These models are direct models of daily asset return volatility based on realised measures constructed from high-frequency data. Our analysis identifies that the models have momentum and mean reversion effects, and that they adjust quickly to structural breaks in the level of the volatility process. We study how to estimate the models and how they perform through the credit crunch, comparing their fit to more traditional GARCH models. We analyse a model-based bootstrap which allows us to estimate the entire predictive distribution of returns. We also provide an analysis of missing data in the context of these models. [source] Reproducibility assessment of metabolic variables characterizing muscle energetics in Vivo: A 31P-MRS studyMAGNETIC RESONANCE IN MEDICINE, Issue 4 2009Gwenael Layec Abstract The purpose of the present study was to assess the reliability of metabolic parameters measured using 31P magnetic resonance spectroscopy (31P MRS) during two standardized rest-exercise-recovery protocols. Twelve healthy subjects performed the standardized protocols at two different intensities; i.e., a moderate intensity (MOD) repeated over a two-month period and heavy intensity (HEAVY) repeated over a year's time. Test-retest reliability was analyzed using coefficient of variation (CV), limits of agreement (LOA), and intraclass correlation coefficients (ICC). During exercise and recovery periods, most of the metabolic parameters exhibited a good reliability. The CVs of individual concentration of phosphocreatine ([PCr]), concentration of adenosine diphosphate ([ADP]), and pH values recorded at end of the HEAVY exercise were lower than 15%. The CV calculated for the rate of PCr resynthesis and the maximal oxidative capacity were less than 13% during the HEAVY protocol. Inferred parameters such as oxidative and total adenosine triphosphate (ATP) production rates exhibited a good reliability (ICC , 0.7; CV < 15% during the HEAVY protocol). Our results demonstrated that measurement error using 31P-MRS during a standardized exercise was low and that biological variability accounted for the vast majority of the measurement variability. [source] Adaptations of an Emergence Trap for Use in Tropical StreamsINTERNATIONAL REVIEW OF HYDROBIOLOGY, Issue 4 2004Hendrik Freitag Abstract An emergence trap based on the MPI Schlitz model was designed for use in the tropics and it was tested over one year in Palawan, the Philippines. Instructions for construction and use are given here. Only commonly available materials were used, except for the collection assembly made of UV-light permeable acrylic glass. Heavy and bulky assemblies were avoided to enable easy transportation in the field. A special modification allows a fast and easy replacement of the screen when damaged by flooding, as is often required when traps are used downstream of headwaters. This type of trap also allows sampling of a wide littoral strip. Problems concerning the use of emergence traps in the humid tropics are discussed based on experience at different longitudinal stream sections in Palawan. BORMAN clima; incendio forestal; pastoreo histórico; pino ponderosa; supresión de fuego Abstract:,Livestock grazing has been implicated as a cause of the unhealthy condition of ponderosa pine forest stands in the western United States. An evaluation of livestock grazing impacts on natural resources requires an understanding of the context in which grazing occurred. Context should include timing of grazing, duration of grazing, intensity of grazing, and species of grazing animal. Historical context, when and under what circumstances grazing occurred, is also an important consideration. Many of the dense ponderosa pine forests and less-than-desirable forest health conditions of today originated in the early 1900s. Contributing to that condition was a convergence of fire, climate, and grazing factors that were unique to that time. During that time period, substantially fewer low-intensity ground fires (those that thinned dense stands of younger trees) were the result of reduced fine fuels (grazing), a substantial reduction in fires initiated by Native Americans, and effective fire-suppression programs. Especially favorable climate years for tree reproduction occurred during the early 1900s. Exceptionally heavy, unregulated, unmanaged grazing by very large numbers of horses, cattle, and sheep during the late nineteenth and early twentieth centuries occurred in most of the U.S. West and beginning earlier in portions of the Southwest. Today, livestock numbers on public lands are substantially lower than they were during this time and grazing is generally managed. Grazing then and grazing now are not the same. Resumen:,El pastoreo de ganado ha sido implicado como una causa de la mala salud de los bosques de pino ponderosa en el occidente de Estados Unidos. La evaluación de los impactos del pastoreo sobre los recursos naturales requiere de conocimiento del contexto en que ocurrió el pastoreo. El contexto debe incluir al período de ocurrencia, la duración y la intensidad del pastoreo, así como la especie de animal que pastoreó. El contexto histórico, cuando y bajo que circunstancias ocurrió el pastoreo, también es una consideración importante. Muchos de los bosques densos de pino ponderosa y de las condiciones, menos que deseables, de salud de los bosques