Selected AbstractsDetection of Cryptosporidium parvum in lettuceINTERNATIONAL JOURNAL OF FOOD SCIENCE & TECHNOLOGY, Issue 4 2007John E. Moore Summary Human cryptosporidiosis has emerged as an important gastrointestinal infection in the 1990s as a result of the ingestion of mainly contaminated water and to a lesser extent foodstuffs containing the protozoan parasite, Cryptosporidium parvum. This pathogen has particular clinical significance for immunocompromised persons, including AIDS patients and cancer patients receiving toxic chemotherapeutic drug regimens. There have been a limited number of studies performed examining the occurrence of the parasite on vegetables, including lettuce. Detection rates are very dependent on the laboratory isolation technique employed and has ranged from 1.2% to 14.5%. Current best practice of laboratory recovery, isolation and detection methods include detergent removal, oocysts concentration by immunomagnetic separation, followed by a combination of immunofluorescent microscopy and a nested PCR approach. Employment of contaminated non-potable water in the production of vegetables, particularly lettuce, may represent an important potential source of entry of pathogens into food processing and the human food chain. Given that lettuce is an important constituent of hamburger dressing, and the size of the fast-food industry, where lettuce is an important constituent, horticultural producers of lettuce should therefore place special emphasis on developing suitable and efficient Hazard Analysis Critical Control Point strategies for the critical control of oocysts depending on the type of unit operation employed and vegetable being processed. This review aims to examine (i) the incidence of C. parvum in vegetables, particularly lettuce and (ii) laboratory detection methods for the isolation and identification of this parasite from lettuce. [source] Why ,Health' is not a Central Category for Public Health PolicyJOURNAL OF APPLIED PHILOSOPHY, Issue 2 2009STEPHEN JOHN abstract We normally think that public health policy is an important political activity. In turn, we normally understand the value of public health policy in terms of the promotion of health or some health-related good (such as opportunity for health), on the basis of the assumption that health is an important constituent or determinant of wellbeing. In this paper, I argue that the assumption that the value of public health policy should be understood in terms of health leads us to overlook important benefits generated by such policy. To capture these benefits we need to understand the ends of public health policy in terms of the promotion of ,physical safety'. I then go on to argue that the idea that ,health' is an important category for evaluating or estimating individuals' wellbeing in the normative context of social policy is confused. I then clarify the relationship between my arguments and QALY-based accounts of health assessment. In the final section of the paper, I defend this surprising conclusion against various attacks. In Iran, Indian white shrimp (Fenneropenaeus indicus) is the major cultured shrimp species as a result of market demand, local availability and growth rate. It is mainly reared using commercial feed. The purpose of this study was to evaluate the effects of replacing 50% of the fish oil by vegetable oils in shrimp feed on total lipid, fatty acid composition and lipid oxidation of shrimp muscle. RESULTS: No significant differences in total lipid content (6.1,7.3 g kg,1) were found between edible tissues of shrimp fed different diets. The major fatty acids in shrimp muscle were palmitic, oleic, lionoleic, stearic, eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids. Higher levels of EPA and DHA were observed in muscle of shrimp fed a diet containing fish oil. Oxidative rancidity, measured as thiobarbituric acid reactive substances, for all shrimps did not exceed 0.2 mg malonaldehyde kg,1 muscle tissue, which was low and acceptable. CONCLUSION: This study had shown that the fatty acid composition of feed directly affects the fatty acid composition of Indian white shrimp muscle. Farmed Indian white shrimp can be considered as a species of low fat and shrimp muscle was quite stable to oxidation during storage. CHEN Summary., Thrombin can amplify inflammation induced by other stimuli, either through ischemia (consequent upon thrombosis), indirectly through generation of downstream mediators such as activated protein C, or directly via signals through protease activated receptors (PAR). This paper will summarize