Tenaud Abstract: The migration of keratinocytes plays an important role in the re-epithelialization of cutaneous wounds. Zinc, copper and manganese are used in vivo for their healing properties and their mechanism of action is still only partially known. Thus, they have been shown both to promote keratinocyte proliferation and to modulate integrins expression. The aim of this study was to determine if trace elements induce an increase of the migration of keratinocytes and if this effect is related to the modulation of integrins. Two independent migration assays were used to study keratinocyte migration: the scratch assay using normal human keratinocytes and the modified Boyden chamber using HaCaT cells. Inhibition studies using function-blocking antibodies directed to ,3, ,6, ,V and ,1 subunits were performed to investigate the modulator effect of trace elements on integrin function. In this way, zinc and copper gluconates increased ,3, ,V and ,1 function whereas manganese gluconate seems mainly able to modulate the function of ,3 and ,1. The stimulating effect of these trace elements on keratinocyte migration does not appear related to ,6 subunit. Thus, zinc, copper and manganese enhanced keratinocyte migration and one of the mechanisms was going through a modulation of integrin functions. [source] A novel method of comparing the healing properties of two hydrogels in chronic leg ulcersJOURNAL OF THE EUROPEAN ACADEMY OF DERMATOLOGY & VENEREOLOGY, Issue 2 2006M De La Brassinne Abstract Skin ulcers on the legs have a chronic, relapsing course and are often a significant management challenge. Novel methods of measuring and comparing the effects of different treatments can be of assistance in addressing this situation. A clinical pilot study using original methods was undertaken to compare the healing properties of the alginate gel Flaminal® (test) and the hydrocolloid gel Intrasite® (control) on chronic leg ulcers. The study was performed over a period of 28 days with two parallel groups of 10 patients. Both the surface (acetate tracing and planimetry) and the volume (Jeltrate® mould impression and weighting) of each wound were measured at baseline and after 7, 14 and 28 days of treatment. On both parameters results were superior with the test product compared to the control, with volume reduction being the first parameter to change. Between groups, difference in wound volume reduction was detected as early as day 7 whereas difference in surface reduction was clearly apparent only at day 28. Correlation between wound surface and volume reductions was also better in the test group (r = 0.843 vs. 0.421) than in the control. In conclusion, this pilot study suggests that combining wound surface and volume evaluations allows a more precise analysis of the healing process in venous leg ulcers and that this method is able to detect very early differences in treatments even with limited sample size. [source] Identification of the alkaloids of Galipea officinalis by gas chromatography,mass spectrometryPHYTOCHEMICAL ANALYSIS, Issue 5 2001Ingrid Jacquemond-Collet Abstract Galipea officinalis Hancock, a Venezuelan shrubby tree which is acclaimed in folk medicine for its many healing properties, is the only species of the genus to contain tetrahydroquinoline alkaloids. A GC-MS method has been developed in order to analyse the essential oil, hexane and chloroform extracts of the trunk bark of this plant, without prior derivatisation of the alkaloidal components. A study of the MS fragmentation patterns of the components permitted the identification of five new minor quinoline alkaloids together with the known alkaloids. [source] Antioxidant and cytoprotective properties of infusions from leaves and inflorescences of Achillea collina Becker ex Rchb.PHYTOTHERAPY RESEARCH, Issue 4 2009Annamaria Giorgi Abstract Plants are the main source of molecules with antioxidant and radical scavenging properties that aid the natural defence systems of cells and may be involved in the preservation of human health, particularly preventing all the physiopathological conditions where oxidative damage is a hallmark. Achillea collina Becker ex Rchb. is a medicinal plant of the Achillea millefolium aggregate (yarrow) traditionally used, particularly in mountain areas, as an infusion or alcohol extract for its digestive, antiinflammatory, analgesic, antipyretic and wound healing properties. The aim of this study was to investigate the antioxidant capacity and cytoprotective activity against oxidative stress of infusions obtained from the leaves and inflorescences of Achillea collina Becker ex Rchb., assessed by chemical (free radical scavenging activity by DPPH and Folin Ciocalteu assay) and biological assays (in vitro model of cytotoxicity and lipid peroxidation in PC12 cells line). Infusions of