Osteopontin expression correlates with invasiveness in cervical cancer

Aim: Osteopontin is a secreted, integrin-binding glycophosphoprotein that is overexpressed in many types of cancers and appears to be involved in carcinogenesis and cancer progression. To understand the role of osteopontin in carcinogenesis of cervical cancer, this study was designed to determine whether osteopontin is expressed in cervical cancer and carcinoma in situ (CIS) tissue as well as in normal cervical tissue. Methods: The expression of osteopontin was immunohistochemically analysed from 68 normal cervix, 55 CIS and 52 invasive cervical cancer tissues using a paraffin-embedded tissue array. Immunostaining was evaluated by intensity and the percentage of stained cells. Results: Osteopontin expression in normal, CIS and cervical cancer tissues was two of 68 (2.9%), 43 of 55 (78.2%) and 46 of 52 (88.4%), respectively (P < 0.01). High intensity (strong positive)/high proportion (more than 50%) staining seen in CIS and cervical cancer tissue samples was 45 of 55 (81.8%)/22 of 55 (40.0%) and 50 of 52 (96.2%)/31 of 52 (59.7%), respectively (P = 0.029 and P = 0.054). There was no significant correlation between the immunostaining score and stage and the immunostaining score and survival. Conclusion: Osteopontin may have a potential use as a diagnostic factor for cervical cancer and osteopontin expression is closely correlated with carcinogenesis and invasion of cervical cancer. Aetiology, clinical course and outcome of sporadic acute viral hepatitis in pregnancy,

summary. Hepatitis E causes large-scale epidemics in endemic areas. The disease, during epidemics, has increased incidence and severity in pregnant women. Sporadic acute viral hepatitis (AVH) is common in endemic areas. The relationship of sporadic AVH and pregnancy has not been well studied. Over a 3-year period we prospectively studied 76 pregnant women and 337 non-pregnant women of childbearing age with sporadic acute viral hepatitis for aetiology, clinical course and outcome of disease. The aetiology in sporadic AVH was hepatitis A virus (HAV) in six (1.5%), hepatitis B virus (HBV) in 62 (15%), hepatitis C virus (HCV) in seven (1.7%), hepatitis D virus (HDV) co-infection in six (1.5%), hepatitis E virus (HEV) in 205 (49.6%), and hepatitis non-A-to-E (HNAE) in 127 (30.7%). Sixty-five (85.5%) pregnant women and 140 (41.5%) nonpregnant women had hepatitis E. The proportion of pregnant women was 31.7% in HEV group and 5.3% in non-HEV group [P < 0.001; OR=8.3 (95%C1 4.2,16.3)]. The prevalence of HEV in pregnant women in first trimester (76.9%), second trimester (88.9%), third trimester (83.8%) and puerperium (100%) did not differ significantly (P=0.09). Forty-seven (61.8%) of the 76 pregnant women developed fulminant hepatic failure (FHF), 69.2% in HEV group and 10% in non-HEV group (P < 0.001). Thirty-four (10.1%) nonpregnant women developed fulminant hepatic failure, 10% in HEV group and 9.7% in non-HEV group (P=0.86). FHF had occurred in four (40%) of 10 patients with HE in first trimester as against 41 (74.5%) of 55 patients in second trimester and beyond (P=0.015). Amongst the major complications of fulminant hepatic failure, cerebral oedema (53.2%) and disseminated intravascular coagulation (21.3%) occurred more often in pregnant women than in nonpregnant women (29.4% and 2.8%; P=0.03 and 0.016, respectively) while infections occurred more often in nonpregnant women (36.1%) than in pregnant women (10.6%; P=0.003). Fifty (61.7%) patients with FHF died [25 (53.2%) pregnant women and 25 (69.5%) nonpregnant women (P=0.06)]. Cerebral oedema and HEV aetiology were independent variables of survival in patients with FHF. Patients with cerebral oedema had worse prognosis and patients with HEV aetiology had best chances of survival. Hence HEV was the most common cause of sporadic AVH in this endemic area. High proportion of pregnant women and increased severity of disease in pregnancy were limited to patients with hepatitis E. Sporadic AVH caused by agents other than HEV did not show any special predilection to or increased severity in pregnancy. FHF in pregnant women caused by HEV was an explosive disease with short pre- encephalopathy period, rapid development of cerebral oedema and high occurrence of disseminated intravascular coagulation and may represent a severe manifestation of a Schwartzmann-like phenomenon. High proportion of protected minke whales sold on Japanese markets is due to illegal, unreported or unregulated exploitation

Abstract Whale meat products sold on Japanese markets originate from two stocks of North Pacific (NP) minke whales Balaenoptera acutorostrata scammoni: the depleted J-stock, which has been protected since 1986 but continues to be killed as fisheries ,bycatch', and the more abundant O-stock, which is hunted under special permit (scientific whaling). We investigated the geographic distribution and temporal changes in stock composition of NP minke whale products sold on Japanese markets between December 1997 and June 2004. From nearly 1200 ,whale meat' products purchased during this time, 250 were identified as NP minke whales by phylogenetic analysis of mitochondrial DNA (mtDNA) sequences. The 250 NP minke whale products were found to represent 201 unique ,market individuals' after exclusion of replicate products using microsatellite genotypes. Market individuals were further classified into four mtDNA haplogroups, three of which are characteristic of the J-stock (J-type) and one characteristic of the O-stock (O-type). There were moderate differences in the proportions of J-type individuals found in coastal prefectures, perhaps reflecting regional differences in the sale of local bycatch, but no significant difference across time. The absence of a change over time was inconsistent with the four- to fivefold increase in reported bycatch, from an average of 25,122 whales year,1, following a 2001 regulation allowing commercial sale of whales taken as bycatch. Using a mixed-stock analysis based on haplogroup frequencies over the entire survey period, we estimated that 46.1% (se, 4.2%) of all market individuals originated from the J-stock. This estimate of illegal, unreported or unregulated (IUU) exploitation is higher than expected from the officially reported bycatch, suggesting either large-scale under-reporting and/or unrecognized takes of J-stock minke whales from Pacific coastal waters by the scientific hunt. Our estimates of the true level of IUU exploitation have important implications for recovery of this depleted coastal stock. [source]


