Quality and Functional Characteristics of Chungkukjang Prepared with Various Bacillus sp.

ABSTRACT: Bacillus circulans, Brevibacillus brevis, B. licheniformis, B. coagulans, B. subtilis, and B. sterothermophillus were isolated and identified from chungkukjangs (Korean traditional soybean paste fermented for a few day). Chungkukjang was prepared on a laboratory scale with soybeans and the isolated strains. Characteristics of the chungkukjangs including slime material content, free amino acid content, sensory qualities, and antimutagenicity were determined. The content of slime material, which is an important indicator of the quality of chungkukjang, was highest in B. licheniformis -inoculated chungkukjang, andlowestin B. sterothermophillus -inoculated chungkukjang. The total content of glycine, glutamic acid, aspartic acid, and arginine, which contribute a savory taste to chungkukjangs, was highest in B. licheniformis-inoculated chungkukjang. The content of leucine, which gives a bitter taste, was highest inB. brevis -inoculated chungkukjang. Sensory evaluation revealed that chungkukjangs made using B. licheniformis and B. subtilishad a weak bitter taste and strong sweet and savory taste and good color, so their overall acceptability was high. Chungkukjang fermented with B. circulans and B. licheniformis inhibited N-Methyl-N,-nitro-N-nitrosoguanidine (MNNG) mutagenicity by more than 80%. B. licheniformis-inoculated chungkukjang exhibited the highest antimutagenicity against and aflatoxin B1 (AFB1) and MNNG. These results suggest that using B. licheniformis to ferment chungkukjang increases the antimutagenic properties and improves the sweet and savory taste by increasing glycine, glutamic acid, aspartic acid, and arginine concentrations. Cerebellar granule cells cultured from adolescent rats express functional NMDA receptors: an in vitro model for studying the developing cerebellum

Abstract In the developing rat cerebellum functional NMDA receptors (NMDARs) expressing the NR2C subunit have been identified on or after postnatal day 19. We obtained primary cultured cells from 19- to 35-day-old rat cerebellum that expressed few oligodendrocytes or astrocytes. Cultured cells were immunoreactive for neuron-specific proteins thus indicating a neuronal population. The primary neuron present was the granule cell as indicated by immunofluorescence for the GABAA alpha 6 subunit. Whole-cell patch-clamp experiments indicated that functional NMDARs were present. Functional characteristics of NMDARs expressed in cerebellar granule cells (CGCs) obtained from adolescent animals were similar to those previously reported for NMDARs expressed in CGCs obtained from neonatal rats. Cultured CGCs obtained from older animals contained NMDARs that were inhibited by EtOH and were less sensitive to the NR2B subunit-specific antagonist Ro 25-6981. Furthermore, NMDA-induced currents were smaller than those observed in CGCs. Western blot analysis indicated the presence of the NMDA NR2A and NR2C subunits, but not the NR2B in cultures obtained from the adolescent rats. CGCs obtained from adolescent rats express functional NMDARs consistent with a developmental profile observed in vivo. [source]


Functional characteristics of antibodies induced by Arg-gingipain (HRgpA) and Lys-gingipain (Kgp) from Porphyromonas gingivalis

Arginine-specific gingipain (HRgpA) and lysine-specific gingipain (Kgp), enzymes produced by Porphyromonas gingivalis, may be candidates for an anti,P. gingivalis vaccine. The purpose of our study was to determine whether HRgpA and Kgp have opsonic target sites and whether these sites are available and accessible on intact P. gingivalis cells. Rabbits were used to generate polyclonal antibodies to both proteins. Animals were immunized and immunoglobulin G (IgG) fractions were isolated from preimmune and immune sera. Functional characteristics of the antibodies were assessed by determining antibody titers by enzyme-linked immunosorbent assay (ELISA), generating Western immunoblots, and measuring antibody enhancement of P. gingivalis opsonization, phagocytosis and killing by polymorphonuclear leukocytes (PMN) of intact cells of strains of P. gingivalis representative of the four serotypes. Strains studied included 33277 (serotype A), A7A1,28 (serotype B), W50 (serotype C) and 381 (serotype D). Both HRgpA and Kgp induced high titers of IgG antibody. Anti-HRgpA and anti-Kgp bound to both HRgpA and Kgp demonstrating a large proportion of shared antigenic epitopes. The two antibodies bound equally well to all four P. gingivalis serotypes with titers ranging from 77 to 205 ELISA units when compared to preimmune IgG set at 1 ELISA unit. The immunoblot patterns of binding of the two antibodies to HRgpA and Kgp and to sonicates of the four P. gingivalis serotypes were virtually identical. Both antibodies detected components in HRgpA at 27, 35 and 45 kDa and in Kgp at 27, 32, 35, 40 and 55 kDa. The antibodies also detected components at or near these same positions in addition to multiple high molecular mass components in the cell sonicates of P. gingivalis. Both proteins induced antibodies that significantly enhanced opsonization as assessed by chemiluminescence, with values ranging from 130 mV to 375 mV for anti-HRgpA IgG and from 240 mV to 475 mV for anti-Kgp IgG. Both antibodies significantly enhanced PMN-mediated bacterial killing of the four P. gingivalis serotypes, although the percentage of killing varied among the serotypes (24,81% for anti-HRgpA and 37,89% for anti-Kgp). Thus, both HRgpA and Kgp express opsonic target sites and induce high titers of antibodies that opsonize and enhance killing of all four serotypes of P. gingivalis. These two proteins appear to be potential candidate antigens for an anti,P. gingivalis vaccine. [source]


