Selected AbstractsThe histology of the Upper Silurian osteostracan Timanaspis kossovoii Obruchev (Agnatha) from North Timan, RussiaACTA ZOOLOGICA, Issue 2009Olga B. Afanassieva Abstract The fine structure of the exoskeleton of Timanaspis kossovoii Obruchev 1962 was investigated based on new well-preserved material. The remains come from the lower part of the Eptarma Formation (Upper Silurian). Timanaspis kossovoii was referred by Obruchev to the family Tremataspididae and most authors agree with the placement of Timanaspis among tremataspidid osteostracans. Histological study of the shield of T. kossovoii has revealed the presence of unusual features in the exoskeletal structure. The smooth and shiny surface of the ventral side of the shield of T. kossovoii is similar to that of species of Tremataspis, but thin sections show differences in histological composition. The exoskeleton of T. kossovoii consists mainly of dense bony tissue in the middle layer and a laminated basal layer. No osteocyte cavities of typical form and size were found in the middle layer. The upper parts of the elongated tubercles of the dorsal side of the shield are formed by a well-developed dentine-like tissue; the arrangement of numerous tubules within this tissue is far more regular than in the mesodentine of Tremataspis. No traces of perforated septa, pore fields or a polygonal model were detected in the structure of the exoskeleton of Timanaspis. We conclude that Timanaspis can be differentiated from conventional tremataspidids by histological features and refer it to the new family Timanaspididae. Moreover, the Timanaspis exoskeleton provides evidence that an acellular condition of bony tissue does not appear to be characteristic of only Late Devonian osteostracans. [source] Histological study of fetal kidney with urethral obstruction and vesicoureteral reflux: A consideration on the etiology of congenital reflux nephropathyINTERNATIONAL JOURNAL OF UROLOGY, Issue 10 2003KENJI SHIMADA Purpose: A recent subject of interest regarding reflux nephropathy is the presence of renal abnormalities in neonates and infants who have no history of urinary tract infections. Debates have centered on the etiology of this renal abnormality , congenital reflux nephropathy; regarding whether it is the result of abnormal ureteral budding or of back pressure effect from sterile reflux. We examined the renal pathology of fetuses with urethral obstruction and vesicoureteral reflux, and we suggest herein a possible etiology of congenital reflux nephropathy. Methods: The renal pathology of seven autopsied fetuses with vesicoureteral reflux was studied. Reflux was demonstrated at autopsy by slow injection of contrast medium into the bladder. Severe urethral obstruction, either atresia or urethral valves, was evident in six of the subjects. Results: In six subjects, abnormality of the urinary tracts was detected by prenatal ultrasonography. Of these six subjects, three revealed characteristics of prune belly syndrome. Reflux was graded as moderate in five subjects, and severe in two. In three subjects autopsied at 21 weeks gestation or earlier, the kidneys were well-developed with normal corticomedullary configuration, and nephrogenesis was retained. In three cases autopsied at over 25 weeks of gestation, the kidneys were grossly cystic, and the nephrogenic zone was completely absent. Contrast medium was observed not only in the dilated ducts and tubules, but also in the subcapsular cysts. Extravasation of the contrast medium was seen in the peritubular space. In the last subject with normal lower urinary tract, abnormal segments among normal cortical structures were observed. Conclusion: Our findings of renal pathology in fetuses with reflux are quite similar to those seen in fetal hydronephrosis. Back pressure from reflux probably damages the developing kidney leading to a degeneration of the ampullae and a reduction in the number of nephrons. Both dilatation of the collecting ducts and tubules, and extravasation of the urine may result in interstitial fibrosis. We postulate that one of the important etiologies of congenital reflux nephropathy may be the result of back pressure from sterile reflux. [source] Antihyperlipidemic activity of 3-hydroxymethyl xylitol, a novel antidiabetic compound isolated from Casearia esculenta (Roxb.) root, in streptozotocin-diabetic ratsJOURNAL OF BIOCHEMICAL AND MOLECULAR TOXICOLOGY, Issue 2 2010Govindasamy Chandramohan Abstract Casearia esculenta root (Roxb.) is widely used in traditional system of medicine to treat diabetes in India. An active compound, 3-hydroxymethyl xylitol (3-HMX), has been isolated, and its optimum dose has been determined in a short duration study and patented. In addition, the long-term effect of 3-HMX in type 2 diabetic rats on carbohydrate metabolism was investigated, and its antihyperglycemic effect was shown previously (Chandramohan et al., Eur J Pharmacol 2008;590:437,443). In this study we investigated the effect of 3-HMX on plasma and tissue lipid profiles in streptozotocin-induced diabetic rats. Diabetes was induced in adult male albino rats of the Wistar strain, weighing 180,200 g, by administration of streptozotocin (40 mg/kg of body weight) intraperitoneally. The normal and diabetic rats were treated with 3-HMX (40 mg/kg BW/day) for 45 days. The levels of total cholesterol, triglycerides, free fatty acids, and phospholipids were assayed in the plasma besides lipoprotein-cholesterol (high-density lipoprotein-cholesterol (HDL-C), low-density lipoprotein-cholesterol (LDL-C), and very low density lipoprotein-cholesterol (VLDL-C)) and tissues (liver, kidney, heart, and brain). Total cholesterol, triglyceride, free fatty acid, and phospholipid (LDL-C and VLDL-C in plasma only) levels increased in plasma and tissues significantly, whereas plasma HDL-C significantly decreased in diabetic rats. Treatment with 3-HMX or glibenclamide reversed the above-mentioned changes and improved toward normalcy. Histological study of liver also confirmed the biochemical findings. Thus administration of 3-HMX is able to reduce hyperglycemia and hyperlipidemia related to the risk of diabetes mellitus. © 2010 Wiley Periodicals, Inc. Roosen MSc Abstract Background and Objectives We have recently shown that repeated low fluence photoepilation (LFP) with intense pulsed light (IPL) leads to effective hair removal, which is fully reversible. Contrary