative to those with low to moderate nutrient concentrations. Regression analyses indicated that phosphorus concentrations, in particular, significantly influenced statoblast concentrations. 3. Concentrations of Lophopus crystallinus statoblasts were also higher in sites characterised by high nutrient concentrations. Logistic regression analysis revealed that the presence of L. crystallinus statoblasts was significantly associated with decreasing altitude and increasing phosphorus concentrations. This apparently rare species was found in nine rivers (out of 20), seven of which were new sites for L. crystallinus. 4. Growth rates of Fredericella sultana in laboratory microcosms increased with increasing nutrient concentration and high mortality rates were associated with low nutrient concentrations. 5. Our results indicate that bryozoans respond to increasing nutrient concentrations by increased growth, resulting in higher biomasses in enriched waters. We also found that an important component of bryozoan diets can derive from food items lacking chlorophyll a. Finally, bryozoans may be used as independent proxies for inferring trophic conditions, a feature that may be especially valuable in reconstructing historical environments by assessing the abundance of statoblasts in sediment cores.     Consequences of maternal yolk testosterone for offspring development and survival: experimental test in a lizard

    Summary 1Hormone-mediated maternal effects and developmental plasticity are important sources of phenotypic variation, with potential consequences for trait evolution. Yet our understanding of the importance of maternal hormones for offspring fitness in natural populations is very limited, particularly in non-avian species. 2We experimentally elevated yolk testosterone by injection of a physiological dose into eggs of the lizard Ctenophorus fordi Storr, to investigate its roles in offspring development, growth and survival. 3Yolk testosterone did not influence incubation period, basic hatchling morphology or survival under natural conditions. However, there was evidence for increased growth in hatchlings from testosterone-treated eggs, suggesting that maternal hormones have potential fitness consequences in natural populations. 4The positive effect of prenatal testosterone exposure on postnatal growth could represent a taxonomically widespread developmental mechanism that has evolved into an adaptive maternal effect in some taxa, but remains deleterious or selectively neutral in others. 5A broader taxonomic perspective should increase our understanding of the role of physiological constraints in the evolution of endocrine maternal effects.     Effects of food limitation and emigration on self-thinning in experimental minnow cohorts

    Summary 1.,The theory of food-regulated self-thinning (FST) for mobile animals predicts population density (N) to be an inverse function of mean body mass (W) scaled to an exponent (b), such that N = k W,b, where k is a constant. FST also predicts energy requirements (or energy flow) to remain constant over time (termed energetic equivalence) as losses to cohorts (e.g. emigration and mortality) are balanced by increased growth of surviving individuals. 2.,To test these predictions, we analysed the dynamics of six experimental minnow cohorts. Replicate populations of fish were held under identical conditions with a constant and limited supply of food over a 126-day period. Half of the cohorts were open to emigration, and half were closed so that fish could only be lost through starvation mortality. 3.,Patterns of self-thinning indicated non-linear changes in population density and energy flow in relation to changes in mean body mass and time, respectively. Non-linear patterns of self-thinning were probably due to a delayed growth response to changes in population density effected through mortality and/or emigration. Contrary to results of similar experiments on other fish, emigration did not have a significant influence on the pattern of self-thinning. 4.,These results may be attributed to trophic interactions within cohorts and the importance of social behaviour to cohort dynamics. Both population density and energy flow in our experimental populations appeared to cycle, with episodes of starvation and mortality alternating with food recovery and weight gain, as predicted by recent models of stepwise die-off and stunted growth in animal cohorts. 5.,Most of the support for FST in mobile animals comes from observational data on mean body mass and population density. Potentially important mechanisms, including the manner in which individuals are lost or retained in populations, are usually not investigated directly. Such tests of FST can only provide equivocal support. Detailed observational study and controlled experiments are needed to understand casual mechanisms. [source]


    High aldehyde dehydrogenase and expression of cancer stem cell markers selects for breast cancer cells with enhanced malignant and metastatic ability

