Can a large metropolis sustain complex herpetofauna communities?

An analysis of the suitability of green space fragments in Rome
Abstract Urban areas are primary causes of species' range fragmentation and reduction. However, relatively few studies have attempted to describe the habitat variables influencing the diversity and conservation of amphibians and reptiles, particularly in Mediterranean Europe and in large metropolitan areas. We explored this broad conservation ecology problem by studying the richness and diversity patterns in relation to a suite of six independent habitat variables in Rome, one of the most ancient cities of the world. We considered all the green remnant areas (n=62) of Rome, ranging 1 to >1000 ha in size, which are interspersed within a sea of urbanized matrix. A total of 10 amphibian and 15 reptile species were studied. Their presence/absence patterns were assessed and the effects of the various habitat variables on each species were predicted by a logistic regression model. A total of 1261 presence records (404 amphibians and 857 reptiles) were analysed. Fragment size and wood size within each fragment did correlate significantly with the species richness of both amphibians and reptiles, and there was a clear threshold effect after 50 ha of wooded surface. The presence of water bodies positively affected the species distribution. One amphibian and three reptiles inhabited exclusively fragments >50 ha. The distance from the centre did not affect fragment species richness. The presence of most species of both amphibians and reptiles was positively influenced by the irregular versus circular shape of the wooded area. The legal protection of a given area did not influence the observed patterns but the total number of sheltered species. Overall, our study suggests that, in order to maintain the current diversity and population viability, it is necessary, in addition to water bodies' maintenance, to (1) preserve the wooded landscapes over 50 ha; (2) promote irregularly shaped increases in the wood surface; (3) maintain ecotonal boundaries. Low Recruitment of Trees Dispersed by Animals in African Forest Fragments

We compared adult and juvenile trees in forest transects in a 3500,ha submontane forest with those in four forest fragments of 521, 30, 9, and 0.5 ha. Preliminary results show that recruitment of seedlings and juveniles of 31 animal-dispersed tree species was more than three times greater in continuous forest and large forest fragments (,30 ha) than in small forest fragments (,9 ha), whereas recruitment of eight wind- and gravity-dispersed trees of the forest interior was unaffected. Recruitment of 10 endemic, animal-dispersed tree species was 40 times lower in small fragments than in continuous forest or large fragments. Counts of diurnal primates and birds in all five sites indicated that frugivorous species have declined with decreasing fragment size. These results are consistent with the idea that loss of dispersal agents depresses tree recruitment in the course of forest fragmentation. Resumen: Investigamos los efectos de la fragmentación del bosque en la desaparición de animales frugívoros y el reclutamiento de árboles dispersados por animales y viento en parches de bosques de 80 años de edad en las montañas del este de Usambara, Tanzania. Comparamos árboles adultos y juveniles en transectos de bosque en un bosque submontañoso de 3500 ha con transectos de cuatro fragmentos de bosque de 521, 30, 9 y 0.5 ha. Los resultados preliminares muestran que el reclutamiento de plántulas y juveniles especies de árboles dispersados por animales fue tres veces mayor en el bosque continuo y fragmentos grandes (,30 ha) que en fragmentos pequeños (,9 ha), mientras que el reclutamiento de ocho árboles dispersados por viento y gravedad del interior del bosque no fue afectado. El reclutamiento de 10 especies endémicas de árboles dispersados por animales fue 40 veces menor en los fragmentos pequeños que en el bosque continuo o en los fragmentos grandes. Los conteos de primates diurnos y aves en los cinco sitios indican que las especies frugívoras han disminuido con la disminución del tamaño del fragmento. Size-related deterioration of semi-natural grassland fragments in Sweden

