Comparison by PAM Fluorometry of Photosynthetic Activity of Nine Marine Phytoplankton Grown Under Identical Conditions,

ABSTRACT The photosynthetic activity of marine phytoplankton from five algal classes (Phaeodactylum tricornutum, Skeletonema costatum, Thalassiosira oceanica, Thalassiosira weissflogii, Dunaliella tertiolecta, Mantoniella squamata, Emiliania huxleyi, Pavlova lutheri and Heterosigma akashiwo) was investigated under identical growth conditions to determine interspecies differences. Primary photochemistry and electron transport capacity of individual species were examined by pulse amplitude-modulated (PAM) fluorescence. Although few differences were found in maximal photosystem II (PSII) photochemical efficiency between various species, large differences were noticed in their PSII-photosystem I (PSI) electron transport activity. We found that species such as T. oceanica and M. squamata have much lower photochemical activity than H. akashiwo. It appeared that processes involved in electron transport activity were more susceptible to change during algal evolution compared with the primary photochemical act close to PSII. Large variations in the nonphotochemical energy dissipation event among species were also observed. Light energy required to saturate photosynthesis was very different between species. We have shown that M. squamata and H. akashiwo required higher light energy (>1300 ,mol m,2 s,1) to saturate photosynthesis compared with S. costatum and E. huxleyi (ca 280 ,mol m,2 s,1). These differences were interpreted to be the result of variations in the size of lightharvesting complexes associated with PSII. These disparities in photosynthetic activity might modulate algal community structure in the natural environment where light energy is highly variable. Our results suggest that for an accurate evaluation of primary productivity from fluorescence measurements, it is essential to know the species composition of the algal community and the individual photosynthetic capacity related to the major phytoplankton species present in the natural phytoplankton assemblage. On sliding mode observers for systems with unknown inputs

Abstract This paper considers the problem of designing an observer for a linear system subject to unknown inputs. This problem has been extensively studied in the literature with respect to both linear and nonlinear (sliding mode) observers. Necessary and sufficient conditions to enable a linear unknown input observer to be designed have been established for many years. One way to express these conditions is that the transfer function matrix between the unknown input and the measured output must be minimum phase and relative degree one. Identical conditions must be met in order to design a ,classical' sliding mode observer for the same problem. This paper shows how the relative degree condition can be weakened if a classical sliding mode observer is combined with sliding mode exact differentiators to essentially generate additional independent output signals from the available measurements. A practical example dedicated to actuator fault detection and identification of a winding machine demonstrates the efficacy of the approach. Copyright © 2007 John Wiley & Sons, Ltd. [source]


Constructive modification of conducting polyaniline characteristics in unusual proportion through nanomaterial blend formation with the neutral polymer poly(vinyl pyrrolidone)

Abstract The present work reports an investigation on the modification of conducting polyaniline (PANI) characteristics favorably on blending with the neutral polymer, poly(vinyl pyrrolidone) (PVP) in a systematic variation of their molar ratios (aniline : PVP = 4 : 1, 2 : 1, 1 : 1, 1 : 2, and 1 : 3). Prepared by precipitation technique by conventional in situ chemical oxidative polymerization with ammonium peroxodisulfate in aqueous H2SO4 medium (pH 1.0), these materials have nanometer sizes (, 50,200 nm) and, depending on the molar ratios, exhibit a distinct deviation in physicochemical characteristics from those of pristine PANI prepared in the identical condition. A gradual trend in characteristics is noticed in first three PANI,PVP blends, while an abnormal hike in conductivity, unusual spectral features in IR and UV,vis, hardened nature, and induction of characteristic morphology, crystallinity, and thermal stability are associated with the last two blends that have excess PVP. Thus a division of two sets of nanoblends, one set with less or equal content of PVP and another with excess of PVP, emerges. Evidently, PVP has a tuning effect on PANI through its dopant, supporting matrix and interpenetrating steric stabilizer acts in proportion quite unusual to its neutral nature. Organizational effects of maternal testosterone on reproductive behavior of adult house sparrows

Abstract Despite the well-known, long-term, organizational actions of sex steroids on phenotypic differences between the sexes, studies of maternal steroids in the vertebrate egg have mainly focused on effects seen in early life. Long-term organizational effects of yolk hormones on adult behavior and the underlying mechanisms that generate them have been largely ignored. Using an experiment in which hand-reared house sparrows (Passer domesticus) from testosterone- or control-treated eggs were kept under identical conditions, we show that testosterone treatment in the egg increased the frequency of aggressive, dominance, and sexual behavior of 1-year-old, reproductively competent house sparrows. We also show that circulating plasma levels of progesterone, testosterone, 5,-dihydrotestosterone, and 17,-estradiol did not differ between treatment groups. Thus, a simple change in adult gonadal hormone secretion is not the primary physiological cause of long-term effects of maternal steroids on adult behavior. Rather, differences in adult behavior caused by exposure to yolk testosterone during embryonic development are likely generated by organizational modifications of brain function. Furthermore, our data provide evidence that hormone-mediated maternal effects are an epigenetic mechanism causing intra-sexual variation in adult behavioral phenotype. © 2008 Wiley Periodicals, Inc. Cerebellar granule cells show age-dependent migratory differences in vitro