The coupling of biological iron cycling and mineral weathering during saprolite formation, Luquillo Mountains, Puerto Rico

ABSTRACT Corestones of quartz diorite bedrock in the Rio Icacos watershed in Puerto Rico weather spheroidally to form concentric sets of partially weathered rock layers (referred to here as rindlets) that slowly transform to saprolite. The rindlet zone (0.2,2 m thick) is overlain by saprolite (2,8 m) topped by soil (0.5,1 m). With the objective of understanding interactions between weathering, substrate availability, and resident micro-organisms, we made geochemical and microbiological measurements as a function of depth in 5 m of regolith (soil + saprolite). We employed direct microscopic counting of total cell densities; enumeration of culturable aerobic heterotrophs; extraction of microbial DNA for yield calculations; and biochemical tests for iron-oxidizing bacteria. Total cell densities, which ranged from 2.5 × 106 to 1.6 × 1010 g,1 regolith, were higher than 108 g,1 at three depths: in the upper 1 m, at 2.1 m, and between 3.7 and 4.9 m, just above the rindlet zone. High proportions of inactive or unculturable cells were indicated throughout the profile by very low percentages of culturable heterotrophs (0.0004% to 0.02% of total cell densities). The observed increases in total and culturable cells and DNA yields at lower depths were not correlated with organic carbon or total iron but were correlated with moisture and HCl-extractable iron. Biochemical tests for aerobic iron-oxidizers were also positive at 0.15,0.6 m, at 2.1,2.4 m, and at 4.9 m depths. To interpret microbial populations within the context of weathering reactions, we developed a model for estimating growth rates of lithoautotrophs and heterotrophs based on measured substrate fluxes. The calculations and observations are consistent with a model wherein electron donor flux driving bacterial growth at the saprolite,bedrock interface is dominated by Fe(II) and where autotrophic iron-oxidizing bacteria support the heterotrophic population and contribute to bedrock disaggregation and saprolite formation. Sibling cannibalism in dorada under experimental conditions.

II. Effect of initial size heterogeneity, light regime on early cannibalism
Cannibalism among embryos and larvae of Brycon moorei (Characidae) occurs during daytime and night-time, and persists under permanent darkness. Embryos and larvae of dorada-provisioned with formulated feed over the first week of exogenous feeding did not survive, except for those exerting cannibalism. When offered alternative fish prey [embryos of Prochilodus magdalenae (0·5,0·8 mg) and Oreochromis niloticus (9,10 mg)], 1-day-old embryos of dorada preferred preying on these, thereby reducing early cannibalism. However, this promoted depensatory growth and more intense cannibalism later in the larval stage. Dorada provisioned with Artemia nauplii in excess showed more homogeneous growth and higher survival, most cannibalistic acts being restricted to the first 24 h of exogenous feeding, just after oral teeth were fully developed (21 h after hatching). Provisioning dorada with Artemia nauplii a few hours before their oral teeth were fully developed reduced early cannibalism from 41 to 15%. High proportions of deformed fish caused higher mortality, both directly and indirectly, as they promoted early cannibalism, depensatory growth and more intense cannibalism among larvae. The initial sorting of embryos, based on their occupation of the water column improved survival significantly during the first week of exogenous feeding, up to 52% in progenies containing <10% of deformed fish. Size-grading of larvae and young juveniles over the next 2 weeks reduced cannibalism to 2·6 and 1·9% day,, in the first and second weeks, respectively. These results indicate that cannibalism in dorada can be mitigated efficiently through appropriate rearing procedures, and open promising perspectives for the intensive culture of this fast-growing tropical species. Effects of dietary lipid levels on the growth, digestive enzyme, feed utilization and fatty acid composition of Japanese sea bass (Lateolabrax japonicus L.) reared in freshwater

Abstract Triplicate groups of 40 Japanese sea bass Lateolabrax japonicus reared in freshwater (average weight, 9.52±0.47 g) were fed with six isonitrogenous (,46% crude protein) diets containing 6%, 8%, 10%, 12%, 14% or 16% lipid for 10 weeks respectively. The results showed that the maximum weight gain (WG), specific growth rate (SGR), feed intake (FI) and protein efficiency ratio (PER) all occurred at the 10% lipid level (P<0.05) and growth depression occurred when the dietary lipid level was over 12%. Whole body and liver lipid concentrations were enhanced with the increase in the dietary lipid levels, but the muscle lipid content did not significantly change with the increase in the dietary lipid levels. Both liver pepsin and trypsin activities increased with dietary lipid levels ranging from 6% to 10%, and then decreased with a further increase in the dietary lipid content. Liver lipase activities showed a positive correlation with dietary lipid levels, but amylase activities were not markedly influenced by dietary lipid levels. High proportions of 18:1n-9, 20:1n-9, eicosapentaenoic acid (20:5n-3; EPA), 22:1n-11 and docosahexaenoic acid (22:6n-3; DHA), and low concentrations of n-6 fatty acids, particularly 18:2n-6 occurring in the liver and muscle, to some extent, reflected the fatty acid composition in experimental diets. Shade-Coffee Plantations as Refuges for Tropical Wild Orchids in Central Veracruz, Mexico