Abstract. One of the most dramatic landscape changes during the 20th century in Sweden, like in most of Europe, has been the reduction and fragmentation of semi-natural grasslands. Using a set of remnant semi-natural grasslands, chosen to be as similar as possible, but differing in size, we have examined whether size of remnant fragments of traditionally managed semi-natural grasslands in Sweden is related to patterns of species richness and composition. We focused on edge-to-interior relationships, since we expected that a possible impact from invasive habitat generalists would be manifested in a gradient from the edge of fragments to their interior. We found no relationship between size of grassland fragments and (a) overall species richness, (b) species richness at different spatial scales, and (c) abundance of some typical invader species or species characteristic of semi-natural grasslands. However, the results indicated that larger grasslands have a comparatively larger number of species in the edges, whereas the opposite pattern was found in smaller grasslands. The similarity in species composition between the edge and the interior of the pastures also increased with grassland size. Thus, even though the overall species richness is still unaffected by reduction in grassland fragment size, the edges of smaller grasslands show signs of degradation, i.e. reduction in species richness and a decreased similarity to the grassland interior. We suggest that these kinds of effects may be early signs of fragmentation effects that in the future will result in species loss even if the present distribution of semi-natural grasslands is maintained. The effects of edge, fragment size and degree of isolation on avian species richness in highly fragmented forest in West Africa

Almost nothing is known of the effects of forest fragmentation on bird diversity within the heavily degraded and fragmented forest remnants in West Africa. We examined the effects of edge, fragment size and isolation on bird species richness in southwestern Nigeria where forest fragmentation is pronounced. In total, 122 km of line transects were used to survey birds and vegetation within 45 forest patches between January 2000 and March 2002: 197 species were recorded. Avian species number and total counts in forest patches were unrelated to fragment area (within the observed range of 14,445 ha), but were negatively influenced by degree of isolation and increasing distance from the edge. As the total area of forested land within 15 km of a patch fell from 4 to 0%, so 21% of species were lost. In total, six and zero species (of 154 recorded more than once) were consistently recorded in the larger and smaller forest fragments, respectively, and four and two bird species were consistently recorded in unisolated and isolated forest fragments, respectively, suggesting that the addition of ,edge' species did not compensate for loss of species sensitive to fragmentation. Diversity index was not affected by either fragment area or degree of isolation, but decreased with distance from the edge. When individual species counts were considered, 68% of species (n = 62) showed no significant effect of distance to edge. Of those 20 species which showed an effect, 12 were less common close to the edge. Most species (65%) did not respond significantly to increasing isolation but of those 22 species that did, 20 were less common in more isolated fragments. Ninety-seven per cent of species showed no significant response to area. As avian diversity and species composition, but not species number, were apparently insensitive to forest fragmentation, our findings suggest that fragmentation reduces the probability of occurrence of a wide range of West African bird species, rather than a subset of fragmentation-sensitive species. The greater apparent sensitivity of present-day West African forest bird communities to fragmentation rather than patch size might reflect previous extinctions of area-sensitive species. Minimizing further forest fragmentation might be the most effective means of conserving avian diversity in current West African landscapes where most remaining forest patches are small (i.e. < 500 ha). Hair breakage during combing.

Hair breakage during combing was evaluated by combing tresses and examining photographs of snags of hair fibers in combs. The resultant hair fiber arrangements suggest that breakage likely involves hair-on-hair interactions, and broken fragment size suggests that breakage occurs primarily at or near the hair-comb interface. Compression forces during combing were also measured, and impact loading of a hair fiber over another hair vs. a hair fiber over a comb tooth shows that compression and abrasion are important to breakage during combing and that impact loading of one hair fiber over another during snagging is a probable and important pathway for hair breakage. The effects of fragmentation on fluctuating asymmetry in passerine birds of Brazilian tropical forests