Protein and the demand for hard wheats,

Wheat protein is one of the most important specifications used in domestic and import purchase contracts and is used partly as a proxy for functional quality. The purpose of this article is to analyse the demand for wheat delineated by protein class. A choice-based econometric model is specified and estimated using a novel dataset of pooled wheat shipments to individual importing countries. Buyers are importing countries that make purchase decisions among different protein levels. The model frames the choice in terms of attributes of the choice and of the importing countries. Results indicate that there have been shifts over time, and purchase probabilities are highly price elastic and vary across importing regions. Functional characteristics including wet gluten content and extraction rates have significant impacts on purchase probabilities. These results have implications for breeders as it clearly illustrates the role of protein and functional characteristics on demand. The results also have implications for analysts modelling wheat trade in that there are many factors impacting market segments that would not be captured in conventional demand specifications. Cultivation and ensilage of yacon plants (Smallanthus sonchifolius [Poepp.

Abstract Yacon (Smallanthus sonchifolius [Poepp. & Endl.] H. Robinson) plant cultivation has extended rapidly throughout Japan since 1985 because of its utility as a tuberous root crop. In this study, strains with high yields of both above-ground and underground parts were identified to investigate the use of above-ground parts for animal feed silage. In addition, we measured the anti-oxidative activity of the silage as a functional characteristic. Data from 5 years of the cultivation of 17 yacon strains suggest that the most suitable strain for planting in the Aso region of Japan is SY217, which has a high yield of both above-ground parts and tuberous roots. The root tuber parts of yacon are used almost entirely as food; actually, most foliage parts are hardly used, except for some above-ground parts, which are used as tea. The fermentation qualities of the silage using all above-ground parts and harvest residue tuberous roots (harvest residues) are high and their dry matter digestibility, investigated using pigs, is 66.7%. Fresh above-ground parts had very strong anti-oxidative activity as compared with butylated hydroxyanisole and ,-tocopherol. In addition, that activity is retained during ensiling for 4 weeks. These results suggest that the entire yacon plant can be used effectively as a feedstuff resource with the added bonus of anti-oxidant activity. [source]


Glutamate-mediated influx of extracellular Ca2+ is coupled with reactive oxygen species generation in cultured hippocampal neurons but not in astrocytes

Abstract Generation of reactive oxygen species (ROS) in brain tissue leads to neurodegeneration. The major source of ROS is the mitochondrial respiratory chain. We studied regulation of Ca2+ level, mitochondrial potential, and ROS generation in defined mixed hippocampal cell cultures exposed to glutamate (100 ,M). Recordings were made from individually identified astrocytes and neurons to compare the physiologic responses in both cell types. Neurons identified by synaptotagmin immunoreactivity were characterized functionally by the fast Ca2+ increase with K+ (50 mM) stimulation, and the astrocytes identified by glial fibrillary acidic protein (GFAP) staining had the functional characteristic of a transient Ca2+ peak in response to ATP (10 ,M) stimulation. We found that the glutamate-mediated Ca2+ response in neurons is due largely to influx of extracellular Ca2+. This is consistent with our finding that in cultured hippocampal neurons, stores depending on the activity of the sarcoendoplasmic reticulum Ca2+ ATPase (SERCA) pump had a low Ca2+ content, regardless of whether the neurons were challenged or not with K+ before applying the SERCA inhibitor cyclopiazonic acid (CPA). Astrocytes displayed a large CPA-mediated Ca2+ response, indicating a high level of Ca2+ load in the stores in astrocytes. Importantly, the rise in ROS generation due to glutamate application was cell-type specific. In neurons, glutamate induced a marked rise in generation of ROS, but not in astrocytes. In both astrocytes and neurons, the mitochondrial potential was increased in response to glutamate challenge. We conclude that in neurons, Ca2+ influx accounts for the increased ROS generation in response to glutamate. This might explain the high vulnerability of neurons to glutamate challenge compared to the vulnerability of astrocytes. Telomerase RNA component (TERC) and telomerase reverse transcriptase (TERT) function together to elongate telomeres and to protect chromosomal ends. Recent studies have discovered that overexpression of telomerase's TERT subunit promoted epidermal stem-cell mobilization, hair growth and stem-cell proliferation without changes in length of telomeres.1,2 This telomerase functional characteristic is TERC independent and is operated through a mechanism other than telomere elongation. These findings open new doors for future explorations to understand telomerase function and its interaction with other cell components in the regulation of cell senescence and tumorigenesis. A robust monitor construct with runtime fault detection,

Abstract The monitor concept provides a structured and flexible high-level programming construct to control concurrent accesses to shared resources. It has been widely used in a concurrent programming environment for implicitly ensuring mutual exclusion and explicitly achieving process synchronization. This paper proposes an extension to the monitor construct for detecting runtime errors in monitor operations. Monitors are studied and classified according to their functional characteristics. A taxonomy of concurrency control faults over a monitor is then defined. The concepts of a monitor event sequence and a monitor state sequence provide a uniform approach to history information recording and fault detection. Rules for detecting various types of faults are defined. Based on these rules, fault-detection algorithms are developed. A prototypical implementation of the proposed monitor construct with runtime fault detection mechanisms has been developed in Java. Human peripheral blood B-cell compartments: A crossroad in B-cell traffic,