actuales se originaron al principio del siglo pasado. Contribuyó a esa condición una convergencia de factores, fuego, clima y pastoreo, que fueron únicos en ese tiempo. Young Abstract Cytokinesis is the process by which a cell physically divides in two at the conclusion of a cell cycle. In animal and fungal cells, this process is mediated by a conserved set of proteins including actin, type II myosin, IQGAP proteins, F-BAR proteins, and the septins. To facilitate biochemical and ultrastructural analysis of cytokinesis, we have isolated and partially purified the Saccharomyces cerevisiae cytokinetic apparatus. The isolated apparatus contains all components of the actomyosin ring for which we tested,actin, myosin heavy and light chain, and IQGAP,as well as septins and the cytokinetic F-BAR protein, Hof1p. We also present evidence indicating that the actomyosin rings associated with isolated cytokinetic apparati may be contractile in vitro, and show preliminary electron microscopic imaging of the cytokinetic apparatus. This first successful isolation of the cytokinetic apparatus from a genetically tractable organism promises to make possible a deeper understanding of cytokinesis. © 2009 Wiley-Liss, Inc. [source] Dynein light chain family in Tetrahymena thermophilaCYTOSKELETON, Issue 2 2007David E. Wilkes Abstract Dyneins are large protein complexes that produce directed movement on microtubules. In situ, dyneins comprise combinations of heavy, intermediate, light-intermediate, and light chains. The light chains regulate the locations and activities of dyneins but their functions are not completely understood. We have searched the recently sequenced Tetrahymena thermophila macronuclear genome to describe the entire family of dynein light chains expressed in this organism. We identified fourteen genes encoding putative dynein light chains and seven genes encoding light chain-like proteins. RNA-directed PCR revealed that all 21 genes were expressed. Quantitative real time reverse transcription PCR showed that many of these genes were upregulated after deciliation, indicating that these proteins are present in cilia. Using the nomenclature developed in Chlamydomonas, Tetrahymena expresses two isoforms each of LC2, LC4, LC7, and Tctex1, three isoforms of p28, and six LC8/LC8-like isoforms. Tetrahymena also expresses two LC3-like genes. No Tetrahymena orthologue was found for Chlamydomonas LC5 or LC6. This study provides a complete description of the different genes and isoforms of the dynein light chains that are expressed in Tetrahymena, a model organism in which the targeted manipulation of genes is straightforward. [source] If cannabis caused schizophrenia,how many cannabis users may need to be prevented in order to prevent one case of schizophrenia?ADDICTION, Issue 11 2009England, Wales calculations ABSTRACT Background We consider how many cannabis users may need to be prevented in order to prevent one case of schizophrenia or psychosis [defined as number needed to prevent (NNP)]. Method Calculation for England and Wales using best available estimates of: incidence of schizophrenia; rates of heavy and light cannabis use; and risk that cannabis causes schizophrenia. Results In men the annual mean NNP for heavy cannabis and schizophrenia ranged from 2800 [90% confidence interval (CI) 2018,4530] in those aged 20,24 years to 4700 (90% CI 3114,8416) in those aged 35,39. In women, mean NNP for heavy cannabis use and schizophrenia ranged from 5470 (90% CI 3640,9839) in those aged 25,29 to 10 870 (90% CI 6786,22 732) in 35,39-year-olds. Equivalent mean NNP for heavy cannabis use and psychosis were lower, from 1360 (90% CI 1007,2124) in men aged 20,24 and 2480 (90% CI 1408,3518) in women aged 16,19. The mean and median number of light cannabis users that would need to be prevented in order to prevent one case of schizophrenia or psychosis per year are four to five times greater than among heavy users. Conclusions The number of young people who need to be exposed to an intervention to generate NNP and prevent one case of schizophrenia will be even larger. The public health importance of preventing cannabis to reduce schizophrenia or psychosis remains uncertain. More attention should be given to testing the hypothesis that cannabis is related causally to psychotic outcomes, and to considering what strategies will be the most effective in reducing heavy cannabis use among young people. [source] Preventing heavy alcohol use in adolescents (PAS): cluster randomized trial of a parent and student intervention offered separately and simultaneouslyADDICTION, Issue 10 2009Ina M. Koning ABSTRACT Aims To evaluate the effectiveness of two preventive interventions to reduce heavy drinking in first- and second-year high school students. Design and setting Cluster randomized controlled trial using four conditions for comparing two active interventions with a control group from 152 classes of 19 high schools in the Netherlands. Participants A total of 3490 first-year high school students (mean 12.68 years, SD = 0.51) and their parents. Intervention conditions (i) Parent intervention (modelled on the Swedish Örebro Prevention Program) aimed at encouraging parental rule-setting concerning their children's alcohol consumption; (ii) student intervention consisting of four digital lessons based on the principles