leaves had the highest antioxidant properties and cytoprotective activity. The antioxidant capacity was significantly correlated with the total phenolic content but not with the cytoprotective profile. Achillea collina Becker ex Rchb. has good antioxidant and cytoprotective properties, suggesting further investigations on its chemical composition and potential health value, particularly for traditionally prepared infusions of leaves. Abstract Aqueous extracts of leaves, rind, fruit pulp and flowers of Hylocereus undatus were studied for their wound healing properties. Wound healing effects were studied on incision (skin breaking strength), excision (percent wound contraction) and the nature of wound granulation tissues, which were removed on day 7 and the collagen, hexosamine, total proteins and DNA contents were determined, in addition to the rates of wound contraction and the period of epithelialization. In streptozotocin diabetic rats, where healing is delayed, topical applications of H. undatus produced increases in hydroxyproline, tensile strength, total proteins, DNA collagen content and better epithelization thereby facilitating healing. [source] The protective and healing effects of a natural antioxidant formulation based on ubiquinol and Aloe vera against dextran sulfate-induced ulcerative colitis in ratsBIOFACTORS, Issue 1-4 2003Ludmila Korkina Abstract Oxygen/nitrogen reactive species (ROS/RNS) are currently implicated in the pathogenesis of ulcerative colitis, drawing attention on the potential prophylactic and healing properties of antioxidants, scavengers, chelators. We evaluated the possible protective/curative effects of a natural antioxidant preparation based on Aloe vera and ubiquinol, against intestinal inflammation, lesions, and pathological alterations of the intestinal electrophysiological activity and motility, in a rat model of DSS-induced colitis. 5% dextrane sulfate (DDS) (3 days), followed by 1% DSS (4 days) was administered in drinking water. The antioxidant formulation (25 mg/kg) was delivered with a pre-treatment protocol, or simultaneously or post-colitis induction. Spontaneous and acetylcholine-stimulated electrical activity were impaired in the small intestine and in distal colon, upon exposure to DSS only. Severe inflammation occurred, with increased myeloperoxidase activity, and significant alterations of the oxidant/antioxidant status in colonic tissue and peritoneal cells. Lipoperoxidation, superoxide production, glutathione peroxidase and glutathione-S-transferase activities, and reduced glutathione content increased, whilst superoxide dismutase and catalase activities were sharply suppressed in colon tissue. ROS/RNS formation in peritoneal cells was strongly inhibited. Inflammation, electrical/mechanical impairment in the gut, and a great majority of oxidative stress parameters were improved substantially by pre-treatment with the antioxidant preparation, but not by simultaneous administration or post-treatment. [source] AMINO ACID AND FATTY ACID COMPOSITION OF AN AQUEOUS EXTRACT OF CHANNA STRIATUS (HARUAN) THAT EXHIBITS ANTINOCICEPTIVE ACTIVITYCLINICAL AND EXPERIMENTAL PHARMACOLOGY AND PHYSIOLOGY, Issue 3 2007ZA Zakaria SUMMARY 1The present study was performed in order to determine the amino acid and fatty acid composition of an aqueous extract of the freshwater fish Channa striatus, obtained by soaking (1 : 2, w/v) fresh fillets overnight in a chloroform : methanol (2 : 1, v/v) solvent, to elucidate the mechanism responsible for its antinociceptive activity and to clarify the relationship between the presence of the amino and fatty acids and the expected activity. 2The aqueous extract was found to contain all amino acids with the major amino acids glycine, alanine, lysine, aspartic acid and proline making up 35.77 ± 0.58, 10.19 ± 1.27, 9.44 ± 0.56, 8.53 ± 1.15 and 6.86 ± 0.78% of the total protein, respectively. 3In addition, the aqueous extract was found to have a high palmitic acid (C16 : 0) content, which contributed approximately 35.93 ± 0.63% to total fatty acids. The other major fatty acids in the aqueous extract were oleic acid (C18 : 1), stearic acid (C18 : 0), linoleic acid (C18 : 2) and arachidonic acid (C20 : 4), contributing 22.96 ± 0.40, 15.31 ± 0.33, 11.45 ± 0.31 and 7.44,±,0.83% of total fatty acids, respectively. 4Furthermore, the aqueous extract was demonstrated to possess concentration-dependent antinociceptive activity, as expected, when assessed using the abdominal constriction test in mice. 5It is concluded that the aqueous extract of C. striatus contains all the important amino acids, but only some of the important fatty acids, which are suggested to play a key role in the observed antinociceptive activity of the extract, as well as in the traditionally claimed wound healing properties of the extract.