Summary 1. ,Fluctuating asymmetry (FA) refers to the difference between the right and left sides in characters that should otherwise be bilaterally symmetrical, but whose expression is affected by epigenetic stress during development. Forest fragmentation may promote an increase in FA in isolated populations, by either genetic or environmental stress. FA may function as a biomonitor index in conservation biology if increased levels were observed in populations from fragmented habitats. 2. ,We tested the hypothesis that FA is higher in birds from small tropical forest patches than in large forest tracts. We measured wing and tarsus FA on mist-netted birds from seven fragments and seven continuous areas from south-eastern Atlantic rain forest in Brazil. We performed FA comparisons between fragments and control groups for the whole community, for individual foraging guilds and for the six most abundant species. 3. ,Wing and tarsus FA were significantly greater in fragments than continuous areas for the whole community and were both negatively correlated with forest fragment size. Differences in FA varied among foraging guilds, being more evident for insectivorous species, especially those feeding in or near the understorey. 4. ,FA levels increased significantly in forest fragments in at least one trait for five of the six most abundant species. There was no correlation between tarsus and wing asymmetries for the individuals of any species nor any difference between the degree of asymmetries of these characters. 5. ,We suggest that FA is a useful tool to assess the effects of fragmentation on forest birds, and may be applied in monitoring neotropical birds. FA indices might be profitably developed, particularly in species most threatened by fragmentation effects and when investigated in different morphological characters. [source]


Bottlenecks, drift and differentiation: the population structure and demographic history of sika deer (Cervus nippon) in the Japanese archipelago

Abstract We assessed genetic differentiation and diversity in 14 populations of sika deer (Cervus nippon) from Japan and four populations of sika deer introduced to the UK, using nine microsatellite loci. We observed extreme levels of differentiation and significant differences in diversity between populations. Our results do not support morphological subspecies designations, but are consistent with previous mitochondrial DNA analyses which suggest the existence of two genetically distinct lineages of sika deer in Japan. The source of sika introduced to the UK was identified as Kyushu. The underlying structure of Japanese populations probably derives from drift in separate glacial refugia and male dispersal limited by distance. This structure has been perturbed by bottlenecks and habitat fragmentation, resulting from human activity from the mid-nineteenth century. Most current genetic differentiation and differences in diversity among populations probably result from recent drift. Coalescent model analysis suggests sika on each of the main Japanese islands have experienced different recent population histories. Hokkaido, which has large areas of continuous habitat, has maintained high levels of gene flow. In Honshu the population is highly fragmented and is likely to have been evolving by drift alone. In Kyushu there has been a balance between gene flow and drift but all the populations have experienced high levels of drift. Habitat fragment size was not significantly associated with genetic diversity in populations but there was a significant correlation between habitat fragment size and effective population size. Gastrointestinal parasites of Howler monkeys (Alouatta palliata) inhabiting the fragmented landscape of the Santa Marta mountain range, Veracruz, Mexico

Abstract In recent years populations of howler monkeys (Alouatta palliata) in southeastern Mexico have decreased substantially due to the transformation and loss of natural habitats. This is especially evident in the Santa Marta mountain range, Veracruz, Mexico where several studies have evaluated the impact of fragmentation on howler monkey populations in order to propose management programs for their conservation. The conditions generated by fragmentation likely change the rates of parasitic infection and could decrease howler survival. In this study, gastrointestinal parasite species richness, prevalence, and egg density of infection were determined in howler groups inhabiting five forest fragments at the Santa Marta mountain range. Two hundred and seventy-eight fresh fecal samples were collected between October 2002 and April 2003. Three parasite species were found during the dry and the wet season in all forest fragments sampled: one unidentified species of Eimeriidae; Trypanoxyuris minutus (Oxyuridae); and Controrchis biliophilus (Dicrocoeliidae). Both the prevalence of T. minutus and infection density for all parasites differed between seasons and fragments (the largest fragment consistently differed from other fragments). Host density, distance to the nearest town, fragment size, fragment shape, and total basal area of food trees explained parasite prevalence, but each species had a different pattern. Although parasite richness was lower, prevalence and density were higher than values reported for howlers in conserved forests. These results suggest that the establishment of biological corridors and animal translocation programs must take into account the parasite ecology of each fragment to avoid higher infection rates and preclude potential consequent mortality. The species,area relationship and confounding variables in a threatened monkey community

