A. Fontem Abstract Experiments were conducted to detect potential hosts of Phytophthora infestans, causal agent of potato late blight among weeds occurring in Cameroon. Isolates of P. infestans isolated from garden huckleberry (Solanum scabrum), potato (S. tuberosum) and tomato (S. lycopersicon) were inoculated on detached leaves of 12 solanaceous and 14 asteraceous species collected from the potato agroecosystem in the western highlands of Cameroon. Isolates of P. infestans from huckleberry and potato infected the same host plants as well as gboma eggplant (S. macrocarpon) and two asteraceous weeds; Billy goatweed (Ageratum conyzoides) and Dichrocephala (Dichrocephala integrifolia). Inoculum from potato caused late blight symptoms on haemorrhage plant (Aspilia africana); while inoculum from tomato resulted in late blight on worowo (Solanecio biafrae). This is the first report of late blight infection on S. macrocarpon, A. conyzoides, Sol. biafrae and Asp. africana in Cameroon. The research results indicate that some asteraceous and solanaceous weeds may be alternative hosts of P. infestans in the potato agroecosystem. [source] Effects of Mycorrhizae and Nontarget Organisms on Restoration of a Seasonal Tropical Forest in Quintana Roo, Mexico: Factors Limiting Tree EstablishmentRESTORATION ECOLOGY, Issue 2 2005Michael F. Allen Abstract We initiated a study of the effects of mycorrhizal fungal community composition on the restoration of tropical dry seasonal forest trees. Tree seedlings were planted in a severely burned experimental site (1995 fire) during the growing season of 1998 at the El Edén Ecological Reserve, in north Quintana Roo, Mexico. Seedlings of Leucaena leucocephala, Guazuma ulmifolia, Caesalpinia violacea, Piscidia piscipula, Gliricidia sepium, and Cochlospermum vitifolium were germinated in steam-sterilized soil and either remained uninoculated (nonmycorrhizal at transplanting) or were inoculated with mycorrhizal fungi in soils from early-seral (recently burned) or late-seral (mature forest) inoculum. Inoculum from the early-seral soil was largely Glomus spp., whereas a diverse community of arbuscular mycorrhizal fungi were reintroduced from the mature forest including species of Scutellospora, Gigaspora, Glomus, Sclerocystis, and Acaulospora. Plants grew better when associated with the mature forest inoculum, unlike a previous experiment in which plants grew taller with the early-seral inoculum. Reasons for the different responses include a less-intense burn resulting in more residual organic matter. In addition to mycorrhizal responses, plants were severely affected by deer browsing. One tree species, C. vitifolium found in the region but not in the reserve, was eliminated by a resident fungal facultative pathogen. Several practical conclusions for restoration can be made. The common nursery practice of soil sterilization may be detrimental because it eliminates beneficial mycorrhizal fungi; species not native to the site may not survive because they may not be adapted to the local pathogens; and herbivory can be severe depending on the landscape context of the restoration. [source] The promise and the potential consequences of the global transport of mycorrhizal fungal inoculumECOLOGY LETTERS, Issue 5 2006Mark W. Schwartz Abstract Advances in ecology during the past decade have led to a much more detailed understanding of the potential negative consequences of species' introductions. Moreover, recent studies of mycorrhizal symbionts have led to an increased knowledge of the potential utility of fungal inoculations in agricultural, horticultural and ecological management. The intentional movement of mycorrhizal fungal species is growing, but the concomitant potential for negative ecological consequences of invasions by mycorrhizal fungi is poorly understood. We assess the degree to which introductions of mycorrhizal fungi may lead to unintended negative, and potentially costly, consequences. Our purpose is to make recommendations regarding appropriate management guidelines and highlight top priority research needs. Given the difficulty in discerning invasive species problems associated with mycorrhizal inoculations, we recommend the following. First, careful assessment documenting the need for inoculation, and the likelihood of success, should be conducted prior to inoculation because inoculations are not universally beneficial. Second, invasive species problems are costly and often impossible to control by the time they are recognized. We recommend using local inoculum sources whenever possible. Third, non-sterile cultures of inoculum can result in the movement of saprobes and pathogens as well as mutualists. We recommend using material that has been produced through sterile culture when local inoculum is not available. Finally, life-history characteristics of inoculated fungi may provide general guidelines relative to the likelihood of establishment and spread. We recommend that, when using non-local fungi, managers choose fungal taxa that carry life-history traits that may minimize the likelihood of deleterious invasive species problems. Additional research is needed on the potential of mycorrhizal fungi to spread to non-target areas and cause ecological damage. [source] Search of Microorganisms that Degrade PAHs under Alkaline ConditionsENGINEERING IN LIFE SCIENCES (ELECTRONIC), Issue 4 2004A. Gerbeth Abstract Bacterial strains were enriched from building rubble contaminated with polycyclic aromatic hydrocarbons (PAHs). These strains were studied as an inoculum in bioremediation processes with contaminated building rubble. The selection criteria for the bacteria were broad profiles in PAH degradation, stable expression of the traits and tolerance to alkaline conditions. Various strains of Micrococcus sp., Dietzia sp., Rhodococcus sp. and Pseudomonas sp. met the selection criteria. In