Selected AbstractsGeneration of the improved recombinant xylose-utilizing Saccharomyces cerevisiae TMB 3400 by random mutagenesis and physiological comparison with Pichia stipitis CBS 6054FEMS YEAST RESEARCH, Issue 3 2003C.Fredrik Wahlbom Abstract The recombinant xylose-utilizing Saccharomyces cerevisiae TMB 3399 was constructed by chromosomal integration of the genes encoding d -xylose reductase (XR), xylitol dehydrogenase (XDH), and xylulokinase (XK). S. cerevisiae TMB 3399 was subjected to chemical mutagenesis with ethyl methanesulfonate and, after enrichment, 33 mutants were selected for improved growth on d -xylose and carbon dioxide formation in Durham tubes. The best-performing mutant was called S. cerevisiae TMB 3400. The novel, recombinant S. cerevisiae strains were compared with Pichia stipitis CBS 6054 through cultivation under aerobic, oxygen-limited, and anaerobic conditions in a defined mineral medium using only d -xylose as carbon and energy source. The mutation led to a more than five-fold increase in maximum specific growth rate, from 0.0255 h,1 for S. cerevisiae TMB 3399 to 0.14 h,1 for S. cerevisiae TMB 3400, whereas P. stipitis grew at a maximum specific growth rate of 0.44 h,1. All yeast strains formed ethanol only under oxygen-limited and anaerobic conditions. The ethanol yields and maximum specific ethanol productivities during oxygen limitation were 0.21, 0.25, and 0.30 g ethanol g xylose,1 and 0.001, 0.10, and 0.16 g ethanol g biomass,1 h,1 for S. cerevisiae TMB 3399, TMB 3400, and P. stipitis CBS 6054, respectively. The xylitol yield under oxygen-limited and anaerobic conditions was two-fold higher for S. cerevisiae TMB 3399 than for TMB 3400, but the glycerol yield was higher for TMB 3400. The specific activity, in U mg protein,1, was higher for XDH than for XR in both S. cerevisiae TMB 3399 and TMB 3400, while P. stipitis CBS 6054 showed the opposite relation. S. cerevisiae TMB 3400 displayed higher specific XR, XDH and XK activities than TMB 3399. Hence, we have demonstrated that a combination of metabolic engineering and random mutagenesis was successful to generate a superior, xylose-utilizing S. cerevisiae, and uncovered distinctive physiological properties of the mutant. [source] Storage of biodegradable polymers by an enriched microbial community in a sequencing batch reactor operated at high organic load rateJOURNAL OF CHEMICAL TECHNOLOGY & BIOTECHNOLOGY, Issue 11 2005Davide Dionisi Abstract The production of polyhydroxyalkanoates (PHAs) from organic acids by mixed bacterial cultures using a process based on aerobic enrichment of activated sludge, that selects for mixed microbial cultures able to store PHAs at high rates and yields, is described. Enrichment resulted from the selective pressure established by periodic feeding the carbon source in a sequencing batch reactor (SBR); a mixture of acetic, lactic and propionic acids was fed at high frequency (2 hourly), high dilution rate (1 d,1), and at high organic load rate (12.75 g chemical oxygen demand (COD) L,1 d,1). The performance of the SBR was assessed by microbial biomass and PHA production as well as the composition and polymer content of the biomass. A final batch stage was used to increase the polymer concentration of the excess sludge produced in the SBR and in which the behaviour of the biomass was investigated by determining PHA production rates and yields. The microbial biomass selected in the SBR produced PHAs at high rate [278 mg PHAs (as COD) g biomass (as COD),1 h,1, with a yield of 0.39 mg PHAs (as COD) mg removed substrates (as COD),1], reaching a polymer content higher than 50% (on a COD basis). The stored polymer was the copolymer poly(3-hydroxybutyrate/3-hydroxyvalerate) [P(HB/HV)], with an HV fraction of 18% mol mol,1. The microbial community selected in the SBR was analysed by DGGE (denaturing gradient gel electrophoresis). The operating conditions of the SBR were shown to select for a restricted microbial population which appeared quite different in terms of composition with respect to the initial microbial cenosis in the activated sludge used as inoculum. On the basis of the sequencing of the major bands