Abstract This study investigates the species,area relationship (SAR) for forest monkeys in a biodiversity hotspot. The Udzungwa Mountains of Tanzania are well-suited to investigate the SAR, with seven monkey species in a range of fragment sizes (0.06,526,km2). We test the relationship between species richness and forest fragment size, relative to human and environmental factors. We distinguish resident and transitory species because the latter have an "effective patch size" beyond the area of forest. Forest area was the strongest (log-linear) predictor of species richness. However, forest area, elevation range and annual moisture index were intercorrelated. Previous knowledge of the relationship between elevation and tree communities suggests that the SAR is largely a result of habitat heterogeneity. Isolation by farmland (matrix habitat) also had a significant negative effect on species richness, probably exacerbated by hunting in small forests. The effect of area and isolation was less for transitory species. The human influence on species' presence/absence was negatively related to the extent of occurrence. Weaker relationships with temperature and precipitation suggest underlying climatic influences, and give some support for the influence of productivity. A reduced area relationship for smaller forests suggests that fragment sizes below 12,40,km2 may not be reliable for determining SAR in forest monkeys. Further practical implications are for management to encourage connectivity, and for future SAR research to consider residency, matrix classification and moisture besides precipitation. Am. J. Modelling species distribution at multiple spatial scales: gibbon habitat preferences in a fragmented landscape

Abstract Conservation of Indochinese primates is hampered by a lack of knowledge of species ecology, habitat preferences and, locally, distribution. Predictive distribution and habitat suitability models, using predictors known to affect the distribution of similar species elsewhere, may, therefore be of great benefit to conservationists within the region. Yellow-cheeked crested gibbon Nomascus gabriellae is an IUCN-listed endangered primate distributed east of the Mekong River in Cambodia, southern Vietnam, and possibly southern Lao PDR. Within Cambodia, yellow-cheeked crested gibbon are naturally restricted to evergreen forest fragments within a landscape matrix of deciduous dipterocarp and semi-evergreen forests. During the 2008 dry season, auditory surveys for yellow-cheeked crested gibbon were conducted within Phnom Prich Wildlife Sanctuary, Mondulkiri province, Cambodia. Predictive distribution models, in which variables were included at the scale at which they best explained gibbon occurrence (multi-grain models), were constructed to examine the species' habitat associations and tolerance of habitat fragmentation within the wildlife sanctuary. Gibbon occupancy (,) was higher in evergreen (0.43±0.26,0.62) than in semi-evergreen forest (0.21±0.09,0.4), with gibbon presence constrained by a critical amount of evergreen forest within 5 km radius of listening posts. Three patches of optimal habitat within Phnom Prich Wildlife Sanctuary were identified. These, and connecting habitats, should be the target of increased enforcement effort to limit hunting and habitat conversion. Similar multi-grain models are likely to be valuable for conservationists within mosaic habitats as they can facilitate identification of the minimum suitable fragment size for species of conservation concern. Investigating the distribution of prairie dogs in an urban landscape

Abstract Habitat fragmentation is a prevalent threat to biological diversity, and urbanization is a primary agent of fragmentation and a leading cause of species endangerment. Landscape biogeographic and local habitat characteristics can be important determinants of the distribution of species in habitat patches in urban landscapes. However, the specifics of which characteristics are most critical to maintaining biological diversity are not fully known for prairie ecosystems, especially in fragmented urban habitat. This study focuses on black-tailed prairie dogs along an urban gradient in Denver, CO. Prairie dogs have declined precipitously throughout the region and are an essential part of the prairie ecosystem, making them excellent study subjects. We identified a series of habitat fragments along a gradient of urbanization in the fully urbanized areas and south suburbs of Denver, CO, both containing and not containing prairie dogs. Local characteristics, including fragment slope and vegetative cover, and landscape characteristics, including fragment size, age and connectivity, were measured on each fragment. We used likelihood-based methods to explore which variables most accurately predicted prairie dog occurrence within our study area. Multiple factors influenced the distribution of prairie dogs in urban settings, with colony connectivity the strongest predictor of occupancy. Large and recently isolated fragments near other prairie dog colonies, flat areas and those with high graminoid cover were most likely to support prairie dog populations. Our study provides the first attempt to model prairie dog occurrence in highly fragmented urban habitat and has important implications for the management and conservation of prairie dogs. [source]


