Retinal capillary basement membrane thickness in diabetic mice genetically modified at the haptoglobin locus

Abstract Background Individuals with diabetes mellitus (DM) homozygous for the haptoglobin (Hp) 1 allele are at decreased risk of retinopathy as compared to DM individuals with the Hp 2 allele. We sought to recapitulate these findings in DM mice genetically modified at the Hp locus. Methods An early morphological characteristic of the microangiopathy seen in diabetic retinal disease is retinal capillary basement membrane (RCBM) thickening. RCBM thickness as assessed by electron microscopy was performed on a total of 12 eyes taken from three mice in each of the four study groups (three eyes from C57Bl/6 Hp 1 and C57Bl/6 Hp 2 mice with and without streptozotocin-induced diabetes). Results The non-parametric Kruskal,Wallis ANOVA test demonstrated that there was a highly significant difference between the four groups of mice (P < 0.0001). Mann,Whitney tests for specific pair-wise comparisons demonstrated that there was no significant difference in the RCBM thickness between Hp 1 and Hp 2 mice (p = 0.70) or between DM Hp 1 and non-DM Hp 1 mice (p = 0.42). However, induction of diabetes resulted in a marked increase in RCBM thickness in Hp 2 mice compared to non-DM Hp 2 mice (p = 0.0004) and compared to DM Hp 1 mice (p = 0.0005). Conclusions A highly significant increase in RCBM thickness was observed in DM mice with the Hp 2 genotype. These data provide important support for association studies done in humans showing an increased prevalence of diabetic retinopathy in individuals with the Hp 2 genotype. Quantitative analysis of argyrophilic nuclear organizer regions in giant cell lesions of jaws

J Oral Pathol Med (2010) 39: 431,434 Background:, Giant cell lesions of the jaws are considerably similar according to histopathologic characteristics yet show different clinical behaviors. These lesions include central giant cell granuloma (CGCG), aneurysmal bone cyst, Cherubism, and Brown tumor associated with hyperparathyroidism. The present study aimed to investigate AgNORs count in these lesions as a proliferative marker and to determine whether it can be used to discriminate between them or not. Methods:, Forty-one cases of giant cell lesions of jaws were retrived from Oral Pathology Department (1987,2007). They included 21 cases of CGCG, eight cases of aneurysmal bone cyst (ABC), six cases of Cherubism, six cases of Brown tumor. The mean AgNORs count was calculated for all cases. To compare mean AgNORs in groups of lesions, ANOVA test was performed. Results:, Mean AgNOR counts were: (0/85 ± 0/29) in CGCG, (0/76 ± 0/32) in ABC (0/87 ± 0/10) in Cherubism and (0/82 ± 0/16) in Brown tumor. A significant difference was not observed in AgNOR counts among these groups of lesions. Conclusions:, Jaws giant cell containing lesions have no acceptable differences in mean AgNORs. [source]


In healthy subjects without knee osteoarthritis, the peak knee adduction moment influences the acute effect of shoe interventions designed to reduce medial compartment knee load

Abstract The purpose of this study was to evaluate shoe sole material stiffness changes and angle changes that are intended to reduce the peak knee adduction moment during walking. Fourteen physically active adults were tested wearing their personal shoes (control) and five intervention pairs, two with stiffness variations, two with angle variations, and a placebo shoe. The intervention shoes were evaluated based on how much they reduced the peak knee adduction moment compared to the control shoe. An ANOVA test was used to detect differences between interventions. Linear regression analysis was used to determine a relationship between the magnitude of the knee adduction moment prior to intervention and the effectiveness of the intervention in reducing the peak knee adduction moment. Peak knee adduction moments were reduced for the altered stiffness and altered angle shoes (p,<,0.010), but not for the placebo shoe (p,=,0.363). Additionally, linear regression analysis showed that subjects with higher knee adduction moments prior to intervention had larger reductions in the peak knee adduction moment (p,<,0.010). These results demonstrate that shoe sole stiffness and angle interventions can be used to reduce the peak knee adduction moment and that subjects with initially higher peak knee adduction moments have higher reductions in their peak knee adduction moments. © 2007 Orthopaedic Research Society. Confocal Examination of Subsurface Cracking in Ceramic Materials

