Selected AbstractsFREQUENCY AND SPATIAL PATTERNING OF CLONAL REPRODUCTION IN LOUISIANA IRIS HYBRID POPULATIONSEVOLUTION, Issue 1 2000John M. Burke Abstract., The plant genera in which natural hybridization is most prevalent tend to be outcrossing perennials with some mechanism for clonal (i.e., asexual) reproduction. Although clonal reproduction in fertile, sexually reproducing hybrid populations could have important evolutionary consequences, little attention has been paid to quantifying this parameter in such populations. In the present study, we examined the frequency and spatial patterning of clonal reproduction in two Louisiana iris hybrid populations. Allozyme analysis of both populations revealed relatively high levels of genotypic diversity. However, a considerable amount of clonality was apparent. Nearly half of all genets (47%) in one population and more than half (61%) in the other had multiple ramets. Furthermore, both populations exhibited relatively high levels of genetic structuring, a pattern that resulted from the aggregation of clonal ramets. The occurrence of clonal reproduction in hybrid populations could not only facilitate introgression through an increase in the number of flowering ramets per genet and/or the survivorship of early generation hybrids, but might also influence the mating system of such populations. Any potential increase in the selfing rate due to cross-pollination among ramets of the same genet may, in turn, increase the likelihood of homoploid hybrid speciation. [source] Allochronic differentiation among Daphnia species, hybrids and backcrosses: the importance of sexual reproduction for population dynamics and genetic architectureJOURNAL OF EVOLUTIONARY BIOLOGY, Issue 2 2004T. Jankowski Abstract Seasonal dynamics of the abundance, sexual reproduction and genetic architecture in a Daphnia hyalina-galeata hybrid complex were studied in the large and deep Lake Constance. We found evidence for the occurrence of first and second order hybridization. Our study revealed strong differences between the parental species not only regarding their seasonal dynamics, genetic architecture and diversity, but also their sexual reproductive behaviour. The overwintering D. hyalina showed low genetic diversity, no genetic differentiation during the season, and reproduced sexually in autumn, whereas D. galeata reached higher levels of genetic diversity, reproduced sexually in early summer, and exhibited changes in genetic structure during the season, but was only present from spring to autumn. However, in both species sexual reproduction was a rare event, and daphnids, including hybrids, reproduced predominantly asexually. This allows long-term persistence of hybrids as well without continuing hybridization events. Within all variables studied, F1 and F2 hybrids showed an intermediate pattern, whereas proposed backcross hybrids were more similar to their respective parentals. These differences in phenotype as well as significant differences in pairwise Fst values between parentals suggest that gene flow seems to be relatively low in the Lake Constance hybrid system. We found evidence for unidirectional introgression by backcrossing from D. galeata to D. hyalina and found a decrease in at least one of the proposed introgressed alleles in the hyalina -backcross while the season progressed. Our findings suggest allochronic differentiation within this hybrid population and different microevolutionary trajectories of the parental species, which will be discussed in the light of the ongoing reoligotrophication process of Lake Constance. [source] Variable number of tandem repeats in the growth hormone gene of Sparus aurata: association with growth and effect on gene transcriptionJOURNAL OF FISH BIOLOGY, Issue 2004R. Almuly The GH gene of Sparus aurata(saGH) contains variable number of tandem repeats (VNTR). The hyper-variable minisatellites in the first and third introns segregate in a Mendelian manner and exhibit numerous alleles. Analysis by PCR and sequencing of the two introns in several wild Sparidae species revealed comparable minisatellites with some variations. ,Zoo blot' with the first intron unit as a probe showed this sequence to be characteristic of several families from the Perciformes order. Unexpectedly, a similar minisatellite was found in the first intron of the GH gene in flounder, which belongs to a different order. Transfection of constructs containing a reporter gene and first intron of different length to four cell lines resulted in an inhibitory effect of the longer intron relative to the short intron. A (CA)n microsatellite (saGHpCA) is found in the GH promoter. A similar repeat at the same location is present in GH promoters of several other fish species. High variability (11 alleles) of the saGHpCA was found in a hatchery population. Full-sib family genotyping showed a Mendelian inheritance of