Shade-coffee plantations with high levels of structural diversity provide refuge for forest-dependent biota (e.g., birds and insects). Orchids typical of natural forest may also be found in the canopy of shade-coffee agroecosystems. It is not known, however, whether these are relicts from the original forest vegetation or if the plantations themselves provide the necessary conditions to support a self-sustained orchid population. We studied the population structure of the epiphytic orchids Jacquiniella teretifolia (Sw.) Britton & Willson, Scaphyglottis livida (Lindl.) Schltr., and Maxillaria densa Lindl. in a shade-coffee plantation (commercial polyculture) in central Veracruz. We also studied the previously undescribed reproductive biology of the latter two species. Our results show that the three orchid species had high population densities (>800 plants/ha). In our study site, 50% to 68% of the orchid plants of the target species were young individuals (less than five shoots). Reproductive structures were present in 80% of individuals larger than 30 shoots in the three species. M. densa is self-incompatible, and the fruit set obtained from cross pollination (42.7%) was higher than that obtained from natural pollination (18.2%), suggesting that this species could be pollinator limited. S. livida is autocompatible, not autogamous, and was not pollinator limited. Our results show that the coffee plantation had abundant orchid populations with log-normal size/age structures. Two of the target species, M. densa and S. livida, depend on pollinators to reproduce. It is clear that pollinators that allow orchids to set a high proportion of fruits persist in shade-coffee plantations. Coffee plantations may not replace the original conditions of a forest, but it is possible that these and other orchid species survive and reproduce in coffee plantations that provide appropriate microclimate conditions for the plants, including pollinators. Resumen:,En el centro de Veracruz, México, las plantaciones de café han reemplazado a extensas áreas de bosque nublado montano. Las plantaciones cafetaleras de sombra con altos niveles de diversidad estructural proporcionan refugio a biota dependiente de bosques (e. g., aves e insectos). En el dosel de agroecosistemas de café de sombra también se pueden encontrar orquídeas típicas de bosques naturales. Sin embargo, no se conoce si son relictos de la vegetación del bosque original o si las plantaciones mismas proporcionan los recursos necesarios para soportar a una población de orquídeas auto sostenida. Estudiamos la estructura de la población de orquídeas epifitas Jacquiniella teretifolia (Sw.) Britton & Willson, Scaphyglottis livida (Lindl.) Rare Species and the Use of Indicator Groups for Conservation Planning

Although tests of the concept have produced varied results, sites selected to address indicator groups can include a high proportion of other species. We tested the hypothesis that species at risk of extinction are not likely to be included in sites selected to protect indicator groups. Using a reserve-selection approach, we compared the ability of seven indicator groups,freshwater fish, birds, mammals, freshwater mussels, reptiles, amphibians, and at-risk species of those six taxa,to provide protection for other species in general and at-risk species in particular in the Middle Atlantic region of the United States. Although sites selected with single taxonomic indicator groups provided protection for between 61% and 82% of all other species, no taxonomic group provided protection for more than 58% of all other at-risk species. The failure to cover at-risk species is likely linked to their rarity. By examining the relationship between a species' probability of coverage by each indicator group and the extent of its geographic range within the study area, we found that species with more restricted ranges were less likely to be protected than more widespread species. Furthermore, we found that although sites selected with indicator groups composed primarily of terrestrial species ( birds and mammals ) included relatively high percentages of those species ( 82,85% ) they included smaller percentages of strictly aquatic species (27,55%). Finally, of both importance and possible utility, we found that at-risk species themselves performed well as an indicator group, covering an average of 84% of all other species. Resumen: Los indicadores de la biodiversidad han sido propuestos como una herramienta potencial en la selección de áreas para conservación cuando la información sobre la distribución de algunas especies es escasa. A pesar de que algunas evaluaciones de este concepto han producido resultados variados, los sitios seleccionados para evaluar grupos indicadores pueden incluir una alta proporción de otras especies. EXECUTING THE INNOCENT AND SUPPORT FOR CAPITAL PUNISHMENT: IMPLICATIONS FOR PUBLIC POLICY

Research Summary: The issue of whether innocent people have been executed is now at the center of the debate concerning the legitimacy of capital punishment. The purpose of this research was to use data collected by the Gallup Organization in 2003 to investigate whether Americans who believed that an innocent person had been executed were less likely to support capital punishment. We also explored whether the association varied by race, given that African Americans are disproportionately affected by the death penalty. Our results indicated that three-quarters of Americans believed that an innocent person had been executed for a crime they did not commit within the last five years and that this belief was associated with lower levels of support for capital punishment, especially among those who thought this sanction was applied unfairly. In addition, our analyses revealed that believing an innocent person had been executed had a stronger association with altering African American than white support for the death penalty. Policy Implications: A key claim of death penalty advocates is that a high proportion of the public supports capital punishment. In this context, scholars opposing this sanction have understood the importance of showing that the public's support for executing offenders is contingent and shallower than portrayed by typical opinion polls. The current research joins this effort by arguing that the prospect of executing innocents potentially impacts public support for the death penalty and, in the least, creates ideological space for a reconsideration of the legitimacy of capital punishment. An audit of ,equivocal' (C3) and ,suspicious' (C4) categories in fine needle aspiration cytology of the breast We have audited the frequency of use and outcome of the ,equivocal/atypia probably benign' (C3) and ,suspicious of malignancy' (C4) category for breast cytology in our Unit. A total of 14 935 cytological specimens were reported by at least one of the three pathologists with a special interest in breast pathology, according to five categories of the NHSBSP guidelines for cytology reporting, 1992; 3.7% (555 cases) and 3.9% (587 cases) of cases were classified as equivocal (C3) and suspicious (C4), respectively, giving a total rate (C3 + C4) of 7.6%. Of the C3 cases, 68% were subsequently benign and 32% were malignant. Of the C4 cases, 19% were subsequently benign and 81% malignant. The commonest benign lesions in both categories were fibroadenomas (7.6% of C3 and 19.8% of C4), fibrocystic change (14.3% of C3 and 12.5% of C4), radial scars (6.2% of C3 and 10.4% of C4) and papillomas (6.2% of C3 and 6.3% of C4). Of the malignant lesions (particularly those classified as C3), a high proportion were low grade or special type cancers. The categories of atypia probably benign (C3) and suspicious of malignancy (C4) in breast cytology provide a strategy for classification of problematic or uncertain cases; this maintains the predictive value of the benign (C2) and malignant (C5) categories, and allows separation of these difficult cases into clinically useful groups with differing probabilities of malignancy. Dental therapists' experience in the immediate management of traumatized teeth