Abstract A relatively high number of different subsets of B-cells are generated through the differentiation of early B-cell precursors into mature B-lymphocytes in the bone marrow (BM) and antigen-triggered maturation of germinal center B-cells into memory B-lymphocytes and plasmablasts in lymphoid tissues. These B-cell subpopulations, which are produced in the BM and lymphoid tissues, recirculate through peripheral blood (PB), into different tissues including mucosa and the BM, where long-living plasma cells produce antibodies. These circulating PB B-cells can be classified according to their maturation stage into i) immature/transitional, ii) naïve, and iii) memory B-lymphocytes, and iv) plasmablasts/plasma cells. Additionally, unique subsets of memory B-lymphocytes and plasmablasts/plasma cells can be identified based on their differential expression of unique Ig-heavy chain isotypes (e.g.: IgM, IgD, IgG, IgA). In the present paper, we review recent data reported in the literature about the distribution, immunophenotypic and functional characteristics of these cell subpopulations, as well as their distribution in PB according to age and seasonal changes. Additional information is also provided in this regard based on the study of a population-based cohort of 600 healthy adults aged from 20 to 80 years, recruited in the Salamanca area in western Spain. Detailed knowledge of the distribution and traffic of B-cell subsets through PB mirrors the immune status of an individual subject and it may also contribute to a better understanding of B-cell disorders related to B-cell biology and homeostasis, such as monoclonal B-cell lymphocytosis (MBL). © 2010 International Clinical Cytometry Society [source]


Sequential activation of transcription factors in lens induction

Since the pioneering work of the early 1900s, the lens has been used as a model system for the study of tissue development in vertebrates. A number of embryological transplantation experiments designed to elucidate the role of tissue interactions in the formation of the lens have led to the proposal of a stepwise determination model. This model has recently been refined through the identification of certain transcription factor genes, which exhibit distinct expression patterns and functional properties in the lens cell lineage. Otx2, Pax6, and Lens1 are induced by the adjacent anterior neural plate and expressed in predifferentiated lens ectoderm. Contact between the optic vesicle and lens ectoderm promotes expression of mafs, Soxs, and Prox1, which are responsible for the initiation of lens differentiation programs including crystallin expression, cell elongation, and cell cycle arrest. Further analysis of the expression and functional characteristics of these transcription factors will allow greater detail when describing the orchestration of genetic programs, which control tissue development from induction to maturation. Development of murine hepatic sinusoidal endothelial cells characterized by the expression of hyaluronan receptors

Abstract Endothelial cells (ECs) display distinct structural and functional characteristics depending on the tissue and developmental stage; however, the development of tissue-specific ECs remains poorly understood. Here, we describe the development of hepatic sinusoids in mice based on the expression of hyaluronan receptors Stab2 and Lyve-1. Flk-1+ cells in and around the liver bud begin to express Stab2 at embryonic day (E) 9.5, before the formation of vascular lumen. Hepatic sinusoidal endothelial cells (HSECs) begin to express Lyve-1 at E10.5, and both markers continue to be expressed in HSECs thereafter. Although HSECs and lymphatic ECs (LECs) are known to share functional and phenotypic characteristics, we clearly show that HSECs can be distinguished from LECs by the expression of molecular markers and higher endocytotic activity. Our results provide new insight into the development of tissue-specific ECs and phenotypic criteria to distinguish HSECs from other types of ECs, including LECs. Preclinical development of hybrid cell vaccines for multiple myeloma

Abstract Immunotherapy may provide alternative or supplementary treatment of multiple myeloma (MM). We propose that hybrid cells, formed by fusing professional antigen-presenting cells with malignant plasma cells, would induce immune responses capable of mediating tumour regression. The human B-lymphoblastoid cell line, HMy2, was fused in vitro with CD138+ bead-separated myeloma plasma cells from five patients with MM. The hybrid cell lines generated in these studies grew stably in tissue culture, and maintained their phenotypic and functional characteristics, providing self-renewing cell lines with potential for therapeutic vaccination. The hybrid cells stimulated allogeneic and autologous T-cell proliferative responses in vitro to a considerably greater degree than their respective parent myeloma plasma cells, and directly activated both CD4+ and CD8+ T-cell responses. The enhanced T-cell stimulation correlated with expression of CD80 on the hybrid cells, and was inhibited by CTLA4-Ig fusion protein. The hybrid cell lines expressed several tumour-associated antigens known to be expressed in myeloma. These data show that self-replicating cell lines with enhanced immunostimulatory properties and potential for therapeutic vaccination can be generated by in vitro fusion of ex vivo myeloma cells and B-lymphoblastoid cell lines. Platelet-derived growth factor (PDGF) in human acute myelogenous leukemia: PDGF receptor expression, endogenous PDGF release and responsiveness to exogenous PDGF isoforms by in vitro cultured acute myelogenous leukemia blasts