in the DGGE profiles, four main genera were identified: a Methylobacteriaceae bacterium, Flavobacterium sp, Candidatus Meganema perideroedes, and Thauera sp. The effects of nitrogen depletion (ie absence of growth) and pH variation were also investigated in the batch stage and compared with the SBR operative mode. Absence of growth did not stimulate higher PHA production, so indicating that the periodic feed regime fully exploited the storage potential of the enriched culture. Polymer production rates remained high between pH 6.5 and 9.5, whereas the HV content in the stored polymer strongly increased as the pH value increased. This study shows that polymer composition in the final batch stage can readily be controlled independently from the feed composition in the SBR. Kliphuis Abstract To be able to study the effect of mixing as well as any other parameter on productivity of algal cultures, we designed a lab-scale photobioreactor in which a short light path (SLP) of (12 mm) is combined with controlled mixing and aeration. Mixing is provided by rotating an inner tube in the cylindrical cultivation vessel creating Taylor vortex flow and as such mixing can be uncoupled from aeration. Gas exchange is monitored on-line to gain insight in growth and productivity. The maximal productivity, hence photosynthetic efficiency, of Chlorella sorokiniana cultures at high light intensities (1,500 ,mol m,1 s,1) was investigated in this Taylor vortex flow SLP photobioreactor. We performed duplicate batch experiments at three different mixing rates: 70, 110, and 140 rpm, all in the turbulent Taylor vortex flow regime. For the mixing rate of 140 rpm, we calculated a quantum requirement for oxygen evolution of 21.2 mol PAR photons per mol O2 and a yield of biomass on light energy of 0.8 g biomass per mol PAR photons. The maximal photosynthetic efficiency was found at relatively low biomass densities (2.3 g L,1) at which light was just attenuated before reaching the rear of the culture. When increasing the mixing rate twofold, we only found a small increase in productivity. On the basis of these results, we conclude that the maximal productivity and photosynthetic efficiency for C. sorokiniana can be found at that biomass concentration where no significant dark zone can develop and that the influence of mixing-induced light/dark fluctuations is marginal. © 2010 American Institute of Chemical Engineers Biotechnol. Prog., 2010 [source] Packed Bed Column Fermenter and Kinetic Modeling for Upgrading the Nutritional Quality of Coffee Husk in Solid-State FermentationBIOTECHNOLOGY PROGRESS, Issue 6 2001Débora Brand Studies were carried out to evaluate solid-state fermentation (SSF) for the upgradation of the nutritional quality of coffee husk by degrading the caffeine and tannins present in it. SSF was carried out by Aspergillus niger LPBx in a glass column fermenter using factorial design experiments and surface response methodology to optimize bioprocess parameters such as the substrate pH and moisture content and aeration rate. The first factorial design showed that the moisture content of the substrate and aeration rate were significant factors for the degradation of toxic compounds, which was confirmed by the second factorial design too. The kinetic study showed that the degradation of toxic compounds was related to the development of the mold and its respiration and also to the consumption of the reducing sugars present in coffee husk. From the values obtained experimentally for the oxygen uptake rate and CO2 evolved, the system determined a biomass yield (Yx/o) of 3.811 (g of biomass)·(g of consumed O2),1 and a maintenance coefficient (m) of 0.0031 (g of consumed O2)·(g biomass of biomass),1·h,1. The best results on the degradation of caffeine (90%) and tannins (57%) were achieved when SSF was carried out with a 30 mL·min,1 aeration rate using coffee husk having a 55% initial moisture content. The inoculation rate did not affect the metabolization of the toxic compounds by the fungal culture. After SSF, the protein content of the husk was increased to 10.6%, which was more than double that of the unfermented husk (5.2%). [source] |