    Abstract Cancer stem cells (CSCs) have recently been identified in leukaemia and solid tumours; however, the role of CSCs in metastasis remains poorly understood. This dearth of knowledge about CSCs and metastasis is due largely to technical challenges associated with the use of primary human cancer cells in pre-clinical models of metastasis. Therefore, the objective of this study was to develop suitable pre-clinical model systems for studying stem-like cells in breast cancer metastasis, and to test the hypothesis that stem-like cells play a key role in metastatic behaviour. We assessed four different human breast cancer cell lines (MDA-MB-435, MDA-MB-231, MDA-MB-468, MCF-7) for expression of prospective CSC markers CD44/CD24 and CD133, and for functional activity of aldehyde dehydrogenase (ALDH), an enzyme involved in stem cell self-protection. We then used fluorescence-activated cell sorting and functional assays to characterize differences in malignant/metastatic behaviour in vitro (proliferation, colony-forming ability, adhesion, migration, invasion) and in vivo (tumorigenicity and metastasis). Sub-populations of cells demonstrating stem-cell-like characteristics (high expression of CSC markers and/or high ALDH) were identified in all cell lines except MCF-7. When isolated and compared to ALDHlowCD44low/, cells, ALDHhiCD44+CD24, (MDA-MB-231) and ALDHhiCD44+CD133+ (MDA-MB-468) cells demonstrated increased growth (P < 0.05), colony formation (P < 0.05), adhesion (P < 0.001), migration (P < 0.001) and invasion (P < 0.001). Furthermore, following tail vein or mammary fat pad injection of NOD/SCID/IL2, receptor null mice, ALDHhiCD44+CD24, and ALDHhiCD44+CD133+ cells showed enhanced tumorigenicity and metastasis relative to ALDHlowCD44low/, cells (P < 0.05). These novel results suggest that stem-like ALDHhiCD44+CD24, and ALDHhiCD44+CD133+ cells may be important mediators of breast cancer metastasis. [source]


    Modifications of the fibroblast growth factor-2 gene led to a marked enhancement in secretion and stability of the recombinant fibroblast growth factor-2 protein

    Abstract Progress in FGF-2 gene therapy has been hampered by the difficulty in achieving therapeutic levels of FGF-2 secretion. This study tested whether the addition of BMP2/4 hybrid secretion signal to the FGF-2 gene and mutation of cys-70 and cys-88 to serine and asparagine, respectively, would increase the stability and secretion of active FGF-2 protein in mammalian cells using MLV-based vectors. Single or double mutations of cys-70 and cys-88 to ser-70 and asp-88, respectively, markedly increased the amounts of FGF-2 protein in conditioned media and cell lysates, which may be due to glycosylation, particularly at the mutated asp-88 residue. Addition of BMP2/4 secretion signal increased FGF-2 secretion, but also suppressed FGF-2 biosynthesis. The combination of BMP2/4 secretion signal and double cys-70 and cys-88 mutations increased the total amount of secreted FGF-2 protein >60-fold. The modifications did not alter its ability to stimulate cell proliferation and Erk1/2 phosphorylation in marrow stromal cells or its ability to bind heparin in vitro, suggesting that the modified FGF-2 protein was functionally as effective as the unmodified FGF-2. An ex vivo application of rat skin fibroblasts (RSF) transduced with the modified FGF-2 vector in a subcutaneous implant model showed that rats with implants containing cells transduced with the modified FGF-2 vector increased serum FGF-2 level >15-fold, increased growth of the implant, and increased vascularization within the implant, compared to rats that received implants containing ,-galactosidase- or wild-type FGF-2-transduced control cells.     Natural disturbance and life history: consequences of winterkill on fathead minnow in boreal lakes

    Age, growth and reproductive characteristics of fathead minnow Pimephales promelas populations inhabiting four lakes that varied in the extent and frequency of winterkill were studied in the boreal region of western Canada. The lifespan of fathead minnows inhabiting lakes prone to winterkill was 1,2 years shorter than those in less disturbed lakes. In populations prone to winterkill, fish displayed faster growth rates and grew to a larger size-at-age, particularly during the first year of life. Although lower population densities in winterkill lakes probably contributed to this increased growth, adults in these populations tended to spawn earlier in the season than the smaller adults in more stable populations. Fathead minnows in lakes prone to winterkill also matured at an earlier age and allocated a greater proportion of their body mass to gonads than conspecifics in the more benign, stable lakes. These trends are consistent with predictions for organisms in variable, unpredictable environments and, because fathead minnows are tolerant to a wide range of environmental conditions, suggest that variation in life-history traits among populations is probably a product of both selection and phenotypic plasticity.     Experimental exposure of zebrafish, Danio rerio (Hamilton), to Mycobacterium marinum and Mycobacterium peregrinum reveals the gastrointestinal tract as the primary route of infection: a potential model for environmental mycobacterial infection