general, degradative activity was limited at higher pH values. Strains of Micrococcus were suitable for practical use as complete degradation of various PAHs was observed at pH values exceeding 10. Strains of Dietzia sp. showed broad PAH degradation profile, but in some cases degradation came to a halt leaving some of the PAHs unutilized. With Dietzia sp. this could be due to inhibitory effects from the accumulation of toxic PAH metabolic products and/or growth-limiting media conditions. [source] Production of a Laccase and Decrease of the Phenolic Content in Canola Meal during the Growth of the Fungus Pleurotus ostreatus in Solid State Fermentation ProcessesENGINEERING IN LIFE SCIENCES (ELECTRONIC), Issue 1 2004J. Hu Abstract Solid state fermentation of canola meal was carried out with the fungus Pleurotus ostreatus DAOM 197961, which is a producer of laccase. The aim of this study was to examine the effects of moisture content, inoculum size, homogenisation of inoculum and particle size of canola meal on the growth of the fungus, the production of a laccase and the decrease of the content of sinapic acid esters (SAE) in a solid state process. The results showed that the optimum moisture content, which was varied in the media between 50% and 75%, for the growth and enzyme production was 60%. The initial rate of SAE content decrease was faster in the media with 70% and 75% moisture than in those with lower moisture levels. In the study of the effects of inoculum concentration in the range of 1.1 mg to 5.5 mg/g of the medium, it was found that larger amounts of biomass and enzyme were produced in the media with inoculum concentrations from 1.1 mg to 3.3 mg/g of the medium than in the media with a higher inoculum concentration. The final and approximately the same concentrations of SAE were reached at the same time regardless of the inoculum concentration. Considering that the fungus formed pellets under the conditions at which it was grown during the inoculum preparation, it was necessary to break them by homogenisation prior to their utilisation as an inoculum. The homogenisation was carried out during a period between 15s and 200s. Although higher biomass concentrations and enzyme activities were obtained in the media which were inoculated with the inoculum homogenised for 15s and 30s, the maximum enzyme activities and biomass concentrations were reached in the media inoculated with the inoculum, which was homogenised for 120s and 200s. The time of inoculum homogenisation did not influence the kinetics of the SAE decrease. When the effects of the particle size of canola meal on the process were studied, it was found that larger particles of the meal in the solid media were more favourable for the production of the biomass and enzyme, and for a faster decrease of the SAE content than those of smaller sizes. From the obtained results it can be concluded that the tested variables have a significant influence on the growth of the fungus Pleurotus ostreatus DAOM 197961, the production of laccase and the decrease of the SAE content in canola meal. The data could be useful for the development of a solid state process for the production of laccase and for the decrease of the phenolics content in canola meal. [source] Resting spore formation of aphid-pathogenic fungus Pandora nouryi depends on the concentration of infective inoculumENVIRONMENTAL MICROBIOLOGY, Issue 7 2008Zhi-Hong Huang Summary Resting spore formation of some aphid-pathogenic Entomophthorales is important for the seasonal pattern of their prevalence and survival but this process is poorly understood. To explore the possible mechanism involved in the process, Pandora nouryi (obligate aphid pathogen) interacted with green peach aphid Myzus persicae on cabbage leaves under favourable conditions. Host nymphs showered with primary conidia of an isolate (LC50: 0.9,6.7 conidia mm,2 4,7 days post shower) from air captures in the low-latitude plateau of China produced resting spores (azygospores), primary conidia or both spore types. Surprisingly, the proportion of mycosed cadavers forming resting spores (Pcfrs) increased sharply within the concentrations (C) of 28,240 conidia mm,2, retained high levels at 240,1760, but was zero or extremely low at 0.3,16. The Pcfrs,C relationship fit well the logistic equation Pcfrs = 0.6774/[1 + exp(3.1229,0.0270C)] (r2 = 0.975). This clarified for the first time the dependence of in vivo resting spore formation of P. nouryi upon the concentration of infective inoculum. A hypothesis is thus proposed that some sort of biochemical signals may exist in the host,pathogen interaction so that the fungal pathogen perceives the signals for prompt response to forthcoming host-density changes by either producing conidia for infecting available hosts or forming resting spores for surviving host absence in situ. [source] Anaerobic degradation of benzene by a marine sulfate-reducing enrichment culture, and cell hybridization of the dominant phylotypeENVIRONMENTAL MICROBIOLOGY, Issue 1 2008Florin Musat Summary The anaerobic biodegradation of benzene, a common constituent of petroleum and one of the least reactive aromatic hydrocarbons, is insufficiently understood with respect to the involved microorganisms and their metabolism. To study these aspects, sulfate-reducing bacteria were enriched with benzene as sole organic substrate using marine sediment as inoculum. Repeated subcultivation yielded a sediment-free enrichment culture constituted of mostly oval-shaped cells and showing benzene-dependent sulfate reduction and growth under strictly anoxic conditions. Amplification and sequencing of 16S rRNA genes from progressively diluted culture samples revealed an abundant phylotype; this was closely related to a clade of Deltaproteobacteria that includes sulfate-reducing bacteria able to degrade naphthalene or other aromatic