A Novel Three-Dimensional In Vitro System to Study Trophoblast,Endothelium Cell Interactions

Introduction Pregnancy complications have been linked to improper trophoblast migration and failure of spiral artery transformation. Endothelial cells play an essential role in directing trophoblast migration and transformation, although by an unknown mechanism. We describe a novel in vitro model to evaluate endothelial,trophoblast interaction and signaling in a three-dimensional system. Method of study Immortalized human endometrial endothelial cell line and first trimester trophoblast cells were co-cultured. Endothelial transformation into vessel-like structures occurred in MatrigelTM OpenLab Image Analysis software was used to monitor labeled trophoblast migration and endothelium transformation. Cytokine/chemokine production was determined using Multiplex. Results Trophoblast migrates toward endothelial cells in Matrigel, aligns on top of the endothelium within 4,8 hr and achieves complete replacement of the endothelium by 72,96 hr. Lipopolysaccharide treatment damages the endothelium and disrupts endothelium,trophoblast interaction. Conclusion We report a novel three-dimensional in vitro and in vivo system of trophoblast,endothelium cell interaction. Significant changes in endothelial cells' phenotype are observed upon differentiation in Matrigel. These changes may be necessary for endothelium to direct trophoblast migration and transformation. Role of computerized morphometric analysis in diagnosis of effusion specimens

Abstract The present study was undertaken to compare morphometric analysis of cells in 100 effusion samples, with unaided cytological diagnosis. All the cases in the form of Leishman's stained smears were subjected to computerized-interactive morphometry (CIM) by usage of Image analysis software. On routine cytological examination, 44 cases were benign, 26 were malignant, and 30 cases were placed in "atypical" category. On morphometry, values for mean nuclear area (MNA) and mean cytoplasmic area (MCA) in benign cases were found to be 58.39 ± 12.22 ,m2 and 185.70 ± 43.69 ,m2, while for malignant cases were 120.12 ± 16.28 ,m2 and 274.65 ± 61.01 ,m2, respectively (P < 0.001, P < 0.001, highly significant). Significant differences were also noted among benign, atypical, and malignant cases for MNA (P < 0.001). The differences between atypical and malignant cases for N/C (nuclear/cytoplasmic) ratio were also found to be statistically significant (P < 0.001). Finally, 46 cases were found to be benign, 39 malignant, and 15 in the category of atypical cases. CIM helps in better and quicker sorting for malignant cells. MNA and N/C ratio are the most significant parameters in differentiating various cells in effusions. MCA constitutes as another useful parameter. Change in supporting tissue following loss of a permanent maxillary incisor in children

Abstract,,, Alveolar bone resorption is an inevitable consequence of tooth loss and may be detrimental to long-term dental aesthetics and function. The aim of the present study was to quantify the degree of tissue resorption following the loss of a permanent incisor in a young population. The study group comprised 11 boys and five girls who all required the extraction of a permanent maxillary central incisor due to trauma-related sequelae. Mean age at tooth loss was 10.8 years. Upper alginate impressions were taken at regular intervals following tooth loss and were cast in yellow dental stone. Study models were sectioned longitudinally through the mid-point of both the maxillary incisor socket and the contra-lateral incisor to provide a thin plaster section. Digital photographs were acquired of the edentulous (A1) and dentate (A2) surfaces of this section and image analysis software was employed to quantify the surface area of both A1 and A2. At 3 months postextraction, mean A1 was 15.7% less than mean A2. By 6 months mean A1 had further reduced and was 25.3% less than that of the corresponding dentate alveolus. However, at subsequent time intervals following tooth extraction (>6 months), tissue loss appeared to stabilise with an overall reduction in tissue area remaining at 22%. This reduction in supporting tissue area was found to be highly statistically significant (P = 0.002, anova). Furthermore, girls appeared to have an overall greater degree of tissue loss than boys (P = 0.015). Further research is indicated to explore factors influencing the degree of tissue loss following incisor extraction and the benefit of therapeutic interventions in limiting this resorption. Proteome analysis of human liver tumor tissue by two-dimensional gel electrophoresis and matrixassisted laser desorption/ionization-mass spectrometry for identification of disease-related proteins