Abstract: We investigated effects of Platelet-derived growth factor (PDGF) and Platelet factor 4 (PF-4) on the functional characteristics of native, human acute myelogenous leukemia (AML) blasts. AML blast expression of the PDGF-receptor ,-chain was detected for a subset of patients (45%), whereas PDGF-receptor ,-chain expression was detected for most patients (90%). Constitutive AML blast release of the PDGF-AB isoform (the major form also derived from normal platelets) was detected for 43% of patients, whereas PDGF-BB release was not detected for any patient. The PDGF isoforms AA, AB and BB had dose-dependent and divergent effects on spontaneous and cytokine-dependent AML blast proliferation, whereas for constitutive cytokine secretion (IL-1,, IL-6, TNF-,) inhibitory effects were rare and all three isoforms usually had no effect or enhanced the constitutive secretion. The PDGF effects were caused by a direct effect on the AML blasts and were not dependent on the presence of serum. The PDGF effects could also be detected after in vitro culture of AML cells in the presence of IL-4+granulocyte-macrophage colony stimulating factor. PF-4 had divergent effects on proliferation and cytokine secretion by native AML blasts. Our results suggest that exogenous (e.g. platelet-secreted) PDGF and PF-4 can function as regulators of leukemic hematopoiesis and possibly also modulate the function of residual AML cells in peripheral blood stem cell grafts. On the other hand, endogenous release of PDGF-AB by native blasts may modulate the function of normal cells in the bone marrow microenvironment (e.g. bone marrow stromal cells). [source]


Germ-line and rearranged Tcrd transcription distinguish bona fide NK cells and NK-like ,,,T cells

Abstract NK cells and ,,,T cells are distinct subsets of lymphocytes that contextually share multiple phenotypic and functional characteristics. However, the acquisition and the extent of these similarities remain poorly understood. Here, using T cell receptor ,,locus-histone,2B-enhanced GFP (Tcrd-H2BEGFP) reporter mice, we show that germ-line transcription of Tcrd occurs in all maturing NK cells. We also describe a population of mouse NK-like cells that are indistinguishable from "bona fide" NK cells using standard protocols. Requirements for V(D)J recombination and a functional thymus, along with very low-level expression of surface TCR,, but high intracellular CD3, define these cells as ,,,T cells. "NK-like ,,,T cells" are CD127+, have a memory-activated phenotype, express multiple NK cell receptors and readily produce interferon-, in response to IL-12/IL-18 stimulation. The close phenotypic resemblance between NK cells and NK-like ,,,T cells is a source of experimental ambiguity in studies bridging NK and T cell biology, such as those on thymic NK cell development. Influence of filling fat type on praline products with nougat filling

Abstract The aim of this research was to examine the composition and functional characteristics of three samples of commercial filling fat intended for the making of nougat fillings for confectionary products. The crystallization rate of the fats was measured under static conditions using the NMR technique and under dynamic conditions as well as using a rotational rheometer. The functional characteristics of the fats were estimated by means of sensory values of the prepared praline product. Under the static conditions, all investigated filling fats showed a fast crystallization rate (,2,4%/min). The sample with the highest lauric acid contents started to crystallize immediately (no induction period). The longest induction period was found for the sample with the lowest saturated fatty acid and trans fatty acid contents. During the crystallization under dynamic conditions, all investigated samples started to crystallize more rapidly (rapid viscosity increase) at practically the same temperature (,17,°C). Measuring solid fat content and viscosity are two approaches of determining the suitability of a filling fat to be used in pralines. Both measurements provide valuable information, and particularly viscosity build-up is very important from the view of production on factory scale. [source]


L-type calcium channels are involved in fast endocytosis at the mouse neuromuscular junction

Abstract We used fluorescence microscopy of FM dyes-labeled synaptic vesicles and electrophysiological recordings to examine the functional characteristics of vesicle recycling and study how different types of voltage-dependent Ca2+ channels (VDCCs) regulate the coupling of exocytosis and endocytosis at mouse neuromuscular junction. Our results demonstrate the presence of at least two different pools of recycling vesicles: a high-probability release pool (i.e. a fast destaining vesicle pool), which is preferentially loaded during the first 5 s (250 action potentials) at 50 Hz; and a low-probability release pool (i.e. a slow destaining vesicle pool), which is loaded during prolonged stimulation and keeps on refilling after end of stimulation. Our results suggest that a fast recycling pool mediates neurotransmitter release when vesicle use is minimal (i.e. during brief high-frequency stimulation), while vesicle mobilization from a reserve pool is the prevailing mechanism when the level of synaptic activity increases. We observed that specific N- and L -type VDCC blockers had no effect on evoked transmitter release upon low-frequency stimulation (5 Hz). However, at high-frequency stimulation (50 Hz), L -type Ca2+ channel blocker increased FM2-10 destaining and at the same time diminished quantal release. Furthermore, when L -type channels were blocked, FM2-10 loading during stimulation was diminished, while the amount of endocytosis after stimulation was increased. Our experiments suggest that L -type VDCCs promote endocytosis of synaptic vesicles, directing the newly formed vesicles to a high-probability release pool where they compete against unused vesicles. Discharge patterns of neurons in the medial pontobulbar reticular formation during fictive mastication in the rabbit