Abstract,,, The optimal immediate management of traumatized teeth is known to be important for long-term success. One hundred and sixty-seven school dental therapists with General Certificate of Education ,Ordinary' (GCE ,O') level qualification were surveyed on their knowledge and experience on immediate management of dental trauma. The results showed only 41.2% felt comfortable with their present knowledge on the subject. A high proportion of the respondents (94.6%) indicated a need for more knowledge. All therapists concurred on the need to replant a permanent tooth and most (85.1%) agreed that this should be done within 30 min. More than half (54.8%) were not sure of the optimal storage medium for avulsed teeth. Their attitude towards acquiring knowledge in this aspect was good (80.5%) and all would take immediate action to settle appointments for trauma cases. [source]

Abstract Major depression affects one out of five women during her lifetime. Depressed mothers with psychiatrically ill children represent an especially vulnerable population. Challenged by the demands of caring for ill children, these mothers often put their own needs last; consequently, their depressions remain untreated. This population is especially difficult to engage in treatment. We have developed a nine-session intervention, an engagement session followed by eight sessions of brief interpersonal psychotherapy designed to increase maternal participation in their own psychotherapy, resolve symptoms of maternal depression, and enhance relationships (IPT-MOMS). This open-label trial assesses the feasibility and acceptability of providing this treatment to depressed mothers. Thirteen mothers meeting DSM-IV criteria for major depression were recruited from a pediatric mental health clinic where their school-age children were receiving psychiatric treatment. Subjects (mothers) were treated openly with IPT-MOMS. Eighty-five percent (11/13) completed the study. Subjects were evaluated with the Hamilton Rating Scale for Depression, and completed self-report measures of quality of life and functioning at three time points: baseline, after treatment completion, and 6-months posttreatment. A signed rank test was used to compare measurement changes between assessment time points. Subjects showed significant improvement from baseline to posttreatment on measures of maternal symptoms and functioning. These gains were maintained at 6-month follow-up. Therapy was well tolerated and accepted by depressed mothers, who are typically difficult to engage in treatment. A high proportion of subjects completed treatment and experienced improvements in functioning. Future randomized clinical trials are needed to establish the efficacy of this approach. Background. Vitiligo therapy is difficult. Depending on its clinical presentation, unilateral or bilateral vitiligo lesions respond well with different repigmentation rates, according to age, affected anatomic area, extension of lesions, time at onset, timing of depigmentation spread, and other associated factors. When stable and refractory to medical treatment, vitiligo lesions may be treated by implanting pigment cells on depigmented areas. Objective. To describe the main events of depigmentation and the fundamentals of surgical techniques for repigmenting vitiligo by implanting noncultured cellular or tissue grafts, in vitro cultured epidermis-bearing pigment cells, or melanocyte suspensions. Methods. A description of the available techniques for repigmentation of vitiligo is done, emphasizing the most important details of each procedure to obtain the best repigmentation and minimize side effects. Results. With most of these techniques, adequate repigmentation is obtained, although there are limitations when applying some methods to clinical practice. Conclusions. Restoration of pigmentation may be accomplished with all available surgical procedures in most anatomic locations, but they are of little value for acral areas. Unilateral vitiligo responds well in a high proportion of patients, and bilateral disease may also respond when stable. Appropriate patient selection is important to achieve the best results. Motor function in 5-year-old children with cerebral palsy in the South Australian population

The aim of this study was to describe the motor function of a population of children at age 5 years enrolled on the South Australian Cerebral Palsy Register. Among children born between 1993 and 1998, there were 333 with confirmed cerebral palsy (prevalence rate 2.2 per 1000 live births), in whom 247 assessments (56.7% males, 43.3% females) were completed. The distribution by Gross Motor Function Classification System (GMFCS) level was: level I, 50.6%; level II, 18.2%; level III, 9.3%; level IV, 9.7%; level V, 12.1%. The most common topographical classification was spastic diplegia (38.5%), followed by spastic hemiplegia (34.8%) and spastic quadriplegia (14.6%). Abnormal movements occurred at rest or with intention in 19.4% of children. A high proportion of the population with relatively mild gross motor impairments have difficulty with everyday bimanual tasks, reinforcing the need to assess upper limb function independently of gross motor function. The use of ankle,foot orthoses was common, particularly across GMFCS levels II to IV. Further refinement is indicated for this population's motor dataset, to include more recently described classification measures as well as future novel measures to better describe the presence of both spasticity and dystonia. A low incidence of Type 1 diabetes between 1977 and 2001 in south-eastern Sweden in areas with high population density and which are more deprived