Abstract Hepatocellular carcinoma (HCC) is a common malignancy worldwide and is a leading cause of death. To contribute to the development and improvement of molecular markers for diagnostics and prognostics and of therapeutic targets for the disease, we have largely expanded the currently available human liver tissue maps and studied the differential expression of proteins in normal and cancer tissues. Reference two-dimensional electrophoresis (2-DE) maps of human liver tumor tissue include labeled 2-DE images for total homogenate and soluble fraction separated on pH 3,10 gels, and also images for soluble fraction separated on pH 4,7 and pH 6,9 gels for a more detailed map. Proteins were separated in the first dimension by isoelectric focusing on immobilized pH gradient (IPG) strips, and by 7.5,17.5% gradient sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gels in the second dimension. Protein identification was done by peptide mass fingerprinting with delayed extraction-matrix assisted laser desorption/ionization-time of flight-mass spectrometry (DE-MALDI-TOF-MS). In total, 212 protein spots (117 spots in pH 4,7 map and 95 spots in pH 6,9) corresponding to 127 different polypeptide chains were identified. In the next step, we analyzed the differential protein expression of liver tumor samples, to find out candidates for liver cancer-associated proteins. Matched pairs of tissues from 11 liver cancer patients were analyzed for their 2-DE profiles. Protein expression was comparatively analyzed by use of image analysis software. Proteins whose expression levels were different by more than three-fold in at least 30% (four) of the patients were further analyzed. Numbers of protein spots overexpressed or underexpressed in tumor tissues as compared with nontumorous regions were 9 and 28, respectively. Among these 37 spots, 1 overexpressed and 15 underexpressed spots, corresponding to 11 proteins, were identified. The physiological significance of the differential expressions is discussed. Hippocampal mossy fiber sprouting and elevated trkB receptor expression following systemic administration of low dose domoic acid during neonatal development

Abstract We have previously reported that serial systemic injections of low-dose (subconvulsive) domoic acid (DOM) during early postnatal development produces changes in both behavior and hippocampal cytoarchitecture in aged rats (17 months) that are similar to those seen in existing animal models of temporal lobe epilepsy. Herein we report further hippocampal changes, consisting of mossy fiber sprouting and associated changes in the trkB receptor population in young adult (3 months) rats, and further, report that these changes show regional variation throughout the septo-temporal axis of the hippocampus. Groups of Sprague Dawley rat pups were injected daily from postnatal day 8,14 with either saline (n = 23) or 20 ,g/kg DOM (n = 25), tested for key indicators of neonatal neurobehavioral development, and then left undisturbed until ,90 days of age, at which time brain tissue was removed, hippocampi were dissected, fixed and processed using either Timm's stain to visualize hippocampal mossy fiber sprouting (MFS) or trkB immunohistochemistry to visualize full length trkB receptors. Multiple sections from dorsal, mid, and ventral hippocampus were analyzed separately and all measures were conducted using image analysis software. The results indicate significant increases in MFS in the inner molecular layer in treated animals with corresponding changes in trkB receptor density. Further we identified significant increases in trkB receptor density in the hilus of the dentate gyrus and area CA3 and report increased mossy fiber terminal density in the stratum lucidum in treated rats. The magnitude of these changes differed between sections from dorsal, mid, and ventral hippocampus. We conclude that low dose neonatal DOM produces cytoarchitectural changes indicative of abnormal development and/or synaptic plasticity that are progressive with age and show regional variation within the hippocampal formation. © 2007 Wiley-Liss, Inc. An in vitro investigation of the bulk flow of fluid through apical foramina during simulated tooth extraction: a potential confounder in microbiological studies?