Abstract Developmental differences between cerebellar granule cells during their migratory period were revealed using dissociated granule cell cultures isolated from 4, 7, or 10 days old (P4, P7, P10) mice. Under all culture conditions, the great majority of cultivated cell populations consisted of those granule cells that had not reach their final destination in the internal granule cell layer (IGL) by the age of isolation. In vitro morphological development and the expression of migratory markers (TAG-1, astrotactin, or EphB2) showed similar characteristics between the cultures. The migration of 1008 granule cells isolated from P4, P7, and P10 cerebella and cultivated under identical conditions were analyzed using statistical methods. In vitro time-lapse videomicroscopy revealed that P4 cells possessed the fastest migratory speed while P10 granule cells retained their migratory activity for the longest time in culture. Cultures obtained from younger postnatal ages showed more random migratory trajectories than P10 cultures. Our observations indicate that despite similar morphological and molecular properties, migratory differences exist in granule cell cultures isolated from different postnatal ages. Does the diapause experience of bumblebee queens Bombus terrestris affect colony characteristics?

Summary 1. Bumblebee colonies show much variation in the number of workers, drones, and queens produced. Because this variation prevails even when colonies are kept under identical conditions, it does not seem to be caused by extrinsic factors but rather by differences between founding queens. 2. The most likely factor that could cause differences between queens is diapause. Although colonies are raised under standardised conditions, the queens often experience diapause of different length. If there are costs associated with diapause that influence post-diapause reproduction, the diapause history of the queens could affect colony characteristics. 3. Here, several colony characteristics are compared: number of first and second brood workers; total number of workers, drones, and queens; energy spent on sexuals; sex ratio; rate of worker production; time to emergence of first reproductive; and colony lifetime. Colonies were used where the queens experienced a diapause treatment of 0 (nondiapause queens), 2, and 4 months. 4. Although no proof was found for the existence of costs associated with diapause, the colony characteristics of nondiapause queens were significantly different from those of diapause queens. Colonies of nondiapause queens produced the lowest number of workers but the highest number of young queens. 5. It is argued that these nondiapause colonies are more time-constrained than diapause colonies because nondiapause colonies produce two generations within the same season and should therefore be more efficient in producing sexual offspring. 6. Moreover, nondiapause colonies should rear a more female-biased sex ratio because they can be certain of the presence of males produced by other (diapause) colonies. [source]


Genetic variability in a population of arbuscular mycorrhizal fungi causes variation in plant growth

Abstract Different species of arbuscular mycorrhizal fungi (AMF) alter plant growth and affect plant coexistence and diversity. Effects of within-AMF species or within-population variation on plant growth have received less attention. High genetic variation exists within AMF populations. However, it is unknown whether genetic variation contributes to differences in plant growth. In our study, a population of AMF was cultivated under identical conditions for several generations prior to the experiments thus avoiding environmental maternal effects. We show that genetically different Glomus intraradices isolates from one AMF population significantly alter plant growth in an axenic system and in greenhouse experiments. Isolates increased or reduced plant growth meaning that plants potentially receive benefits or are subject to costs by forming associations with different individuals in the AMF population. This shows that genetic variability in AMF populations could affect host-plant fitness and should be considered in future research to understand these important soil organisms. [source]


Sensitive Biomimetic Sensor Based on Molecular Imprinting at Functionalized Indium Tin Oxide Electrodes

Abstract We initially report an electrochemical sensing platform based on molecularly imprinted polymers (MIPs) at functionalized Indium Tin Oxide Electrodes (ITO). In this research, aminopropyl-derivatized organosilane aminopropyltriethoxysilane (APTES), which plays the role of functional monomers for template recognition, was firstly self-assembled on an ITO electrode and then dopamine-imprinted sol was spin-coated on the modified surface. APTES which can interact with template dopamine (DA) through hydrogen bonds brought more binding sites located closely to the surface of the ITO electrode, thus made the prepared sensor more sensitive for DA detection. Potential scanning is presented to extract DA from the modified film, thus DA can rapidly and completely leach out. The affinity and selectivity of the resulting biomimetic sensor were characterized using cyclic voltammetry (CV). It exhibited an increased affinity for DA over that of structurally related molecules, the anodic current for DA oxidation depended on the concentration of DA in the linear range from 2×10,6 M to 0.8×10,3 M with a correlation coefficient of 0.9927. In contrast, DA-templated film prepared under identical conditions on a bare ITO showed obviously lower response toward dopamine in solution. It should be noted that potential scanning is a very effective approach for DA extraction, and surface modification of the electrochemical transducer with functional monomers is responsible for the development of MIPs-based highly sensitive biomimetic sensor. Internal electrolyte temperatures for polymer and fused-silica capillaries used in capillary electrophoresis