The response of tree squirrels to fragmentation: a review and synthesis

Habitat fragmentation is often considered a major threat to biodiversity; however, our understanding of how fragmentation impacts populations is poor. Identifying appropriate models for such studies is difficult. Tree squirrels are dependent on mature forests for food, cover and nests; these are habitats that are being fragmented rapidly and that are easily defined by humans. Squirrels represent excellent models for study of fragmentation. The literature on tree squirrels was reviewed to glean data on density and home-range size in forest fragments. Sufficient data were available on four species (Sciurus carolinensis, S. niger, S. vulgaris, Tamiasciurus hudsonicus). Density was negatively related to fragment size for S. carolinensis and S. niger and marginally so for T. hudsonicus. Sciurus vulgaris did not exhibit this relationship. Home-range size was analysed for three species of Sciurus and was positively related to forest fragment size for S. carolinensis and S. niger. Again, only S. vulgaris did not to show this relationship. Sciurus vulgaris is rarely found in small forest fragments and is believed to be especially sensitive to fragmentation; other tree squirrels appear to be sensitive to fragmentation in more subtle ways. Home range compaction provides a mechanism by which densities may increase in small fragments. The demographic consequences resultant from the high densities of squirrels found in small woodlots are not known but may explain the forest damage, avian nest predation and reduced diversity often cited to occur in woodland fragments. A proposal for standardization in forensic equine DNA typing: allele nomenclature for 17 equine-specific STR loci

Summary In this study, a proposal is presented for the allele nomenclature of 17 polymorphic STR loci (AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, HTG10, LEX3 and VHL20) for equine genotyping (Equus caballus). The nomenclature is based on sequence data of the polymorphic region of the STR loci as recommended by the DNA commission of the International Society for Forensic Genetics for human DNA typing. For each STR locus, several alleles were selected and animals homozygous for those alleles were subjected to sequence analysis. The alleles of the 17 STR loci consisted either of simple (10), compound (6) or complex repeat patterns (1). Only a limited number of alleles with the same fragment size showed different repeat structures. The allele designation described here was based on the number of repeats, including all variable regions within the amplified fragment. [source]

To cover commonly and rarely occurring alleles, a selection of animals homozygous for the alleles at these STR loci were analysed and subjected to sequence studies. The alleles of the STR loci consisted either of simple or compound dinucleotide repeat patterns. Only a limited number of alleles with the same fragment size showed different repeat structures. The allele designation described here was based on the number of repeats including all variable regions within the amplified fragment. The set of 16 STR markers should be propagated for the use in all bovine applications including forensic analysis. [source]


Characteristics of PCR fragments amplified using five microsatellite markers for identifying the Nagoya breed

ABSTRACT The Nagoya breed is a native chicken of Aichi Prefecture, Japan, a dual-purpose breed for eggs and meat. A method for distinguishing the Nagoya breed from Aichi Prefecture from other chickens using five microsatellite markers (ABR0015, ABR0257, ABR0417, ABR0495 and ADL0262) has already been utilized in order to check the authenticity of Nagoya breed-labeled chicken on the market. The present study was conducted to investigate nucleotide sequences and sizes of PCR fragments containing the five microsatellite regions for the Nagoya breed and to confirm that the genomic identification could continue to be applied in the future. The DNA sequencing of fragments containing the five markers showed that ABR0015, ABR0417 and ABR0495 had a single haplotype, ABR0257 had three haplotypes, and ADL0262 had two haplotypes, although all the markers exhibited one fixed fragment size each upon sequencing of the fragments and fragment analysis. The results of the fragment analysis of each marker using DNA samples of 28 Nagoya breed males (G0 generation) reared in 2000,2001 and 20 of their offspring males (G8) reared in 2008,2009 showed one fixed fragment size in both populations. Therefore, we confirmed that the five microsatellite markers are useful tools for accurately distinguishing the Nagoya breed from other chickens. [source]


Mating success of the endemic Des Murs' Wiretail (Sylviorthorhynchus desmursii, Furnariidae) in fragmented Chilean rainforests