Abstract Purpose: The original ceramic surface finish and its microstructure may have an effect on crack propagation. The purpose of this study was to investigate the relation between crack propagation and ceramic microstructure following cyclic fatigue loading, and to qualitatively evaluate and quantitatively measure the surface and subsurface crack depths of three types of ceramic restorations with different microstructures using a Confocal Laser Scanning Microscope (CLSM) and Scanning Electron Microscope (SEM). Materials and Methods: Twenty (8 × 4 × 2 mm3) blocks of AllCeram (AC), experimental ceramic (EC, IPS e.max Press), and Sensation SL (SSL) were prepared, ten glazed and ten polished of each material. Sixty antagonist enamel specimens were made from the labial surfaces of permanent incisors. The ceramic abraders were attached to a wear machine, so that each enamel specimen presented at 45 degrees to the vertical movement of the abraders, and immersed in artificial saliva. Wear was induced for 80K cycles at 60 cycles/min with a load of 40 N and 2-mm horizontal deflection. The specimens were examined for cracks at baseline, 5K, 10K, 20K, 40K, and 80K cycles. Results: Twenty- to 30-,m deep subsurface cracking appeared in SSL, with 8 to 10 ,m in AC, and 7 ,m close to the margin of the wear facets in glazed EC after 5K cycles. The EC showed no cracks with increasing wear cycles. Seventy-,m deep subsurface cracks were detected in SSL and 45 ,m in AC after 80K cycles. Statistically, there was significant difference among the three materials (p < 0.05). Bonferroni multiple comparison of means test confirmed the ANOVA test and showed that there was no statistical difference (p > 0.05) in crack depth within the same ceramic material with different surface finishes. Conclusions: The ceramic materials with different microstructures showed different patterns of subsurface cracking. [source]


Assessment of grape skin hardness by a puncture test

Abstract BACKGROUND: The release of grape components during wine making might be related to the mechanical properties of the skin, in particular its hardness. Samples from three varieties collected during the 2005 vintage season in Piedmont, Italy, were tested for their skin hardness using a texture analyser. The goal was to understand the statistical interactions between three factors,variety, cluster position and puncture point,and their influence on the grape skin hardness. A discussion on the relationship between the size of the sample used and the confidence level is also provided. RESULTS: Results of the ANOVA test showed that there is an interaction between the variety and the puncture point when measuring the skin hardness with the break energy. The position of the berry on the cluster does not affect the berry skin break energy. We also show that a sample size depend on the variety tested. CONCLUSION: The break energy is more useful in understanding the effect of the three factors on the skin hardness. Other factors that might affect the puncture test applied to grapes need to be studied in the future and the usefulness of the test in winemaking will need to be further developed. The fate of seeds in Mediterranean soil seed banks in relation to their traits

Abstract Question: Is there any change in seed density and species richness in Mediterranean surface soil banks during summer? Are there any relationships between these summer variations and seed traits (weight, length and shape), without and with controlling the phylogenetic effect? Location: Central Spain. Methods: Samples of the surface soil seed bank were collected in two Mediterranean systems, grassland and scrubland, at two points in the year: immediately after the summer production peak and immediately prior to the autumn germination peak. We used Canonical Correspondence Analysis ordination to check for changes in floristic composition and ANOVAs to check for changes in seed density and species richness between summer and autumn samples. We used multiple regression analysis to analyse the relationship between summer variations in soil seed density and form traits, with and without controlling phylogenetic relations. Results: Soil seed density dropped significantly during the summer in the two systems (28% in grasslands, repeated measures ANOVA test; F = 58.19, P < 0.01; 72% in scrublands, repeated measures ANOVA test; F = 75.67, P < 0.001). Species richness, however, only dropped significantly in the scrubland (32%; repeated measures ANOVA test; F = 9.17, P < 0.05). Variation in the floristic composition of the shallow banks was only significant in the scrubland. Changes in shallow bank density were related significantly to seed morphology features, with greater drops in species with heavier seeds in grasslands and species with longer seeds in scrubland. Conclusions: Our results show a substantial loss of seeds in the uppermost soil layer during the summer period between the point of peak production and the autumn germination peak. This drop was clearer in the scrubland than in the grassland. Longer and/or heavier seeds underwent the greatest declines in density at the end of summer, indicating a more intense effect of post-dispersal predation on large-seeded species. [source]


Effect of weak, interrupted sinusoidal low frequency magnetic field on neural regeneration in rats: Functional evaluation