Abstract Polymers are important as materials for manufacturing microfluidic devices for electrodriven separations, in which Joule heating is an unavoidable phenomenon. Heating effects were investigated in polymer capillaries using a CE setup. This study is the first step toward the longer-term objective of the study of heating effects occurring in polymeric microfluidic devices. The thermal conductivity of polymers is much smaller than that of fused silica (FS), resulting in less efficient dissipation of heat in polymeric capillaries. This study used conductance measurements as a temperature probe to determine the mean electrolyte temperatures in CE capillaries of different materials. Values for mean electrolyte temperatures in capillaries made of New Generation FluoroPolymer (NGFP), poly-(methylmethacrylate) (PMMA), and poly(ether ether ketone) (PEEK) capillaries were compared with those obtained for FS capillaries. Extrapolation of plots of conductance versus power per unit length (P/L) to zero power was used to obtain conductance values free of Joule heating effects. The ratio of the measured conductance values at different power levels to the conductance at zero power was used to determine the mean temperature of the electrolyte. For each type of capillary material, it was found that the average increase in the mean temperature of the electrolyte (,TMean) was directly proportional to P/L and inversely proportional to the thermal conductivity (,) of the capillary material. At 7.5,W/m, values for ,TMean for NGFP, PMMA, and PEEK were determined to be 36.6, 33.8, and 30.7°C, respectively. Under identical conditions, ,TMean for FS capillaries was 20.4°C. [source]


Toxicity of methyl tert butyl ether to soil invertebrates (springtails: Folsomia candida, Proisotoma minuta, and Onychiurus folsomi) and lettuce (Lactuca sativa)

Abstract Experiments were conducted to assess the toxicity of methyl tert butyl ether (MTBE) to three species of Collembola (Proisotoma minuta, Folsomia candida, and Onychiurus folsomi) and lettuce (Lactuca sativa L.) using an artificial Organization for Economic Cooperation and Development (OECD) soil and field-collected sandy loam and silt loam soil samples. Soil invertebrate tests were carried out in airtight vials to prevent volatilization of MTBE out of the test units and to allow for direct head-space sampling and gas chromatography-mass spectrometry (GC-MS) analysis for residual MTBE. The use of the airtight vial protocol proved to be very successful, in that the measured MTBE concentrations at the beginning of the experiments were within 95% of nominal concentrations. The test methods used in this study could be used to test the toxicity of other volatile organic compounds to Collembola. The soil invertebrates tested had inhibitory concentration (ICx) and lethal concentration (LCx) values that ranged from 242 to 844 mg MTBE/kg dry soil. When the three test species of Collembola were tested under identical conditions in the artificial OECD soil, O. folsomi was the most sensitive collembolan, with a median inhibitory concentration (IC50; reproduction) of 296 mg MTBE/kg dry soil. The most sensitive endpoint for lettuce was an IC50 for root length of 81 mg MTBE/kg dry soil after 5 d of germination in OECD soil. Thermostability of genetically modified sunflower oils differing in fatty acid and tocopherol compositions

Abstract The objective of the study was to investigate the performance at frying temperature of a new sunflower oil with high content of oleic and palmitic acid (HOHPSO) and containing ,-tocopherol as the most abundant natural antioxidant. HOHPSO either containing ,- or ,-tocopherol (HOHPSO-, and HOHPSO-,, respectively) were obtained from genetically modified sunflower seeds and refined under identical conditions. The oil stability against oxidation, as measured by Rancimat at 120,°C, was much higher for the oil containing ,-tocopherol, suggesting the higher effectiveness of ,-tocopherol as compared to ,-tocopherol to delay oxidation. Experiments at high temperature (180,°C) simulating the conditions applied in the frying process clearly demonstrated that, for the same periods of heating, the oil degradation and the loss of natural tocopherol were significantly lower for the oil containing ,-tocopherol. Comparison of different genetically modified sunflower oils with different fatty acid compositions confirmed that oil degradation depended on the fatty acid composition, being higher at a higher degree of unsaturation. However, the loss of tocopherol for a similar level of oil degradation was higher as the degree of unsaturation decreased. Overall, the results showed that HOHPSO-, had a very high stability at frying temperatures and that mixtures of HOHPSO-, and HOHPSO-, would be an excellent alternative to fulfill the frying performance required by the processors and the vitamin,E content claimed by the consumers. How to deal with pitfalls of immunohistochemistry
Abstract Immunohistochemistry is a sensitive and versatile method widely used to investigate the cyto- and chemoarchitecture of the brain. It is based on the high affinity and selectivity of antibodies for a single epitope. However, it is now recognized that the specificity of antibodies needs to be tested in control experiments to avoid false-positive results due to non-specific binding to tissue components or recognition of epitopes shared by several molecules. This ,Technical Spotlight' discusses other pitfalls, which are often overlooked, although they can strongly influence the outcome of immunohistochemical experiments. It also recapitulates the minimal set of information that should be provided in scientific publications to allow proper evaluation and replication of immunohistochemical experiments. In particular, tissue fixation and processing can have a strong impact on antigenicity by producing conformational changes to the epitopes, limiting their accessibility (epitope masking) or generating high non-specific background. These effects are illustrated for an immunoperoxidase staining experiment with three antibodies differing in susceptibility to fixation, using tissue from mice processed under identical conditions, except for slight variations in tissue fixation. In these examples, specific immunostaining can be abolished depending on fixation strength, or detected only after prolonged postfixation. As a consequence, antibody characterization in immunohistochemistry should include their susceptibility towards fixation and determination of the optimal conditions for their use. [source]