Aim,The ,pumping action' induced during tooth extraction may cause bacteria suspended in tissue fluids to be transposed from one anatomical compartment to another. Apart from causing bacteraemia, this may lead to inaccuracies in studies evaluating the presence and distribution of bacteria in and around tooth apices. The aim was to investigate the bulk flow of fluid through apical foramina during simulated extraction of teeth in an in vitro model. The influence of the presence or absence of a coronal restoration was also evaluated. Methodology,Twenty extracted single-rooted, human, mature, permanent teeth were used. Standard access cavities were prepared and the root canals located. Standardized micrographs of the apical foramina were obtained and their area (µm2) was calculated by image analysis software. The teeth were then set and sealed into polyvinylsiloxane (rubber base) impression material. Crystal violet dye was inoculated into the coronal half of the root canal system. Tooth extraction movements were simulated in the impression matrix and the leakage of dyes with and without the presence of a coronal restoration was examined. The procedure was repeated, following application of safranin dye in a coronal trough within the simulated rubber base gingival margin at the CEJ. The results were analysed statistically with the independent-samples t -test and the McNemar test. Results,In the absence of a coronal restoration crystal violet leaked out of the apical foramina in 18/20 teeth; conversely safranin leaked into the teeth through the apical foramina in 11/20 cases when applied to the external root surface. In the presence of an intact coronal restoration crystal violet dye leaked out in 6/20 teeth and conversely safranin leaked into 7/20 teeth. The presence of a coronal restoration significantly reduced (P = 0.002) dye leakage out of the root canal system. No associations were found for leakage of dye into the root canal system when applied externally. In addition, the amount of dye leakage was positively correlated with the area of the apical foramen in the presence of a coronal restoration (P = 0.009). Conclusion,The presence of a coronal restoration significantly reduced leakage of dye out of the apical foramen. Microbiological studies on root canals and periapical lesions using extracted teeth should take potential contamination from this source into account. [source]


Canal shapes produced sequentially during instrumentation with Quantec SC rotary nickel,titanium instruments: a study in simulated canals

Abstract Aim The aim of this study was to determine the shaping ability of Quantec SC nickel,titanium rotary instruments in simulated root canals. Methodology Forty simulated canals consisting of four different shapes in terms of angle and position of curvature were prepared with Quantec SC instruments. Sequential still images were taken of the canals using a video camera attached to a computer with image analysis software. Images were taken preoperatively, and then after instrument 7 (Size 25, 0.05 taper), instrument 8 (size 25, 0.06 taper), and instrument 10 (size 45, 0.02 taper) were taken to length. Each sequential postoperative image was superimposed individually over the preoperative image in order to highlight the amount and position of material removed during preparation. Results Overall, the mean preparation time to size 10 was 3.6 min with 12 mm canals taking on average less time than 8 mm canals. There was a highly significant difference between the canal types (P < 0.0001). No instruments fractured within the canal or deformed, although one instrument separated from the latch grip. All canals remained patent. Following preparation to size 10, 19 canals (48%) retained their length, eight (20%) lost length, and 13 (32%) gained length; the magnitude of the change in length was always 0.5 mm or below. Following preparation to size 7 instruments all canals showed aberrant shapes. Excess removal of material along the outer aspect of the curve between the beginning of the curve and the end-point (outer widening) was found in 26 canals (65%) after instrument 7. At the same stage of preparation six canals (15%) had zips, three (8%) had ledges and five (13%) had perforations. Following preparation to size 10, 27 (68%) canals were perforated. Conclusions Under the conditions of the study, Quantec SC instruments consistently produced aberrations when canals were enlarged to size 7 (size 25, 0.05 taper) or above. Care should be exercised when using these instruments in real teeth. [source]


Shaping ability of Hero 642 rotary nickel,titanium instruments in simulated root canals: Part 2

Abstract Aim To determine the shaping ability of Hero 642 nickel,titanium rotary instruments during the preparation of simulated canals. Methodology A total of 40 simulated root canals made up of four different shapes, in terms of angle and position of curvature, were prepared by Hero 642 instruments using a crown-down preparation sequence. Pre- and postoperative images of the canals were taken using a video camera attached to a computer with image analysis software. The pre- and postoperative views were superimposed to highlight the amount and position of material removed during preparation. This report describes the efficacy of the instruments in terms of prevalence of canal aberrations, the amount and direction of canal transportation and overall postoperative shape. Results Four zips and four elbows were created during preparation, all in canals with 40°, 12 mm curves. No perforations or danger zones were created. Highly significant differences (P < 0.001) were apparent between the canal shapes in total canal width at the apex and beginning of the curve, and in the amount of resin removed from the inner and outer aspects of the curve at the orifice. Canal transportation was most frequently directed toward the outer aspect of the curve at specific points along the canal, except at the orifice, where it was apparent that canals with 20° curves transported toward the inner. Overall, mean absolute transportation was always less than 0.15 mm; however, significant differences occurred between canal shapes at the end-point (P < 0.01), apex of the curve (P < 0.01) and at the orifice (P < 0.01). Conclusions Under the conditions of this study, Hero 642 rotary nickel,titanium instruments created canals with few aberrations and no perforations. The relatively high proportion of aberrations in canals with short, acute curves may indicate that instruments with increased taper should be used with caution at or near the full working distance. Further research in real teeth is necessary to elucidate the full potential of these new rotary instruments for use in root canal preparation. [source]