Abstract: We studied the effects of fragment size, vegetation structure and presence of habitat corridors on the reproductive success of the Des Murs' Wiretail (Sylviorthorhynchus desmursii Des Murs, Furnariidae), a small (10 g) understorey bird, endemic to South American forests. In a rural landscape of Chiloé Island, southern Chile (42°S; 70°W), we determined the mating and nesting success of wiretails in 28 territories distributed in seven small (1,20 ha) and two large (>300 ha) forest fragments during the 1997,1998 breeding season. Wiretails inhabited dense bamboo thickets in the understorey of forest patches, dense shrublands covering old fields, and dense early successional forest vegetation. Wiretails avoided open pastures. Reproductive success depended solely on the probability of finding mates, and the main factor affecting mating success was the presence of corridors. Mated individuals occupied 72% of the territories in forest patches <20 ha connected by corridors, 73% of the territories in large (>300 ha) fragments, but only 20% of territories in isolated fragments surrounded by pastures. Because of the rapid expansion of pastures in southern Chile, the conservation of wiretails and other understorey birds will depend on the maintenance of travel corridors with dense understorey vegetation between forest fragments. Forest Fragment and Breeding Habitat Characteristics Explain Frog Diversity and Abundance in Singapore

ABSTRACT Habitat loss and fragmentation can have severe negative and irreversible effects on biodiversity. We investigated the effects of forest fragmentation on frog diversity in Singapore because of its high rates of deforestation and the demonstration that frogs are some of the most sensitive species to habitat degradation. We surveyed frog species in 12 forest fragments varying from 11 to 935 ha. We compared differences in species richness, abundance, and Shannon's index in relation to forest fragment size, connectivity (distance between fragments), and breeding habitat heterogeneity. A total of 20 species from 12 genera and five families were encountered in 12 fragments. Larger fragments and those closer to larger fragments had higher species richness. Abundance, however, was not correlated with forest area or connectivity, but we found fewer individual frogs in the larger fragments. We also found that breeding habitat heterogeneity best explained frog species diversity and abundance in forest fragments. Fragments with a high diversity of breeding habitats had more species. We found no evidence to suggest that abundance and diversity are strongly correlated, particularly in disturbed areas, but that breeding habitat heterogeneity is an under-appreciated factor that should be considered when prioritizing areas for anuran conservation. Enriching breeding habitat heterogeneity, creating corridors between fragments, and reforesting degraded areas are some of the most beneficial strategies for preserving urban frog biodiversity. ABSTRACT Very little is known about the diversity of arthropods in the fast-disappearing fragments of natural forests in sub-Saharan Africa. This study investigated: (1) the influence of forest fragment characteristics on dung beetle species richness, composition, abundance, and diversity; and (2) the relationship between dung beetle assemblages and rainfall pattern. Beetles were sampled through 12 mo using dung baited pitfall traps. A total of 18,073 dung beetles belonging to three subfamilies and 45 species were captured. The subfamily Scarabaeinae was the most abundant (99%) and species rich (89%). Fast-burying tunnellers (paracoprids) were the most dominant functional group. Catharsius sesostris, Copris nepos, and Heliocopris punctiventris were the three most abundant species, and had the highest contributions to dissimilarities between forests. With few exceptions, dung beetle abundance, species richness, and diversity were generally higher in larger forest fragments (100,150 ha) than in smaller ones (10,50 ha) and the nature reserve (1042 ha). Forest fragment size had a highly significant positive relationship with beetle abundance, but only when the nature reserve is excluded in the analysis. Dung beetle abundance and species richness showed direct weak relationships with litter depth (positive) and groundcover (negative) but not tree density, tree species richness, and fragment isolation distance. Dung beetle abundance and species richness were strongly correlated with monthly changes in rainfall. Results of this study indicate that forest fragments on agricultural lands in the Budongo landscape, especially medium-sized (100,150 ha) ones, represent important conservation areas for dung beetles. Facioscapulohumeral muscular dystrophy: epidemiological and molecular study in a north-east Italian population sample