Abstract A study of the effect of weak, interrupted sinusoidal low frequency magnetic field (ISMF) stimulation on regeneration of the rat sciatic nerve was carried out. In the experiment, 60 Wistar rats were used: 24 rats underwent unilateral sciatic nerve transection injury and immediate surgical nerve repair, 24 rats underwent unilateral sciatic nerve crush injury, and the remaining 12 rats underwent a sham surgery. Half of the animals (n,=,12) with either sciatic nerve lesion were randomly chosen and exposed between a pair of Helmholtz coils for 3 weeks post-injury, 4 h/day, to an interrupted (active period to pause ratio,=,1.4 s/0.8 s) sinusoidal 50 Hz magnetic field of 0.5 mT. The other half of the animals (n,=,12) and six rats with sham surgery were used for two separate controls. Functional recovery was followed for 6 weeks for the crush injuries and 7˝ months for the transection injuries by video assisted footprint analysis in static conditions and quantified using a recently revised static sciatic index (SSI) formula. We ascertained that the magnetic field influence was weak, but certainly detectable in both injury models. The accuracy of ISMF influence detection, determined by the one-way repeated measures ANOVA test, was better for the crush injury model: F(1, 198),=,9.0144, P,=,.003, than for the transection injury model: F(1, 198),=,6.4826, P,=,.012. The Student,Newman,Keuls range test for each response day yielded significant differences (P,<,.05) between the exposed and control groups early in the beginning of functional recovery and later on from the points adjacent to the beginning of the plateau, or 95% of functional recovery, and the end of observation. These differences probably reflect the ISMF systemic effect on the neuron cell bodies and increased and more efficient reinnervation of the periphery. Effectiveness of an oral health education programme in primary schools in Zimbabwe after 3.5 years

Abstract , Many medical and dental professionals in African nations believe that school teachers, through attendance of a short workshop, can be trained to provide oral health education. This increases the number of professionals available and is regarded as an important way forward in improving oral health. Objectives: The current study assesses the effectiveness of an oral health education programme administered by schoolteachers in a district in Zimbabwe over a period of 3.5 years. Methods: The experimental group consisted of schools that had sent representatives to a regional workshop on oral health held in 1992. The control group was selected at random from schools not having attended the workshop. A total number of 439 boys and 526 girls were examined in 1992. Follow-up evaluations were carried out in 1993, 1994 and 1996. The dependent variables were plaque accumulation and caries increment in grade 2 and grade 4 children of experimental and control schools. Results: ANOVA test with year of evaluation (1992,94), experiment/control school, age and gender as independent variables showed no statistically significant difference in mean plaque scores in longitudinally examined original grade 2 (P>0.20) and grade 4 children (P=0.06) from experimental and control schools. The mean caries increment score in the experimental and control schools was 0.04 and 0.19, respectively. ANOVA test with fluoride levels and gender as independent variables on caries increment in experimental and control schools did not show a statistically significant difference (P=0.06). Conclusion: The one-time training of teachers in aspects of oral health was ineffective in lowering plaque levels over a period of 3.5 years. Considering the low caries increment observed over the study period, the effect of the oral health programme on caries levels in the study group was inconclusive. [source]


The Effect of Using Variable Frequency Trains During Functional Electrical Stimulation Cycling

ABSTRACT Objectives., This paper describes an experimental investigation of variable frequency stimulation patterns as a means of increasing torque production and, hence, performance in cycling induced by functional electrical stimulation. Materials and Methods., Experiments were conducted on six able-bodied subjects stimulating both quadriceps during isokinetic trials. Constant-frequency trains (CFT) with 50-msec interpulse intervals and four catchlike-inducing trains (CIT) were tested. The CITs had an initial, brief, high-frequency burst of two pulses at the onset of or within a subtetanic low-frequency stimulation train. Each stimulation train consisted of the same number of pulses. The active torques produced by each train were compared. Parametric main effect ANOVA tests were performed on the active torque-time integral (TTI), on the active torque peaks and on the time needed to reach those peaks (T2P). Results., The electrical stimulation of the quadriceps produced active torques with mean peak values in the range of 1.6,3.5 Nm and a standard error below 0.2 Nm. CITs produced a significant increase of TTI and torque peaks compared with CFTs in all the experimental conditions. In particular, during the postfatigue trials, the CITs with the doublet placed in the middle of the train produced TTIs and torque peaks about 61% and 28% larger than the CFT pattern, respectively. In addition, the CITs showed the lowest reduction of the performance between prefatigue and postfatigue conditions. Conclusions., The use of CITs improves the functional electrical stimulation cycling performance compared with CFT stimulation. This application might have a relevant clinical importance for individuals with stroke where the residual sensation is still present and thus the maximization of the performance without an excessive increase of the stimulation intensity is advisable. Therefore, exercise intensity can be increased yielding a better muscle strength and endurance that may be beneficially for later gait training in individuals with stroke. [source]