these alleles. A significant association was found between allele distribution and body mass in large and average size fishes from a hatchery population. The intron minisatellites may serve as markers for hybrid population and parental assignment. Its presence in families and orders of the higher teleosts may help solving classification uncertainties. Their conservation and inhibitory effect suggest a biological role. The saGHpCA is correlated with growth and may be a good candidate for predicting growth performance. [source] Intimately linked or hardly speaking?MOLECULAR ECOLOGY, Issue 3 2001The relationship between genotype, environmental gradients in a Louisiana Iris hybrid population Abstract Several models of hybrid zone evolution predict the same spatial patterns of genotypic distribution whether or not structuring is due to environment-dependent or -independent selection. In this study, we tested for evidence of environment-dependent selection in an Iris fulva×Iris brevicaulis hybrid population by examining the distribution of genotypes in relation to environmental gradients. We selected 201 Louisiana Iris plants from within a known hybrid population (80 m × 80 m) and placed them in four different genotypic classes (I. fulva, I. fulva -like hybrid, I. brevicaulis -like hybrid and I. brevicaulis) based on seven species-specific random amplified polymorphic DNA (RAPD) markers and two chloroplast DNA haplotypes. Environmental variables were then measured. These variables included percentage cover by tree canopy, elevation from the high water mark, soil pH and percentage soil organic matter. Each variable was sampled for all 201 plants. Canonical discriminant analysis (CDA) was used to infer the environmental factors most strongly associated with the different genotypic groups. Slight differences in elevation (,0.5 m to +0.4 m) were important for distinguishing habitat distributions described by CDA, even though there were no statistical differences between mean elevations alone. I. brevicaulis occurred in a broad range of habitats, while I. fulva had a narrower distribution. Of all the possible combinations, I. fulva -like hybrids and I. brevicaulis -like hybrids occurred in the most distinct habitat types relative to one another. Each hybrid class was not significantly different from its closest parent with regard to habitat occupied, but was statistically unique from its more distant parental species. Within the hybrid genotypes, most, but not all, RAPD loci were individually correlated with environmental variables. This study suggests that, at a very fine spatial scale, environment-dependent selection contributed to the genetic structuring of this hybrid zone. [source] Mapping polygenes for tuber resistance to late blight in a diploid Solanum phureja × S. stenotomum hybrid populationPLANT BREEDING, Issue 4 2006I. Simko Abstract Potato tuber blight is a disease caused by the oomycete Phytophthora infestans (Mont.) de Bary. Due to the significant economic impact of this disease, introgression of durable resistance into the cultivated potato is one of the top priorities of breeding programmes worldwide. Though numerous resistance loci against this devastating disease have already been mapped, most of the detected loci are contributing towards foliar resistance while specific information on tuber resistance is limited. To identify the genetic components of tuber resistance and its relationship to foliar resistance and plant maturity we have investigated the host-pathogen interaction in a segregating diploid hybrid Solanum phureja × S. stenotomum family. Mature tubers from this mapping family were inoculated with a sporangial suspension of P. infestans (US-8 clonal lineage) and evaluated for lesion expansion. No significant correlation was detected between late blight resistance in foliage and tubers, and between plant maturity and tuber resistance. Four chromosomal regions were significantly associated with tuber resistance to the disease. The largest effect was detected near the marker locus PSC (LOD 10.7) located on chromosome 10. This locus explained about 63% of the total phenotypic variation of the trait. The other three resistance-related loci were mapped on chromosomes 8 (GP1282, LOD 4.4), 6 (CP18, LOD 4.0) and 2 (CP157, LOD 3.8). None of the four tuber resistance loci coincides with the foliage resistance loci detected in this same family. Tuber blight resistance quantitative trait loci (QTL) on chromosomes 2, 8 and 10 are distinct from the maturity QTLs and have an additive effect on tuber resistance. These results indicate that different genes are involved in foliar and tuber resistance to P. infestans in the present family and that some of the resistance genes might be associated with late maturity. [source] Evaluation and analysis of reduction of late blight disease in a diploid potato progenyPLANT PATHOLOGY, Issue 