Abstract Aims To explore how socioeconomic factors and population density may contribute to the geographical variation of incidence of Type 1 diabetes in children in south-eastern Sweden. Method All children diagnosed with Type 1 diabetes in south-eastern Sweden during 1977,2001 were defined geographically to their place of residence and were allocated x and y coordinates in the national grid. The population at risk and socioeconomic data were aggregated in 82 000 200-m squares and geocoded likewise. A socioeconomic index was calculated using a signed ,2 method. Rural,urban gradients were defined by overlay analysis in a geographic information system. Results The incidence during the past 25 years has been rising steadily, particularly in the last 6 years. The incidence was highest in areas with a high proportion of small families, of families with a high family income and better education, and this was found both at the time of diagnosis and at the time of birth. In the rural,urban analysis, the lowest incidence was found in the urban area with > 20 000 inhabitants, where there was also a higher frequency of deprivation. Conclusions Our findings indicate that geographical variations in incidence rates of Type 1 diabetes in children are associated with socioeconomic factors and population density, although other contributing factors remain to be explained. Characteristics of women seeking treatment for premenstrual syndrome in Taiwan

Objective:,To analyze the characteristics of 150 women who visited a premenstrual syndrome (PMS) clinic in a Taiwanese medical center staffed by both psychiatrists and gynecologists. Method:,All subjects were interviewed and assessed for premenstrual dysphoric disorder (PMDD) using DSM-IV criteria, a PMS questionnaire, and the structured Mini-International Neuropsychiatric Interview (MINI). Where PMDD was suspected, subjects were followed using a daily symptom record. Results:,A total of 110 subjects (73%) met the ICD-10 criteria for PMS. The most common PMS symptoms were minor psychological discomfort, muscular tension, and aches or pains. For 129 subjects (86%), other concurrent psychiatric disorders were diagnosed using the MINI. Of these, 48 (37%) reported premenstrual exacerbation (PME) of a previously diagnosed psychiatric condition. Conclusion:,The results of this study indicate that women who complain of PMS may be at a high risk of other psychiatric dysfunction, especially mood disorder. Further, the high proportion of PME cases determined in this study suggests that further investigation is required. [source]


Rapid review of liquid-based smears as a quality control measure

Abstract The objective of this study was to investigate the effectiveness of a standardized method of rapid review (RR) of monolayer preparations for the identification of abnormalities, the presence of an endocervical component and infectious agents. A total of 200 ThinPrep (Cytyc, Boxborough, MA) slides representing the spectrum of abnormalities commonly encountered in cervical/vaginal cytologic specimens was retrieved from archive. The study set comprised 129 cases within normal limits (WNL); 36 low-grade epithelial abnormalities (LGEA); 28 high-grade epithelial abnormalities (HGEA), including 2 endocervical adenocarcinomas in situ (AIS) and 7 carcinomas. Eighteen false negative (FN) cases were also included for study. Originally missed on initial review, these cases were found to be abnormal on quality control review (17 LGEA; 1 AIS). Commonly encountered infectious agents were represented and included Candida albicans, Trichomonas vaginalis, herpes simplex virus, and Actinomyces. The slides were reviewed using a standardized method of RR (turret technique, for 60 sec) by three experienced screeners masked to the original reference diagnosis. Median sensitivity for LGEA was 70% (range, 67,72%); HGEA, 69% (range, 54,80%); and FN, 65% (range, 56,78%). Specificity remained high, median specificity for LGEA was 95%; HGEA, 97%; and FN, 100%. There was no significant overcalling of any diagnostic category. The chi-square test at P < 0.05 showed no significant difference between RR and full manual rescreen of the ThinPrep smears in this study. While no statistical difference was proven, the sensitivity measurements for all categories of abnormality were moderate due to the high proportion of atypical cases included into the study set. Abnormalities on the monolayer preparations frequently displayed fewer, smaller groups of disaggregated cells with rounded cytoplasmic outlines that were difficult to discern on RR. Interobserver variation was noted. Monolayers with a paucity of diagnostic cells and those displaying subtle nuclear atypia were often overlooked. Diagn. Disentangling the relative effects of environmental versus human factors on the abundance of native and alien plant species in Mediterranean sandy shores

Abstract Aim, Mediterranean coastal sand dunes are characterized by both very stressful environmental conditions and intense human pressure. This work aims to separate the relative contributions of environmental and human factors in determining the presence/abundance of native and alien plant species in such an extreme environment at a regional scale. Location, 250 km of the Italian Tyrrhenian coast (Region Lazio). Methods, We analysed alien and native plant richness and fitted generalized additive models in a multimodel-inference framework with comprehensive randomizations to evaluate the relative contribution of environmental and human correlates in explaining the observed patterns. Results, Native and alien richness are positively correlated, but different variables influence their spatial patterns. For natives, human population density is the most important factor and is negatively related to richness. Numbers of natives are unexpectedly lower in areas with a high proportion of natural land cover (probably attributable to local farming practices) and, to a lesser degree, affected by the movement of the coastline. On the other hand, alien species richness is strongly related to climatic factors, and more aliens are found in sectors with high rainfall. Secondarily, alien introductions appear to be related to recent urban sprawl and associated gardening. Main conclusions, Well-adapted native species in a fragile equilibrium with their natural environment are extremely sensitive to human-driven modifications. On the contrary, for more generalist alien species, the availability of limited resources plays a predominant role. [source]