hydrocarbons. Cell hybridization with two specifically designed 16S rRNA-targeted fluorescent oligonucleotide probes showed that the retrieved phylotype accounted for more than 85% of the cells detectable via DAPI staining (general cell staining) in the enrichment culture. The result suggests that the detected dominant phylotype is the ,candidate species' responsible for the anaerobic degradation of benzene. Quantitative growth experiments revealed complete oxidation of benzene with stoichiometric coupling to the reduction of sulfate to sulfide. Suspensions of benzene-grown cells did not show metabolic activity towards phenol or toluene. This observation suggests that benzene degradation by the enriched sulfate-reducing bacteria does not proceed via anaerobic hydroxylation (mediated through dehydrogenation) to free phenol or methylation to toluene, respectively, which are formerly proposed alternative mechanisms for benzene activation. [source] Use of a site-specific recombination-based biosensor for detecting bioavailable toluene and related compounds on rootsENVIRONMENTAL MICROBIOLOGY, Issue 4 2003N. Carol Casavant Summary We constructed and characterized a plasmid-based genetic system that reports the expression of a toluene-responsive promoter (PtbuA1) by effecting an irreversible, heritable change in the biosensor cell. Expression of the reporter gene gfp is strongly repressed in the absence of expression from the PtbuA1 promoter, and high level gfp expression in the original cell and its progeny is mediated by the site-specific recombination machinery of bacteriophage P22 to initiate removal of a repressor cassette. The reporter plasmid pTolLHB was functional in two soil saprophytes, Pseudomonas fluorescens A506 and Enterobacter cloacae JL1157, with the efficiency and sensitivity to low toluene concentrations being optimal in P. fluorescens A506. In culture, 80,100% of the A506 (pTolLHB) population expressed gfp following exposure to 0.2 µm toluene for one to three hours. Compared to the response of A506 containing a plasmid-borne PtbuA1 - gfp fusion, the recombination-based biosensor was more sensitive at detecting low toluene and trichloroethylene concentrations. An A506 (pTolLHB) inoculum, which had a background of 2.5% of the cells expressing gfp, was introduced onto barley roots in soil microcosms. If toluene was introduced into the microcosms, after 24 h, 72% of the A506 (pTolLHB) cells recovered from roots expressed gfp, indicating bioavailable toluene to rhizosphere bacteria. When toluene was not introduced, 16.5% of the A506 (pTolLHB) cells recovered from the roots expressed gfp, indicating that natural inducers of the PtbuA1 promoter were present in the barley rhizosphere. When introduced into rhizotrons containing barley plants and toluene vapours, the biosensor allowed localization of the availability of toluene along the seminal roots. In rhizotrons that were not exposed to toluene vapours, the biosensor exhibited high PtbuA1 -promoter activity in distinct regions along the seminal roots, indicating spatial heterogeneity plant- or rhizosphere microbial community-derived inducers of the PtbuA1 promoter. This recombination-based toluene biosensor thus was useful in identifying bacterial exposure to transient or low levels of toluene, or related compounds, directly in the environment. [source] Influence of hydroxypropyl-,-cyclodextrin on the extraction and biodegradation of phenanthrene in soilENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 3 2004Brian J. Reid Abstract A study was conducted to investigate the effect of hydroxypropyl-,-cyclodextrin (HPCD) on the aging and biodegradation of phenanthrene (PHE) in soil. Soil was spiked with PHE at 25 mgPHE/kgSOIL and HPCD at a range of concentrations from 0 to 3.5 gHPCD/kgSOIL and aged for 1, 84, and 322 d. At each time point, a variety of analyses were performed to assess the loss and aging of the PHE in the soil. Methods included determination of total PHE remaining, dichloromethane (DCM) and butan-1-ol (BuOH) extractions, and determination of PHE extractable by an aqueous HPCD shake extraction. Mineralization assays were also carried out to assess the availability of the PHE to a PHE-degrading bacterial inoculum. It was found that the presence of HPCD in the soils increased PHE loss from the aged soil systems, particularly at the higher application rates. Dichloromethane and BuOH extractabilities were reduced with aging and increasing HPCD concentration, as was the amount of PHE that was extractable using an aqueous HPCD shake extraction or that was available for mineralization. The DCM and BuOH extraction yielded similar results, and both greatly overestimated the availability of the PHE to the degraders, whereas the HPCD extraction results were very similar to that of PHE biodegradation. This study indicates that cyclodextrins have potential for use as alternatives to surfactants in enhancing the desorption/solubilization and degradation of recalcitrant organic contaminants in soil. [source] Pyrene and chrysene fate in surface soil and sand microcosmsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 2 2001J. Chadwick Roper Abstract Polycyclic aromatic hydrocarbons (PAHs) are major components of wastes from municipal gas plants and many wood preservatives. Soil contaminated with these wastes is a potential threat to human health because of the carcinogenicity of many PAHs. This study follows the fate of two four-ring PAHs, pyrene and chrysene, in three matrices: an adapted soil (obtained from a site contaminated with PAHs for more than 75 years), an uncontaminated soil (with and without an inoculum of adapted soil), and sand mixed with an inoculum of adapted soil. Radiolabeled pyrene, chrysene, and salicylic acid (a metabolite of PAH biodegradation) were used to trace the mineralization, transformation, extractability, and