Abstract In this study, we describe functional characteristics of neurons forming networks generating oral ingestive motor behaviours. Neurons in medial reticular nuclei on the right side of the brainstem between the trigeminal and hypoglossal motor nuclei were recorded in anaesthetized and paralysed rabbits during two types of masticatory-like motor patterns induced by electrical stimulation of the left (contralateral) or right (ipsilateral) cortical masticatory areas. Sixty-seven neurons in nucleus reticularis pontis caudalis (nPontc), nucleus reticularis parvocellularis (nParv), and nucleus reticularis gigantocellularis (Rgc) were studied. These were classified as phasic or tonic depending on their firing pattern during the fictive jaw movement cycle. Phasic neurons located in the dorsal part of nPontc were active during the jaw opening phase, whilst those in dorsal nParv tended to fire during the closing phase. In most neurons, burst duration and firing frequency changed between the two motor patterns, but there was little change in phase of firing. Tonic units were mainly recorded in the ventral half of nPontc, and at the junction between Rgc and caudal nParv. Cortical inputs with short latency from the contralateral masticatory area were more frequent in phasic (82%) than tonic (44%) neurons, whilst inputs from the ipsilateral cortex were equal in the two subgroups (57% and 56%). Phasic neurons had significantly shorter mean contralateral than ipsilateral cortical latencies, whilst there was no difference among tonic neurons. Intra- and perioral primary afferent inputs activated both types of neurons at oligo-synaptic latencies. Our results show that subpopulations of neurons in medial reticular nuclei extending from the caudal part of the trigeminal motor nucleus to the rostral third of the hypoglossal motor nucleus are active during the fictive masticatory motor behaviour. Unlike masticatory neurons in the lateral tegmentum, the medial subpopulations are spatially organized according to discharge pattern. [source]

This study analyses the relevance and the meaning given by Italians to the political labels ,left' and ,right' between 1975 and 2006. Based on responses to the open-ended question ,What do you mean by "left/right" in politics?', the study compares five alternative hypotheses on the meaning of the left-right axis and show that, despite the alleged end of ideologies, the relevance of the axis has increased over time. A core of abstract meanings persists throughout the thirty-year period considered. As the importance of abstract meanings has increased over time, reference to more concrete contents (such as ,parties' and ,leaders') has decreased. The findings thus support the hypothesis that the left-right axis has the functional characteristics of social representations. [source]


Xylanases, xylanase families and extremophilic xylanases

Abstract Xylanases are hydrolytic enzymes which randomly cleave the , 1,4 backbone of the complex plant cell wall polysaccharide xylan. Diverse forms of these enzymes exist, displaying varying folds, mechanisms of action, substrate specificities, hydrolytic activities (yields, rates and products) and physicochemical characteristics. Research has mainly focused on only two of the xylanase containing glycoside hydrolase families, namely families 10 and 11, yet enzymes with xylanase activity belonging to families 5, 7, 8 and 43 have also been identified and studied, albeit to a lesser extent. Driven by industrial demands for enzymes that can operate under process conditions, a number of extremophilic xylanases have been isolated, in particular those from thermophiles, alkaliphiles and acidiphiles, while little attention has been paid to cold-adapted xylanases. Here, the diverse physicochemical and functional characteristics, as well as the folds and mechanisms of action of all six xylanase containing families will be discussed. The adaptation strategies of the extremophilic xylanases isolated to date and the potential industrial applications of these enzymes will also be presented. [source]


Functional biodiversity of macroinvertebrate assemblages along major ecological gradients of boreal headwater streams

Summary 1. Biodiversity,environment relationships are increasingly well-understood in the context of species richness and species composition, whereas other aspects of biodiversity, including variability in functional diversity (FD), have received rather little rigorous attention. For streams, most studies to date have examined either taxonomic assemblage patterns or have experimentally addressed the importance of species richness for ecosystem functioning. 2. I examined the relationships of the functional biodiversity of stream macroinvertebrates to major environmental and spatial gradients across 111 boreal headwater streams in Finland. Functional biodiversity encompassed functional richness (FR , the number of functional groups derived from a combination of functional feeding groups and habit trait groups), FD , the number of functional groups and division of individuals among these groups, and functional evenness (FE , the division of individuals among functional groups). Furthermore, functional structure (FS) comprised the composition and abundance of functional groups at each site. 3. FR increased with increasing pH, with additional variation related to moss cover, total nitrogen, water colour and substratum particle size. FD similarly increased with increasing pH and decreased with increasing canopy cover. FE decreased with increasing canopy cover and water colour. Significant variation in FS was attributable to pH, stream width, moss cover, substratum particle size, nitrogen, water colour with the dominant pattern in FS being related to the increase of shredder-sprawlers and the decrease of scraper-swimmers in acidic conditions. 4. In regression analysis and redundancy analysis, variation in functional biodiversity was not only related to local environmental factors, but a considerable proportion of variability was also attributable to spatial patterning of environmental variables and pure spatial gradients. For FR, 23.4% was related to pure environmental effects, 15.0% to shared environmental and spatial effects and 8.0% to spatial trends. For FD, 13.8% was attributable to environmental effects, 15.2% to shared environmental and spatial effects and 5% to spatial trends. For FE, 9.0% was related to environmental variables, 12.7% to shared effects of environmental and spatial variables and 4.5% to spatial variables. For FS, 13.5% was related to environmental effects, 16.9% to shared environmental and spatial effects and 15.4% to spatial trends. 5. Given that functional biodiversity should portray variability in ecosystem functioning, one might expect to find functionally rather differing ecosystems at the opposite ends of major environmental gradients (e.g. acidity, stream size). However, the degree to which variation in the functional biodiversity of stream macroinvertebrates truly portrays variability in ecosystem functioning is difficult to judge because species traits, such as feeding roles and habit traits, are themselves strongly affected by the habitat template. 6. If functional characteristics show strong responses to natural environmental gradients, they also are likely to do so to anthropogenic environmental changes, including changes in habitat structure, organic inputs and acidifying elements. However, given the considerable degree of spatial structure in functional biodiversity, one should not expect that only the local environment and anthropogenic changes therein are responsible for this variability. Rather, the spatial context, as well as natural variability along environmental gradients, should also be explicitly considered in applied research. [source]


