    Photodynamic Therapy of Cutaneous Lymphoma Using 5-Aminolevulinic Acid Topical Application

    Background. Photodynamic therapy (PDT) with topical application of 5-aminolevulinic acid (ALA) is a new and effective modality for treatment of superficial basal and squamous cell carcinomas. Objective. We present the kinetics of ALA-induced protoporphyrin IX (PP) accumulation and the results of ALA PDT treatment on two patients with different stages (stage I and stage III) of mycosis fungoides (MF)-type cutaneous T-cell lymphoma (CTCL). Methods. ALA-Decoderm cream was applied to the lesions for 16 hours. Spectrofluorescence measurements of PP accumulation were carried out before, during, and 1 hour after photoirradiation (580,720 nm) using the VersaLight system. Results. Different patterns of PP fluorescence kinetics were observed in patients with early and advanced stages of the disease. During photoirradiation the intensity of fluorescence decreased depending on the lesion thickness. One hour after the photoirradiation procedure no PP fluorescence was observed in the stage I MF lesion, while in the thick stage III MF lesions, PP fluorescence reappeared; after an additional 10,15 minutes of irradiation PP fluorescence disappeared. Complete response with excellent cosmetic results was observed in the stage I lesion after a single irradiation with a light dose of 170 J/cm2; in five stage III lesions, complete response was achieved after fractionated irradiation with a total light dose of 380 J/cm2 (follow-up at 27 and 24 months, respectively). Conclusion. The results showed a high response of both stage I and stage III MF lesions to ALA PDT. This modality appears to be very effective and can be used successfully for MF treatment.     Electrocatalytic Oxidation of Sulfur Containing Amino Acids at Renewable Ni-Powder Doped Carbon Ceramic Electrode: Application to Amperometric Detection L -Cystine, L -Cysteine and L -Methionine

    Abstract A sol-gel technique was used here to prepare a renewable carbon ceramic electrode modified with nickel powder. Cyclic voltammograms of the resulting modified electrode show stable and a well defined redox couple due to Ni(II)/Ni(III) system with surface confined characteristics. The modified electrode shows excellent catalytic activity toward L -cystine, L -cysteine and L -methionine oxidation at reduced overpotential in alkaline solutions. In addition the antifouling properties at the modified electrode toward the above analytes and their oxidation products increases the reproducibility of results. L -cystine, L -cysteine and L -methionine were determined chronoamperometricaly at the surface of this modified electrode at pH range 9,13. Under the optimized conditions the calibration curves are linear in the concentration range 1,450,,M, 2,90,,M and 0.2,75,,M for L -cystine, L -methionine and L -cysteine determination, respectively. The detection limit and sensitivity were 0.64,,M, 3.8,nA/ ,M for L -cystine, 2,,M, 5.6,nA/ ,M for L -methionine and 0.2,,M and 8.1,nA/,M for L -cysteine. The advantageous of this modified electrode is high response, good stability and reproducibility, excellent catalytic activity for oxidation inert molecules at reduced overpotential and possibility of regeneration of the electrode surface by potential cycling for 5,minutes. Furthermore, the modified electrode has been prepared without using specific reagents. This sensor can be used as an amperometric detector for disulfides detection in chromatographic or flow systems.     Inclusion Complexes for Use in Room-Temperature Gas-Sensor Design

    Abstract The inclusion complex [{Co(bpy)(H2O)4}·(fcds)]n (1), which has been constructed using the guest molecule ferrocene-1,1,-disulfonate (fcds), the bridging ligand 4,4,-bipyridine (bpy) and d7 Co2+, contains an infinite zigzag chain formed by the central CoII ion and the bridging bpy ligand. Guest fcds molecules lie between two adjacent zigzag chains. The highly conjugated structure of complex 1 means that it can be used as a metal-organic semiconductor, and it also shows a high response to liquefied petroleum gas (LPG) and ethanol/petroleum ether (EP) at room temperature. The inclusion complexes [{Co(bpp)2(H2O)2}·(fcds)·4H2O]n [2; bpp = 1,3-bis(4-pyridyl)propane] and [{Zn(bpy)(H2O)4}·(fcds)]n (3), on the other hand, cannot be employed as room-temperature gas sensors because they are insulators. The electrical resistivity of inclusion complex [{Ni(bpy)(H2O)4}·(fcds)]n (4) is 621 M,, whereas that of [{Co(bpy)(H2O)4}SO4·(4-abaH)2·3H2O]n (5) (4-abaH = 4-aminobenzoic acid) is only 137 M,. This means that the semi-conducting properties of such inclusion complexes depend on both the conjugated structure and the central metal ions. Furthermore, conjugated inclusion complexes with an odd number of electrons could be useful for the design of highly selective room-temperature gas sensors.(© Wiley-VCH Verlag GmbH & Co.     Evaluation of tissue culture response from mature seeds of Panicum spp.