formation of an unextractable residual over time. Linear approximations of the rates of these processes were made. High-performance liquid chromatography (HPLC) analysis of extracts from inoculated soil showed the transient formation of two known metabolites: 1-hydroxypyrene (from pyrene) and 1-hydroxy-2-naphthoic acid (from chrysene). The amount of extractable label diminished steadily over the course of the study in systems that were not inhibited with sodium azide, whereas the amount of extractable label remained relatively constant in inhibited systems. Correspondingly, the amount of nonextractable residual label generally increased during each incubation in uninhibited systems, whereas the amount of this residual label remained relatively constant in inhibited systems. In contrast, the rate and extent of mineralization varied widely across matrix types. This suggests that alterations of the PAH that impact extractability and residual formation are common, in contrast to mineralization, which was apparently limited to adapted communities. [source] Epidemiology of sharka disease in FranceEPPO BULLETIN, Issue 2 2006G. Labonne Plum pox virus was first detected in France in the 1960s. Both PPV-D and PPV-M strains are present but epidemics related to the PPV-M strain detected in the late 1980s are the most problematic. The two PPV strains have unequal distributions in peach and apricot orchards and different prevalences. More than 20 different aphid species have been identified as vectors of PPV but most of them do not colonize Prunus species. Thus, aphids involved in the spread of PPV in orchards are essentially visiting aphids. The main sources of inoculum for the vectors are leaves and fruits of infected stone-fruit trees. Spontaneous, wild and ornamental Prunus species such as Prunus dulcis, P. spinosa or P. pissardii are susceptible to PPV isolates found in France but their role as a reservoir in sharka epidemics is probably negligible. The disease spreads rapidly in orchards but the rate of progression may vary according to the identity of the PPV strain and the Prunus species. Analysis of spatial patterns of disease has shown that secondary spread by aphids frequently occurs over short distances in the orchards (aggregated patterns) but also that dissemination at longer distances (of several hundred metres) is a common event. [source] Resistance of potato cultivars to Synchytrium endobioticum in field and laboratory tests, risk of secondary infection, and implications for phytosanitary regulationsEPPO BULLETIN, Issue 1 2005R. P. Baayen Laboratory (Spieckermann) tests, pot tests and field tests provided concordant evidence for the partial nature of resistance of potatoes to pathotypes 1 (D1) and 6 (O1) of Synchytrium endobioticum. Susceptible potato cultivars produced large warts (> 16 mm in diameter) in Spieckermann tests and had low field resistance levels (1,6). Field-resistant cultivars (levels 7,9) produced small warts or no warts at all in Spieckermann and field tests. In pot tests, at low inoculum levels (1 sporangium per 25 g soil) susceptible cultivars still developed warts, whereas field-resistant ones did not develop any warts below 25 sporangia per g soil. Above 35 sporangia per g soil, 100% disease incidence was observed in susceptible cultivars but only minimal wart development in field-resistant ones. Tests with continuous cultivation of potato cultivars in infected soil during three consecutive years showed that field-resistant cultivars will not support build-up of inoculum in soil. It is concluded that field-resistant cultivars do not create a risk of secondary infection, the criterion given for resistance in EU Directive 69/464/EC. [source] WITHIN-HOST POPULATION DYNAMICS AND THE EVOLUTION OF MICROPARASITES IN A HETEROGENEOUS HOST POPULATIONEVOLUTION, Issue 2 2002Vitaly V. Ganusov Abstract Why do parasites harm their hosts? The general understanding is that if the transmission rate and virulence of a parasite are linked, then the parasite must harm its host to maximize its transmission. The exact nature of such trade-offs remains largely unclear, but for vertebrate hosts it probably involves interactions between a microparasite and the host immune system. Previous results have suggested that in a homogeneous host population in the absence of super- or coinfection, within-host dynamics lead to selection of the parasite with an intermediate growth rate that is just being controlled by the immune system before it kills the host (Antia et al. 1994). In this paper, we examine how this result changes when heterogeneity is introduced to the host population. We incorporate the simplest form of heterogeneity,random heterogeneity in the parameters describing the size of the initial parasite inoculum, the immune response of the host, and the lethal density at which the parasite kills the host. We find that the general conclusion of the previous model holds: parasites evolve some intermediate growth rate. However, in contrast with the generally accepted view, we find that virulence (measured by the case mortality or the rate of parasite-induced host mortality) increases with heterogeneity. Finally, we link the within-host and between-host dynamics of parasites. We show how the parameters for epidemiological spread of the disease can be estimated from the within-host dynamics, and in doing so examine the way in which trade-offs between these epidemiological parameters arise as a consequence of the interaction of the parasite and the immune response of the host. [source] Bacterial competition between a bacteriocin-producing and a bacteriocin-negative strain of Streptococcus bovis in batch and continuous cultureFEMS MICROBIOLOGY ECOLOGY, Issue 3 2006Bruno M. Xavier Abstract A bacteriocin-producing Streptococcus bovis strain (HC5) outcompeted a sensitive strain (JB1) before it reached stationary