Cloning, distribution and functional analysis of the type III sodium channel from human brain

Abstract The type III voltage-gated sodium channel was cloned from human brain. The full-length cDNA has 89% identity with rat type III, and the predicted protein (1951 amino acids) has 55 differences. The expression pattern of human type III mRNA was determined in adult brain tissue and, in contrast to rat, was detected in many regions, including caudate nucleus, cerebellum, hippocampus and frontal lobe. The human type III channel was stably expressed in Chinese hamster ovary (CHO) cells and its biophysical properties compared to the human type II channel using identical conditions. The voltage dependence and kinetics of activation were found to be similar to that of type II. The kinetics of inactivation of the two human subtypes were also similar. However, type III channels inactivated at more hyperpolarized potentials and were slower to recover from inactivation than type II. When expressed in human embryonic kidney (HEK293T) cells, type III channels produced currents with a prominent persistent component, which were similar to those reported for rat type II [Ma et al. (1997) Neuron, 19, 443,452]. However, unlike type II, this was prominent even in the absence of coexpressed G-proteins, suggesting type III may adopt this gating mode more readily. The distinct properties of the channel, together with its wide distribution in adult brain, suggest that in humans, type III may have important physiological roles under normal, and perhaps also pathological conditions. COEVOLUTION OF COLOR PATTERN AND THERMOREGULATORY BEHAVIOR IN POLYMORPHIC PYGMY GRASSHOPPERS TETRIX UNDULATA

Abstract Ectothermic organisms, such as insects and reptiles, rely on external heat sources to control body temperature and possess physiological and behavioral traits that are temperature dependent. It has therefore been hypothesised that differences in body temperature resulting from phenotypic properties, such as color pattern, may translate into selection against thermally inferior phenotypes. We tested for costs and benefits of pale versus dark coloration by comparing the behaviors (i.e., basking duration and bouts) of pygmy grasshopper (Tetrix undulata) individuals exposed to experimental situations imposing a trade-off between temperature regulation and feeding. We used pairs consisting of two full-siblings of the same sex that represented different (genetically coded) color morphs but had shared identical conditions from the time of fertilization. Our results revealed significant differences in behavioral thermoregulation between dark and pale individuals in females, but not in males. Pale females spent more time feeding than dark females, regardless of whether feeding was associated with a risk of either hypothermia or overheating. In contrast, only minor differences in behavior (if any) were evident between individuals that belonged to the same color morph but had been painted black or gray to increase and decrease their heating rates. This suggests that the behavioral differences between individuals belonging to different color morphs are genetically determined, rather than simply reflecting a response to different heating rates. To test for effects of acclimation on behaviors, we used pairs of individuals that had been reared from hatchlings to adults under controlled conditions in either low or high temperature. The thermal regime experienced during rearing had little effect on behaviors during the experiments reported above, but significantly influenced the body temperatures selected in a laboratory thermal gradient. In females (but not in males) preferred body temperature also varied among individuals born to mothers belonging to different color morphs, suggesting that a genetic correlation exists between color pattern and temperature preferences. Collectively, these findings, at least in females, are consistent with the hypothesis of multiple-trait coevolution and suggest that the different color morphs represent alternative evolutionary strategies. Modelling the response of sprinklers in compartment fires

Alpert's expressions are used extensively to calculate the maximum temperature and velocity in the ceiling-jet at any distance, r, from the fire axis. These expressions are valid for unconfined ceilings insofar as the environment outside the ceiling-jet is uniform in temperature and is atmospheric ambient. In compartments the outward flow of hot gases is checked by the presence of bounding walls and so a layer of hot gases is formed in the upper portion of the compartment. The environment outside the ceiling-jet is no longer ambient atmospheric. Expressions due to Alpert becomes unsuitable, unless the effects of the accumulated hot layer are included in these expressions. The effect of the upper layer temperature has been taken by assuming that since the ceiling-jet is completely submerged in the upper hot gas layer, the entrained gases are drawn from this layer only instead of the ambient atmospheric air at T,. Thus T, has been replaced by TL in the Alpert's empirical expression for ceiling-jet temperatures. The proposed equation is then used to calculate the response time of sprinklers fitted in compartments of varying sizes. Calculations have been done for identical conditions to those employed by Evans and compared. Comparison has also been made with Cooper's LAVENT model predictions. The study shows that the present model is capable of predicting the maximum, as well as average, temperatures in the ceiling-jets submerged in the hot gas layer. Magnetic and Structural Investigation of ZnSe Semiconductor Nanoparticles Doped With Isolated and Core-Concentrated Mn2+ Ions