3 2001B. R. Trognitz The rate of late blight disease was analysed for individuals of a diploid Solanum phureja,Solanum tuberosum dihaploid hybrid population (PD), using three different assessment techniques, in the laboratory, screenhouse, and field. These hybrids expressed low disease rates in the field, comparable to resistance based on intact R genes. However, none of the parents of PD expressed any R genes and the pattern of segregation within the PD population was not indicative of R-gene inheritance. The foliage (or leaflet) area diseased had the largest broad-sense heritability of all criteria analysed, in all tests performed. In the field evaluations, the PD population showed intermediate levels of broad-sense heritability for foliage area diseased, relative to the much larger heritability detected for the group of controls possessing R genes. Resistance in the field of the PD hybrids had very little genotype-environment (G × E) interaction, indicating stability of its expression. All genotypes without R genes exhibited heritable, reduced rates of late blight disease in the field, but they were susceptible with low heritability in screenhouse and laboratory tests. This differential expression of disease indicates that the plants' indirect response to unknown environmental stress in the field may have been measured. The value of the PD hybrids for breeding of late blight resistant potato and the use of the disease rate data for detection of the underlying quantitative trait loci are discussed. [source] Fitness consequences of floral variation in Senecio jacobaea (Asteraceae): evidence from a segregating hybrid population and a resource manipulation experimentBIOLOGICAL JOURNAL OF THE LINNEAN SOCIETY, Issue 1 2001STEFAN ANDERSSON The present study examines some of the ecological factors that might exert selection on floral morphology in Senecio jacobaea, a self-sterile composite which exhibits geographic variation in the frequency of rayed and discoid individuals. Regression analyses of phenotypic data from a large, segregating hybrid population, established in a semi-natural (garden) environment and studied over a 2-year period, revealed a negative relationship between the size of the rays and the average germination rate of the maternal seed crop, a pattern that can be attributed to the reduced germination speed of achenes from ray florets. There was no effect of ray size on the amount of cross-pollination achieved, the proportion of heads infested by larvae of seed flies (Pegohylemyia) and the amount of resources retained for the next flowering season. The lack of resource costs was also apparent in a manipulation experiment with greenhouse-grown plants of the rayed phenotype: artificial removal of all rays at the early bud or flowering stage had no detectable effect on subsequent flower and fruit development, regardless of whether the plants experienced high or low water stress. Given these and other observations, I hypothesize that plant-animal interactions and resource costs sometimes play a minor role in exerting selection on flower morphology and that spatially varying selection on germination behaviour accounts for some of the morph frequency variation in S. jacobaea. [source] CROSSING RELATIONSHIPS AMONG ANCIENT AND EXPERIMENTAL SUNFLOWER HYBRID LINEAGESEVOLUTION, Issue 3 2000Loren H. Rieseberg Abstract., Reproductive barrier formation between newly derived hybrid taxa and their parental species represents a major evolutionary hurdle. Here, I examine the development of a sterility barrier during hybrid speciation by examining the fertility of progeny from all combinations of crosses involving three experimentally synthesized sunflower hybrid lineages, their natural hybrid counterpart, Helianthus anomalus, and their parents, H. annuus and H. petiolaris. Crosses between the parental species and H. anomalus generated almost completely sterile offspring (pollen viability < 5%; seed set < 1%). A fairly strong sterility barrier also has developed between three hybrid lineages and both parental species (pollen viability 11.1,41.6%; seed set 0.84,20.1%). In contrast, the three hybrid lineages are almost fully interfertile (pollen viabilities 83.1,88.6%; seed set 72.1,75.3%), as predicted by molecular mapping studies that indicate they have converged on a similar set of gene combinations and chromosomal rearrangements. A modest decline in compability is observed in crosses between the three hybrid lineages and H. anomalus (pollen viabilities 64.1,70.7%; seed set 37,43%), a result that agrees well with prior data demonstrating significant congruence between the genomes of the natural and experimental hybrid lineages. These observations not only indicate that reproductive isolation can arise as a by-product of fertility selection in hybrid populations, but also testify to the repeatability of this mode of speciation. Campbell