to permanent hair removal treatments, LFP does not induce severe damage to the hair follicle. The purpose of the current study is to investigate the impact of LFP on the structure and the physiology of the hair follicle. Study Design/Materials and Methods Single pulses of IPL with a fluence of 9 J/cm2 and duration of 15 milliseconds were applied to one lower leg of 12 female subjects, followed by taking a single biopsy per person, either immediately, or after 3 or 7 days. Additionally, we present a novel approach to examine the effects of LFP, in which ex vivo hairy human scalp skin was exposed to IPL pulses with the same parameters as above, followed by isolation and culturing of the hair follicles over several days. Samples were examined histologically and morphologically. Results The majority of the cultured follicles that had been exposed to LFP treatment showed a marked treatment effect. The melanin containing part of the hair follicle bulb was the target and a catagen-like transformation was observed demonstrating that hair formation had ceased. The other follicles that had been exposed to LFP showed a less strong or no response. The skin biopsies also revealed that the melanin-rich region of the hair follicle bulb matrix was targeted; other parts of the follicle and the skin remained unaffected. Catagen/telogen hair follicles were visible with unusual melanin clumping, indicating this cycle phase was induced by the IPL treatment. Conclusions Low fluence photoepilation targets the pigmented matrix area of the anagen hair follicle bulb, causing a highly localized but mild trauma that interrupts the hair cycle, induces a catagen-like state and eventually leads to temporary loss of the hair. [source] Influence of extracorporeal porcine liver perfusion on nonhuman primates: Minimizing hemolysis improves subsequent survivalLIVER TRANSPLANTATION, Issue 7 2001Ryuta Nishitai MD The aim of this study is to detect and analyze risk factors of direct cross-circulation between porcine liver and nonhuman primates before a clinical application of extracorporeal liver perfusion (ECLP) as a liver-assist method. Porcine livers were perfused with baboon blood in an ECLP system. Six healthy baboons were directly connected to the ECLP system with continuous prostaglandin E1 administration. Cross-circulation was terminated in the following circumstances: (1) hepatic arterial or portal perfusion pressures elevated to 200 or 60 mm Hg, respectively; (2) massive exudative bleeding from the graft surface; or (3) bile output decreased to less than 5 ,L/h/g of liver weight. In case 1, cross-circulation was continued for 10 hours. Severe macroscopic hemolysis occurred, and serum hemoglobin (s-Hb) concentration reached a peak of 47 mg/dL. The baboon died of acute renal failure 2 days later. Histological study of the perfused porcine liver showed marked microthrombi formation. In 3 of the later 5 cases, cross-circulation was discontinued when mild macroscopic hemolysis was observed. The duration of the 5 cross-circulations was maximally 6 hours (mean, 4.4 ± 1.2 [SD] hours). Mean s-Hb concentration in the 5 cases was elevated to 14.8 ± 5.8 mg/dL at the end of cross-circulation and decreased to the baseline level within 24 hours. These 5 baboons survived without organ dysfunction or immunologic disturbance. When severe hemolysis is avoided, direct cross-circulation using the ECLP system can be achieved without serious complications in nonhuman primates. [source] Case of acute ileus caused by a spirurina larvaPATHOLOGY INTERNATIONAL, Issue 9 2004Toshihiko Miyake A growing body of clinical cases suggests that a kind of nematode larva, type X larva of the suborder Spirurina that inhabits firefly squids (Watasenia scintillans, or ,Hotaru-ika' in Japanese), can cause acute ileus in humans. However, the larva itself has rarely been found in the wall of the obstructed intestine. We describe here a case of acute ileus, in which a type X spirurina larva was found histologically. A 60-year-old Japanese man suffered from acute abdomen, and an emergency laparotomy revealed a marked stenosis of the ileum. Histological study of the surgically resected ileum showed severe eosinophilic enteritis and a nematode larva. The morphological features of this larva were identical to those of the type X spirurina larva. Interestingly, the larva that was found existed within a small blood vessel, suggesting that the larva migrans of type X spirurina can take place via vasculature. The patient in the present case did not recall ingesting raw squids prior to the onset of his disease. Hence, this indicates that even if the ingestion of raw firefly squids is uncertain, spirurina infection should be included in the differential diagnosis of acute ileus or eosinophilic enteritis. [source] Histological Studies of Growing and Mature Antlers of Red Deer Stags (Cervus elaphus)ANATOMIA, HISTOLOGIA, EMBRYOLOGIA, Issue 3 2009M. Cegielski Summary This study aims at presenting histology of growing and mature antlers in red deer stag (Cervus elaphus). Growing antlers constitute a model organ for examining regeneration processes of tissues because they are the only mammalian appendages capable of regeneration. Histological study revealed that the tip of a growing antler consists of hairy skin, perichondrium, mesenchyme and chondroprogenitors area. By performing immunochistochemistry, we found that cell expressing Ki-67 and PCNA antigens were localized in basal layer of epidermis, skin glands and beneath their secretory sections, mesenchyme as well as within and in the vicinity of central blood vessels. Ultrastructurally, cells from chondroprogenitors zone have chondroblast-like morphology and take part in producing of collagen fibres followed by the process of cartilage mineralization. However, mature antlers also consist of lamellar osseous tissue. [source] Interferon-gamma is causatively involved in experimental inflammatory bowel disease in miceCLINICAL & EXPERIMENTAL IMMUNOLOGY, Issue 2 2006R. Ito Summary Cytokines may be crucially involved in the pathogenesis of inflammatory bowel diseases (IBD), but it remains controversial whether interferon (IFN)-,, a typical proinflammatory cytokine, is an essential mediator to cause the disorders. In the present study, IFN-,,/, and wild-type (WT) C57BL/6 mice were fed 2·5% dextran sodium sulphate (DSS) in drinking water for 7 days, in order to investigate