X-Ray magnetic circular dichroism (XMCD) experiments on diluted magnetic semiconductor nanocrystals (2,7,nm) are reported in order to study their local electronic structure and magnetic properties. ZnSe nanoparticles containing either single manganese ions (Mn2+) distributed in the lattice of the entire particle or a MnSe core in the center are prepared using high temperature approaches. The Mn2+ concentration is varied between less than one to several tens of manganese ions per nanocrystal. For all samples it is shown that the Mn2+ is exclusively present in the bulk of ZnSe nanoparticles with no evidence for oxidation to higher Mn-oxidation states. The magnetic ions are highly polarized inside the nanocrystals reaching about 80% of the theoretical value of a pure d5 state under identical conditions for the case of isolated manganese ions. Nanocrystals with a MnSe core ZnSe shell structure reach <50% of this value. Thus, their polarization is significantly more hindered, which is due to the significantly enhanced Mn,Mn interactions and a more distorted crystalline lattice. In contrast, no coupling between the manganese centers is observed in the nanoparticles doped samples with low concentrations of Mn2+, indicating that these ions are isolated in the bulk of the nanoparticles. Converse Bergmann cline in a Eucalyptus herbivore, Paropsis atomaria Olivier (Coleoptera: Chrysomelidae): phenotypic plasticity or local adaptation?

ABSTRACT Aim, To measure latitude-related body size variation in field-collected Paropsis atomaria Olivier (Coleoptera: Chrysomelidae) individuals and to conduct common-garden experiments to determine whether such variation is due to phenotypic plasticity or local adaptation. Location, Four collection sites from the east coast of Australia were selected for our present field collections: Canberra (latitude 35°19, S), Bangalow (latitude 28°43, S), Beerburrum (latitude 26°58, S) and Lowmead (latitude 24°29, S). Museum specimens collected over the past 100 years and covering the same geographical area as the present field collections came from one state, one national and one private collection. Methods, Body size (pronotum width) was measured for 118 field-collected beetles and 302 specimens from collections. We then reared larvae from the latitudinal extremes (Canberra and Lowmead) to determine whether the size cline was the result of phenotypic plasticity or evolved differences (= local adaptation) between sites. Results, Beetles decreased in size with increasing latitude, representing a converse Bergmann cline. A decrease in developmental temperature produced larger adults for both Lowmead (low latitude) and Canberra (high latitude) individuals, and those from Lowmead were larger than those from Canberra when reared under identical conditions. Main conclusions, The converse Bergmann cline in P. atomaria is likely to be the result of local adaptation to season length. [source]

Summary Polymeric immunoglobulins (pIgs) that are present at mucosal surfaces play key roles in both the innate and adaptive immune responses. These pIgs are delivered to the mucosal surface via transcytosis across the epithelium, a process mediated by the polymeric immunoglobulin receptor (pIgR). Previous studies demonstrate that expression of the pIgR is regulated by multiple immunomodulatory factors including interleukin-4 (IL-4) and interferon-, (IFN-,). In studies using human intestinal epithelial cells (HT29), multiple inhibitors of the transcription factor nuclear factor-,B (NF-,B), including a dominant negative I,B,-serine mutant, inhibited both IL-4- and IFN-dependent increases in pIgR expression. Under identical conditions, NF-,B inhibitors had no effect on cytokine-dependent increases in expression of the transcription factor interferon regulatory factor-1. Over-expression of the I,B,-serine mutant also inhibited reporter gene expression in response to IL-4, TNF-,, IL-1,, and in some cases IFN-, using constructs with sequences from the pIgR promoter. Reduced levels of pIgR were observed even when inhibitors were added ,24 hr after cytokines suggesting that prolonged activation of NF-,B is required. Finally, reporter gene studies with NF-,B enhancer elements indicated that IFN-, alone and IL-4 in combination with other cytokines activated NF-,B in HT29 cells. Together, these studies provide additional insight into the signalling pathways that contribute to expression of the pIgR, a critical player in mucosal immunity. [source]

A light-emitting transistor (LEFET) architecture with a distributed feedback (DFB) resonator structure nanoimprinted into the gain medium offers a route to achieving an electrically pumped plastic laser. Effects of food limitation and emigration on self-thinning in experimental minnow cohorts

