Dynamics of the SAR11 bacterioplankton lineage in relation to environmental conditions in the oligotrophic North Pacific subtropical gyre

Summary A quantitative PCR assay for the SAR11 clade of marine Alphaproteobacteria was applied to nucleic acids extracted from monthly depth profiles sampled over a 3-year period (2004,2007) at the open-ocean Station ALOHA (A Long-term Oligotrophic Habitat Assessment; 22°45,N, 158°00,W) in the oligotrophic North Pacific Ocean. This analysis revealed a high contribution (averaging 36% of 16S rRNA gene copies) of SAR11 to the total detected 16S rRNA gene copies over depths ranging from the surface layer to 4000 m, and revealed consistent spatial and temporal variation in the relative abundance of SAR11 16S rRNA gene copies. On average, a higher proportion of SAR11 rRNA gene copies were detected in the photic zone (< 175 m depth; mean = 38%) compared with aphotic (> 175 m depth; mean = 30%), and in the winter months compared with the summer (mean = 44% versus 33%, integrated over 175 m depth). Partial least square to latent structure projections identified environmental variables that correlate with variation in the absolute abundance of SAR11, and provided tools for developing a predictive model to explain time and depth-dependent variations in SAR11. Moreover, this information was used to hindcast temporal dynamics of the SAR11 clade between 1997 and 2006 using the existing HOT data set, which suggested that interannual variations in upper ocean SAR11 abundances were related to ocean-climate variability such as the El Niño Southern Oscillation. Application of Nanostructured ASP Precursors for Processing CaCuMn6O12 Colossal Magnetoresistance Ceramics

Nanostructured precursors prepared by ultrasonic aerosol spray pyrolisys at 750,950°C in air were applied for the first time to obtain Ca(Cu,Mn)7O12 colossal magnetoresistance ceramics with a unique submicron necked-grain microstructure. It was observed that a high contribution of intergrain tunneling magnetoresistance results in a 10,20% difference in functional properties as dependent on morphological features of hollow precursor microspheres ASP processed at a given temperature. Frequencies of the -330 (T , G) IL-2 and -590 (T , C) IL-4 gene polymorphisms in a population from south-eastern Brazil

Summary Polymorphisms in the promoter regions of cytokine genes may affect their transcription. A T/G substitution at position ,330 of the interleukin-2 (IL-2) gene and a T/C substitution at position ,590 of the interleukin-4 (IL-4) gene have been described previously. The ,590 (T , C) IL-4 gene polymorphism was associated with asthma and atopy in US and Japanese populations. Population genetics is a useful tool for determination of the biological significance of genetic polymorphisms. The aim of this study was to investigate the frequencies of polymorphisms in the promoter regions of the IL-2 and IL-4 genes in a population from south-eastern Brazil and to compare them with those published for other populations. Allele frequencies were estimated in 114 unrelated individuals from São Paulo State. These subjects had an average age of 41.2 years (± 12.4 years) and the ethnic composition of the sample was: 78.07% Caucasian, 11.4% Black and 10.53% Mulatto. DNA from subjects was extracted from epithelial buccal cells, and the PCR-RFLP technique was employed to investigate the ,330 (T , G) IL-2 and ,590 (T , C) IL-4 gene polymorphisms. The allele frequency of the IL-2 gene polymorphism obtained in our study was similar to that found in UK Caucasoid groups. The T allele frequency of the IL-4 gene polymorphism observed in the Caucasian Brazilian group was similar to that found in UK and Australian populations, while the frequency observed for the Black Brazilian group was similar to that found in Japanese and Kuwaiti Arab populations. The results for the ,330 (T , G) IL-2 and ,590 (T , C) IL-4 polymorphisms are consistent with the high contribution of European lineages to the population in south-eastern Brazil. [source]


FTIR-spectroscopic characterization of humic acids and humin fractions obtained by advanced NaOH, Na4P2O7, and Na2CO3 extraction procedures