of the theory of planned behaviour and social cognitive theory; (iii) interventions 1 and 2 combined; and (iv) the regular curriculum as control condition. Main outcome measures Incidence of (heavy) weekly alcohol use and frequency of monthly drinking at 10 and 22 months after baseline measurement. Findings A total of 2937 students were eligible for analyses in this study. At first follow-up, only the combined student,parent intervention showed substantial and statistically significant effects on heavy weekly drinking, weekly drinking and frequency of drinking. At second follow-up these results were replicated, except for the effects of the combined intervention on heavy weekly drinking. These findings were consistent across intention-to-treat and completers-only analyses. Conclusions Results suggest that adolescents as well as their parents should be targeted in order to delay the onset of drinking, preferably prior to onset of weekly drinking. [source] The impact of alcohol use on depressive symptoms in human immunodeficiency virus-infected patients,ADDICTION, Issue 9 2008Lynn E. Sullivan ABSTRACT Aims To examine the impact of alcohol use on depressive symptoms in human immunodeficiency virus (HIV)-infected patients. Design Data were collected at 6-month intervals and analyzed to evaluate the association between alcohol dependence and consumption on depressive symptoms using longitudinal mixed-effects regression models controlling for specified covariates. Measurements The two independent variables were current alcohol dependence assessed using the Composite International Diagnostic Interview (CIDI) and past month consumption (heavy versus not heavy drinking) using a validated calendar-based method. The primary outcome was depressive symptoms as measured by the Center for Epidemiologic Studies Depression Scale (CES-D). Participants HIV-infected adults with current or past alcohol problems. Findings Alcohol dependence and heavy alcohol use were significantly associated with higher CES-D scores in unadjusted models. In adjusted analyses, the association of current alcohol dependence persisted [mean difference in CES-D was 3.49 for dependence versus non-dependence; 95% confidence interval (CI): 1.76,5.22]; however, the effect of heavy drinking was no longer statistically significant (mean difference in CES-D was 1.04 for heavy versus not heavy drinking; 95% CI: ,0.24,2.32). Conclusions Alcohol use is associated with more depressive symptoms in HIV-infected patients with alcohol problems. This association remains significant after adjusting for potential confounders only when alcohol use meets the criteria for alcohol dependence. [source] Clonally expanded plasma cells in the cerebrospinal fluid of MS patients produce myelin-specific antibodiesEUROPEAN JOURNAL OF IMMUNOLOGY, Issue 7 2008Hans-Christian von Büdingen Abstract Clonally expanded plasma cells (cePC) and their presumed products, oligoclonal immunoglobulin,G bands (OCB), are characteristic findings in the cerebrospinal fluid (CSF) of patients with multiple sclerosis (MS). While cePC and OCB strongly suggest an involvement of B cell-dependent immune mechanisms in the pathogenesis of MS, their actual pathological relevance and target antigens remain unknown. To further understand the potential role played by cePC, we generated a panel of monoclonal antibodies (MS-mAb) from CSF-derived cePC from four patients with early or definite MS. Single-cell RT-PCR of correctly paired heavy and light chain immunoglobulin genes from individual cePC ensured the subsequent resurrection of their original antigen specificity. Immunofluorescence stainings of MS lesion tissue with MS-mAb revealed myelin reactivity in the cePC repertoire of all four patients and intracellular filament reactivity in one patient. While myelin staining by MS-mAb was only rarely detectable in non-MS CNS white matter tissue, it was greatly enhanced at the edge of demyelinating lesions in MS brain tissue. Our findings provide conclusive evidence for the presence of an antigen-driven B cell response in the CSF of MS patients directed against epitopes present in areas of myelin degradation. [source] REVIEW: Ethanol consumption: how should we measure it?ADDICTION BIOLOGY, Issue 2 2010Achieving consilience between human, animal phenotypes ABSTRACT There is only modest overlap in the most common alcohol consumption phenotypes measured in animal studies and those typically studied in humans. To address this issue, we identified a number of alcohol consumption phenotypes of importance to the field that have potential for consilience between human and animal models. These phenotypes can be broken down into three categories: (1) abstinence/the decision to drink or abstain; (2) the actual amount of alcohol consumed; and (3) heavy drinking. A number of suggestions for human and animal researchers are made in order to address these phenotypes and enhance consilience. Laboratory studies of the decision to drink or to abstain are needed in both human and animal research. In human laboratory studies, heavy or binge drinking that meets cut-offs used in epidemiological and clinical studies should be reported. Greater attention to patterns of drinking over time is needed in both animal and human studies. Individual differences pertaining to all consumption phenotypes should be addressed