    Abstract The natural route by which fish become infected with mycobacteria is unknown. Danio rerio (Hamilton) were exposed by bath immersion and intubation to Mycobacterium marinum and Mycobacterium peregrinum isolates obtained from diseased zebrafish. Exposed fish were collected over the course of 8 weeks and examined for the presence of mycobacteriosis. Mycobacteria were consistently cultured from the intestines, and often from the livers and spleens of fish exposed by both methods. Mycobacteria were not observed in the gills. Histological analysis revealed that fish infected with M. marinum often developed granulomas accompanied by clinical signs of mycobacteriosis, while infection with M. peregrinum infrequently led to clinical signs of disease. Passage of the bacteria through environmental amoebae (Acanthamoeba castellani) was associated with increased growth of M. peregrinum over the course of 8 weeks, when compared to infection with the bacteria not passed through amoebae. The results provide evidence that zebrafish acquire mycobacteria primarily through the intestinal tract, resulting in mycobacterial dissemination. [source]


    Metronidazole Increases Intracolonic but Not Peripheral Blood Acetaldehyde in Chronic Ethanol-Treated Rats

    Background: Metronidazole leads to the overgrowth of aerobic flora in the large intestine by reducing the number of anaerobes. According to our previous studies, this shift may increase intracolonic bacterial acetaldehyde formation if ethanol is present. Metronidazole is also reported to cause disulfiram-like effects after alcohol intake, although the mechanism behind this is obscure. Therefore, the aim was to study the effect of long-term metronidazole and alcohol treatment on intracolonic acetaldehyde levels and to explore the possible role of intestinal bacteria in the metronidazole related disulfiram-like reaction. Methods: A total of 32 rats were divided into four groups: controls (n= 6), controls receiving metronidazole (n= 6), ethanol group (n= 10), and ethanol and metronidazole group (n= 10). All rats were pair-fed with the liquid diet for 6-weeks, whereafter blood and intracolonic acetaldehyde levels and liver and colonic mucosal alcohol (ADH) and aldehyde dehydrogenase (ALDH) activities were analyzed. Results: The rats receiving ethanol and metronidazole had five times higher intracolonic acetaldehyde levels than the rats receiving only ethanol (431.4 ± 163.5 ,M vs. 84.7 ± 14.4 ,M, p= 0.0035). In contrast, blood acetaldehyde levels were equal. Cecal cultures showed the increased growth of Enterobacteriaceae in the metronidazole groups. Metronidazole had no inhibitory effect on hepatic or colonic mucosal ADH and ALDH activities. Conclusions: The increase in intracolonic acetaldehyde after metronidazole treatment is probably due to the replacement of intestinal anaerobes by ADH-containing aerobes. Unlike disulfiram, metronidazole neither inhibits liver ALDH nor increases blood acetaldehyde. Thus, our findings suggested that the mechanism behind metronidazole related disulfiram-like reaction might be located in the gut flora instead of the liver. [source]


    Cognitive styles and hypermedia navigation: Development of a learning model

    There has been an increased growth in the use of hypermedia to deliver learning and teaching material. However, much remains to be learned about how different learners perceive such systems. Therefore, it is essential to build robust learning models to illustrate how hypermedia features are experienced by different learners. Research into individual differences suggests cognitive styles have a significant effect on student learning in hypermedia systems. In particular, Witkin's Field Dependence has been extensively examined in previous studies. This article reviews the published findings from empirical studies of hypermedia learning. Specifically, the review classifies the research into five themes: nonlinear learning, learner control, navigation in hyperspace, matching and mismatching, and learning effectiveness. A learning model, developed from an analysis of findings of the previous studies, is presented. Finally, implications for the design of hypermedia learning systems are discussed. [source]