phase (pH 6.4), even though it grew 10% slower and cell-free bovicin HC5 could not yet be detected. The success of bacteriocin-negative S. bovis isolates was enhanced by the presence of another sensitive bacterium (Clostridium sticklandii SR). PCR based on repetitive DNA sequences indicated that S. bovis HC5 was not simply transferring bacteriocin genes to S. bovis JB1. When the two S. bovis strains were coinoculated into minimal medium, bacteriocin-negative isolates predominated, and this effect could be explained by the longer lag time (0.5 vs. 1.5 h) of S. bovis HC5. If the glucose concentration of the minimal medium was increased from 2 to 7 mg mL,1, the effect of lag time was diminished and bacteriocin-producing isolates once again dominated the coculture. When the competition was examined in continuous culture, it became apparent that batch culture inocula were never able to displace a strain that had already reached steady state, even if the inoculum was large. This result indicated that bacterial selection for substrate affinity was even more important than bacteriocin production. [source] On the reproducibility of microcosm experiments , different community composition in parallel phototrophic biofilm microcosmsFEMS MICROBIOLOGY ECOLOGY, Issue 2 2006Guus Roeselers Abstract Phototrophic biofilms were cultivated simultaneously using the same inoculum in three identical flow-lane microcosms located in different laboratories. The growth rates of the biofilms were similar in the different microcosms, but denaturing gradient gel electrophoresis (DGGE) analysis of both 16S and 18S rRNA gene fragments showed that the communities developed differently in terms of species richness and community composition. One microcosm was dominated by Microcoleus and Phormidium species, the second microcosm was dominated by Synechocystis and Phormidium species, and the third microcosm was dominated by Microcoleus- and Planktothrix -affiliated species. No clear effect of light intensity on the cyanobacterial community composition was observed. In addition, DGGE profiles obtained from the cultivated biofilms showed a low resemblance with the profiles derived from the inoculum. These findings demonstrate that validation of reproducibility is essential for the use of microcosm systems in microbial ecology studies. [source] Salinity as a structuring factor for the composition and performance of bacterioplankton degrading riverine DOCFEMS MICROBIOLOGY ECOLOGY, Issue 2 2003Silke Langenheder Abstract The impact of salinity on the composition and functional performance (biomass production, growth efficiency and growth rates) of bacterial communities was investigated using batch cultures growing on dissolved organic carbon from a river draining into the Northern Baltic Sea. The cultures were adjusted to riverine or estuarine salinity levels and inoculated with bacteria from these two environments. Bacterial growth efficiencies differed in response to salinity and the origin of the inoculum. When salinity was adjusted to correspond to the salinity at the site where the inoculum was retrieved, growth efficiency was relatively high (11.5±2.6%). However, when bacteria were confronted with a shift in salinity, growth efficiency was lower (7.5±2.0%) and more of the utilized carbon was respired. In contrast, growth rates were higher when bacteria were exposed to a change in salinity. The composition of the bacterial communities developing in the batch cultures differed, as shown by 16S rDNA DGGE, depending on the origin of the inoculum and salinity. Reverse and direct DNA,DNA hybridization revealed salinity optima in the growth of specific bacterial strains as well as broader phylogenetic groups. Strains belonging to the ,- and ,- Proteobacteria, Actinobacteria and ,- Proteobacteria other than the genus Pseudomonas showed higher relative abundance under freshwater conditions, whereas strains of the genus Pseudomonas and the Cytophaga,Flavobacterium,Bacteroides group were favored by estuarine conditions. Generally, our results demonstrate functional changes associated with changes in community composition. We suggest that even moderate changes in salinity affect bacterial community composition, which subsequently leads to altered growth characteristics. Shah Abstract Nutrition influenced growth, sporulation and virulence of the insect pathogenic fungus, Metarhizium anisopliae. Virulent conidia were produced on susceptible insect hosts, 1% yeast extract, 2% peptone, osmotic stress medium (OSM) and CN 10:1 medium. Several strain independent markers were identified that could be used to predict the virulence of M. anisopliae conidia. Virulent conidia typically had high levels of spore bound Pr1, an important cuticle degrading protease, and high germination rates. We also show for the first time that virulent conidia have an endogenous CN ratio below 5.2:1. Real Time PCR revealed that virulent conidia from insects contained significantly higher levels of transcripts of pr1 A and other pathogenicity-related genes than inoculum from artificial media. Of the artificial media studied, 1% yeast extract medium yielded the most virulent conidia, these had higher levels of transcripts of these pathogenicity-related genes than the least virulent conidia from the high conidia yielding CN 35:1 medium (= SDA), however, the levels were significantly lower than those in insect-derived conidia. Our study shows for the first time that the passaged inoculum is virulent irrespective of the original culture medium or insect host. Virulent conidia were consistently produced on OSM even though growth and sporulation were poor. We postulate that starvation conditions, whether in vivo or in vitro, results in de-repression of Pr1 and that elevated levels of this enzyme enhance fungal virulence. Gibson Abstract During brewery handling, production strains of yeast