DSS-induced intestinal inflammation. The DSS-treated WT mice exhibited a robust production of IFN-, in the gut, a remarkable loss of body weight, as well as high rate of mortality (60%). In striking contrast, IFN-, deficient mice did not develop DSS-induced colitis, as indicated by the maintenance of body weight and survival rate of 100%. Severe intestinal inflammation was demonstrated exclusively in WT animals in terms of the shortening of the bowel as well as the elevation of the disease activity index, myeloperoxidase (MPO) activity and serum haptoglobin level. Histological study of DSS-treated WT intestine revealed disruption of mucosal epithelium and massive infiltration of inflammatory cells, while the organ from IFN-,,/, mice remained virtually normal in appearance. Enzyme-linked immunosorbent assay (ELISA) analyses indicated abundant production of three chemokines, i.e. monokine induced by interferon-, (MIG), interferon-inducible protein 10 (IP-10) and monocyte chemoattractant protein-1 (MCP-1), in the DSS-irritated intestine of WT but not of IFN-,,/, mice. The present results demonstrate clearly that IFN-, plays indispensable roles in the initiation of DSS colitis, and some chemokines are produced in an IFN-,-dependent fashion. [source] Comparison of histologic, biochemical, and mechanical properties of murine skin treated with the 1064-nm and 1320-nm Nd:YAG lasersEXPERIMENTAL DERMATOLOGY, Issue 12 2005Yong-Yan Dang Abstract:, The goal of this study was to compare the effects of the Q-switched 1064-nm Nd:YAG laser and the 1320-nm Nd:YAG laser non-ablative treatments on mouse skin in vivo. Skin elasticity measurements were carried out with a Reviscometer, and skin samples were taken for histological study, hydroxyproline content assay and estimation of collagen type I and III. By the second month after non-ablative treatments, the 1064-nm laser treatment resulted in an average of 25% greater improvement of skin elasticity, 6% more increase of dermal thickness, and 11% higher synthesis of hydroxyproline than the 1320-nm laser. Collagen type III increased markedly after the 1064-nm laser treatment whereas more collagen type I was elicited by the 1320-nm laser. Our results demonstrated that the 1064-nm laser was more effective than the 1320-nm Nd:YAG laser in non-ablative treatments, but the results needed to be confirmed in humans. It appeared that photo-mechanic reaction could cause more collagen type III synthesis whereas the photo-thermal effect was in favor of the formation of collagen type I. [source] High Efficacy of Ranitidine Bismuth Citrate, Amoxicillin, Clarithromycin and Metronidazole Twice Daily for Only Five Days in Helicobacter pylori EradicationHELICOBACTER, Issue 2 2001Javier P. Gisbert ABSTRACT Aim. The combination of a proton pump inhibitor (PPI) or ranitidine-bismuth-citrate (Rbc) and two antibiotics for 7,10 days are, at present, the preferred treatments in Helicobacter pylori eradication. However, therapies for fewer than 7 days have been scarcely evaluated and it is unknown whether the length of treatment can be shortened, without a lost of efficacy, if three instead of two antibiotics are used. The aim of our study was to evaluate the efficacy of Rbc plus three antibiotics for only 5 days in H. pylori eradication. Methods. We prospectively studied 80 patients (34% duodenal ulcer, 66% functional dyspepsia) infected by H. pylori. At endoscopy, biopsies were obtained for histological study and rapid urease test, and a 13C-urea breath test was carried out. Urea breath test was repeated 4 weeks after completing eradication treatment with Rbc [400 mg twice a day (bid)], amoxicillin (1 g bid), clarithromycin (500 mg bid) and metronidazole (500 mg bid). All drugs were administered together after breakfast and dinner for 5 days only, and no treatment was administered thereafter. Compliance with therapy was determined from the interrogatory and the recovery of empty envelopes of medications. Results. In 79 out of the 80 patients, H. pylori eradication success or failure was assessed after therapy (one patient was lost from follow-up). All but one of these 79 patients took all the medications (one patient stopped treatment on the day 3 due to nausea/vomiting). Per protocol eradication was achieved in 72/78 (92%; 95% CI, 84,96%) and in 72/80 (90%; 81,95%) by intention-to-treat. Therapy was more effective in patients with duodenal ulcer than in those with functional dyspepsia [100% (87,100%) vs. 85% (73,92%) by intention-to-treat; p < .05]. Adverse effects were described in ten patients (12%), and included the perception of a metallic taste (eight patients), nausea/vomiting (two patients, one of them abandoned the treatment due to this), and diarrhea (two patients). Conclusion. The combination of Rbc, amoxicillin, clarithromycin and metronidazole for only 5 days represents a promising therapy for H. pylori infection, due to its high efficacy, simple posology, low cost and excellent tolerance. Mjör Abstract Aim To study the structure of the apical region of human teeth with emphasis on dentinal tubules and their branches. Methodology This descriptive histological study employed demineralized stained sections for light microscopy, demineralized unstained sections for scanning electron microscopy, and undemineralized, acid-etched specimens for confocal tandem scanning microscopy. Results The apical portion of human teeth showed marked variations in structure, including accessory root canals, areas of resorption and repaired resorptions, occasional attached, embedded and free pulp stones, varied amounts of irregular secondary dentine, and even cementum-like tissue lining the apical root canal wall. The apex often deviated from the long axis of the root canal. Primary dentinal tubules were irregular in direction and density. Some areas were devoid of tubules. Conclusions The irregular and variable structure of the apical region of human teeth represent special challenges during endodontic therapy. Obturation techniques based on the penetration of adhesives into dentinal tubules are unlikely to be successful and adhesive techniques must depend on impregnation of a hybrid layer. [source] The functional anatomy of Kager's fat pad in relation to retrocalcaneal problems and other hindfoot disordersJOURNAL OF ANATOMY, Issue 1 2006P. Theobald Abstract Kager's fat pad is a mass of adipose tissue occupying Kager's triangle. By means