Assembly rules and functional groups at global biogeographical scales

Summary 1The taxonomic and functional composition of reef fish assemblages are quantified in three biogeographical regions: Great Barrier Reef, French Polynesia and Caribbean. Assemblages are described in three habitats of differing wave exposure. Functional abilities are estimated based on published analyses linking fin morphology and swimming performance. 2Two questions were addressed: (1) To what extent are labrid assemblages similar among habitats and regions? (2) To what extent are functional characteristics of fish assemblages shaped by differences in biodiversity, evolutionary history and species composition? 3All three regions display highly congruent patterns of habitat use, in terms of assemblage structure and functional characteristics, despite a five-fold difference in species richness, limited or no species overlap, and a 3·2-Myear history of isolation. Exposed reef crest assemblages were dominated by fishes with fins reflecting lift-based high-speed locomotion. In contrast to abundance-based patterns, species presence/absence data were uninformative. 4The relationship between swimming ability and habitat use reveals underlying assembly rules at a functional level, emphasizing the utility of functional attributes as a metric for comparing system-level properties in taxonomically distinct faunas. The carboxy-terminal tail region of human Cav2.1 (P/Q-type) channel is not an essential determinant for its subcellular localization in cultured neurones

A recent report on the mechanism of synaptic targeting of Cav2.2 channel suggested that this process depends upon the presence of long C-terminal tail and that protein interactions mediated by SH3-binding and PDZ-binding motifs in the tail region are important. To examine the possibility that C-terminal tail of the Cav2.1 channel and the polyglutamine stretch therein are also involved in the mechanism for channel localization, we constructed several expression plasmids for human Cav2.1 channel tagged with enhanced green fluorescent protein (EGFP) and introduced them into mouse hippocampal neuronal culture. HC construct encodes short version of Cav2.1, and HS and HL encode Cav2.1 channel with a long C-terminal tail, which contains polyglutamine tract of 13 (normal range) and 28 (SCA6 disease range) repeat units, respectively. Surprisingly, transfection with HC, HS, and HL gave essentially the same results: EGFP signal was observed in cell soma, dendrites, and the axon as well. Furthermore, mutation of the PDZ-binding motif located at the C-terminus of the long version of Cav2.1, by adding FLAG tag, did not affect the localization patterns of HS and HL as well. Therefore, the C-terminal region is not indispensable for the subcellular localization of Cav2.1 channel, nor expansion of polyglutamine length affected the localization of the channel. Thus, it is possible that the localization mechanism of Cav2.1 channel is different from that of Cav2.2, though these channels share various structural and functional characteristics. [source]


Differential contributions of the parahippocampal place area and the anterior hippocampus to human memory for scenes

Abstract Past neuroimaging research has identified a parahippocampal place area (PPA) in the posterior medial temporal lobe (MTL), which responds preferentially to visual scenes and plays a role in episodic memory for this class of stimuli. In the present positron emission tomography study, we examined to what extent the functional characteristics of the PPA resemble those of other, more anterior MTL regions across various learning and recognition-memory tasks. We also determined whether the involvement of the PPA in recognition of previously studied scenes is specific to a particular type of scene information. We found that, like the PPA, anterior hippocampal regions showed a novelty response (higher activation for novel than repeated scenes) and a stimulus-related response (higher activation for scenes than objects) during learning, indicating that MTL structures other than the PPA contribute to the encoding of novel stimulus relationships in scenes. However, these anterior hippocampal regions showed no involvement during recognition of either spatial or nonspatial information contained in scenes. The PPA, by contrast, was consistently involved in recognition of all types of scene details, presumably through interactions with co-activated parietal and occipitotemporal cortices. We suggest that MTL contributions from the PPA are sufficient to support recognition of scenes when the task can be based on a perceptually based familiarity process. Expression and functional characterization of FOXP3+CD4+ regulatory T cells in ulcerative colitis,

Abstract Background: CD4+CD25+ regulatory T cells (TR) can prevent or treat experimental murine colitis but little is known about their potential role in human inflammatory bowel disease (IBD). FOXP3 is a transcription factor that plays a critical role in the development and function of CD4+CD25+ TR. The aim of this study was to examine the presence and functional characteristics of TR cells in colonic lymphoid tissues in patients with ulcerative colitis (UC). Methods: FOXP3 expression was assessed by flow cytometry, immunohistochemistry, and reverse-transcriptase polymerase chain reaction (RT-PCR). Functional characterization of CD4+CD25+ cells was analyzed by suppression of proliferation and secretion of cytokines by cocultured effector CD4+CD25, T cells. Results: FOXP3+CD4+ T cells are increased in the lamina propria (LP) of inflamed and noninflamed areas of UC colon compared to normal colon. CD4+CD25+ T cells in UC mesenteric lymph nodes (MLN) express FOXP3 mRNA and protein and suppress the proliferation of autologous MLN CD4+CD25, T cells. The suppressor activity of MLN CD4+CD25+ T cells is cell contact-dependent but cytokine-independent. In addition, CD4+CD25+ T cells potently suppress the production of both Th1 (IFN-,, IL-2) and Th2 (IL-5, IL-13) cytokines by cocultured CD4+CD25, T cells. FOXP3+ cells localized in the T-cell-rich areas of MLN and occasionally present in the follicles. Conclusions: There is an expansion of FOXP3+CD4+ T cells in mucosal lymphoid tissues in UC. CD4+CD25+ isolated from UC MLN express FOXP3 and display features of TR cells in spite of active mucosal inflammation. Traffic flow continuum modeling by hypersingular boundary integral equations