Facioscapulohumeral muscular dystrophy (FSHD) is an autosomal dominant disease associated with a partial deletion on chromosome 4q35. Few relevant investigations have been reported on its epidemiology and were essentially based on clinical diagnosis, having been performed before recognition of the molecular mutation. We report an epidemiological survey on FSHD patients, in which the diagnosis was obtained by combined clinical and molecular evaluation. The survey concerned the north-east Italian province of Padova, an area of 871,190 inhabitants (1 January 2004). We identified 40 patients affected by FSHD based on clinical diagnosis. In 33 of them, the EcoRI fragment size in the 4q35 region ranged from 14 to 35 kb. Four other patients belonging to the same family harbored a 38-kb fragment. In these four cases, the relationship between the borderline deletion with the mild FSHD phenotype was corroborated by additional haplotype reconstruction and segregation analysis. Interestingly, the same mild facial-sparing clinical pattern was apparent only in one other patient with an EcoRI fragment of 32 kb, suggesting that this unusual FSHD phenotype may be due to very small 4q35 deletions. On the whole, estimating a prevalence rate of 44 × 10,6, our survey confirmed FSHD as one of the most frequent neuromuscular disorders in Western populations. [source]


Reactive oxygen species induce RNA damage in human atherosclerosis

Abstract Background, Reactive oxygen species (ROS)-induced DNA damage has recently been identified in both human and experimental atherosclerosis. This study was undertaken to investigate whether RNA damage occurs in human atherosclerotic plaques and whether this could be related to oxidative stress. Materials and methods, The integrity of total RNA isolated from carotid endarterectomy specimens (n = 20) and nonatherosclerotic mammary arteries (n = 20) was analyzed using an Agilent 2100 Bioanalyser (Agilent Technologies, Palo Alto, CA). Oxidative modifications of RNA were detected by immunohistochemistry. Results, Eleven out of 20 atherosclerotic plaques showed a significant reduction of the 18S/28S rRNA peaks and a shift in the RNA electropherogram to shorter fragment sizes. In contrast, all mammary arteries showed good-quality RNA with clear 18S and 28S rRNA peaks. Strong nuclear and cytoplasmic immunoreactivity for oxidative damage marker 7,8-dihydro-8-oxo-2,-guanosine (8-oxoG) could be detected in the entire plaque in smooth muscle cells (SMCs), macrophages and endothelial cells, but not in SMCs of adjacent normal media or in mammary arteries. Cytoplasmic 8-oxoG staining in the plaque clearly diminished when tissue sections were pretreated with RNase A, suggesting oxidative base damage of RNA. In vitro treatment of total RNA with ROS-releasing compounds induced RNA degradation. Conclusion, Both loss of RNA integrity and 8-oxoG oxidative modifications were found in human atherosclerotic plaques. Because RNA damage may affect in vitro transcript quantification, RT-PCR results must be interpreted cautiously if independent experimental validation (e.g. evaluation of RNA integrity) is lacking. Degradation of transgenic Cry1Ab DNA and protein in Bt-176 maize during the ensiling process

Summary Maize silage is commonly used as feed for farm animals. The aim of this study was to monitor the time-dependent degradation of non-recombinant chloroplast DNA (exemplified by the rubisco gene) in comparison with the recombinant cry1Ab gene in the course of the ensiling process. In parallel, the Cry1Ab protein content and fragment sizes were determined. Fragments of the rubisco (173, 896, 1197, 1753 and 2521 bp) and of the cry1Ab gene (211, 420, 727 and 1423 bp) were selected to investigate the DNA degradation process. The detection of the Cry1Ab protein was performed using an enzyme-linked immunosorbent assay (ELISA) and immunoblotting. Rubisco gene fragments of 173 bp were still detectable after 61 days, while fragments of 1197 and 2521 bp were detectable up to 30 days and on the first day only respectively. Polymerase chain reaction (PCR) analyses revealed that fragments of the cry1Ab gene with sizes of 211 and 420 bp were detectable up to 61 days, fragments with sizes of 727 and 1423 bp, 30 and 6 days respectively. The ELISA showed a decrease of the Cry1Ab protein in maize silage during the ensiling process. No marked degradation was observed during the first 43 h. Thereafter, a sharp decrease was measured. After 61 days, 23.6 ± 0.9% of the initial Cry1Ab protein was still detectable. Immunoblotting confirmed the results of the ELISA showing a positive signal of approximately 60 kDa size for 8 days of ensiling; no further immunoactive fragments were detectable by immunoblotting. In conclusion, the ensiling process markedly decreases the presence of long functional cry1Ab gene fragments and full size Cry1Ab protein. [source]