recent data from our laboratory indicating that thrombin is required to initiate CCR2-dependent leukocyte recruitment and that it is the principal determinant of the outcome after vascular injury, via PAR-1 activation of a distinct subset of smooth muscle cell progenitors. In both, tissue factor (TF) initiates thrombin generation and the thrombin acts locally, exemplifying that the initiation phase can generate autocrine or paracrine signalling molecules. Thrombin is an important constituent of innate immunity, able to amplify and modify responses to invading pathogens or tissue damage. With novel anti-thrombin therapeutics and agents to target PAR, a new understanding of the importance of thrombin may allow the development of innovative anti-inflammatory strategies. Kidney-resident DC (KDC) are an important constituent of passenger leucocytes that initiate the direct component of allograft rejection in transplantation and form a central element of the innate immune response following injurious stimuli to the kidney. DC are recruited to the kidney in pathophysiological states such as glomerulonephritis and ischaemia,reperfusion injury. However, the exact mechanism for engaging and attracting DC to infectious and transplant antigens, and whether specific DC subsets are involved remains unresolved. In addition, the extent to which resident and infiltrating DC contribute to the propagation of injury or rejection is also unclear. Despite consistently expanding published work regarding DC location, phenotype and function, there are a number of deficiencies in our knowledge base, particularly in relation to KDC. [source] Psychosocial Work Characteristics as Predictors of Affective Organisational Commitment: A Longitudinal Multi-Level Analysis of Occupational Well-BeingAPPLIED PSYCHOLOGY: HEALTH AND WELL-BEING, Issue 2 2010Thomas Clausen The purpose of this study was to identify longitudinal associations between psychosocial work characteristics and affective organisational commitment among 6,299 employees in the Danish eldercare services. Individual-level measures and group-level measures of psychosocial work characteristics were included in multi-level analyses. At the workgroup level, quality of leadership, influence at work, emotional demands, and work pace predicted affective organisational commitment at follow-up. At the individual level, quality of leadership, influence at work, team climate, role ambiguity, and work pace predicted affective organisational commitment at follow-up. Finally, a multi-level model including both individual- and group-level measures showed that quality of leadership measured at the group level and influence at work and quality of leadership measured at the individual level contributed to predicting affective organisational commitment at follow-up, while adjusting for baseline levels of affective organisational commitment. The results thus imply that affective organisational commitment is conditioned by individual and contextual factors in the psychosocial work environment and that multi-level models add to our understanding of complex organisational phenomena. As affective organisational commitment can be considered an important constituent of occupational well-being, implications for interventions in the psychosocial work environment to increase affective organisational commitment are also discussed. [source] Volatile constituents of different organs of Psoralea bituminosa L.FLAVOUR AND FRAGRANCE JOURNAL, Issue 2 2004Alessandra Bertoli Abstract The essential oil and SPME samples of the leaves, ,owers and seeds of Psoralea bituminosa L. were analysed by GC and GC,MS. We have investigated also the presence of monoterpene or aliphatic alcohol glucosides. The essential oils showed both qualitative and quantitative differences. The main constituents of the leaf and the ,ower essential oils were caryophyllene (23% and 18%, respectively), , -farnesene (15% and 6%, respectively), and germacrene D (24% and 18%, respectively). Signi,cant amounts (7%) of the same compounds were also directed in the seed essential oil, but tricyclene (11%) and , -pinene (50%) were the most important constituents of this oil. The volatile fractions of remaining leaf aqueous extracts after treatment with , -glucosidase revealed qualitative differences in comparison with the composition of the corresponding essential oils, and high levels of 3-hexen-1-ol (37%) and 1-octen-3-ol (27%) were observed. The SPME analysis of the fresh leaves, ,owers and seeds of P. bituminosa con,rmed the qualitative composition of the volatile oils, even