Abstract The quantity of data necessary in order to study traffic in dense urban areas through a traffic network, and the large volume of information that is provided as a result, causes managerial difficulties for the said model. A study of this kind is expensive and complex, with many sources of error connected to each step carried out. A simplification like the continuous medium is a reasonable approximation and, for certain dimensions of the actual problem, may be an alternative to be kept in mind. The hypotheses of the continuous model introduce errors comparable to those associated with geometric inaccuracies in the transport network, with the grouping of hundreds of streets in one same type of link and therefore having the same functional characteristics, with the centralization of all journey departure points and destinations in discrete centroids and with the uncertainty produced by a huge origin/destination matrix that is quickly phased out, etc. In the course of this work, a new model for characterizing traffic in dense network cities as a continuous medium, the diffusion,advection model, is put forward. The model is approached by means of the boundary element method, which has the fundamental characteristic of only requiring the contour of the problem to be discretized, thereby reducing the complexity and need for information into one order versus other more widespread methods, such as finite differences and the finite element method. On the other hand, the boundary elements method tends to give a more complex mathematical formulation. Location-related differences in structure and function of the stratum corneum with special emphasis on those of the facial skin

Synopsis Between the two different kinds of the skin covering the body, the glabrous skin is found only on the palmo-plantar surface because of its rather simple function to protect the underlying living tissue with its remarkably thick stratum corneum (SC) from strong external force and friction. Thus, its barrier function is extremely poor. In contrast, the hair-bearing skin covers almost all over the body surface regardless of the presence of long hair or vellus hair. In regard to its SC, many dermatologists and skin scientists think that it is too thin to show any site-specific differences, because the SC is just present as an efficient barrier membrane to protect our body from desiccation as well as against the invasion by external injurious agents. However, there are remarkable regional differences not only in the living skin tissue but also even in such thin SC reflecting the function of each anatomical location. These differences in the SC have been mostly disclosed with the advent of non-invasive biophysical instruments, particularly the one that enables us to measure transepidermal water loss (TEWL), the parameter of the SC barrier function, and the one that evaluates the hydration state of the skin surface, the parameter of the water-holding capacity of the SC that brings about softness and smoothness to the skin surface. These in vivo instrumental measurements of the SC have disclosed the presence of remarkable differences in the functional properties of the SC particularly between the face and other portions of the body. The SC of the facial skin is thinner, being composed of smaller layers of corneocytes than that of the trunk and limbs. It shows unique functional characteristics to provide hydrated skin surface but relatively poor barrier function, which is similar to that observed in retinoid-treated skin or to that of fresh scar or keloidal scars. Moreover, there even exist unexpected, site-dependent differences in the SC of the facial skin such as the forehead, eyelid, cheek, nose and perioral regions, although each location occupies only a small area. Between these locations, the cheek shows the lowest TEWL in contrast to the perioral region that reveals the highest one. Moreover, these features are not static but change with age particularly between children and adults and maybe also between genders. Among various facial locations, the eyelid skin is distinct from others because its SC is associated with poor skin surface lipids and a thin SC cell layer composed of large corneocytes that brings about high surface hydration state but poor barrier function, whereas the vermillion borders of the lips that are covered by an exposed part of the oral mucosa exhibit remarkably poor barrier function and low hydration state. Future studies aiming at the establishment of the functional mapping in each facial region and in other body regions will shed light on more delicate site-dependent differences, which will provide us important information in planning the strategy to start so called tailor-made skin care for each location of the body. Résumé Entre les deux types différents de peau couvrant le crops, on trouve la peau glabre uniquement sur la surface palmo-plantaire du fait de sa fonction plutôt simple de protection du tissu vivant sous-jacent par un stratum corneum (SC) trés épais vis-à-vis des forces extérieures et de la friction. De ce fait, sa fonction barrière est extrêmement pauvre. Au contraire, la peau velue courve la presque totalité de la surface du crops, que ce soit par la présence de longs cheveux ou de duvet. En ce qui concerne son SC, la plupart des dermatologues et des scientifiques de la peau pensent qu'il est trop mince pour montrer une différence spécifique au site, attendu que le SC est simplement présent en tant que membrane barriére efficace pour protéger notre corps de la dessiccation ainsi que pour lutter contre l'invasion d'agents nuisibles externes. Cependant, il existe des différences importantes entre les sites, non seulement dans la peau vivante, mais également dans ce SC aussi mince, qui révèlent la fonctin de chaque site anatomique. Ces différences dans le SC ont surtout été révélées avec l'apparition d'instruments biophysiques non invasifs, en particulier celui qui nous permet de mesurer la perte transépidermale en eau (TEWL), le paramétre de la fonction barrière du SC et celui qui évalue l'état d'hydratation de la surface de peau, le paramètre de la capacité en rétention de l'eau du SC qui est liéà la souplesse et à la douceur à la surface de peau. Ces mesures instrumentales in vivo du SC ont révélé la présence de différences remarquables entre les propriétés fonctionnelles du SC particulièrement entre le visage et d'autres parties du corps. Le SC de la peau de la face est plus mince, car li est composé de couches plus petites de corneocytes que celui du tronc et des membres. Il montre des caractéristiques fonctionnelles uniques pour permettre l'hydratation de la surface de peau, mais une fonction barrière relativement faible, semblable à celle observée dans la peau traitée avec un rétinoïde ou à celle d'une cicatrice récente ou de cicatrices kéloidales. De plus, il existe des différences sites-dépendantes inattendues dans le SC de la peau de la face comme le front, la paupière, la joue, le nez et les régions périorales, et ce, bien que chaque emplacement occupe seulement un petit secteur. Entre ces divers emplacements, la joue montre le TEWL le plus bas par comparaison avec la région périorale qui montre le plus élevé. De plus, ces caractéristiques ne sont pas fixes, mais changent avec l'âge en particulier entre enfants et adultes et peut-être aussi entre sexes. Entre les diverses régions de la face, la peau de la paupière se distingue parce que son SC est associéà une peau pauvre en lipides de surface constituée par une mince couche de cellule composée de grand cornéocytes qui provoquent un haut état d'hydratation superficiel, mais une faible fonction barrière. A l'inverse les bordures vermillion des lévres recouvertes par une partie exposée de muqueuse orale, possèdent une fonction barrière très faible et un état d'hydratation bas. Les études futures visant àétablir la configuration fonctionnelle de chaque région de la face et d'autres régions du corps mettrons en lumière des différences sites-dépendantes plus subtiles, qui nous fourniront des informations importantes pour planifier la stratégie pour commencer le soin de la peau sur mesure si attendu pour chaque partie du corps. A short history of sweat gland biology