Genetic analysis of ratmouth barbell (Ptychidio jordani Myers) from different geographic sub-populations in the Pearl River Basin using microsatellite markers

Summary Thirteen polymorphic microsatellite markers were used to study the genetic variation of Ptychidio jordani sampled from the three sites of Liuzhou, Guiping and Yunan in the Pearl River Basin with the help of local fishers. Allele numbers ranged from 2 to 19 and fragment sizes ranged from 108 to 288. Average polymorphism information content (PIC), average observed heterozygosity (Ho) and average expected heterozygosity (He) were 0.439917, 0.4147 and 0.4986, respectively, which indicate a comparatively high level of genetic diversity. The coefficient of genetic differentiation (Gst) between the two sub-populations ranged from 0.0074 to 0.0156, which shows little differentiation between sub-populations. amova analysis also offered evidence of weak differentiation with 0.53% variation between sub-populations, 99.47% variation within sub-populations and the Fst = 0.00531. The morphological variation among geographic sub-populations might result from different habitats. The correlation between genotype and morphological characters was analyzed and a significant correlation found between the genotype of microsatellite locus Hlj038 and the ratio of body depth and body length. The results suggest a possible linkage between this locus and these morphological characters. [source]


The species,area relationship and confounding variables in a threatened monkey community

Abstract This study investigates the species,area relationship (SAR) for forest monkeys in a biodiversity hotspot. The Udzungwa Mountains of Tanzania are well-suited to investigate the SAR, with seven monkey species in a range of fragment sizes (0.06,526,km2). We test the relationship between species richness and forest fragment size, relative to human and environmental factors. We distinguish resident and transitory species because the latter have an "effective patch size" beyond the area of forest. Forest area was the strongest (log-linear) predictor of species richness. However, forest area, elevation range and annual moisture index were intercorrelated. Previous knowledge of the relationship between elevation and tree communities suggests that the SAR is largely a result of habitat heterogeneity. Isolation by farmland (matrix habitat) also had a significant negative effect on species richness, probably exacerbated by hunting in small forests. The effect of area and isolation was less for transitory species. The human influence on species' presence/absence was negatively related to the extent of occurrence. Weaker relationships with temperature and precipitation suggest underlying climatic influences, and give some support for the influence of productivity. A reduced area relationship for smaller forests suggests that fragment sizes below 12,40,km2 may not be reliable for determining SAR in forest monkeys. Further practical implications are for management to encourage connectivity, and for future SAR research to consider residency, matrix classification and moisture besides precipitation. Am. J. Demography of an Afrotropical passerine in a highly fragmented landscape

Abstract Demographic attributes of discrete subpopulations of animals and plants that constitute a larger (meta)population network may affect the strength and direction of local population responses to habitat loss or degradation. To address this question in an Afrotropical context, we studied survival rates, population densities, sex ratios and age distributions in seven white-starred robin Pogonocichla stellata populations inhabiting differently sized forest remnants in a highly fragmented Kenyan landscape. Sex ratios were strongly male biased, especially during the non-breeding season, but the level of bias did not differ between age groups nor fragment sizes. Juvenile to adult ratios were smallest in the medium-sized fragment, but did not differ between the largest and smallest fragments. Low population density combined with a skewed sex ratio in the medium-sized fragment pointed towards a local scarcity of females, which was supported by the presence of unmated territorial males. Based on capture,recapture analysis, all populations were considered stable on average. When combining demographic patterns with those emerging from a recent population genetic study and removal experiment, our results support the notion that small populations inhabiting tiny habitat remnants may play an important role in augmenting the long-term survival of spatially structured populations.