of a combined magnetic resonance imaging, ultrasound, gross anatomical and histological study, we show that it has three regions that are closely related to the sides of the triangle. Thus, it has parts related to the Achilles and flexor hallucis longus (FHL) tendons and a wedge of fat adjacent to the calcaneus. The calcaneal wedge moves into the bursa during plantarflexion, as a consequence of both an upward displacement of the calcaneus relative to the wedge and a downward displacement of the wedge relative to the calcaneus. During dorsiflexion, the bursal wedge is retracted. The movements are promoted by the tapering shape of the bursal wedge and by its deep synovial infolds. Fibrous connections linking the fat to the Achilles tendon anchor and stabilize it proximally and thus contribute to the motility of its tip. We conclude that the three regions of Kager's fat pad have specialized functions: an FHL part which contributes to moving the bursal wedge during plantarflexion, an Achilles part which protects blood vessels entering this tendon, and a bursal wedge which we suggest minimizes pressure changes in the bursa. All three regions contribute to reducing the risk of tendon kinking and each may be implicated in heel pain syndromes. [source] Effect of dietary linoleic acid concentration and vitamin E supplementation on cell desquamation and susceptibility to oxidative damage of pig jejunal mucosaJOURNAL OF ANIMAL PHYSIOLOGY AND NUTRITION, Issue 1-2 2001C. J. López Bote Sixty Large White × Great York pigs weighing approximately 60 kg were individually fed on six experimental diets. The experiment was organized in a 3 × 2 factorial arrangement design with three different fat sources and a basal or supplemented (200 mg/kg diet) level of ,-tocopheryl acetate. All diets contained the same concentration of saturated fatty acids (15 ± 0.2 g/kg dry matter) but differed in the concentration of C18 : 2 (14 ± 0.5, 18 ± 0.4 and 21 ± 0.6 g/kg) and monounsaturated fatty acids (19 ± 0.2, 15 ± 1.2 and 10 ± 1.3 g/kg). No effect of dietary treatment was observed on weight gain and feed consumption. A histological study of the jejunal mucosa showed lower cell desquamation in groups containing a supplemental level of ,-tocopheryl acetate (p=0.080). A higher cell desquamation was found in the groups fed diets containing the higher concentration of C18 : 2 (p=0.087). We also observed an interaction effect (p < 0.001) of dietary fat source and vitamin E supplementation on jejunal cell desquamation in which the effect of dietary vitamin E was lower when diets with a low concentration of C18 : 2 were administered. In vitro- induced oxidation of jejunal mucosa homogenates was lower in pigs fed diets supplemented with ,-tocopheryl acetate (p < 0.002). The dietary concentration of C18 : 2 significantly affected oxidation of pig jejunal mucosa (p < 0.002). [source] On the vascularization and structure of the skin of a Korean bullhead Pseudobagrus brevicorpus (Bagridae, Teleostei) based on its entire body and appendagesJOURNAL OF APPLIED ICHTHYOLOGY, Issue 1 2010J. Park Summary To investigate the vascularization and structure of the skin and its relationship to cutaneous respiration in Pseudobagrus brevicorpus, a histological study by light microscopy was carried out on 15 regions of the skin, including eight body regions, six fins and the barbel. The skin consisted of the epidermis, dermis and subcutis in all regions, except for the barbel that had a relatively thin dermis and subcutis. The epidermis was composed of the outermost layer, the middle layer and the stratum germinativum. There were two kinds of gland cells: the unicellular mucus cells and large club cells. The middle layer had a small number of fine blood capillaries accompanied by dermal collagen in all regions; the mean number of blood capillaries ranged from 0.9 to 5.9. The mean diffusion distance between the capillary endothelial cells and the surface of the epidermis ranged from 50.6 to 126.8 ,m. Based on these intra-epithelial blood capillaries, the relative surface area of the respiratory epithelium ranged from 0.1 to a maximum value of 1.2%. The dermis lacking scales had collagen bundles arranged parallel to each other, but vertical fiber bundles around the dorso-lateral regions were seen at intervals. Sensory organs such as taste buds, pit organs and lateral canals were found whereby the taste buds in particular were more abundant in the epidermis of the barbel. The vascularization of the skin may be closely related to an additional respiratory system used to deal with an extreme hypoxic condition during dry seasons. [source] Chitosan scaffolds for in vitro buffalo embryonic stem-like cell culture: An approach to tissue engineeringJOURNAL OF BIOMEDICAL MATERIALS RESEARCH, Issue 1 2007Wah W. Thein-Han Abstract Three-dimensional (3D) porous chitosan scaffolds are attractive candidates for tissue engineering applications. Chitosan scaffolds of 70, 88, and 95% degree of deacetylation (% DD) with the same molecular weight were developed and their properties with buffalo embryonic stem-like (ES-like) cells were investigated in vitro. Scaffolds were fabricated by freezing and lyophilization. They showed open pore structure with interconnecting pores under scanning electron microscopy (SEM). Higher % DD chitosan scaffolds had greater mechanical strength, slower degradation rate, lower water uptake ability, but similar water retention ability, when compared to lower % DD chitosan. As a strategy to tissue engineering, buffalo ES-like cells were cultured on scaffolds for 28 days. It appeared that chitosan was cytocompatible and cells proliferated well on 88 and 95% DD scaffolds. In addition, the buffalo ES-like cells maintained their pluripotency during the culture period. Furthermore, the SEM and histological study showed that the polygonal buffalo ES-like cells proliferated well and attached to the pores. This study proved that 3D biodegradable highly deacetylated chitosan scaffolds are promising candidates for ES-like cell based tissue engineering and this chitosan scaffold and ES cell based system can be used as in vitro model for subsequent clinical applications. © 2006 Wiley Periodicals, Inc. J Biomed Mater Res Part B: Appl Biomater, 2007 [source] Non-alcoholic fatty liver disease in Malaysia: A demographic, anthropometric, metabolic and histological studyJOURNAL OF DIGESTIVE DISEASES, Issue 