Comparison of characteristics of treated and non-treated patients with Hepatitis C infection,

Abstract Objectives This study compares the characteristics of treated and non-treated patients with Hepatitis C (HCV) infection. Methods Information on patient demographics, clinical data, and treatment regimens were collected from a retrospective medical record review of 998 patients diagnosed with HCV, representing a diverse geographic sample of 200 U.S. physicians including 130 gastroenterologists, 50 infectious disease physicians, and 20 hepatologists. A total of 551 patients were randomly selected and 447 were provided as an augmented sample in an intent-to-treat analysis based on treatment decisions. Pretreatment factors examined included age, gender, race, weight, HCV genotype, HCV viral load, serum ALT levels, liver biopsy results, cirrhosis, HIV co-infection, HBV co-infection, IV drug use, and insurance status. Univariate analyses were performed using Chi-squared or ANOVA tests. Factors that were significant in univariate analyses were entered into a multivariate logistic regression model with HCV treatment as the outcome variable. Results Of the 998 patients reviewed, 778 were treated for HCV and 220 were not treated. In univariate analyses, non-treated patients were more likely to be African American, HBV co-infected, HIV co-infected, IDUs, alcoholics, Medicaid insured, and were less likely to have had biopsies. The multivariate regression analysis demonstrated that performance of a liver biopsy, treatment with psychiatric medications (antidepressants and anxiolytics), and patient weight were independently associated with treatment, while Medicaid insurance and HIV co-infection were independently associated with a decreased likelihood of receiving HCV therapy. Conclusion This study suggests that it is not the clinical stage of HCV infection but the patient's demographic characteristics and co-morbid conditions that impact the decision to initiate HCV therapy. Application of a digital technique in evaluating the reliability of shade guides

summary, There appears to be a need for a reliable method for quantification of tooth colour and analysis of shade. Therefore, the primary objective of this study was to show the applicability of graphic software in colour analysis and secondly to investigate the reliability of commercial shade guides produced by the same manufacturer, using this digital technique. After confirming the reliability and reproducibility of the digital method by using self-assessed coloured images, three shade guides of the same manufacturer were photographed in daylight and in studio environments with a digital camera and saved in tagged image file format (TIFF) format. Colour analysis of each photograph was performed using the Adobe Photoshop 4·0 graphic program. Luminosity, and red, green, blue (L and RGB) values of each shade tab of each shade guide were measured and the data were subjected to statistical analysis using the repeated measure Anova test. The L and RGB values of the images taken in daylight differed significantly from those of the images taken in studio environment (P < 0·05). In both environments, the luminosity and red values of the shade tabs were significantly different from each other (P < 0·05). It was concluded that, when the environmental conditions were kept constant, the Adobe Photoshop 4·0 colour analysis program could be used to analyse the colour of images. On the other hand, the results revealed that the accuracy of shade tabs widely being used in colour matching should be readdressed. [source]


Biocompatibility of various root canal filling materials ex vivo

Abstract Aim, To evaluate the biocompatibility of a resin-based endodontic filler (RealSeal) using the indirect cytotoxicity test. Methodology, Human gingival fibroblasts were cultured ex vivo. Pellets of the materials to be tested were incubated for 24, 48, and 72 h at 37 °C under sterile conditions to obtain their eluates. The fibroblasts were exposed to either diluted (50%) or undiluted eluates for 24 h. A culture medium with foetal calf serum was added to the control wells. Cell viability was estimated by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide method. The data concerning cell viability were statistically analyzed using one-way anova test and Bonferroni multiple comparisons test. Results, Eluates obtained after 24 h of incubation with the resin filler did not reduce cellular viability. An increase in cellular viability, as compared with control cells, was observed in the gutta-percha group. The undiluted eluate from the polyether material was cytotoxic, causing an 82 ± 4% decrease in cellular viability. Eluates obtained after 48 h of incubation with the resin filler increased cellular viability, whereas the polyether significantly reduced viability. Gutta-percha did not cause any detectable change. After 72 h of incubation the eluate of the resin filler caused an increase in cellular viability, as did gutta-percha, whereas polyether caused a significant decrease. Conclusions, RealSeal resin filler was nontoxic in this laboratory model. Further investigations are necessary to verify its usefulness in clinical applications. [source]