Summary 1.,The theory of food-regulated self-thinning (FST) for mobile animals predicts population density (N) to be an inverse function of mean body mass (W) scaled to an exponent (b), such that N = k W,b, where k is a constant. FST also predicts energy requirements (or energy flow) to remain constant over time (termed energetic equivalence) as losses to cohorts (e.g. emigration and mortality) are balanced by increased growth of surviving individuals. 2.,To test these predictions, we analysed the dynamics of six experimental minnow cohorts. Replicate populations of fish were held under identical conditions with a constant and limited supply of food over a 126-day period. Half of the cohorts were open to emigration, and half were closed so that fish could only be lost through starvation mortality. 3.,Patterns of self-thinning indicated non-linear changes in population density and energy flow in relation to changes in mean body mass and time, respectively. Non-linear patterns of self-thinning were probably due to a delayed growth response to changes in population density effected through mortality and/or emigration. Contrary to results of similar experiments on other fish, emigration did not have a significant influence on the pattern of self-thinning. 4.,These results may be attributed to trophic interactions within cohorts and the importance of social behaviour to cohort dynamics. Both population density and energy flow in our experimental populations appeared to cycle, with episodes of starvation and mortality alternating with food recovery and weight gain, as predicted by recent models of stepwise die-off and stunted growth in animal cohorts. 5.,Most of the support for FST in mobile animals comes from observational data on mean body mass and population density. Potentially important mechanisms, including the manner in which individuals are lost or retained in populations, are usually not investigated directly. Such tests of FST can only provide equivocal support. Detailed observational study and controlled experiments are needed to understand casual mechanisms. [source]


Oviposition deterrence of shoots and essential oils of Minthostachys spp. (Lamiaceae) against the potato tuber moth

Abstract:, The potato tuber moth is a noxious pest of potato in stores, where the use of repellent plants is an environmentally sound alternative to the application of chemical pesticides. We evaluated the protective effect of native Minthostachys species (Lamiaceae) against tuber infestation by the potato tuber moth in a rustic store in Cusco, Perú. We covered potato tubers with dried shoots of Minthostachys spicata and Minthostachys glabrescens and compared tuber damage with a control treatment of maize straw. We also conducted a no-choice oviposition bioassay in the laboratory, testing the oviposition deterrence of essential oils of M. spicata, M. glabrescens and Minthostachys mollis at natural concentrations. We recorded the number of eggs laid by mated moths on filter paper treated with essential oils of each of the three species and on two control treatments: hexane and untreated blank. Finally, we tested for differences in oviposition deterrence among five full-sib families of potato tuber moth raised under identical conditions. We found that dried, chopped leaves and flowers of Minthostachys species reduced the percentage of tuber damage in stores in comparison with the control (5% vs. 12%), but no difference in protection was found between species. Essential oils at natural concentrations deterred moth oviposition, reducing the number of eggs laid by about 80% compared with the control treatments; again, there were no significant differences between Minthostachys species. Finally, whereas we detected among-family variation in oviposition on filter papers treated with essential oils, no difference was found in the number of eggs laid on control substrates. Therefore, there was genetic variation for oviposition deterrence in the potato tuber moth and resistance to repellent plants might evolve thereafter. [source]


Influence of compatibilizer on notched impact strength and fractography of HDPE,organoclay composites

Abstract The focus of this study was the notched impact property of high-density polyethylene (HDPE),organoclay composites and the resultant morphology of impact-fractured surfaces. Composites with a different organoclay content and degree of organoclay dispersion were compared with neat HDPE under identical conditions. The degree of organoclay dispersion was controlled through the use of a compatibilizer, maleic anhydride grafted polyethylene. It was found that the addition of organoclay can slightly increase the elastic modulus and notched impact strength of the composite. When the level of organoclay dispersion was improved by using compatibilizer, elastic modulus and toughness further increased. A significant increase in yield strength was also notable. The presence of organoclay was found to suppress strain hardening of the matrix during tensile testing. The impact-fractured surfaces of failed specimens were studied with scanning electron microscopy. Ceric ammonium sulfate/sodium disulfite initiated grafting of acrylamide on to Cassia reticulata seed gum

Abstract Ceric ammonium sulfate/sodium disulfite redox system was evaluated for the poly(acrylamide) (PAM) grafting on to Cassia reticulata (CR) seed gum. Grafting conditions were optimized and the maximum %Grafting (%G) and %Efficiency (%E) achieved were 152 and 97.2%, respectively, using [disulfite] 0.005M; [ceric ammonium sulfate] 0.026M; [acrylamide] 0.11M; [gum] 0.125 g/25mL at 40 ± 0.2°C. Representative CR-grafted gum (CRPAM) was characterized by Fourier transform infrared spectrometry (FTIR), X-ray diffraction (XRD), and thermogravimetric analysis (TGA). Under identical conditions, the redox initiator could result 142.6 %G and 91.2 %E on to guar gum (GG). Various physical properties of the CR gum/grafted CR gum, such as viscosity, water retention, and saline retention, were studied and compared with GG/grafted GG to find out the potential industrial use of CR gum and PAM- grafted -CR gum. © 2008 Wiley Periodicals, Inc. Raman spectroscopy for spinline crystallinity measurements.