Traffic as dispersal vector across urban, rural gradients
ABSTRACT Urban areas are among the land use types with the highes richness in plant species. A main feature of urban floras is the high proportion of non-native species with often divergent distribution patterns along urban,rural gradients. Urban impacts on plant species richness are usually associated with increasing human activity along rural-to-urban gradients. As an important stimulus of urban plant diversity, human-mediated seed dispersal may drive the process of increasing the similarity between urban and rural floras by moving species across urban,rural gradients. We used long motorway tunnels as sampling sites for propagules that are released by vehicles to test for the impact of traffic on seed dispersal along an urban,rural gradient. Opposite lanes of the tunnels are separated by solid walls, allowing us to differentiate seed deposition associated with traffic into vs. out of the city. Both the magnitude of seed deposition and the species richness in seed samples from two motorway tunnels were higher in lanes leading out of the city, indicating an ,export' of urban biodiversity by traffic. As proportions of seeds of non-native species were also higher in the outbound lanes, traffic may foster invasion processes starting from cities to the surrounding landscapes. Indicator species analysis revealed that only a few species were confined to samples from lanes leading into the city, while mostly species of urban habitats were significantly associated with samples from the outbound lanes. The findings demonstrate that dispersal by traffic reflects different seed sources that are associated with different traffic directions, and traffic may thus exchange propagules along the urban,rural gradient. Land-cover and land-use change and its contribution to the large-scale organization of Puerto Rico's bird assemblages

ABSTRACT Global biodiversity is changing rapidly driven by human alteration of habitat, and nowhere this is more dramatic than in insular habitats. Yet land-cover change is a complex phenomenon that not only involves habitat destruction but also forest recovery over different time scales. Therefore, we might expect species to respond in diverse ways with likely consequences for the reorganization of regional assemblages. These changes, however, may be different in tropical islands because of their low species richness, generalist habits and high proportion of endemics. Here, we focus on the island of Puerto Rico and ask how island-wide changes in land cover and land use has influenced the large-scale organization of bird assemblages. To address this question, we combined in a Geographical Information System (GIS) the first 6 years (1997,2002) of the Puerto Rican Breeding Bird Survey (PR-BBS) with land-cover and land-use data extracted from a published digital map derived from the classification of Landsat images. A Non-metric Multidimensional Scaling (NMS) ordination based on the composition and abundance of birds, and percentage land-use types showed that land use followed by climate could explain most of the variation observed among routes in terms of species composition and abundance. Moreover, endemic and exotic species were widely distributed throughout the island, but the proportion of endemic species is higher in closed forests while exotic species are more abundant in open habitats. However, historical accounts from the early 1900s indicate that endemic species were distributed across the entire island. Today, most of the land cover transformation in Puerto Rico occurs in the lowlands which may explain the high abundance of endemic species in cloud forests and the high abundance of exotic species in open habitats in the lowlands. The distribution and prevalence of sponges in relation to environmental gradients within a temperate sea lough: inclined cliff surfaces

Abstract. Sponge communities on inclined cliff surfaces (40°) at Lough Hyne Marine Nature Reserve, Co. Cork, Ireland were sampled at five sites (four inside the lough and one on the adjacent Atlantic coast). Each site varied in sedimentation rate and flow regime. Sites ranged from turbulent (with negligible sedimentation) to very low flow (< 3 cm,1) and highly sedimented regimes. Sponge species showed variation between sites and depth. The greatest difference in sponge communities was observed between the most turbulent and most sedimented sites. The distinct zonation patterns, present at all sites, were most pronounced at the highly sedimented sites. Encrusting forms constituted a high proportion of the sponges at all sites. However encrusting species found at the turbulent site were different to those at the sedimented sites. Arborescent species were common, mainly at the sedimented sites within Lough Hyne. Distributions of sponge species are considered with respect to morphological adaptation, competition and physiological adaptation. The distributions of sponge species on inclined surfaces are also compared with those on vertical cliff faces. [source]


Prognostic Value of Exercise Stress Test and Dobutamine Stress Echo in Patients with Known Coronary Artery Disease

Background: The aim of this study was to compare the feasibility of dobutamine stress echocardiography (DSE) and exercise stress test (EST) between patients in different age groups and to evaluate their proportional prognostic value in a population with established coronary artery disease (CAD). Methods: The study sample included 323 subjects, subdivided in group 1 (G1), comprising 246 patients aged <75 years, and group 2 (G2), with 77 subjects aged ,75 years. DSE and EST were performed before enrollment in a cardiac rehabilitation program; for prognostic assessment, end points were all-cause mortality and hard cardiac events (cardiac death or nonfatal myocardial infarction). Results: During DSE, G2 patients showed worse wall motion score index (WMSI), but the test was stopped for complications in a comparable proportion of cases (54 G1 and 19 G2 patients, P = NS). EST was inconclusive in similarly high proportion of patients in both groups (76% in G1 vs. 84% in G2, P = NS); G2 patients reached a significantly lower total workload (6 ± 1.6 METs in G1 vs. 5 ± 1.2 METs in G2, P < 0.001). At multivariate analysis, a lower peak exercise capacity (HR 0.566, CI 0.351,0.914, P = 0.020) was associated with higher mortality, while a high-dose WMSI >2 (HR 5.123, CI 1.559,16.833, P = 0.007), viability (HR 3.354, CI 1.162,9.678, P = 0.025), and nonprescription of beta-blockers (HR 0.328, CI 0.114,0.945, P = 0.039) predicted hard cardiac events. Conclusion: In patients with known CAD, EST and DSE maintain a significant prognostic role in terms of peak exercise capacity for EST and of presence of viability and an extensive wall motion abnormalities at peak DSE. The significance of geographic range size for spatial diversity patterns in Neotropical palms