The percentage of gutta-percha-filled area in simulated curved canals when filled using Endo Twinn, a new heat device source

Abstract Aim, To compare the percentage of gutta-percha-filled area (PGP) in simulated root canals when varying the penetration depth and function of the pluggers (heat versus heat plus vibration) using Endo Twinn. Methodology, Sixty-four resin blocks with simulated 34,35° curved canals were randomly divided into two groups in order to obtain two canal shapes: group A with 0.8 taper and group B with 0.4 taper. The apical portion of each canal was prepared to a size 20 K-file. The canals were filled with gutta-percha in combination with a root canal sealer. In each group 16 canals were filled using the Endo Twinn heat function and 16 canals by means of both the heat and the vibration function. All samples were sectioned horizontally at three levels (1.25, 2.5 and 4.0 mm from the working length) and the PGP was measured. Data were analysed using anova test. Results, At the 1.25 mm level PGP was significantly greater using the vibration function (P = 0.0329) and in 0.8 taper canals (P < 0.0001). At the 2.5 mm level the PGP was greater in the canals with 0.8 taper compared with a 0.4 taper with or without vibration (vibration, P = 0.0056; interaction taper-vibration, P = 0.0020). In 0.4 taper canals the PGP was greater when the vibration function was activated. At the 4 mm level in 0.8 taper canals there was no significant difference in PGP with or without the vibration (P = 0.6742). Conclusions, 0.8 taper canals had significantly greater PGP than 0.4 taper canals. At the 1.25 mm level there was significantly greater PGP when the vibration function was activated. [source]


Effects of endodontic instrument handle diameter on electromyographic activity of forearm and hand muscles

Abstract Aim To determine the influence of the handle diameter of endodontic instruments on forearm and hand muscle activity using electromyographic (EMG) recording. Methodology Size 45 K-type files were fitted with four different handle diameters; 3.5, 4.0, 5.0, and 6.0 mm. Seven dentists then attempted to negotiate to the working length acrylic resin root canals with each of the four handle sizes using a reaming motion. EMG activities were recorded from the flexor pollicis brevis muscle (f.p.b.), the flexor carpi radialis muscle (f.c.r.), and the brachioradialis muscle (b) with bipolar surface electrodes. The time taken to negotiate the canals, the area of integrated EMG that corresponded to the amount of EMG activity required during penetration and the maximum amplitude of EMG were measured using the EMG data. Results were analysed statistically using a one-way factorial anova test and multiple comparison tests. Results Reaming time and integrated EMG area of each muscle decreased with an increase in handle diameter. The most significant difference in time and area of integrated EMG was detected between handles of 6 mm and 3.5 mm diameter (time: P < 0.01, area of the f.p.b.: P < 0.01, area of the f.c.r. and b: P < 0.05), and between handles of 5 mm and 3.5 mm diameter (P < 0.05). Both 5 mm and 6 mm handles significantly decreased the maximum amplitude of EMG recorded from the f.p.b. compared with 3.5 mm handles (between 3.5 mm and 6 mm: P < 0.01, between 3.5 mm and 5 mm: P < 0.05). Conclusion The results indicate that handle diameter has an effect on reaming time as well as on muscle activity. As a consequence, handle diameter influenced operator performance during instrumentation. [source]


A comparative study of oral health knowledge, attitude and behaviour of first and final year dental students of Udaipur city, Rajasthan, India

Abstract:, Objective:, To assess and compare differences in oral health knowledge, attitudes and behaviour among first and final year dental students in Udaipur city, Rajasthan, India. Materials and methods:, In a cross-sectional study, 182 first year students (59 males and 123 females) and 157 final year students (75 males and 82 females) of the two dental colleges in Udaipur city; Rajasthan, were surveyed using a self-administered structured questionnaire including 37 multiple choice questions pretested through a pilot survey. The data was analysed using the spss version 10.0. The mean percentage scores, standard deviation and frequency distribution were calculated. The Student's t -test and anova test were used as test of significance. The linear regression analysis were used to assess the relation of knowledge, attitude and behaviour. Results:, The mean % score for oral health knowledge, attitude and behaviour were significantly higher in final year students compared to first year students (P < 0.001). The linear regression analysis showed a statistically significant linear relationship of attitude with the knowledge (P < 0.001) and behaviour with the attitude (P < 0.001) of the students. Conclusion:, Besides the positive changes revealed in the oral health knowledge, attitude and behaviour, among the students from first to final year of dental studies, preventive behaviour among the students could still be improved. Effect of administration mode (patient vs physician) and patient's educational level on the Turkish version of the International Prostate Symptom Score