1 2007Abdul MALIK BACKGROUND AND OBJECTIVES: Nonalcoholic fatty liver disease (NAFLD) is increasing rapidly in the Asia,Pacific region. There has been a paucity of studies from the region. The aims of this study were to define the demographic, anthropometric, metabolic and histological characteristics of patients with NAFLD in our local population and to determine independent predictors of severe liver fibrosis. METHODS: Patients with persistently raised liver enzymes and/or fatty liver detected on ultrasonography with exclusion of other liver disorders were prospectively recruited. Their insulin resistance was assessed using the homeostasis model assessment of insulin resistance score. A liver biopsy was performed in all cases for grading (for steatohepatitis) and staging (for fibrosis) of NAFLD. Independent risk factors for fibrosis were determined using multiple logistic regression analysis. RESULTS: Seventy-five patients were recruited: 39 men (52%) and 36 women (48%). The mean age of the patients was 47.0 ± 12.2 years. Of these, 58 patients (77.3%) were centrally obese, 29 patients (38.7%) were diabetic and 15 patients (20.0%) had impaired glucose tolerance. Insulin resistance was diagnosed in 62 out of 64 (96.9%) patients. Benign steatosis, nonalcoholic steatohepatitis and cirrhosis were diagnosed in three (4.3%), 59 (84.3%) and eight (11.4%) of 70 patients, respectively. Significant independent predictors of liver fibrosis were; male sex (P = 0.019, OR = 5.55, CI = 1.33,23.18) and Indian race (P = 0.013, OR = 8.21, CI = 1.56,43.16). CONCLUSIONS: The full histological spectrum of NAFLD was seen in our patients. The majority of patients were insulin resistant, centrally obese and either diabetic or had impaired glucose tolerance. The predictors of severe liver fibrosis were male sex and Indian race. The disease has complete penetrance in adults and variable expressivity. It is caused by mutations in the ATP2A2 gene, which encodes the sarco/endoplasmic reticulum Ca2+ ATPase type 2 isoform (SERCA2). Objective, We report histological investigations of six unrelated Tunisian families including 15 affected individuals with Darier's disease mutations. Results, The typical histological features of Darier's disease have been observed in the 15 patients. Variable histological features have been observed among Tunisian patients ranging from mild to moderate lesions of Darier's disease. A significant correlation has been observed between the clinical presentation of the Darier's disease (mild or moderate) and the intensity of the histological features. Isolated acral form of Darier's disease was seen in one case. Two distinct original associations have been observed: Darier's disease/pemphigus vulgaris in one patient and Darier's disease/ichtyosis in the other patient. Conclusion, Our findings confirmed the clinical heterogeneity of Darier's disease on the basis of histological study. The intensity of the histological features could be closely correlated to the severity of Darier's disease clinical presentation. This histological study compares laser class V preparations with conventional treatment to evaluate the resulting formation at the cavity walls. Study Design/Materials and Methods Four class V preparations (one made with a diamond drill and three with the CO2 laser (9.6 ,m, 60 microseconds pulse width, 40 mJ pulse energy, 100 Hz, integrated scanner system, water cooling) were performed on ten extracted teeth. The cavities were filled with a composite resin partly including enamel and dentine conditioning. Results After laser preparation, no cracks or signs of carbonisation were detected. The results were comparable to those attained with conventional treatment. Following cavity filling without prior conditioning, gaps were noted at the cavosurface indicating a lack of adhesion. [source] Antivascular Tumor Eradication by Hypericin-mediated Photodynamic Therapy,PHOTOCHEMISTRY & PHOTOBIOLOGY, Issue 5 2002Bin Chen ABSTRACT Photodynamic therapy (PDT) with hypericin has been shown to inhibit tumor growth in different tumor models, and tumor vascular damage was suggested to be mainly responsible for the antitumoral effect. Here, we demonstrate tumor vascular damage and its consequence on local tumor control after hypericin-mediated PDT by using both short and long drug,light intervals. Radiation-induced fibrosarcoma-1 tumors were exposed to laser light at either 0.5 or 6 h after a 5 mg/kg dose of hypericin. Tumor perfusion was monitored by fluorescein dye,exclusion assay and by Hoechst 33342 staining of functional blood vessels. Significant reduction in tumor perfusion was found immediately after both PDT treatments. A complete arrest of vascular perfusion was detected by 15 h after the 0.5 h-interval PDT, whereas well-perfused areas could still be found at this time in tumors after the 6 h-interval PDT. A histological study confirmed that primary vascular damage was involved in both PDT treatments. Tumor cells appeared intact shortly after light treatment, degenerated at later hours and became extensively pycnotic at 24 h after the 0.5 h-interval PDT. PDT under this condition led to complete tumor cure. In contrast, significant numbers of viable tumor cells, especially at the tumor periphery, were found histologically at 24 h after the 6 h-interval PDT. No tumor cure was obtained when PDT was performed at this time. Our results strongly suggest that targeting the tumor vasculature by applying short drug,light interval PDT with hypericin might be a promising way to eradicate solid tumors. [source] Follicular, Oocyte and Embryo Features Related to Metabolic Status in Primiparous Lactating does Fed with High-Fibre Rearing DietsREPRODUCTION IN DOMESTIC ANIMALS, Issue 5 2010M Arias-Álvarez Contents Fertility of primiparous lactating does in the early postpartum (pp) period is very low mainly due to pronounced deficient energy intake, influencing oocyte and embryo developmental competence. The hypothesis used in this work was that high-lignin fibre diet supplied during the rearing period could increase feed intake and, consequently, improve the reproductive physiology and metabolic status of primiparous does in the early pp period. Diets with high-lignin [HL: 15.8% dry matter (DM)] or standard-lignin content (SL: 4.9% DM) were supplied until parturition time. No diet effects in serum oestradiol, progesterone concentrations and follicle categories