if we detected signi,cative differences in the percentage ratio between monoterpenes and sesquiterpenes in comparison with the oils, where sesquiterpenes were the main components. In fact the variation of the monoterpenes, tricyclene, , -pinene and camphene between the leaf oil and the corresponding headspace sample was remarkable: tricyclene increased from 0.1% to 8%, , -pinene from 0.1% to 16% and camphene from 0.3% to 10% in the SPME samples. WIGNALL ABSTRACT A record of the Early Jurassic mass extinction event is reported from eastern Tethyan (Tibetan) locations for the first time. In the Mount Everest region a thick Lower Jurassic carbonate formation, here named the Yungjia Formation, is developed within the predominantly clastic Triassic,Jurassic succession. Within the formation a sharp transition from peloidal packstones/grainstones to thin-bedded, pyritic micrite-shales interbeds records a sharp pulse of deepening and development of dysoxic bottom waters. Both the lithiotid bivalves and the lituolid foraminifera are important constituents of the lower Yungjia Formation but they disappear at this flooding surface or a short distance below it. This extinction event is comparable to that seen at the base of the Pliensbachian/Toarcian boundary in western Tethyan platform carbonates but the Tibetan events occurred late in the Toarcian Stage as indicated by nannofossil biostratigraphy and C isotope chemostratigraphy. The Early Jurassic extinction event (and the associated spread of oxygen-poor waters) was therefore not synchronous throughout the Tethyan region. [source] The riverscape of Western Amazonia , a quantitative approach to the fluvial biogeography of the regionJOURNAL OF BIOGEOGRAPHY, Issue 8 2007Tuuli Toivonen Abstract Aim, To provide a quantitative spatial analysis of the riverscape (open-water bodies and their surrounding areas) of the Western Amazonian lowlands using a consistent surface of remotely sensed imagery. Taking into account the essential significance of fluvial environments for the Amazonian biota, we propose that an enhanced understanding of the Amazonian riverscape will provide new insight for biogeographical studies in the region and contribute to the understanding of these megadiverse tropical lowlands. Location, An area of 2.2 million km2 covering the Western Amazonian lowlands of the Andean foreland region, i.e. the upper reaches of the Amazon river system. Areas in Colombia, Venezuela, Ecuador, Peru, Brazil and Bolivia between longitudes 83 °W and 65 °W and latitudes 5 °N and 12 °S are included. Methods, A mosaic of 120 Landsat TM satellite images was created with 100-m resolution, and water areas of over 1 ha in size or c. 60 m in width were extracted using a simple ratio threshold applicable to a large set of data. With this method, 99.1% of the water areas present in 30-m imagery were mapped with images with 100-m resolution. Water pixels of distinct river segments were assigned to river classes on the basis of their channel properties, and islands and lakes were distinguished separately and classified. Measures of water patterns such as structure, composition, richness and remoteness were provided for various spatial units. Riverine corridors were computed from the open-water mask by outer limits of active channels and floodplain lakes. Analytical results are shown as both thematic maps and statistics. Results, A total of 1.1% of Western Amazonia is covered by open-water bodies over 1 ha in size or 60 m in width. River-bound waters comprise 98% of the total water surface. Whilst isolated lakes are scarce, river-bound oxbow and backchannel lakes are plentiful, comprising 17.5% of all waters. They are particularly frequent along meandering channels, which dominate both in area and length. The riverine corridors including active channels and floodplain lakes cover 17% of the land area. The average distance from any point of land to the nearest water is 12 km. Geographically speaking, the distribution of waters is uneven across the region, and the detailed characteristics of the riverscape are geographically highly variable. Three major, fluvially distinct regions can be identified: central Western Amazonia, the south, and the north-east. The proportional surface areas of the riverine corridors, numbers of lakes, sizes of islands and their distributions depend largely on the types and sizes of the rivers. Main conclusions, Our results support the notion of Western Amazonia as a dynamic, highly fluvial environment, highlighting and quantifying considerable internal variation within the region