Abstract When species hybridize, offspring typically exhibit reduced fitness and maladapted phenotypes. This situation has biosafety implications regarding the unintended spread of novel transgenes, and risk assessments of crop-wild hybrids often assume that poorly adapted hybrid progeny will not evolve adaptive phenotypes. We explored the evolutionary potential of early generation hybrids using nontransgenic wild and cultivated radish (Raphanus raphanistrum, Raphanus sativus) as a model system. We imposed four generations of selection for two weedy traits , early flowering or large size , and measured responses in a common garden in Michigan, USA. Under selection for early flowering, hybrids evolved to flower as early as wild lineages, which changed little. These early-flowering hybrids also recovered wild-type pollen fertility, suggesting a genetic correlation that could accelerate the loss of crop traits when a short life cycle is advantageous. Under selection for large size at reproduction, hybrids evolved longer leaves faster than wild lineages, a potentially advantageous phenotype under longer growing seasons. Although early generation hybrid offspring have reduced fitness, our findings provide novel support for rapid adaptation in crop-wild hybrid populations. Biosafety risk assessment programs should consider the possibility of rapid evolution of weedy traits from early generations of seemingly unfit crop-wild hybrids. FITZPATRICK Abstract Patterns of divergence and polymorphism across hybrid zones can provide important clues as to their origin and maintenance. Unimodal hybrid zones or hybrid swarms are composed predominantly of recombinant individuals whose genomes are patchworks of alleles derived from each parental lineage. In contrast, bimodal hybrid zones contain few identifiable hybrids; most individuals fall within distinct genetic clusters. Distinguishing between hybrid swarms and bimodal hybrid zones can be important for taxonomic and conservation decisions regarding the status and value of hybrid populations. In addition, the causes of bimodality are important in understanding the generation and maintenance of biological diversity. For example, are distinct clusters mostly reproductively isolated and co-adapted gene complexes, or can distinctiveness be maintained by a few ,genomic islands' despite rampant gene flow across much of the genome? Here we focus on three patterns of distinctiveness in the face of gene flow between gartersnake taxa in the Great Lakes region of North America. Bimodality, the persistence of distinct clusters of genotypes, requires strong barriers to gene flow and supports recognition of distinct specialist (Thamnophis butleri) and generalist (Thamnophis radix) taxa. Concordance of DNA-based clusters with morphometrics supports the hypothesis that trophic morphology is a key component of divergence. Finally, disparity in the level of differentiation across molecular markers (amplified fragment length polymorphisms) indicates that distinctiveness is maintained by strong selection on a few traits despite high gene flow currently or in the recent past. HALFHILL Abstract Concerns exist that transgenic crop × weed hybrid populations will be more vigorous and competitive with crops compared with the parental weed species. Hydroponic, glasshouse, and field experiments were performed to evaluate the effects of introgression of Bacillus thuringiensis (Bt) cry1Ac and green fluorescent protein (GFP) transgenes on hybrid productivity and competitiveness in four experimental Brassica rapa × transgenic Brassica napus hybrid generations (F1, BC1F1, BC2F1 and BC2F2). The average vegetative growth and nitrogen (N) use efficiency of transgenic hybrid generations grown under high N hydroponic conditions were lower than that of the weed parent (Brassica rapa, AA, 2n = 20), but similar to the transgenic crop parent, oilseed rape (Brassica napus, AACC, 2n = 38). No generational differences were detected under low N conditions. In two noncompetitive glasshouse experiments, both transgenic and nontransgenic BC2F2 hybrids had on average less vegetative growth and seed production than B. rapa. In two high intraspecific competition field experiments with varied herbivore pressure, BC2F2 hybrids produced less vegetative dry weight than B. rapa. The competitive ability of transgenic and nontransgenic BC2F2 hybrids against a neighbouring crop species were quantified in competition experiments that assayed wheat (Triticum aestivum) yield reductions under agronomic field conditions. The hybrids were the least competitive with wheat compared with parental Brassica competitors, although differences between transgenic and nontransgenic hybrids varied with location. Hybridization, with or without transgene introgression, resulted in less productive and competitive populations. [source] Anténor Firmin: Haitian Pioneer of