in animal research. Lastly, improved biomarkers need to be developed in future research for use with both humans and animals. Greater precision in estimating blood alcohol levels in the field, together with consistent measurement of breath/blood alcohol levels in human laboratory and animal studies, provides one means of achieving greater consilience of alcohol consumption phenotypes. [source] Peripheral B cell receptor editing may promote the production of high-affinity autoantibodies in CD22-deficient miceEUROPEAN JOURNAL OF IMMUNOLOGY, Issue 10 2006Yuval Yarkoni Abstract CD22-deficient mice are characterized by B cell hyperactivity and autoimmunity. We have constructed knock-in CD22,/, mice, expressing an anti-DNA heavy (H) chain (D42), alone or combined with V,1-J,1 or V,8-J,5 light (L) chains. The Ig-targeted mice produced a lupus-like serology that was age- and sex-dependent. High-affinity IgG autoantibodies were largely dependent on the selection of B cells with a particular H/L combination, in which a non-transgenic, endogenous L,chain was assembled by secondary rearrangements through the mechanism of receptor editing. Moreover, we present evidence that these secondary rearrangements are very prominent in splenic peripheral B cells. Since CD22 is primarily expressed on the surface of peripheral B cells, we propose a model for the development of a lupus-like autoimmune disease by a combination of peripheral receptor editing and abnormal B cell activation. Recent evidence indicates that recruitment and selection of MZ-B,cells occurs on the basis of positive selection constraints that likely operate via B,cell receptor (BCR) signaling. Previous studies have shown that MZ-B,cells carry relatively shorter immunoglobulin (Ig) heavy (H) chain complementarity-determining region,3 (H-CDR3) sequences and express BCR which are thought to be polyreactive. In this scenario, MZ-B,cell selection proceeds via engagement of the BCR with exogenous (i.e. microbial gut flora-derived) and/or endogenous (self) antigens. Here, we studied the influence of exogenous antigens on the selection process of MZ-B,cells using non-genetically manipulated adult germ-free and conventionally reared infant rats. This study was carried out by H-CDR3 spectratype analysis of VH(PC7183)-encoded Ig VHDJH -, transcripts expressed by purified splenic MZ-B,cells and other B,cell subsets. We show that MZ-B,cells in both adult germ-free and conventionally reared infant (14-day-old) rats are H-CDR3-selected cells, providing strong evidence that recruitment and selection of MZ-B,cells is driven by self antigens. Regardless of their Igh haplotype, B6.,,,SEG mice show a restricted B cell distribution by light chain subtype with ,x dominance in all peripheral compartments except peritoneal cavity where ,2 is dominant. This distribution suggests that selection mechanisms act differently in different B cell compartments on ,2 and ,x bearing B cells. Sequence analysis before or following immunization did not reveal unusual mechanisms of diversification. B6.,,,SEG mice still respond to various challenging antigens using new Ab patterns. In particular, regardless of Igha or Ighb haplotypes, the anti-2,4-dinitrophenyl response is characterized by a restricted diversity for both heavy and light chains and a delayed IgG response when compared to B6 and B6.,, mice. We suggest that the delayed IgG response is due to the expansion of marginal zone B cells whereas follicular B cells are strongly reduced. [source] Expression of individual immunoglobulin genes occurs in an unusual system consisting of multiple independent lociEUROPEAN JOURNAL OF IMMUNOLOGY, Issue 9 2004Donna Abstract Humoral immunity is effected through the rearrangement of immunoglobulin (Ig) genes in individual somatic cells committed to the B,lymphocyte lineage. Haplotype or allelic exclusion restricts B,lymphocytes to the expression of a single Ig receptor that can sustain further somatic modification. In most species, a specific Ig chain is encoded at a single genetic locus. However, in cartilaginous fish, hundreds of independent Ig heavy- (IgH) and Ig light-chain (IgL) gene loci are present, many of which are joined in the germ line. Ig gene transcripts have been amplified from single peripheral blood lymphocytes isolated from the clearnose skate (Raja eglanteria) using reverse-transcription PCR, and a single productive IgH transcript was detected in the majority of cells analyzed. Similarly, only a single IgL transcript was detected in over half of the individual cells. Taken together, these findings suggest that a mechanism for haplotype exclusion arose early in the evolution of antibody diversity and is independent of a single genetic locus. [source] GENETIC STUDY: The interaction between the dopamine transporter gene and age at onset in relation to tobacco and alcohol use among 19-year-oldsADDICTION BIOLOGY, Issue 4 2009Brigitte Schmid ABSTRACT Recent evidence suggests that heterogeneity in the age at onset could explain the inconsistent findings of association studies relating the dopamine transporter (DAT1) gene with alcohol and nicotine consumption. The aim of this study was to examine interactions between two DAT1 polymorphisms and different initiation ages with regard to alcohol and tobacco consumption levels and dependence. Two hundred and ninety-one young