in terms of fluvial patterns and the processes that they reflect and control. Biogeographically, the variety of types of fluvial environments and their characteristics are important constituents of what influences the distribution of species and dynamics of terrestrial habitats. Spatially consistent riverscape data can serve as a consistent and scalable source of relevant information for other biogeographical approaches in the region. [source] Pharmacological and therapeutic effects of Berberis vulgaris and its active constituent, berberinePHYTOTHERAPY RESEARCH, Issue 8 2008Mohsen Imanshahidi Abstract Barberry (Berberis vulgaris L. family Berberidaceae) is well known in Iran and various parts of this plant including its root, bark, leaf and fruit have been used as folk medicine. The two decades of research has demonstrated different pharmacological and therapeutic effects of B. vulgaris and its isoquinoline alkaloids (particularly berberine). Studies carried out on the chemical composition of the plant show that the most important constituents of this plant are isoquinoline alkaloids such as berberine, berbamine and palmatine. Berberine represents one of the most studied among the naturally occurring protoberberine alkaloids. In addition to B. vulgaris (barberry), berberine is present in many other plants and is used for the treatment of different diseases. [source] New Bradoriid Arthropods from the Early Cambrian Balang Formation of Eastern Guizhou, South ChinaACTA GEOLOGICA SINICA (ENGLISH EDITION), Issue 1 2010Jin PENG Abstract: The Early Cambrian Balang Formation is comprised of mudrock and shale, which was deposited in a shelf environment in the eastern part of Guizhou, south China. The Balang Fauna, which consists of seven phyla, occurs in the middle and upper parts of the Balang Formation. Arthropods are important constituents of the Balang Fauna and include a great number of trilobites, large bivalved arthropods, and newly-discovered well-preserved bradoriid fossils. The bradoriids present include three genera and four species: Comptaluta inflate (Cheng, 1974) emend Hou et al., 2002; Comptaluta kailiensis sp. nov, and Alutella elongeta sp. nov, Aluta sp. This faunal assemblage in the Balang Formation is distinguished from the Tsunyiella Chang, 1964, Songlinella Yin, 1978 and Kunmingella Hou, 1956 assemblage which occurs in the Niutitang and Mingxinsi formations of the Yangtze Platform in middle region of Guizhou and which is earlier than the Balang Formation in age. However, this assemblage resembles the ComptalutaÖpik, 1968 assemblage from the Early Cambrian Heilinpu Formation in Wuding County, Yuanan Province and from the Ordian Stage of the Cambrian of Australia. The great abundance of ComptalutaÖpik, 1968 and overall taxonomic diversity of the ComptalutaÖpik, 1968 assemblage set it distinctly apart from the Alutella Kobayashi et Kato, 1951 and Aluta Hou, 1956 assemblages of the Balang Formation. Alutella Kobayashi et Kato, 1951 and Aluta Hou, 1956 also occur in the Early Cambrian Niutitang Formation of the Yangtze Platform of Guizhou. Individual Bradoriids from the Balang Formation are characterized by large size (>3 mm). The discovery of new Bradoriid assemblages not only expands the group's geographical range and assemblage affinities, but also indicates that Bradoriids migrated eastward from shallow-water to deeper-water environments during the Early Cambrian, indicating that they were capable of life in deeper-water, and adaptation to a new ecological setting. [source] Adherence and invasion of Bacteroidales isolated from the human intestinal tractCLINICAL MICROBIOLOGY AND INFECTION, Issue 10 2008V. Nakano Abstract Members of the genera Bacteroides and Parabacteroides are important constituents of both human and animal intestinal microbiota, and are significant facultative pathogens. In this study, the ability of Bacteroides spp. and Parabacteroides distasonis isolated from both diarrhoeal and normal stools (n = 114) to adhere to and invade HEp-2 cells was evaluated. The presence of putative virulence factors such as capsule and fimbriae was also investigated. Adherence to HEp-2 cells was observed in 75.4% of the strains, which displayed non-localized clusters. Invasion was observed in 37.5% and 26% of the strains isolated from diarrhoeal and non-diarrhoeal stools, respectively. All strains displayed a capsule, whereas none of them showed fimbriae-like structures. This is the first report of the ability of Bacteroides spp. and P. distasonis to adhere to and invade cultured HEp-2 epithelial cells. [source] |
