[source] Alcoholism Susceptibility Loci: Confirmation Studies in a Replicate Sample and Further MappingALCOHOLISM, Issue 7 2000Tatiana Foroud Background: There is substantial evidence for a significant genetic component to the risk for alcoholism. A previous study reported linkage to chromosomes 1, 2, and 7 in a large data set that consisted of 105 families, each with at least three alcoholic members. Methods: Additional, genotyping in the 105 families has been completed in the chromosomal regions identified in the initial analyses, and a replication sample of 157 alcoholic families ascertained under identical criteria has been genotyped. Two hierarchical definitions of alcoholism were employed in the linkage analyses: (1) Individuals who met both Feighner and DSM-III-R criteria for alcohol dependence represented a broad definition of disease; and (2) individuals who met ICD-10 criteria for alcoholism were considered affected under a more severe definition of disease. Results: Genetic analyses of affected sibling pairs supported linkage to chromosome 1 (LOD = 1.6) in the replication data set as well as in a combined analysis of the two samples (LOD = 2.6). Evidence of linkage to chromosome 7 increased in the combined data (LOD = 2.9). The LOD score on chromosome 2 in the initial data set increased after genotyping of additional markers; however, combined analyses of the two data sets resulted in overall lower LOD scores (LOD = 1.8) on chromosome 2. A new finding of linkage to chromosome 3 was identified in the replication data set (LOD = 3.4). Conclusions: Analyses of a second large sample of alcoholic families provided further evidence of genetic susceptibility loci on chromosomes 1 and 7. Genetic analyses also have identified susceptibility loci on chromosomes 2 and 3 that may act only in one of the two data sets. [source] Stratified resampling of phytosociological databases: some strategies for obtaining more representative data sets for classification studiesJOURNAL OF VEGETATION SCIENCE, Issue 4 2005Ilona Knollová Abstract Question: The heterogeneous origin of the data in large phytosociological databases may seriously influence the results of their analysis. Therefore we propose some strategies for stratified resampling of such databases, which may improve the representativeness of the data. We also explore the effects of different resampling options on vegetation classification. Methods: We used 6050 plot samples (relevés) of mesic grasslands from the Czech Republic. We stratified this database using (1) geographical stratification in a grid; (2) habitat stratification created by an overlay of digital maps in GIS; (3) habitat stratification with strata defined by traditional phytosociological associations; (4) habitat stratification by numerical classification and (5) habitat stratification by Ellenberg indicator values. Each time we resampled the database, taking equal numbers of relevés per stratum. We then carried out cluster analyses for the resampled data sets and compared the resulting classifications using a newly developed procedure. Results: Random resampling of the initial data set and geographically stratified resampling resulted in similar classifications. By contrast, classifications of the resampled data sets that were based on habitat stratifications (2,5) differed from each other and from the initial data set. Stratification 2 resulted in classifications that strongly reflected environmental factors with a coarse grain of spatial heterogeneity (e.g. macroclimate), whereas stratification 5 resulted in classifications emphasizing fine-grained factors (e.g. soil nutrient status). Stratification 3 led to the most deviating results, possibly due to the subjective nature of the traditional phytosociological classifications. Conclusions: Stratified resampling may increase the representativeness of phytosociological data sets, but different types of stratification may result in different classifications. No single resampling strategy is optimal or superior: the appropriate stratification method must be selected according to the objectives of specific studies. Ferreira N -Acetylglucosamine 6-phosphate deacetylase (E.C. 3.5.1.25), an enzyme from Escherichia coli involved in aminosugar catabolism, has been crystallized by the vapour-diffusion technique using phosphate as precipitant. X-ray diffraction experiments show the crystals to belong to the orthorhombic crystal system, with space group P21212. The unit-cell parameters are a = 82.09,(2), b = 114.50,(1), c = 80.17,(1),Å. The crystals diffract to a maximum resolution of 1.8,Å and an initial data set was collected to 2.0,Å. [source] |