    Abstract The genus Panicum contains important warm-season forage grasses and species with potential as biomass crops. We selected Panicum genotypes with high response to tissue culture for genetic improvement. The highest frequency of callus induction from mature seed of Panicum maximum cultivar Natsukaze was obtained on MS medium containing 4.0 mg L,1 2,4-dichlorophenoxyacetic acid and solidified with 0.3% Gelrite. We compared germination frequencies and callus induction capacities among 24 genotypes of 11 Panicum species on this medium. Callus induction frequencies varied among genotypes. Those with high germination frequencies generally had high callus induction frequencies. On the other hand, especially in P. maximum, the callus induction ratio (callus induction frequency/germination frequency) depended on the reproductive mode and ploidy. The callus induction ratio of three sexual accessions of P. maximum were very low compared to apomictic accessions, and besides, a tetraploid sexual accession Noh PL1 had very low germination and callus induction frequencies. Callus induction and regeneration capacities were independent of each other. For shoot regeneration, we transferred callus derived from the 24 genotypes onto MS medium supplemented with 1.0 mg L,1 kinetin and 0.4% Gelrite. Six of the genotypes regenerated plantlets. Among them, Panicum meyerianum produced the highest shoot regeneration frequency of 61.6% and the maximum number of shoots callus,1 in the shortest time. The callus of P. meyerianum also showed vigorous proliferation. We thus selected high-response genotypes of P. meyerianum. [source]


    Basal serum testosterone as an indicator of response to clomiphene treatment in human epididymis, seminal vesicles and prostate

    Summary. The present study was designed to determine the response of human epididymis, seminal vesicles and prostate function after a 5-day course of clomiphene citrate in men attending an infertility service. In 45 men, the secretions of the epididymis, seminal vesicles and prostate were assessed by measurements of seminal ,-glucosidase, fructose and acid phosphatase, respectively. Subjects were classified as normal or abnormal: abnormal men were defined as those who either had history of a sexually transmitted disease (STD), leukocytosper-mia, hypoandrogenism, or a low response of Leydig cells to clomiphene stimulation; and normal subjects were those who did not have these conditions. Mean serum testosterone luteinizing hormone (LH) and follicle stimulating hormone (FSH) levels were significantly increased after the short course with clomiphene citrate. After clomiphene citrate stimulation, the men in the normal group showed significantly increased marker levels of the seminal vesicles (P <0.02) and prostate (P <0.05), but not of the epididymis (P: NS). Men classified as abnormal showed no response according to the markers of the seminal vesicles and epididymis. Men with history of STD and abnormal basal values of acid phosphatase did not respond to the treatment. Men with history of STD but normal basal values of seminal acid phosphatase increased significantly in their levels of seminal acid phosphatase after clomiphene stimulation (P <0.02). Multivariate analysis showed that the basal serum testosterone level was the only variable in predicting the probability of response to the clomiphene in terms of true-corrected seminal fructose, seminal acid phosphatase and seminal ,-glucosidase levels. In fact, a high response of the seminal vesicles was observed in men with the highest basal serum testosterone levels (0.45 *** 0.17; coefficient of regression *** standard error; P <0.018). However, a high response in terms of seminal acid phosphatase (P <0.004) or ,-glucosidase (P <0.037) was observed in men with low basal serum testosterone levels. In conclusion, in the normal men, true-corrected fructose and acid phosphatase but not ,-glucosidase in semen increased after duplicate androgen stimulation. An absence of response was observed in cases with history of STD/leukocytospermia or hypoandrogenism. [source]


    Should fimbriae be included in pertussis vaccines?