must respond to fluctuations in dissolved oxygen concentration, pH, osmolarity, ethanol concentration, nutrient supply and temperature. Fermentation performance of brewing yeast strains is dependent on their ability to adapt to these changes, particularly during batch brewery fermentation which involves the recycling (repitching) of a single yeast culture (slurry) over a number of fermentations (generations). Modern practices, such as the use of high-gravity worts and preparation of dried yeast for use as an inoculum, have increased the magnitude of the stresses to which the cell is subjected. The ability of yeast to respond effectively to these conditions is essential not only for beer production but also for maintaining the fermentation fitness of yeast for use in subsequent fermentations. During brewery handling, cells inhabit a complex environment and our understanding of stress responses under such conditions is limited. [source] Utilization of essential oil as natural antifungal against nail-infective fungiFLAVOUR AND FRAGRANCE JOURNAL, Issue 2 2002Mamta Patra Abstract During antifungal screening of some essential oils, Foeniculum vulgare exhibited the strongest activity, completely inhibiting the mycelial growth of the nail-infective fungi, Trichophyton rubrum, T. mentagrophytes and Scytalidium dimidiatum. The essential oil was found to be fungicidal at 0.2, 0.4 and 0.5 µl/ml concentrations. The oil was efficiently active against heavy doses of inoculum at minimum fungicidal concentrations. The fungicidal activity of the oil was found to be thermostable up to 80 °C, with no descramble decrease in activity after 48 months of storage. The oil also showed a broad fungitoxic spectrum, inhibiting the mycelial growth of other nail-infective fungi, viz. Aspergillus flavus, A. fumigatus, A. niger,A. ustus, Candida albicans, Epidermophyton floccosum, Microporum audouinii, M. canis, M. gypseum, M. nanum, Rhizopus nigricans, Trichophyton tonsurans and T. violaceum. Moreover, it did not exhibit any adverse effects on mammalian skin and nails up to 5% concentration. Hietala Summary Rhizoctonia solani was frequently isolated in the Italian Alps from nursery-grown European beech (Fagus sylvatica) seedlings displaying symptoms of cotyledon rot. Koch's postulates were verified and mode of infection of the associated isolates was investigated with light and scanning electron microscopy. Population structure of the pathogen was investigated by scoring the anastomosis reaction type in pairings between different isolates from the same seedbed. One pathogen genotype showed a large distribution area within the seedbed, this implying that the inoculum had been building up in the seedbed over a longer time period. Hyphal anastomosis tests and sequence analysis of the internal transcribed spacer (ITS) region of ribosomal DNA indicated that the pathogen belongs to AG 2-1 of R. solani. ITS sequence analysis indicates that the isolates from beech are closely related to R. solani isolates causing a disease on tulip. The striking similarities between the two diseases are discussed. La structure de la population de l'agent pathogène a étéétudiée en examinant les réactions d'anastomoses dans les confrontations par paires des isolats d'un même lit de semences. Un génotype particulier s'est avéré largement distribué dans le lit de semence, suggérant soit une accumulation de l'inoculum pendant une longue période soit que ce génotype est capable de reproduction homocaryotique, favorisant sa dispersion. Les tests d'anastomose et l'analyse de la séquence de la région ITS de l'ADN ribosomal indiquent que l'agent pathogène appartient au groupe AG 2-1 de R. solani. L'analyse de la séquence de l'ITS montre que les isolats de hêtre sont proches d'isolats de R. solani pathogènes sur tulipe. Ein Kompatibilitätstyp war innerhalb des Saatbeetes weit verbreitet, was darauf hindeutet, dass sich das Inokulum über einen längeren Zeitraum dort angereichert hatte und/oder der Genotyp homokaryotisch fruchtet, was seine Ausbreitung fördert. [source] Seasonal and spatial mortality patterns of holm oak seedlings in a reforested soil infected with Phytophthora cinnamomiFOREST PATHOLOGY, Issue 6 2005M. C. Rodríguez-Molina Summary The viability of 1-year-old holm oak (Quercus ilex) seedlings in a soil naturally infected with Phytophthora cinnamomi was studied during 2 consecutive years in a plot located in south-western Spain. In both years, total mortality during autumn and winter was not noticeable (<2.1%). In spring, mortality levels were higher (8.3,4.6%), especially the first year. A steep increase in total mortality occurred in summer, both in the first (11.4%) and second (24.2%) year, but mortality attributable to P. cinnamomi was 1.9 and 7.6%, respectively. Thus, 2 years after planting, total cumulative mortality was 43.4%, and that attributable to P. cinnamomi 9.6% (i.e. 22.1% of total mortality). Fungus-derived mortality followed a spatially aggregated pattern in the reforestation plot, suggesting a clustered distribution of the inoculum in the soil. Furthermore, mortality by P. cinnamomi was also associated with nearness of infected adult trees in the plot. Results obtained are discussed in the framework of seasonal water deficit, P. cinnamomi damage, weed competition and sanitation techniques to be used in declined holm oak stands in Spain. Résumé La viabilité de semis de chênes verts d'un an dans un sol naturellement infecté par P. cinnamomi a été suivie pendant deux années consécutives dans une parcelle du sud-ouest de l'Espagne. La mortalité en automne et hiver a été négligeable (<2.1%) au cours des deux années. La mortalité a été plus forte au printemps (8.3 ,4.6%), particulièrement la première année. Un pic de mortalité a été observé pendant l'été, aussi bien la première (11.4%) que la deuxième année (24.2%) mais la mortalité attribuable