adults (135 males, 156 females) participating in the Mannheim Study of Children at Risk were genotyped for the 40-bp variable number of tandem repeats (VNTR) and rs27072 polymorphisms of DAT1. Age at initiation was assessed at age 15 and 19 years. Information about current alcohol and tobacco consumption was obtained at age 19 years using self-report measures and structured interviews. Results suggest that age at onset of intensive consumption moderated the association of the DAT1 gene with early adult substance use and dependence, revealing a DAT1 effect only among individuals homozygous for the 10r allele of the 40-bp VNTR who had started daily smoking or being intoxicated early in life. Equally, carriers of the T allele of the rs27072 polymorphism reporting an early age at first intoxication showed higher current alcohol consumption at age 19 years. In contrast, no interaction between rs27072 and the age at first cigarette with regard to later smoking was observed. These findings provide evidence that the DAT1 gene interacts with an early heavy or regular drug exposure of the maturing adolescent brain to predict substance (ab)use in young adulthood. Further studies are required to confirm these findings. [source] GENETIC STUDY: BclI glucocorticoid receptor polymorphism and smoking in the general populationADDICTION BIOLOGY, Issue 3 2009Satu J. Siiskonen ABSTRACT We studied the hypothesis that the BclI polymorphism of the glucocorticoid receptor gene is associated with an increased probability of being a (heavy) smoker and a decreased ability to quit smoking. The study cohort consisted of all subjects in the Rotterdam Study, a Dutch population-based cohort of people aged 55 years and older, for whom BclI genotyping and smoking status at baseline were available. In prospective analyses, the smoking status was reassessed during three additional examination rounds. Logistic regression analysis was used to study the association between BclI polymorphism and being a smoker or a heavy smoker at baseline. Furthermore, the relationship between BclI polymorphism and incident smoking cessation was tested with Cox proportional hazards analysis within those who smoked at baseline. In total, 6358 subjects were included in the study. The presence of a G-allele was not associated with current smoking at baseline [odds ratio (OR) = 0.96, 95%confidence interval (CI): 0.85,1.09] or with the incidence of smoking cessation during follow-up [hazard ratio (HR) = 0.98, 95%CI: 0.80,1.19]. Within current smokers, having a G-allele was not significantly associated with the risk of being a heavy smoker when measured by pack-years smoked (OR = 1.07, 95%CI: 0.85,1.35) or daily consumption of tobacco (OR = 1.10, 95%CI: 0.88,1.37). We were not able to replicate the earlier findings indicating that the proportion of current smokers is lower among carriers of the CC-genotype of the BclI glucocorticoid receptor. Furthermore, the BclI glucocorticoid receptor polymorphism did not predict the incidence of smoking cessation in the general elderly population. [source] Changes in shrinkage of restored soil caused by compaction beneath heavy agricultural machineryEUROPEAN JOURNAL OF SOIL SCIENCE, Issue 4 2008B. Schäffer Summary Compaction is a major cause of soil degradation. It affects not only the porosity of the soil, but also the soil's hydrostructural stability. Soil that is restored after temporary removal is particularly sensitive to compaction. We investigated the effects of trafficking with a heavy combine harvester on the shrinkage behaviour of a restored soil that had been gently cultivated for several years. We tested the hypothesis that compaction decreases the hydrostructural stability of restored soil by analysing simultaneously measured shrinkage and water retention curves of undisturbed soil samples. Shrinkage strongly depended on clay and organic carbon content. Taking account of this influence and normalizing the shrinkage parameters with respect to these soil properties, we found pronounced effects of trafficking on shrinkage. Ten passes with the combine harvester decreased the structural porosity by about 40% at maximum swelling and by about 30% at the shrinkage limit and increased the bulk density by 8% at maximum swelling and by 10% at the shrinkage limit, but did not significantly affect the porosity of the soil plasma. Moreover, trafficking modified shrinkage, increasing the slopes of the shrinkage curve in the basic and structural shrinkage domains by about 30% and more than 150% after 10 passes, respectively. Evidently the aggregate structure was strongly destabilized. The results indicate that the hydrostructural stability of the soil was still very sensitive to compaction by trafficking even 5 years after restoration. MurphyArticle first published online: 13 OCT 200 Summary Phosphorus loss from agricultural soils to water is recognized as a major contributor to eutrophication of surface water bodies. There is much evidence to suggest that liming, a common agricultural practice, may decrease the risk of P loss by decreasing P solubility. An unsaturated leaching column experiment, with treatments of control and two lime rates, was carried out to investigate the effects of liming on P mobility in a low-P acid Irish soil, which was sieved and then packed in columns. Phosphorus was applied at