We examined the effect of range size in commonly applied macroecological analyses using continental distribution data for all 550 Neotropical palm species (Arecaceae) at varying grain sizes from 0.5° to 5°. First, we evaluated the relative contribution of range-restricted and widespread species on the patterns of species richness and endemism. Second, we analysed the impact of range size on the predictive value of commonly used predictor variables. Species sequences were produced arranging species according to their range size in ascending, descending, and random order. Correlations between the cumulative species richness patterns of these sequences and environmental predictors were performed in order to analyse the effect of range size. Despite the high proportion of rare species, patterns of species richness were found to be dominated by a minority of widespread species (,20%) which contained 80% of the spatial information. Climatic factors related to energy and water availability and productivity accounted for much of the spatial variation of species richness of widespread species. In contrast, species richness of range-restricted species was to a larger extent determined by topographical complexity. However, this effect was much more difficult to detect due to a dominant influence of widespread species. Although the strength of different environmental predictors changed with spatial scale, the general patterns and trends proved to be relatively stabile at the examined grain sizes. Our results highlight the difficulties to approximate causal explanations for the occurrence of a majority of species and to distinguish between contemporary climatic factors and history. [source]


Patterns of commonness and rarity in central European birds: reliability of the core-satellite hypothesis within a large scale

The frequency distribution of species' area of occupancy is often bimodal, most species being either very rare or very common in terms of number of occupied sites. This pattern has been attributed to the nonlinearity associated with metapopulation dynamics of the species, but there are also other explanations comprising sampling artifact and frequency distribution of suitable habitats. We tested whether the bimodal frequency distribution of occupied squares in central European birds could be derived solely from the frequency distribution of species population sizes (i.e. the sampling artifact hypothesis) or from the spatial distribution of their preferred habitats. Both models predict high proportion of very common species, i.e. the right side of frequency distribution. Bimodality itself is well predicted by models based on random placement of individuals according to their abundances but neither model predicts the observed prevalence of rare species. Even the combined models that assume random placement of individuals within the squares with suitable habitat do not predict such a high proportion of rare species. The observed distribution is more aggregated, rare species occupying a smaller portion of suitable habitat than predicted on the basis of their abundance. The pattern is consistent with metapopulation processes involving local population extinctions. The involvement of these processes is supported by two further observations. First, species rarity is associated with significant population trend and/or location on the edge of their ranges within central Europe, both situations presumably associated with metapopulation processes. Second, suitable habitats seem to be either saturated or almost unoccupied, which is consistent with the predictions of the metapopulation model based on nonlinear dynamics of extinction and colonization. Although the habitat suitability is an important determinant of species distribution, the rarity of many species of birds within this scale of observation seems to be affected by other factors, including local population extinctions associated with fragmentation of species' habitats. Landscape composition and vole outbreaks: evidence from an eight year study of Arvicola terrestris

This study investigates the relationships between landscape composition and the population dynamics of the fossorial water vole Arvicola terrestris. Land use patterns were studied based on agricultural and forestry data from the French Ministry of Agriculture collected in 1955 and 1988. In the Massif Central, France, water vole populations were monitored from 1985 to 1993 by using index methods. Outbreaks of water vole populations occurred in many dispersed epicentres and spread suddenly and widely over > 7500 km2. At a regional scale, the fluctuation lasts six years on average with an outbreak period lasting from two to three years. Density variation patterns are positively correlated with the proportion of permanent grassland to agricultural land. A high risk of outbreak is linked to a high proportion of permanent grassland (over 90%), whereas a low risk of outbreak is linked to a proportion of < 80%, Conversely, density variation patterns are negatively correlated with the proportion of temporary grassland to agricultural land and with the proportion of forest to total land in the western (major) part of the study area. Temporary grassland thus appears to be a marginal habitat for water voles and extensive forests could act as a brake on outbreaks. The increase in the area of permanent grassland from 1955 to 1988 was apparently the major cause of chronic high densities of water voles. Therefore, land use and landscape management could be one way to control water vole outbreaks. [source]

Habitat specificity in a naturally fragmented landscape
Abstract., 1. Metapopulation and island biogeography theory assume that landscapes consist of habitat patches set in a matrix of non-habitat. If only a small proportion of species conform to the patch,matrix assumptions then metapopulation theory may only describe special cases rather than being of more general ecological importance. 2. As an initial step towards understanding the prevalence of metapopulation dynamics in a naturally fragmented landscape, the distribution of beetle species in three replicates of three habitat types was examined, including rainforest and eucalypt forest (the habitat patches), and buttongrass sedgeland (the matrix), in south-west Tasmania, Australia. 3. Ordination methods indicated that the buttongrass fauna was extremely divergent from the fauna of forested habitats. Permutation tests showed that the abundance of 13 of 17 commonly captured species varied significantly among habitats, with eight species confined to eucalypts or rainforest, and three species found only in buttongrass. Approximately 60% of species were confined to forested habitat implying that metapopulation theory has the potential to be very important in the forest,buttongrass landscape. 4. Although floristically the rainforest and eucalypts were extremely distinct, the beetle faunas from eucalypts and rainforests overlapped substantially. Therefore rainforest patches connected by eucalypt forest represent continuous habitat for most species. 5. Other studies report a wide range of values for the proportion of patch-specific species in fragmented landscapes. Understanding the environmental or historical conditions under which a high proportion of species become patch specialists would help to identify where spatial dynamic theory may be especially applicable, and where habitat loss and fragmentation poses the greatest threat to biodiversity. Physical and chemical cues influencing the oviposition behaviour of Aphidius ervi