were found in the histological study. Metaphase II rate of in vitro -matured oocytes was significantly higher in the SL vs the HL group (p < 0.001). Cytoplasmically degenerated oocytes (in terms of abnormal distribution of cortical granules) and follicular atresia rate were significantly lower in the SL group than in the HL group (p < 0.05 and p < 0.005 respectively). In addition, HL-fed does showed lower number of viable embryos and higher rate of retarded in vivo -recovered embryos compared with the SL group (p < 0.05). Neither in vitro embryo development of viable embryos nor conception rate was significantly different between groups. Feed intake increased during the first pregnancy in the HL group (p < 0.05), but not during early lactation. Serum protein, non-esterified fatty acid and leptin concentrations, as well as estimated body composition were similar in does fed with both diets. In conclusion, the enhancement of reproductive management by using highly lignified products in rearing diets does not seem to report physiological reproductive benefits affecting oocyte maturation rate and embryo viability in primiparous lactating does. [source] Effects of antisense interleukin-5 gene transferred by recombinant adeno-associated virus to allergic ratsRESPIROLOGY, Issue 1 2010Daxiong ZENG ABSTRACT Background and objective: The accumulation of eosinophils in airways is an important characteristic of asthma. The process is primarily mediated by interleukin-5 (IL-5) secreted by Th2 lymphocytes. This study explored a new approach to asthma therapy in which allergic rats were transfected with the IL-5 antisense gene delivered by the recombinant adeno-associated virus (rAAV-ASIL-5). Methods: The viral vector rAAV-ASIL-5 was constructed and the IL-5 antisense gene transfected into allergic rats. The levels of IL-5, IgE, eotaxin and eosinophilic cationic protein (ECP) in sera and bronchoalveolar lavage fluid (BALF) were measured by ELISA. The inflammatory responses in lung tissues were evaluated by histological study. Results: The levels of IL-5 protein in serum and BALF were significantly decreased in the allergic rats treated with rAAV-ASIL-5 (P < 0.05). Serum ovalbumin-specific IgE was reduced in treated rats compared with untreated rats (P < 0.05). rAAV-ASIL-5 treatment also reduced eosinophils in the peripheral blood and BALF, as well as the ECP and eotaxin levels in serum and BALF (P < 0.05). There was significantly less inflammation in the lungs of rAAV-ASIL-5-treated rats than in those of untreated rats. No obvious pathological damage to the kidneys and livers of the rats treated with rAAV was observed. Conclusions: Treatment with rAAV-ASIL-5 inhibited the accumulation of eosinophils and airway inflammation in the rat model of allergic asthma by suppressing IL-5 production. These results suggest that rAAV-ASIL-5-based gene therapy may be used for the treatment of allergic asthma. The aims of the present study were to directly detect ROS in the tympanic membrane and middle ear mucosa of rats by measuring luminol amplified chemiluminescence, to evaluate the changes in the levels of ROS after treatment with vitamin E, and to examine the possible changes in the tympanic membranes otomicroscopically and histologically. Study Design Prospective controlled animal study. Methods Forty healthy Sprague-Dawley rats were divided into five groups of eight animals each. Animals in all groups except group 1 were bilaterally myringotomized. Group 2 received no treatment, group 3 was treated with topical olive oil, group 4 received topical vitamin E, and group 5 received intramuscular vitamin E. After 24 hours of myringotomy, tympanic membranes were examined otomicroscopically; thereafter, tympanic membranes and middle ear mucosa were peeled off. The right ears of the animals were used for biochemical assay, and the left ears were used for histological study. Results Reactive oxygen species levels were significantly decreased in group 4 with topical application of vitamin E compared with untreated and myringotomized animals in group 2. Reactive oxygen species levels were also decreased in group 5, although the decrease was not statistically significant when compared with groups 2 and 3. Histological studies confirmed sclerotic changes in the untreated myringotomized animals. The tympanic membranes of animals in groups 2 and 3 showed a white, chalk-like pattern of sclerotic changes, whereas animals in groups 4 and 5, with the exception of two animals in group 5, lacked these changes. Conclusion Although the relationship between the development of myringosclerosis and ROS had been well documented previously, the present study is the first that has directly measured the levels of ROS in the tympanic membrane and middle ear mucosa. These results are relevant because they correlate with histological findings. It has also been demonstrated that topically applied vitamin E is effective in decreasing the ROS levels. [source] Effects of sublethal levels of tributyltin chloride on a new toxicity test organism, Liza saliens (osteichthyes, mugilidae): a histological studyAPPLIED ORGANOMETALLIC CHEMISTRY, Issue 6 2006P. D'Agati Abstract The histopathological effects of 10,7 and 10,9M tributyltin(IV)chloride,TBTCl, solutions on different Liza saliens organs have been studied by light microscope. The fish were sacrificed after 3,4 h incubation in 10,7M TBTCl solution or after 15 days incubation in 10,9M solution. The observed histopathological changes were dose- and time-dependent. The 10,7M TBTCl concentration resulted in major damage to the gill epithelium, indicating that TBTCl primarily interfered with the respiration, osmoregulation, acid balance and nitrogenous waste excretion processes. After incubation in 10,9M TBTCl solution the fish lived 20 or more days, but many of the organs were altered. Thymus atrophy, reduced spleen and altered head kidney were observed. These histological results indicated that TBTCl interfered with organ immunodefense and altered main metabolic pathways in Liza saliens. The presence of melano-macrophage centers, only in TBT-treated liver and spleen, can be considered a tool to facilitate, with other biomarkers, the detection of alterations by toxicants. Regarding the pancreas activity in 10,7M solutions, it has been noted that, in the exocrine cells, very few zymogen granules were still present and the Langerhans islets were more