Synopsis The axilla, especially its microflora and axillary sweat glands as well as their secretions, is the main target of cosmetic compositions such as deodorants or antiperspirants. There are three types of sweat glands present in the axillary skin, namely apocrine, eccrine and apoeccrine sweat glands. Here, we provide an overview of the morphological, structural and functional characteristics of the different gland types and present techniques that allow their clear distinction. Moreover, we describe different forms of perspiration as physical reactions to external and internal stimuli. Dans la peau axillaire, il y a trois formes différentes de glandes sudoripares appelées apocrines, eccrines et apoeccrines. Nous donnons une vue d'ensemble des propriétés fonctionnelles, morphologiques et structurelles des différentes glandes et présentons des techniques qui permettent leur distinction. Effect of manufacturing treatments on the rheological character of Mozzarella cheese made from microfiltration retentate depleted of whey proteins,

Low-moisture, part-skim (LMPS) Mozzarella cheeses were made from highly concentrated skim milk microfiltration (MF) retentate and butteroil. Differing combinations of rennet concentration, coagulation temperature and post-coagulation curd cutting time were used, with comparisons made of the rheological and functional characteristics of cheeses during ageing. Lubricated squeezing flow, stress relaxation, melt and stretch measurements demonstrated that commercial LMPS Mozzarella had substantial textural and functional change between 7 and 30 days of age, while the rheological and functional behaviour of LMPS MF Mozzarella (MFM) exhibited substantial change between 30 and 60 days of age. Rennet concentration was the only experimental factor to significantly affect MFM rheological and functional development. The delayed development of MFM was partially attributed to inhibition of normal cheese proteolysis via whey protein (WP) incorporation, lack of starter culture protease, and homogenization of MF cheesemilk. Use of starter culture in MFM led to improved rheological and functional behaviour. [source]


S100A6 (calcyclin) deficiency induces senescence-like changes in cell cycle, morphology and functional characteristics of mouse NIH 3T3 fibroblasts

Abstract S100A6 (calcyclin) is a calcium binding protein with two EF-hand structures expressed mostly in fibroblasts and epithelial cells. We have established a NIH 3T3 fibroblast cell line stably transfected with siRNA against S100A6 to examine the effect of S100A6 deficiency on non-transformed cell physiology. We found that NIH 3T3 fibroblasts with decreased level of S100A6 manifested altered cell morphology and proliferated at a much slower pace than the control cells. Cell cycle analysis showed that a large population of these cells lost the ability to respond to serum and persisted in the G0/G1 phase. Furthermore, fibroblasts with diminished S100A6 level exhibited morphological changes and biochemical features of cellular senescence as revealed by ,-galactosidase and gelatinase assays. Also, S100A6 deficiency induced changes in the actin cytoskeleton and had a profound impact on cell adhesion and migration. Thus, we have shown that the S100A6 protein is involved in multiple aspects of fibroblast physiology and that its presence ensures normal fibroblast proliferation and function. J. Cell. Genetic and epigenetic aspects of bladder cancer

Abstract Transitional cell carcinoma of the urinary bladder has a diverse collection of biologic and functional characteristics. This is reflected in differing clinical courses. The diagnosis of bladder cancer is based on the information provided by cystoscopy, the gold standard in combination with urinary cytology findings. Many tumor markers have been evaluated for detecting and monitoring the disease in serum, bladder washes, and urinary specimens. However, none of these biomarkers reported to date has shown sufficient sensitivity and specificity for the detection of the whole spectrum of bladder cancer diseases in routine clinical practice. The limited value of established prognostic markers requires the analysis of new molecular parameters of interest in predicting the prognosis of bladder cancer patients; in particular, the high-risk patient groups at risk of progression and recurrence. Over the past decade, there has been major progress elucidating of the molecular genetic and epigenetic changes leading to the development of transitional cell carcinoma. This review focuses on the recent advances of genetic and epigenetic aspects in bladder cancer, and emphasizes how molecular biology would be likely to affect the future therapies. © 2005 Wiley-Liss, Inc.