Abstract Online Raman spectra, obtained at different points along the spinline during the melt spinning of polypropylene homopolymer (hPP) fibers, are presented. The percentage crystallinity corresponding to each spectrum was determined from the normalized intensity of the 809-cm,1 Raman band. A calibration curve for propylene crystallinity was established offline with compression-molded films and fibers spun under different processing conditions. Several hPPs and propylene,ethylene copolymers (with 5,15% ethylene) were used to cover a wide calibration range for propylene crystallinity (9.5,60.9%) with an R2 value of 0.989. This calibration curve was subsequently used to predict the polypropylene crystallinity in the spinline as a function of distance from the spinneret. Under identical conditions of quench and throughput, at a fixed point along the spinline, the overall crystallinity developed in the fiber was found to increase with an increase in the spinning speed. As the spinning speed increased, the point of the onset of crystallization moved closer to the spinneret. The rise in crystallinity was more gradual, at 750 m/min as opposed to 1500 m/min. Increasing the throughput at constant spinning speed was found to decrease the rate of crystallization because of a decrease in the spinline stress. At a fixed distance from the spinneret under identical conditions of quench and spinning speed, fibers spun at a higher throughput showed less overall crystallinity. The onset and rate of crystallization was found to be faster in the lower melt index H502-25RG resin as compared to the 5D49 resin under the spinning conditions explored. The experimental data presented here were used to validate fundamental fiber-spinning models (see part II of this series of articles). The validated models and experimental observations can be used to guide the fiber spinning of isotactic polypropylene for rapid product development. © 2008 Wiley Periodicals, Inc. Moisture absorption behavior of epoxies and their S2 glass composites

Abstract The influence of moisture exposure on the behavior of three toughened epoxy,amine systems (scrimp resins SC11, SC15, and SC79, Applied Poleramic, Inc., Benicia, CA) was investigated. Neat resin samples were conditioned by immersion in distilled water at 71°C and in an environmental chamber at 85% relative humidity and 87.8°C until saturation. The equilibrium weight gain ranged from 1.8 to 3.8% for the resins. The long-chain, low-crosslink-density epoxy system (SC11) absorbed the highest amount of water and was saturated first, and it was followed by the medium-crosslink-density (SC15) and high-crosslink-density materials (SC79). The moisture diffusivity decreased with the increasing crosslink density of the resins. The percentage reduction of the glass-transition temperature (Tg) at equilibrium moisture absorption was highest for the low-crosslink molecule. The percentage reductions for the medium-crosslink and higher crosslink systems were comparable. A net weight loss after drying was observed for the SC11 and SC79 resin systems. Fourier transform infrared analysis confirmed the segment breakage and leaching of molecules from the epoxy,amine network. The effects of moisture cycling on Tg were dependent on the epoxy,amine morphology. During the drying stage, Tg increased to a value higher than that of the unaged dry systems. The S2 glass composite samples were conditioned under identical conditions for the resin system. Pralidoxime and l -lactate effects in vitro on the inhibition of acetylcholinesterase by paraoxon: pralidoxime does not confer superior protection

Abstract Intoxication with the organophosphorus compound paraoxon (POX), an inhibitor of serine hydrolases, is frequent. Although oximes are the only enzyme reactivators presently available, clinical experience with their use was rather disappointing. Recent work has shown that under certain conditions l -lactate is also able to reduce in vitro the POX inhibition of butyrylcholine- and acetylcholineesterase (BChE and AChE). To assess the practical relevance, if any, of these findings, the protective effects of pralidoxime (PRX) and those of lactate had to be compared in the same in vitro model. Effects of PRX on the inhibition of AChE by POX were assessed in vitro in plasma of 12 (six male and six female) healthy human volunteers. The determinations were repeated using different oxime and different POX concentrations. The AChE activity determinations were performed using the following sampler: sample BL,baseline (or untreated plasma); sample a,after addition of POX to plasma (pl + POX); sample b,after POX and plasma were incubated and then oxime was added (pl + POX/PRX); sample c,after addition of oxime to plasma (pl + PRX); sample d,after oxime and plasma were incubated and then POX was added (pl + PRX/POX); sample e,after oxime and POX were incubated and then added to plasma (PRX + POX/pl). Results were corrected for spurious enzyme ,pseudo-activity' due to interaction between PRX and substrate (acetylthiocholine) in the absence of enzyme. In the micro- and millimolar ranges, PRX is able to protect in vitro AChE from inhibition by POX when added to human plasma prior to POX or when incubated with POX prior to addition to plasma. Adding PRX to plasma after POX has no protective effect. The PRX results were compared statistically with historical lactate data (obtained under identical conditions) using the Wilcoxon matched pairs test, with significance assumed for p = 0.01. No difference between PRX and lactate's protective effect on the AChE inhibition by POX was found in the in vitro model used. O -acetylation of GD3 prevents its apoptotic effect and promotes survival of lymphoblasts in childhood acute lymphoblastic leukaemia