Abstract The oviposition behaviour of the aphid parasitoid Aphidius ervi Haliday is influenced by both chemical and physical cues. Oviposition attack responses were elicited by paint pigments sealed into the tip of a glass capillary tube. Parasitoids reacted to yellow pigments with repeated oviposition attack responses, but they did not react to green pigments. The spectrum of reflected light from the yellow pigments was very similar to that from the ,green' natural host Acyrthosiphon pisum (Harris), with a high proportion of the total radiation energy being emitted in the yellow-orange wavebands (580,660 nm). Pea aphid cornicle secretion also elicited oviposition attack responses, which were not exclusively induced by its pale yellow-green colour. In fact, the oviposition attack response to capillary tips coated with cornicle secretion remained evident under red light conditions, which, in contrast, nearly completely suppressed the response to yellow pigments. Chemical compounds from cornicle secretion do not appear to be involved in parasitoid orientation, even though they stimulate intense oviposition attack responses. Olfactometer experiments showed that the putative kairomone involved acts only at very short range or on contact. Host exuviae, which also elicited strong and persistent oviposition reactions from A. ervi females, appear to be a good alternative source of ovipositional kairomone(s). This work confirms the existence of an aphid cuticular kairomone. [source]


Phylogenetic diversity and metagenomics of candidate division OP3

Summary Except for environmental 16S rRNA gene sequences, no information is available for members of the candidate division OP3. These bacteria appear to thrive in anoxic environments, such as marine sediments, hypersaline deep sea, freshwater lakes, aquifers, flooded paddy soils and methanogenic bioreactors. The 16S rRNA phylogeny suggests that OP3 belongs to the Planctomycetes/Verrucomicrobia/Chlamydiae (PVC) superphylum. Metagenomic fosmid libraries were constructed from flooded paddy soil and screened for 16S rRNA gene-containing fragments affiliated with the PVC superphylum. The screening of 63 000 clones resulted in 23 assay-positive fosmids, of which three clones were affiliated with OP3. The 16S rRNA gene sequence divergence between the fragments OP3/1, OP3/2 and OP3/3 ranges from 18% to 25%, indicating that they belong to different OP3 subdivisions. The 23S rRNA phylogeny confirmed the membership of OP3 in the PVC superphylum. Sequencing the OP3 fragments resulted in a total of 105 kb of genomic information and 90 ORFs, of which 47 could be assigned a putative function and 11 were conserved hypothetical. Using BLASTP searches, a high proportion of ORFs had best matches to homologues from Deltaproteobacteria, rather than to those of members of the PVC superphylum. On the fragment OP3/3, a cluster of nine ORFs was predicted to encode the bacterial NADH dehydrogenase I. Given the high proportion of homologues present in deltaproteobacteria and anoxic conditions in the natural environment of OP3 bacteria, the detection of NADH dehydrogenase I may suggest an anaerobic respiration mode. Oligonucleotide frequencies calculated for OP3/1, OP3/2 and OP/3 show high intraphylum correlations. This novel sequence information could therefore be used to identify OP3-related fragments in large metagenomic data sets using marker gene-independent procedures in the future. In addition to the OP3 fragments, a single metagenomic fragment affiliated with the candidate division BRC1 was obtained and analysed. [source]


Vertical distribution of picoeukaryotic diversity in the Sargasso Sea

Summary Eukaryotic molecular diversity within the picoplanktonic size-fraction has primarily been studied in marine surface waters. Here, the vertical distribution of picoeukaryotic diversity was investigated in the Sargasso Sea from euphotic to abyssal waters, using size-fractionated samples (< 2 ,m). 18S rRNA gene clone libraries were used to generate sequences from euphotic zone samples (deep chlorophyll maximum to the surface); the permanent thermocline (500 m); and the pelagic deep-sea (3000 m). Euphotic zone and deep-sea data contrasted strongly, the former displaying greater diversity at the first-rank taxon level, based on 232 nearly full-length sequences. Deep-sea sequences belonged almost exclusively to the Alveolata and Radiolaria, while surface samples also contained known and putative photosynthetic groups, such as unique Chlorarachniophyta and Chrysophyceae sequences. Phylogenetic analyses placed most Alveolata and Stramenopile sequences within previously reported ,environmental' clades, i.e. clades within the Novel Alveolate groups I and II (NAI and NAII), or the novel Marine Stramenopiles (MAST). However, some deep-sea NAII formed distinct, bootstrap supported clades. Stramenopiles were recovered from the euphotic zone only, although many MAST are reportedly heterotrophic, making the observed distribution a point for further investigation. An unexpectedly high proportion of radiolarian sequences were recovered. From these, five environmental radiolarian clades, RAD-I to RAD-V, were identified. RAD-IV and RAD-V were composed of Taxopodida-like sequences, with the former solely containing Sargasso Sea sequences, although from all depth zones sampled. Our findings highlight the vast diversity of these protists, most of which remain uncultured and of unknown ecological function. [source]

Summary This laboratory study of a variably mineralized and hydrothermally altered granite outcrop investigated the influences of rock-surface chemistry and heavy metal content on resident bacterial populations. Results indicated that elevated heavy metal concentrations had a profound impact on bacterial community structure, with strong relationships found between certain ribotypes and particular chemical/heavy metal elements. Automated ribosomal intergenic sequence analysis (ARISA) was used to assess the nature and extent of bacterial diversity, and this was combined with chemical analysis and multivariate statistics to identify the main geochemical factors influencing bacterial community structure. A randomization test revealed significant changes in bacterial structure between samples, while canonical correspondence analysis (CCA) related each individual ARISA profile to linear combinations of the chemical variables (mineralogy, major element and heavy metal concentrations) revealing the geochemical factors that correlated with changes in the ARISA data. anova was performed to further explore interactions between individual ribotypes and chemical/heavy metal composition, and revealed that a high proportion of ribotypes correlated significantly with heavy metals.