The biometry of gills of 0-group European flounder

The gill surface area of 0-group, post-metamorphic Pleuronectes flesus L. was examined using digital image analysis software and expressed in relation to body mass according to the equation log Y=loga+c logW (a=239·02; c=0·723). The components that constitute gill area, total filament length, interlamellar space and unilateral lamellar area were measured. The measurement of the length of every filament on all eight arches showed that commonly used methods of calculation can lead to an under-estimation of up to 24% of total filament length. Direct measurements of unilateral lamellar area with digital image analysis showed that previously reported gill area data for the same species was over-estimated by as much as 58%. In addition, in this species the neglect of gill pouch asymmetry after metamorphosis, can bring about a 14% over-estimation of total gill area. [source]

ABSTRACT: A laboratory experiment is described that has been designed to allow students to gather meaningful structural and mechanical data with limited equipment. Images are acquired using a computer-interfaced flatbed scanner. Although intended for bread, this approach can be applied to other food products as well. This experiment may be as broad or narrow and as complex or simple as desired. Students have the decided advantage of gathering data themselves, not merely viewing a demonstration of expensive research-grade instrumentation. Experience with image analysis software facilitates a better understanding of quantifying structural data than can be obtained from lecture or text material. Students should become aware of the dependence a specific property, texture, on the underlying structure of food materials and gain an appreciation of the role food structure has in determining many quality parameters. Analysis of Particle Size Distribution by Particle Tracking

Abstract Particle tracking is performed using a combination of dark field or fluorescence video microscopy with automatic image analysis. The optical detection together with the image analysis software allows for the time resolved localization of individual particles with diameters between 100 and 1000,nm. Observation of their Brownian motion over a set of time intervals leads to the determination of their mean square displacements under the given room temperature and viscosity. Hereby, the radii of a set of particles visible within a given optical frame are derived simultaneously. Rapid data analysis leads to reliable particle size histograms. The applicability of this method is demonstrated on polystyrene latices and PMMA nanospheres with radii between 51,nm and 202,nm. [source]


Validation of multi-detector computed tomography as a non-invasive method for measuring ovarian volume in macaques (Macaca fascicularis)

Abstract The purpose of this study was to validate low radiation dose, contrast-enhanced, multi-detector computed tomography (MDCT) as a non-invasive method for measuring ovarian volume in macaques. Computed tomography scans of four known-volume phantoms and nine mature female cynomolgus macaques were acquired using a previously described, low radiation dose scanning protocol, intravenous contrast enhancement, and a 32-slice MDCT scanner. Immediately following MDCT, ovaries were surgically removed and the ovarian weights were measured. The ovarian volumes were determined using water displacement. A veterinary radiologist who was unaware of actual volumes measured ovarian CT volumes three times, using a laptop computer, pen display tablet, hand-traced regions of interest, and free image analysis software. A statistician selected and performed all tests comparing the actual and CT data. Ovaries were successfully located in all MDCT scans. The iliac arteries and veins, uterus, fallopian tubes, cervix, ureters, urinary bladder, rectum, and colon were also consistently visualized. Large antral follicles were detected in six ovaries. Phantom mean CT volume was 0.702±SD 0.504,cc and the mean actual volume was 0.743±SD 0.526,cc. Ovary mean CT volume was 0.258±SD 0.159,cc and mean water displacement volume was 0.257±SD 0.145,cc. For phantoms, the mean coefficient of variation for CT volumes was 2.5%. For ovaries, the least squares mean coefficient of variation for CT volumes was 5.4%. The ovarian CT volume was significantly associated with actual ovarian volume (ICC coefficient 0.79, regression coefficient 0.5, P=0.0006) and the actual ovarian weight (ICC coefficient 0.62, regression coefficient 0.6, P=0.015). There was no association between the CT volume accuracy and mean ovarian CT density (degree of intravenous contrast enhancement), and there was no proportional or fixed bias in the CT volume measurements. CT analysis after distraction osteogenesis in Pierre Robin Sequence