Abstract We have previously demonstrated induction of O -acetylated sialoglycoproteins on lymphoblasts of childhood acute lymphoblastic leukaemia (ALL). These molecules promote survival of lymphoblasts by preventing apoptosis. Although O -acetylated sialoglycoproteins are over expressed, the status of O -acetylation of gangliosides and their role in lymphoblasts survival remains to be explored in ALL patients. Here, we have observed enhanced levels of 9- O -acetylated GD3 (9- O -AcGD3) in the lymphoblasts of patients and leukaemic cell line versus disialoganglioside GD3 in comparison to the normal cells. Localization of GD3 and 9- O -AcGD3 on mitochondria of patient's lymphoblasts has been demonstrated by immuno-electron microscopy. The exogenous administration of GD3-induced apoptosis in lymphoblasts as evident from the nuclear fragmentation and sub G0/G1 apoptotic peak. In contrast, 9- O -AcGD3 failed to induce such apoptosis. We further explored the mitochondria-dependent pathway triggered during GD3-induced apoptosis in lymphoblasts. GD3 caused a time-dependent depolarization of mitochondrial membrane potential, release of cytochrome c and 7.4- and 8-fold increased in caspase 9 and caspase 3 activity respectively. However, under identical conditions, an equimolar concentration of 9- O -AcGD3 failed to induce similar effects. Interestingly, 9- O -AcGD3 protected the lymphoblasts from GD3-induced apoptosis when administered in equimolar concentrations simultaneously. In situ de- O -acetylation of 9- O -AcGD3 with sodium salicylate restores the GD3-responsiveness to apoptotic signals. Although both GD3 and 9- O -acetyl GD3 localize to mitochondria, these two structurally related molecules may play different roles in ALL-disease biology. Taken together, our results suggest that O -acetylation of GD3, like that of O -acetylated sialoglycoproteins, might be a general strategy adopted by leukaemic blasts towards survival in ALL. J. Plasma cortisol and metabolite level profiles in two isogenic strains of common carp during confinement

A rapid increase in common carp Cyprinus carpio plasma cortisol levels was noted, in two experiments, after 30 mins of a 3 h net confinement, which was sustained while the fish were held in the nets. After release from the nets, cortisol levels returned to control values in 1 h. Plasma glucose and free fatty acid levels were elevated by the confinement. Glucose was increased after 30 min but returned to basal levels after 22 h of recovery while free fatty acids were not elevated until 3 h of confinement and remained high for the duration of the recovery period. After confinement for 3 h, plasma lactate levels were reduced and remained low for a further 1 h. No change in either plasma triglyceride or cholesterol levels were found during the study. Confinement had no effect on haematocrit levels but blood haemoglobin levels were reduced. In both experiments hypochloraemia occurred in response to confinement. However, values returned to pre-confinement levels 22 h after confinement. These results show that rearing isogenic carp strains, under identical conditions, results in a reproducable response to an acute stressor and that these carp respond in a similar manner to other teleost species. SUPERHEATED STEAM-DRYING OF MATE LEAVES AND EFFECT OF DRYING CONDITIONS ON THE PHENOL CONTENT

ABSTRACT Four drying experiments of mate leaves (Ilex paraguariensis St. Hilaire) were carried out in a packed bed superheated steam dryer by varying outlet steam temperature from 120 to 140C and equivalent particle diameter from 4.7 × 10,3 to 6.95 × 10,3 m. The influence of these variables on the drying coefficient calculated by assuming a simplified drying kinetic model was investigated. A classical statistical approach revealed the significant effect of both factors on this parameter. Two additional drying runs were performed at identical conditions with conventional hot air and low pressure superheated steam, respectively. The influence of drying atmosphere on the total content of phenols was evaluated. A conventional process of extraction by using an aqueous methanol solution was adopted. Analyses for total phenols were performed by spectrophotometry at 715 nm by applying Folin-Denis assay. The leaves dried with superheated steam had approximately 47% higher retention of these compounds. [source]


COMPUTATIONAL FLUID DYNAMICS MODELING OF FLUID FLOW IN HELICAL TUBES

ABSTRACT The effect of different processing parameters on the degree of mixing and axial and radial pressure drops, during single-phase flow in helical tubes was investigated by using CFD software. Correlations were developed to calculate axial and radial pressure drops, and also the ratio of maximum to average fluid velocities. All of these quantities were found to be dependent on curvature ratio (ratio of tube diameter to coil diameter). Flow visualization experiments were performed to assess the degree of mixing in different configurations. At identical conditions, the degree of mixing was higher in the system with the large curvature ratio, which is in agreement with the simulation results. A minimum ratio of maximum to average fluid velocities of 1.61 was achieved, representing a 20% reduction in hold tube length for Newtonian fluid in laminar flow. [source]