Abstract Aim of our study was the development of the methodological basis for the characterization of humic fractions of a long-term field experiment. Humic acids (HAs) were extracted from three layers of a nontilled soil using three different extractants (1 M NaOH, 0.1 M Na4P2O7, 1 M Na2CO3), and the humin fraction was enriched. NaOH as extractant for FTIR analysis of humic substances yields higher resolved IR spectra, especially in the important regions of stretching vibrations including aromatic and aliphatic groups and in the fingerprint area including amides, aliphats, and aromats than the other extractants. The NaOH extraction has lower extraction yields as compared to Na4P2O7 and Na2CO3 and represents a different part of the soil organic matter (SOM). This is reflected by lower C : N ratios and higher E4 : E6 and fulvic acid,to,humic acid ratios as compared to the other extractants. The FTIR band areas of HA fraction obtained by NaOH showed an increase of the aromatic and carbonyl groups and a decrease of amide groups with increasing soil depth. Aliphatic groups showed contradicting results: The bands of the stretching vibrations increased, and the band of the bending vibrations decreased. We assume that band interactions in the bending vibrations were responsible for that phenomenon under the assumption of an increase of aliphatic groups with increasing soil depth. The IR bands of the enriched humin fraction showed a decreasing trend in case of both aliphatic bands deriving from stretching vibrations and an increase of aromatic characteristics with depth. Our study led to the conclusion that HA fractions obtained by 1 M NaOH represent a small and dynamic fraction indicated by the measured yields in combination with values of Nt, C : N, E4 : E6 ratios, and ratios of fulvic acids (FA) to HA. The humin fraction has a high contribution to the total organic C and represents a more stabilized fraction of SOM which still shows changes in its aromatic and aliphatic characteristics with soil depth. Identification of the food sources of sympatric ghost shrimp (Trypaea australiensis) and soldier crab (Mictyris longicarpus) populations using a lipid biomarker, dual stable isotope approach

Abstract Fatty acids (FAs) profiles and stable isotope signatures of the ghost shrimp, Trypaea australiensis and the soldier crab, Mictyris longicarpus were determined at an unvegetated sandbank of Southport (Gold Coast, Queensland, Australia), in November 2005 and February 2006. Additionally, the FAs composition of the faeces and feeding pellets of M. longicarpus and the surface sediment at the study site were also analysed. Trypaea australiensis was found to selectively feed principally on benthic diatoms, as revealed by the high contribution of the marker lipid (20:5 (n-3)) to tissue total FAs and the ,13C and ,15N isotopic signatures of shrimp tissues. Although the diet of T. australiensis did not change between the two sampling periods, the shrimps appeared to reduce their feeding activity in summer, presumably in relation to a restricted metabolism, as revealed by a decrease in the contribution of the microalgal markers in their tissues. The FAs composition of the tissues of the soldier crab indicated that bacteria and diatoms constituted the base of its diet (contributions of branched 15:0 and 17:0, 18:1 (n-7) and 20:5 (n-3)). However, the isotopic signatures of the crabs suggested that meiofauna may represent an intermediate link between the crab and these micro-organisms. [source]


The radiation of the Cape flora, southern Africa

ABSTRACT The flora of the south-western tip of southern Africa, the Cape flora, with some 9000 species in an area of 90 000 km2 is much more speciose than can be expected from its area or latitude, and is comparable to that expected from the most diverse equatorial areas. The endemism of almost 70%, on the other hand, is comparable to that found on islands. This high endemism is accounted for by the ecological and geographical isolation of the Cape Floristic Region, but explanations for the high species richness are not so easily found. The high species richness is accentuated when its taxonomic distribution is investigated: almost half of the total species richness of the area is accounted for by 33,Cape floral clades'. These are clades which may have initially diversified in the region, and of which at least half the species are still found in the Cape Floristic Region. Such a high contribution by a very small number of clades is typical of island floras, not of mainland floras. The start of the radiation of these clades has been dated by molecular clock techniques to between 18 million years ago (Mya)(Pelargonium) and 8 Mya (Phylica), but only six radiations have been dated to date. The fossil evidence for the dating of the radiation is shown to be largely speculative. The Cenozoic environmental history of southern Africa is reviewed in search of possible triggers for the radiations, climatic changes emerge as the most likely candidate. Due to a very poor fossil record, the climatic history has to be inferred from larger scale patterns, these suggest large-scale fluctuations between summer wet (Palaeocene, Early Miocene)and summer dry climates (Oligocene, Middle Miocene to present). The massive speciation in the Cape flora might be accounted for by the diverse limitations to gene flow (dissected landscapes, pollinator specialisation, long flowering times allowing much phenological specialisation), as well as a richly complex environment providing a diversity of selective forces (geographically variable climate, much altitude variation, different soil types, rocky terrain providing many micro-niches, and regular fires providing both intermediate disturbances, as well as different ways of surviving the fires). However, much of this is based on correlation, and there is a great need for (a)experimental testing of the proposed speciation mechanisms, (b)more molecular clock estimates of the age and pattern of the radiations, and (c)more fossil evidence bearing on the past climates. Keeping up with the Joneses: The relationship of perceived descriptive social norms, social information, and charitable giving

We study the influence of perceived descriptive social norms on subsequent giving behavior to nonprofits, explore how social information can influence these norms, and provide insight for fundraising practice. A survey conducted in a nonprofit organization first shows that donors use their beliefs about the descriptive social norm to inform their own donation behavior. Donors who believe that others make high contributions tend to make high contributions themselves. Next, a laboratory experiment demonstrates the influence of social information on the descriptive social norm and consequently on giving. These results suggest strategies for fundraising practice. Informing donors of contributions made by another person influences their perceptions about the descriptive social norm, which in turn influences their giving behavior. We conclude with a discussion of theoretical and practical implications. [source]
