Heterocyclic compounds from 4h -3,1-benzoxazin-4-one derivatives as anticancer agent

JOURNAL OF HETEROCYCLIC CHEMISTRY, Issue 7 2005
    Taha M Abdel-Rahman
Behaviour of 2-(4-oxo-4H -benzo[d][l,3]oxazin-2-yl)-benzoic acid (1) towards nitrogen nucleophiles namely, hydrazine hydrate, in different solvents, ammonium acetate, and o -phenylenediamine has been investigated to give aminoquinazolin-4-one, benzotriazepinone, spiro-type compound, and nitrogen bridgehead compounds 3-5, respectively. Also, reactivity of the aminoquinazolin-4-one 2 towards carbon elec-trophiles such as ethyl acetoacetate, ethyl phenylacetate, ethyl chloroacetate, and aromatic aldehydes has been discussed. Reaction of Schiff s base 8 with sulfur nucleophiles namely o -aminothiophenol and/or thio-glycolic acid afforded Michael type adducts. Structural assignments, of products 1-24 have been confirmed by elemental analysis and spectral data (1H- and 13C -NMR and MS fragmentation). The bioassay indicates that some of the target compounds obtained have good selective anticancer activity. [source]

    Synthesis of 1,3-Diphospha-2,3-dihydro-1H -phenalenes

EUROPEAN JOURNAL OF INORGANIC CHEMISTRY, Issue 10 2010
    Arkadii Tarasevych
Abstract The reaction of 1,8-dilithionaphthalene (1) with methylenebis[(dialkylamino)chlorophosphanes] [R2N(Cl)PCH2P(Cl)NR2; R = Me (2a), Et (2b)] leads to the formation of new heterocyclic compounds, 1,3-diphospha-2,3-dihydro-1H -phenalenes 3a,b as a mixture of cis and trans isomers. DFT calculations indicate that the cis isomers are thermodynamically more stable by about 1,3 kcal/mol than the trans isomers. Compounds 3a,b can be converted into dithio the derivatives 5a,b and the borane complexes 6a,b, which were characterized by NMR spectroscopy and investigated by X-ray diffraction analysis. The dialkylamino groups in 3 can be substituted by chlorine to give the chlorophosphane 7. [source]

Gold(I) Catalysis: Selective Synthesis of Six- or Seven-Membered Heterocycles from Epoxy Alkynes

EUROPEAN JOURNAL OF ORGANIC CHEMISTRY, Issue 19 2009
    Lun-Zhi Dai
Abstract Gold(I)-catalyzed intramolecular cycloisomerization of ketone-substituted epoxides with alkynes to six- or seven-membered heterocyclic compounds is firstly introduced in this paper. This procedure involves a cascade isomerization of the ketone-substituted epoxides into 1,3-diketones in the presence of a Lewis acid and subsequent gold(I)-catalyzed selective intramolecular addition of an oxygen or a carbon nucleophile to the alkynes. Reactions of Allyl Alcohols of the Pinane Series and of Their Epoxides in the Presence of Montmorillonite Clay

HELVETICA CHIMICA ACTA, Issue 2 2007
    Irina
Abstract The reactivity of allyl alcohols of the pinane series and of their epoxides in the presence of montmorillonite clay in intra- and intermolecular reactions was studied. Mutual transformations of (+)- trans -pinocarveol ((+)- 2) and (,)-myrtenol ((,)- 3a) were major reactions of these compounds on askanite, bentonite clay (Schemes,1 and 2). However, the two reactions gave different isomerization products, indicating that the reactivity of the starting alcohol (+)- 2 or (,)- 3a was different from that of the same compound (+)- 2 or (,)- 3 formed in the course of the reactions. (,)- cis - and (+)- trans -Verbenol ((,)- 16 and (+)- 12, resp.), as well as (,)- cis -verbenol epoxide ((,)- 20) reacted with both aliphatic and aromatic aldehydes on askanite, bentonite clay giving various heterocyclic compounds (Schemes,4, 5 and 7); the reaction path depended on the structure of both the terpenoid and the aldehyde. Chalcogenation reactions of a stable ferrocenyldiphosphene: Formation of thia-, selena-, and telluradiphosphiranes,

    HETEROATOM CHEMISTRY, Issue 5 2008
    Noriyoshi Nagahora
The reactions of TbtPPTbt (Tbt = 2,4,6-tris[bis(trimethylsilyl)methyl]phenyl) with elemental sulfur and selenium in the presence of triethylamine resulted in the formation of the corresponding thia- and selenadiphosphiranes in 71% and 65%, respectively, whereas the tellurization reaction was unsuccessful probably due to the steric reason. On the other hand, treatment of TbtPPFc (Fc = ferrocenyl) with elemental sulfur, selenium, and tributylphosphine telluride afforded the corresponding thia-, selena-, and telluradiphosphiranes in 84%, 88%, and 66% yields, respectively. The molecular structures of these three-membered heterocyclic compounds were confirmed by spectroscopic analysis. © 2008 Wiley Periodicals, Inc. Copper(I)-Catalyzed Synthesis of Novel 4-(Trifluoromethyl)-[1,2,3]triazolo[1,5- a]quinoxalines via Cascade Reactions of N -(o -Haloaryl)alkynylimine with Sodium Azide

ADVANCED SYNTHESIS & CATALYSIS (PREVIOUSLY: JOURNAL FUER PRAKTISCHE CHEMIE), Issue 8 2010
    Zixian Chen
Abstract Novel tricyclic 4-(trifluoromethyl)-[1,2,3]triazolo[1,5- a]quinoxalines were readily prepared from N -(o -haloaryl)alkynylimines and sodium azide via copper(I)-catalyzed tandem reactions. This synthetic strategy provides an efficient way to access a library of novel heterocyclic compounds that are of interest in drug discovery. [source]

    Gold(I)-Catalyzed One-Pot Tandem Coupling/Cyclization: An Efficient Synthesis of Pyrrolo-/Pyrido[2,1- b]benzo[d][1,3]oxazin- 1-ones

ADVANCED SYNTHESIS & CATALYSIS (PREVIOUSLY: JOURNAL FUER PRAKTISCHE CHEMIE), Issue 2-3 2010
    Yu Zhou
Abstract A highly efficient method has been developed for the one-pot synthesis of multi-ring heterocyclic compounds such as pyrrolo-/pyrido[2,1- b]benzo[d][1,3]oxazin-1-ones from o -aminobenzyl alcohols via a gold(I)-catalyzed tandem coupling/cyclization reaction. Significantly, the strategy presents a straightforward and efficient approach to construct novel tricyclic or polycyclic molecular architectures in which two new CN bonds and one CO bond are formed in a one-pot reaction operation from two simple starting materials. Moreover, a broad spectrum of substrates can participate in the process effectively to produce the desired products in good yields. [source]