    Studies on ELISA IgG anti-Fim2/3 antibodies after vaccination, infection
    The anti-Fim response and long-term persistence after vaccination and infection may be of importance in understanding population immunity. Longitudinal serum samples (n = 1330) from 542 non-infected children related to a Swedish vaccine trial showed that the post vaccination (DTPa5) antibody decay curve for pertussis ELISA IgG anti-fimbriae2/3 (anti-Fim2/3) was bi-phasic. A slower one followed an initial rapid decay approximately 5,6 months after the third dose at 12 months of age. After 71 months, however, 60% still had concentrations above ,5 EU/ml, a level that had been shown to correlate with decreased risk of disease. Booster responses after re-vaccination with DTPa5 at 4, 5 and 6 years of age were strong and appeared within 1 week after vaccination, indicating immune memory. Ninety-six young children with verified pertussis infection, for whom we had serum samples both before, during and after the infection, showed a high response if they had been primed with fimbriae (either DTPa5 or DTPwc). In contrast, 76% of infected children not primed with fimbriae (a DTPa2 or DT group) only had concentrations below the minimum level of detection in all samples taken during and after the infection. In two Swedish seroepidemiological surveys, one from 1997 just after reintroduction of universal childhood vaccination against pertussis and one from 2007, the proportion of children 2,3 years with anti-Fim2/3 concentrations <5 EU/ml was similar and above 90%. This reflects that the two- or three-component pertussis vaccines (DTPa2 and DTPa3) that were introduced in Sweden in 1996 do not induce anti-Fim2/3 antibodies. In previous studies it was shown in multivariate analyses that levels of IgG anti-Fim2/3 ,5 EU/ml reduced short-term risk of pertussis in small children. As the antibody response to Fim2/3 after infection is poor in children who have not been primed earlier in life, inclusion of immunogenic Fim2/3 in future pertussis vaccines should be considered.     Olfactory responses of two species of grasshoppers to plant odours

    Abstract Electroantennogram (EAG) responses were recorded from two species of oedipodine grasshoppers, Oedaleus decorus asiaticus L. (graminivorous) and Angaracris barabensis Pall. (forbivorous), to volatiles emitted by chopped leaves of ten plant species. Male O. d. asiaticus showed much stronger EAG responses than conspecific females and both sexes of A. barabensis. Sexual differences in EAG responses correspond to different numbers of antennal sensilla of both sexes and to certain behavioural and morphological factors as well. The overall EAG response profiles of the two grasshopper species to the ten plant odours were similar. However, adaptation to host odour might have occurred because of differences in their feeding habits. Females of the graminivorous O. d. asiaticus possess a significantly higher olfactory sensitivity for poaceous plant species than A. barabensis, while the forbivorous A. barabensis showed significantly higher EAG responses to Allium senescens (Liliaceae), and a tendency of high responses to composite plant species in comparison with O. d. asiaticus.     Predictive model for the outcome of infliximab therapy in Crohn's disease based on apoptotic pharmacogenetic index and clinical predictors

    Abstract Background: Infliximab (IFX) is an effective therapy for refractory luminal and fistulizing Crohn's disease (CD). Predictors of response could improve selection of patients with a higher probability of favorable outcomes and could improve the safety profile. We aimed to develop a predictive model for the response to infliximab in CD. Methods: Genetic and clinical data collected in a previous pharmacogenetic study of apoptosis genes were analyzed using SAS Enterprise miner modeling software and SPSS 12.0. We proposed a novel apoptotic pharmacogenetic index (API) with a score ranging from 0 (low apoptotic response) to 3 (high apoptotic response) and subsequently developed a decision tree model. Results: Response and remission rates significantly increased with API score (P = 0.005 in the group of patients with luminal CD, P = 0.02 in the group of patients with fistulizing CD). Patients with an API , 1 (n = 59) had the lowest response and remission rates in both the luminal CD (50% and 39.5%, respectively) and fistulizing CD (61.9% and 28.6%, respectively) groups, compared to those with an API of 2 (n = 158), whose response and remission rates were 73.8% and 56.1%, respectively, in the luminal CD group and 85.7% and 44.9%, respectively, in the fistulizing CD group; and those with an API of 3 (n = 10), whose response and remission rates were 100% and 85.7%, respectively, in the luminal CD group and 100% and 0% in the fistulizing CD group. Response in patients with an API , 1 was significantly influenced by concurrent azathioprine therapy in the luminal CD (21.4% versus 78.9%, P < 0.001) and in the fistulizing CD (46.6% versus 100%, P = 0.04) groups. In patients with an API of 2, we saw an interaction with age older than 40 years and location of disease (response 52.2% versus 83.9%, P = 0.008) in the luminal CD group and with baseline CRP greater than 5 mg/L (73.9% versus 93.9%, P = 0.04) in the fistulizing CD group. Conclusions: From our newly proposed apoptotic pharmacogenetic index and clinical predictors, we developed a model for prediction of low, medium, and high responses to the first infusion of IFX in patients with CD.