the soil surface in the form of KH2PO4 in solution or as cow slurry. Soil solution was sampled at time intervals over depth and analysed for P fractions. Organic P (OP) was the dominant form of P mobile in soil solution. Liming increased OP mobility, probably through increased dispersion of OP with increased pH. Slurry application also increased OP mobility. Results indicated the potential for OP loss following heavy (100 m,3 ha,1) cow slurry application, even from low-P soils, and suggested that liming may increase this risk. Reactive P (RP) was sorbed strongly and rapidly by the soil and did not move substantially below 5 cm depth. As a result, Olsen-P values in the top 2 cm were greatly increased, which indicates an increased risk of RP loss in overland flow. Lime showed little potential as a soil amendment to reduce the risk of P loss. In particular, the focus of the therapist is on the partners' interactions, their hesitations and their surprises. Working in this way opens space for the partners to reflect on what they experience as crucial in their bond. The protocol is illustrated with two detailed case examples. Fishing pressure has been heavy over a prolonged period and the recent decades have shown dramatic decline in abundance of many stocks. The Arcto-Norwegian (or North-east Arctic) cod stock in the Barents Sea is now the largest stock of Atlantic cod. Recruitment to this stock has varied extensively during the last 60 yr. There is evidence for fluctuations in climate, particularly sea temperature, being a main cause for this variability, higher temperatures being favourable for survival throughout the critical early life stages. Our studies of time series present compelling evidence for a strengthening of the climate,cod recruitment link during the last decades. We suggest this is an effect of the age and length composition of the spawning stock having changed distinctly. The age of the average spawner has decreased by more than 3 yr from between 10 and 11 in the late 1940s to 7,8 in the 1990s, average length from just above 90 cm to around 80 cm. The number of age classes contributing to the spawning stock has also decreased, while the number of length groups present increased slightly. Significant decrease in age of spawners has frequently been described for other heavily fished stocks worldwide. We therefore find it likely that the proposed mechanism of increased influence of climate on recruitment through changes in the spawning stock age and size composition is of a general nature and might be found in other systems. [source] Relationship between presence of basidiomes, above-ground symptoms and root infection by Collybia fusipes in oaksFOREST PATHOLOGY, Issue 1 2000B. Marçis Summary Collybia fusipes is a common cause of root rot on oak in the north of France. Collybia fusipes basidiomes can be as frequent on oaks in stands where no decline of the trees occurs compared with stands where the decline is chronic. This might be explained by differences in the amount of roots damaged by the parasite. To test that hypothesis, 430 oak trees, Quercus petraea, Quercus robur and Quercus rubra, located in six forests were selected. Half of them showed C. fusipes basidiomes at the trunk base. The association between presence of basidiomes and decline of affected trees depended on the forest. The level of infection of each tree by C. fusipes, as well as the crown appearance, the tree height : diameter at breast height ratio, age and sapwood width were determined. The presence of C. fusipes basidiomes was always associated with significant root infections. The crowns of the trees deteriorated with increasing level of root infection and the decline was severe only when the root damage was heavy. Although the decline of trees that were heavily damaged by C. fusipes was severe in some of the stands, in others, it was only mild, and so the differences in tree decline between the stands could not be attributed solely to differences in root infection severity. Cells of neuronal origin are acutely sensitive to glutathione depletion, providing an avenue for studying the mechanisms invoked for neuronal survival in response to oxidant challenge. We investigated the changes in mRNA profile in HT22 hippocampal cells following administration of homocysteic acid (HCA), a glutathione-depleting drug. [source] Beyond stratigraphic noise: Unraveling the evolution of stratified assemblages in faunalturbated sitesGEOARCHAEOLOGY: AN INTERNATIONAL JOURNAL, Issue 6 2006Eugène Morin This study explores the stratigraphic evolution of archaeological assemblages in faunalturbated sites. These sites are sometimes described as having limited archaeological value because of varying degrees of occupation mixing. It is argued here that the vertical distribution of assemblages is predictable in faunalturbated contexts. Understanding this vertical evolution may lead to a better assessment of the archaeological potential of a cultural sequence. In faunalturbated soils, the stratigraphic evolution of the assemblages is largely dictated by faunal activity. Tunneling by small animals tend to "sink" cultural remains through the deposits. Although objects sink at a relatively fast pace near the surface, the process slows down as the objects reach the bottom of the biomantle where the sediments are more compact. This