altered. In 10,9M solution the exocrine pancreatic cells had no granules and the islet cells presented degenerative alterations. In addition, TBTCl, which altered the pancreas and gonad morphology, could again be considered an endocrine disrupter even if biochemical data are still necessary. Finally, the Liza saliens juveniles could be considered an interesting biological model for experiments with contaminants, due to their ease of adaptation to experimental conditions and food chain position. One restenosis lesion had a bilayer structure of hyperechoic outer layers and highly echolucent inner layers on OCT images and histologically exhibited myxomatous neointima tissue in the highly echolucent regions; another restenosis case showed patchy and highly echolucent regions throughout the layers and its histology revealed fibrin thrombosis. We should be aware that patterns of echolucent on OCT images may have various histology. OCT allows the visualization of fine lesions that conventional intravascular ultrasound (IVUS) cannot provide. The OCT images of drug-eluting stent restenosis lesions often show echolucent regions [Shuzoh et al., EuroInterv 2006;1:484]. However, no histological study of the lesions has been reported to date. The purpose of the present study was to determine the relationship between oral Mg supplementation and changes in plasma glucose, calcium, haemogolobin, Ca/Mg ratio, blood pressure and the histology of the pancreas and vascular system in streptozotocin-induced diabetic rats. 2.,Ten days after the induction of diabetes in male Wistar rats, half the diabetic animals were divided into six groups, receiving 0, 1, 3, 10, 30 or 50 g/L MgSO4 added into the drinking water for 8 weeks. Plasma glucose and Mg were measured at days 1, 2, 3, 5, 7, 14 and 21 to find the optimum dose of Mg and the time-course of its effect. In addition, histological observations were undertaken. Eight weeks later, all animals were decapitated, the pancreas and thoracic aorta were removed carefully and immersed immediately in 10% formaldehyde for histological study. 3.,To evaluate the effects of Mg on plasma glucose, calcium, haemoglobin, Mg and blood pressure, another group of animals was divided into four experimental groups, as follows: (i) non-diabetic controls received tap water for 8 weeks; (ii) acute diabetics received tap water for 10 days; (iii) chronic diabetic controls received tap water for 8 weeks; and (iv) Mg-treated chronic diabetic rats received 10 g/L MgSO4 added into the drinking water 10 days after the induction of diabetes for 8 weeks. 4.,Magnesium dose dependently affects plasma glucose levels. The peak effect was reached during the first 24 h following oral administration. Administration of 10 g/L MgSO4 results in the return of normal structure in the diabetic pancreas and aorta. Moreover, this concentration of MgSO4 causes glucose, haemoglobin, calcium, the Ca/Mg ratio and blood pressure to reach normal levels. Although the Mg level increases slightly following the administration of 10 g/L MgSO4 to diabetic rats, it never reaches control levels. 5.,On the basis of the results of the present study, it may be concluded that chronic Mg administration may have beneficial effects on diabetes. [source] Utility of fine-needle aspiration for diagnosis of carcinoma associated with multinodular goitreCLINICAL ENDOCRINOLOGY, Issue 6 2004Antonio Ríos Summary background, Fine-needle aspiration (FNA) is a useful method for evaluating a solitary thyroid nodule; however, this is not an agreed method for a multinodular goitre (MNG). The aim of this study was to assess the utility of preoperative FNA for detecting malignancy in MNG. patients and method, We analysed operated MNGs in which FNA had been performed. Puncture was carried out on the dominant nodule and any other nodules with features suggesting malignancy. The diagnosis was classed as colloid, follicular or Hürthle proliferation, suggestive of malignancy, haematic and inadequate. The thyroid FNA results, grouped into suggestive of malignancy (positive result) and other diagnoses (negative result), were compared to those of the final histological study in order to calculate the value of the test in diagnosing malignancy. results, FNA was performed in 432 MNGs, of which 42 (9·7%) were associated with carcinoma. Overall, the results of the test were poor, revealing a sensitivity of 17%, specificity of 96% and diagnostic accuracy of 88%, with a positive predictive value of 32% and negative predictive value of 88%. When the values were recalculated with the exclusion of microcarcinomas ,considering their minor clinical importance , there was a slight improvement in the results: the sensitivity increased to 26%, diagnostic accuracy to 93% and negative predictive value to 96%. However, the specificity remained at 96%, and the positive predictive value fell from 32% to 25%. The results of the test improved in multifocal carcinomas. conclusions, Thyroid fine needle aspiration is not useful for differentiating MNG with malignant degeneration from benign MNG, as more than 80% of carcinomas go unnoticed; it provides a sensitivity of 17% for detecting carcinomas, rising to 26% if microcarcinomas are excluded. We therefore suggest that clinical criteria should prevail over FNA. [source] Early cortical bone healing around loaded titanium implants: a histological study in the rabbitCLINICAL ORAL IMPLANTS RESEARCH, Issue 2 2009Elke Slaets Abstract Objectives: To identify the role of immediate implant loading on the early phases of the bone healing responses. Material and methods: Implants were placed in rabbit tibial diaphyses and left to heal for 3, 7, 14, 28 or 42 days. Half of the animals received an immediate loading protocol of 2.2 N at 3 Hz for 1800 cycles and 5 days/week, whereas the others served as unloaded controls. Histological assessment was combined with histomorphometrical measurements. Results: At early time-points, an endosteal and periosteal new bone formation was found, while the cortex itself contained damaged osteocytes. At later time-points, new bone formation was also found at the cortical level itself. Differences between groups were found mainly in this new bone formation process, with larger reactions for the endosteal and periosteal bone in the loaded group after 28 and 42 days, respectively. At the end-point of the experiment, bone formation at the cortical level was