Abstract Objectives/Hypothesis: Early mandibular lengthening by distraction osteogenesis provides an alternative to traditional methods of airway management in infants with Pierre Robin sequence (PRS). Little evidence in the medical literature quantitatively demonstrates the changes in skeletal, soft tissue, and hypopharyngeal spaces with mandibular distraction. Study Design: Prospective analysis of a cohort of three patients with PRS. Methods: We reviewed a series of infants with PRS and severe upper airway obstruction who underwent mandibular distraction. The infants underwent mandibular lengthening with the same internal, unidirectional distraction osteogenesis device. Standardized serial computed tomography (CT) scans were obtained according to established protocol. Computed tomography data were extracted and analyzed with medical image analysis software for mandibulo-maxillary arch harmony, symmetry, hypopharyngeal airway volume, geniohyoid distance, distraction osteogenesis bone volume, and mandibular length. Results: Mandibulo-maxillary alveolar ridge distances were corrected to 0.5 mm after distraction. Clinical examination showed good arch harmony without open-bite or cross-bite deformities. Mandibular ramus was lengthened by 19.5%; the body, 43.4%. After distraction, total mandibular length was increased by 26.2%; hypopharyngeal airway volume, 192%; posterior distance from pharyngeal wall to tongue base, 198.9%; and geniohyoid distance, 14.1%. Conclusions: Unidirectional internal microdistractors can achieve good mandibulo-maxillary arch harmony. Hypopharyngeal airway volume increases substantially, with an even greater increase in distance between tongue base and posterior pharyngeal wall. As the distal mandibular segment is distracted, the hyoid moves anteriorly, with minor increase in geniohyoid relationship. Impact of topical anaesthesia on pain alleviation and wound healing in lambs after mulesing

Objective To investigate the impact of using the topical anaesthetic preparation Tri-Solfen® on pain alleviation and wound healing in lambs undergoing mulesing. Design Three separate trials, placebo controlled and/or randomised, were carried out over a 5 month period on three mobs of between 60 and 263 merino lambs undergoing routine mulesing. Procedure Wound pain was assessed using 10 and 75 g calibrated Von-Frey monofilaments to determine sensitivity to light touch and pain stimulation over a 4 to 8 h period. Pain-related behaviour was documented by trained, blinded observers using a numerical rating scale. Wound healing rates were determined using scaled digital photography and image analysis software to calculate contraction in wound surface area 2 and 4 weeks after mulesing. Results There was rapid (3 min) and prolonged (up to 8 h) wound analgesia as shown by pain response scores (P , 0.01), with absent or significantly diminished primary and secondary hyperalgesia (P , 0.01) and significant reduction in pain-related behaviour (P < 0.001) in treated versus untreated lambs. In addition there was improved wound healing in the treated lambs (P , 0.05). Conclusion Tri-Solfen® effects rapid and prolonged wound analgesia, reduction in pain-related behaviour and improved wound healing in lambs undergoing routine mulesing, providing effective alleviation of pain associated with routine mulesing in sheep. Visceral adiposity is closely correlated with neck circumference and represents a significant indicator of insulin resistance in WHO grade III obesity

Summary Objective, Although associations between visceral adiposity (intra-abdominal fat mass) and insulin resistance are well established, previous data include few subjects with WHO grade III obesity [body mass index (BMI) > 40 kg/m2]. We have investigated the relationship between visceral adiposity and insulin resistance using computed tomography (CT)-quantified fat mass and the homeostasis model assessment for insulin resistance (HOMA-IR) in patients with severe obesity. Patients and methods, Eighteen nondiabetic subjects with BMI > 40 kg/m2 were recruited. BMI, and waist, hip and neck circumferences were measured. Fasting plasma insulin and glucose were measured to calculate HOMA-IR. A single slice CT scan was taken at L4 and visceral and abdominal subcutaneous adipose tissue (VAT and ASAT, respectively) quantified using ,SliceOmatic' image analysis software. Results, A close correlation was demonstrated between VAT and HOMA-IR (r2 = 0·46, P = 0·002), whereas ASAT showed no relationship. Neck circumference correlated with both VAT (r2 = 0·67, P < 0·0001) and HOMA-IR (r2 = 0·35, P = 0·01). Waist circumference only correlated significantly with VAT (r2 = 0·25, P = 0·03). Conclusions, Visceral adiposity remains a strongly significant indicator of insulin resistance in WHO grade III obesity. Neck circumference surpasses other anthropometric measurements as a powerful marker of both VAT and insulin resistance. [source]