Abstract Objectives: To compare the effectiveness of the International Prostate Symptom Score (IPSS) when administered by the physician to when self-administered by the patient. The effect of the patient's educational level on the IPSS was also evaluated. Methods: One hundred and seven previously untreated patients with symptomatic benign prostatic hyperplasia (BPH) completed the Turkish version of the International Prostate Symptom Score (Turkish I-PSS) and quality of life (QOL) questionnaires during a single office visit, first on their own and then with an interviewing physician. The patients were categorized into three groups according to their educational levels. Paired t -tests were performed to compare the total IPSS (tIPSS) and QOL results between the two testing modes. IPSS and QOL scores resulting from both modes were compared using a kappa test. Differences between the physician-assisted and self-administered scores among the different educational groups were further compared using a one-way anova test and Post Hoc Multiple Comparisons. To compare the objective effectiveness of tIPSS and QOL between the two testing modes, we selected the positive actual state, which was maximum urine flow (Qmax) of 15 mL/s or less and constructed receiver operating characteristics (ROC) curves for all patients. This estimation was constructed for each educational level. Results: There were no statistical differences in IPSS and QOL values obtained by the patients or physicians (P > 0.05). The ROC areas for tIPSS were 0.94 and 0.93, and the ROC areas for QOL scores were 0.97 and 0.91 for information obtained by physicians and patients, respectively. When IPSS answers and QOL scores were evaluated separately, consistency was found across both modes of administration. However, there were lower levels of consistency in answers to IPSS questions 2, 5 and 6 (P = 0.59;0.42; 0.52, respectively). There was no significant difference among the aforementioned data in the educational groups. Conclusion: Although the total IPSS and QOL scores were not affected by the different modes of administration, we recommend that the physicians should evaluate answers to questions 2, 5 and 6 carefully. The present study demonstrates that the educational level did not affect the IPSS and QOL when administered either by the physician or the patient. [source]


Plasma lipoprotein concentrations in the dog: the effects of gender, age, breed and diet

Summary Earlier studies of canine lipoprotein metabolism have frequently not taken into account such variables as age, gender, lifestyle or feeding status. In the last years, many changes to lifestyle and feeding of dogs have occurred. In this study, C-tot, C-HDL, C-LDL, triglycerides and lipoprotein fractions were determined in 251 healthy dogs by means of enzymatic methods and through the electrophoretic technique. All data were analysed by multifactor anova test to determine which factors (age, gender, breed and diet) have a statistically significant effect (p < 0.05) on the determined parameter and subsequently Bonferroni's test was applied where necessary. Gender, age, breed and diet can significantly affect lipid metabolism, in particular lipoproteins involved in cholesterol plasma transport; on the contrary, triglycerides are not influenced by the same factors. The most important observation about age is the high level of C-LDL in puppies under 1 year of age. The highest cholesterol concentrations are found in Rottweiler but high values of plasma cholesterol are found also in Pyrenees Mountain dog and a great level of C-LDL in Labrador. Diet has shown a great influence on lipidic metabolism: dogs fed with different high-quality dry foods had significant differences in plasma cholesterol values (C-tot, C-HDL, C-LDL,), in particular, dogs fed with a diet rich in fish and fish-by-products have shown the lowest levels of C-tot, C-HDL and C-LDL. Origin and Chemical Coding of Primary Afferent Neurones Supplying the Prostate of the Dog