àP. cinnamomi n'est que de 1.9% et 7.6% respectivement. Deux ans après plantation, la mortalité cumulée est de 43.4%, dont 9.6% attribuable àP. cinnamomi (22% de la mortalité totale). La mortalité associée à l'agent pathogène présente une agrégation spatiale dans la parcelle reboisée, suggérant une distribution en agrégats de l'inoculum dans le sol. De plus cette mortalité est associée à la proximitéà des arbres infectés dans la parcelle. Im Sommer stieg die Gesamtmortalität stark an, im ersten Jahr betrug sie 11,4%, im zweiten Jahr 24,2%, jedoch lagen die P. cinnamomi zuzuordnenden Raten bei nur 1,9 bzw. 7,6%. Yuan A study of lesion development in stems of Eucalyptus nitens following artificial inoculations with canker fungi was carried out on 16-year-old plantation trees. In a first trial cambium bark wounds on smooth- and rough-barked trees were inoculated with the mycelium of nine species of canker fungi, including Endothia gyrosa. In a second trial spores or mycelium of E. gyrosa were applied directly onto undamaged or superficially wounded bark surfaces. Infection subsequent to artificial inoculation via wounding (whatever the wounding technique or type of inoculum) resulted in significantly larger external lesions (mean lesion area up to 35.6 cm2 20 months after inoculation) on smooth bark compared with those on rough bark (up to 19.0 cm2). Microscopic studies of infected rough and smooth bark suggest that, once smooth bark is compromised by wounding and artificial inoculation, the particular anatomical structure of smooth bark may offer less mechanical resistance to post-penetration hyphal spread in comparison with rough bark. It is suggested that at a pre-penetration stage under natural conditions spores of E. gyrosa more easily infect rough bark via cracks associated with this type of bark but not present in smooth bark. Competitive ability (the ability to capture or co-occupy an opponent's inoculum) varied according to species and inoculum age. In wood block pairings in the absence of soil there was evidence that P. velutina opportunistically utilized C previously mobilized within an opponent's inoculum. 3.,In soil systems, short-term (28-day) respiratory losses of preloaded 14C (supplied as glucose) indicated that interaction could have a substantial C cost, depending on the resource quality of the opponents' inocula. Miller Abstract 1Two semiaquatic grasses, Panicum hemitomon Schultes and Leersia hexandra Schwartz, were grown for 12 weeks in sterilized soil in experimental mesocosms, with and without the addition of arbuscular mycorrhizal (AM) fungal inoculum (as nonsterilized soil), under the following rooting-zone flood regimes: waterlogged (W), free-draining (D), beginning W and ending D (W,D), and beginning D and ending W (D,W). The purpose of the experiment was to determine whether these controlled water regimes affected both colonization of wetland grasses by AM fungi and the effects of the colonization on various plant parameters. 2Water regime, addition of inoculum, and their interaction were highly significant effects on total and proportion of root length colonized by AM fungi. Trends were very similar for the two grass species. Colonization was less and plants smaller in the W and W,D than in the D and D,W treatments. The viability of mycorrhiza at the end of the experiment, as measured by vital staining techniques, was not affected by changes in water level. 3Colonized plants in all water level treatments showed an improvement in phosphorus (P) nutrition over noncolonized plants. Colonized grasses of both species gained consistently more P per plant and had greater tissue P concentrations, with the greatest P concentration in the most heavily colonized plants (from the D and D,W treatments). 4The effect of flooding on the mycorrhizal association depended largely on the extent to which the association was already established when the flooding occurred. SHEPARD The roles of biology in the morphogenesis of microbial mats and stromatolites remain enigmatic due to the vast array of physical and chemical influences on morphology. However, certain microbial behaviors produce complex morphological features that can be directly attributed to motility patterns. Specifically, laboratory experiments with a strain of the cyanobacteria Pseudanabaena demonstrate that distinctive morphologies arise from the undirected gliding and colliding of filaments. When filamentous cells collide, they align and clump, producing intersecting ridges surrounding areas with low cell density, i.e. reticulate structures. Cell motility is essential for the development of reticulates and associated structures: filaments organize into reticulates faster than cell division and growth, and conditions that inhibit motility also inhibit reticulate formation. Cell density of the inoculum affects the frequency of cell,cell collisions, and thus the time required for biofilm organization into reticulate structures. This also affects the specific geometry of the reticulates. These patterns are propagated into larger structures as cyanobacterial cell numbers increase and cells remain motile. Thus, cell motility is important for templating and maintaining the morphology of these microbial communities, demonstrating a direct link between a microbial behavior and a community morphology. Reticulate geometries have been identified in natural microbial mats as well as in the fossil record, and these structures can be attributed to the motility of filamentous bacteria. [source] Role of Immune Serum in the Killing of Helicobacter pylori by MacrophagesHELICOBACTER, Issue 3 2010Stacey Keep Abstract Background:,Helicobacter pylori infection can lead to the development of gastritis, peptic ulcers and gastric cancer, which makes this bacterium an important concern for human health. Despite evoking a strong immune response in the