AnthropologyAMERICAN ANTHROPOLOGIST, Issue 3 2000Carolyn Fluehr-Lobban Anténor Firmin published De I'Égalité des Races Humaines in 1885 in Paris as a response both to Arthur de Gobineau's racist tome L'lnégalité des Races (1853-55) and to the racialist anthropology of the nineteenth century. This pioneering work of anthropology has been translated for the first time into English by Assclin Charles as The Equality of the Human Races (Firmin [1885]2000). In 662 pages of the original text, Firmin systematically critiqued the anthropometry and craniometry that dominated the anthropology of his day, while he envisioned a broad, synthetic discipline that would follow once this narrow approach to the study of man was abandoned. He challenged virtually every extant racial myth and laid a basis for the understanding of human variation as adaptation to climate and environment. Contrary to the polygenist doctrines of the infertility of interracial matings, Firmin extolled the value of racial mixture, especially in the vigorous New World hybrid populations. He developed a critical view of racial classifications and of race that foreshadowed much later social constructions of race. In the book he also articulated early Pan-Africanist ideas as well as an analytical framework for what would become postcolonial studies. The Equality of the Human Races is a text that lies historically at the foundations of the birth of the discipline of anthropology, yet it is unknown to the field. It is a pioneering work in critical anthropology that awaits recognition 115 years after it was first published. [Anténor Firmin, history of racism, antiracism, historical texts, Haitian anthropologist, critical anthropology, nineteenth-century pioneer] [source] Viability and bar expression are negatively correlated in Oregon Wolfe Barley Dominant hybridsPLANT BIOTECHNOLOGY JOURNAL, Issue 3 2007Phil Bregitzer Summary The expression level of bar, which encodes phosphinothricin acetyltransferase (PAT), was correlated with the inviability of barley hybrids between 20 Golden Promise-derived transgenic lines (Ds-bar lines) and a specialized genetic marker stock, Oregon Wolfe Barley Dominant (OWBD). Each Ds-bar line was homozygous for a modified maize Ds element that encoded bar and that had been delivered via transposition to a unique location. All Ds-bar lines were viable and morphologically similar. Only four of the 20 hybrid populations were viable. The remaining populations died prior to producing seed. Phenotypic, enzyme-linked immunosorbent assay and quantitative reverse transcriptase-polymerase chain reaction analyses of these lines, and of lines from unrelated transformation events that also expressed bar, showed that viability was negatively correlated with bar expression. Analysis of crosses of a high- bar -expressing line with the OWB mapping population showed that the sensitivity of OWBD to PAT segregated as a single locus on chromosome 6HL. No sensitivity to PAT could be detected in several other lines and cultivars. OWBD has been shown to be genetically divergent from other germplasm groups within cultivated barley; therefore, the observed sensitivity may be peculiar to OWBD and thus would not impact generally on the utility of bar as a selectable marker or source of herbicide resistance in barley. Nevertheless, these results demonstrate the extent of allelic variability present in Hordeum vulgare, and suggest an additional variable for consideration when devising protocols for the transformation of Hordeum cultivars or landraces that are not known to be tolerant to PAT. [source] Milk protein polymorphisms in cattle (Bos indicus), mithun (Bos frontalis) and yak (Bos grunniens) breeds and their hybrids indigenous to BhutanANIMAL SCIENCE JOURNAL, Issue 5 2010Tashi DORJI ABSTRACT In the current study, milk protein variation was examined in cattle (Bos indicus), mithun (Bos frontalis), yak (Bos grunniens) and their hybrid populations in Bhutan to estimate genetic variability, conduct genetic characterization and assess the possibility of gene flow between mithun and cattle. Isoelectric focusing of 372 milk samples from 11 populations detected four molecular types of ,- lactoglobulin (A, B, E and M), five molecular types of ,S1 -casein (A, B, C, E and X) and three molecular types of k -casein (A, B and X). Mithun and yak shared alleles but were found to exhibit different allele frequencies for the proteins studied. The degree of genetic variability within populations was measured by average heterozygosity and ranged from 24,40% in cattle, 26% for yak and 33% for mithun. We also resolved the traditional mithun and cattle hybridization system via principal component analysis. Our results suggested secondary introgression of mithun genes to the village Thrabum population, and a close genetic relationship between Bhutanese indigenous cattle and Indian cattle. [source] |