    Nitrogen fertilization reduces Sphagnum production in bog communities

    The effects of increased nitrogen influx on Sphagnum growth and on interspecific competition between Sphagnum species were studied in a 3-yr experiment in mires situated in two areas with different rates of airborne N deposition. Sphagnum growth was recorded after various supplementary N influxes (0, 1, 3, 5 and 10 g m ,2 yr,1) in hummocks and lawn communities. Sphagnum biomass production decreased with increasing N influx in both areas. After the first season at the low-deposition site, Sphagnum showed an increased growth in length with the intermediate N treatment, but in the second and third seasons the control treatment had the highest growth in length. Capitulum dry mass increased with increasing N influx. Sphagnum N concentration and N/P quotient were higher at the high- than at the low-deposition site. The low quotient at the low-deposition site, together with the initial growth increase with intermediate N supplements, indicates that growth was N-limited at this site, but our lowest N supplement was sufficient to reduce growth. The N treatments had no effect on interspecific competition between the Sphagnum species. This indicates that the species have similar responses to N. The species studied all occur naturally on ombrotrophic, N-poor sites and show low tolerances to increased N influx. Reduced Sphagnum production may affect the carbon balance, changing the mires from C sinks to sources. [source]


    Skeletal effects of bite jumping therapy on the mandible , removable vs. fixed functional appliances

    Structured Abstract Authors , Shen G, Hägg U, Darendeliler MA Objective , Based on an extensive review of the literature, the aim of this study was to explore the mainstream consensus on the controversial topic of whether the bite jumping treatment could enhance mandibular growth. Design , The data for removable and fixed functional appliances were respectively comprehended and analyzed with regard to their attributes in mandibular growth modification. Furthermore, numerous reported findings were assessed by relating them to some important factors influencing the effects of bite jumping, such as treatment timing, treatment duration and post-treatment follow-up, to allow for a more objective and accurate evaluation. Results , The key differences between removable and fixed appliances are working hours (intermittent vs. continuous), length of treatment time (long vs. short), optimal treatment timing (before puberty growth vs. at or after puberty spurt), and mode of bite-jumping (considerable vertical opening vs. limited vertical opening). These different features lead to different treatment effects on mandibular and TMJ growth, such as the intensity of possibly increased growth (clinically less significant vs. significant), the direction of enhanced growth (vertical vs. horizontal), and the stability of treatment changes (unstable vs. stable). The short-term or long-term post-treatment relapse mainly relates to the rebound of dental position. Conclusion , The immediate effects of bite jumping functional appliances on the mandibular growth enhancement are convincing during actual treatment. This extra gain of growth might be sustainable during the short-term and long-term post-treatment period. [source]


    Antiproliferative effect of Scutellaria barbata D. Don. on cultured human uterine leiomyoma cells by down-regulation of the expression of Bcl-2 protein

    Abstract Scutellaria barbata D. Don (Lamiaceae; SB) inhibited the growth of leiomyomal cells (LM). A time-dependent antiproliferative effect was noted when 10,5m buserelin, gonadotrophin-releasing hormone (GnRH) agonist or 20,40 µg/mL SB was added. The inhibition of cell growth decreased with the addition of the PKC activator (12-O-tetradecanoylphorbor-13-acetate; TPA) much as it did with the addition of SB, and the decreases in the viable cells caused by the addition of SB were reversed completely by pretreatment with a protein kinase C (PKC) inhibitor (calphostin C). The findings suggest that SB inhibits cell proliferation in cultured human uterine leiomyoma cells accompanied by PKC activation. Next, the study investigated the effect of SB on fetal development for toxicity. Pregnant Sprague-Dawley rats, from gestation day 6,15, were administered 20 g/L or 50 g/L SB in the drinking water and then killed on day 20. No maternal toxicity was observed, however, embryonic loss in the treatment groups was double that of the controls (p < 0.05). No gross morphologic malformations were seen in the treated fetuses. Fetuses exposed to SB were found to be significantly heavier than the controls, an effect that was greater in female fetuses and was not correlated with increased placental size. The results suggest that the SB had no toxicity and that in utero exposure to SB resulted in increased early embryo loss with increased growth in surviving fetuses. On the other hand, Western blot analyses revealed that Bcl-2 protein of a 26 kDa was abundant in leiomyomal cells, but not in normal myometrial cells. The addition of progesterone (100 ng/mL) resulted in a striking increase in Bcl-2 protein expression in the cultured leiomyoma cells. However, the addition of SB (20 µg/mL) resulted in a significant reduction in Bcl-2 protein expression in the cells. The results indicated that human uterine leiomyomal cells express Bcl-2 protein and progesterone enhances its expression, however, SB reduces the expression of Bcl-2 protein in human uterine leiomyoma cells.     Axin2 expression identifies progenitor cells in the murine prostate