Summary Retrograde tracing technique combined with the double-fluorescent immunohistochemistry were used to investigate the distribution and chemical coding of primary afferent neurones supplying the canine prostate. After the injection of Fast Blue (FB) into the prostatic tissue retrogradely-labelled (FB+) primary afferent neurones were localized in bilateral L1,Ca1 dorsal root ganglia (DRG). Statistical analysis using anova test showed that there are two major sources of afferent prostate innervation. The vast majority of prostate-supplying primary afferent neurones were located in bilateral L2,L4 DRG (56.9 ± 0.6%). The second source of the afferent innervation of canine prostate were bilateral S1,Ca1 DRG (40.6 ± 1.0%). No statistically significant differences were found between average number of FB+ neurones localized in the left and right DRG (49.5 ± 1.7 and 50.5 ± 1.7%, respectively). Immunohistochemistry revealed that FB+ primary afferent neurones contain several neuropeptides in various combinations. In the prostate-supplying neurones of lumbar and sacro-caudal DRG the immunoreactivity to substance P (SP) and calcitonin gene-related peptide (CGRP) was found most frequently (50 ± 3.7 and 37.3 ± 1.9%, respectively). Both in the lumbar and sacro-caudal DRG, considerable population of FB+ neurones immunoreactive neither to SP nor CGRP were also found (23 ± 2.6 and 32.8 ± 2.3%, respectively). In the lumbar DRG 10.7 ± 1.1% of SP-immunoreactive FB+ neurones also contained galanin (GAL). In 9.2 ± 2.2% of the prostate-supplying primary afferent neurones located in the sacro-caudal DRG the co-localization of SP and GAL was also reported. Results of the retrograde tracing experiment demonstrated for the first time sources of afferent innervation of the canine prostate. Double immunohistochemistry revealed that many of the prostate-supplying primary afferent neurones express some of sensory neuropeptides which presumably may be involved in nociception and some pathological processes like inflammation or nerve injury. Subcutaneous histamine versus sodium valproate in migraine prophylaxis: a randomized, controlled, double-blind study

Histamine has a selective affinity for H3-receptors and it may specifically inhibit the neurogenic edema response involved in migraine pathophysiology. The objective of this study was to evaluate the therapeutic potential of subcutaneous administration of histamine in migraine prophylaxis, compared with oral administration of sodium valproate, in an open clinical trial. Ninety-two patients with migraine were selected under criteria established by the International Headache Society and enrolled in a 12-week double-blind controlled clinical trial to evaluate the efficacy of subcutaneous administration of histamine (1,10 ng twice a week; n = 46) compared with oral administration of sodium valproate (500 mg daily dose; n = 46). The variables studied were headache intensity, frequency, duration, analgesic intake and migraine disability assessment (MIDAS). Two-tailed Student's t - test was used to compare means and the Mann,Whitney U and anova tests were used. The data collected during the 4th, 8th and 12th weeks of treatment revealed that histamine caused a significantly greater reduction (P < 0.001) in intensity and duration of migraine attacks as well as in analgesic intake. No difference was detected in the frequency of attacks or in MIDAS. The present study provides evidence of the superior efficacy of histamine applied subcutaneously in migraine prophylaxis when compared with sodium valproate taken orally. Subcutaneously applied histamine may represent a novel and effective therapeutic alternative in resistant migraine patients. [source]


Effect of Er:YAG laser and EDTAC on the adhesiveness to dentine of different sealers containing calcium hydroxide

Abstract Aim, To evaluate the effect of 15% EDTAC solution and Er:YAG laser irradiation on the adhesiveness to dentine of root canal sealers containing calcium hydroxide. Methodology, The crowns of 60 maxillary human molars were ground until dentine was exposed. The teeth were divided into three groups of 20 teeth: group I, the dentine surface received no treatment; group II, 15% EDTAC solution was applied to the dentine; group III, the dentine received Er:YAG laser application (11 mm focal distance with perpendicular incidence to dentine surface; 4 Hz frequency; 200 mJ energy; 2.25 W potency; 62 J total energy; 1 min application time). Aluminium cylinders filled with the sealers, Sealer 26, Apexit, Sealapex and CRCS, were then applied to the treated surfaces. Adhesiveness was measured with a universal testing machine, with traction results given in MegaPascals (MPa). These results were submitted to anova tests. Results, Statistical analysis showed significant differences (P < 0.01) amongst adhesiveness values of the sealers and treatments tested. Thus, sealers could be ranked in decreasing adhesiveness values: Sealer 26, CRCS, Apexit, Sealapex. Er:YAG laser irradiation and EDTAC solution application increased adhesiveness values only for Sealer 26 and Apexit. Laser irradiation was superior to EDTAC application only for Sealer 26 adhesiveness values. Conclusions, Er:YAG laser is as efficient as EDTAC solution in increasing adhesiveness of root canal sealers containing calcium hydroxide to human dentine. [source]