reduced in the loaded group compared with the control group. Conclusions: These results show that the immediate loading protocol caused no differences in the sequential events leading to osseointegration in cortical bone. However, the processes of new bone formation originating from the endosteum and the periosteum lasted longer compared with the unloaded controls. [source] The effect of three different calcium phosphate implant coatings on bone deposition and coating resorption: a long-term histological study in sheepCLINICAL ORAL IMPLANTS RESEARCH, Issue 3 2005Christian Schopper Abstract: The present study investigated the hypothesis that hydroxyapatite (HA), tricalcium phosphate (TCP), and a HA-gel coated on endosseous titanium (Ti) implants by spark discharging (SD) and dip coating would achieve predictable osseointegration without evident bioresorption of the coatings on the long term. A costal sheep model was used for the implantation of the HA/SD, HA/TCP/SD, and HA-gel/SD specimens, which were retrieved 6 and 12 months following implantation. HA and Ti coatings on implants obtained by conventional plasma spraying (HA/PS, Ti/PS) were used as controls. Microscopy showed that osseointegration was achieved from all types of implants. No evidence for bioresorption of the HA/SD, HA/TCP/SD, and HA-gel/SD coatings was present but cohesive failure with disruption of the coating/implant interface was seen. A statistical analysis of the histomorphometrical data showed no time-dependent effect, however. HA/PS coatings achieved significantly higher bone,implant contact (BIC) percentages of the total implant surface (toBIC) than the other types of coatings (P=0.01). If the BIC percentages were traced separately for implant portions placed into cortical and cancellous bone (coBIC and caBIC, respectively), detailed analysis showed that the caBIC values of HA-gel/SD and HA/PS coatings were significantly higher than that of the other types of coatings (P=0.01). CaBIC values were highly correlated with toBIC values (P<0.001). The present study showed that the preparation techniques used produced thin, dense, and unresorbable coatings that achieved osseointegration. Compared with the control coatings, however, only HA-gel/SD coating can be recommended from the investigated preparation techniques for a future clinical use if a better coating cohesion is achieved. Résumé L'étude présente a étudié l'hypothèse que le recouvrement par de l'hydroxyapatite, du phosphate tricalcique et un gel d'hydroxyapatite sur les implants en titane par décharges spark et recouvrement par trempage pourrait apporter une ostéïntégration prévisible sans biorésorption importante des recouvrements à long terme. Un modèle de mouton a été utilisé pour l'implantation de spécimens HA/SD, HA/TCP/SD et gel-HA/SD qui ont été enlevés six et douze mois après leur insertion. Les implants recouverts d'hydroxyapatite et de titane obtenus par plasma-spray conventionnel (HA/PS et Ti/PS) ont été utilisés comme contrôles. La microscopie a montré que l'ostéoïntégration a été réalisée pour tous les types d'implants. Aucune évidence pour la biorésorption de HA/SD, HATCP/SD, et gel-HA/SD n'était présente mais un échec de cohésion avec destruction de l'interface implant/recouvrement a été mis en évidence. Une analyse statistique des données histomorphométriques ne montrait cependant aucun effet dépendant du temps. Die Proben wurden 6 und 12 Monate nach Implantation entnommen. Als Kontrolle dienten Hydroxyapatit- und Titanbeschichtungen (HA/PS und Ti/PS), welche mittels Plasmaspray aufgetragen worden waren. Die mikroskopische Untersuchung zeigte, das bei allen Implantattypen eine Osseointegration erreicht wurde. Bei den HA/SD, HA/TCP und HA-Gel/SD Beschichtungen waren keine Anzeichen von Bioresorption vorhanden, aber es konnten kohäsive Misserfolge mit Abrissen im Bereich der Implantat/Beschichtung-Berührungsfläche gesehen werden. Eine statistische Analyse der histomorphometrischen Daten zeigte jedoch keinen zeitabhängigen Effekt. Die caBIC Werte zeigten eine starke Korrelation mit den toBIC Werten (P<0.001). Die Studie zeigte, dass das verwendete Herstellungsverfahren dünne, dichte und nicht resorbierbare Beschichtungen ergab, welche eine Osseointegration erreichten. Im Vergleich mit den Kontrollbeschichtungen können jedoch nur die HA-Gel/SD Beschichtungen der untersuchten Herstellungsverfahren für den weiteren klinischen Gebrauch empfohlen werden, falls eine bessere Kohäsion der Beschichtung erreicht werden kann. Resumen El presente estudio investigó la hipótesis de que la hidroxiapatita, el fosfato tricálcico y un gel de hidroxiapatita cubriendo implantes endoóseos de Ti por medio de chisporroteo e inmersión pueden lograr una osteointegración predecible sin una biorreabsorción evidente de las cubiertas a largo plazo. Se usó un modelo costal de oveja para la implantación de especímenes HA/SD, HA/TCP/SD, y gel-HA/SD que se retiraron a los 6 y a los 12 meses de la implantación. Como control se usaron cubiertas de hidroxiapatita y titanio en implantes obtenidos por medio de pulverización de plasma convencional (HA/SD, Ti/PS). La microscopía demostró que la osteointegración se logró en todos los tipos de implantes. No existió evidencia de biorreabsorción de las cubiertas HA/SD, HA/TCP/SD, y gel-HA/SD pero se observó fallos en la cohesión con disrupción de la interfase cubierta/implante. Un análisis estadístico de los datos histomorfométricos no mostró, sin embargo efectos dependientes del tiempo. Las cubiertas HA/PS lograron unos porcentajes de contacto hueso-implante significativamente mayores del total de la superficie del implante (toBIC) que los otros tipos de cubiertas (P=0.01). Si se ubicaran los porcentajes de contacto hueso-implante separadamente para porciones situadas dentro de hueso cortical o esponjoso (coBIC y caBIC respectivamente), un análisis detallado mostró que los valores caBIC de las cubiertas de HA-gel/SD y HA/PS fueron significativamente mayores que aquellos de los otros tipos de cubiertas (P<0.001). El presente estudio mostró que las técnicas de preparación usadas produjeron cubiertas finas, densas y no reabsorbibles que alcanzaron la osteointegración. De todos modos, comparadas con las cubiertas de control, solo la cubierta HA-gel/SD pude ser recomendada desde las técnicas de preparación investigadas para un futuro uso clínico si se lograse una mejor cohesión de cubierta.