    Abstract BACKGROUND We previously reported that prostatic stem/progenitor cells are concentrated in the proximal region of prostatic ducts and express stem cell antigen 1 (Sca-1). As Wnt signaling is important for the maintenance of stem cells, we determined whether Sca-1 expressing cells also express Axin2, as Axin2 expression is highly suggestive of active Wnt signaling. METHODS Axin2 promoter reporter mice were used for whole mount and fluorescence activated cell sorting (FACS) analysis to determine its expression in the prostate. Axin2 expressing cells were also examined for the co-expression of Sca-1. We also used a chemical activator of Wnt signaling, BIO, to determine the effects of Wnt signaling on the growth of primary prostate cells in vitro. RESULTS We show that Axin2 expression is present in all lobes and is regulated by androgens with the highest Axin2 expression in the lateral and dorsal prostate. Furthermore, a fraction of Axin2 expressing cells co-express Sca-1, suggesting that some progenitor cells have active Wnt signaling. Lastly, we demonstrate that activation of the Wnt pathway may result in increased growth, consistent with a role for Wnt signaling in maintenance and/or expansion of the progenitor cell population. CONCLUSION Axin2 expressing cells that co-express Sca-1 are present in all prostate lobes suggesting that progenitor cells reside within the Wnt active population. An understanding of the basic biology of signaling pathways mediating growth in the prostate may lead to rational therapies to treat benign prostatic hyperplasia and prostate cancer.     Association of the melanocortin 4 receptor with feed intake and daily gain in F2 Mangalitsa × Piétrain pigs

    Summary The melanocortin 4 receptor (MC4R) is a key factor in the regulation of energy balance and body weight. Hence it is a candidate for feed intake and energy homeostasis-related traits. Studies in humans and swine have revealed several sequence variants in the gene that are associated with some of these traits. In pigs the coding non-synonymous missense variant Asp298Asn in MC4R has been associated with feed intake, fatness and growth. Here we confirm the association of this Piétrain-derived polymorphism with feed intake and daily gain in the F2 generation of a Mangalitsa × Piétrain cross. In one Piétrain founder animal, we detected an additional non-synonymous missense variant Arg236His. Thus, the MC4R gene could be a useful marker for increased growth in the relatively slow-growing Piétrain breed. [source]


    Influence of previous frost damage on tree growth and insect herbivory of Eucalyptus globulus globulus

    Abstract The plant stress hypothesis suggests that some herbivores favour stressed plants, whereas the plant vigour hypothesis proposes that other herbivores prefer vigorous plants. The effects of a prior stress, that of frost damage, were examined on the subsequent growth of Eucalyptus globulus globulus and on the response of insect herbivores. Frost damage affected tree growth by reducing new leaf area and increasing specific leaf area (SLA). However, herbivore abundance was not affected by prior frost damage. Two feeding trials using Anoplognathus chloropyrus and Hyalarcta huebneri and a morphometric study of Ctenarytaina eucalypti were conducted to assess the performance of herbivores on trees that had suffered more or less frost damage. Consumption by A. chloropyrus and H. huebneri was unaffected by foliage origin (damaged versus healthy). Hyalarcta huebneri grew faster when fed leaves from previously damaged trees, and C. eucalypti from previously damaged trees were larger than those from healthy trees. Enhanced insect performance on frost damaged plants may have resulted from the high specific leaf area (most likely thinner) leaves. The herbivore abundance data did not support the hypothesis that previously frost damaged plants are preferred by insects. However, increased growth of H. huebneri and larger body size of C. eucalypti on damaged trees indicates that previously stressed trees may produce leaves of higher nutritional value. [source]