host, H. pylori persists, requiring complex antibiotic therapy for eradication. Here we have studied the impact of a patient's immune serum on H. pylori in relation to macrophage uptake, phagosome maturation, and bacterial killing. Materials and Methods:, Primary human macrophages were infected in vitro with both immune serum-treated and control H. pylori. The ability of primary human macrophages to kill H. pylori was characterized at various time points after infection. H. pylori phagosome maturation was analyzed by confocal immune fluorescence microscopy using markers specific for H. pylori, early endosomes (EEA1), late endosomes (CD63) and lysosomes (LAMP-1). Results:, Immune serum enhanced H. pylori uptake into macrophages when compared to control bacteria. However, a sufficient inoculum remained for recovery of viable H. pylori from macrophages, at 8 hours after infection, for both the serum-treated and control groups. Both serum-treated and control H. pylori phagosomes acquired EEA1 (15 minutes), CD63 and LAMP-1 (30 minutes). These markers were then retained for the rest of an 8 hour time course. Conclusions:, While immune sera appeared to have a slight positive effect on bacterial uptake, both serum-treated and control H. pylori were not eliminated by macrophages. Furthermore, the same disruptions to phagosome maturation were observed for both serum-treated and control H. pylori. We conclude that to eliminate H. pylori, a strategy is required to restore the normal process of phagosome maturation and enable effective macrophage killing of H. pylori, following a host immune response. Davenport Summary:, A large number of experimental studies have been performed over the past decade in an attempt to develop a vaccine for human immunodeficiency virus (HIV). These studies have used a variety of approaches aimed at stimulating both antibody-mediated and cell-mediated immunity. Many of these experiments have been performed in macaque models of HIV. Analysis and modeling of the results of these studies provide the opportunity to investigate the mechanisms and limitations of viral control by humoral and cell-mediated immunity. These studies suggest that CD8+ T cells do ,too little too late' to prevent the establishment of viral infection and latency. By contrast, passively administered antibody acts extremely early to reduce the initial inoculum and slow viral growth. González-García Summary Effectiveness of fibrolytic enzymes supplementing a range of forage to concentrate (F:C) diets was assessed with goat (G) or cow (C) inoculum using the gas production (GP) technique. Four F:C diets were evaluated: forage (1:0), high forage (0.7:0.3), medium forage (0.5:0.5) and low forage (0.3:0.7) diets, supplemented or not with PromoteTM (PRO) at 1 or 2 ml/kg dry matter (DM). The GP kinetic was different between F:C (1:0 < 0.7:0.3 < 0.5:0.5 < 0.3:0.7) and inoculum. Responses to enzyme were positively related to forage level and differed with inoculum. The neutral detergent fibre and acid detergent fibre degradation were depressed by the concentrate in the substrates fermented with C and were not altered or even enhanced in G sets. Results confirm that increasing starch proportion modified the pattern of microbial fermentation, while no influences were detected in the improvement of cell wall degradation with fibrolytic enzymes. Another in vitro experiment was conducted to investigate factors by which endo-,-1,4-glucanase activity (EA) of PRO is compromised in a factorial design (3 × 4 × 3) for three pH (4.0, 5.5 and 6.5), four temperatures (30, 40, 50 and 70 °C) and three doses (1, 2 and 3 ml/kg DM of substrate). Maximum EA were obtained for pH 4.0, 50 °C and 3 ml/kg DM. Optimal conditions for PRO proved to be outside the normal ranges in ruminal environment. The production of the crude by-product may reach more than 4 millions t/year in Spain (MAPA, 2003), most of it in areas (Southeast) with shortage of pastures and conventional feeds for ruminants. Six continuous fermenters, inoculated with ruminal liquor from wethers or goats, were fed lucerne hay (LH); LH plus a concentrate including dried two-phase olive cake (LHCO) and; diet LHCO added with polyethylene glycol (LHCOP). The digestibility of carbohydrates (CHO) was affected (p < 0.001) by inoculum (67.0 and 58.8%, respectively, for goats and wethers) the lowest values being for diet LHCO (53.2 and 57.0% with inoculum from wethers and goats, respectively). The main volatile fatty acid (VFA) was acetic acid with higher (p < 0.01) values in fermenters with inoculum from goats than from wethers (80.2 and 63.0 mmol/day respectively). The efficiency of bacterial protein synthesis (EBS) was not different (p > 0.05) with inoculum from wethers and goats [26.4 and 28.1 and 35.2 and 33.5 g bacterial N/kg digested CHO, respectively, obtained by using diamino pimelic acid (DAPA) and purine bases (PB) as microbial markers]. The lowest (p < 0.05) values were found in fermenters fed diets LHCOP and LH, estimated, respectively, from DAPA and PB (21.9 and 29.0 g bacterial N/kg digested CHO). The substitution of a part of lucerne hay by a concentrate including dried 2POC does not seem to cause important differences in efficiency of VFA production. Results concerning bacterial protein synthesis are not so clear since values estimated from DAPA and PB did not show similar trends. Neither inoculum origin nor PEG had important effects on fermentation characteristics. The DAPA could be an adequate marker in continuous fermenters, with comparative purposes, as protozoa are not present and, estimated protein synthesis values are similar to those obtained in vivo with similar diets using urinary excretion of PD (Yáñez Ruiz et al., 2004b). Further research is needed to state the optimal proportion of 2POC in practical diets for ruminants at both maintenance and production stages. [source] |