process affects the shape of the artifact vertical distribution. The analysis of a multicomponent site from southern Québec is used to model how assemblages may evolve temporally in faunalturbated settings. At Station 3-avant, the stratigraphic distribution of the cultural remains appears to evolve from an upward-skewed distribution to a bottom-skewed distribution through time. This evolution is interpreted to result from the fact that objects are often too large and/or heavy to be moved upward by the pedofauna. In situations in which this stage of stratigraphic evolution is not yet reached, enough information about temporal successions may be retained for the site to be useful for investigating diachronic change. Clua Abstract Narrow-leaved bird's-foot-trefoil (Lotus tenuis) is a perennial forage legume adapted to waterlogged and heavy and infertile soils and can replace alfalfa (Medicago sativa) in areas with these soils in Argentina. Its seeds are hard and water-impermeable but the effects of environmental factors on seed dormancy and germination are not known. The objective was to evaluate the hypothesis that water availability during seed development and maturation affects the degree of hardseededness in L. tenuis by changing seed coat properties, conditioning water uptake through the seed coat; and subsequently affecting dormancy, germination and speed of germination. Seeds were harvested in December/January and in February in both 1993/1994 and 1994/1995 from a permanent pasture of L. tenuis growing in a Hapludol soil in San Miguel del Monte province of Buenos Aires. Environmental conditions of each anthesis-harvest period were determined. Seeds of each harvest were subjected to chilling, washing and mechanical scarification. After 12 months seeds from each harvest were observed in a scanning electron microscope. The water deficit of the soil and relative humidity were greater in the second than the first anthesis-harvest period in both seasons. In 1993/1994 the control treatment in December had a higher germination rate than the February control seeds (0·40 vs. 0·20) and a faster germination rate. Mechanical scarification and chilling significantly enhanced the germination rate (0·95) and its speed in seeds of both harvests. Low temperatures significantly enhanced germination rate, starting after 60 d for the seeds harvested in December, and 90 d for the seeds harvested in February. In 1994/1995 the results were similar but both the January and February control treatments had higher germination rates (0·60 vs. 0·40) than in the previous year. Seeds harvested in February were more dormant in both years. These differences could be explained by the conditions in February anthesis-harvest period in both years that could have hastened the natural dehydration process of seed, changing integument structure and enhancing its impermeability. [source] Study of Five Discrete-Interval-Type Ground Water Sampling DevicesGROUND WATER MONITORING & REMEDIATION, Issue 3 2004Louise V. Parker Five relatively newly developed discrete-interval-type ground water sampling devices,the KABIS, HydraSleeve, Discrete Interval, Pneumo-Bailer, and USGS passive diffusion bag (PDB) samplers,were tested to determine their ability to recover representative concentrations of a variety of analytes,volatile organics, explosives, pesticides, and metals,from a standpipe and trichloroethylene (TCE) from a deep monitoring well. Samples taken from a well were compared with samples taken using low-flow sampling. The PDB sampler was the easiest to use, but could only be used to sample volatile organic compounds (VOCs). The HydraSleeve and the KABIS samplers were also easy to use; these devices produced representative concentrations of explosives, pesticides, and metals in the standpipe experiments, but elevated the turbidity in our monitoring well. Although there were statistically significant differences for some VOCs with both devices, these differences were generally very small (< 5%). The one exception was an 18% loss of TCE with the KABIS sampler. The Discrete Interval sampler and the Pneumo-Bailer are pressurized devices that are designed to only collect a sample when activated. The Pneumo-Bailer was heavy and awkward to handle, required a nitrogen tank in the field, and involved many steps to operate. The Discrete Interval sampler only required a bicycle pump to pressurize the chamber and was lighter and easier to handle and operate than its counterpart. Both devices generally delivered representative concentrations of all the analytes tested. [source] What is the current evidence for antigen involvement in the development of chronic lymphocytic leukemia?HEMATOLOGICAL ONCOLOGY, Issue 1 2006Gerard Tobin Abstract For many years it has been evident that B-cell chronic lymphocytic leukemia (CLL) displays preferential usage of individual immunoglobulin (Ig) variable heavy chain (VH) genes. The VH1-69 gene was the first to be reported overrepresented in a large number of CLL patients, where the VH1-69+ CLL rearrangements showed characteristic molecular features, such as unmutated VH genes, usage of specific diversity/joining gene segments, and a longer than average complementarity determining region (CDR) 3 with certain common amino acid motifs. Also, biased usage of the VH3-07 and VH4-34 genes with specific rearrangement characteristics was reported in CLL. These findings led to the speculation that antigens could be involved during CLL development by triggering proliferation of B-cells with specific B-cell receptors (BCRs) leading to an increased risk of transforming events. Recently, we characterized a subset of CLL utilizing the VH3-21 gene that also displayed peculiar Ig features, e.g. very short and homologous CDR3s, predominant , expression and preferential V,2-14 gene usage. This VH3-21+ subgroup also had poor prognosis despite the fact that two-thirds of cases carried mutated VH genes. Moreover, we and others have thereafter described further CLL subsets with very similar heavy and light chain gene rearrangement features. These latter findings of subsets expressing restricted BCRs have emphasized the hypothesis that antigens could play a role during the pathogenesis of CLL. Interestingly, recombinant antibodies produced from these restricted subsets showed similar cytoplasmatic reactivity within each group, thus suggesting recognition of a limited number of autoantigens. Sometimes, they show low and varying levels of heavy and light chains synthesis. Therefore it is reasonable to preserve generated specificities of light and heavy chains by cloning them to phagemid vector and creating phage display library. The aim of this study was to construct phage display library of Fab fragments recognizing sperm surface antigens. The source of mRNA constituted seven hybridoma cell lines producing antisperm antibodies which was proved by ELISA, and agglutination test as well as by inhibition of sperm to penetrate hamster oocytes. Fragments of cDNA encoding ,/, and , chains were cloned into pComb3HSS phagemid vector and amplified in XL-1Blue. The library was panned against whole unfixed sperm cells. Three positive clones selected after fourth round of panning showed heavy chain belonging to VH4 family, two of them (G28, K61) possessed lambda chain from VL2 family and one (H43) kappa chain from VK1 family. As these Fabs revealed similarities to antibodies against some proteins involved in sperm motility and cell fusion it can be suggested that these Fabs may be a cause of infertility. Finally, we proved that it is feasible to preserve specificities produced by human hybridomas using phage display technique and we recovered some Fabs which may be of diagnostic and research value, and may also have some value for contraceptive vaccine. [source] Carcinogenetic impact of ADH1B and ALDH2 genes on squamous cell carcinoma risk of the esophagus with regard to the consumption of alcohol, tobacco and betel quidINTERNATIONAL JOURNAL OF CANCER, Issue 6 2008Chien-Hung Lee Abstract The consumption of alcohol, tobacco and betel quid has been found to be an important contributor to esophageal squamous cell carcinoma (ESCC) in Taiwan. The genotoxic effect of the ADH1B and ALDH2 genes modulating an individual's alcohol-metabolizing capacity on ESCC may be linked to drinking behavior, intake pattern and other exogenous factors. To investigate the interplay of these genetic and environmental factors in determining the risk of ESCC, a multicenter case-control study was conducted. Here, 406 patients with pathology-proven ESCC, as well as 656 gender, age and study hospital matched controls were recruited. Genetic polymorphisms of ADH1B and ALDH2 appeared to correlate with the abstinence of alcohol, though not with tobacco and betel quid. Within the same levels of alcohol consumption, carcinoma risks increased along with an increase in the numbers of ADH1B*1 and ALDH2*2 alleles. The inactive ALDH2*1/*2 genotype was found to multiplicatively interact with a low-to-moderate (0.1,30 g/day) and a heavy (>30 g/day) ethanol intake to increase the ESCC risk (the joint aOR = 14.5 and 102.6, respectively). Among low-to-moderate drinkers, a smoking-dependent carcinogenetic effect for the ADH1B*1/*1 and ALDH2*1/*2+*2/*2 genotypes was recognized, with significant risks found in smokers, but not in nonsmokers. Further, a supra-multiplicative combined risk of ESCC for alcohol and tobacco use was identified among carriers of the ADH1B*1/*1 genotype (p for interaction = 0.042). In conclusion, the interplay of the ADH1B and ALDH2 genotypes, in conjunction with a behaved drinking habit and a practiced drinking pattern, along with continued tobacco consumption, plays an important pathogenic role in modulating ESCC risk. © 2007 Wiley-Liss, Inc. [source] China's snow disaster in 2008, who is the principal player?INTERNATIONAL JOURNAL OF CLIMATOLOGY, Issue 14 2009Gao Hui Abstract The unprecedented snow disaster in January 2008 brought serious human and economic losses to China. It has been suggested that the La Nina event is the principal cause. But analysis indicates that in December 2007, the circulation patterns in the tropical regions are quite similar with those in January 2008. In contrast large differences existed at high latitudes, especially the Siberia high (SH) and the north polar vortex (NPV). The differences can also be found between other extreme heavy and light snow years. In the extreme heavy (light) snow years, the SH is stronger (weaker) and the NPV is deeper (shallower). But these extreme snow events don't correspond to ENSO events well. Statistical results also indicate that both the SH and the NPV are independent of ENSO.
